Abstract:Fe3O4@C nanocapsules were synthesized via a sacrificial-template method by coating SiO2 nanospheres with an Fe3O4@C double-shell structure, followed by dissolving SiO2 nanospheres through ammonia water under hydrothermal conditions. The nanocapsules show a loading capacity as high as 1300 mg g-1 for doxorubicin (DOX), and the DOX loaded on the surface of the carbon shells displays pH-sensitive release behavior. Drug release experiments were carried out at pH 7.4, 6.2 and 5.0. It was found that the drug release rate at pH 6.2 was about two times as fast as that at pH 7.4, and even faster at pH 5.0. A MTT assay was used to test the cytotoxicity of the DOX, nanocapsules and DOX-nanocapsules, which indicated the low cytotoxicity of the nanocapsules towards cells. The DOX-nanocapsules can be taken up by cancer cells through endocytosis, releasing DOX into the cytoplasm, which was observed by both transmission electron microscopy and confocal microscopy. In vitro magnetic resonance imaging (MRI) experiments validated the potential use of these nanocapsules as MRI contrast agents.
A novel high doxorubicin-loaded Fe3O4@void@ZnO nanocomposite: pH-controlled drug release and targeted antitumor activity

Abstract:A novel ferroferric oxide (Fe3O4)@void@zinc oxide (ZnO) nanocomposite was constructed through a template-assisted selective etching method. The experiment results showed that the magnetic saturation value of nanocomposite could reach to 44.07 emu g−1, indicating the favorable magnetic targeting function. The presence of the void endowed the nanocomposite with high drug doxorubicin (DOX) loading capacity (85.65 μg mg−1). The pH-sensitive drug release was realized due to the gating effect of ZnO, and the DOX release rates were 92.7%, 60.8% and 23.4% at the pH values of 5.0, 6.5 and 7.4, respectively (37 °C). Besides, the Fe3O4@void@ZnO-DOX exhibited desirable antitumor effects attributed to their cellular uptake and above advantages. Therefore, the Fe3O4@void@ZnO nanocomposite may serve as a promising drug carrier in cancer therapy and other biomedical applications.

Abstract:Nanoplatforms used for the loading of anticancer drugs constitute a promising approach to cancer treatment and reducing the side effects of these drugs. Among the cutting-edge systems used in this area are magnetic nanocomposites (MNCs) and nanocapsules (NCs). MNCs are considered to constitute a smart tool for magnetic-field-guided targeted drug delivery, magnetic resonance imaging, and hyperthermia therapy. Nanocapsules offer great potential due to their ability to control drug-loading capacity, their release efficiency, their stability, and the ease with which their surfaces can be modified. This study proposes a method for the development of nylon-6-coated MNCs and nylon-6 polymeric membrane NCs. A biocompatible nylon-6 polymer was first used for NC synthesis. Oleic-acid-modified and non-modified Fe3O4 nanoparticles were synthesized for the production of nylon-coated MNCs. Dynamic light scattering (DLS), transmission electron microscopy (TEM), and ζ-potential measurements were used to perform size, morphology, and charge analyses. The above-mentioned two types of MNCs were considered templates for the manufacture of nylon nanocapsules, leading to NCs with different charges and structures. The developed oleic-acid-coated nylon-6 MNCs and NCs showed excellent loading values of the chemotherapy drug doxorubicin (DOX) of up to 732 and 943 μg/mg (DOX/MNC or NC), respectively. On the contrary, the capacity of the nano-construction that was not modified with oleic acid did not exceed 140 μg/mg. The DOX-loaded nanosystems displayed pH-sensitive drug release properties, for which the highest efficiency was observed at an acidic pH. The series of DOX-loaded MNCs and NCs inhibited A549 and HEK 293FT cell lines, with the lowest IC50 value of 0.31 μM observed for the nanocapsules, which is a 1.5-fold lower concentration than the free DOX. Therefore, the presented nanoscale systems offer great potential for cancer treatment.

