Abstract:Zinc oxide nanoparticles (nano-ZnOs) are widely used and possess great potentials in agriculture and biomedicine. It is inevitable for human exposure to these nanoparticles. However, no study had been conducted to investigate the long term effects of nano-ZnOs. This study aimed at investigating effects of nano-ZnOs on development, zinc metabolism and biodistribution of minerals (Zn, Fe, Cu, and Mn) in mice from week 3 to 35. After the characteristics of nano-ZnOs were determined, they were added into the basal diet at 0, 50, 500 and 5000 mg/kg. Results indicated that added 50 and 500 mg/kg nano-ZnOs showed minimal toxicity. However, 5000 mg/kg nano-ZnOs significantly decreased body weight (from week 4 to 16) and increased the relative weights of the pancreas, brain and lung. Added 5000 mg/kg nano-ZnOs significantly increased the serum glutamic-pyruvic transaminase activity and zinc content, and significantly enhanced mRNA expression of zinc metabolism-related genes, including metallothionein 1(32.66 folds), metallothionein 2 (31.42 folds), ZIP8 (2.21folds), ZIP14 (2.45 folds), ZnT1 (4.76 folds), ZnT2 (6.19 folds) and ZnT4 (1.82 folds). The biodistribution determination showed that there was a significant accumulation of zinc in the liver, pancreas, kidney, and bones (tibia and fibula) after receiving 5000 mg/kg nano-ZnO diet, while no significant effects on Cu, Fe, and Mn levels, except for liver Fe content and pancreas Mn level. Our results demonstrated that long term exposure to 50 and 500 mg/kg nano-ZnO diets showed minimal toxicity. However, high dose of nano-ZnOs (5000 mg/kg) caused toxicity on development, and altered the zinc metabolism and biodistribution in mice.
Zinc oxide induces the stringent response and major reorientations in the central metabolism of Bacillus subtilis

Abstract:Microorganisms, such as bacteria, are one of the first targets of nanoparticles in the environment. In this study, we tested the effect of two nanoparticles, ZnO and TiO2, with the salt ZnSO4 as the control, on the Gram-positive bacterium Bacillus subtilis by 2D gel electrophoresis-based proteomics. Despite a significant effect on viability (LD50), TiO2 NPs had no detectable effect on the proteomic pattern, while ZnO NPs and ZnSO4 significantly modified B. subtilis metabolism. These results allowed us to conclude that the effects of ZnO observed in this work were mainly attributable to Zn dissolution in the culture media. Proteomic analysis highlighted twelve modulated proteins related to central metabolism: MetE and MccB (cysteine metabolism), OdhA, AspB, IolD, AnsB, PdhB and YtsJ (Krebs cycle) and XylA, YqjI, Drm and Tal (pentose phosphate pathway). Biochemical assays, such as free sulfhydryl, CoA-SH and malate dehydrogenase assays corroborated the observed central metabolism reorientation and showed that Zn stress induced oxidative stress, probably as a consequence of thiol chelation stress by Zn ions. The other patterns affected by ZnO and ZnSO4 were the stringent response and the general stress response. Nine proteins involved in or controlled by the stringent response showed a modified expression profile in the presence of ZnO NPs or ZnSO4: YwaC, SigH, YtxH, YtzB, TufA, RplJ, RpsB, PdhB and Mbl. An increase in the ppGpp concentration confirmed the involvement of the stringent response during a Zn stress. All these metabolic reorientations in response to Zn stress were probably the result of complex regulatory mechanisms including at least the stringent response via YwaC.
Ecotoxicological Effect of Zinc Oxide Nanoparticles on Soil Microorganisms

