Abstract:Background: Viscum album L. (Santalaceae), commonly known as mistletoe, is a hemiparasitic plant traditionally used in complementary cancer treatment. Its antitumor potential is mostly attributed to the presence of aqueous soluble metabolites; however, the use of ethanol as solvent also permits the extraction of pharmacological compounds with antitumor potential. The clinical efficacy of mistletoe therapy inspired the present work, which focuses on ethanolic extracts (V. album "mother tinctures", MT) prepared from different host trees. Methods: Samples from three European subspecies (album, austriacum, and abietis) were harvested, and five different V. album-MT strains were prepared. The following phytochemical analyses were performed: thin layer chromatography (TLC), high-performance liquid chromatography (HPLC) and liquid chromatography-high resolution mass spectrometry (LC-HRMS). The proliferation assay was performed with WST-1 after incubation of tumor (Yoshida and Molt-4) and fibroblast cell lines (NIH/3 T3) with different MT concentrations (0.5 to 0.05% v/v). The cell death mechanism was investigated by flow cytometry (FACS) using Annexin V-7AAD. Results: Chemical analyses of MT showed the presence of phenolic acids, flavonoids and lignans. The MT flavonoid and viscotoxin contents (mg/g fresh weight) were highest in Quercus robur (9.67 ± 0.85 mg/g) and Malus domestica (3.95 ± 0.58 mg/mg), respectively. The viscotoxin isoform proportions (% total) were also different among the VA subspecies with a higher content of A3 in V. album growing on Abies alba (60.57 ± 2.13). The phytochemical compounds as well as the viscotoxin contents are probably related to the antitumor effects of MT. The cell death mechanisms evaluated by colorimetric and FACS methodologies involved necrotic damage, which was host tree-, time- and dose- dependent, with different selectivity to tumor cells. Mother tincture from V. album ssp. abietis was the most effective at inducing in vitro cellular effects, even when incubated at the smallest concentration tested, probably because of the higher content of VT A3. Conclusion: Our results indicate the promising antitumor potential of Viscum album ethanolic extracts and the importance of botanical and phytochemical characterization for in vitro anti-proliferative effects.
Proteome analysis of leaf, stem and callus in Viscum album and identification of lectins and viscotoxins with bioactive properties

Abstract:Mistletoes are semiparasite plants containing pharmaceutical proteins with applications in cancer treatment. Previous research has demonstrated that somaclonal variation can lead to the biosynthesis of novel proteins from mistletoe callus cultures. The protein content of Viscum album subsp. abietis tissues and biotechnologically propagated calluses, was analyzed to identify proteins with putative anticancer properties. In addition, evolutionary relations among linked species to Viscum were studied. Calluses were propagated from stem explants. The protein extracts mass spectra were processed with Proteome Discoverer and a search was performed using as reference the Uniprot V. album reviewed database. A phylogenetic tree was reconstructed using the LG amino acid substitution model by homologous sequences for Beta galactoside-specific lectin 2. The homology modeling of the Beta-galactoside-specific lectin 2 was carried out using Modeller software. Considerable differences were observed by comparing the protein content of the calluses and the maternal tissues. Four mistletoe lectins, six viscotoxins and the chitin binding lectin-cbML were identified within the species tissues. An in silico phylogenetic and structural study provides insights to the role of these lectins and the mechanism of semiparasite survival and evolution, towards a novel anticancer and immune system modulation pipeline. Callogenesis exhibited protein biosynthesis alterations and novel protein isoforms expression. Phyllogenetic analysis revealed evolutionary relations primarily within the Viscum genus and other species containing 2-ribosome inactivating proteins. The homology modeling of the mistletoe lectin 2 revealed possible structure related anticancer properties. In conclusion, mistletoe calluses were shown to possess a unique protein biosynthetic profile compared to donor plant tissues.

