Abstract:BACKGROUND The widespread use of chemical insecticides has resulted in the development of resistance in German cockroaches worldwide, and biopesticides based on entomopathogenic fungi as active ingredients have become a promising alternative strategy. Resistance can change many of the physiological and biochemical characteristics of insect pests, such as cuticle thickness, detoxification enzyme activity, and even intestinal flora composition. Thus, potential interactions between pathogenic fungi and insecticide resistance may lead to unpredictable changes in pest susceptibility to fungi. RESULTS Beta-cypermethrin-resistant German cockroaches were more susceptible to infection with the fungus Metarhizium anisopliae regardless of age and sex. Histopathological results showed that the infection of resistant strains (R) by M. anisopliae was visibly faster than that of susceptible strains (S). The gut microbiota of the S strain indicated a stronger ability to inhibit fungi in vitro. The abundance of Parabacteroides, Lachnoclostridium, and Tyzzerella_3 decreased significantly in the R strain, and most demonstrated the ability to regulate glucose and lipid metabolism, and antifungal infections. The expression levels of Akirin, BgTPS, and BgPo genes in the R strain were significantly lower than those in the S strain, while BgChi and CYP4G19 gene expression were significantly higher. The mortality of cockroaches infected with M. anisopliae decreased to varying degrees after RNA interference, reflecting the role of these genes in antifungal infection. CONCLUSIONS Results confirmed that insecticide resistance may enhance cockroach susceptibility to fungi by altering intestinal flora and gene expression. Fungal biopesticides have high utilization value in pest control and insecticide resistance management strategies.
Entomopathogenic fungi based microbial insecticides and their physiological and biochemical effects on Spodoptera frugiperda (J.E. Smith)

Abstract:Background: 'The fall armyworm, Spodoptera frugiperda', represents a significant threat to maize production, a major staple crop in Asian countries. Methods: In pursuit of more effective control of this insect pest, our study assessed the physiological and biochemical effects of the entomopathogenic fungus Metarhizium anisopliae against the larvae of S. frugiperda. Results: Results revealed that, following nine days of treatment, a high concentration of conidia (1.5x107 conidia/mL-1) was toxic to all stages of larvae (second to fifth instar), resulting in 97% mortality of the second instar, 89% mortality of the third instar, 77% mortality of the fourth instar, and 72% mortality of fifth instar. All larval instars were found to have dose-dependent mortality effects. Treated S. frugiperda larvae further displayed significant physiological, morphological, and behavioral changes. Here, treated larvae displayed significantly lower levels of acetylcholinesterase, α-carboxylesterase, and β-carboxylesterase enzyme activity when compared to control groups. Treated larvae underwent an outward morphological change as the result of a decrease in the exterior cuticle of the anal papillae and a demelanization of the interior cuticle. Treated larvae also exhibited abnormal feeding behaviors as a consequence of the negative impact of conidia treatment on the neuromuscular system. Investigation into the effect of M. anisopliae on the non-target organism, the earthworm Eudrilus eugeniae, revealed that M. anisopliae conidia did not produce significant pathogenicity following three days of treatment. Furthermore, histological analysis revealed no significant effect of the entomopathogenic fungi on the gut tissue of the non-target organism. Conclusion: This study highlights the potential of M. anisopliae in the control of S. frugiperda.

Abstract:This study examined the impact of Metarhizium anisopliae (Hypocreales: Clavicipitaceae) conidia on the eggs, larvae, pupae, and adults of Spodoptera frugiperda. The results showed that eggs, larvae, pupae, and adults exhibited mortality rates that were dependent on the dose. An increased amount of conidia (1.5 × 109 conidia/mL) was found to be toxic to larvae, pupae, and adults after 9 days of treatment, resulting in a 100% mortality rate in eggs, 98% in larvae, 76% in pupae, and 85% in adults. A study using earthworms as bioindicators found that after 3 days of exposure, M. anisopliae conidia did not cause any harmful effects on the earthworms. In contrast, the chemical treatment (positive control) resulted in 100% mortality at a concentration of 40 ppm. Histopathological studies showed that earthworm gut tissues treated with fungal conidia did not show significant differences compared with those of the negative control. The gut tissues of earthworms treated with monocrotophos exhibited significant damage, and notable differences were observed in the chemical treatment. The treatments with 70 and 100 μg/mL solutions of Eudrilus eugeniae epidermal mucus showed no fungal growth. An analysis of the enzymes at a biochemical level revealed a decrease in the levels of acetylcholinesterase, α‐carboxylesterase, and β‐carboxylesterase in S. frugiperda larvae after exposure to fungal conidia. This study found that M. anisopliae is effective against S. frugiperda, highlighting the potential of this entomopathogenic fungus in controlling this agricultural insect pest.