Abstract:Due to the limited efficacy and evident side effects of traditional chemotherapy drugs attributed to their lack of specificity and selectivity, novel strategies are essential for improving cancer treatment outcomes. Here, we successfully engineered Fe3O4 magnetic nanoparticles coated with zeolitic imidazolate framework-8 (ZIF-8). The resulting nanocomposite (Fe3O4@ZIF-8) demonstrates efficient adsorption of a substantial amount of doxorubicin (DOX) due to the porous nature of ZIF-8. The drug-loaded nanoparticles, Fe3O4@ZIF-8/DOX, exhibit significant accumulation at the tumor site in SW620 colon-cancer-bearing mice when guided by an external magnetic field. Within the acidic microenvironment of the tumor, the ZIF-8 framework collapses, releasing DOX and effectively inducing tumor cell death, thereby inhibiting cancer progression while not causing undesired side effects, as confirmed by a variety of in vitro and in vivo characterizations. In comparison to free DOX, Fe3O4@ZIF-8/DOX nanoparticles show superior efficacy in colon cancer treatment. Our findings suggest that Fe3O4@ZIF-8 holds promise as a carrier for small-molecule drug adsorption and its ferromagnetic properties provide drug targeting capabilities, thereby enhancing therapeutic effects on tumors at the same drug dosage. With excellent biocompatibility, Fe3O4@ZIF-8 demonstrates potential as a drug carrier in targeted cancer chemotherapy. Our work suggests that a combination of magnetic targeting and acid-responsiveness holds great promise for advancing targeted cancer therapy in precision nanomedicine.

Abstract:This study describes the development of an innovative pH-sensitive drug delivery system (DDS) utilizing ferrocene (Fc) and magnetic nanoparticles (MNPs) for cancer therapy. One potential method for generating a diverse range of functional NPs involves attaching functional substances to MNPs. In this investigation, we display the successful modification of Fe 3 O 4 NPs by an Fc derivative (4-Ferrocenylbutyl)dimethylsilane through a hydrosilylation reaction toward the synthesis of the Fe 3 O 4 @SiO 2 @VTES@Fc nanocarrier (NC). The doxorubicin (DOX) drug was loaded onto Fe 3 O 4 @SiO 2 @VTES@Fc NC with a drug loading capacity of about 80 % via π-π stacking interactions. Also, the prepared materials were thoroughly characterized using FT-IR, XRD, FE-SEM, EDX-MAP, VSM, DAPI, MTT, antioxidants, and CV techniques. In vitro studies on the release of DOX from Fe 3 O 4 @SiO 2 @VTES@Fc NC indicated a pH-responsive behavior with a lower release rate (< 10 %) at pH 7.4 compared to pH 5 (approximately 70 %) over a period of 96 h. Moreover, cytotoxicity assessments demonstrated notable cytotoxic effects of DOX-loaded Fe 3 O 4 @SiO 2 @VTES@Fc on MCF-7 cells (IC50 value: ∼8 μg/mL). The study findings suggest that the fabricated magnetic core-shell NC (Fe 3 O 4 @SiO 2 @VTES@Fc/DOX) exhibits stimulus-responsive behavior and holds promise as a sophisticated approach to specifically transport therapeutic agents into tumor sites while minimizing exposure to healthy tissues.

Abstract:Construction of multifunctional stimuli-responsive nanotherapeutics enabling improved intratumoral penetration of therapeutics and reversal of multiple-drug resistance (MDR) is potent to achieve effective cancer treatment. Herein, we report a general method to synthesize pH-dissociable calcium carbonate (CaCO3) hollow nanoparticles with amorphous CaCO3 as the template, gallic acid (GA) as the organic ligand, and ferrous ions as the metallic center via a one-pot coordination reaction. The obtained GA–Fe@CaCO3 exhibits high loading efficiencies to both oxidized cisplatin prodrug and doxorubicin, yielding drug loaded GA–Fe@CaCO3 nanotherapeutics featured in pH-responsive size shrinkage, drug release, and Fenton catalytic activity. Compared to nonresponsive GA–Fe@silica nanoparticles prepared with silica nanoparticles as the template, such GA–Fe@CaCO3 confers significantly improved intratumoral penetration capacity. Moreover, both types of drug-loaded GA–Fe@CaCO3 nanotherapeutics exhibit synergistic therapeutic efficacies to corresponding MDR cancer cells because of the GA–Fe mediated intracellular oxidative stress amplification that could reduce the efflux of engulfed drugs by impairing the mitochondrial-mediated production of adenosine triphosphate (ATP). As a result, it is found that the doxorubicin loaded GA–Fe@CaCO3 exhibits superior therapeutic effect towards doxorubicin-resistant 4T1 breast tumors via combined chemodynamic and chemo-therapies. This work highlights the preparation of pH-dissociable CaCO3-based nanotherapeutics to enable effective tumor penetration for enhanced treatment of drug-resistant tumors.