Abstract:The widespread production and use of zinc oxide nanoparticles (ZnO-NPs) in recent years have posed potential threat to the ecosystem. This study aimed to investigate the ecotoxicological effect of ZnO-NPs on soil microorganisms using laboratory microcosm test. Respiration, ammonification, dehydrogenase (DH) activity, and fluorescent diacetate hydrolase (FDAH) activity were used as ecotoxicological parameters. The results showed that in the neutral soil treated with 1 mg ZnO-NPs per g soil (fresh, neutral), ammonification was significantly inhibited during the study period of three months, but the inhibition rate decreased over increasing time. Inhibition in respiration was observed in the first month of the test. In various ZnO-NPs treatments (1 mg, 5 mg, and 10 mg ZnO-NPs per g soil), DH activity and FDAH activity were inhibited during the study period of one month. For both enzyme activities, there were positive dose–response relationships between the concentration of ZnO-NPs and the inhibition rates, but the curves changed over time due to changes of ZnO-NPs toxicity. Soil type affected the toxicity of ZnONPs in soil. The toxicity was highest in the acid soil, followed by the neutral soil. The toxicity was relatively low in the alkaline soil. The toxicity was not accounted for by the Zn2+ released from the ZnO-NPs. Direct interaction of ZnO-NPs with biologic targets might be one of the reasons. The adverse effect of ZnO-NPs on soil microorganisms in neutral and acid soils is worthy of attention.
Evaluation of the Effects of Ag, Cu, ZnO and TiO2 Nanoparticles on the Expression Level of Oxidative Stress-Related Genes and the Activity of Antioxidant Enzymes in Escherichia coli, Bacillus cereus and Staphylococcus epidermidis

Abstract:Although the molecular response of bacteria exposed to metal nanoparticles (NPs) is intensively studied, many phenomena related to their survival, metal uptake, gene expression and protein production are not fully understood. Therefore, this work aimed to study Ag-NPs, Cu-NPs, ZnO-NPs and TiO2-NPs-induced alterations in the expression level of selected oxidative stress-related genes in connection with the activity of antioxidant enzymes: catalase (CAT), peroxidase (PER) and superoxide dismutase (SOD) in Escherichia coli, Bacillus cereus and Staphylococcus epidermidis. The methodology used included: the extraction of total RNA and cDNA synthesis, the preparation of primers for selected housekeeping and oxidative stress genes, RT-qPCR reaction and the measurements of CAT, PER and SOD activities. It was established that the treatment of E. coli and S. epidermidis with NPs resulted mainly in the down-regulation of targeted genes, whilst the up-regulation of genes was confirmed in B. cereus. The greatest differences in the relative expression levels of tested genes occurred in B. cereus and S. epidermidis treated with TiO2-NPs, while in E. coli, they were observed under ZnO-NPs exposure. The changes found were mostly related to the expression of genes encoding proteins with PER and CAT-like activity. Among NPs, ZnO-NPs and Cu-NPs increased the activity of antioxidants in E. coli and B. cereus. In turn, TiO2-NPs had a major effect on enzymes activity in S. epidermidis. Considering all of the collected results for tested bacteria, it can be emphasised that the impact of NPs on the antioxidant system functioning was dependent on their type and concentration.

Abstract:ABSTRACT In this study, we investigated the impact of zinc oxide (ZnO) and zinc oxide nanoparticles (ZnONPs) on Escherichia coli . These compounds are commonly used as fertilizers and feed additives and may have unintended consequences on bacteria. We conducted phenotypic and multi-omics analyses of E. coli exposed to sub-lethal concentrations of ZnO and ZnONPs for 40 sub-culturings and 20 sub-culturings with zinc followed by 20 sub-culturings without zinc in an attempt to reverse zinc effects. An extended treatment with ZnO for 40 sub-culturings had the greatest impact leading to bacterial resistance to aminoglycosides, cephalosporins, and sulfonamides while the treatment with ZnONPs for 40 sub-culturings led to the elevated MIC to chloramphenicol only. Cells exposed to these treatments were thicker and had retarded growth in elevated temperatures. Importantly, zinc withdrawal reversed most phenotypic changes. Zinc exposure caused dramatic changes in cell transcripts and proteins with a role in antibiotic response, heat stress, growth regulation, cell shape, and biofilm formation. An extended zinc exposure led to overall major alterations in E. coli biology and resulted in multi-drug resistance. IMPORTANCE A long-term exposure of bacteria to zinc oxide and zinc oxide nanoparticles leads to major alterations in bacterial morphology and physiology. These included biochemical and physiological processes promoting the emergence of strains with multi-drug resistance and virulence traits. After the removal of zinc pressure, bacterial phenotype reversed back to the original state; however, certain changes at the genomic, transcriptomic, and proteomic level remained. Why is this important? The extensive and intensive use of supplements in animal feed effects the intestinal microbiota of livestock and this may negatively impact the health of animals and people. Therefore, it is crucial to understand and monitor the impact of feed supplements on intestinal microorganisms in order to adequately assess and prevent potential health risks.