Abstract:Insect pests of agricultural crops have establish immunological tolerance against fungal infection caused by pathogens via different humoral and cellular processes. Fungal infection can be prevented by insect antioxidant and detoxifying enzymes, but there is no clear understanding of how they physiologically and biochemically interact. Our study aims to examine the antioxidant and detoxifying enzyme defense systems of the pest insect Spodoptera litura in response to infection by Metarhizium flavoviride. At 48 h following exposure to M. flavoviride, antioxidant enzyme levels were modified, and phenoloxidase and total hemocyte count were decreased significantly. The amount of detoxifying enzymes increased significantly. M. flavoviride appears to directly affect the S. litura immune system and results in decreased immunity. In a bioassay, M. flavoviride was found to be harmful to S. litura larvae in their third and fourth instar stage. M. flavoviride may be an effective tool in the control of S. litura larvae. Such entomopathogenic fungi represent cheaper, pollution free, target specific, promising alternatives to synthetic chemical tools in the for control insect pests.
Biocontrol effect of entomopathogenic fungi Metarhizium anisopliae ethyl acetate-derived chemical molecules: An eco-friendly anti-malarial drug and insecticide

Abstract:Insect pests represent a major threat to human health and agricultural production. With a current over-dependence on chemical insecticides in the control of insect pests, leading to increased chemical resistance in target organisms, as well as side effects on nontarget organisms, the wider environment, and human health, finding alternative solutions is paramount. The employment of entomopathogenic fungi is one such potential avenue in the pursuit of greener, more target-specific methods of insect pest control. To this end, the present study tested the chemical constituents of Metarhizium anisopliae fungi against the unicellular protozoan malaria parasite Plasmodium falciparum, the insect pests Anopheles stephensi Listen, Spodoptera litura Fabricius, and Tenebrio molitor Linnaeus, as well as the nontarget bioindicator species, Eudrilus eugeniae Kinberg. Fungal crude chemical molecules caused a noticeable anti-plasmodial effect against P. falciparum, with IC50 and IC90 values of 11.53 and 7.65 µg/mL, respectively. The crude chemical molecules caused significant larvicidal activity against insect pests, with LC50 and LC90 values of 49.228-71.846 µg/mL in A. stephensi, 32.542-76.510 µg/mL in S. litura, and 38.503-88.826 µg/mL in T. molitor at 24 h posttreatment. Based on the results of the nontarget bioassay, it was revealed that the fungal-derived crude extract exhibited no histopathological sublethal effects on the earthworm E. eugeniae. LC-MS analysis of M. anisopliae-derived crude metabolites revealed the presence of 10 chemical constituents. Of these chemicals, three major chemical constituents, namely, camphor (15.91%), caprolactam (13.27%), and monobutyl phthalate (19.65%), were highlighted for potential insecticidal and anti-malarial activity. The entomopathogenic fungal-derived crude extracts thus represent promising tools in the control of insect pests and malarial parasites.
Novel report on soil infection with Metarhizium rileyi against soil‐dwelling life stages of insect pests