Abstract:Achieving accumulation of a target drug in tumor tissue, optimal release behavior and concurrent diagnostic imaging with therapy (theranostic) requires an ideal chemotherapy protocol that involves the responsive control of drug delivery. In the current study, we report the construction of a novel theranostic drug delivery system based on radial mesoporous silica, which is hybridized with multiscale magnetic nanoparticles for MRI-guided and alternative magnetic field (AMF) responsive chemotherapy for breast cancer. Superparamagnetic iron oxide nanoparticles (IONPs) with multiscale sizes were prepared via a hydrothermal method (for larger IONPs, lIO NPs) and a thermal decomposition method (for ultra-small IONPs, uIO NPs). lIO NPs act as the heat mediator that is responsive to the AMF, while uIO NPs act as the T-2 contrast agent for MRI. Mesoporous silica nanoparticles with radially oriented mesochannels were further in situ grown onto the surfaces of the lIO NPs, and both the uIO NPs and doxorubicin, an anti-cancer drug, can be readily incorporated within the mesochannels. To endow the as-prepared doxorubicin-loaded nano-hybrids with a good targeting ability, the surfaces of the particles were modified with folic acid (FA). This prepared drug delivery system based on magnetic/silica nanocomposites showed high drug loading efficiency and AMF stimuli responsive release properties. Meanwhile, in vitro MRI measurements revealed an excellent MRI contrast effect with a high relaxation value of 308 mM(-1) s(-1). Furthermore, both in vitro cytotoxicity analysis and in vivo anti-tumor effect evaluation demonstrated that the drug delivery system possesses great potential as an MRI-guided stimuli-responsive theranostic platform for effective active targeting chemotherapy of cancer.

Abstract:Fabrication of magnetic nanocarriers that demonstrate enhanced biocompatibility and excellent colloidal stability is critical for the application of magnetic-motored drug delivery, and it remains a challenge. Herein, a novel approach to synthesize mesoporous magnetic colloidal nanocrystal clusters (MMCNCs) that are stabilized by agarose is described; these clusters demonstrate high magnetization, large surface area and pore volume, excellent colloidal stability, enhanced biocompatibility, and acid degradability. The hydroxyl groups of agarose, which cover the surface of the magnetic nanocrystals, are modified with vinyl groups, followed by click reaction with mercaptoacetyl hydrazine to form the terminal hydrazide (-CONHNH(2)). The anticancer agent doxorubicin (DOX) is then conjugated to MMCNCs through a hydrazone bond. The resulting hydrazone is acid cleavable, thereby providing a pH-sensitive drug release capability. This novel carrier provides an important step towards the construction of a new family of magnetic-motored drug-delivery systems. The experimental results show that the release rate of DOX from the DOX-conjugated MMCNCs (MMCNCs-DOX) is dramatically improved at low pH (tumor cell: pH 4-5 in the late stage of endolysosome and pH 5-6 from the early to late endosome), while almost no DOX is released at neutral pH (blood plasma). The cell cytotoxicity of the MMCNCs-DOX measured by MTT assay exhibits a comparable antitumor efficacy but lower cytotoxicity for normal cell lines, when measured against the free drug, thus achieving the aim of reducing side effects to normal tissues associated with controlled drug release.

