Abstract:For revealing the mechanism of allelopathic influence on phytoplankton by aquatic macrophytes, the growth and photosynthetic activities of cyanobacteria Microcystis aeruginosa and the chlorophyte Selenastrum capricornutum were investigated when they coexisted with submerged macrophyte Myriophyllum spicatum and were exposed to allelopathic polyphenols: pyrogallic acid (PA), gallic acid (GA), ellagic acid (EA) and (+)-catechin (CA). According to the results of coexistence assays, the non-photochemical quenching (NPQ) and effective quantum efficiency (YII) of M. aeruginosa were affected earlier and more rapidly than the cell density. However, the influence of M. spicatum on S. capricornutum was not found. When the Toxicity Index (TI) was applied to evaluate the combined effects of binary and multiple mixtures of polyphenols, it was found that the four tested polyphenols with the proportion identified in the M. spicatum-cultured solution were observed to present synergistic effect (0.36-0.49) according to the cell density, NPQ and YII of M. aeruginosa. With the combined effects of polyphenols on S. capricornutum, only additive action (0.52-1.62) was found. On the other hand, PA (2.97mgL(-1)), GA (2.65mgL(-1)) caused significant reductions of photosystem II (PSII) and whole electron transport chain activities of M. aeruginosa by 71.43 and 18.37%, 70.95 and 40.77% (P<0.05), respectively, after 24-h exposure, but no inhibition effect was found in S. capricornutum. The dark respiration and photosystem I (PSI) activities of M. aeruginosa were significantly increased by exposure to PA and GA (P<0.05). Nevertheless, EA and CA had no influence on the electron transport activities of the tested organisms. These results indicate that the reduction in photosynthetic activity of M. aeruginosa and the synergistic effect of allelochemicals may be two important causes for the inhibition of undesired phytoplankton by submersed macrophytes in natural aquatic ecosystems, and PSII in cyanobacteria is considered to be one of the target sites attacked by allelopathic polyphenols.
The Growth Suppression Effects of UV-C Irradiation on Microcystis Aeruginosa and Chlorella Vulgaris under Solo-Culture and Co-Culture Conditions in Reclaimed Water.

Abstract:Harmful algal blooms (HAB5) are serious problems in landscape waters sourced from reclaimed water. In this study, the suppression effects of UV-C irradiation on microalgal growth were researched to find a possible preventive approach. Microcystis aeruginosa and Chtorella vulgaris were exposed to UV-C irradiation and then cultured in real reclaimed water for 7-18 d. UV-C irradiation at 50-200 mJ cm(-)(2) could inhibit the growth of M. aeruginosa, C. vulgaris, and both microalgae in co-culture for 3-14,1-3, and 1-5 d respectively. In addition, UV-C irradiation could cause damage to the cell integrity. At 100-200 mJ cm(-2) UV-C, the proportion of microalgal membrane damage (P-md) in M. aeruginosa cells increased rapidly to 56%-76% from day 3. whereas that in C. vulgaris cells increased to 23%-62% within 3 d. The photochemical efficiency (represented by Y value) of the irradiated groups was negatively affected immediately after UV-C irradiation and recovered gradually during the incubation. The Y value of M. aeruginosa cells began to recover from days 3 to 14, whereas that of C. vulgaris recovered much more quickly, from days 0.1 to 1. Overall, the irradiation-induced suppressive effects on algal growth correlated positively with the UV-C doses. Because M. aeruginosa was more sensitive to UV-C irradiation. UV-C irradiation not only controlled the total biomass of the mixed algae but also selectively reestablished the dominance of the nontoxic C. vulgaris. (C) 2020 Elsevier B.V. All rights reserved.
Enantioselective Toxicity of Chiral Herbicide Metolachlor to Microcystis Aeruginosa

