Abstract:Endophytic fungal molecules have the potential to be a cost-effective chemical source for developing eco-friendly disease-controlling pharmaceuticals that target mosquito-borne illnesses. The primary aims of the study were to identify the fungus Fusarium begoniae larvicidal ability against Aedes aegypti, Culex quinquefasciatus, and Anopheles stephensi. The ethyl acetate extract demonstrated lethal concentrations that kill 50% of exposed larvae (LC50) and 90% of exposed larvae (LC90) for the 1st to 4th instar larvae of An. stephensi (LC50 = 54.821, 66.525, 68.250, and 73.614; LC90 = 104.56, 138.205, 150.415, and 159.466 μg/mL), Cx. quinquefasciatus (LC50 = 64.981, 36.505, 42.230, and 36.514; LC90 = 180.46, 157.105, 140.318, and 153.366 μg/ mL), and Ae. aegypti (LC50 = 74.890, 33.607, 52.173, and 26.974; LC90 = 202.56, 162.205, 130.518, and 163.286 μg/mL). Mycelium metabolites were evaluated for their pupicidal activity towards Ae. aegypti (LC50 = 80.669, LC90 = 119.904), Cx. quinquefasciatus (LC50 = 70.569, LC90 = 109.840), and An. stephensi (LC50 = 73.269, LC90 = 110.590 μg/mL). The highest larvicidal activity was recorded at 300 µg/mL, with 100% mortality against first and second-instar larvae of Cx. quinquefasciatus. Metabolite exposure to larvae exhibited several abnormal behavioral changes. The exposure to F. begoniae metabolite, key esterases such as acetylcholinesterase, α-and-β-carboxylesterase, and acid and alkaline phosphatase activity significantly decreased compared to control larvae. The outcomes of the histology analysis revealed that the mycelium metabolites-treated targeted larvae had a disorganized abdominal mid and hindgut epithelial cells. The is first-hand information on study of ethyl-acetate-derived metabolites from F. begoniae tested against larvae and pupae of Ae. aegypti, Cx. quinquefasciatus and An. stephensi. Bio-indicator toxicity findings demonstrate that A. nauplii displayed no mortality. Insecticidal activities of Streptomyces sp. KSF103 ethyl acetate extract against medically important mosquitoes and non-target organisms

Abstract:A potentially novel actinobacterium isolated from forest soil, Streptomyces sp. KSF103 was evaluated for its insecticidal effect against several mosquito species namely Aedes aegypti , Aedes albopictus , Anopheles cracens and Culex quinquefasciatus . Mosquito larvae and adults were exposed to various concentrations of the ethyl acetate (EA) extract for 24 h. Considerable mortality was evident after the EA extract treatment for all four important vector mosquitoes. Larvicidal activity of the EA extract resulted in LC 50 at 0.045 mg/mL and LC 90 at 0.080 mg/mL for Ae. aegypti ; LC 50 at 0.060 mg/mL and LC 90 at 0.247 mg/mL for Ae. albopictus ; LC 50 at 2.141 mg/mL and LC 90 at 6.345 mg/mL for An. cracens ; and LC 50 at 0.272 mg/mL and LC 90 at 0.980 mg/mL for Cx. quinquefasciatus . In adulticidal tests, the EA extract was the most toxic to Ae. albopictus adults (LD 50 = 2.445 mg/mL; LD 90 = 20.004 mg/mL), followed by An. cracens (LD 50 = 5.121 mg/mL; LD 90 = 147.854 mg/mL) and then Ae. aegypti (LD 50 = 28.873 mg/mL; LD 90 = 274.823 mg/mL). Additionally, the EA extract exhibited ovicidal activity against Ae. aegypti (LC 50 = 0.715 mg/mL; LC 90 = 6.956 mg/mL), Ae. albopictus (LC 50 = 0.715 mg/mL; LC 90 = 6.956 mg/mL), and An. cracens (LC 50 = 0.715 mg/mL; LC 90 = 6.956 mg/mL), evaluated up to 168 h post-treatment. It displayed no toxicity on the freshwater microalga Chlorella sp. Beijerinck UMACC 313, marine microalga Chlorella sp. Beijerinck UMACC 258 and the ant Odontoponera denticulata . In conclusion, the EA extract showed promising larvicidal, adulticidal and ovicidal activity against Ae. aegypti , Ae. albopictus , An. cracens , and Cx. quinquefasciatus (larvae only). The results suggest that the EA extract of Streptomyces sp. KSF103 has the potential to be used as an environmental-friendly approach in mosquito control. The current study would serve as an initial step toward complementing microbe-based bioinsecticides for synthetic insecticides against medically important mosquitoes.

