Abstract:Out-of-season breeding is an effective method for addressing seasonal shortages of fry in aquaculture species such as largemouth bass (LMB) for year-round production. Off-season breeding of LMB can be achieved by subjecting breeding LMB to prolonged low-temperature conditions; however, this can alter reproductive rhythms, affecting the quality of their sperm and leading to a decrease in reproductive efficiency. Therefore, it is crucial to investigate issues such as the damage to the testes and the related mechanisms caused by low-temperature stress during out-of-season breeding. In this experiment, we assessed the changes in the testes during this time in LMB by comparing reproductive rhythms, testicular histomorphology, ultrastructure, antioxidant capacity and apoptosis. We synthesized measurements of LMB from three identically treated cement ponds and fish exposed to water temperatures of 13–16 °C to assess the changes in the testes. The results showed that (1) out-of-season reproduction delayed sperm production and promoted sperm redevelopment in LMB, various hormone levels have changed over time (e.g., LH, FSH, and T). (2) The head plasma membrane of LMB spermatozoa was separated, and the middle mitochondria were swollen. (3) The expression levels of antioxidant enzymes (cat, sod, and gpx) were upregulated, and oxidative stress occurred in LMB. (4) The expression levels of apoptosis genes (e.g., bax, bcl2, and caspase3) were upregulated, and apoptosis occurred in LMB due to off-season breeding. Moreover, important genes of the mitochondrial apoptosis pathway (bid, CYT-C) were upregulated, indicating that spermatozoan apoptosis in LMB was probably achieved through the mitochondrial apoptosis pathway. These results suggest the delays, damage, and regeneration of LMB testes. Our findings provide new insights into the molecular mechanisms that trigger changes in sperm quality during out-of-season breeding in fish.

Effects of Different Light Spectra on the Growth Performance and Survival of Largemouth Bass (micropterus Salmoides) Larvae in Recirculating Aquaculture Systems

Abstract:The effects of light-emitting diode (LED) spectra on the survival rate and growth performance of largemouth bass in the nursery stage were explored in this study. By means of recirculating aquaculture systems, largemouth bass larvae (initial body length of 6.5 +/- 0.23 mm) were reared under four different light spectra: blue (half-peak bandwidth = 429 to 450 nm), green (half-peak bandwidth = 503 to 535 nm), white (320 < lambda < 950), and full spectrum. After the 16-day experiment, the results showed that the survival rate was highest for the larvae in the full spectrum group (33.5% +/- 3.5%) and lowest for the larvae in the blue light group (11.6% +/- 0.7%). The final body lengths of the larvae under different light spectra was as follows: the full spectrum group had the longest average body length (12.4 +/- 0.3 mm), followed by the white light group and the green light group, and the larvae in the blue light group had the shortest average body length (12.1 +/- 0.4 mm). The dorsal and anal fins of the larvae differentiated fastest in the full spectrum group, and the caudal fins differentiated fastest in the green light group. The maximal and minimal malformation rates of the larvae occurred under white light (26.7% +/- 0.2%) and full spectrum light (16.7% +/- 0.9%), respectively. The results demonstrated that a full spectrum light environment can improve the survival of largemouth bass larvae and promote their growth and development.
Effects of Stocking Density During Simulated Transport on Physiology and Behavior of Largemouth Bass (micropterus Salmoides)

Abstract:Abstract The live transport of farmed fish is an important practice in Chinese aquaculture due to consumer preferences in its domestic market. However, live transport can be stressful for fish and may cause many welfare issues. This study aimed to examine the effects of transport density on the welfare of largemouth bass (Micropterus salmoides). Adult fish were allocated to four experimental treatments in a two‐factor design (N = 5 groups per treatment). Factor one was stocking density of either 39 (low density) or 78 (high density) g L−1. Factor two was transport, with half of the fish groups exposed to simulated transport stress for 3 h using a motion platform, and the other half kept in static tanks for the same length of time. Baseline behavior and physiology were determined from a subset of fish after acclimation but before selection into groups and application of transport. To determine the effects of transport, stocking density, and their interaction on the experimental groups, physiological sampling and behavioral recording were carried out immediately after simulated transport, and water quality was measured both before and after transport. Changes in water quality were observed between two timepoints, with decreased dissolved oxygen levels and increased water temperature. Total ammonia nitrogen levels significantly increased in all treatments over time, particularly in the high‐density groups. Fish ventilation rates were significantly elevated in the groups kept at a high stocking density regardless of whether they also experienced transport stress. Fish transported at a high stocking density also showed more fear‐related behaviors such as freezing and thigmotaxis than non‐transported groups. Serum cortisol, catalase, and aspartate transaminase levels were significantly higher in the transported treatment groups compared to the baseline fish, but no difference was found for serum glucose, lactate, and glutathione peroxidase. We concluded that 3 h of simulated transport was stressful to largemouth bass kept at 78 g L−1, as evidenced by altered fish physiological and behavioral indicators of stress, as well as deteriorated water quality. Additionally, we identified a positive correlation between serum and skin mucus samples for cortisol, glucose, and lactate in transported fish, providing important information for developing less invasive methods for the welfare assessment of largemouth bass.

