Abstract:The gene of the NAD-dependent formate dehydrogenase (FDH) from the yeast Candida boidinii was cloned by PCR using genomic DNA as a template. Expression of the gene in Escherichia coli yielded functional FDH with about 20% of the soluble cell protein. To confirm the hypothesis of a thiol-coupled inactivation process, both cysteine residues in the primary structure of the enzyme have been exchanged by site-directed mutagenesis using a homology model based on the 3D structure of FDH from Pseudomonas sp. 101 and from related dehydrogenases. Compared to the wt enzyme, most of the mutants were significantly more stable towards oxidative stress in the presence of Cu(II) ions, whereas the temperature optima and kinetic constants of the enzymatic reaction are not significantly altered by the mutations. Determination of the Tm values revealed that the stability at temperatures above 50 degrees C is optimal for the native and the recombinant wt enzyme (Tm 57 degrees C), whereas the Tm values of the mutant enzymes vary in the range 44-52 degrees C. Best results in initial tests concerning the application of the enzyme for regeneration of NADH in biotransformation of trimethyl pyruvate to Ltert leucine were obtained with two mutants, FDHC23S and FDHC23S/C262A, which are significantly more stable than the wt enzyme.

Abstract:Previous research has found that the traditional approach to measuring hazard perception, as used by the UK Government, does not necessarily transfer well to other countries. The speeded response times of a hazard perception test were susceptible to threshold bias, differentiating drivers according to cultural background more than driving experience. An alternative method - the hazard prediction test - appears to mitigate these problems when used across different countries. The current study applies the hazard prediction methodology to a new cultural context (Israel), delivered via an online platform. A further innovation was to include both hazardous and non-hazardous trials allowing measures of criterion and sensitivity parameters of the Signal Detection Theory to be calculated.In total, 74 participants (43 experienced and 31 novices) watched 26 hazard prediction clips that had been filmed in Tel Aviv (Israel), 13 of which contained cues to genuine hazards. Each hazardous clip was occluded prior to the hazardous situation fully materialising, though with sufficient evidence that any participant who was looking in the right place at the right time would be able to correctly identify the unfolding hazard. Participants were first asked to identify whether there was a hazard (a binary decision), and then to predict how the traffic situation was going to develop by choosing one of four options provided after each video.As hypothesised, experienced drivers outperformed novices in the multiple-choice prediction question, provided that they had correctly identified that the clip contained a hazard in the first question. Novices were poor at selecting the correct multiple-choice option, regardless of whether they correctly identified that the clip contained a hazard. Both driving experience and hazard sensitivity significantly predicted multiple-choice accuracy, though criterion did not. The newly developed online test was successful in differentiating between experienced and novice driver groups for accuracy in prediction. These results support the roll-out of the hazard prediction methodology across different countries. Practitioner summary: This study provides new insights into how to develop an online hazard prediction test suitable for international export. The results suggest that this newly developed test is successful in differentiating between experienced and novice drivers. This raises the possibilit y of assessing and training drivers on a global scale.
Road Hazard Reaction Testing Using Driving Simulation: the Novice Vs. the Experienced Drivers

Abstract:It is believed that the novice drivers are less prepared for potential road hazards and their reaction mode to hazards is different from the experienced drivers. Main road hazards in China were identified using focus group discussions and questionnaires. Then virtual reality technology was used to develop five most important hazards for experimental study. In this preliminary study, two groups of Chinese drivers (8 novice and 8 experienced) were tested on a driving simulation system. Their reaction to the hazards in terms of average driving speed, braking operation against suddenly appeared hazards, and wheel steering were automatically collected and analyzed. Results indicate that experienced drivers drive slower before potential hazards and their reaction to sudden appeared hazards are more appropriate. Potential impacts of this method and result include developing interventions for novice driver training program and developing hazard warning devices.
Development and Evaluation of a Virtual Environment to Assess Cycling Hazard Perception Skills

