Abstract:Background: Currently, balanced diet and healthy foods are attracting increasing attention due to their great health-beneficial effects. The tuber of Dioscorea (yam) is both edible and medicinal, as it contains various functional compounds with multiple pharmacological activities and disease targets. Moreover, yam tuber can serve as a vegetable or staple food due to its high starch content. Therefore, exploring the functional activities of yam tuber and its application in food and medicine industries is significant. Scope and approach: This review comprehensively summarizes the characteristics and application of yam tuber, as well as the findings about its primary nutrients, distinct functional components, extraction technologies, detection methods and biological activities, and provides some new prospects for further investigation. Besides, the development and comprehensive utilization of yam tuber products are elucidated. Finally, some future research directions for better utilization of yam tubers are suggested. Key findings and conclusions: The Dioscorea genus is distributed worldwide with a wide variety of species. Yam tuber is rich in nutrients and bioactive compounds, such as starch, protein, non-starch polysaccharides, steroid saponin, allantoin, and phenolic compounds. The most important health benefits include antioxidant activity, anti-inflammatory activity, gastrointestinal protection, gut microbiota regulation, cardiac diseases treatment, hypoglycemic activity, anti-tumor activity, and estrogen-like effect. We also summarize the applications of yam tuber in foods and medicine. Finally, the review indicates that better utilization of yam tuber is hindered by poor variety protection, limited research on its pharmacological activities, browning phenomenon, and some safety concerns.
Dioscorea spp.: Comprehensive Review of Antioxidant Properties and Their Relation to Phytochemicals and Health Benefits

Abstract:Dioscorea, consisting of over 600 species, is the most important genus in the Dioscoreaceae family; however, the practically used plants, which are commonly called yam, are restricted to a remarkably smaller number of species. Numerous studies have reported the high nutritional value of yam, particularly as an alternative source of starch and some important micronutrients. Several Dioscorea species are widely used for various medicinal purposes as well. In many studies, the bioactivities and health benefits of Dioscorea extracts and other preparations have been related to the presence of phytochemicals, which possess antioxidant properties; they are related mainly to radical-scavenging capacity in chemical assays and positive effects on the endogenous antioxidant system in cell-based and in vivo assays. Considering the increasing number of publications on this topic and the absence of comprehensive and focused review papers on antioxidant potential, this article summarizes the results of studies on the antioxidant properties of Dioscorea spp. and their relation to phytochemicals and health benefits. A comprehensive survey of the published articles has revealed that the majority of studies have been performed with plant tubers (rhizomes, roots), while reports on leaves are rather scarce. In general, leaf extracts demonstrated stronger antioxidant potential than tuber preparations. This may be related to the differences in phytochemical composition: saponins, phenanthrenes and, for some pigment-rich species (purple yams), anthocyanins are important constituents in tubers, while phenolic acids and flavonoids are characteristic phytochemicals in the leaves. The review may assist in explaining ethnopharmacological knowledge on the health benefits of Dioscorea plants and their preparations; moreover, it may foster further studies of poorly investigated species, as well as their wider application in developing new functional foods and nutraceuticals.