Abstract:The feeding method is of great significance for aquaculture production and cost. In recent years, research on intelligent feeding, a method based on fish appetite, has been trending. Few studies, however, have focused on fish welfare issues that can result from intelligent feeding. In this study, an adaptive feeder based on a practical intelligent feeding method was designed to evaluate whether this intelligent feeding method would impair fish welfare compared to traditional automatic feeding. The results indicated that the amount of residual feed and size inhomogeneity in the traditional group was significantly higher than that in the intelligent group. The results of the growth indicator showed that the weight gain rate (WGR) in the intelligent feeding group was significantly higher (30.17%) than in the traditional feeding group. Although no significant differences were observed in the survival rate (SR), the condition factor (CF) and the hepatosomatic index (HSI), the specific growth rate (SGR) in the intelligent feeding group was significantly increased by 8.33% while the feed conversion rate (FCR) was reduced by a remarkable 17.07% compared to the traditional feeding group. Moreover, intelligent feeding significantly improved the pepsin activity of the bass. In terms of immunity and antioxidant capacity, however, the fish in the intelligent feeding group had a significantly lower lysozyme (LZM) level than those fed with the traditional method. For superoxide dismutase (SOD), the intelligent group also displayed a lower activity value, although it was not significant. The intelligent feeding group had 0.14 nmol/mg protein higher malondialdehyde (MDA) activity than the traditional group. Regarding stress, although no statistical significance was observed, the cortisol level in the intelligent group was 1.9 ng/ml higher than in the traditional group. Together, these data suggested that the intelligent feeding method can significantly improve fish growth but may also result in stress and suppress innate immunity.

Abstract:Largemouth bass (LMB) production exceeded 0.7 million tons in 2021 and has become one of the most important freshwater aquaculture species in China. The stable and fixed culture cycle led to regular and drastic price fluctuation during the past decade. Strong price fluctuation provides opportunities and challenges for the LMB industry, and out-of-season spawning (OSS) and culture will provide technical support for the opportunities. To induce OSS at a low cost, we established a controllable recirculating system that allows precise thermo-photoperiod manipulation. In the system, four experimental groups were assigned, 18NP (18°C overwintering water temperature, natural photoperiod), 18CP (18°C overwintering water temperature, controlled photoperiod), 16CP (16°C overwintering water temperature, controlled photoperiod), and NTNP (natural water temperature and natural photoperiod), to determine the effects of chilling temperature and photoperiod on spawning performance. OSS was observed in all the experimental groups without significant differences, except NTNP. The manipulated broodstock can re-spawn 3 months later in the next spring in advance. Further analysis of the volume percentage of different stages of oocytes provides a base for excellent regression between the volume percentage of the primary growth stage, cortical alveoli stage, vitellogenesis/maturation stage, and gonadal development/maturation. The results suggest that the volume percentage of oocytes is a better indicator of gonadal development and maturation than the gonadosomatic index. We also found that LMB prefers palm fiber as a spawning nest over gravel. The findings of this work provide important technique guidance for practical OSS of the LMB aquaculture industry and standardization of ovary development and maturation in fish with asynchronous developmental oocytes.