Among various species, the lipophilic extract of the Viscum album subsp. austriacum (Wiesb.) Vollm. (VALE) has shown promising therapeutic potential. The present work aimed to qualify the plant source and characterize the extract's chemical profile. In addition, a self-nanoemulsifying drug delivery system (SNEDDS) containing VALE (SNEDDS-VALE) was developed. Methods: V. album subsp. austriacum histochemistry was performed, and the chemical profile of VALE was analyzed by GC-MS. After the SNEEDS-VALE development, its morphology was visualized by transmission electron microscopy (TEM), while its stability was evaluated by the average droplet size, polydispersity index (PdI) and pH. Lastly, SNEDDS-VALE chemical stability was evaluated by LC-DAD-MS. Results: The histochemical analysis showed the presence of lipophilic compounds in the leaves and stems. The major compound in the VALE was oleanolic acid, followed by lupeol acetate and ursolic acid. SNEDDS was composed of medium chain triglyceride and Kolliphor ® RH 40 (PEG-40 hydrogenated castor oil). A homogeneous, isotropic and stable nanoemulsion was obtained, with an average size of 36.87 ± 1.04 nm and PdI of 0.14 ± 0.02, for 14 weeks. Conclusion: This is the first histochemistry analysis of V. album subsp. austriacum growing on Pinus sylvestris L . which provided detailed information regarding its lipophilic compounds. A homogeneous, isotropic and stable SNEDDS-VALE was obtained to improve the low water solubility of VALE. Further, in vitro and in vivo experiments should be performed, in order to evaluate the antitumoral potential of SNEDDS-VALE. Abstract:We investigated the association of time to prostate-specific antigen nadir (TTPN) and logarithm of prostate-specific antigen velocity after progression Log(PSAVAP) in metastatic prostate cancer with prior primary androgen deprivation therapy (ADT). All metastatic prostate cancer patients treated with primary ADT from 2000 to 2009 were reviewed. Patients who developed disease progression were included in the subsequent analyses. Patients were categorized into three groups according to their TTPN: TTPN of <3 months, 3-17 months, and >17 months. We compared the Log(PSAVAP) between the different TTPN groups using Mann-Whitney U-test and Kruskal-Wallis test. Further multiple linear regression analyses on Log(PSAVAP) were performed to adjust for other potential confounding factors. Among 419 patients who were treated with primary ADT, 306 patients developed disease progression with a median follow-up of 28 months. Longer TTPN was associated with lower Log(PSAVAP) (P = 0.008) within all subgroup analyses (TTPN of <3 vs 3-17 months, P= 0.020; TTPN of 3-17 vs >17 months, P= 0.009; and TTPN of <3 vs >17 months, P= 0.001). Upon multiple linear regression analyses, baseline PSA (regression coefficient 0.001, P= 0.045), PSA nadir (regression coefficient 0.002, P= 0.040), and TTPN (regression coefficient −0.030, P= 0.001) were the three factors that were significantly associated with Log(PSAVAP). In conclusion, a longer TTPN was associated with lower Log(PSAVAP) in metastatic prostate cancer patients following primary ADT. TTPN cut-offs at 3 months and 17 months appeared to have prognostic significance in predicting Log(PSAVAP). TTPN may serve as a good prognostic indicator in deciding the treatment strategy in patients with disease progression.

Abstract:Objective: comparative study of the amino acid and elemental composition of leaves Viscum album L. and host plants. Materials and methods. The amino acid and elemental composition in raw materials and extracts from leaves Viscum album and the corresponding host plant, obtained by extraction with 50% ethyl alcohol, was determined by capillary electrophoresis. Results. Glycine, alanine, leucine, proline, methionine, valine, asparagine, aspartic and glutamic acids were previously identified in the leaves Viscum album by ascending paper chromatography. By capillary electrophoresis, it was found that the raw materials and extracts contained proline, valine, alanine, threonine and methionine in the greatest amount. It has been established that in a semi-parasitic plant, the content of proline is 16 times higher than in Malus domestica Borkh. leaves and 5 times higher than in Pyrus communis L. leaves In alcohol-water extracts, the proline content is 5 and 1,5 times higher, respectively. It should be noted that in raw materials and extracts from Viscum album, obtained by extraction with 50% ethyl alcohol, potassium ions predominates quantitatively. Compared to leaves Malus domestica, leaves Viscum album accumulate 4,5 times more potassium, and 1,5 times more than in Pyrus communis leaves; in extracts, the content is also higher by 3 and 2 times, respectively. Conclusion. In raw materials and extracts from Viscum album, 13 free amino acids were found, of which 6 are essential (Arg, Phe, Leu, Met, Thr, Val). It was found that potassium predominates quantitatively in raw materials and extracts from Viscum album.
Separation and Purification Process of Total Flavonoids of Mistletoe with Macroporous Resins