Abstract:The Genus Diospyros has garnered significant scientific interest due to its diverse bioactivities, attributed to various phytochemicals like flavonoids, terpenoids, and xanthones. Several species, including D. lotus, D. kaki, and D. tricolor, exhibit promising anti‐cancer properties through in vitro and in vivo studies, demonstrating inhibition of proliferation, induction of apoptosis, and mitigation of angiogenesis. Integrating traditional knowledge with modern research, Diospyros shows potential as a source for novel and effective anti‐cancer therapeutics in ethno‐pharmacology. The genus Diospyros has gained significant attention in the scientific community owing to its diverse bioactivities ascribed to specific bioactive constituents present in different species of this plant. Phytochemicals like flavonoids, terpenoids, and xanthones have been reported to be present in other Diospyros species responsible for their pharmacological properties. These compounds are well known for their diverse potent therapeutic potentials, such as antimicrobial, antioxidant, anti‐inflammatory, and anticancer properties. This review enlightens the details of the Genus Diospyros, ranging from an overview of its species to an in‐depth analysis of phytochemistry, ethnopharmacology, and their potential as anticancer agents. Different species, including Diospyros lotus, Diospyros kaki, Diospyros maritima, Diospyros mespiliformis, and Diospyros tricolor, presented with an enormous range of anticancer activities against human cancer cell cultures. Moreover, this review highlights the results of various in vitro (antiproliferative, cytotoxic effects against), in vivo (inhibition of tumor, apoptosis), and in silico (GLU234, GLU278, and LYS158 protein residues) studies, elucidating its preclinical anticancer potential. The anticancer potential displays inhibition of cellular proliferation, induction of apoptosis, and mitigation of angiogenesis. Furthermore, this review may elaborate the use of traditional knowledge, modern research, and potential therapeutic applications in the field of anticancer ethnopharmacology. As the modern‐day research approaches novel alternatives to combat diseases like cancer, the Genus Diospyros may emerge as a promising avenue with the potential to yield innovative and effective therapeutic agents.

Abstract:This review will summarize some of the important reports on the chemistry and the biological activities of Dioscorea steroid saponins from the literature data of recent years (2000–2006) and from the authors’ studies. These discoveries became possible as a result of the scientific development of isolation, structure elucidation, and the development of in vitro bioassays. Over 50 steroid saponins of furostane-, spirostane-, and pregnane-type skeleton have been discovered and characterized from 13 Dioscorea species, namely, D. bulbifera L., D. cayenensis Lam.-Holl, D. colletii Hook. F. var. hypoglauca (Palib.) Pei et Ting, D. deltoidea Wall var. orbiculata, D. futschauensis R. Abstract:The quest for a food secure and safe world has led to continuous effort toward improvements of global food and health systems. While the developed countries seem to have these systems stabilized, some parts of the world still face enormous challenges. Yam (Dioscorea species) is an orphan crop, widely distributed globally; and has contributed enormously to food security especially in sub-Saharan Africa because of its role in providing nutritional benefits and income. Additionally, yam has non-nutritional components called bioactive compounds, which offer numerous health benefits ranging from prevention to treatment of degenerative diseases. Pharmaceutical application of diosgenin and dioscorin, among other compounds isolated from yam, has shown more prospects recently. Despite the benefits embedded in yam, reports on the nutritional and therapeutic potentials of yam have been fragmented and the diversity within the genus has led to much confusion. An overview of the nutritional and health importance of yam will harness the crop to meet its potential towards combating hunger and malnutrition, while improving global health. This review makes a conscious attempt to provide an overview regarding the nutritional, bioactive compositions and therapeutic potentials of yam diversity. Insights on how to increase its utilization for a greater impact are elucidated.

Abstract:Dioscorea nipponica Makino is a perennial twining herbs belonging to the family Dioscoreaceae, which is mainly distributed in the northeastern, northern, eastern and central regions of China. Traditionally, the rhizome of this herb has been commonly used by Miao and Meng ethnic groups of China to treat rheumatoid arthritis, pain in the legs and lumbar area, Kashin Beck disease, bruises, sprains, chronic bronchitis, cough and asthma. Modern pharmacological studies have discovered that this herb possesses anti-tumor, anti-inflammatory, anti-diuretic, analgesic, anti-tussive, panting-calming and phlegm-dispelling activities, along with enhancing immune function and improving cardiovascular health. In recent years, both fat-soluble and water-soluble steroidal saponins were isolated from the rhizomes of D. nipponica using silica gel column chromatography, thin layer chromatography and high performance liquid chromatography methods. Saponin and sapogenins are mainly responsible for most of the pharmacological effects of this plant. Further, the chemical components of the aboveground parts contain more than 10 kinds of phenanthrene derivatives. The present review summarizes the knowledge concerning the geographical distribution, chemical composition, pharmacological effects, toxicology studies and clinical applications of D. nipponica.