Abstract:The enantioselective effects of chiral herbicides on aquatic organisms have received increasing attention. As one kind of freshwater algae responsible for most algal blooms, Microcystis aeruginosa can produce hepatotoxic microcystin and cause serious health concerns for drinking water. Thus, the effects of chiral herbicides on M. aeruginosa are of vital significance but poorly understood, especially as the structures of chiral herbicides become more complex. In this study, the enantioselective effects of four metolachlor enantiomers based on carbon center and axis chirality on M. aeruginosa were investigated for the first time at an enantiomeric level. The results of the investigation into algal growth inhibition, chlorophyll a content, and cell integrity indicated that ( S)-metolachlor [( S)-Met] was significantly more toxic than any other isomer. The toxicity ranking of different enantiomers at the highest concentration (15 mg/L) against M. aeruginosa was ( S)-Met > (α R,1' S)-Met > (α S,1' S)-Met > (α S,1' R)-Met > (α R,1' R)-Met, with (α S,1' S)-Met and (α R,1' S)-Met displaying a synergistic effect. Additionally, the Fe distribution in M. aeruginosa presented distinct enantioselectivity, which may contribute to the enantioselective toxicity of metolachlor. Furthermore, metolachlor upregulated the expression of genes mcyD and mcyH in an enantioselective manner, indicating that this herbicide can potentially promote the synthesis and efflux of microcystin, thus aggravating agricultural water contamination to different extents. Overall, this study will help to understand the ecotoxicity of metolachlor at a deeper level and provide theoretical insights into the enantioselective behaviors of metolachlor.
Quercetin Potentiates the Concurrent Hyper-Accumulation of Cellular Biomass and Lipids in Chlorella Vulgaris.

Abstract:Provision of chemical modulators has emerged as an effective strategy to govern cell growth and development. Here, the impact of flavonoid quercetin on algal growth, lipid accumulation and transcriptional patterns was investigated in the green microalga Chlorella vulgaris. These results demonstrated that quercetin (15 mu g/l) significantly enhanced the cellular biomass and photosynthetic efficiency, with up to 2.5-fold in the biomass in the stationary phase. Lipidomic analyses revealed that lipid content was increased by 1.8-fold. Furthermore, the functional mechanism of quercetin on the molecular level was dissected by transcriptomic analysis. Results revealed that quercetin upregulated the expression pattern of key genes involved in cellular signaling mechanisms such as phosphatidylinositol 4-kinase alpha, thus consequently enhanced cell growth. Altogether, the data present in this study demonstrate the dramatic role of quercetin on enhancing microalgal biomass and lipid accumulation by unprecedented regulation, of key metabolic nodes, for the first time and provide a novel insight into microalgal metabolism and regulation.
Physiological and biochemical responses of Microcystis aeruginosa to glyphosate and its Roundup® formulation

Abstract:Glyphosate may have dual effect on bloom algae as a phosphorus source or pesticide. The physiological and biochemical responses of Microcystis aeruginosa (M. aeruginosa) to glyphosate and its formulation in the common herbicide, Roundup(®), were compared. The result suggested that both the cell numbers and Chl-a content of M. aeruginosa increased when the glyphosate concentration increased from 0.01 to 5mg P L(-1). However, Roundup(®) showed low-dose (below 1mg P L(-1)) stimulation and high-dose (above 1mg P L(-1)) inhibition on M. aeruginosa cell density and Chl-a content (hormesis effect). Phosphate was more available than glyphosate or Roundup(®), and Roundup(®) was more toxic than glyphosate itself at 3mg P L(-1). Analysis of the maximum yield of PSII indicated that glyphosate stimulated the photosynthesis process while Roundup(®) inhibited the photosynthesis of M. aeruginosa. The photosynthesis process was enhanced on the 21st day compared with that on the 14th day in all P mediums. The extracellular alkaline phosphatase activity (APA) decreased with the increasing glyphosate or Roundup(®) concentration. The change pattern of APA was similar in both the glyphosate and Roundup(®) mediums.
The Synergistic Energy and Carbon Metabolism under Mixotrophic Cultivation Reveals the Coordination Between Photosynthesis and Aerobic Respiration in Chlorella Zofingiensis

