Abstract:Enantioselective control of free radical reactions has eluded organic chemists for decades. Echoed with the re-naissance of photo-induced processes, or so called photocatalysis or photoredox catalysis in organic synthesis, photo-induced organic radical chemistry has regained its prominence in developing catalytic asymmetric radical reaction. The generally mild conditions inherited with photochemistry, particularly visible light photo-processes, have allowed for controllable generation of free radicals as well as the subsequent bond formations. The past five years have witnessed dramatic advances in exploring photo-induced catalytic asymmetric free radical reactions, and enormous potentials along this line are envisaged. This perspective gives a brief summary on the important advances in this field. Accordingly, the major advances are classified based on different radical species including α -amino/oxyl radicals, radicals generated from enones and its analogues, benzyl radicals, α -carbonyl radicals, polyhalogenated alkyl radicals and nitrogen radicals. Brief discussion of mechanism is presented whenever relevant.
Photoinduced Chemomimetic Biocatalysis for Enantioselective Intermolecular Radical Conjugate Addition

Abstract:Exploiting nature’s catalysts for non-natural transformations that are inaccessible to chemocatalysis is highly desirable but challenging. On the one hand, the widespread nicotinamide-dependent oxidoreductases have not been utilized for single-electron-transfer-induced bimolecular cross-couplings; on the other, the addition of catalytic asymmetric radical conjugate to terminal alkenes remains a challenge owing to strong racemic background reaction and unselective termination of prochiral radical species. Here we report a chemomimetic biocatalysitic approach for construction of alpha-carbonyl stereocentres via an unnatural intermolecular conjugate addition of N -(acyloxy)phthalimides-derived radicals with acceptor-substituted terminal alkenes, by combination of visible-light excitation and nicotinamide-dependent ketoreductases (KREDs). Based on protein crystal structure, we engineered KREDs via a semi-rational mutagenesis strategy to improve reaction outcomes with a small and high-quality variants library. Mechanistic investigations combining wet experiments, crystallographic studies and computational simulations demonstrate that the repurposed biocatalyst can suppress racemic background reaction and unselected side reactions, yielding enantioselectivity that is challenging to achieve by chemocatalysis.
Photoredox Catalysis of Aromatic β-Ketoester in Situ toward Transient and Persistent Radicals for Organic Transformation.

Abstract:Radical formation is an initial step for conventional radical chemistry. Reported herein is a unified strategy to generate radicals in situ from aromatic β -ketoester only by a photocatalyst. Under visible light irradiation, a small amount of photocatalyst fac -Ir(ppy) 3 succeeds in the generation of transient α -carbonyl radical and persistent ketyl radical in situ , for the first time. In contrast to the well-established approaches, neither stoichiometric external oxidant nor reductant is required for this reaction. The synthetic utility is demonstrated by pinacol-coupling of ketyl radicals and benzannulation of α -carbonyl radicals with alkynes to give a series of highly substituted 1-naphthols in good to excellent yields. Provided the readily available photocatalyst, extremely mild reaction condition, broad substrate scope and functionality tolerance offer a unique perspective in synthetic planning and implementation.

Abstract:Carbohydrates, one of the three primary macromolecules of living organisms, play significant roles in various biological processes such as intercellular communication, cell recognition, and immune activity. While the majority of established methods for the installation of carbohydrates through the anomeric carbon rely on nucleophilic displacement, anomeric radicals represent an attractive alternative because of their functional group compatibility and high anomeric selectivities. Herein, we demonstrate that anomeric nucleophiles such as C1 stannanes can be converted into anomeric radicals by merging Cu(I) catalysis with blue light irradiation to achieve highly stereoselective C(sp3)-S cross-coupling reactions. Mechanistic studies and DFT calculations revealed that the C-S bond-forming step occurs via the transfer of the anomeric radical directly to a sulfur electrophile bound to Cu(II) species. This pathway complements a radical chain observed for photochemical metal-free conditions where a disulfide initiator can be activated by a Lewis base additive. Both strategies utilize anomeric nucleophiles as efficient radical donors and achieve a switch from an ionic to a radical pathway. Taken together, the stability of glycosyl nucleophiles, a broad substrate scope, and high anomeric selectivities observed for the thermal and photochemical protocols make this novel C-S cross coupling a practical tool for late-stage glycodiversification of bioactive natural products and drug candidates.