Abstract:Largemouth bass (LMB) is one of the two fastest-growing fish species and viewed as one of the five most successful alien species in Chinese aquaculture. Global aquaculture production of LMB reached 0.458 million tons in 2017 and China produced 99.8% of the total amount. Currently, imbalanced regional production, strong price fluctuation, and universal poor seed quality are three main stumbling blocks for steady and healthy aquaculture industry development of LMB. In the present work, we have analyzed interannual and seasonal price volatility between 2015 through 2018 and the production mode of LMB and concluded that off-season spawning is one of the best solutions for these issues. We have established an off-season spawning regimen for LMB through a single manipulation of temperature, which is suitable for large-scale production of LMB fry in the autumn (September and October). We also found as a species displaying a mixed-breeding (capital breeding and income breeding) strategy, LMB is able to acclimate itself rapidly to compressed reproductive cycle using different energy allocation strategies. Our results combined with previous reports suggest photoperiod is more important for the process of oogenesis in LMB, while the temperature is more critical for vitellogenesis and the final stage. The off-season spawning technique allows us to produce LMB fry twice in a year using the same broodstock population by single temperature manipulation.

Functional Properties of Protein Hydrolysates on Growth, Digestive Enzyme Activities, Protein Metabolism, and Intestinal Health of Larval Largemouth Bass (<i>Micropterus salmoides</i>)

Abstract:The present study was conducted to investigate the effects of dietary inclusion of protein hydrolysates on growth performance, digestive enzyme activities, protein metabolism, and intestinal health in larval largemouth bass (Micropterus salmoides). The experimental feeding trial presented in this study was based on five isonitrogenous and isolipidic diets formulated with graded inclusion levels of protein hydrolysates, and it showed that protein hydrolysates improved growth performance, reduced larval deformity rate, and increased the activity of digestive enzymes, including pepsin and trypsin. Gene expression results revealed that the supplementation of protein hydrolysates upregulated the expression of intestinal amino acid transporters LAT2 and peptide transporter 2 (PepT2), as well as the amino acid transporters LAT1 in muscle. Dietary provision of protein hydrolysates activated the target of rapamycin (TOR) pathway including the up-regulation of TOR and AKT1, and down-regulation of 4EBP1. Additionally, the expression of genes involved in the amino acids response (AAR) pathway, ATF4 and REDD1, were inhibited. Protein hydrolysates inhibited the transcription of some pro-inflammatory cytokines, including IL-8 and 5-LOX, but promoted the expression of anti-inflammatory cytokines TGF-beta and IL-10. The 16S rRNA analysis, using V3-V4 region, indicated that dietary protein hydrolysates supplementation reduced the diversity of the intestine microbial community, increased the enrichment of Plesiomonas and reduced the enrichment of Staphylococcus at the genus level. In summary, protein hydrolysates have been shown to be an active and useful supplement to positively complement other protein sources in the diets for largemouth bass larvae, and this study provided novel insights on the beneficial roles and possible mechanisms of action of dietary protein hydrolysates in improving the overall performance of fish larvae.