Abstract:Microalgal biofuels have attracted much attention in recent years. Nevertheless, the relatively low growth rate and low cell density limits its industrial application. Mixotrophic cultivation has been considered as a good strategy to increase the specific growth rate and biomass yield in some microalgal species. However, the metabolic mechanism has not been well elucidated. To address it, Chlorella zofingiensis was employed in the present study. The biomass productivity and maximum dry weight achieved under mixotrophic condition were greater than the sum of those under photoautotrophic and heterotrophic conditions. Interestingly, compared with photoautotrophic cultures, the RuBisCO activity was reduced for mixotrophic cultures, indicative of the down-regulation of the Calvin cycle. When compared with heterotrophic cultures, the citrate synthase activity was attenuated for mixotrophic cultures, indicative of the down-regulation of Tricarboxylic Acid cycle. Biochemical analysis showed that mixotrophic cells contained more starch but less total fatty acid content than heterotrophic cells. Besides, the biomass yield on glucose was 0.789 g g− 1 for mixotrophic cultures, 63.7% higher than that for heterotrophic cultures, and the enhancement effect of light under mixotrophic cultivation was 7.35-fold. Based on these results, a novel mechanism for the synergistic effects in mixotrophic cultivation was proposed, in which glucose was more efficiently utilized for biomass production by the possible coordination of energy and carbon metabolism between photosynthesis and aerobic respiration.
Mechanistic insights into the effects of diuron exposure on Alexandrium pacificum

Abstract:Diuron (N-(3,4-dichlorophenyl)-N,N‐dimethylurea, DCMU), a ureic herbicide, is extensively used in agriculture to boost crop productivity; however, its extensive application culminates in notable environmental pollution, especially in aquatic habitats. Therefore, the present study investigated the effect of diuron on the dinoflagellate Alexandrium pacificum , which is known to induce harmful algal blooms (HAB), and its potential to biodegrade DCMU. Following a four-day DCMU exposure, our results revealed that A. pacificum proficiently assimilated DCMU at concentrations of 0.05 mg/L and 0.1 mg/L in seawater, attaining a complete reduction (100 % efficiency) after 96 h for both concentrations. Moreover, evaluations of paralytic shellfish toxins content indicated that cells subjected to higher DCMU concentrations (0.1 mg/L) exhibited reductions of 73.4 %, 86.7 %, and 75 % in GTX1, GTX4, and NEO, respectively. Exposure to DCMU led to a notable decrease in A. pacificum 's photosynthetic efficacy, accompanied by increased levels of reactive oxygen species (ROS) and suppressed cell growth, with a growth inhibition rate of 41.1 % at 72 h. Proteomic investigations pinpointed the diminished expression levels of specific proteins like SxtV and SxtW, linked to paralytic shellfish toxins (PSTs) synthesis, as well as key proteins associated with Photosystem II, namely PsbA, PsbD, PsbO, and PsbU. Conversely, proteins central to the cysteine biosynthesis pathways exhibited enhanced expression. In summary, our results preliminarily resolved the molecular mechanisms underlying the response of A. pacificum to DCMU and revealed that DCMU affected the synthesis of PSTs. Meanwhile, our data suggested that A. pacificum has great potential in scavenging DCMU.