Abstract:Entomopathogenic fungi are the most effective control remedy against a wide range of medical and agricultural important pests. The present study aimed to isolate, identify, and assess the virulence of Metarhizium rileyi against Spodoptera litura and Spodoptera frugiperda pupae under soil conditions. The biotechnological methods were used to identify the isolate as M. rileyi. The fungal conidial pathogenicity (2.0 × 107, 2.0 × 108, 2.0 × 109, 2.0 × 1010, and 2.0 × 1011 conidia/mL−1) was tested against prepupae of S. litura and S. frugiperda at 3, 6, 9, and 12 days after treatments. Additionally, the artificial soil‐conidial assay was performed on a nontarget species earthworm Eudrilus eugeniae, using M. rileyi conidia. The present results showed that the M. rileyi caused significant mortality rates in S. litura pupae (61‐90%), and S. litura pupae were more susceptible than S. frugiperda pupae (46%–73%) at 12 day posttreatment. The LC50 and LC90 of M. rileyi against S. litura, were 3.4 × 1014–9.9 × 1017 conidia/mL−1 and 6.6 × 105–4.6 × 1014 conidia/mL−1 in S. frugiperda, respectively. The conidia of M. rileyi did not exhibit any sublethal effect on the adult stage of E. eugeniae, and Artemia salina following a 12‐day treatment period. Moreover, in the histopathological evaluation no discernible harm was observed in the gut tissues of E. eugeniae, including the lumen and epithelial cells, as well as the muscles, setae, nucleus, mitochondria, and coelom. The present findings provide clear evidence that M. rileyi fungal conidia can be used as the foundation for the development of effective bio‐insecticides to combat the pupae of S. litura and S. frugiperda agricultural pests.

Fine specificity of antibodies produced in rhesus monkeys following in vivo treatment with anti‐T cell murine monoclonal antibodies

Abstract:The immune response of 23 rhesus monkeys against different murine monoclonal antibodies (mAb) administered in vivo as immunosuppressive agents has been analyzed. Seven mAb specific for either helper‐inducer (CD4 molecules) or cytotoxic/suppressor (CD8 molecules) T cells, that cross‐react with monkey lymphocytes, were administered i.v. for 10 consecutive days in rhesus monkeys. Nineteen of the animals were recipients of a skin or renal allotransplant. Nineteen out of the 23 monkeys developed a significant immune response against the injected monoclonal. This response was restricted in its specificity since unrelated murine monoclonals were not recognized by the monkeys' anti‐monoclonal immunoglobulins. Fine analysis of the monkeys' sera revealed that the antibodies produced against the xenogeneic proteins selectively exhibited two major specificities i.e., anti‐isotypic and anti‐idiotypic. On practical basis, these results suggest that an animal already immunized against a given mAb should still be sensitive to the therapeutic effect of another monoclonal sharing the same specificity but different idiotype.
Bio-Efficacy of Fusarium humuli and Fusarium incarnatum (Hypocreales: Nectriaceae) Against Larvae of Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) Under Controlled Conditions

Abstract:Spodoptera frugiperda has impaired the productivity of staple crops in Tanzania since it was first reported in 2017. The overutilization of synthetic insecticides as an immediate remedy has led to new ecological problems along effects on public health. Using entomopathogenic fungi (EPF) is perceived as a safer way, and therefore, Fusarium incarnatum (Vw3) and Fusarium humuli (Vw5) from the SUA germplasm collection were tested against S. frugiperda larvae at levelled concentrations; C1, C2 and C3 alongside 0.1% Triton X-100 and Banophos 720 EC (20 ml/20 L) as negative (C-) and positive (C+) controls, respectively. The findings showed C+ inflicted the highest death rates (84% and 97% for Vw3 and Vw5, respectively), followed by C1 (48.8% and 55.5% for Vw3 and Vw5, respectively) and C2 (22.2%, 28.8% for Vw3 and Vw5, respectively). Moreover, C3 inflicted mortalities by 11.1% and 12.2% for Vw3 and Vw5 respectively, while C- inflicted mortalities by 7.7% and 1.1% for Vw3 and Vw5, respectively. Significant differences were observed in concentrations and ages of the insects. However, more studies are recommended to validate their safety and compatibility with other IPM components prior to qualifying the EPF for the control of S. frugiperda.
Selection of Global Metarhizium Isolates for the Control of the Rice Pest Nilaparvata Lugens (homoptera : Delphacidae)