Biocontrol effect of entomopathogenic fungi Metarhizium anisopliae ethyl acetate-derived chemical molecules: An eco-friendly anti-malarial drug and insecticide

Abstract:Insect pests represent a major threat to human health and agricultural production. With a current over-dependence on chemical insecticides in the control of insect pests, leading to increased chemical resistance in target organisms, as well as side effects on nontarget organisms, the wider environment, and human health, finding alternative solutions is paramount. The employment of entomopathogenic fungi is one such potential avenue in the pursuit of greener, more target-specific methods of insect pest control. To this end, the present study tested the chemical constituents of Metarhizium anisopliae fungi against the unicellular protozoan malaria parasite Plasmodium falciparum, the insect pests Anopheles stephensi Listen, Spodoptera litura Fabricius, and Tenebrio molitor Linnaeus, as well as the nontarget bioindicator species, Eudrilus eugeniae Kinberg. Fungal crude chemical molecules caused a noticeable anti-plasmodial effect against P. falciparum, with IC50 and IC90 values of 11.53 and 7.65 µg/mL, respectively. The crude chemical molecules caused significant larvicidal activity against insect pests, with LC50 and LC90 values of 49.228-71.846 µg/mL in A. stephensi, 32.542-76.510 µg/mL in S. litura, and 38.503-88.826 µg/mL in T. molitor at 24 h posttreatment. Based on the results of the nontarget bioassay, it was revealed that the fungal-derived crude extract exhibited no histopathological sublethal effects on the earthworm E. eugeniae. LC-MS analysis of M. anisopliae-derived crude metabolites revealed the presence of 10 chemical constituents. Of these chemicals, three major chemical constituents, namely, camphor (15.91%), caprolactam (13.27%), and monobutyl phthalate (19.65%), were highlighted for potential insecticidal and anti-malarial activity. The entomopathogenic fungal-derived crude extracts thus represent promising tools in the control of insect pests and malarial parasites.

Abstract:Vector-borne diseases pose a significant public health challenge in economically disadvantaged nations. Malaria, dengue fever, chikungunya, Zika, yellow fever, Japanese encephalitis, and lymphatic filariasis are spread by mosquitoes. Consequently, the most effective method of preventing these diseases is to eliminate the mosquito population. Historically, the majority of control programs have depended on chemical pesticides, including organochlorines, organophosphates, carbamates, and pyrethroids. Synthetic insecticides used to eradicate pests have the potential to contaminate groundwater, surface water, beneficial soil organisms, and non-target species. Nanotechnology is an innovative technology that has the potential to be used in insect control with great precision. The goal of this study was to test the in vitro anti-dengue potential and mosquitocidal activity of Chaetomorpha aerea and C. aerea –synthesized Mn-doped superparamagnetic iron oxide nanoparticles ( CA -Mn-SPIONs). The synthesis of CA -Mn-SPIONs using C. aerea extract was verified by the observable alteration in the colour of the reaction mixture, transitioning from a pale green colour to a brown. The study of UV–Vis spectra revealed absorbance peaks at approximately 290 nm, which can be attributed to the surface Plasmon resonance of the CA -Mn-SPIONs. The SEM, TEM, EDX, FTIR, vibrating sample magnetometry, and XRD analyses provided evidence that confirmed the presence of CA -Mn-SPIONs. In the present study, results revealed that C. aerea aqueous extract LC 50 values against Ae. aegypti ranged from 222.942 (first instar larvae) to 349.877 ppm in bioassays (pupae). CA -Mn-SPIONs had LC 50 ranging from 20.199 (first instar larvae) to 26.918 ppm (pupae). After treatment with 40 ppm CA -Mn-SPIONs and 500 ppm C. aerea extract in ovicidal tests, egg hatchability was lowered by 100%. Oviposition deterrence experiments showed that in Ae. aegypti , oviposition rates were lowered by more than 66% by 100 ppm of green algal extract and by more than 71% by 10 ppm of CA -Mn-SPIONs (oviposition activity index values were 0.50 and 0.55, respectively). Moreover, in vitro anti-dengue activity of CA -Mn-SPIONs has good anti-viral property against dengue viral cell lines. In addition, GC–MS analysis showed that 21 intriguing chemicals were discovered. Two significant phytoconstituents in the methanol extract of C. aerea include butanoic acid and palmitic acid. These two substances were examined using an in silico methodology against the NS5 methyltransferase protein and demonstrated good glide scores and binding affinities. Finally, we looked into the morphological damage and fluorescent emission of third instar Ae. aegypti larvae treated with CA -Mn-SPIONs. Fluorescent emission is consistent with ROS formation of CA -Mn-SPIONs against Ae. aegypti larvae. The present study determines that the key variables for the successful development of new insecticidal agents are rooted in the eco-compatibility and the provision of alternative tool for the pesticide manufacturing sector.