Abstract:Abstract Background: The intestine is the first line of defense against bacteria, toxins and antigens presents in the lumen of the instine. Aeromonas veronii is an opportunistic pathogen that has been related to mortality in fish. However, the protective mechanisms of intestinal damage caused by Aeromonas veronii are poorly defined. Thus, the objetive of this study was to evaluate the influence of dietary valine (Val) on growth, antioxidant capacity, and endoplasmic reticulum stress-mediated apoptosis in intestines of the juvenile largemouth bass Micropterus punctulatus after infection with Aeromonas veronii (A. veronii). Methods: A total of 720 fish (37.99 ± 0.01 g) were randomly allocated to six groups with three replicates each group, with different levels of Val (11.0, 14.0, 17.0, 20.1, 23.1, and 26.1 g/kg diet, respectively) for 77 d, and then challenged with A. veronii. Results: In the present study, we demonstrated that dietary Val: (1) increased the percent weight gain (PWG), specific growth rate (SGR), feed intake (FI), feed efficiency (FE), protein efficiency ratio (PER), protein production value (PPV), lipid production value (LPV), and ash production value (APV) (P< 0.05); (2) improved survival rate (SR) and alleviated intestinal injury; (3) reduced the levels of reactive oxygen species (ROS), malondialdehyde (MDA), and protein carbonyl (PC) (P< 0.05), and improved the intestinal antioxidant capacity by enhancing the mRNA levels of Cu/Zn superoxide dismutase (SOD1), catalase (CAT), glutathione-S-transferase (GST), glutathione peroxidase (GPx), glutathione reductase (GR), and γ-glutamyl cysteine ligase catalytic subunit (GCLC) (P< 0.05); (4) increased nuclear NFE2-related factor 2 (Nrf2) mRNA and protein levels (P< 0.05); (5) reduced intestinal DNA fragments, and down-regulated the mRNA levels of 78 KD glucose regulated protein (GRP78), activating transcription factor 6 (ATF6), inositol requiring enzyme-1 (IRE1), protein kinase RNA-dependent-like endoplasmic reticulum kinase (PERK), eukaryotic initiation factor 2α (eIF2α), activating transcription factor 4 (ATF4), C/EBP homologous protein (CHOP), cysteine-aspartic protease 9 (caspase 9), and cysteine-aspartic protease 3 (caspase 3), while up-regulating that of B-cell lymphoma-2 (Bcl2) (P< 0.05); (6) decreased phosphorylation of PERK (p-PERK) and phosphorylation of eIF2α (p-eIF2α), caspase 9, and caspase 3 protein levels. Conclusion: Dietary optimal Val level enhanced the growth performance and the intestinal antioxidant capacity via the Nrf2 signaling pathway after infection with A. veronii. Furthermore, this study is the first to demonstrate the role of Val in reducing ERS-mediated apoptosis through modulation of the PERK/eIF2α signaling pathway. Finally, Quadratic regression analysis of the SGR indicated that the dietary Val requirement of largemouth bass (37.99-162.92 g) was 21.50 g/kg diet, corresponding to 46.64 g/kg protein.
Optimal conditions for cryopreservation by vitrification of largemouth bass (Micropterus salmoides) embryos

Abstract:The largemouth bass (Micropterus salmoides) is one of the important freshwater aquaculture species in the world. However, due to limitations on introduction scale, high-density farming, inbreeding, and species hybridization, the germplasm resources of largemouth bass face threats such as degradation and susceptibility to diseases. Therefore, it is urgent to conduct research on the conservation of its original and good germplasm resources. We optimized the conditions of cryopreservation to vitrify and revive largemouth bass embryos, including the mixing ratio of cryoprotectants, embryo stage, equilibration step and temperature, and washing regent. The results showed that the least toxic single, binary, and ternary mixed permeating cryoprotectants were PG, PM (PG: MeOH = 2:1), and PMD (PM: DMSO = 3:1), respectively. The least toxic non-permeating cryoprotectant was 5 % glucose. The optimal vitrification solution selected was PMDG (30 % PMD + 5 % glucose) with an 80.67 % survival rate of embryos. Embryos at the heartbeat stage exhibited strong tolerance to the PMDG solution, which is the optimal embryo stage for cryopreservation. During the equilibration process, either the five-step equilibration method or pre-cooling the cryoprotectant to 4°C could reduce its toxicity. During the washing process, a 0.125 mol·L-1 sucrose solution yielded the best results. Based on the optimized conditions, 650 embryos at the heartbeat stage were subjected to cryopreservation by vitrification, resulting in a total of 350 intact transparent eggs, two of which hatched successfully. The results provide a reference for further improving the efficiency of cryopreservation by vitrification of largemouth bass and other fish species.
[School bullying and its association with health and lifestyle among schoolchildren].

Abstract:UNLABELLED The purpose of the study was to assess the association of bullying and being bullied with indicators of subjective health and problem behaviors among schoolchildren aged 11-15 years. MATERIAL AND METHODS The data of the anonymous survey of 5645 filth-, seventh-, and ninth-grade students (aged 11, 13, and 15 years, respectively), conducted in the spring of 2002, were analyzed. The students completed the World Health Organization's Health Behavior in School-aged Children questionnaire that included self-report of involvement in bullying and being bullied by others as well as subjective health and well-being estimates, health complains (headache, stomachache, back pain, anxiety, etc.), and problem behaviors (smoking, alcohol and drug use, suicidal ideation). The response rate was 95%. RESULTS More than half (52.3%) of students were involved in bullying process at least two times per month: 17.9% were involved as bullies ("victims"), 18.3% were bullied ("aggressors"), and 16.1% bullied others and were bullied themselves. A significant association between experiencing bullying and adverse health outcomes was found. Perpetrating bullying increased the odds of smoking, alcohol and drug use. Perpetrating and/or experiencing bullying increased the risk of high suicidal ideation that had cumulative effect. The established associations varied between genders and groups of students defined as "victims," "aggressors," and "victims/aggressors." CONCLUSIONS In Lithuania, school bullying is extremely prevalent and is associated with health disorders, poorer well-being, and problem behavior of schoolchildren. Urgent antibullying efforts, including both research and preventive measures, are needed in order to deal with this social phenomenon.
Earlier Activation of Interferon and Pro-Inflammatory Response Is Beneficial to Largemouth Bass (Micropterus salmoides) against Rhabdovirus Infection