Abstract:Single-electron reduction of a carbonyl by photoredox catalysis enables the generation of ketyl radicals, providing a benign procedure for constructing C-C bonds. However, external photocatalysts are required. In this paper, photoinduced generation of ketyl radicals and application in C-C coupling are reported, which require no use of any external photocatalyst. Irradiation of carbonyl with purple light in the presence of a tertiary amine generates highly reactive ketyl radicals, enabling a series of important reactions in synthetic chemistry, such as the reductive coupling of aldehydes, ketones, and imines, the allylation of aldehydes, the Barbier reaction, reductive arylation, and aldehyde-aniline coupling. We also present evidence to support the ketyl radicals being formed via photoinduced intermolecular electron transfer from the tertiary amine to aryl aldehydes.

Abstract:Enzymes are recognized as exceptional catalysts for achieving high stereoselectivities 1,2,3 , but their ability to control the reactivity and stereoinduction of free radicals lags behind that of chemical catalysts 4 . Thiamine diphosphate (ThDP)-dependent enzymes 5 are well-characterized systems that inspired the development of N -heterocyclic carbenes (NHCs) 6,7,8 but have not yet been proved viable in asymmetric radical transformations. There is a lack of a biocompatible and general radical-generation mechanism, as nature prefers to avoid radicals that may be harmful to biological systems 9 . Here we repurpose a ThDP-dependent lyase as a stereoselective radical acyl transferase (RAT) through protein engineering and combination with organophotoredox catalysis 10 . Enzyme-bound ThDP-derived ketyl radicals are selectively generated through single-electron oxidation by a photoexcited organic dye and then cross-coupled with prochiral alkyl radicals with high enantioselectivity. Diverse chiral ketones are prepared from aldehydes and redox-active esters (35 examples, up to 97% enantiomeric excess (e.e.)) by this method. Mechanistic studies reveal that this previously elusive dual-enzyme catalysis/photocatalysis directs radicals with the unique ThDP cofactor and evolvable active site. This work not only expands the repertoire of biocatalysis but also provides a unique strategy for controlling radicals with enzymes, complementing existing chemical tools.

Abstract:The selective coupling of two different radicals is a long-standing challenge due to the process occurring statistically. Generally, the cross-coupling selectivity is achieved by the persistent radical effect (PRE), resulting in limited radical precursors. In this paper, a highly selective cross-coupling of alkyl radicals with acyl radicals to construct C(sp2)-C(sp3) bonds, or with alkyl radicals to construct C(sp3)-C(sp3) bonds have been achieved with the readily available carboxylic acids and their derivatives (NHPI ester) as coupling partners. The success originates from the use of tridentate ligand (L1) to enable radical cross-coupling process to Ni-mediated organometallic mechanism. This protocol offers a facile and flexible access to structurally diverse ketones (up to 90% yield), and also a new solution for the challenging double decarboxylative C(sp3)-C(sp3) coupling. The broad utility and functional group tolerance are further illustrated by the late-stage functionalization of natural-occurring carboxylic acids and drugs.

A Photoenzyme for Challenging Lactam Radical Cyclizations

Abstract:Reductive radical cyclizations are ubiquitous in organic synthesis and have been applied to the synthesis of structurally complex molecules. N-heterocyclic motifs can be prepared through the cyclization of α-haloamides; however, slow rotation around the amide C-N bond results in preferential formation of an acyclic hydrodehalogenated product. Here, we compare four different methods for preparing γ, δ, ε, and ζ-lactams via radical cyclization. We found that a photoenzymatic method using flavin-dependent 'ene'-reductases affords the highest level of product selectivity. We suggest that through selective binding of the cis amide isomer, the enzyme preorganizes the substrate for cyclization, helping to avoid premature radical termination.

Abstract:Photoenzymatic catalysts are attractive for stereoselective radical reactions because the transformation occurs within tunable enzyme active sites. When using flavoproteins for non-natural photoenzymatic reactions, reductive mechanisms are often used for radical initiation. Oxidative mechanisms for radical formation would enable abundant functional groups, such as amines and carboxylic acids, to serve as radical precursors. However, excited state flavin is short-lived in many proteins because of rapid quenching by the protein scaffold. Here we report that adding an exogenous Ru(bpy) 3 2+ cofactor to flavin-dependent 'ene'-reductases enables the redox-neutral decarboxylative coupling of amino acids with vinylpyridines with high yield and enantioselectivity. Additionally, stereo-complementary enzymes are found to provide access to both enantiomers of the product. Mechanistic studies indicate that Ru(bpy) 3 2+ binds to the protein, helping to localize radical formation to the enzyme's active site. This work expands the types of transformation that can be rendered asymmetric using photoenzymatic catalysis and provides an intriguing mechanism of radical initiation.

