Abstract:In this paper we present recent advances in Forster resonance energy transfer (FRET) sensing and bioimaging using nontoxic silicon quantum dots. (SiQDs) In our work, we prepare SiQDs-dye conjugates, with SiQDs serving as the donor which are covalently attached to organic dye acceptors via self-assembled monolayer linkers. Enzymatic cleavage of the peptide leads to changes in FRET response which was monitored using fluorescence lifetime imaging microscopy (FLIM-FRET). The combination of interfacial design and optical imaging presented in this work opens new opportunities for bio-applications using nontoxic silicon quantum dots.

Abstract:Concerns over possible toxicities of conventional metal-containing quantum dots have inspired growing research interests in colloidal silicon nanocrystals (SiNCs), or silicon quantum dots (SiQDs). This is related to their potential applications in a number of fields such as solar cells, optoelectronic devices and fluorescent bio-labelling agents. The past decade has seen significant progress in the understanding of fundamental physics and surface properties of silicon nanocrystals. Such understanding is based on the advances in the preparation and characterization of surface passivated colloidal silicon nanocrystals. In this critical review, we summarize recent advances in the methods of preparing high quality silicon nanocrystals and strategies for forming self-assembled monolayers (SAMs), with a focus on their bioapplications. We highlight some of the major challenges that remain, as well as lessons learnt when working with silicon nanocrystals (239 references).

Abstract:Fluorescence lifetime imaging microscopy is successfully demonstrated in both one-and two-photon cases with surface modified, nanocrystalline silicon quantum dots in the context of bioimaging. The technique is further demonstrated in combination with Forster resonance energy transfer studies where the color of the nanoparticles is tuned by using organic dye acceptors directly conjugated onto the nanoparticle surface.

Abstract:In this study, we describe a solution procedure for the preparation and surface modification of photostable colloidal silicon quantum dots (SiQDs) for imaging of cancer cells. Photoluminescent SiQDs were synthesized by reduction of halogenated silane precursors using a microemulsion process. It was shown that 1,8-nonadiyne molecules could be grafted onto the surface of hydrogen-terminated SiQDs via ultraviolet (UV)-promoted hydrosilylation, demonstrated by Fourier transform infrared spectroscopy (FTIR) measurements. In addition, various azide molecules were coupled onto nonadiyne-functionalized particles, rendering particles dispersible in selected polar and nonpolar solvents. The photoluminescence of functionalized SiQDs was stable against photobleaching and did not vary appreciably within biologically applicable pH and temperature ranges. To demonstrate compatibility with biological systems, water-soluble SiQDs were used for fluorescent imaging of HeLa cells. In addition, the SiQDs were shown to be non-cytotoxic at concentrations up to 240 μg/mL. The results presented herein provide good evidence for the versatility of functionalized SiQDs for fluorescent bioimaging application.

Abstract:The rapid advance and growth of the point-of-care diagnosis industry has provided an impetus for the development of novel signal labels for highly sensitive bio-molecule detections. Colloidal quantum dots (QDs) exhibit superior brightness, facile surface functionalization and exceptional photostability, making them the preferred option for these biological applications. However, the significantly reduced fluorescence intensity and limited photochemical stability in complex biological environments have greatly hampered their further use. Herein, with an innovative alloying engineering strategy, the high-quality water-soluble CdSe/CdxZn1−xS QDs with near-unity PL quantum yield and monoexponential PL decay dynamics are obtained. Notably, for the first time, a record-breaking stability at the single QD level in water with nonblinking behavior persisting for an hour is achieved, which approaching those of state-of-the-art hydrophobic QDs. Furthermore, the nanocomposites formulated with these novel alloyed QDs demonstrate a remarkable PL QY of 96 % and provide an ultrasensitive detection for prostate specific antigen on lateral flow immunoassays. These findings presented here shed new light on the design of high-brightness water-soluble QDs in single-molecule level and QDs-based nanocomposites, significantly pushing ahead toward high-sensitivity biomedical detections and diagnosis.

