Abstract:This study investigated the indigenous functional microbial communities associated with the degradation of chiral fungicide mandipropamid enantiomers in soils repeatedly treated with a single enantiomer. The R-enantiomer degraded faster than the S-enantiomer, with degradation half-lives ranging from 10.2 d to 79.2 d for the R-enantiomer and 10.4 d to 130.5 d for the S-enantiomer. Six bacterial genera, (Burkholderia, Paraburkholderia, Hyphomicrobium, Methylobacterium, Caballeronia, and Ralstonia) with R-enantiomer substrate preference and three bacterial genera (Haliangium, Sorangium, and Sandaracinus) with S-enantiomer substate preference were responsible for the preferential degradation of the R-enantiomer and S-enantiomer, respectively. KEGG analysis indicated that Burkholderia, Paraburkholderia, Hyphomicrobium, and Methylobacterium were the dominant contributors to soil microbial metabolic functions. Notably, six microbial metabolic pathways and twelve functional enzyme genes were associated with the preferential degradation of the R-enantiomer, whose relative abundances in the R-enantiomer treatment were higher than those in the S-enantiomer treatment. A constructed biodegradation gene (BDG) protein database analysis further confirmed that Burkholderia, Paraburkholderia, Hyphomicrobium, Methylobacterium, and Ralstonia were the potential hosts of five dominant BDGs, bphA1, benA, bph, p450, and ppah. We concluded that bacterial genera Burkholderia, Paraburkholderia, Hyphomicrobium, and Methylobacterium may play pivotal roles in the preferential degradation of mandipropamid R-enantiomer in repeatedly treated soils.

Abstract:Pesticides residues in soils and on vegetables are a public safety concern. Pretreatment with microorganisms degrading pesticides has the potential to alleviate the conditions. For this purpose, the degradation characteristics of chlorpyrifos by an isolated fungal strain Verticillium sp. DSP in pure cultures, soil, and on pakchoi (Brassica chinensis L.) were investigated. Degradation rate of chlorpyrifos in the mineral salts medium was proportional to the concentrations of chlorpyrifos ranging from 1 to 100mgl−1. The rate of degradation for chlorpyrifos (1mgl−1) in the mineral salts medium was 1.12 and 1.04 times faster at pH 7.0 than those at pHs 5.0 and 9.0, and the degradation at 35°C was 1.15 and 1.12 times faster, respectively, than those at 15 and 20°C. The addition of the fungal strain DSP into the contaminated soils was found to significantly increase the degradation of chlorpyrifos. Degradation rates of chlorpyrifos in inoculated soils were 3.61, 1.50 and 1.10 times faster in comparison with the sterilized soil, previously chlorpyrifos-untreated soil, and previously chlorpyrifos-treated soil under laboratory conditions. In contrast to the controls, the half-lives of chlorpyrifos were significantly shortened by 10.9% and 17.6% on treated pakchoi, 12.0% and 37.1% in inoculated soils, respectively, in the greenhouse and open field. The results indicate that the fungal strain DSP can be used successfully for the removal or detoxification of chlorpyrifos residues in/on contaminated soil and vegetable.

Abstract:The application of the herbicide chlorimuron-ethyl has a lasting toxic effect on some succession crops. Here, a bacterium capable of utilizing chlorimuron-ethyl as the sole source of nitrogen was isolated from the contaminated soil and was identified as Enterobacter ludwigii sp. CE-1, and its detoxification and degradation of the herbicide were then examined. The biodegradation of chlorimuron-ethyl by the isolate CE-1 was significantly accelerated with increasing concentration (1–10mg/l) and temperature (20–40°C). The optimal pH for the degradation of chlorimuron-ethyl by the isolate CE-1 was pH 7.0. A pathway for the biodegradation of chlorimuron-ethyl by the isolate CE-1 was proposed, in which it could be first converted into 2-amino-4-chloro-6-methoxypyrimidine and an intermediate product by the cleavage of the sulfonylurea bridge and then transformed into saccharin via hydrolysis and amidation. The plant height and fresh weight of corn that had been incubated in nutrient solution containing 0.2mg/l of chlorimuron-ethyl significantly recovered to 83.9% and 83.1% compared with those in the uninoculated control, although the root growth inhibition of chlorimuron-ethyl could not be alleviated after inoculation for 14 d. The results indicate that the isolate CE-1 is a promising bacterial resource for the biodegradation and detoxification of chlorimuron-ethyl.