Abstract:BACKGROUND: Dioscorea alata L. tubers (DA) are suspected to prevent diabetes mellitus (DM) because they have a low glycemic index. However, only a few reports about the anti-diabetic effect of DA were reported up to date. This study aims to analyze the effect of DA consumption on several diabetic biomarkers through in vitro, in vivo, and in silico analysis.METHODS: In vitro experiments were conducted by observing the anti-inflammatory activity of DA extract, steroidal saponins (SDA) isolated from DA, and diosgenin in lymphocyte cell cultures. The tumor necrosis factor (TNF)-α and interferon (IFN)-γ percentages were analyzed by flow cytometry. In vivo study involved 24 healthy adolescents that were given a boiled DA 10 hours post-prandial. The blood sugar levels were measured at 0, 30, 60, and 120 min after treatment. Furthermore, the in silico study was carried out by analyzing the active compounds and predicting the biological activity, the target proteins, and interactions of target proteins with compounds contained in DA .RESULTS: DA extract, SDA isolated from DA, and diosgenin at 50 μg/mL significantly reduced pro-inflammatory cytokines TNF-α and IFN-γ in lymphocyte cell culture. The blood glucose levels in the DA group were lower at 30 and 60 min after treatment. Based on the in silico study, the anti-diabetic activity of DA was speculated to be attributed to the mechanisms of anti-hyperglycemia, prevention of mitochondrial dysfunction, anti-inflammation, and treated insulin resistance. Several proteins included in the DM pathway became the protein target of compounds contained in DA.CONCLUSION: DA potentially have an anti-diabetic activity through several mechanisms.KEYWORDS: hyperglycemia, inflammation, insulin resistance, yam

Abstract:Diosgenin is an aglycone of dioscin, a major bioactive steroidal saponin found in plants, including Himalayan Paris (Paris polyphylla), fenugreek (Trigonella foenum-graecum), and yam (Dioscorea spp.). We have previously demonstrated that a species of natural yam, Dioscorea japonica, contains a promising bioactive compound diosgenin, which induces anti-carcinogenic and anti-hypertriacylglycerolemic activities. Here, we found for the first time that Japanese yam (D. japonica) bulbils are richer in diosgenin than the edible tubers (rhizomes) and leaves. LC-MS and imaging-MS analyses revealed that diosgenin accumulated in the peripheral region of D. japonica bulbils. Additionally, we performed RNA-seq analysis of D. japonica, and multiple sequence alignment identified D. japonica CYP90 (DjCYP90), the orthologous gene of CYP90G4 in P. polyphylla, CYP90B50 in T. foenum-graecum, CYP90G6 in Dioscorea zingiberensis, and CYP90G in Dioscorea villosa, which encodes a diosgenin biosynthetic rate-limiting enzyme. The expression levels of DjCYP90 were significantly upregulated in D. japonica bulbils than in its rhizomes and leaves. Since diosgenin is one of the most promising functional food factors executing several favorable bioactivities, D. japonica bulbils rich in diosgenin would be a beneficial natural resource.

Abstract:Dioscorea bulbifera (Family: Dioscoreaceae) also referred to as 'Air potato,' carries significant importance in the traditional medicine of China, West Africa and India. It is a common ingredient in numerous herbals and Ayurvedic formulations used to treat a variety of ailments. The literature review extensively examined the historical usage, phytoconstituents, characterisation of phytoconstituents, and medicinal uses of tubers, leaves, rhizomes and bulbils, with a particular focus on comparing research findings. Among the bioactive constituents, aglycone forms of steroidal saponins such as dioscorine, dioscin, diosbulbins, and diosgenin exhibit significant biological activities. Extracts from different parts of the plant exhibited pharmacological activities like anti-viral, anti-malarial, analgesic, antidiabetic, and anticancer. It is necessary to conduct an in-depth investigation to bridge between traditional knowledge and scientific evidence. This comprehensive review aims to provide a detailed understanding of the ethnobotany, phytoconstituent, chemical characterization and pharmacological potential of D. bulbifera, highlighting its prospects and challenges for future research and medicinal application.
Protection effect of Dioscoreae Rhizoma against ethanol-induced gastric injury in vitro and in vivo: A phytochemical and pharmacological study