Abstract:The unique properties and growing usage of zinc oxide nanoparticles increase their release in municipal wastewater treatment plants. Therefore, these nanoparticles, by interacting with microorganisms, can fail the suitable functioning of biological systems in treatment plants. For this reason, research into the toxicity of ZnO is urgent. In the present study, the toxicity mechanism of ZnO-NPs towards microbial communities central to granular activated sludge (GAS) performance was assessed over 120-day exposure. The results demonstrate that the biotoxicity of ZnO-NPs is dependent upon its dosage, exposure time, and the extent of reactive oxygen species (ROS) production. Furthermore, GAS performance and the extracellular polymeric substances (EPS) content were significantly reduced at 50 mg/L ZnO-NPs. This exposure led to decreases in the activity of ammonia monooxygenase (25.2%) and nitrate reductase (11.9%) activity. The Field emission scanning electron microscopy images confirmed that ZnO-NPs were able to disrupt the cell membrane integrity and lead to cell/bacterial death via intracellular ROS generation which was confirmed by the Confocal Laser Scanning Microscopy analysis. After exposure to the NPs, the bacterial community composition shifted to one dominated by Gram-positive bacteria. The results of this study could help to develop environmental standards and regulations for NPs applications and emissions.

Abstract:The biosynthesis of zinc oxide nanoparticles (ZnONPs) offers great potential for plant disease management due to their potent antimicrobial properties and environmental safety. However, the precise mechanisms underlying their antifungal mode of action and role in suppressing mycotoxins remain unclear. This study aims to elucidate the mechanisms by which ZnONPs suppress the pathogenic fungus Fusarium graminearium, known to cause Fusarium head blight in wheat. Additionally, it investigates how ZnONPs mitigate the production of mycotoxins, which pose risks to humans and ruminants. The study demonstrates that ZnONPs, bioproduced by Pseudomonas poae (P. poae), inhibit not only fungal growth, colony formation, and spore germination, but also significantly reduce mycotoxin production of F. graminearium by inhibiting the synthesis of deoxynivalenol (DON), downregulating the FgTRI gene, and causing morphological alterations of the toxisomes. The results also highlight that ZnONPs exert significant effects on fungi through multiple mechanisms, including cell wall damage and the generation of reactive oxygen species (ROS). Moreover, ZnONPs effectively inhibit F. graminearium in wheat leaves and coleoptiles. Fluorescence microscopy, scanning electron microscopy (SEM), transmission electron microscopy (TEM), and optical microscopy all show that ZnONPs stop F. graminearium from getting into wheat plants and colonising them. Overall, the findings of this study provide evidence that ZnONPs are highly effective in reducing F. graminearium colonization in wheat plants and effectively decreasing mycotoxin production through multiple pathways.
Impact of bovine serum albumin - A protein corona on toxicity of ZnO NPs in environmental model systems of plant, bacteria, algae and crustaceans