Abstract:Photoenzymatic catalysis is an emerging platform for asymmetric synthesis. In most of these reactions, the protein templates a charge transfer complex between the cofactor and substrate, which absorbs in the blue region of the electromagnetic spectrum. Here, we report the engineering of a photoenzymatic ‘ene’-reductase to utilize red light (620 nm) for a radical cyclization reaction. Mechanistic studies indicate that red light ac-tivity is achieved by introducing a broadly absorbing shoulder off the previously identified cyan absorption feature. Molecular dynamics simulations, docking, and excited-state calculations suggest that red light absorption is a 𝜋→ 𝜋* transition from flavin to the substrate, while the cyan feature is the red-shift of the flavin 𝜋→ 𝜋* transition, which occurs upon substrate binding. Differences in the excitation event help to disfavor alkylation of the flavin cofactor, a pathway for catalyst decomposition observed with cyan light but not red.

Abstract:Radical/radical cross-coupling reactions represent an efficient and straightforward approach for the construction of chemical bonds and accordingly have drawn increasing attention over the past decades. In order to achieve synthetically useful transformations, a persistent radical should be coupled with a transient radical in accordance with the persistent radical effect (PRE). However, known transient radicals outnumber by far the known types of free persistent radicals, which limits the widespread application of the PRE, until today. Thus, the development of efficient cross-coupling reactions between transient radicals has been in focus, and meanwhile transition-metal catalysis has been successfully implemented to artificially prolong radical lifetimes, allowing their utilization in formal radical/radical cross-couplings. Complementary research in the field recently uncovered that organocatalytically generated NHC-derived ketyl radicals are a type of catalytically generated free persistent radicals. NHC-catalyzed radical transformations of aldehydes and carboxylic acid derivatives have enabled the disclosure of an ever-increasing number of interesting reactions, which are different from traditional NHC-catalyzed ionic processes, offering otherwise inaccessible activation modes. These discoveries have opened a door to NHC organocatalysis for the manipulation of radical reactions. Due to its obvious potential in synthetic organic chemistry, it is timely to provide a perspective on this emerging field.

Abstract:Photocatalysis opens up a new window for carbonyl chemistry. Despite a multitude of photochemical reactions of carbonyl compounds, visible light-induced catalytic asymmetric transformations remain elusive and pose a formidable challenge. Accordingly, the development of simple, efficient, and economic catalytic systems is the ideal pursuit for chemists. Herein, we report an enantioselective radical photoaddition to ketones through a Lewis acid-enabled photoredox catalysis wherein the in situ formed chiral N,N '-dioxide/ Sc(III)-ketone complex serves as a temporary photocatalyst to trigger single-electron transfer oxidation of silanes for the generation of nucleophilic radical species, including primary, secondary, and tertiary alkyl radicals, giving various enantioenriched aza-heterocycle-based tertiary alcohols in good to excellent yields and enantioselectivities. The results of electron paramagnetic resonance (EPR) and high-resolution mass spectrum (HRMS) measurements provided favorable evidence for the stereocontrolled radical addition process involved in this reaction.

Abstract:State-of-the-art non-natural photoenzymatic catalysis typically relies on either ultraviolet light activation or the formation of electron donor–acceptor complexes. Inspired by nature, herein we develop a biocatalytic scheme based on direct visible-light excitation of flavoprotein to achieve stereocontrolled intermolecular radical hydroarylation of alkenes with electron-rich arenes. Following visible-light activation, flavin-dependent ene-reductases—which naturally catalyse the two-electron reduction of activated alkenes—are repurposed to trigger single-electron oxidation of arenes and afford C( sp 2 )–C( sp 3 ) bond-forming products in a redox-neutral and enantiodivergent fashion while both enantiomers are obtained with different enzymes. Experiments and computational simulations demonstrate that the reaction is initiated from the single-electron oxidation of substrate by the direct visible-light-excited flavoprotein, and explain the origin of enantioselectivity in this radical hydroarylation that is otherwise notoriously challenging to achieve by chemocatalysis. This work expands the repertoire of photoenzymatic catalysis and will stimulate the development of further biotransformations.

Abstract:Enzymes are increasingly explored for use in asymmetric synthesis1–3, but their applications are generally limited by the reactions available to naturally occurring enzymes. Recently, interest in photocatalysis4 has spurred the discovery of novel reactivity from known enzymes5. However, so far photoinduced enzymatic catalysis6 has not been used for the cross-coupling of two molecules. For example, the intermolecular coupling of alkenes with α-halo carbonyl compounds through a visible-light-induced radical hydroalkylation, which could provide access to important γ-chiral carbonyl compounds, has not yet been achieved by enzymes. The major challenges are the inherent poor photoreactivity of enzymes and the difficulty in achieving stereochemical control of the remote prochiral radical intermediate7. Here we report a visible-light-induced intermolecular radical hydroalkylation of terminal alkenes that does not occur naturally, catalysed by an ‘ene’ reductase using readily available α-halo carbonyl compounds as reactants. This method provides an efficient approach to the synthesis of various carbonyl compounds bearing a γ-stereocentre with excellent yields and enantioselectivities (up to 99 per cent yield with 99 per cent enantiomeric excess), which otherwise are difficult to access using chemocatalysis. Mechanistic studies suggest that the formation of the complex of the substrates (α-halo carbonyl compounds) and the ‘ene’ reductase triggers the enantioselective photoinduced radical reaction. Our work further expands the reactivity repertoire of biocatalytic, synthetically useful asymmetric transformations by the merger of photocatalysis and enzyme catalysis. A transformation in which an ‘ene’ reductase catalyses the visible-light-induced intermolecular radical hydroalkylation of alkenes gives carbonyl compounds with a remote stereocentre in high yield and enantioselectivity.