Abstract:Objective:To choose the optimum resin for separation and purification of mistletoe (Viscum album) flavonoids and to analyze influenceable factors of separation and purification process. Methods:Four macroporous resins including AB-8, NKA-9, NKA-II and D101 were used to separation of total flavonoids of mistletoe (Viscum album). During the adsorption and desorption process, with UV to assay total flavonoids, the different macroporous resins and their capacities were studied. Results:AB-8 resin possesses much higher adsorption and desorption capacity. The optimum conditions are:the feed solution pH 4, the flow rate 2 BV/h, 40% alcohol as eluant for desorption, the flow rate 1 BV/h and eluant volume 4 BV. An enriched total flavonoids extract with 43.56% purity can be separated from crude mistletoe extract (about 12.16% purity) in one run. Conclusion:It is feasible to separate and purify mistletoe flavonoids with AB-8 resin.

Abstract:Mistletoe is an evergreen woody shrub with stems measuring 30–100 cm. It has leathery, yellowish-green strap-shaped leaves, yellowish-green flowers, and bears typical berries. The most common species is Viscum album L., mainly present in Europe and Asia. It is commonly known as European mistletoe or simply, mistletoe. Scientific interest in mistletoe was awakened in the XX century. Mistletoe, especially V. album L., has historically been used in the treatment and prophylaxis of CVD, with its properties being confirmed in recent studies. This mini-review describes new aspects of the cardioprotective properties of various species of mistletoe, especially V. album L. The effect of oral and subcutaneous application of fresh V. album L. extracts on blood pressure has been studied in various models; while the data suggests that mistletoe may be a promising herbal extract with cardioprotective properties, the species has only been tested in vitro and in vivo , on animals. In addition, it is unclear whether the cardioprotective activity of mistletoe may be due to particular chemical components, as the chemical composition of mistletoe extracts can vary depending on inter alia the time of harvest, extraction method and plant part. Hence, this activity may instead result from synergistic interactions between various secondary metabolites. Therefore, further studies are needed to identify the mechanisms of action of mistletoe compounds on CVDs, and determine their interactions with other cardioprotective drugs, their metabolic mechanisms, pharamacokinetics and adverse effects. More research is also needed to determine the therapeutic doses of active ingredients for use in clinical trials; this would require an accurate understanding of the chemical composition of extracts from different species of mistletoe (not only V. album L.) and from various host trees.

Abstract:Introduction: European mistletoe ( Viscum album L.) has been gaining increasing interest in the field of oncology as a clinically relevant adjunctive treatment in many forms of cancer. In the field of phytopharmacology, harvesting time is pivotal. In the last century, a form of metabolomic fingerprinting based on pattern formation was proposed as a way to determine optimal harvesting times to ensure high quality of mistletoe as raw material for pharmaceutical use. In order to further evaluate the information obtained with this metabolomic fingerprinting method, we analysed a large time series of previously undigitised daily mistletoe chromatograms dating back to the 1950s. Methods: These chromatograms were scanned and evaluated using computerized image analysis, resulting in 12 descriptors for each individual chromatogram. We performed a statistical analysis of the data obtained, investigating statistical distributions, cross-correlations and time self-correlations. Results: The analysed dataset spanning about 27 years, contains 19,037 evaluable chromatograms in daily resolution. Based on the distribution and cross-correlation analyses, the 12 descriptors could be clustered into six independent groups describing different aspects of the chromatograms. One descriptor was found to mirror the annual rhythm being well correlated with temperature and a phase shift of 10 days. The time self-correlation analysis showed that most other descriptors had a characteristic self-correlation of ∼50 days, which points to further infradian rhythms (i.e., more than 24 h). Discussion: To our knowledge, this dataset is the largest of its type. The combination of this form of metabolomic fingerprinting with the proposed computer analysis seems to be a promising tool to characterise biological variations of mistletoe. Additional research is underway to further analyse the different rhythms present in this dataset.