Abstract:Doxorubicin (DOX) is an effective and widely used cancer chemotherapy drug, though its use is extremely dangerous due to side effects such as cardiotoxicity resulting from high doses. Herein, a sustainable and biocompatible starch/polyvinyl pyrrolidone/hydroxyapatite nanocomposite was prepared and loaded with DOX by water-in-oil-in water (W/O/W) emulsification method. Fourier-transform infrared spectroscopy (FT-IR) and X-ray diffraction (XRD) confirmed the successful synthesis of the nanocomposite and indicated strong interactions between its components. Scanning electron microscopy (SEM) images revealed the quasi-spherical shape of the nanocomposite and dynamic light scattering (DLS) showed an average particle size of 258 nm. The zeta potential at pH 5.4 was −37.28 mV, corroborating its colloidal stability. Doxorubicin release in the ex vivo environment was investigated at pH 5.4 and 7.4, mimicking tumour and physiological conditions, respectively, and the results showed no initial burst release and a faster release kinetics in the acidic environment owed to the repulsive forces among protonated oxygenated groups of the drug and the polymers. The W/O/W improved entrapment efficiency and enabled a gradual DOX release, thus increasing the drug stability. The cell biocompatibility and anticancer activity of the drug-loaded nanocomposite were proved by MTT analysis on breast cancer fibroblast cells (7-MCF). The synthesized nanocomposite shows great potential as an effective nanocarrier of anticancer drugs for controlled release in the acidic environment of the tumour. Abstract:The challenging part of this work was to research the potential aspects of sodium alginate (SA)-polyvinyl alcohol (PVA)-bovin serum albumin (BSA) coated Fe3O4 nanoparticles (Fe3O4-SA-PVA-BSA) as a drug delivery system for doxorubicin (DOX). The anticancer drug doxorubicin was selected as a model drug which is powerful for numerous cancer treatments. Superparamagnetic Fe3O4 nanoparticles were prepared by co-precipitation method. The mixture solution of Fe3O4-sodium alginate (SA) - doxorubicin (DOX) was crosslinked with Ca2+ to form (Fe3O4-SA-DOX) nanoparticles and addition of PVA and BSA with (Fe3O4-SA-DOX) nanoparticles was prepared by coating procedure. Doxorubicin drug loaded NPs were prepared by a simple crosslinking method by calcium chloride solution. The prepared polymer coated magnetic nanoparticles (Fe3O4-SA-PVA-BSA) were characterized by using SEM, AFM, FT-IR, XRD and VSM. The mean sizes of the obtained drug loaded nanoparticles (Fe3O4-SA-DOX, Fe3O4-SA-DOX-PVA and Fe3O4-SA-DOX-PVA-BSA) were between 240±8.3 and 460±8.7nm and zeta potential of the particles also was evaluated using Malvern Zetasizer which ranged between -48.1±2.3 and -22.4±4.1mV. The encapsulation efficiency, was between 36.2±0.01 and 96.45±2.12. Moreover drug loading and drug release properties of the polymer coated magnetic nanoparticles loaded with doxorubicin (Fe3O4-SA-DOX-PVA-BSA) were also studied. In addition, the cytotoxicity of the created nanoparticles was performed by using MTT assay analysis which showed that DOX loaded nanoparticles (Fe3O4-SA-DOX-PVA-BSA) were toxic to HepG2 cell lines and non-toxic to L02 cell lines. The in-vitro drug release was studied by using UV-Visible spectrophotometer at acidic environment (pH5.0) and basic environment (pH7.4) as well as at different temperatures (37°C and 42°C). It was found that DOX drug is released much faster in acidic environment (pH5.0) than in the basic environment (pH7.4). The results propose that prepared polymer coated magnetic (Fe3O4-SA-PVA-BSA) nanoparticles are suitable for controlled and targeted release of anticancer drugs reducing side effects and attaining higher efficacy.