Abstract:BACKGROUND: This study was initiated to search for fungal candidates for microbial control of brown planthopper (BPH) Nilaparvata lugens Stal, to which little attention has been paid in the past two decades.RESULTS: Thirty-five isolates of Metarhizium anisopliae (Metschnikoff) Sorokin and M. flavoviride Gams & Rozsypal from different host insects worldwide were bioassayed for their lethal effects against third-instar BPH nymphs at 25 degrees C and a 14:10 h light:dark photoperiod at ca 1000 conidia mm(-2). On day 9 post-treatment, mortality attributable to mycosis ranged from 6.5 to 64.2% and differed significantly among the tested isolates with no apparent relationship to their host origin. Only two BPH-derived M. anisopliae isolates from the Philippines (ARSEF456) and Indonesia (ARSEF576) killed > 50% of the nymphs. Both isolates were further bioassayed for time-concentration-mortality responses of the nymphs to the sprays of 19-29, 118-164 and 978-1088 conidia mm(-2) in repeated bioassays. The resultant data fitted a time-concentration-mortality model very well. Their LC50 values were estimated as 731 and 1124 conidia mm(-2) on day 7 and fell to 284 and 306 conidia mm(-2), respectively, on day 10.CONCLUSION: The two M. anisopliae isolates are potential biocontrol agents of BPH for further research. This is the first report of the lethal effects of global Metarhizium isolates on the rice pest. (C) 2008 Society of Chemical Industry.

Abstract:Cydia pomonella L. is a highly destructive pest inflicting substantial damage on pome fruits globally. Resistance to chemical pesticides by C. pomonella poses a significant challenge to the effective management strategies. Utilising entomopathogenic fungi (EPF) for the management of insect pests has emerged as one of the promising and environmentally friendly alternatives. Against this backdrop, we conducted laboratory bioassays to assess the efficacy of three EPF, namely Beauveria bassiana (Bals.-Criv.) Vuill., Cordyceps fumosorosea (Wize) and Metarhizium anisopliae (Metschn.) Sorokīn against C. pomonella using three effective concentrations (1 × 10 4 , 1 × 10 5 , and 1 × 10 6 conidia/ml). Highest larval (98.62 ± 10.00%) and pupal (37.35 ± 2.15%) mortality was observed when treated with B. bassiana while lowest larval (51.55 ± 6.30%) and pupal (17.83 ± 7.28%) mortality was seen in C. fumosorosea. Maximum adult emergence (16.26 ± 3.24) was recorded in C. fumosorosea and the minimum (5.72 ± 0.09) in B. bassiana . LC 50 values of C. fumosorosea , M. anisopliae and B. bassiana were 5.482, 2.683 and 1.891 conidia/ml, respectively. All three tested EPF were effective against C. pomonella under controlled laboratory conditions.

Abstract:Anopheles stephensi (Diptera: Culicidae) is the vector of urban malaria in India and has a significant impact in transmitting infection in cities and towns. Further, WHO has also alarmed its invasive nature as a threat to African countries. Entomopathogenic fungi such as Beauveria bassiana and Metarhizium anisopliae have been found to be highly effective in controlling vector mosquito populations and therefore could be used in integrated vector control programs. Before employing the entomopathogenic fungi into the control programs, an effective isolate must be selected. Two separate experiments were conducted to evaluate the efficacy of Beauveria bassiana (Bb5a and Bb-NBAIR) and Metarhizium anisopliae (Ma4 and Ma-NBAIR) isolates against An. stephensi. Cement and mud panels were treated with fungal conidia with the concentration of 1 × 107 conidia/mL and adult An. stephensi mosquitoes were exposed to the treated panels (24 h after conidia were applied) by conducting WHO cone bioassay tests. The survival of the mosquitoes was monitored daily until the 10th day. In the second experiment, second instar larvae of An. stephensi were treated with fungal (Bb5a, Bb-NBAIR, Ma4 and Ma-NBAIR) conidia and blastospores with the spore concentration of 1 × 107 spores/mL. The survival of larvae was monitored until pupation. All the fungal isolates tested caused mortality in the adult mosquitoes, with varying median survival times. The Bb5a isolate reported lesser median survival times on both cement and mud panels (6 days). The treated mosquitoes showed similar survival rates for each fungal isolate irrespective of the panel type. There was no mortality in the treated larvae; however, a delay in larval development to pupae was observed compared with the untreated control larvae. Ma4-treated larvae took 11 days (95% CI = 10.7–11.2) to become pupae when compared with the untreated control larvae (6 days [95% CI = 5.6–6.3]). The findings of this study will be useful to consider EPF as a tool for the management of vector mosquitoes.