Abstract:The intensive use of agrochemicals has led to nutrient loss, greenhouse gas emissions, and resource depletion, thus the development of sustainable agricultural solutions is required. Microalgal biomass has the potential to provide nutrients such as nitrogen, phosphorus, and potassium, along with various plant growth promoters, to enhance crop productivity and impart disease resistance. This study provides a comprehensive assessment of the potential applications of microalgal extracts and biomass in the contexts of seed germination, hydroponic systems, and soil-based crop cultivation. The results revealed that the extracts from Chlorella sp. and Anabaena sp. have no significant impact on the germination of wheat seeds. High concentrations of Chlorella sp. and Anabaena sp. cell extracts in hydroponics enhanced the length of cucumber seedling stems by 81.7 % and 58.3 %, respectively. Additionally, the use of microalgal cell extracts hindered root elongation while stimulating the growth of lateral and fibrous roots. Furthermore, the study compared the performance of 5 different fertilizers: 1) inorganic fertilizer (IF), 2) organic fertilizer (OF), 3) microalgae-based biofertilizer (MF), 4) inorganic fertilizer + microalgae-based biofertilizer (IM), 5) organic fertilizer + microalgae-based biofertilizer (OM). The findings indicate that the plant growth and soil physicochemical properties in the groups supplied with different fertilizers are comparable and significantly higher than those in the control group. The levels of protein, chlorophyll A, and chlorophyll B in the MF group increased significantly by 40 %, 29.2 %, and 33.5 %, respectively, compared to the control group. However, it remained notably lower compared to groups supplied with inorganic and organic fertilizers ( p < 0.05). Combining microalgae with organic fertilizer can simultaneously enhance the yield and quality of Chinese cabbage, representing a promising source of crop nutrition. In conclusion, this study suggests that it is promising to use microalgae to reduce the use of conventional fertilizers in hydroponics and soil-based cultivation.

Abstract:Ubiquitous distribution of pharmaceutical contaminants in environment has caused unexpected adverse effects on ecological organisms; however, how microorganisms recover from their toxicities remains largely unknown. In this study, we comprehensively investigated the effect of a representative pollutant, doxylamine (DOX) on a freshwater microalgal species, Chlorella sp. by analyzing the growth patterns, biochemical changes (total chlorophyll, carotenoid, carbohydrate, protein, and antioxidant enzymes), and transcriptomics. We found toxicity of DOX on Chlorella sp. was mainly caused by disrupting synthesis of ribosomes in nucleolus, and r/t RNA binding and processing. Intriguingly, additional bicarbonate enhanced the toxicity of DOX with decreasing the half-maximum effective concentrations from 15.34 mg L-1 to 4.63 mg L-1, which can be caused by inhibiting fatty acid oxidation and amino acid metabolism. Microalgal cells can recover from this stress via upregulating antioxidant enzymatic activities to neutralize oxidative stresses, and photosynthetic pathways and nitrogen metabolism to supply more energies and cellular signaling molecules. This study extended our understanding on how microalgae can recover from chemical toxicity, and also emphasized the effect of environmental factors on the toxicity of these contaminants on aquatic microorganisms.
Synergetic Suppression Effects Upon the Combination of UV-C Irradiation and Berberine on Microcystis Aeruginosa and Scenedesmus Obliquus in Reclaimed Water: Effectiveness and Mechanisms.

Abstract:The risk of harmful algal blooms (HABs) in the water recharged with reclaimed water is a bottleneck for water reuse. The suppression effects and mechanisms of the combination of UV-C and berberine on Microcystis aeruginosa and Scenedesmus obliquus in reclaimed water were investigated. Mono UV-C irradiation at 75 mJ cm(-2) could suppress the growth of M. aeruginosa for 7 d and that at 90 mJ cm(-2) could suppress the growth of S. obliquus for 5 d. UV-C irradiation combined with 02-2 mg L-1 berberine lengthened the inhibition period of M. aeruginosa to 10- > 22 d and that of S. obliquus to 7- > 22 d and induced more rapid lethal effects on the harmful microalgal cells, in significant synergetic patterns. The combination of UV-C and berberine suppressed total, intracellular and extracellular microcystin-LR (MC-LR) more effectively and decreased the MC-1.12 quota significantly, which further reduced the risks of microcystin production and release. Furthermore, synergetic mechanisms of the combined treatments were systematically investigated from the aspects of photosynthetic system (photosynthetic activity and pigments), metabolic activity (ATP and membrane potential), oxidation stress (reactive oxygen species (ROS) and glutathione (GSH)), and apoptosis-like cell death (phosphatidylserine (PS) ectropion, caspase-3 activity and terminal deoxynudeotidyl transferase dUTP nick end labeling (TUNEL) positive rate). The combination treatment provided a joint attack of UV-C and berberine on photosynthetic transport chain of photosynthetic system II (PS II), and a synergetic pathway to achieve more severe disruptions in energy metabolism as well as aggravated oxidative stress. The accumulated ROS enhanced increases in programmed cell death (PCD) indicators of both microalgal species, which contributed to the enhancement effects on growth suppression. The results showed that the combination treatment achieved lower dose requirements of both UV-C irradiation and berberine for inducing the same inhibition effects on microalgal cells, which was promising to be applied in the HABs control of reclaimed water. Neglected methane production and toxicity risk in low-frequency ultrasound for controlling harmful algal blooms