Abstract:The hemiparasitic flowering plant Viscum album (European mistletoe) is known for its very special life cycle, extraordinary biochemical properties, and extremely large genome. The size of its genome is estimated to be 30 times larger than the human genome and 600 times larger than the genome of the model plant Arabidopsis thaliana. To achieve insights into the Gene Space of the genome, which is defined as the space including and surrounding protein-coding regions, a transcriptome project based on PacBio sequencing has recently been conducted. A database resulting from this project contains sequences of 39,092 different open reading frames encoding 32,064 distinct proteins. Based on 'Benchmarking Universal Single-Copy Orthologs' (BUSCO) analysis, the completeness of the database was estimated to be in the range of 78%. To further develop this database, we performed a transcriptome project of V. album organs harvested in summer and winter based on Illumina sequencing. Data from both sequencing strategies were combined. The new V. album Gene Space database II (VaGs II) contains 90,039 sequences and has a completeness of 93% as revealed by BUSCO analysis. Sequences from other organisms, particularly fungi, which are known to colonize mistletoe leaves, have been removed. To evaluate the quality of the new database, proteome data of a mitochondrial fraction of V. album were re-analyzed. Compared to the original evaluation published five years ago, nearly 1000 additional proteins could be identified in the mitochondrial fraction, providing new insights into the Oxidative Phosphorylation System of V. album. The VaGs II database is available at https://viscumalbum.pflanzenproteomik.de/. Furthermore, all V. album sequences have been uploaded at the European Nucleotide Archive (ENA).

Abstract:There were only non-significant relationships between NSC concentrations in mistletoe and in its host species across the nine mistletoe-host pairs, suggesting the carbon condition of ssp. is determined by both the heterotrophic carbon transfer and self-photosynthetic capacity among different mistletoe-host pairs. However, mistletoe leaf morphological traits (single leaf area and mass, and leaf mass per unit leaf area) did not change across the nine mistletoe-host pairs, and mistletoe leaf δ13C, water content and macronutrient concentrations were linearly correlated with those in the host leaves. Macronutrients showed accumulations in mistletoe across the nine pairs. Further, tissue N concentrations were significantly higher in mistletoe grown on N-fixing hosts than on non-N-fixing hosts. Finally, leaf N:P in mistletoe was significantly correlated with the ratio in the host across the nine mistletoe-host pairs. Overall, our results indicate strong relationships between mistletoe and its hosts for water- and nutrient-related traits, but not for carbon-related traits, which demonstrates that ssp. album can adjust its physiology to survive on different deciduous tree species hosts and under different site conditions.
Comparative study of three local mistletoes: insights from photosynthetic, ionomic, and metabolomic attributes

Abstract:Mistletoes are adapted to parasitize aerial parts of their hosts, and they have a great morphological variation and wide global distribution. Their effects on hosts were more or less investigated; however, no comparative interspecific study had been reported. This study aimed to compare three locale mistletoes Phragmanthera austroarabica , Viscum schimperi , and Plicosepalus acacia based on photosynthetic activity, major-trace- and ultra-trace elements' distribution and metabolome profiling. P. acacia exhibited the highest net photosynthetic assimilation rate (3.3-folds), water use efficiency (5.5-folds), and accumulated major elements ( 44 Ca, 24 Mg, and 23 Na). Equally, the untargeted metabolomic method showed a substantial difference in metabolome profiling, with P. acacia being the lesser accumulative of major metabolites than P. austroarabica and V. schimperi . Based on the few identified common metabolite analysis, P. acacia had the ability to avoid the accumulation of the pollutant mono(2-ethylhexyl) phthalate which, P. austroarabica and V. schimperi , cannot do. Equally, P. acacia had more defence metabolites against stressors than the other mistletoes. Accordingly, P. acacia displayed higher potentialities to be more vigor and more adapted to the parasitism's life than P. austroarabica and V. schimperi.