Abstract:Ten isolates of Beauveria bassiana (Bb), Metarhizium anisopliae (Ma) and Paecilomyces fumosoroseus (Pfr) with different host insect origins were assayed for their biocontrol potential against the female adults of carmine spider mite Tetranychus cinnabarinus using a novel bioassay system, which was featured with paired fava bean leaves supported upright by the petioles individually inserted into rhizocaline-inclusive agar in caged bottles and provided a nearly natural habitant for free activity of the highly mobile mite adults. Each pair of the leaves harboring 15 females (⩽2-day-old) was exposed to a spray of each isolate at the concentration of 21–78 (low), 119–308 (medium) or 740–1946 (high) conidia/mm2 plus a blank control (sprayed with 0.02% Tween-80), maintained in a top-meshed cage at 25°C and 12:12 L:D and observed daily for 9-day mortality records. Each of the bioassays was repeated 6–8 times. The three fungal concentrations resulted in the mite mortalities of 31.9–87.7% for B. bassiana, 24.2–80.3% for M. anisopliae and 19.4–77.7% for P. fumosoroseus, respectively. The LC50s of the tested isolates determined by the fitted time-concentration-mortality relationships declined over days after spray while their LT50s shortened as the concentration increased. The two isolates Bb734 and Bb2860 had the LC50s of 42–180 and 177–351 conidia/mm2 on days 7–9 and the LT50s of 5.8–6.9 and 3.6–8.4 days at 200–1500 conidia/mm2. These were followed by Ma456 with the LC50s of 179–925 conidia/mm2 on days 7–9 and the LT50s of 6.5–7.2 days at 750–1500 conidia/mm2. Other isolates had larger LC50s and longer LT50s, which represent intermediate or low virulence to the mite species. Thus, the mentioned three among the tested isolates are promising candidates for microbial control of the mite pest.

Abstract:The thromboxane A2 receptor antagonist, ramatroban (BAY u 3405), was orally administered for 4 weeks at a daily dose of 150 mg (b.i.d.) to 10 patients with perennial allergic rhinitis who had a positive reaction to house dust challenge on nasal mucosa. Nasal cavity volume and minimum cross-sectional area were measured, and changes in nasal mucosal swelling were determined following allergen challenge with house dust. The influence on nasal mucosal hemodynamics was also investigated. Nasal cavity volume and minimum cross-sectional area were measured by acoustic rhinometry, and blood flow in the nasal mucosa was measured by laser Doppler flowmetry. Percent changes in values from baseline nasal cavity volume were significantly decreased by allergen challenge before ramatroban administration, but no significant decrease was noted after ramatroban administration. Similarly, percent changes in values from baseline nasal cavity minimum cross-sectional area were significantly decreased by allergen challenge before administration of ramatroban, but not after administration. Percent changes in values from baseline nasal mucosal hemodynamics were significantly increased by allergen challenge both before and after ramatroban administration, which thus had no effect on mucosal hemodynamics. These findings suggest that ramatroban might inhibit the increase in nasal mucosal swelling but has no effect on nasal mucosal hemodynamics.

Abstract:Abstract Background The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a noxious pest of cruciferous crops all over the world causing serious economic damage. Management of insect pest generally depends on chemical control; however, due to development of resistance against all types of insecticides, alternative approaches especially utilization of a microbial agent is inevitable. Results Potential of 2 entomopathogenic fungi (EPF), viz., Beauveria bassiana and Metarhizium anisopliae , was evaluated against 2 nd and 3 rd larval instars of P. xylostella by adopting leaf dip and direct spraying methods under laboratory conditions. Significant mortality rate was achieved by each fungus under adopted methodologies. However, B. bassiana was found to be more effective in both conditions than M. anisopliae . Highest mean corrected mortality (77.80%) was recorded, when spores of B. bassiana were sprayed on the 2 nd instar larvae (LC 50 =1.78×10 4 /ml) after the 6th day of treatment. Similarly, incase of M. anisopliae LC 50 for the 2 nd instar at the same methodology was 2.78×10 4 /ml with a mortality percentage of 70.0%. Offspring sex ratio was non-significantly related to treatment concentration and methodology, except for the control. Conclusion Beauveria bassiana and M. anisopliae had potential to suppress P. xylostella infestations when applied appropriately. Present findings suggested that B. bassiana and M. anisopliae when sprayed on immatures of host insect had more effect as compared to leaf dip procedure. Furthermore, no significant effect of concentrations was observed on sex ratio.