Abstract:Recently, there has been a lot of interest in silicon quantum dots (Sil-QDts) because of their special opto-electronic properties. These qualities include broad absorption spectra, strong photo-bleaching stability, and size-tunable photoluminescence, which can range from visible to near-infrared depending on its size. Because of their high biocompatibility, low cytotoxicity, and vast surface modification capacity, these nanoparticles have the potential to be used in a wide range of biological and biomedical applications, including sensing, bio-imaging, and photodynamic therapy. This paper presents a thorough analysis of current developments using Sil-QDts in sensor technology. Various types of sensors that use Sil-QDts are examined in detail, such as metal ion sensors, biosensors, food quality sensors, and pesticide and pollution detectors. This paper also addresses prospective applications and future possibilities of Sil-QDt-based sensors.

Abstract:Quantum dots (QDs) have been facilitating the development of sensitive fluorescence biosensors over the past two decades due to their high quantum yield, narrow and tunable emission spectrum and good photostability. The new emerging QDs with improved biocompatibility further promote their biological applications. In this review, we first briefly introduce the prevalently used QDs and their preparation and bioconjugation approaches. Then we summarize QDs-based fluorescent biosensing for proteins and nucleic acids, and QDs-based applications in cellular and in vivo targeting and imaging. Last but not the least, we envision the potential QDs-based applications in future perspectives.

Abstract:Silicon nanomaterials are one of the most representative inorganic nanomedicines in therapeutic biomedicine. Silicon quantum dots (Si QDs) have aroused wide attention in the biomedical field owing to their abundant natural reserves, favorable biocompatibility, and attractive optical properties. In recent years, extensive efforts have been devoted to exploring efficient strategies for the synthesis of Si QDs suitable for versatile biomedical applications. Herein, this Review focuses on the recent exciting advances in how Si QDs can be designed into promising biomedical materials. Rational design of the fabrication methods and modifications of Si QDs is discussed, which provides a generic idea for the biomedical exploration of modified Si QDs. In addition, up-to-date research hotspots of Si QD applications in the biomedical field are also summarized, including disease diagnosis/therapeutics, bioanalyte sensing, and tissue engineering. This Review aims to provide material researchers with deep insights into the design of novel Si QDs for biomedical applications.
One-Pot Synthesis of Silicon Quantum Dots-Based Fluorescent Nanomaterial and Its Application

Abstract:Conventionally obtained silicon quantum dots (Si QDs) generally suffer from the disadvantages of a cumbersome preparation process, large fluctuation in the quality of Si QDs, poor water solubility, and aggregation-caused quenching (ACQ) phenomenon. Here we report a facile one-pot strategy to synthesize a novel Si QDs-based fluorescent nanomaterial in which Si QDs are confined into dendritic mesoporous silica, named as SiQDs@DMSNs. The prepared SiQDs@DMSNs, with adjustable particle sizes ranging from 140 to 300 nm, emit blue fluorescence around 410 nm upon excitation by ultraviolet light at a wavelength of 300 nm. It is found that the addition of sodium salicylate (NaSAL) plays a crucial role in the in situ generation of Si QDs. The obtained SiQDs@DMSNs exhibit excellent fluorescence intensity, water solubility, and stability, facilitating easy surface modification, without being limited by the ACQ phenomenon. It is expected to be widely used in many fields such as biosensors, nanomedicines, in vivo imaging, fingerprint identification, and anticounterfeiting labels.

Abstract:Silicon quantum dots (Si QDs) are usually smaller than 10 nm. They have drawn much attention from researchers because of their novel electronic and optical properties caused by quantum confinement effect and surface effect, which are different from those of bulk silicon materials. In recent years, Si QDs have been applied in the field of optoelectronics because of their novel electronic and optical properties, and a series of research progress have been achieved. The electronic and optical properties of Si QDs are overviewed. The use of Si QDs in optoelectronic devices such as light-emitting diodes, solar cells and photodetectors is introduced in detail. The performance of different types of Si QDs in optoelectronic devices is analyzed as well. It is believed that if continuous efforts in the research on Si QDs are made, Si QDs will play a crucial role in the innovation of optoelectronic devices in the future.