Abstract:Responsive nanomaterials have emerged as promising candidates as drug delivery vehicles in order to address biomedical diseases such as cancer. In this work, polymer-based responsive nanoparticles prepared by a supramolecular approach are loaded with doxorubicin (DOX) for the cancer therapy. The nanoparticles contain disulfide bonds within the polymer network, allowing the release of the DOX payload in a reducing environment within the endoplasm of cancer cells. In addition, the loaded drug can also be released under acidic environment. In vitro anticancer studies using redox and pH dual responsive nanoparticles show excellent performance in inducing cell death and apoptosis. Zebrafish larvae treated with DOX-loaded nanoparticles exhibit an improved viability as compared with the cases treated with free DOX by the end of a 3 d treatment. Confocal imaging is utilized to provide the daily assessment of tumor size on zebrafish larva models treated with DOX-loaded nanoparticles, presenting sustainable reduction of tumor. This work demonstrates the development of functional nanoparticles with dual responsive properties for both in vitro and in vivo drug delivery in the cancer therapy.

Abstract:Herbal medicine is now globally accepted as a valid alternative system of pharmaceutical therapies. Various studies around the world have been initiated to develop scientific evidence-based herbal therapies. Recently, the therapeutic potential of medicinal plant derived miRNAs has attracted great attraction. MicroRNAs have been indicated as new bioactive ingredients in medicinal plants. (European mistletoe) has been widely used in folk medicine for the treatment of cancer and cardiovascular diseases. Our previous study has indicated the therapeutic potential of mistletoe miRNAs by using bioinformatics tools. To evaluate the stability of these miRNAs, various mistletoe extracts that mimic the clinical medicinal use as well as traditional folk medicinal use were prepared. The mistletoe miRNAs including miR166a-3p, miR159a, miR831-5p, val-miR218 and val-miR11 were quantified by stem-loop qRT-PCR. As a result, miRNAs were detectable in the majority of the extracts, indicating that consumption of medicinal plant preparations might introduce miRNAs into mammals. The factors that might cause miRNA degradation include ultrasonic treatment, extreme heat, especially RNase treatment, while to be associated with plant molecules (e.g., proteins, exosomes) might be an efficient way to protect miRNAs against degradation. Our study confirmed the stability of plant derived miRNAs during herb preparations, suggesting the possibility of functionally intact medicinal plant miRNAs in mammals.

Abstract:This study is about a morphoanatomical and histochemical analysis Symplocos tetrandra (Mart.) Mart. The leaves and stem were analyzed using light and scanning electron microscopy, and qualitative chemical analysis. The investigation revealed prismatic crystals, calcium oxalate druses and brachysclereids. These findings suggest potential anatomical markers for species identification and differentiation. The Symplocaceae family comprises 406 species, all of which are classified within a single genus, Symplocos Jacq., which is distributed across multiple continents. In Brazil, this genus is predominantly found in the Atlantic Forest and Cerrado regions and is characterized by the presence of secondary metabolites, such as iridoids, terpenoids, and saponins with biological activities. Moreover, the literature research has unveiled the potential of this genus for antimicrobial, antitumor, and anti‐HIV applications. This research conducted morphoanatomical, histochemical studies using light microscopy and scanning electron microscopy (SEM) analysis of the leaves and stems of Symplocos tetrandra (Mart.) Mart. Cross‐sections and paradermal were performed. The investigation revealed the presence of prismatic calcium oxalate crystal in petiole and druses calcium oxalate crystals, in petiole and leaf blade. Paracytic stomata on the abaxial surface, lignified pith in the stem, which was detected with the phloroglucinol staining. Oil bodies, uniseriate epidermis, and a thin lipidic cuticle were confirmed with Sudan III. These findings suggest potential anatomical markers for taxonomic classification of the species. The data obtained in this study contributes for species identification, and the presence of metabolites can guide future pharmacological investigation with S. tetrandra.

Abstract:This research investigates the morpho-anatomical characteristics of seven Baccharis species, namely B. articulata, B. milleflora, B. myriocephala, B. pentaptera, B. riograndensis, B. sagittalis and B. trimera. Commonly called carquejas, these species have aerial photosynthetic winged stems known as cladodes and are widely used traditionally to treat digestive and diuretic disorders. This study aimed to characterize these commonly misidentified species using morphological and microscopic techniques, including light and scanning electron microscopy coupled with energy-dispersive X-ray spectroscopy. Trichomes, the presence or absence of oil bodies, and a subepidermal collenchyma layer at the wing edge were identified as primary anatomical markers that can help differentiate the studied species.