Abstract:Zinc oxide nanoparticles (ZnO NPs) are widely applied in industrial, household and medical areas that lead to its discharge and accumulation in ecosystem. Here, the toxic effect of ZnO NPs in presence and absence of bovine serum albumin (BSA) was analyzed. The difference in toxicity of bare ZnO and BSA interacted ZnO was studied with different environmental models. P. aeruginosa and S. aureus were used as model bacterial systems. Toxicity against bacteria was determined by employing plate count method. C. pyrenoidsa was used as algal system for evaluating toxicity and it was determined by chlorophyll estimation assay. Daphnia sp. was chosen as crustacean system model. A. cepa root cells were chosen as plant model. ZnO NPs increased the ROS formation, lipid peroxidation and oxidative stress and it reduced in the presence of BSA. The cytotoxicity, chromosomal aberrations and micronuclei (MN) index of A. cepa were increased after ZnO NPs treatment. Same time the toxic effect was decreased in case of BSA coated ZnO NPs. The NPs toxic potential on the organisms decreased in the order of P. aeruginosa (LC50-0.092 mg/L) > S. aureus (LC50-0.33 mg/L) > Daphnia sp (LC50-0.35 mg/L) > C. pyrenoidosa (LC50-8.17 mg/L). LC50 in presence of BSA was determined to be 18.45, 26.24, 17.27 and 53.97 mg/L for P. aeruginosa, S. aureus, Daphnia sp and C. pyrenoidosa respectively. Therefore, the report suggests that BSA stabilized ZnO NPs could be more amenable towards applications in biotechnology and bioengineering.

Abstract:Burkholderia glumae and B. gladioli are seed-borne rice pathogens that cause bacterial panicle blight (BPB) disease, resulting in huge rice yield losses worldwide. However, the excessive use of chemical pesticides in agriculture has led to an increase in environmental toxicity. Microbe-mediated nanoparticles (NPs) have recently gained significant attention owing to their promising application in plant disease control. In the current study, we biologically synthesize zinc oxide nanoparticles (ZnONPs) from a native Bacillus cereus RNT6 strain, which was taxonomically identified using 16S rRNA gene analysis. The biosynthesis of ZnONPs in the reaction mixture was confirmed by using UV–Vis spectroscopy. Moreover, XRD, FTIR, SEM-EDS, and TEM analysis revealed the functional groups, crystalline nature, and spherical shape of ZnONPs with sizes ranging from 21 to 35 nm, respectively. Biogenic ZnONPs showed significant antibacterial activity at 50 µg mL−1 against B. glumae and B. gladioli with a 2.83 cm and 2.18 cm zone of inhibition, respectively, while cell numbers (measured by OD600) of the two pathogens in broth culture were reduced by 71.2% and 68.1%, respectively. The ultrastructure studies revealed the morphological damage in ZnONPs-treated B. glumae and B. gladioli cells as compared to the corresponding control. The results of this study revealed that ZnONPs could be considered as promising nanopesticides to control BPB disease in rice.

Abstract:Zinc oxide nanoparticles (ZnO NPs) show adverse impacts on aerobic denitrifying bacteria, little is known about the response of these bacteria to ZnO NPs exposure at cellular level. This study assessed the multiple responses of Pseudomonas aeruginosa PCN-2 under ZnO NPs exposure. We demonstrated that ZnO NPs exposure could inhibit the intracellular metabolism and stimulate the antioxidant defence capability of PCN-2. At lower exposure concentration (5 mg/L), exogenous ROS generated and resulted in the inhibition of pyruvate metabolism and citrate cycle, which caused deficient energy for aerobic denitrification. At higher concentrations (50 mg/L), endogenous ROS additionally generated and triggered to stronger down-regulation of oxidative phosphorylation, which caused suppressed electron transfers for aerobic denitrification. Meanwhile, ZnO NPs exposure promoted EPS production and biofilm formation, and antioxidases was especially particularly stimulated at higher concentration. Our findings are significant for understanding of microbial bacterial susceptibility, tolerance and resistance under the exposure of ZnO NPs.Aerobic denitrification is suppressed with increased ZnO NPs concentrations.ZnO NPs induce inhibition of pyruvate metabolism and citrate cycle at 5 mg/L.Higher ZnO NPs concentration induces the generation of endogenous and exogenous ROS.Higher NPs concentration leads to the trigger of antioxidant system.ZnO NPs stimulate metabolisms of EPS production.GRAPHICAL