Abstract:ZJ0273,Propyl 4-(2-(4,6-dimethoxypyrimidin-2-yloxy) benzylamino) benzoate,is a novel herbicide with the property of high activity,broad spectrum and low toxicity to animals,and it is developed for pre-and post-emergence weed control in the oilseed rape field.The highly efficient ZJ0273 degrading bacterial strain M3-1 was screened,isolated and separated from farmland of long-term using pyrimidinylthiobenzoate herbicides by applying modern molecular biological techniques together with the traditional microbiological methods,and it can grow on by using ZJ0273 as the sole carbon and energy source.The strain M3-1 was identified as Amycolatopsis sp.by physiological,biochemical tests and 16s rDNA sequence.The optimum ZJ0273 degrading conditions of using strain M3-1 and their application in soil were studied.The results show that with condition of 35℃,pH 6.0,8% of inoculation and 100 mg?L?1 of ZJ0273,the degrading ratio in 25 d is 63.3%.Using strain M3-1,the degradation ratio of ZJ0273 is decreased with the increasing ZJ0273 concentration;300 mg?L?1 of ZJ0273 can strongly inhibit bio-activity of strain M3-1,which means that higher concentration of ZJ0273 has higher toxicity to M3-1.The strain M3-1also has a good degradation effect to ZJ0273 in soil,the degradation ratio could up to 53.7% after 25 days.The physicochemical properties of strain M3-1 may open new possibilities for its biotechnological applications and allow the use of Amycolatopsis sp.M3-1 in the soil bioremediation.

Abstract:In this study, the fate of paichongding was investigated in three soils with contrasting soil properties. In general, low soil pH has the potential to retard the mineralization and promote the dissipation of paichongding and the formation of its primary transformation product and to accelerate the formation of bound residue. The dissipation of paichongding stereoisomers was very fast and diastereoselective. This selectivity was found only between diastereomers and not between enantiomers and was observed to be soil dependent. In the acidic soil, the enantiomers (5R,7R)- and (5S,7S)-paichongding were degraded more quickly than (5R,7S)- and (5S,7R)-paichongding, whereas a contrary trend was observed in the neutral soil, and such selectivity did not occur in the alkaline soil. The OM and clay contents also played important roles in the fate of paichongding. This effect of soil properties should be considered in risk assessment of chiral pesticides and their application in the field.

Abstract:A bacterium, isolated from contaminated soils around a chemical factory and named strain DSP3 was capable of biodegrading both chlorpyrifos and 3,5,6-trichloro-2-pyridinol. Based on the results of phenotypic features, phylogenetic similarity of 16S rRNA gene sequences, DNA G+C content, and DNA homology between strain DSP3 and reference strains, strain DSP3 was identified as Alcaligenes faecalis. Chlorpyrifos was utilized as the sole source of carbon and phosphorus by strain DSP3. We examined the role of strain DSP3 in the degradation of chlorpyrifos and 3,5,6-trichloro-2-pyridinol under different culture conditions. Parathion and diazinon could also be degraded by strain DSP3 when provided as the sole sources of carbon and phosphorus. An addition of strain DSP3 (108cellsg−1) to soil with chlorpyrifos (100mgkg−1) resulted in a higher degradation rate than the one obtained from non-inoculated soils. Different degradation rates of chlorpyrifos in six types of treated soils suggested that soils used for cabbage growing in combination with inoculation of strain DSP3 showed enhanced microbial degradation of chlorpyrifos.
[Isolation and Characterization of a Chlorpyrifos Degrading Bacteria and Its Bioremediation Application in the Soil].