Abstract:Six viscotoxins in pure form from Viscum album L. have been isolated and their toxicity compared by using a viscotoxin sensitive Yoshida cell line. Four of them are the known viscotoxins A2, A3, B and 1‐PS, whereas the other two appear to be new. The six viscotoxins were cytotoxic but varied in potency. Cytotoxicity of the extracts from the three European subspecies of V. album depended on the qualitative and quantitative composition of these viscotoxins. A good correlation of the cytotoxicity calculated on the basis of the content of viscotoxins present in the extracts with that measured by the Yoshida cell assay was observed. The viscotoxin sensitive Yoshida cell line is a suitable system for detecting the viscotoxin dependent cytotoxicity of mistletoe extracts

Abstract:Vachellia gummifera (Willd.) Kyal. & Boatwr. is a medicinal plant endemic to Morocco that has no documented studies on its chemical composition. In this study, the chemical composition of the water/methanol (4:1) extracts of air‐dried leaf and stem samples of Moroccan V. gummifera was determined using UHPLC‐MS and NMR. In total, over 100 metabolites were identified in our study. Pinitol was the major compound in both the leaf and stem extracts, being significantly more abundant in the former. Asparagine and 3‐hydroxyheteroendrin were the second most abundant compounds in the stem and leaf extracts, respectively, though both compounds were present in each tissue. The other compounds included flavonoids based on quercetin, and phenolic derivatives. Eucomic acid, only identified in the stems and was the major aromatic compound distinguishing the leaf and stem profiles. Quercetin 3‐O‐(6′′‐O‐malonyl)‐β‐D‐glucopyranoside was identified as the major flavonoid in the leaves but was also present in the stems. Other malonylated derivatives that were all flavonol glycosides based on myricetin, kaempferol, and isorhamnetin in addition to quercetin were also identified. This is the first report of eucomic acid and malonylated compounds in Vachellia species. This report provides valuable insights into the chemotaxonomic significance of the Vachellia genus.

Abstract:The total alkaloids were ultrasonic-assistedly extracted from Viscum coloratum (Kom.) Nakai., and were systematically separated into five constituent groups namely phenol weak alkaloids, non-phenol weak alkaloids, phenol tertiary amine alkaloids, non-phenol tertiary amine alkaloids, and lipid alkaloids. Then, their antimicrobial activities were assayed. The results showed that five mistletoe alkaloids groups all have antimicrobial on the E.coli, but no-obvious activity on the Staphylococcus aureus.

Abstract:Viscum continuum E. Mey. Ex Sprague (Mistletoe) is a semi-parasitic plant that grows on the branches of other trees with reported numerous biological activities. This study was aimed at isolating a compound/s that will be used as a standard reference for quality control of South African-based commercialized mistletoe products and to further perform antioxidant and antimicrobial tests on the isolated compound. A dried sample of mistletoe was ground and extracted successively with hexane, dichloromethane (DCM), acetone and methanol using a serial exhaustive cold maceration procedure. The compound was isolated using column chromatography, and its chemical structure was elucidated using two-dimensional nuclear magnetic resonance (2D NMR) and ultrahigh-performance liquid chromatography-mass spectrometry (UPLC-MS). The antioxidant activity of the compound was determined using DPPH, hydrogen radical scavenging activity and reducing power assays, whereas antimicrobial activity was assessed using the minimum inhibitory concentration (MIC) method. Subjection of the DCM extract to column chromatography resulted in the isolation of a compound elucidated as olean-12-en-28-butanol-1-one, 3-hydroxy-4,4,10, 14, 20-pentamethyl (D4). Both the DPPH, H2O2 radical scavenging activity and reducing power assays revealed a significant antioxidant potential of compound D4 with an IC50 of 0.701 mg/mL, lower than that of gallic acid (0.793 mg/mL) for the H2O2 radical scavenging assay. The results also indicated good antibacterial activity of D4 with an IC50 of 0.25 mg/mL, compared to ciprofloxacin with an IC50 of 0.0039 mg/mL, against two Gram-negative (Pseudomonas aeruginosa, Escherichia coli) and three Gram-positive (Streptococcus pyogenes, Bacillus cereus and Staphylococcus aureus) bacteria. This study is the first to report on the isolation of the olean-12-en-28-butanol derivative from mistletoe of the South African ecotype.