Abstract:In this paper, a novel pH-responsive drug delivery system based on hierarchical hollow mesoporous MnFe 2 O 4 nanospheres was developed for potential application in cancer treatment. The well-defined hollow mesoporous MnFe 2 O 4 nanospheres were directly fabricated via a facile one-step solvothermal approach without the involvement of additional templates. A mechanism of inside-out Ostwald ripening was suggested to account for the formation of hollow mesoporous nanostructures. The morphology, crystal structure, chemical composition, magnetic properties, thermal stability and mesoporous characteristics of the fabricated MnFe 2 O 4 nanospheres were comprehensively assessed based on various analysis techniques. The resulting hollow mesoporous MnFe 2 O 4 nanospheres with a high biocompatibility were able to be served as a nanocarrier for loading and delivering of anticancer drug doxorubicin (DOX). In vitro drug release study revealed that DOX release from DOX-loaded MnFe 2 O 4 nanospheres was significantly restrained in pH 7.4 phosphate buffer solution (PBS), while it was markedly accelerated in pH 4.5 PBS. The results of cytotoxicity assay and inverted biological microscopy demonstrated that the DOX-loaded MnFe 2 O 4 nanospheres were more cytotoxic against HepG2 cells than MnFe 2 O 4 nanospheres due to the evident inhibition effect of released DOX from the constructed drug delivery system. Therefore, the developed pH-responsive delivery system will have a great potential for its practical application in cancer treatment.

Abstract:This article presented a facile two-step hydrothermal strategy to anchor the Fe3O4 nanoparticles on the surface of ZnAl LDH nanosheets, and investigated systematically the potential application of the constructed Fe3O4/Zn-Al LDH nanocomposites in the biomedical field of controlled drug delivery. It was revealed that the formed Fe3O4/Zn-Al LDH nanocomposites exhibited uniform mesopores, high surface area of 121.7 m2/g, good superparamagnetism and appropriate saturation magnetization of 40.7 emu/g. Due to these fascinating properties, this work explored the application of Fe3O4/Zn-Al LDH in the field of nanocarrier using the doxorubicin hydrochloride (DOX) as a model anticancer drug for the detailed investigations of drug loading capacity, controlled release behavior and cytotoxic effects under in vitro conditions. The Fe3O4/Zn-Al LDH showed a good drug storage capacity and a pH-responsive drug release profile. After loading the anti-cancer drugs, the DOX-loaded Fe3O4/Zn-Al LDH exhibited enhanced therapeutic effect on human hepatocellular carcinoma cells (HepG2), demonstrating that the fabricated Fe3O4/Zn-Al LDH nanocomposites have a great potential in cancer therapy.

Abstract:Background Drug resistance is one of the prime reasons of chemotherapy failure in breast cancer and is also an important factor affecting prognosis. Purpose In this study, we constructed a functional magnetic mesoporous silica-based nanocomposite (MMSN) for breast cancer chemotherapy/photodynamic therapy. Methods MMSN was characterized by scanning electron microscopy and transmission electron microscopy to observe the morphology. The size distribution and zeta potential of the MSNs were determined using Malvern Particle Size Analyzer. Anti-tumor activity in vitro was investigated by CCK-8 assay, flow cytometry and transwell experiment, and the anti-tumor activity in vivo was probed into by magnetic targeting, toxicity, and antitumor effects in breast cancer-bearing BABL/c nude mice. Results The results showed that the release of doxorubicin in the nanocomposites was pH sensitive, and the cumulative release rate reached 80.53% at 60 h under acidic conditions. The nanocomposites had a high cellular uptake ability in MCF-7/ADR cells, and the IC50 value of the nanocomposites on MCF-7/ADR cells was 4.23 μg/mL, much smaller than that of free DOX (363.2 μg/mL). The nanocomposites could effectively reverse resistance and induce apoptosis of MCF-7/ADR cells. The blood biochemistry parameters and H&E staining results showed no serious adverse effects after treatment with the nanocomposites. Prussian blue staining showed that the nanocomposites were able to target tumor tissues in tumor-bearing mice under a magnetic field. The combined chemical/photodynamic therapy significantly inhibited tumor growth in vivo. Conclusion Nanocomposites with magnetic and pH dual-responsive performance has shown a promising platform for enhanced drug-resistant breast cancer treatment.
Fe-Mediated Self-Assembled Nanodrug for Tumor Microenvironment Activated Synergistic Ferroptosis-Based-Chemodynamic/Chemo Therapy and Magnetic Resonance Imaging