Abstract:A strain DSP3 capable of utilizing chlorpyrifos as the sole carbon and energy sources was isolated. Based on the results of phenotypic features, phylogenetic of 16S rDNA sequence, DNA (G + C) mol% and DNA homology between strain DSP3 and reference strains, the strain DSP3 is identified as Alcaligenes faecalis. The degradation rate of chlorpyrifos was at 98.6% (100 mg/L) in liquid culture medium within 18 days and nearly 100% (100 mg/kg) in soil within 20 days respectively. An addition of strain DSP3 (10(8) cells/g) to soil resulted in a higher degradation rate than noninoculated soils. The different degrading rate of chlorpyrifos in four types of treated soils suggests that the dissipation is mediated by the activity of the soil microorganisms.
Influence of Isolated Bacterial Strains on the in Situ Biodegradation of Endosulfan and the Reduction of Endosulfan- Contaminated Soil Toxicity.

Abstract:The recently discovered endosulfan-degrading bacterial strains Pusillimonas sp. JW2 and Bordetella petrii NS were isolated from endosulfan-polluted water and soil environments. The optimal conditions for the growth and biodegradation activity of the strains JW2 and NS were studied in detail. In addition, the ability of the strains JW2 and NS to biodegrade endosulfan in soils during in situ bioremediation experiments was investigated. At a concentration of 2 mg of endosulfan per kilogram of soil, both JW2 and NS had positive effects on the degradation of endosulfan; JW2 degraded 100% and 91.5% of α- and β-endosulfan, respectively, and NS degraded 95.1% and 90.3% of α- and β-endosulfan, respectively. Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) of soil samples showed the successful colonization of JW2 and NS, and the toxicity of the soil decreased, as determined by single-cell gel electrophoresis (SCGE) assays of Eiseniafetida and micronucleus (MN) assays of Viciafaba root tip cells. Furthermore, the metabolic products of the bacterially degraded endosulfan from the in situ experiments were identified as endosulfan ether and lactone. This study provided potentially foundational backgrounds information for the remediation of endosulfan-contaminated soil.
Enhanced Mineralization of Chlorpyrifos Bound Residues in Soil Through Inoculation of Two Synergistic Degrading Strains.

Abstract:Bioaugmentation methods are frequently employed for pesticide pollution remediation; however, it is not clear whether the introduced bacteria affect the pesticide bound residue (BRs) composition and whether the BRs can be catabolized by the introduced strains. This study aimed at answering these questions by using C-14-chlorpyrifos (C-14-CPF) and two CPF-degrading strains (Pseudomonas sp. DSP-1 and Cupriavidus sp. P2). The results showed that the BRs can be up to 83.0%, and that the CPF-BRs formed can be further transformed into (CO2)-C-14 by the strains. Indeed, the microbial inoculation can increase the CPF mineralization by 1.0-22.1 times and can decrease the BRs by up to similar to 50% of the control (on day 20). Compared with the control without bio-augmentation, microbial inoculation enhanced the release of BRs by 2.2-18.0 times. Adding biochar to the soil can greatly inhibit CPF mineralization and maintain the BR content at a relatively stable level. The CPF residue can affect the composition of the indigenous soil microbial community, but the introduction of bacteria for remediation did not have a significant effect. The results indicate that Pseudomonas sp. DSP-1 and Cupriavidus sp. P2 are useful for remediating both CPF extractable and bound residues.
Pseudomonas aeruginosa based concurrent degradation of beta-cypermethrin and metabolite 3-phenoxybenzaldehyde, and its bioremediation efficacy in contaminated soils