Abstract:Biofilm formation by the pathogenic bacteria generates a serious threat to the public health as it can increase the virulence potential, resistance to drugs, and escape from the host immune response mechanisms. Among the environmental factors that influence the biofilm formation, there are only limited reports available on the role of antimicrobial agents. During the antimicrobial drug administration or application for any purpose, the microbial population can expect to get exposed to the sub-minimum inhibitory concentration (sub-MIC) of the drug which will have an unprecedented impact on microbial responses. Hence, the study has been conducted to investigate the effects of sub-MIC levels of zinc oxide nanoparticles (ZnO NPs) on the biofilm formation of Klebsiella pneumoniae and Staphylococcus aureus. Here, the selected bacteria were primarily screened for the biofilm formation by using the Congo red agar method, and their susceptibility to ZnO NPs was also evaluated. Quantitative difference in biofilm formation by the selected organisms in the presence of ZnO NPs at the sub-MIC level was further carried out by using the microtiter plate-crystal violet assay. Further, the samples were subjected to atomic force microscopy (AFM) analysis to evaluate the properties and pattern of the biofilm modulated under the experimental conditions used. From these, the organisms treated with sub-MIC levels of ZnO NPs were found to have enhanced biofilm formation when compared with the untreated sample. Also, no microbial growth could be observed for the samples treated with the minimum inhibitory concentration (MIC) of ZnO NPs. The results observed in the study provide key insights into the impact of nanomaterials on clinically important microorganisms which demands critical thinking on the antimicrobial use of nanomaterials.

Selective inhibitory activity of multidrug-resistant bacteria by zinc oxide nanoparticles

Abstract:The emergence of multidrug-resistant bacteria has attracted much attention from the global community due to their potential to cause harm to health. In this context, ZnO nanoparticles (NPs) produced under different synthesis conditions were successfully applied to inhibit several species of multiresistant bacteria from clinical isolates. Five gram-positive ( Staphylococcus epidermidis ATCC 35984, Staphylococcus aureus ATCC 25923, S. aureus ATCC 8095, Enterococcus faecalis ATCC 29212, and Enterococcus faecium ATCC 700221) and four gram-negative bacterial isolates ( Klebsiella pneumoniae ATCC 700603, Escherichia coli ATCC 25922, Acinetobacter baumannii ATCC 19606, and Pseudomonas aeruginosa ATCC 27853), known to be involved in healthcare-associated infections and multidrug resistance, were selected for this study. Due to their physicochemical properties , we found that ZnO NPs can exhibit selective inhibition. The minimum inhibitory concentration (MIC) for multidrug-resistant bacteria sensitive to ZnO NPS was determined from varying contact time and catalyst concentration from exhaustive tests. Different characterization techniques were studied to determine the crystalline, optical, morphological, and photocatalytic characteristics of ZnO NPs. The bacterial inhibition mechanisms were discussed in detail with support of photocatalytic assays and cytotoxicity of plant species. In addition, ZnO NPs have also shown great potential for application in the environmental remediation of ecosystems contaminated by organic pollutants due to their efficiency in degrading the dye methylene blue (MB) under UV light with a photocatalytic efficiency of 75% in 60 min. The ZnO NPs showed great potential for multidrug-resistant bacteria, with MICs ranging from 256 to 512 mg/L. This demonstrates that the ZnO NPs produced here have the potential to be used as a technology with great application potential in materials used in hospital environments to mitigate the growth and development of these classes of multi-resistant bacteria.