Abstract:Safe cycling requires situational awareness to identify and perceive hazards in the environment to react to and avoid dangerous situations. Concurrently, tending to external distractions leads to a failure to identify hazards or to respond appropriately in a time-constrained manner. Hazard perception training can enhance the ability to identify and react to potential dangers while cycling. Although cycling on the road in the presence of driving cars provides an excellent opportunity to develop and evaluate hazard perception skills, there are obvious ethical and practical risks, requiring extensive resources to facilitate safety, particularly when involving children. Therefore, we developed a Cycling and Hazard Perception virtual reality (VR) simulator (CHP-VR simulator) to create a safe environment where hazard perception can be evaluated and/or trained in a real-time setting. The player interacts in the virtual environment through a stationary bike, where sensors on the bike transfer the player's position and actions (speed and road positioning) into the virtual environment. A VR headset provides a real-world experience for the player, and a procedural content generation (PCG) algorithm enables the generation of playable artifacts. Pilot data using experienced adult cyclists was collected to develop and evaluate the VR simulator through measuring gaze behavior, both in VR and in situ. A comparable scene (cycling past a parked bus) in VR and in situ was used. In this scenario, cyclists fixated 20% longer at the bus in VR compared to in situ. However, limited agreement identified that the mean differences fell within 95% confidence intervals. The observed differences were likely attributed to a lower number of concurrently appearing elements (i.e., cars) in the VR environment compared with in situ. Future work will explore feasibility testing in young children by increasing assets and incorporating a game scoring system to direct attention to overt and covert hazards.
Correlations among self-report, static image, and video-based hazard perception assessments: The validity of a new Lithuanian hazard prediction test

Abstract:Scholars suggest that improving hazard perception (HP) skills among different road users can lead to safer driving. To this end, a small number of countries have developed a national hazard perception test for their licensing procedures. The purpose of the current research is to develop and validate a new video-based hazard prediction test for the Lithuanian driving context. Eighty-eight drivers participated in the study. Initially, 25 naturalistic clips were included and edited into a hazard prediction test following the "what happens next?" paradigm. The overall score of the test was higher for experienced drivers when compared with novices. On further inspection the overall group difference was driven by 12 clips. The Lithuanian Hazard Perception test HPT-LIT featuring static images (Tūskė et al., 2019) and Hazard Perception Questionnaire developed by White et al. (2011) were used to validate the new test. This new version of the test (12 clips, LHP12) showed acceptable psychometric properties. Drivers with less driving experience demonstrated poorer hazard prediction skills than experienced drivers. The results revealed a negative correlation between hazard prediction scores on LHP12 and crash involvement in the novice driver group, as well as a positive correlation between prediction scores and self-reported hazard prediction skills. It was concluded that the new Lithuanian hazard test shows potential for future research.

Abstract:Hazard perception, as a dynamic indicator, intuitively characterizes drivers' ability to detect potential hazard events in complex driving environments. However, current quantitative research on the hazard perceptions of drivers in the human–machine codriving phase is lacking. A total of 30 drivers were recruited to carry out a driving simulator experiment, and a total of 8 risky driving scenarios were experienced. The physiological characteristics and eye movement characteristics of the drivers were collected. An expert evaluation method was selected to quantify the situational hazard degree (SHD). The questionnaire was used to collect the drivers' subjective hazard evaluations (SHEs) of the scenarios. Paired samples t test and correlation analysis were adopted to screen the evaluation indexes of the drivers' hazard perceptions. A gray near-optimal comprehensive evaluation model was established to quantify the human–machine codrivers' hazard perceptions in various risky driving scenarios. The results show that the drivers' SHEs increased with the SHD. The standard deviation of heart rate, mean pupil diameter, and mean fixation duration can be used as evaluation indexes for the human–machine codrivers' hazard perceptions. The proposed quantitative method is expected to shift the development of human–machine codrivers' hazard perception from qualitative analysis to quantitative calculation. The research results can provide a theoretical basis for increasing safety in the human–machine codriving phase, improving road safety and reducing the incidence of accidents.

Abstract:Road traffic crashes are the leading cause of death for young people, among whom cyclists account for a higher percentage of injuries and deaths than any other road users. This study aimed to examine the factor structure of the Young Cyclist Behavior Questionnaire (YCBQ) and investigate the relationships among demographic characteristics, cycling use‐related variables, perceived risk, perceived cycling skills, and risky cycling behaviors among young people. A sample of 448 cyclists (mean age of 20.37 years) completed the questionnaire. Exploratory factor analysis, confirmatory factor analysis, and structural equation modeling were utilized. The YCBQ had a clear factorial structure, items with high factor loadings, and good internal consistency. The five‐factor structure included traffic violations, impulsive behaviors, ordinary violations, distractions, and errors. Risky cycling behaviors could be explained by gender, age, perceived risk, and perceived cycling skills, with the model explaining 37% of the variance. Gender had the greatest impact on risky cycling behaviors; male individuals were more likely to engage in risky behaviors. Young cyclists with higher levels of perceived risk had lower probabilities of engaging in risky cycling behaviors. Cyclists with lower scores on perceived cycling skills were more likely to report engaging in risky cycling behaviors. Age significantly explained risky behaviors; the younger the cyclist was, the higher his or her risky behaviors score. This research provides a theoretical foundation for the prevention of risky behaviors among young cyclists. Regarding intervention design, attention to the identified gender differences, the need to strengthen the ability to perceive risk, and the importance of road safety education for young cyclists may promote safer cycling.