Abstract:Silicon quantum dots (SiQDs) have fully demonstrated their applicability in light of the fluorescence, the extension of their applications to other fields, especially considering their excellent biocompatibility, would be more appealing. Herein, a kind of versatile nitrogen-doped silicon quantum dots (N-SiQDs) was facilely synthesized via one-pot hydrothermal method with 3-aminopropyltrimethoxysilane (APTMS) and tetraethylpentylamine (TEPA) as sources. The N-SiQDs were used as a probe for the bacterial imaging owing to its good fluorescence property, stability and biocompatibility. Besides, thanks to N doping rendering the N-SiQDs stronger reducibility and Au affinity, the N-SiQDs displayed unique reduction capability, and were attempted as a reducing agent and stabilizer for the synthesis of the nanocomposite, i.e. N-SiQDs stabilized Au nanoparticles (N-SiQDs-AuNPs), under mild conditions. The N-SiQDs-AuNPs showed superior catalytic performance to citric-AuNPs due to the synergistical catalytic effect. In addition, the N-SiQDs exhibited good antibacterial properties against Gram-positive (S. aureus) and Gram-negative bacteria (E. coli) without obvious negative influence on the cells, particularly avoiding the use of any other external stimulation. This study may open a new avenue for using the SiQDs for synthesis of catalyst and other biomedicine applications beyond as the fluorescence probe.

Comprehensive Reviews in Food Science and Food Safety

Abstract:Food quality and safety are crucial public health concerns with global significance. In recent years, a series of fluorescence detection technologies have been widely used in the detection/monitoring of food quality and safety. Due to the advantages of wide detection range, high sensitivity, convenient and fast detection, and strong specificity, quantum dot (QD)-based fluorescent nanosensors have emerged as preferred candidates for food quality and safety analysis. In this comprehensive review, several common types of QD production methods are introduced, including colloidal synthesis, self-assembly, plasma synthesis, viral assembly, electrochemical assembly, and heavy-metal-free synthesis. The optoelectronic properties of QDs are described in detail at the electronic level, and the effect of food matrices on QDs was summarized. Recent advancements in the field of QD-based fluorescent nanosensors for trace level detection and monitoring of volatile components, heavy metal ions, food additives, pesticide residues, veterinary-drug residues, other chemical components, mycotoxins, foodborne pathogens, humidity, and temperature are also thoroughly summarized. Moreover, we discuss the limitations of the QD-based fluorescent nanosensors and present the challenges and future prospects for developing QD-based fluorescent nanosensors. As shown by numerous publications in the field, QD sensors have the advantages of strong anti-interference ability, convenient and quick operation, good linear response, and wide detection range. However, the reported assays are laboratory-focused and have not been industrialized and commercialized. Promising research needs to examine the potential applications of bionanotechnology in QD-based fluorescent nanosensors, and focus on the development of smart packaging films, labeled test strips, and portable kits-based sensors.

Colloidally stable silicon quantum dots as temperature biosensors

Abstract:Among semiconductor quantum dots, silicon nanocrystals (SiNCs) are gaining interest due to their high biocompatibility and natural abundance of silicon. The optical properties make SiNCs optimal candidates for luminescent bioprobes: (i) emission energy tuneable to the red and NIR spectral region, compatible with the biological window; (ii) high photoluminescence quantum yield; (iii) no sensitivity to molecular oxygen despite the long lifetime of emission; (iv) long-lived luminescence that enables time-gated detection in the hundreds of μs timescale, which allows removal of scattered excitation light and autofluorescence of the biological sample with a low-cost equipment (gating times of the order of hundreds of μs). SiNCs can be passivated through covalent bond formation between Si and C atoms giving extremely robust systems. The two major issues for SiNCs are the poor absorption outside the UV spectral region and the difficulty to obtain water suspendable SiNCs that maintain their photo physical properties. Our group addressed the first problem by introducing light absorbing units on the SiNCs surface; these dyes can be excited and transfer the energy to the silicon core behaving like a light-harvesting antenna. Regarding the second issue, several attempts are reported in literature to make SiNCs water dispersible, but these approaches often suffer of scarce stability in aqueous environment, and loss of SiNCs photophysical properties. Our approach is based on covalent functionalization of SiNCs via two-step synthesis involving a first coating step in organic solvent and a post-functionalization through thiol-ene click chemistry in order to introduce poly(ethyleneglycol) (PEG) as a water soluble group. SiNCs obtained with this approach are colloidally stable and retain NIR emission with long emission lifetimes (tens of microsecond).