Abstract:Algal blooms are regarded as a significant source of CH4 emissions. Ultrasound has been gradually employed as a fast and efficient algae removal technology in recent years. However, the changes in water environment and potential ecological effects caused by ultrasonic algae removal are not fully clear. Here, a 40-day microcosm study was performed to simulate the collapse of Microcystis aeruginosa blooms after ultrasonic treatment. The results showed that low-frequency ultrasound at 29.4 kHz for 15 min removed 33.49% of M. aeruginosa and contributed to the destruction of cell structure, but it intensified the leakage of intracellular algal organic matter and microcystins. The accelerated collapse of M. aeruginosa blooms after ultrasonication promoted the rapid formation of anaerobic and reductive methanogenesis conditions, and elevated dissolved organic carbon content. Moreover, the release of labile organics, including tyrosine, tryptophan, protein-like compositions, and aromatic proteins, was facilitated by the collapse of M. aeruginosa blooms after ultrasonic treatment, and they supported the growth of anaerobic fermentation bacteria and hydrogenotrophic Methanobacteriales. This was also demonstrated by the increase in methyl-coenzyme M reductase (mcrA) genes in sonicated algae added treatments at the end of incubation. Finally, the CH4 production in sonicated algae added treatments was 1.43-fold higher than that in non-sonicated algae added treatments. These observations suggested that ultrasound for algal bloom control potentially increased the toxicity of treated water and its greenhouse gas emissions. This study can provide new insights and guidance to evaluate environmental effects of ultrasonic algae removal.
Growth Optimisation of Microalga Mutant at High CO2 Concentration to Purify Undiluted Anaerobic Digestion Effluent of Swine Manure

Abstract:Growth rate of the microalga Chlorella PY-ZU1 mutated by nuclear irradiation was optimised for use in the purification of undiluted anaerobic digestion effluent of swine manure (UADESM) with 3745 mg L (1) chemical oxygen demand (COD) and 1135 mg L (1) total nitrogen content. The problem of accessible carbon in UADESM was solved by continuous introduction of 15% (v/v) CO2. Adding phosphorus to UADESM and aeration of UADESM before inoculation both markedly reduced the lag phase of microalgal growth. In addition, the biomass yield and average growth rate of Chlorella PY-ZU1 increased significantly to 4.81 g L (1) and 601.2 mg L (1) d (1), respectively, while the removal efficiencies of total phosphorus, COD and ammonia nitrogen increased to 95%, 79% and 73%, respectively. Thus, the findings indicate that Chlorella PY-ZU1 can be used for effective purification of UADESM, while the biomass can be safely used as animal feed supplement. (C) 2014 Elsevier Ltd. Water reuse and growth inhibition mechanisms for cultivation of microalga Euglena gracilis