To be able to better evaluate the toxicity of NPs, exposure in agar and on filter paper was proposed for enabling a comparison of the importance of different uptake routes. A dose-related increase in mortality was observed in earthworms exposed in agar with almost 100% mortality after 96 h exposure to the highest concentration (1000 mg ZnO/kg agar). Scanning electron microscopy (SEM) showed that the addition of salts enhanced the aggregation of ZnO NPs in agar and consequently affected the dissolution behavior and biological availability of the particles. On filter paper, mortality was the highest at the lowest exposure concentration (50 mg ZnO/L) and seemed to decrease with increasing exposure levels. TEM images of ZnO showed that the solubility and morphology of NPs were changed dramatically upon the addition of humic acids (HA). The subcellular distribution pattern of Zn in earthworms after 96 h exposure in agar and on filter paper showed that the Zn taken up via dietary ZnO particles (from agar) was mainly found in organelles and the cytosol while the Zn accumulated as soluble Zn from filter paper was mainly distributed in cell membranes and tissues. Antioxidant enzymatic activities (SOD, CAT, and GSH-px) were investigated in the worms surviving the toxicity tests. A slight increase of SOD activities was observed at the lowest exposure dose of ZnO (50mg/kg), followed by a decrease at 100mg/kg in the agar cubes. Activities of both CAT and GSH-Px enzymes were not significantly influenced in the worms exposed to agar, although a slight decrease at 500 and 1000 mg ZnO/kg agar was observed. A similar change trend of SOD activities was observed for the earthworms on filter paper, but a significant decrease began at a higher ZnO NP concentration of 500 mg ZnO/L. The use of soil extracts instead of deionized water (DW) to simulate a realistic exposure system significantly reduced the toxicity of the ZnO NPs on filter paper, which increases the predictive power of filter paper toxicity tests for the environmental risk assessment of NPs.
Antibacterial Mechanisms of Zinc Oxide Nanoparticle against Bacterial Food Pathogens Resistant to Beta-Lactam Antibiotics

Abstract:The increase in β-lactam-resistant Gram-negative bacteria is a severe recurrent problem in the food industry for both producers and consumers. The development of nanotechnology and nanomaterial applications has transformed many features in food science. The antibacterial activity of zinc oxide nanoparticles (ZnO NPs) and their mechanism of action on β-lactam-resistant Gram-negative food pathogens, such as Escherichia coli, Pseudomonas aeruginosa, Salmonella typhi, Serratia marcescens, Klebsiella pneumoniae, and Proteus mirabilis, are investigated in the present paper. The study results demonstrate that ZnO NPs possesses broad-spectrum action against these β-lactamase-producing strains. The minimal inhibitory and minimal bactericidal concentrations vary from 0.04 to 0.08 and 0.12 to 0.24 mg/mL, respectively. The ZnO NPs elevate the level of reactive oxygen species (ROS) and malondialdehyde in the bacterial cells as membrane lipid peroxidation. It has been confirmed from the transmission electron microscopy image of the treated bacterial cells that ZnO NPs diminish the permeable membrane, denature the intracellular proteins, cause DNA damage, and cause membrane leakage. Based on these findings, the action of ZnO NPs has been attributed to the fact that broad-spectrum antibacterial action against β-lactam-resistant Gram-negative food pathogens is mediated by Zn2+ ion-induced oxidative stress, actions via lipid peroxidation and membrane damage, subsequently resulting in depletion, leading to β-lactamase enzyme inhibition, intracellular protein inactivation, DNA damage, and eventually cell death. Based on the findings of the present study, ZnO NPs can be recommended as potent broad-spectrum antibacterial agents against β-lactam-resistant Gram-negative pathogenic strains.

Investigation of Genomic Effect of Zirconium Oxide Nanoparticles in Escherichia coli Bacteria

Abstract:Due to the concerns of the society about the increase of antibiotic resistant infections, many studies and research have been done on nanoparticles and applications of nano-biotechnology. Zirconium Oxide ($\text{ZrO}_{2}$) in which called zirconia, is a white oxide of zirconium metal that its diameter is 20 nm. The colloidal size of these particles is often smaller than bacterial and eukaryotic cells. The main intention of this paper is to investigate the effect of different doses of $\text{ZrO}_{2}$ NPs on the sequences changes for the $\textit{Escherichia coli}$ ($\textit{E. coli}$) genome. At the first step, $\textit{E. coli}$ was cultured in eosin methylene blue agar and brain heart broth (BHB) mediums, respectively. Then, bacteria were treated with $\text{ZrO}_{2}$ NPs at concentrations of 100, 250, and 350 $\mu$g/ml. After treatment, the growth of bacteria was evaluated by utilizing spectrophotometry at 600 nm after incubation times including 2, 4, 6, 8, and 24 hours at 37 $^{\circ}$C. At the second step, the extraction of DNA was performed by using control and treated samples. Then, the changes in the sequence of bacterial genome were investigated using RAPD markers. Finally, NTSYS-PC platform was employed in order to analyze of the results extracted by electrophoresis of products on agarose gel. In this paper, it was observed that $\text{ZrO}_{2}$ NPs can inhibit the growth of bacteria at concentrations of 250 and 350 $\mu$g/ml after 8 hours of treatment. It was also found that the $\text{ZrO}_{2}$ NPs at different concentrations have not changed the genome sequence of $\textit{E. coli}$. Furthermore, it was concluded that the $\text{ZrO}_{2}$ NPs with the concentration of 350 $\mu$g/ml had the highest inhibitory properties without significant changing in the genomic sequence of $\textit{E. coli}$.