Abstract:Road traffic accidents resulting in group deaths and injuries are often related to coach drivers' inappropriate operations and behaviors. Thus, the evaluation of coach drivers' fitness to drive is an important measure for improving the safety of public transportation. Previous related research focused on drivers' age and health condition. Comprehensive studies about commercial drivers' cognitive capacities are limited. This study developed a toolkit consisting of nine cognition measurements across driver perception/sensation, attention, and reaction. A total of 1413 licensed coach drivers in Jiangsu Province, China were investigated and tested. Results indicated that drivers with accident history within three years performed overwhelmingly worse (p<0.001) on dark adaptation, dynamic visual acuity, depth perception, attention concentration, attention span, and significantly worse (p<0.05) on reaction to complex tasks compared with drivers with clear accident records. These findings supported that in the assessment of fitness to drive, cognitive capacities are sensitive to the detection of drivers with accident proneness. We first developed a simple evaluation model based on the percentile distribution of all single measurements, which defined the normal range of "fit-to-drive" by eliminating a 5% tail of each measurement. A comprehensive evaluation model was later constructed based on the kernel principal component analysis, in which the eliminated 5% tail was calculated from on integrated index. Methods to categorizing qualified, good, and excellent coach drivers and criteria for evaluating and training Chinese coach drivers' fitness to drive were also proposed.

Abstract:Road safety has become one of the major social problems in China now with the rapid motorization in both urban and rural areas in recent years. Literatures indicate that human error is the most important reason for road fatalities. Education of drivers for typical and common road hazards could be an effective approach to reduce fatalities. Using virtual reality technologies, this study designed 5 typical road hazard scenarios which are both common and potentially dangerous for the consequence in China. Totally 24 participants of experienced automobile drivers in Beijing(12) and Hengyang (12) were recruited for the test in comparative study experiment. The objective of this research is to compare whether there are some differences in their response-to these five road hazards. Findings of the study are important in understanding how the experienced gain their knowledge and experience in dealing with the common road hazards as well as why there are differences in these two areas. The further impact would be how to design corresponding intervention training program to reflect the differences.
Potential Risk and Its Influencing Factors for Separated Bicycle Paths.

Abstract:In this paper, we propose two potential risk indicators to define and evaluate the safety of bicycle path at the microscopic level. Field bicycle data were collected from three survey sites under different traffic conditions. These two risk indicators based on speed dispersion were proposed and calculated during each 5-min interval. The risk influences of various widths of bicycle path and traffic conditions were analyzed by using one-way ANOVA. We further proposed a generalized linear model (GLM) for modeling and analyzing the relationships between bicycle risks and v/c ratio and percentages of electric bicycles, male cyclists, young cyclists, and loaded cyclists. The stepwise regression models were applied for determination of coefficients. The results show that the influences of gender and age of cyclists on potential risks are not significant. The risks increase with the width of bicycle path and percentage of electric bicycles, while only for wider bicycle path (4-lane case in this study), the risks are associated with whether or not cyclists are loaded. The findings could contribute for analysis and evaluation of the safety for bicycle path.

Abstract:Vehicle-pedestrian accidents are one of the main types of road traffic accidents in China because of their mixed traffic features. By analyzing the characteristics of vehicle-pedestrian accidents, the head injury criterion (HIC) was selected as a quantitative index of pedestrian head injury risk, and vehicle-pedestrian collision simulation tests were carried out using PC-Crash. From the collected test data, the multivariate relationship models between the HIC, vehicle speed, and collision angle were fitted for different vehicle types. A risk assessment method for vehicle-pedestrian accidents based on the HIC was proposed by the Fisher optimal segmentation algorithm. Finally, a new index for evaluating the accuracy of accident risk classification, the degree of error classification, was proposed to verify the validity of the accident risk assessment method. The results show that vehicle speed, collision angle, and vehicle type play a key role in pedestrian injury. Flat-headed vehicles are more likely to cause head injuries to pedestrians than high-headed and low-headed vehicles. Rear-end collisions cause more injuries to pedestrians than side collisions. The research results can provide guidance and a basis for accident liability determination, speed limit management, vehicle safety design, and human injury mechanism analysis.

A novel protein-based anticancer drug encapsulating nanosphere: apoferritin-doxorubicin complex.