Abstract:November 2007 393 The last column in this series on antipsychotics focused on the early history of antipsychotics, from reserpine and chlorpromazine to haloperidol.1 This column returns to the issue of relative binding affinity, which was first discussed with reference to antidepressants in a column almost a decade ago.2 The focus of this column will be antipsychotics, using quetiapine and ziprasidone as examples to illustrate the clinical relevance of relative receptor binding affinity. This discussion will set the stage for subsequent columns that will discuss the concept of “atypical” antipsychotics. The regular reader will recognize Equations 1 and 2 (see next page), which are hallmarks of this column. Equation 1 shows that the effect of any drug is a function of three variables: 1) its affinity for and intrinsic activity at one or more sites of actions (almost always a regulatory protein), 2) the concentration achieved at that (or those) site(s) of action, and 3) the specific biology of the patient being treated. Equation 2 specifies that the concentration achieved at that (or those) site(s) of action is in turn determined by the dosing rate going into the patient divided by the ability of that specific patient to clear the drug from his or her body.
Delving into the Causes and Effects of Entomopathogenic Endophytic Metarhizium brunneum Foliar Application-Related Mortality in Spodoptera littoralis Larvae

Abstract:The aim of the current study was to delve into the causes of mortality of Spodoptera littoralis larvae feeding on Metarhizium-colonized plants in the absence of fungal outgrowth on the cadavers as previous studies reported and to elucidate the possible indirect effects of this fungus-colonized diet. The effect was evaluated in experiments conducted using leaf discs of colonized plants and in planta using fungus-colonized whole plants. The mortality rates of larvae fed on Metarhizium-colonized melon leaves were 45.0% and 87.5%, and the average survival times were 6.6 and 3.1 days in experiments performed with discs and in planta, respectively. Notably, these mortality levels were not associated with observed apoptosis mediated by caspases 1, 3-7 and 8; thus, further investigation into the possible immune system reaction of the insect after the ingestion of colonized plants is required. The leaf consumption of S. littoralis larvae fed on melon-colonized leaves was lower than that on control plants in the disc experiments but not in experiments conducted in planta. In this regard, in experiments performed in planta, plant damage increased larval mortality in both fungally challenged and control larvae. There was also a meaningful effect of exposure to Metarhizium-colonized melon leaf discs on S. littoralis fitness, with significant reductions in 39.0% and 22.0% in female fecundity and egg fertility, respectively, detected in females emerging from pupae developing from larvae surviving exposure to colonized plant discs; all larvae died in the in planta experiments. Hence, the present work presents new findings revealing the high potential of endophytic entomopathogenic fungi to improve the outcome of foliar applications against chewing insects in the short, mid- and long term, by the reduction of the reproductive potential of surviving adults and reveals new insights into the development of bioassays with whole plants for more detailed evaluation of the impact of these fungi as endophytes used for plant protection.
Evaluation of key parameters for developing a Metarhizium rileyi-based biopesticide against Spodoptera frugiperda (Lepidoptera: Noctuidae) in maize: laboratory, greenhouse, and field trials