Abstract:Due to the low drug concentration, glutathione (GSH)-based oxidative stress regulating system in target tissues, and serious side effects, doxorubicin (DOX) usually shows a suboptimal efficacy in clinical practice. The synergistic combination of DOX-based chemotherapy with iron ion-based chemodynamic therapy (CDT), sensitization of cancer cells by GSH depletion, and responsive targeted delivery of DOX have been regarded as a potential efficient strategy to improve the efficacy. Herein, benefiting from the strong lipoic acid-Fe3+ coordination, we synthesized the DOX@Fe3+-LA (DOX@FL) nanodrug with a one-pot method based on the Fe3+-DOX chelation, disulfide open-ring polymerization and self-assembly behavior of lipoic acid. The nanodrug showed a spherical, uniform morphology and a high loading of DOX and Fe. Under the tumor microenvironment, the nanodrug could synchronously release DOX and Fe, and then induce center dot OH generation and intracellular GSH depletion efficiently, showing a multimodality synergistic therapeutic effect in vitro and in vivo. Additionally, the DOX@FL showed pH- and GSH-responsive MRI due to the paramagnetism of Fe3+, suggesting that DOX@FL NPs be a simple, efficient, and multifunctional nanoplatform for cancer targeting treatment and MRI.

Abstract:Research on biocompatible metal-organic frameworks (MOFs) has recently attracted much attention from researchers as drug delivery systems. In this study, for the first time, Cu MOFs grown on graphene oxide capped with magnetic hydroxyapatite (MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 ), and then the surface of the resulting nanocomposites was modified with the l -lysine amino acid (Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 ) to prepare a controlled and pH-sensitive nanocomposite for targeted delivery of doxorubicin (DOX) to MDA-MB-231 breast cancer cells and MCF-10A normal cells. Different techniques confirmed the successful preparation of the designed nanocomposites. The values of encapsulation efficiencies (EE%) for MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 and Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 was found to be 79.3 % and 96.5 %, respectively. The in vitro DOX release results showed that the Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 has a more controlled release than that of MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 due to the slow diffusion of the drug through the Lys shell. In addition, both nanocomposites showed a pH-response release behavior. The release mechanism of DOX by the designed nanocomposites also revealed that the Korsmeyer-Peppas model is the best model to describe the release of the drug. Furthermore, in vitro cytotoxicity investigation confirmed that the prepared MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 and Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 nanocomposite had good biocompatibility against MDA-MB-231 and MCF-10A cells. Whereas, DOX-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 and DOX-Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 against MDA-MB-231 cells increased the cytotoxicity as a result of the targeting and controlled delivery of the DOX to the cancer cells. The in vitro antioxidant activity and hemolysis experiments revealed that the designed Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 have excellent antioxidant activity and good haemocompatibility, respectively, as a suitable nanocomposites for drug applications. Therefore, it is suggested that the prepared nanocomposites can be good for biological applications.