Abstract:Ethnopharmacological relevance: Dioscoreae Rhizoma, a kind of Chinese yam, is a medicinal and edible plant used in China for strengthening the spleen and stomach. However, there is a lack of modern pharmacology studies regarding its anti-gastric injury activity. Aim of the study: This study aimed to investigate the phytochemical composition of Chinese yam aqueous extract (CYW) and evaluate its gastroprotective effects against ethanol-induced gastric injury in vitro and in vivo. Materials and methods: The active components of CYW were identified using HPLC-QTOF-MS/MS in combination with the GNPS molecular networking and network pharmacology. In vitro studies were performed in the RAW264.7/GES-1 cell coculture system. In vivo study, mice were treated with CYW (0.31, 0.63, and 3.14 g/kg BW, orally) for 14 days, followed by a single oral dose of ethanol (10 mL/kg BW) to induce gastric injury. The biochemical, inflammation and oxidative stress markers were analyzed using commercial kits. Histopathology was used to assess the degree of gastric injury. Gene and protein expressions were studied using RT-qPCR and western blotting, respectively. Results: CYW significantly restored the levels of SOD, GPx and CAT, and reduced the MDA content. Further analyses showed that CYW significantly alleviated the gastric oxidative stress by inhibiting the inflammation via decreasing p-NF-κB and p-IκB-α expression levels and inhibiting the generation of IL-6, TNF-α, and IL-1β. At the same time, the fraction remarkably upregulated Bcl-2, downregulated Bax and increased growth factor secretion, thereby prevented gastric mucous cell. Besides, The combination of HPLC-QTOF-MS/MS, GNPS molecular networking analysis, and network pharmacology demonstrated that linoleic acid, 3-acetyl-11-keto-beta-boswellic acid, adenosine, aminocaproic acid, tyramine, DL-tryptophan, cycloleucine, lactulose, melibiose, alpha-beta-trehalose, and sucrose would be the main active compounds of CYW against ethanol-induced gastric injury. Conclusion: This study showed that CYW is potentially rich source of anti-oxidant and anti-inflammatory bioactive compounds. It showed efficacy against ethanol-induced gastric injury by inhibiting inflammation, oxidative stress, and apoptosis in the stomach. The results of the current work indicate that Dioscoreae Rhizoma could be utilized as a type of natural resource for production of new medicine and functional foods to prevent and/or ameliorate ethanol-induced gastric injury.
Isolation and identification of active ingredients and biological activity of Dioscorea nipponica Makino

Abstract:This study reported the isolation and identification of bioactive compounds from Dioscorea nipponica Makino, a plant used in traditional medicine for various ailments. Nine compounds were isolated, including a new compound named as diosniposide E, which was elucidated by analyzing its 1 H-NMR, 13 C-NMR, DEPT, COSY, HMBC and MS data and comparing them with data available in literature. The other eight compounds were identified as known compounds. Theoretical calculations of energy and the generation of a molecular electrostatic potential surface map were employed to assess the antioxidant capacity of nine compounds, the calculation results exhibited that compounds 5 and 6 had strong antioxidant capacities. To further evaluate the antioxidant activities of the investigated compounds, the DPPH and ABTS assays were conducted. The results from the DPPH scavenging activity test revealed that compounds 4 – 6 exhibited enhanced scavenging activities compared to L-ascorbic acid, while displaying similar efficacy to trolox. Moreover, the ABTS scavenging activities of compounds 4 – 6 were found to surpass those of L-ascorbic acid and trolox. In terms of α-glucosidase inhibition, compounds 3 and 4 displayed remarkable inhibitory activities that surpassed the effects of acarbose. Additionally, compound 2 exhibited potent anticholinesterase activities, outperforming donepezil. This research provides insights into the potential bioactive compounds present in Dioscorea nipponica Makino and may contribute to its use in traditional medicine.
Insights into diosgenin against inflammatory bowel disease as functional food based on network pharmacology and molecular docking