Abstract:Background: Microalgae can contribute to more than 40% of global primary biomass production and are suitable candidates for various biotechnology applications such as food, feed products, drugs, fuels, and wastewater treatment. However, the primary limitation for large-scale algae production is the fact that algae requires large amounts of fresh water for cultivation. To address this issue, scientists around the world are working on ways to reuse the water to grow microalgae so that it can be grown in successive cycles without the need for fresh water. Results: In this study, we present the results when we cultivate microalgae with cultivation water that is purified and reused. Specifically, we purify the cultivation water using an ultrafiltration membrane (UFM) treatment and investigate how this treatment affects: the biomass and biochemical components of the microalgae; characteristics of microalgae growth inhibitors; the mechanism whereby potential growth inhibitors are secreted (followed using metabolomics analysis); the effect of activated carbon (AC) treatment and advanced oxidation processes (AOPs) on the removal of growth inhibitors of Euglena gracilis. Firstly, the results show that E. gracilis can be only cultivated through two growth cycles with water that has been filtered and reused, and the growth of E. gracilis is significantly inhibited when the water is used a third time. Secondly, as the number of reused water cycles increases, the Cl- concentration gradually increases in the cultivation water. When the Cl- concentration accumulates to a level of fivefold higher than that of the control, growth of E. gracilis is inhibited as the osmolality tolerance range is exceeded. Interestingly, the osmolality of the reused water can be reduced by replacing NH4Cl with urea as the source of nitrogen in the cultivation water. Thirdly, E. gracilis secretes humic acid (HA)-which is produced by the metabolic pathways for valine, leucine, and isoleucine biosynthesis and by linoleic acid metabolism-into the cultivation water. Because HA contains large fluorescent functional groups, specifically extended π(pi)-systems containing C=C and C=O groups and aromatic rings, we were able to observe a positive correlation between HA concentration and the rate of inhibition of E. gracilis growth using fluorescence spectroscopy. Moreover, photosynthetic efficiency is adversely interfered by HA, thereby reductions in the synthetic efficiency of paramylon and lipid in E. gracilis. In this way, we are able to confirm that HA is the main growth inhibitor of E. gracilis. Finally, we verify that all the HA is removed or converted into nutrients efficiently by AC or UV/H2O2/O3 treatments, respectively. As a result of these treatments, growth of E. gracilis is restored (AC treatment) and the amount of biomass is promoted (UV/H2O2/O3 treatment). Conclusions: These studies have important practical and theoretical significance for the cyclic cultivation of E. gracilis and for saving water resources. Our work may also provide a useful reference for other microalgae cultivation.

Abstract:Quizalofop-Ethyl (QE) is a widely used phenoxy herbicide. It contains two enantiomers, and only its R-enantiomer quizalofop-p-ethyl (QpE) has herbicidal activity. Cyanobacterial blooms caused by Microcystis aeruginosa pose a threat to human health and aquatic environments, but the potential effects of chiral herbicides on M. aeruginosa are rarely studied. In this study, we carried out experiments using 0, 0.5, 1, 2, and 5 mg/L of QE and QpE to explore the effects on the growth, metabolism, synthesis, and release of microcystin-LR (MC-LR) in M. aeruginosa, which is the dominant species of algal blooms and indicator organism of water contamination. The biomass and chlorophyll a and protein content were consistent, showing that QpE at high concentrations (2 and 5 mg/L) showed greater acute toxicity than QE. On the expression of photosynthesis-related genes psaB and psbD1, QpE at 2 and 5 mg/L showed a stronger inhibition effect than QE. The oxidative stress caused by QE and QpE indicated that 144 h of exposure increased the concentration of MDA and enhanced the activities of superoxide dismutase and catalase. The synthesis of intracellular MC-LR in M. aeruginosa was stimulated by 144 h of QpE and QE exposure, and QpE showed a stronger stimulation effect than QE at high concentrations. The content of extracellular MC-LR increased after 144 h of QpE exposure and remained stable under QE exposure compared with that in the control group. Moreover, the expression of MC-LR-related gene mcyH was upregulated by QpE (144 h exposure) but downregulated by QE. These results are helpful to understand the toxicological effects of chiral herbicides on nontarget aquatic organisms, thus providing a theoretical basis for the rational use and the ecological risk assessment of chiral pesticides.