Abstract:In order to understand the immune response of largemouth bass against Micropterus salmoides Rhabdovirus (MSRV), assisting disease resistance breeding, three largemouth bass breeding varieties Micropterus salmoides “Youlu No 3” (U3), “Youlu No 1” (U1) and “Zhelu No 1” (P1) were challenged intraperitoneally with MSRV. Serum and tissues were sampled to study the changes in non-specific immune parameters, viral loads, and transcript levels of immune-related genes, and the cumulative mortality rate was recorded daily for 14 days. The results showed that the cumulative mortality rates in the U1, P1, and U3 groups were 6.66% ± 2.89%, 3.33% ± 2.89%, and 0, respectively. The higher mortality may attribute to the increased viral loads after infection in the liver (2.79 × 105 and 2.38 × 105 vs. 1.3 × 104 copies/mg), spleen (2.14 × 105 and 9.40 × 104 vs. 4.21 × 103 copies/mg), and kidney (3.59 × 104 and 8.40 × 103 vs. 2.42 × 103 copies/mg) in the U1 and P1 groups compared to the U3 group. The serum non-specific immune parameters (lysozyme, catalase, and acid phosphatase) were found to be increased significantly in the U3 group. In addition, the transcripts of interferon-related genes (IFN-γ, IRF3, and IRF7) and pro-inflammatory-related genes (TNF-α and IL-1β) exhibited up-regulation and peaked at 6 h post infection in the U3 group, which also exhibited up-regulation but peaked at 12–24 h post infection in the U1 and P1 groups. In conclusion, these findings indicate that earlier activation of interferon and pro-inflammatory response is beneficial to largemouth bass against MSRV infection. This experiment may provide an insight into understanding the immune mechanism of largemouth bass against MSRV infection and contributes to molecular-assisted selection.

Abstract:Omega‐3 long‐chain polyunsaturated fatty acid (n‐3 LC‐PUFA) increases in aquatic products contributes to improving meat quality, thereby positively impacting human health. Different from marine fish which primarily obtain n‐3 LC‐PUFAs directly from zooplankton and algae, freshwater fish mainly utilize dietary linolenic acid (ALA) as a substrate to synthesize n‐3 LC‐PUFAs. Our team has successfully created a transgenic rapeseed oil (TRO) with high ALA content. Therefore, we here assessed the impacts of four different diets (LR, low‐fat rapeseed oil (RO) diet; HR, high‐fat RO diet; LTR, low‐fat TRO diet; HTR, high‐fat TRO diet) on growth performance, lipid accumulation, fatty acid composition, antioxidant capacity, immunity and serum biochemical indexes of juvenile largemouth bass (Micropterus salmoides), an economically valuable freshwater fish. The results showed no significant difference in survival rate among the four dietary groups. No significant differences in body weight gain and final weight were found between the LR and LTR groups, as well as between HR and HTR groups. No matter if it was a high‐fat or low‐fat diet, compared with the RO diet, TRO diets significantly increased the content of n‐3 LC‐PUFA, improved meat quality, effectively alleviated lipid accumulation in livers and muscles of juvenile largemouth bass. In addition, using high‐fat diets, TRO diet improved the antioxidant capacity and immune ability of juvenile largemouth bass, thereby promoting the overall health of fish. This study provides novel insights for fish feed formulation optimization from the perspective of genetically modified feed ingredients, and high‐quality aquatic products for human consumption.