Abstract:Inflammatory bowel disease (IBD) is a growing global health problem. IBD is commonly prevalent in Europe and America and the incidence rate in Asia is on the rise due to altered dietary structure. Diosgenin is a natural steroidal saponin derived from Dioscorea plants. Diosgenin is the main active ingredient of some Chinese medicines which are mainly used to treat coronary heart disease, angina and hyperlipidemia. Recently, growing evidence has exhibited a crucial role of diosgenin and dioscin in alleviating IBD in multiple ways. However, the precise mechanism of diosgenin against IBD needs further exploration. In this study, network pharmacological and systematic bioinformatic analyses were performed to investigate the diosgenin's targets against IBD. 71 targets such as SRC, TNF and STAT3 were identified as overlapped genes between diosgenin and IBD. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis exhibited their involvement in the tyrosine kinase signaling pathway and its membrane receptors. Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor resistance and its downstream Ras-MAPK pathway and PI3K-Akt pathway might become the mechanism of diosgenin against IBD. In addition, molecular docking analysis showed that diosgenin has the massive potential of direct binding to tyrosine kinase and its receptors such as SRC, EGFR, FGFR1 and VEGFR. The results above collectively provided evidence that diosgenin is a promising nutraceutical food against IBD.

Abstract:Diosgenin, a steroidal saponin, is a natural product found in many plants. Diosgenin has a wide range of pharmacological activities, and has been used to treat cancer, nervous system diseases, inflammation, and infections. Numerous studies have shown that diosgenin has potential therapeutic value for lipid metabolism diseases via various pathways and mechanisms, such as controlling lipid synthesis, absorption, and inhibition of oxidative stress. These mechanisms and pathways have provided ideas for researchers to develop related drugs. In this review, we focus on data from animal and clinical studies, summarizing the toxicity of diosgenin, its pharmacological mechanism, recent research advances, and the related mechanisms of diosgenin as a drug for the treatment of lipid metabolism, especially in obesity, hyperlipidemia, nonalcoholic fatty liver disease, atherosclerosis, and diabetes. This systematic review will briefly describe the advantages of diosgenin as a potential therapeutic drug and seek to enhance our understanding of the pharmacological mechanism, recipe‐construction, and the development of novel therapeutics against lipid metabolism diseases.