Abstract:Background: The entomopathogenic fungus Metarhizium rileyi is known to cause natural infections in some caterpillars. In this multiyear study, we carried out laboratory, greenhouse and three field trials with the aim of developing a sustainable option for control of the fall armyworm (FAW) in maize. Association of M. rileyi with Spodoptera frugiperda baculovirus (SfMNPV) and delivery strategies were also investigated. Results: The selected isolate (CG381) was effective in the laboratory at a low concentration (~ 4 × 103 conidia cm-2 ), killing >95% of FAW larvae within 8 days. In the greenhouse assay, applications of conidia suspended in water or as dry powder in maize whorls (~ 6.3 × 106 conidia per plant) produced similar larval mortalities (88%-96%). In the field trials, conventional spraying of unformulated conidia (0.6 and 1.2 × 1012 conidia ha-1 ) caused low larval mortalities (27-31%). Simultaneous application of either unformulated or oil-based formulations of M. rileyi conidia and S. frugiperda baculovirus (SfMNPV) to plant rows caused larval mortalities comparable with each of the pathogens applied alone. However, when a formulation containing both pathogens was sprayed directly into the whorls, the overall mortality of S. frugiperda larvae due to pathogens (mostly to M. rileyi infections) reached ~ 59%, twofold higher than the other treatments with conventional spraying, whereas in the control, the overall mortality was only 1%. Conclusion: Optimizing exposure of S. frugiperda larvae to an inoculum of virulent entomopathogens through directed applications to maize whorls is critical to produce satisfactory mortality levels and is promising for integrated pest management.
Entomopathogenic bacteria and fungi as eco-safe substitutes to chemical insecticides against leaf eating caterpillars of Spodoptera litura

Abstract:The present study was carried out in laboratory conditions (25 ± 10°C and 70% humidity) for studying the bioefficacy of entomopathogenic fungi and Bacillus thuringiensis against Spodoptera litura using the leaf dip method. The insecticidal attributes of three different commercial bioinsecticides (Bacillus thuringiensis 0.5% W.P., Beauveria bassiana 1.0% W.P., and Metarhizium anisopliae 1.0% W.P.) at different concentrations (0.5, 1.0, and 1.5%) were evaluated against the 3rd and 4th larval instars of S. litura. The analysis of variance results revealed that the highest concentration (1.5% concentration) of B. thuringiensis against the 3rd instar larvae of S. litura caused a maximum 80 ± 0.28% larval mortality, followed by B. bassiana and M. anisopliae, which caused 72 ± 0.21 and 64 ± 0.20% larval mortality after 120 h exposure. The calculated median lethal concentration in probit analysis demonstrated the lowest median lethal concentration of 1.22/10 larvae (95% fiducial limit 4.71–11.45) in the 3rd instar larvae after 72 h. Similarly, in 4th instar larvae of S. litura, the highest concentration (1.5% concentration) of B. thuringiensis caused maximum 76 ± 0.23% larval mortality, followed by B. bassiana and M. anisopliae, causing 70 ± 0.21% and 64 ± 0.20% larval mortality after 120 h. The calculated median lethal concentration in probit analysis demonstrated the lowest LC50 = 1.59/10 larvae (95% FL: 1.08–2.36) in the 4th instar larvae after 72 h. Based on these studies, it was revealed that B. thuringiensis and B. bassiana showed maximum efficacy against S. litura larvae. Although M. anisopliae also exhibits insecticidal properties, all of these bioinsecticides can be used to manage the population of S. litura. These bioinsecticides can be included in integrated pest management programs as they do not pose any toxic hazards to the environment.

Abstract:The use of entomopathogenic fungi (EPF) like Beauveria, Metarhizium, Lecanicillium , and Isaria is upsurging in recent years for the management of crop insect pests. EPF are considered better than synthetic insecticides as they are safe for humans, sustainable to the environment, and target-specific in nature. Many of these EPF are pathogenic to economically important insect pests and thus are capable of controlling them. They are cheaper in long run, show lesser residual effects, and are able to overcome the problem of resistance. EPF degrades the host cuticle and proliferates in hemolymph as hyphal bodies, secreting the toxins responsible for the death of host insects. The later saprophytic growth leads to the production of fungal spores capable of reinfecting other hosts. Different commercial formulations of EPF are available globally such as liquid formulation, wettable powder, suspensible granules, and so on. These available formulations under different trade names can be used for several crops and pests at the recommended dosage to obtain optimum results. The storage conditions should be maintained to retain the viability of EPF. Modern biotechnological interventions could be vital in enhancing the efficacy of these entomopathogens by manipulating their traits. Specialized researches are necessary to understand the interaction between EPF, host insects, crops, and their environment in order to explore the best formulation of mycoinsecticides. This review explores the overview of EPF, its mode of action, significance, commercial formulations, future prospects, and the summary of recent findings. Readers could realize the essence of EPF in sustainable agriculture through this review.