Abstract:The antimicrobial activity of ZnO nanoparticles (NPS) was investigated under aquatic and aerosol exposure modes. DO NIPS in aquatic media aggregated to micrometer-sized particles and did not interact with microorganisms effectively. Hence, the inhibition of microbial growth by nano-ZnO NIPS (e.g., Mycobacterium smegmatis and Cyanothece 51142) in aquatic media was mainly attributable to dissolved zinc species. Shewanella oneidensis MR-1 and Escherichia coli were able to produce large amounts of extracellular polymeric substances, and their growth was not inhibited by ZnO NIPS in aquatic media, even at high concentrations (>40 mg/L). On the other hand, when ZnO NIPS were electrosprayed onto an E. coli biofilm so that NIPS could be directly deposited onto the cell surface, the aerosol exposure dramatically reduced cellular viability. For example, an electrospray of ZnO NIPS (20 nm) reduced the total number of viable E.coli cells by 57% compared to the control case, in which we electrosprayed only the buffer solution. However, electrospraying large-sized ZnO particles (480 nm) or nonsoluble TiO2 NPs (20 nm) caused much less lethality to E coli cells. The above observation implies that the aerosol method of exposing ZnO NPs to biological systems appears to have a much higher antimicrobial activity, and thus may lead to practical applications of employing a novel antimicrobial agent for airborne disease control.

Abstract:As antibiotic resistance increases and antibiotic development dwindles, new antimicrobial agents are needed. Recent advances in nanoscale engineering have increased interest in metal oxide nanoparticles, particularly zinc oxide nanoparticles, as antimicrobial agents. Zinc oxide nanoparticles are promising due to their broad-spectrum antibacterial activity and low production cost. Despite many studies demonstrating the effectiveness of zinc oxide nanoparticles, the antibacterial mechanism is still unknown. Previous work has implicated the role of reactive oxygen species such as hydrogen peroxide, physical damage of the cell envelope, and/or release of toxic Zn 2+ ions as possible mechanisms of action. To evaluate the role of these proposed methods, we assessed the susceptibility of S. aureus mutant strains, Δ katA and Δ mprF , to zinc oxide nanoparticles of approximately 50 nm in size. These assays demonstrated that hydrogen peroxide and electrostatic interactions are not crucial for mediating zinc oxide nanoparticle toxicity. Instead, we found that Zn 2+ accumulates in Mueller-Hinton Broth over time and that removal of Zn 2+ through chelation reverses this toxicity. Furthermore, we found that the physical separation of zinc oxide nanoparticles and bacterial cells using a semi-permeable membrane still allows for growth inhibition. We concluded that soluble Zn 2+ is the primary mechanism by which zinc oxide nanoparticles mediate toxicity in Mueller-Hinton Broth. Future work investigating how factors such as particle morphology (e.g., size, polarity, surface defects) and media contribute to Zn 2+ dissolution could allow for the synthesis of zinc oxide nanoparticles that possess chemical and morphological properties best suited for antibacterial efficacy.