Abstract:Developing a drug delivery system, which is uniform, biocompatible, stable and non-toxic, is a challenging issue in anticancer drug delivery strategies. Ferritin is a nano-size spherical protein with an internal cavity where drug molecules can be encapsulated. The apoferritin-doxorubicin complex has been formed by 'opening' and 'closing' the apoferritin sphere in the presence of doxorubicin. The doxorubicin encapsulation was carried out using direct and step-wise change of pH of the solution from 2.5 to 7.4. Non-denaturing polyacrylamide gels showed that the protein cage of the complex successfully self-assembles into its nanosphere form. It was found that up to 28 molecules of doxorubicin can be capsulated per apoferritin protein and no significant drug leakage occurs during the first two days. The apoferritin-doxorubicin complex is a promising nanocarrier for the delivery of anticancer drugs.

Abstract:(1) Background: As cycling gains popularity as a mode of transportation, the frequency of accidents involving cyclists also rises. This has become a major concern for traffic safety, sustainability, and city planning. Identifying the risk factors that contribute to bicycle road accidents remains a significant challenge. This study aims to figure out which risk factors make some road segments more dangerous for cyclists than others. (2) Methods: This study introduces the use of a bicycle simulator to test different road segments involving thirty-nine participants. The impact of demographics and some risk factors related to infrastructure were analyzed in terms of their influence on the perceived level of risk through pre- and post-surveys. (3) Results: The findings showed that the bicycle facility type affects the perceived level of risk. Shared-use roads were ranked as riskiest, while separated bike lanes were least risky. Bicycle roads with no separated safety barriers had higher risks. Heavy traffic jams increased danger among cyclists. Women gave higher risk ratings than men. The perceived levels of risk were then compared with the previously developed risk index and they correlated well. (4) Conclusions: This confirms that the risk index can reliably evaluate the degree of risk of each road segment.

Road user hazard perception tests: A systematic review of current methodologies

Abstract:Background: Poor hazard perception, or the ability to anticipate potentially dangerous road and traffic situations, has been linked to an increased crash risk. Novice and younger road users are typically poorer at hazard perception than experienced and older road users. Road traffic authorities have recognised the importance of hazard perception skills, with the inclusion of a hazard perception test in most Graduated Driver Licensing (GDL) systems. Objectives: This review synthesises studies of hazard perception tests in order to determine best practice methodologies that discriminate between novice/younger and experienced/older road users. Data sources: Published studies available on PsychInfo, Scopus and Medline as at April 2018 were included in the review. Studies included a hazard perception test methodology and compared non-clinical populations of road users (car drivers, motorcyclists, bicyclists and pedestrians), based on age and experience, or compared methodologies. Results: 49 studies met the inclusion criteria. There was a high degree of heterogeneity in the studies. However all methodologies - video, static image, simulator and real-world test-drive were able to discriminate road user groups categorised by age and/or experience, on at least one measure of hazard perception. Conclusions: Whilst there was a high level of heterogeneity of studies, video methodology utilising temporal responses (e.g. press a button when detecting the potential hazard) are a consistent measure of hazard perception across road user groups, whereas spatial measures (e.g. locate potential hazard in the scenario) were inconsistent. Staged footage was found to discriminate as well as unstaged footage, with static images also adding valuable information on hazard perception. There were considerable inconsistencies in the categorising of participants based on age and experience, limited application of theoretical frameworks, and a considerable lack of detail regarding post hoc amendments of hazardous scenarios. This research can guide further developments in hazard perception testing that may improve driver licensing and outcomes for road users.
Mutations in the vitamin D receptor gene in three kindreds associated with hereditary vitamin D resistant rickets.

Abstract:Hereditary vitamin D resistant rickets has been associated with a number of mutations within the DNA and ligand binding domains of vitamin D receptors (VDR). The aim of our study was to identify and characterize the causative mutations in three kindreds with this condition. Resistance of 1,25(OH)2D3 was confirmed in cultured skin fibroblasts in which there was no induction of 24-hydroxylase activity; binding of 1,25(OH)2D3 to VDR was undetectable in patients 1 and 2, but normal in patients 3 and 4. The coding region of the VDR gene was sequenced to seek mutations. A mutation in the VDR gene of patient 1 resulted in a STOP codon, patient 2 showed a 56 bp deletion leading to frameshift and premature termination of VDR; a point mutation of A to C lying within the hormone-binding domain was shown for patients 3 and 4, who were siblings. Transactivation studies confirmed that these were functional mutations. Gel shift assays using nuclear extract from patient 3 demonstrated that the mutation that altered a conserved amino acid (glutamine-259) known to be involved in heterodimerization with other nuclear receptors affected protein: protein interactions.
Predicting and factor analysis of rider injury severity in two-wheeled motorcycle and vehicle crash accidents based on an interpretable machine learning framework