Abstract:BACKGROUND: Quantum dots (QDs) are generally nanosized inorganic particles. They have distinctive size-dependent optical properties due to their very small size (mostly < 10 nm). QDs are regarded as promising new fluorescent materials for biological labeling and imaging because of their superior properties compared with traditional organic molecular dyes. These properties include high quantum efficiency, long-term photostability and very narrow emission but broad absorption spectra.OBJECTIVE/METHODS: Recent developments in synthesizing high quality semiconductor QDs (mainly metal-chalcogenide compounds) and forming biocompatible structures for biomedical applications are discussed in this paper.RESULTS/CONCLUSIONS: This information may facilitate the research to create new materials/technologies for future clinical applications.

Abstract:Fluorescent semiconductor quantum dots (QDs) have attracted tremendous attention over the last decade. The superior optical properties of QDs over conventional organic dyes make them attractive labels for a wide variety of biomedical applications, whereas their potential toxicity and instability in biological environment have puzzled scientific researchers. Much research effort has been devoted to surface modification and functionalization of QDs to make them versatile probes for biomedical applications, and significant progress has been made over the last several years. This review article aims to describe the current state-of-the-art of the synthesis, modification, bioconjugation, and applications of QDs for in vivo targeted imaging. In addition, QD-based multifunctional nanoprobes are also summarized.

Abstract:In-situ, ultra-sensitive, real-time and dynamic acquisition of biochemical information of life is one of the pending key problems for bioscience. It is crucial to develop new related methodologies for life sciences. Fluorescent semiconductor quantum dots bring opportunities to the field, whose novel fluorescence properties are attracting more and more interest. However, some problems involved in quantum dot applications still need to be solved, including controllable solubilization of quantum dots, orientation of biomolecules on their surfaces, and the stability and specificity of quantum dot-labeled bioprobes etc. This review comments briefly on the labeling technique based on quantum dots, construction of quantum dot-labeled bioprobes and their applications in biomedical fields.

Abstract:One of the fastest moving and most exciting interfaces of nanotechnology is the use of quantum dots (QDs) in biology. The unique optical properties of QDs make them appealing as in vivo and in vitro fluorophores in a variety of biological investigations, in which traditional fluorescent labels based on organic molecules fall short of providing long-term stability and simultaneous detection of multiple signals. The ability to make QDs water soluble and target them to specific biomolecules has led to promising applications in cellular labelling, deep-tissue imaging, assay labelling and as efficient fluorescence resonance energy transfer donors. Despite recent progress, much work still needs to be done to achieve reproducible and robust surface functionalization and develop flexible bioconjugation techniques. In this review, we look at current methods for preparing QD bioconjugates as well as presenting an overview of applications. The potential of QDs in biology has just begun to be realized and new avenues will arise as our ability to manipulate these materials improves.

Abstract:Bacterial infections have become a global public health problem. Rapid and sensitive bacterial detection is of great importance for human health. Among various sensor systems, fluorescence sensor is rapid, portable, multiplexed, and cost-efficient. Herein, we reviewed the current trends of fluorescent sensors for bacterial detection from three aspects (response materials, target and recognition way). The fluorescent materials have the advantages of high fluorescent strength, high stability, and good biocompatibility. They provide a new path for bacterial detection. Several recent fluorescent nanomaterials for bacterial detection, including semiconductor quantum dots (QDs), carbon dots (CDs), up-conversion nanoparticles (UCNPs) and metal organic frameworks (MOFs), were introduced. Their optical properties and detection mechanisms were analyzed and compared. For different response targets in the detection process, we studied the fluorescence strategy using DNA, bacteria, and metabolites as the response target. In addition, we classified the recognition way between nanomaterial and target, including specific recognition methods based on aptamers, antibodies, bacteriophages, and non-specific recognition methods based on biological functional materials. The characteristics of different recognition methods were summarized. Finally, the weaknesses and future development of bacterial fluorescence sensor were discussed. This review provides new insights into the application of fluorescent sensing systems as an important tool for bacterial detection.