Abstract:Zinc oxide nanoparticles (ZnO NPs) have been widely used in biomedical applications due to their high biocompatibility and low toxicity to humans. The present work aimed to investigate the antibacterial effects of different concentrations of ZnO NPs on two opportunistic pathogens, Serratia marcescens and Enterococcus faecalis . The surface interaction between nanoparticles and bacterial cell wall, and the subsequent morphological alterations on the bacterial surface, were examined through Fourier transform infrared spectroscopy and scanning electron microscope. The energy dispersive X-ray analysis was used to confirm the elemental composition of ZnO NPs and the cellular accumulation of ZnO NPs in bacteria. The growth-inhibitory test demonstrated a dose-dependent growth inhibitory effect of ZnO NPs against both the test bacteria, as the higher concentration of nanoparticles caused the higher bacterial growth inhibition. The results showed that ZnO NPs caused a higher growth inhibition (63.50 ± 2.50%) on the Gram-positive bacterium E . faecalis compared to the Gram-negative bacterium S. marcescens (51.27 ± 4.56%). Fourier transform infrared spectrum revealed the possible involvement of hydroxyl, carboxyl, amides, methylene, and phosphate groups from the biomolecules of bacterial cell wall such as proteins, carbohydrates, lipids, and phospholipids in the interaction of ZnO NPs on bacterial cell surface. Energy dispersive X-ray analysis showed the higher accumulation of ZnO NPs in E. faecalis than S. marcescens analogous to the bacterial growth inhibition. Scanning electron microscopy images confirmed the antibacterial properties of ZnO NPs, showing the loss of integrity of cell membrane and distortion of bacterial cells. Hence, the potential of ZnO NP as an antibacterial agent against S. marcescens and E. faecalis has been confirmed.

Abstract:Excessive production and utilization of nanoparticles (NPs) at industrial and household levels releases substantial quantities of NPs into the environment. These can be harmful to different types of organisms and cause adverse effects on ecosystems. Purchased TiO2 and ZnO NPs were characterized via XRD, XPS, FESEM, and Zeta potential. This study elucidates how multidrug resistant Escherichia coli LM13, which was recovered from livestock manure, counteracts the antibacterial activities of TiO2 and ZnO NPs to survive in the environment. E. coli ATCC25922, which is susceptible to antibiotics, was used as control. A dose-response experiment showed that the antibacterial activity of TiO2 was lower than that of ZnO NPs and, LM13 was more resistant to NPs than ATCC25922. An AcrAB-TolC efflux pump along with its regulation genes helped LM13 to minimize NP toxicity. Flow cytometry findings also indicated that the intensity of the side-scatter light parameter increased with TiO2 and ZnO NPs in a dose dependent manner, suggesting NP uptake by the both strains. The generation of reactive oxygen species in LM13 was several-fold lower than in ATCC25922, suggesting that reactive oxygen species mainly contribute to the toxicity mechanism. These results illustrate the necessity to evaluate the impacts of NPs on the survival capacity of bacteria and on the resistance genes in bacteria with higher NP resistance than NP susceptible bacteria.

Abstract:Water chemistry can be a major factor regulating the toxicity mechanism of ZnO nanoparticles (nano-ZnO) in water. The effect of five commonly used aqueous media with various chemical properties on the toxicity of nano-ZnO to Escherichia coli O111 (E. coli) was investigated, including ultrapure water, 0.85% NaCl, phosphate-buffered saline (PBS), minimal Davis (MD), and Luria-Bertani (LB). Combined results of physicochemical characterization and antibacterial tests of nano-ZnO in the five media suggest that the toxicity of nano-ZnO is mainly due to the free zinc ions and labile zinc complexes. The toxicity of nano-ZnO in the five media deceased as follows: ultrapure water > NaCl > MD > LB > PBS. The generation of precipitates (Zn(3)(PO(4))(2) in PBS) and zinc complexes (of zinc with citrate and amino acids in MD and LB, respectively) dramatically decreased the concentration of Zn(2+) ions, resulting in the lower toxicity in these media. Additionally, the isotonic and rich nutrient conditions improved the tolerance of E. coli to toxicants. Considering the dramatic difference of the toxicity of nano-ZnO in various aqueous media, the effect of water chemistry on the physicochemical properties of nanoparticles should be paid more attention in future nanotoxicity evaluations.