Abstract:An 8-week feeding trial was conducted to investigate the effects of high-starch diets and the supplementation of an olive extract (OE) on the growth performance, liver health and lipid metabolism of largemouth bass (Micropterus salmoides). Four isonitrogenous and isolipidic diets were prepared: two basal diets containing low (9.0%) and high (14.4%) levels of starch (named as LS and HS), and 0.125% OE was supplemented to each basal diet (named LSOE and HSOE). The results show that high-starch diets had significant negative effects on growth performance, with lower FR, SGR and higher FCR, whereas OE significantly lowered FCR, determined by two-way ANOVA analysis. High-starch diets induced oxidative stress, inflammatory response and liver function injury, with significant increases in the content of plasmatic AKP, AST, ALT, hepatic SOD and MDA, and up-regulation of hepatic TNFα, IL1β, and TGFβ1 gene expression. In addition, a high-starch diet decreased the phosphorylation of AMPK and upregulated the expression of SREBP, together with higher hepatic liver lipid and HSI. The oxidative stress and lipid metabolism disorders indicate metabolic liver disease (MLD) of largemouth bass fed high-starch diets. Feeding on OE-supplemented diets increased the hepatic antioxidant capacity by decreasing the content of MDA and SOD. Fish fed the HSOE diet had an activated phosphorylation of JNK and decreased expression of pro-inflammatory IL1β compared with those fed the HS diet, which strongly indicated that the degree of inflammatory responses was reduced after OE supplementation. Interestingly, this study demonstrated that OE regulates hepatic lipid metabolism in fish by inhibiting the expression of hepatic lipogenesis genes (ACC1 and FASN) and promoting lipolysis (ATGL) and β-oxidation (CPT1α) to prevent TG accumulation. In conclusion, high-starch feed induced oxidative stress and lipid metabolic disorder of largemouth bass, while supplementation with OE improved its antioxidant capacity, anti-inflammatory responses and lipid metabolism. However, hepatic histopathological results suggested that OE supplementation could not completely repair the MLD caused by the high level of starch in largemouth bass.

Abstract:The aim of this study was to investigate the effects of sodium butyrate (SB) supplementation on growth performance, antioxidant enzyme activities, inflammatory factors, and hypoxic stress in largemouth bass ( Micropterus salmoides ). Diets were supplemented with different doses of SB at 0 (SB0), 0.5 (SB1), 1.0 (SB2) and 2.0 (SB3) g/kg. The hypoxic stress experiment was performed after 56 days of culture. The results showed that compared with the SB0 group, the final body weight, weight gain rate and protein deposition rate of the SB3 group were significantly increased ( P <0.05), while FCR was significantly decreased ( P <0.05). The contents of dry matter, crude lipids, and ash in the SB2 group were significantly higher than those in the SB0 group (P<0.05). The urea level was significantly decreased ( P <0.05), and the glucose content was significantly increased ( P <0.05) in the SB supplement group. Compared with the SB0 group, the SB2 group had significant reductions in the levels of serum triglyceride, cholesterol, elevated-density lipoprotein cholesterol, and low-density lipoprotein ( P <0.05), and significant reductions in the levels of liver alkaline phosphatase and malondialdehyde ( P <0.05). The total antioxidant capacity of the SB1 group was higher than that of other groups ( P <0.05). Compared with the SB0 group, the mRNA expression of TLR 22, MyD 88, TGF-β 1, IL -1 β and IL -8 in the SB2 group significantly decreased ( P <0.05). The cumulative mortality rate was significantly decreased in the SB2 and SB3 groups in comparison with that in the SB0 group after three hours of hypoxic stress ( P <0.05). In a 56-day feeding trial, SB enhanced largemouth bass growth by increasing antioxidant enzyme activity and inhibiting TLR 22- MyD 88 signaling, therefore increasing cumulative mortality from hypoxic stress in largemouth bass.