Abstract:The development of wastewater treatment processes capable of reducing and fixing carbon is currently a hot topic in the wastewater treatment field. Microalgae possess a natural carbon-fixing advantage, and microalgae that can symbiotically coexist with indigenous bacteria in actual wastewater attract more significant attention. Ultraviolet (UV) mutagenesis and dissolved organic carbon (DOC) acclimation were applied to strengthen the carbon-fixing performance of microalgae in this study. The mechanisms associated with microalgal water purification ability, gene regulation at the molecular level and photosynthetic potential under different trophic modes resulting from carbon fixation and transformation were disclosed. The superior performance of Chlorella sp. MHQ2 was eventually screened out among a large number of mutants generated from 3 wild-type Chlorella strains. Results indicated that the dry cell weight of the optimal species Chlorella sp. HQ mutant MHQ2 was 1.91 times that of the wild strain in the pure algal system, more carbon from municipal wastewater (MW) were transferred to the microalgae and re-entered into the biological cycle through resource utilization. In addition, COD, NH 3 -N and TP removal efficiencies of MW by Chlorella sp. MHQ2 were found to increase to 95.8% (1.1-times), 96.4% (1.4-times), and 92.9% (1.2-times), respectively, under the extra DOC supply and the assistance of indigenous bacteria in the MW. In the transcriptome analysis of the logarithmic phase, the glycolytic pathway was inhibited, and the pentose phosphate pathway was mainly carried out for microalgal life activities, further promoting efficient energy utilization. Upon analysis of carbon capture capacity and photosynthetic potential in trophic mode, the addition of NaHCO 3 increased the photosynthetic rate of Chlorella sp. MHQ2 in mixotrophy whereas it was attenuated in autotrophy. This study could provide a new perspective for the study of resource utilization and microalgae carbon- fixing mechanisms in the actual wastewater treatment process.

Abstract:The effects of UV-C irradiation on algal growth and cell integrity were investigated to develop a potential method for preventing cyanobacterial blooms. The toxic cyanobacterium Microcystis aeruginosa and three common freshwater green algae Chlorella ellipsoidea, Chlorella vulgaris, and Scenedesmus quadricanda were exposed to UV-C irradiation at 0–200mJcm−2 and subsequently incubated for 9–15 d under normal culture conditions. Cell density and cell integrity were assessed using flow cytometry. The results suggested that UV-C irradiation at 20–200mJcm−2 can suppress M. aeruginosa growth for 3–13 d in a dose-dependent manner. UV-C irradiation at 20 and 50mJcm−2 is sub-lethal to M. aeruginosa cells as over 80% of the exposed cells remained intact. However, UV-C irradiation at 100 and 200mJcm−2 induced severe cell disintegration in more than 70% of the irradiated cells. Neither significant suppression nor disintegration effects on green algae were observed for UV-C irradiation at 20–200mJcm−2 in this study. Taken together, the sensitivity of M. aeruginosa to UV-C irradiation was significantly higher than that of the non-toxic C. ellipsoidea, C. vulgaris, and S. quadricauda, suggesting the potential application of sub-lethal UV-C irradiation for M. aeruginosa bloom control with a predictable low ecological risk.

Abstract:Cultivation of high-lipid microalgae using municipal sewage can both treat polluted water and gain biodiesel. Rapid cultivation of microalgae is crucial for the entire process. In this study, four stimulants (phytohormone 2,4-d; Fe3+; photosynthetic bacteria—Rhodopseudomonas palustris; CO2) were firstly chosen to investigate their effects on microalgae Chlorella pyrenoidosa cultivation in municipal sewage. Meanwhile, orthogonal experiments were designed to compare the enhancing effects on the growth of microalgae C. pyrenoidosa and lipid accumulation by different combinations of stimulants, which were also conducted for outdoor culture. The optimum stimulant combination was 1.0 mg/L 2,4-d; 10−7 mol/L Fe3+; 4.5 mL (109 cells/mL) photosynthetic bacteria; and 8% CO2. Among the four kinds of stimulants, phytohormone 2,4-d and photosynthetic bacteria played more important roles on the microalgae growth and lipid accumulation. The optimum combination of stimulants could also promote the microalgae biomass and lipid yield during outdoor cultivation. The maximum OD681 of microalgae was 37.9% higher than that of the control group without addition of stimulants. The obtained microalgae dry weight and lipid production were 0.3 and 0.13 g /L, which were 80.0% and 85.7% higher than that of the control group, respectively. The promotion of microalgae photosynthetic rate was the main reason for the stimulant-enhanced microalgae cultivation. This study exerts positive effects on the development of microalgae biodiesel.