Abstract:Beta-cypermethrin is one of the widely used pyrethroid insecticides, and problems associated with the accumulation of its residues have aroused public attention. Thus, there is an urgent need to effectively remove the beta-cypermethrin that is present in the environment. Biodegradation is considered a cost-effective and environmentally friendly method for removing pesticide residues. However, the beta-cypermethrin-degrading microbes that are currently available are not optimal. In this study, Pseudomonas aeruginosa PAO1 was capable of efficiently degrading beta-cypermethrin and its major metabolite 3-phenoxybenzaldehyde in water/soil environments. Strain PAO1 could remove 91.4% of beta-cypermethrin (50 mg/L) in mineral salt medium within 120 h. At the same time, it also possesses a significant ability to metabolize 3-phenoxybenzaldehyde-a toxic intermediate of beta-cypermethrin. The Andrews equation showed that the maximum substrate utilization concentrations of beta-cypermethrin and 3-phenoxybenzaldehyde by PAO1 were 65.3558 and 49.6808 mg/L, respectively. Box-Behnken design-based response surface methodology revealed optimum conditions for the PAO1 strain-based degradation of beta-cypermethrin as temperature 30.6 °C, pH 7.7, and 0.2 g/L inoculum size. The results of soil remediation experiments showed that indigenous micro-organisms helped to promote the biodegradation of beta-cypermethrin in soil, and beta-cypermethrin half-life in non-sterilized soil was 6.84 days. The bacterium transformed beta-cypermethrin to produce five possible metabolites, including 3-phenoxybenzyl alcohol, methyl 2-(4-hydroxyphenoxy)benzoate, diisobutyl phthalate, 3,5-dimethoxyphenol, and 2,2-dimethyl-1-(4-phenoxyphenyl)propanone. Among them, methyl 2-(4-hydroxyphenoxy)benzoate and 3,5-dimethoxyphenol were first identified as the intermediate products during the beta-cypermethrin degradation. In addition, we propose a degradation pathway for beta-cypermethrin that is metabolized by strain PAO1. Beta-cypermethrin could be biotransformed firstly by hydrolysis of its carboxylester linkage, followed by cleavage of the diaryl bond and subsequent metabolism. Based on the above results, P. aeruginosa PAO1 could be a potent candidate for the beta-cypermethrin-contaminated environmental bioremediation.
High-efficiency degradation of methomyl by the novel bacterial consortium MF0904: Performance, structural analysis, metabolic pathways, and environmental bioremediation

Abstract:Methomyl is a widely used carbamate pesticide, which has adverse biological effects and poses a serious threat to ecological environments and human health. Several bacterial isolates have been investigated for removing methomyl from environment. However, low degradation efficiency and poor environmental adaptability of pure cultures severely limits their potential for bioremediation of methomyl-contaminated environment. Here, a novel microbial consortium, MF0904, can degrade 100% of 25 mg/L methomyl within 96 h, an efficiency higher than that of any other consortia or pure microbes reported so far. The sequencing analysis revealed that Pandoraea, Stenotrophomonas and Paracoccus were the predominant members of MF0904 in the degradation process, suggesting that these genera might play pivotal roles in methomyl biodegradation. Moreover, five new metabolites including ethanamine, 1,2-dimethyldisulfane, 2-hydroxyacetonitrile, N-hydroxyacetamide, and acetaldehyde were identified using gas chromatography-mass spectrometry, indicating that methomyl could be degraded firstly by hydrolysis of its ester bond, followed by cleavage of the C-S ring and subsequent metabolism. Furthermore, MF0904 can successfully colonize and substantially enhance methomyl degradation in different soils, with complete degradation of 25 mg/L methomyl within 96 and 72 h in sterile and nonsterile soil, respectively. Together, the discovery of microbial consortium MF0904 fills a gap in the synergistic metabolism of methomyl at the community level and provides a potential candidate for bioremediation applications.
Selective removal of the non-herbicidal (S)-enantiomer of dichlorprop from agricultural soil by an in-situ enriched consortium