Abstract:To investigate the activities of intestinal digestive enzymes, liver antioxidant enzymes, immunological enzymes, and glucometabolic enzymes in largemouth bass (Micropterus salmoides) under the biofloc model, an experiment was conducted in 300-liter glass tanks. The experiment comprised a control group, which was fed a basal diet, and a biofloc group, where glucose was added to maintain a C/N ratio of 15. Each group had three parallel setups, with a stocking density of 20 fish per tank. The experiment ran for 60 days, employing a zero-water exchange aquaculture model. The results showed that at the end of the culture period, there were no significant differences between the initial weight, final weight, WGR, SGR, and SR of the biofloc group and the control group of largemouth bass (p > 0.05), whereas the lower FCR and the higher PER in the biofloc group were significant (p < 0.05); intestinal α-amylase, trypsin, and lipase activities of largemouth bass in the biofloc group were significantly increased by 37.20%, 64.11%, and 51.69%, respectively, compared with the control group (p < 0.05); liver superoxide dismutase and catalase activities, and total antioxidant capacity of largemouth bass in the biofloc group were significantly increased by 49.26%, 46.87%, and 98.94% (p < 0.05), while the malondialdehyde content was significantly reduced by 19.91% (p < 0.05); liver lysozyme, alkaline phosphatase, and acid phosphatase activities of largemouth bass in the biofloc group were significantly increased by 62.66%, 41.22%, and 29.66%, respectively (p < 0.05); liver glucokinase, pyruvate kinase, glucose-6-phosphate kinase, pyruvate kinase, glucose-6-phosphatase, and glycogen synthase activities were significantly increased by 46.29%, 99.33%, 32.54%, and 26.89%, respectively (p < 0.05). The study showed that the biofloc model of culturing largemouth bass can not only enhance digestive enzyme activities, antioxidant capacity, and immune response but can also promote the process of glucose metabolism and reduce feeding costs. This study provides data support for healthy culturing of largemouth bass in future production, provides a theoretical reference for optimizing the biofloc technology culture model, and is crucial for promoting the healthy and green development of aquaculture.

Abstract:This research revealed that low-salinity environments have positive effects on largemouth bass (Micropterus salmoides), with the study finding that low-salinity levels can have beneficial impacts on this species. A 8-week experiment was conducted using four salinity gradients S0, S1, S2, S3 (0, 2, 4, 6 psu), and biochemical indicators and 16S rRNA sequencing were analyzed. Results showed relatively good growth performance at S0 and S1, and no significant antioxidant stress response at S3. Na+–K+–ATPase activity was significantly lower in the S2 group. The diversity showed no significant differences between the treatment groups. However, with increasing salinity, the proportion of potentially pathogenic bacteria in freshwater decreased significantly while that in the seawater increased. Indicator species analysis showed a significantly higher abundance of positive microbial species in the 4 psu group. In conclusion, although growth performance decreased at 4 psu, antioxidant damage was relatively small and disease risk was significantly reduced, indicating a positive effect on the largemouth bass.

Abstract:Probiotics play an essential role in the largemouth bass (Micropterus salmoides) aquaculture sector. They aid the fish in sickness prevention, intestinal structure improvement, food absorption, and immune system strengthening. In this experiment, Bacillus subtilis (BS, 107 CFU/g) and Lactobacillus reuteri (LR, 107 CFU/g) were added to the feed and then fed to M. salmoides for 35 days. The effects of two probiotics on the growth, immunity, and metabolism of M. salmoides organisms were studied. The results revealed that the BS group significantly increased the growth rate and specific growth rate of M. salmoides, while both the BS and LR groups significantly increase the length of villi M. salmoides intestines. The BS group significantly increased the levels of AKP, T-AOC, and CAT in the blood of M. salmoides, as well as AKP levels in the intestine. Furthermore, the BS group significantly increased the expression of intestinal genes Nrf2, SOD1, GPX, and CAT, while significantly decreasing the expression of the keap1 gene. M. salmoides gut microbial analysis showed that the abundance of Planctomycetota was significantly different in both control and experimental groups. Analyzed at the genus level, the abundance of Citrobacter, Paracoccus, Luedemannella， Sphingomonas, Streptomyces and Xanthomonas in the both control and experimental groups were significantly different. The BS group's differentially expressed genes were predominantly enriched in oxidative phosphorylation pathways in the intestine, indicating that they had a good influence on intestinal metabolism and inflammation suppression. In contrast, differentially expressed genes in the LR group were primarily enriched in the insulin signaling and linoleic acid metabolism pathways, indicating improved intestine metabolic performance. In conclusion, B. subtilis and L. reuteri improve the growth and health of M. salmoides, indicating tremendous potential for enhancing intestinal metabolism and providing significant application value.