Abstract:OBJECTIVE: As one of the vulnerable road users in accidents, how to improve the two-wheeled motorcyclist's driving safety and reduce accident injury is a public health issue. Accurate identification of the factors influencing the severity of accidents is an important prerequisite for mitigating injury from crashes.METHODS: Based on a vehicle and a two-wheeled motorcycle crash accident data from the China in-depth accident study database (CIDAS), this study uses the performance evaluation indicators of accuracy, precision, recall, F1-score, AUC, and the ROC curve. The classification and prediction performances of the six machine learning methods on the dataset are compared, and the LightGBM algorithm with the best performance is selected to model the accident injury severity of the motorcyclists. The SHAP method is used to extend the interpretability of the LightGBM model results. Based on the SHAP method, the importance, main effect, and the interaction effect of factors under each accident injury severity are quantitatively analyzed.RESULTS: The model prediction accuracy is 92.6%, the F1-Score is 92.8%, and the AUC value is 0.986. The importance of factors varies with the accident injury severity of motorcyclists. The kilometers traveled per year by the driver, the throwing distance of the motorcyclist, and the road speed limit are the three most important factors. The motorcyclist is more likely to suffer fatal injuries when the throwing distance is >1,000 cm.CONCLUSIONS: The prediction model of driver injury severity based on LightGBM algorithm has a good prediction performance. It can be used to analyze the influence factors of injury severity in two-wheeled motorcyclist accident by combining the model with SHAP method. These results could help the traffic management department to take measures to reduce accident injury of motorcyclists.

Abstract:Two-wheeled traffic (referring to electric bikes and regular bikes in this paper) plays an important role in urban transport system of China. Their safety problems have become an issue of increasing public concern in recent years. Red light running (RLR) is one kind of frequent and highly dangerous behaviors for two-wheeled riders. The primary purpose of this research was to better understand this risky behavior using the planned behavior (TPB) framework. A questionnaire based on TPB approach, which measures two-wheeled riders' intentions to cross against red light in different situations, their attitude towards the behavior, subjective norms, perceived behavioral control, risk perception, utility perception and self-identity, was administered to 232 respondents. It was found that male riders had stronger RLR intentions than the females, and those riders who reported having an accident in the past three years also reported higher RLR intentions. A predictive model including traditional TPB variables, extended TPB variables and demographic measures could explain 33.3 of the variance in RLR intention. Subjective norm, perceived behavioral control, utility perception, self-identity, and gender all emerged as significant predictors. Finally, practical implications for these results were discussed. © 2011 ASCE.
Bicyclist's violation and risk perception in different situations

Abstract:The aim of this study was to describe bicyclists' violating behavior and perception of risk in different situations in mixed traffic and to clarify whether or not bicyclists were familiar with the traffic rules and aware of their violating behavior in China. One observation and one on-line survey were done in this study. The focus of the observation was on the passing lane for bicyclists crossing. An on-line survey was conducted to explore bicyclers' risk perception and knowledge about traffic rules. Both Chinese and international participants who had the cycling experience in China responded to the survey. The results showed that bicyclists committed a lot of violations and often put themselves in dangerous situations. International participants perceived the potential risk of riding bicycle in China significantly higher than Chinese participants (p = 0.000). The knowledge of traffic rules and regulations was very well.

Abstract:The radiotherapy centres are using linear accelerators equipped with multi-leaf collimators (MLCs) for treatments of various types of cancer. For superficial cancers located at a maximum depth of 3 cm high-energy electrons are often used, but MLC cannot be used together with electron applicators. Due to the fact that the tumour shape is not square (as electron applicators), searching for different materials that can be used as absorbents or shields for the protection of adjacent organs is of paramount importance. This study presents an experimental study regarding the transmitted dose through some laboratory-made materials when subjected to electron beams of various energies (ranging from 6 to 15 MeV). The investigated samples were composite materials consisting of silicon rubber and micrometre aluminium particles with different thicknesses and various mass fraction of aluminium. The measurements were performed at a source surface distance of 100 cm in the acrylic phantom. The experimental results show that the transmitted dose through tested samples is ranging between ∼1.8 and 90%, depending on the electron beam energy, sample thickness and sample composition. These preliminary results suggest that the analysed materials can be used as absorbers or shields in different applications in radiotherapy and radioprotection.