Abstract:Rac-dichlorprop [(R,S)-2-(2,4-dichlorophenoxy)propanoic acid], a commonly used chiral phenoxyalkanoic acid herbicide, is frequently detected in various environments, posing threats to human health and environmental safety. The (S)-enantiomer of dichlorprop shows a lower herbicidal activity but a higher toxicity as compared to the (R)-enantiomer of dichlorprop. It is of great importance to enantioselectively remove the non-herbicidal (S)-dichlorprop from the rac-dichlorprop in the application sites. In this study, the dissipation of (R)- and (S)-dichlorprop in an agricultural soil was investigated. We found that the dissipation rate of (S)-dichlorprop was higher than that of (R)-dichlorprop in an agricultural soil. Although the influences of different enantiomers of dichlorprop on soil bacterial community were not different, Sphingobium was significantly enriched in the (S)-dichlorprop-treated soil as compared to (R)-dichlorprop, indicating that Sphingobium species played important roles in the dissipation of (S)-dichlorprop. Furthermore, a bacterial consortium containing Sphingobium sp. strain SP-2 (enantioselectively transforming (S)-dichlorprop to 2,4-dichlorophenol) and Sphingopyxis sp. strain DP-21 (mineralizing 2,4-dichlorophenol), which synergistically mineralized the (S)-enantiomer of dichlorprop, was isolated from the agricultural soil. Bioaugmentation with the in-situ isolated consortium (strains SP-2 and DP-21) resulted in the enhanced removal of the non-herbicidal (S)-dichlorprop from the soil and in the adaptation of the inoculated degrading strains. This study offers new insights into the enhanced bioremediation of the non-herbicidal (S)-dichlorprop contaminated soils while simultaneously ensuring environmental safety and crop yield.

The key sulfometuron-methyl degrading bacteria isolation based on soil bacterial phylogenetic molecular ecological networks and application for bioremediation of contaminated soil by immobilization

Abstract:The analysis of soil bacterial community has guiding significance for fully utilization of soil microbial resources. The results of high-throughput sequencing (HTS) showed that the bacteria in the three sulfometuron-methyl contaminated soil samples were mainly composed of 677 genera, including Phenylobacterium, Bacillus, belonging to 28 phyla, including Proteobacteria, Firmicutes. The diversity and richness of bacterial community decreased with the increase in sulfometuron-methyl concentration. In addition, sulfometuron-methyl could also affect the soil bacterial function based on PICRUSt functional predictive analysis. Combined with the results of HTS and phylogenetic molecular ecological networks (pMENs), 12 genera, including Ralstonia (Pi=0.64), were identified as the key soil microflora (intra-module connectivity Zi ≥ 2.5 or inter-module connectivity Pi ≥ 0.62), and the abundance of Ralstonia significantly increased with the concentration of sulfometuron-methyl, indicating that the strains of this genus might be the potential degrading bacteria and could form a stable relationship with indigenous microorganisms. Among the isolated bacteria of genus Ralstonia, one strain, named Ralstonia sp. JM-1, was verified to possess higher sulfometuron-methyl degradation efficiency, which completely degraded 20 mg L−1 of sulfometuron-methyl within 96 h. Furthermore, the immobilized strains generated by the mixture of 2.0 g bamboo charcoal and 3.0 mL bacterial suspension for 24 h had the highest sulfometuron-methyl degradation rate than that under other conditions, and the dynamic process degrading 10–30 mg L−1 of sulfometuron-methyl conforms to the zero-order kinetic equation. The bioremediation of contaminated soil showed the immobilized strains could completely degrade sulfometuron-methyl (1.39 mg kg−1) in contaminated soil within 9 d, which is higher than that application of strains in the free state (74.8%). This study could provide ideas for the isolation of functional strains and a theoretical basis for the bioremediation of STM and other contaminated soils.

Abstract:Fomesafen is a long residual herbicide and poses a potential risk to environmental safety, leading to an increasing need to find eco-friendly and cost-effective techniques to remediate fomesafen-contaminated soils. In this article, a novel strain of Bacillus sp., FE-1 was isolated from paddy field soil. This strain was found to degrade fomesafen both in liquid medium and in soil. >82.9% of fomesafen, at concentrations of 0.5, 1 and 10mgL-1, was degraded by Bacillus sp. FE-1 in liquid medium within 14h. The optimal pH and temperature for degradation were 7.0 and 35°C, respectively. Soil samples inoculated with strain FE-1 showed significantly increased rates of fomesafen degradation. Two metabolites of fomesafen degradation were detected and identified as amino-fomesafen and 5-[2-chloro-4-(trifluoromethyl) phenoxy]-2-amino-benzoic acid. This is the first report of a novel fomesafen biodegradation pathway involving the reduction of a nitro group followed by the hydrolysis of an amide bond. The excellent remediation capability of the isolate FE-1 to detoxify fomesafen-contaminated soil was shown by bioassay of the sensitive aftercrop corn. The results indicate that Bacillus sp. FE-1 has potential for use in the bioremediation of fomesafen-contaminated soil.
Biodegradation of chlorpyrifos using isolates from contaminated agricultural soil, its kinetic studies