Abstract:Yams are the edible subterranean rhizomes, or tubers, of plants from the genus Dioscorea. There are approximately 600 species of yam plants in the world, with more than 90 of these growing in East Asia. One particular species, Dioscorea opposita Thunb., is highly praised as "the Chinese yam". This distinction arises from millennia of storied history, both as a nutritional food source and as a principal ingredient in traditional Chinese medicine. Among the many cultivars of Dioscorea opposita Thunb., Huai Shanyao has been widely regarded as the best. This review surveyed the historical background, physiochemical composition, applications as food and medicine, and research prospects for the Chinese yam. Modern science is finally beginning to confirm the remarkable health benefits of this yam plant, long-known to the Chinese people. Chinese yam promises anti-diabetic, anti-oxidative, anti-inflammatory, immunomodulatory, anti-hyperlipidemic, anti-hypertensive, anti-cancer, and combination treatment applications, both as a functional food and as medicine.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Ethnopharmacological relevance</h3><p>Dysfunction of glucose metabolism is associated with the occurrence of metabolic syndromes, including type 2 diabetes mellitus (T2DM). In this study, we investigated the anti-diabetic effects of yam extract and allantoin in high-fat-diet (HFD) and streptozotocin (STZ)-induced diabetic mice and the mechanism of action on the dysfunction of the liver, pancreas, and skeletal muscle.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Materials and methods</h3><p>Male C57BL/6 mice were induced into a diabetic condition by HFD for 16 weeks and a single injection of STZ (120 mg/kg) and then orally administered yam extract (500 and 1000 mg/kg) or allantoin (20 and 50 mg/kg) once daily for 4 weeks. The changes in physiological parameters, serological parameters, and morphology of tissues were investigated. The expression levels of antioxidant enzymes, biogenetic proteins, and myogenetic proteins were determined in the liver, pancreas and skeletal muscle tissues of mice.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The administration of yam extract and allantoin at high doses in HFD-/STZ-induced diabetic mice compared with the control group significantly decreased the increase in body weight, caloric intake, and water intake. Yam extract and allantoin significantly decreased high glucose and leptin, total cholesterol, triglyceride, low-density lipoprotein-cholesterol, aspartate transaminase, alanine aminotransferase levels and increased insulin and albumin levels in the plasma of mice. Yam extract and allantoin inhibited the structural damage of the liver with regard to fat accumulation, the pancreas with regard to hyperplasia of Langerhans' islets, and skeletal muscle with regard to atrophy and significantly increased the expression of antioxidant enzymes and mitochondria-mediated biogenetic factors in the liver, pancreas, and muscle tissues. In addition, yam extract and allantoin significantly increased the expression of myogenetic proteins in skeletal muscle tissues.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>Our results indicated that yam extract and allantoin improve diabetic symptoms through the regulation of oxidation and glucose imbalance in liver, pancreas, and skeletal muscle tissues in mice. These findings suggest that yam extract and allantoin can be used as antidiabetic factors in supplementary foods and medications for T2DM patients.</p>

Structural and functional properties of polysaccharides extracted from three Dioscorea species

Abstract:Dioscorea has a history spanning over 2000 years for both medicinal and edible purposes in China. It contains rich polysaccharides, which are frequently utilized as thickening and stabilizing agents in the food industry. However, there has been relatively little focus on polysaccharides from common Dioscorea species besides D. opposita, such as D. alata and D. esculenta. In this study, non-starch crude polysaccharides were isolated from D. opposita (BD), D. alata (WC), and D. esculenta (GZ). Their structures, physicochemical compositions, and functional properties were characterized and compared. The results indicated three polysaccharides all exhibited characteristic peaks of polysaccharides and possessed triple-helix structures. The Glc (36.78-83.90 %), Man (6.71-26.68 %), and GalA (8.54-10.22 %) were identified as the primary monosaccharide components. In terms of functionality, three polysaccharide solutions demonstrated non-Newtonian flow characteristics and displayed commendable thermal stability. It is worth noting that the antioxidant and emulsifying properties of polysaccharides isolated from D. opposita (BD) and D. alata (WC) were superior to those of D. esculenta (GZ), making them more suitable for use as antioxidants and stabilizers. By comparing polysaccharides derived from different Dioscorea species, this study provides valuable insights into the food, cosmetic, and pharmaceutical industries based on the unique properties of these different polysaccharides.

Abstract:Pruritus is a frequent, distressing and sometimes disabling symptom of liver and biliary tract disorders. Results of treatment are sometimes disappointing and the pathophysiology is still largely unknown. It was recently discovered that endogenous opioids contribute to the perception of itching and that opiate receptor antagonists can reduce the overstimulation of these receptors and thereby attenuate the itching. A stepwise treatment strategy focusing successively on ion exchange resins, rifampicin and opiate receptor antagonists leads to effective alleviation of itching in most patients.