Abstract:High temperatures and low oxygen in aquatic environments, such as intensive aquaculture or in natural watersheds, inevitably cause stress in fish. Fish are exposed to high temperatures during the summer, which exacerbates hypoxia. Hypoxia (1.2 +/- 0.2 mg/L) under 20 degrees C (20 HG) and 26 degrees C (26 HG) was simulated to induce stress in largemouth bass (Micropterus salmoides). Related enzymes and genes involved in antioxidant, immune, and apoptotic responses were selected to explore the interactive effects of temperature and hypoxia on largemouth bass. The results showed that malondialdehyde (MDA) levels in plasma, gill, and liver increased in the 26 HG (p < 0.05). Liver superoxide dismutase (SOD) activity increased in the 26 HG. Peak SOD (SOD1, SOD2, SOD3a, and SOD3b), CAT, and GSH-Px mRNA levels in the gill and liver were observed at 12-24 h of stress. The levels of gill and liver total antioxidant capacity, catalase (CAT), glutathione peroxidase (GSH-Px) activities and other enzyme activities and genes in the 26 HG were higher than those in the 20 HG (p < 0.05). The gill and liver add phosphatase and alkaline phosphatase activities increased with time in the 26 HG (p < 0.05), while gill and liver lysozyme activities in the 26 HG were lower than those in the 20 HG (p < 0.05). Tumor necrosis factor-alpha mRNA level was upregulated in the gill and downregulated in the liver at 24 h in the 26 HG. Interleukin (IL)-1 beta and IL-8 mRNA levels were upregulated in the gill and liver in the 26 HG at 24 h, whereas IL-15 mRNA level was downregulated in the 26 HG at 12 h. Transforming growth factor-beta 1 mRNA level was upregulated in the gill in the 20 HG at 24 h, but downregulated in gill and liver in the 26 HG at 24 h. Similarly, IL-10, Hepcidin-1, and Hepcidin-2 showed lower expression levels in the 26 HG. Gill and liver caspase-3 activities were higher in the 26 HG (p < 0.05), and gill caspase-3 activity was higher than that in the liver. The mRNA levels of proapoptotic genes (caspase-3, caspase-8, and caspase-9) were higher in the 26 HG. The present study demonstrates the interactive effects of temperature and hypoxia on stress in largemouth bass gill and liver. These results will be helpful to understand the mechanisms of stress induced by temperature and hypoxia in fish and provide a theoretical basis for aquaculture management.

Modulation of growth performance, antioxidant capacity, non-specific immunity and disease resistance in largemouth bass (Micropterus salmoides) upon compound probiotic cultures inclusion

Abstract:An 8-week feeding trial was conducted to evaluate the effects of dietary supplementation of compound probiotic cultures (CPC; Bacillus subtilis, Lactobacillus plantarum and Saccharomyces cerevisiae) on the growth performance, antioxidant capacity, non-specific immunity and disease resistance of juvenile largemouth bass. Triplicate groups of largemouth bass (average weight 42.05 ± 0.02 g), with a destiny of 30 individuals per tank, were fed diets supplemented with different concentration of compound probiotic cultures (CPC) (0%, CPC (0.0); 0.5%, CPC (0.5); 1.0%, CPC (1.0); 2.0%, CPC (2.0)). After the feeding trial, tissue samples of largemouth bass were collected and the challenge test with Aeromonas hydrophila was performed. Results indicated that the CPC supplementation produced no significant difference on the growth performance, feed utilization and body composition of largemouth bass, while significantly increased the cumulative survival rate in the Aeromonas hydrophila challenge test. Meanwhile, the inclusion of CPC elevated the hepatic antioxidant capacity, and the highest activity of antioxidant enzymes, including T-AOC, CAT, GPx and T-SOD, was observed in the CPC (2.0) group. Meanwhile, the transcription of Nrf2/keap1 and antioxidant related genes, including CAT, GPx, GST, SOD1 and SOD2, was significantly elevated with the inclusion of CPC. In addition, the inclusion of CPC improved the non-specific immunity of largemouth bass. The activity of serum lysozyme was significantly elevated in the CPC (2.0) group, while the transcription of RelA and pro-inflammatory factors, including TNF-α and IL-1β, was inhibited with the inclusion of CPC. Meanwhile, related genes potentially linked to RelA, including TLR2 and p38 MAPK, were detected that their relative expression was significantly inhibited with the inclusion of CPC. The current findings indicated that the inclusion of 2% CPC improved the antioxidant capacity, non-specific immunity and disease resistance of juvenile largemouth bass, and suggested that 2% CPC as a functional additive could be applied to the diet of juvenile largemouth bass in aquaculture practice.