Abstract:Abstract Extensive pesticides use is negatively disturbing the environment and humans. Pesticide bioremediation with eco-friendly techniques bears prime importance. This study evaluates the bioremediation of chlorpyrifos in soil using indigenous Bacillus cereus Ct3, isolated from cotton growing soils. Strains were identified through ribotyping (16s rRNA) by Macrogen (Macrogen Inc. Geumchen-gu, South Korea). Bacillus cereus Ct3 was resistant up to 125 mg L −1 of chlorpyrifos and successfully degraded 88% of chlorpyfifos in 8 days at pH 8. Bacillus cereus Ct3 tolerated about 30–40 °C of temperature, this is a good sign for in situ bioremediation. Green compost, farmyard manure and rice husk were tested, where ANOVA ( P < 0.05) and Plackett–Burman design, results indicated that the farm yard manure has significant impact on degradation. It reduced the lag phase and brought maximum degradation up to 88%. Inoculum size is a statistically significant ( P < 0.05) factor and below 10 6 (CFU g −1 ) show lag phase of 4–6 days. Michaelis–Menten model results were as follows; R 2 = 0.9919, V max = 18.8, K s = 121.4 and V max /K s = 0.1546. GC–MS study revealed that chlorpyrifos first converted into diethylthiophosphoric acid and 3,5,6-trichloro-2-pyridinol (TCP). Later, TCP ring was broken and it was completely mineralized without any toxic byproduct. Plackett–Burman design was employed to investigate the effect of five factors. The correlation coefficient (R 2 ) between experimental and predicted value is 0.94. Central composite design (CBD) was employed with design matrix of thirty one predicted and experimental values of chlorpyrifos degradation, having “lack of fit P value” of “0.00”. The regression coefficient obtained was R 2 = 0.93 which indicate that the experimental vales and the predicted values are closely fitted. The most significant factors highlighted in CBD/ANOVA and surface response plots were chlorpyrifor concentration and inoculum size. Bacillus cereus Ct3 effectively degraded chlorpyrifos and can successfully be used for bioremediation of chlorpyrifos contaminated soils.

Abstract:This study investigated indigenous functional microbial communities associated with the degradation of chloroacetamide herbicides acetochlor (ACE), S-metolachlor (S-MET) and their enantiomers in repeatedly treated soils. The results showed that biodegradation was the main process for the degradation of ACE, S-MET and their enantiomers. Eight dominant bacterial genera associated with the degradation were found: Amycolatopsis, Saccharomonospora, Mycoplasma, Myroides, Mycobacterium, Burkholderia, Afipia, and Kribbella. The S-enantiomers of ACE and S-MET were preferentially degraded, which mainly relied on Amycolatopsis, Saccharomonospora and Kribbella for the ACE S-enantiomer and Amycolatopsis and Saccharomonospora for the S-MET S-enantiomer. Importantly, the relative abundances of Amycolatopsis and Saccharomonospora increased by 146.3%–4467.2% in the S-enantiomer treatments of ACE and S-MET compared with the control, which were significantly higher than that in the corresponding R-enantiomer treatments (25.3%–4168.2%). Both metagenomic and qPCR analyses demonstrated that four genes, ppah, alkb, benA, and p450, were the dominant biodegradation genes (BDGs) potentially involved in the preferential degradation of the S-enantiomers of ACE and S-MET. Furthermore, network analysis suggested that Amycolatopsis, Saccharomonospora, Mycoplasma, Myroides, and Mycobacterium were the potential hosts of these four BDGs. Our findings indicated that Amycolatopsis and Saccharomonospora might play pivotal roles in the preferential degradation of the S-enantiomers of ACE and S-MET.