Abstract:Using low strength wastewater for microalgae cultivation is challenged by slow growth and biomass harvesting issue in suspended systems, and growth-promoting effects of phytohormones at currently recommended dosages could neither obtain high enough biomass concentrations nor economic feasibility. This study aims to solve the issues of slow growth, biomass harvest, and phytohormone costs altogether by supplementing low dosage phytohormones in an improved capillary-driven attached cultivation device. The device displayed nutrients-condensing properties, and dosages of indole acetic acid (IAA), 6-benzylaminopurine (6-BA), and salicylic acid (SA) for highest microalgal growth were respectively 10 -6 M, 10 -6 M, and 10 -7 M, being at least one order of magnitude lower than in suspended cultures. SA was most effective in growth-promoting (up to 7.0 g/m 2 biomass density) and nutrients uptake (up to 98.6 % from the bulk environment), while IAA was most effective in antioxidative defenses. These results provided new insights in cost-effective and harvesting-convenient microalgae production.

Abstract:This study investigated the use of Dracontomelon duperreanum leaf litter extract (DDLLE) in inhibiting the growth and photosynthesis of the algae Microcystis aeruginosa. The goal of the study was to evaluate a potential solution for cyanobacterial bloom prevention. M. aeruginosa was exposed to extract concentrations from 0.4 to 2.0 g L-1. Chlorophyll-a (Chl-a) content and photosynthesis levels were assessed using pulse amplitude modulated fluorimetry phytoplankton analyzer. Results suggested that the extract could efficiently suppress M. aeruginosa growth. The content of Chl-a was only 19.0 µg L-1 and achieved 96.0% inhibition rate when exposed to 2.0 g L-1 on day 15. Growth rate in response to different extract concentrations were consistent with changes in the photosynthesis efficiency (alpha), maximal relative electron transport rate and maximal photochemical efficiency of photosystem II (F v /F m ). Furthermore, several kinds of volatile chemicals and their concentrations in DDLLE had been identified by GC-MS, which of them play major role to suppress the growth of M. aeruginosa should be further studied.

Abstract:Heavy metal contamination negatively affects plants and animals in water as well as soils. Some microalgae can remove heavy metal contaminants from wastewater. The aim of this study was to screen green microalgae (GM) to identify those that tolerate high concentrations of toxic heavy metals in water as possible candidates for phytoremediation. Analyses of the tolerance, physiological parameters, ultrastructure, and transcriptomes of GM under Cd/Pb treatments were conducted. Compared with the other GM, Chlorella pyrenoidosa showed stronger tolerance to high concentrations of Cd/Pb. The reduced glutathione content and peroxidase activity were higher in C. pyrenoidosa than those in the other GM. Ultrastructural observations showed that, compared with other GM, C. pyrenoidosa had less damage to the cell surface and interior under Cd/Pb toxicity. Transcriptome analyses indicated that the "peroxisome" and "sulfur metabolism" pathways were enriched with differentially expressed genes under Cd/Pb treatments, and that CpSAT, CpSBP, CpKAT2, Cp2HPCL, CpACOX, CpACOX2, and CpACOX4, all of which encode antioxidant enzymes, were up-regulated under Cd/Pb treatments. These results show that C. pyrenoidosa has potential applications in the remediation of polluted water, and indicate that antioxidant enzymes contribute to Cd/Pb detoxification. These findings will be useful for producing algal strains for the purpose of bioremediation in water contamination.