Abstract:Since the discovery of Hofmann–Löffler–Freytag reaction more than 130 years ago, both the structure and reactivity of nitrogen-centred radicals have been widely studied. Nevertheless, catalytic enantioselective intermolecular radical hydroamination remains a challenge due to the existence of side reactions, the short lifetime of nitrogen-centred radicals and lack of understanding of the fundamental catalytic steps. In the laboratory, nitrogen-centred radicals are produced with radical initiators, photocatalysts or electrocatalysts. In contrast, their generation and reaction are unknown in nature. Here we report a pure biocatalytic system for the photoenzymatic production of nitrogen-centred radicals and enantioselective intermolecular radical hydroaminations by successfully repurposing an ene-reductase through directed evolution. These reactions progress efficiently at room temperature under visible light without any external photocatalysts and exhibit excellent enantioselectivities. A detailed mechanistic study reveals that the enantioselectivity originates from the radical-addition step while the reactivity originates from the ultrafast photoinduced electron transfer from reduced flavin mononucleotide to nitrogen-containing substrates.

Stereodivergent photobiocatalytic radical cyclization through the repurposing and directed evolution of fatty acid photodecarboxylases

Abstract:Despite their intriguing photophysical and photochemical activities, naturally occurring photoenzymes have not yet been repurposed for new-to-nature activities. Here we engineered fatty acid photodecarboxylases to catalyse unnatural photoredox radical C-C bond formation by leveraging the strongly oxidizing excited-state flavoquinone cofactor. Through genome mining, rational engineering and directed evolution, we developed a panel of radical photocyclases to facilitate decarboxylative radical cyclization with excellent chemo-, enantio- and diastereoselectivities. Our high-throughput experimental workflow allowed for the directed evolution of fatty acid photodecarboxylases. An orthogonal set of radical photocyclases was engineered to access all four possible stereoisomers of the stereochemical dyad, affording fully diastereo- and enantiodivergent biotransformations in asymmetric radical biocatalysis. Molecular dynamics simulations show that our evolved radical photocyclases allow near-attack conformations to be easily accessed, enabling chemoselective radical cyclization. The development of stereoselective radical photocyclases provides unnatural C-C-bond-forming activities in natural photoenzyme families, which can be used to tame the stereochemistry of free-radical-mediated reactions.

Abstract:The difunctionalization of alkenes represents a powerful tool to incorporate two functional groups into the alkene bones for increasing molecular complexity and has been widely utilizations in chemical synthesis. Upon the catalysis of the green, sustainable, mild photo-/electrochemistry technologies, much attentions have been attracted to the development of new tactics for the transformations of the important alkene and alkane feedstocks driven by C-H radical functionalization. Herein, we summarize recent advances in the photo-/electrocatalytic difunctionalization of alkenes enabled by C-H radical functionalization. We detailedly discuss the substrate scope and the mechanisms of the photo-/electrocatalytic alkene difunctionalization reactions by selecting impressive synthetic examples, which are divided into four sections based on the final terminated step, including oxidative radical-polar crossover coupling, reductive radical-polar crossover coupling, radical-radical coupling, and transition-metal-catalyzed coupling.

Abstract:N-Heterocyclic carbenes (NHCs) are unique Lewis basic catalysts that mediate various organic transformations by means of polarity reversal. Although the scope of research on two-electron reactions mediated by NHC catalysts has been expanding, the types of these reactions are limited by the inability of NHCs to activate electrophiles with sp3 carbons. However, the revival of photocatalysis has accelerated the development of free-radical chemistry, and combining photoredox catalysis and NHC catalysis to achieve NHC-mediated radical reactions under mild conditions could overcome the above-mentioned limitation. This review summarizes recent advances in combining photoredox and NHC catalysis, focusing on elucidation and exploration of mechanisms, with the aim of identifying challenges and opportunities to develop more types of catalytic models.