Abstract:A micro segmented-flow approach was utilized for the isolation soil bacteria that can degrade synthetic polymers as polyethylene glycols (PEG) and polyacrylamide (PAM). We had been able to obtain many strains; among them, five Achromobacter spanius strains from soil samples of specific sampling sites that were connected with ancient human impacts. In addition to the characterization of community responses and isolating single strains, this microfluidic approach allowed for investigation of the susceptibility of Achromobacter spanius strains against three synthetic polymers, including PEG, PAM, and Polyvinylpyrrolidone (PVP) and two organic solvents known as 1,4-dioxane and diglyme. The small stepwise variation of effector concentrations in 500 nL droplets provides a detailed reflection of the concentration-dependent response of bacterial growth and endogenous autofluorescence activity. As a result, all five strains can use PEG600 as carbon source. Furthermore, all strains showed similar dose-response characteristics in 1,4-dioxane and diglyme. However, significantly different PAM- and PVP-tolerances were found for these strains. Samples from the surface soil of prehistorical rampart areas supplied a strain capable of degradation of PEG, PVP, and PAM. This study demonstrates on the one hand, the potential of microsegment flow for miniaturized dose-response screening studies and its ability to detect novel strains, and on the other hand, two of five isolated Achromobacter spanius strains may be useful in providing optimal growth conditions in bioremediation and biodegradation processes.

Abstract:Chlorimuron-ethyl is currently the primary herbicide used for chemical weed control in a soybean field. In this study, a solid microbial inoculum (corn stalk-white rot fungus (W-1)) was prepared for the remediation of farmland soil contaminated by chlorimuron-ethyl. Firstly, the preparation method of the microbial inoculum was studied. Secondly, the degradation rate of the chlorimuron-ethyl in the ground by the solid microbial inoculum is improved by optimizing the proportion of the protective agent. Then the effects of applying solid microbial inoculum, free bacteria and corn straw on the degradation rate of chlorimuron-ethyl in soil were weighed. Finally, Illumina MiSeq sequencing was used to measure the composition and diversity of bacterial and fungal communities in the ground before and after using microbial inoculum. The degradation rate of chlorimuron-ethyl in soil by solid microbial inoculum was 84.87% after 20 d using corn straw as the support, room temperature drying, 4% Ca 3 (PO 4 ) 2 as the protective drying agent, and 1%(w) dextrin as the ultraviolet protective agent. Inoculation of white rot fungi could significantly affect the community structure of bacteria and fungi in the soil, making the chlorimuron-ethyl degrading communities become the dominant communities and playing an essential role in the degradation of chlorimuron-ethyl. The results showed that using solid microbial inoculum was an effective way to repair farmland soil polluted by chlorimuron-ethyl.

Enantioselective Biodegradation and Enantiomerization of Dichlorprop in Soils.

Abstract:The dissipation of racemic, R-, and S- dichlorprop (DCPP) in four soils were studied in the laboratory. The half-lives of racemic DCPP were from 10.5 to 19.8 days. Preferential degradation of R- or S-DCPP was detected in all soils, even in one soil that the apparent enantiomeric fraction remained constant during incubation. The enantiomerization of DCPP was found to proceed in both directions, except in forest soil that no enantiomerization of S- to R-DCPP was observed. The isomerization equilibrium constant (K = k(RS)/k(SR)) in two vegetable soils were 0.54 and 0.53, respectively, favoring herbicidally active R enantiomer, while in paddy soil K was 1.60, favoring an inversion of R into S enantiomer. Real-time PCR showed that the rdpA gene was not detected in all indigenous and DCPP amended microcosms probably because of relative short incubation time and low amendment concentrations. In contrast, the sdpA gene was present in indigenous soils and significantly elevated after DCPP addition with the highest relative abundance around day 10 in all microcosms. Illumina sequencing of the 16S rRNA gene showed that the relative abundance of Proteobacteria significantly increased in all DCPP treated soils. DCPP-degrading related families, Sphingomonadaceae and Comamonadaceae, enhanced in all soils, while Burkholder-iaceae elevated only in paddy soil with preferential degradation of S-DCPP and Pseudomonadaceae only in forest soil with R-enantiomer preference. The sdpA gene sequencing revealed that about 92%-99% of bacteria harboring sdpA genes in studied soils belong to Alphaproteobacteria. (C) 2020 Elsevier Ltd. All rights reserved.