Abstract:Super Paramagnetic Iron Oxide Nanoparticles have attracted much attention in drug delivery systems because of their considerable surface-to-volume ratio and content as well. In this regard, in this study, four different formulations of magnetic nanoparticles were synthesized via various polymeric coating layer (beta-cyclodextrin, Chitosan, Gum Arabic, and Alginate) in order to find an appropriate polymer in terms of higher drug loading efficiency and capacity. Molecular dynamics simulation and Quantum mechanics were manipulated to examine the interaction mechanism between the polysaccharide polymers and Doxorubicin. Based on the calculated results, As the best candidate, βCD was chosen for DOX loading. DOX, a hydrophilic drug, was loaded into each of the four formulations. The optimum formulation, Fe 3 O 4 -ßCD, possessed loading efficiency and capacity, with 97% and 13%, respectively. The selected nanocarrier was fully characterized by Fourier Transform Infrared, VSM, TGA, XRD, SEM, UV-Vis, DLS, and TEM. TEM result exhibited the prepared Fe 3 O 4 -ßCD-DOX nanoparticles had a spherical shape (core and shell) and the size of around 15 nm. In vitro release investigation was conducted using HPLC analysis in the phosphate-buffered saline in acidic (pH = 5.4) and neutral (pH = 7.4) medium. The obtained results demonstrated that a sustain release performance for more than 72 h with the amount of 50% DOX release in pH = 5.4. Cytotoxicity assay showed the toxicity of bare Fe 3 O 4 and Fe 3 O 4 -ßCDNPsagainst MDA-MB468 cancerous cell and MCF10A normal cell was negligible and DOX-loaded NPs (Fe 3 O 4 -ßCD-DOX) illustrated a higher toxicity in MDA-MB468 cancerous cells than MCF 10 A normal cells. The results showed that Fe 3 O 4 -ßCD-DOXNPs could improve the therapeutic efficacy and also reduce the adversarial effects; accordingly, the prepared system could be served as a promising drug delivery system in cancer treatment. Abstract:The visual fixation of two groups of healthy newborn infants was tested at a mean age of three days. Mothers of the infants in one group received a single dose of meperidine (pethidine) during labour and delivery; those in the second group received no medication. Unlike infants of mothers who received a single dose of alphaprodine hydrochloride in a previous study, there were no differences between the groups in the present study. Moreover, longer drug‐to‐delivery intervals did not appear to potentiate the effects of meperidine. A single dose of meperidine appears to have few effects on neonates' sensory functioning, which is contrary to other reports.

Abstract:Since multifunctional nanomaterials have exhibited significant ability as carriers in biomedical applications, it is valuableto create such a drug delivery system for cancer treatment. In this context, a novel type of trifunctional nanocomposite (high magnetization (26.25 emu.g -1 ) and size about 24 nm)) with pH-responsive, luminescent and magnetic properties (NaCeF 4 :Yb/Er@SiO 2 @Fe 3 O 4 -NH 2 @poly(MAA-co-IA)), has been designed as a new platformfor efficient loading and the pH-responsive concurrent release of methotrexate (MTX) and doxorubicin (DOX). The luminescent and hollow NaCeF 4 :Yb/Er as a core is more suitable for deep-tissue diagnosis while Fe 3 O 4 in outer shell (instead of core) caused its magnetic property retain in comparison with other core shell drug nanocarriers. MTX and DOX were loaded into nanocarrier, the loading capacity being 35 and 99%, respectively) The structure and morphology of the prepared nanocarrier were characterized by XRD, FT‐IR, VSM, TEM, FE-SEM, PL, EDAX, DLS and Zeta potential analyses. The in vitro release studies at different pH values (7.4 and 5) exhibited the pH-responsivesystem. The biocompatibility of the drug free nanocarrier and the cytotoxicity of DOX, MTX, the mixture of drugs (MTX@DOX) and MTX@DOX-nanocarrier were investigated in different concentration (0.01 to 100 μg/mL) by MTT test on human breast epithelial adenocarcinoma (MDA-MB-231) cell line. The anti-cancer studies of the nanocarrier exhibited very low cytotoxicity and high cell viability (70%) against MDA-MB-231 cell line after 72 h. In vitro cytotoxicity tests of the MTX@DOX-nanocarrier against MDA-MB-231 cell line revealed low cell viability (11%) after 72 h. Cellular uptake depicted notably higher uptake percentage for DOX-NPs than free DOX within 3 h while DAPI staining results demonstrated the higher apoptotic effects of drugs-loaded nanocarrier than free form of the drugs. Based on the enticingresults attained, the NaCeF 4 :Yb/Er@SiO 2 @Fe 3 O 4 -NH 2 @poly(MAA-co-IA) nanocomposite could be used as a pH-sensitive and non-toxic carrier for the targeted therapy of aggressive MDA-MB-231 breast cancer cells and is suitable for controlled drug release and monitoring drug delivery.