Efficacy of ethyl acetate extract of Alangium salviifolium fruit pericarp against Culex quinquefasciatus larvae

Abstract:Mosquitoes transmit major human diseases, resulting in millions of fatalities each year and the development of chemical insecticide resistance, leading to a rebound in vectorial capacity. Plants could be used as a mosquito repellent alternative. The purpose of this study was to evaluate the biocontrol potentiality of ethyl acetate extract of fruit pericarp of Alangium salviifolium against Culex quinquefasciatus larvae. 100% larval mortality was recorded after 72 h of exposure with 50, 40 ppm, and 30 ppm concentrations of ethyl acetate extract against 3rd instar mosquito larvae. The bioactive compound responsible for larval mortality was isolated by TLC (Rf value of 0.33). 3rd instar larvae were found to be the most susceptible (LC50 = 3.60 ppm) among all the instars and corresponding LC50 values were 4.45 ppm and 4.52 ppm for 2nd and 4th instars larvae respectively after 72 h of exposure. The mortality rate of all larval instars was directly proportional to the concentration of bioactive compounds. Three-way ANOVA analysis revealed that larval instars, the concentration of bioactive compound, and time of exposure had a significant effect on larval mortality. In the bioactive TLC fraction of A. salviifolium (Rf value of 0.33), FT-IR spectroscopy analysis revealed the presence of numerous functional groups. GC-MS analysis revealed the presence of Benzoyl bromide and 3-Amino -5 (2-Furyl) Pyrazole in the extract. The compounds were also studied on non-target organisms such as Anisops sardea, 4th instar larvae of Chironomus sp. and Diplonychus annulatum, and in all the cases no abnormalities were recorded.
Bioactive components in Psidium guajava extracts elicit biotoxic attributes and distinct antioxidant enzyme modulation in the larvae of vectors of lymphatic filariasis and dengue

Abstract:Control of mosquito vectors, which have caused a global disease burden, has employed various methods. However, the challenges posed by current physical and chemical methods have raised concerns about vector control programs, leading to the search for alternative methods that are less toxic, eco-friendly, and cost-effective. This study investigated the larvicidal potential of aqueous, methanol, and ethylacetate extracts of Guava (Psidium guajava) against Aedes aegypti and Culex quinquefasciatus larvae. Functional group and phytochemical characterization were performed using Fourier-Transform Infrared Spectroscopy (FTIR) and GC-MS analysis to identify the bioactive compounds in the extracts. Larval bioassays were conducted using WHO standard procedures at concentrations of 12.5, 25, 50, 125, and 250 mg/L, and mortality was recorded after 24, 48, and 72 h. Additionally, antioxidant enzyme profiles in the larvae were studied. All of the solvent extracts showed larvicidal activity, with the methanol extract exhibiting the highest mortality against Ae. aegypti and Cx. quinquefasciatus larvae, followed by aqueous and ethylacetate extracts. FTIR spectroscopic analysis revealed the presence of OH, C-H of methyl and methylene, CO and CC. The GC-MS analysis indicated that the methanol, aqueous, and ethylacetate extracts all had 27, 34, and 43 phytoactive compounds that were effective at causing larvicidal effects, respectively. Different concentrations of each extract significantly modulated the levels of superoxide dismutase, catalase, glutathione peroxidase, and reduced glutathione in larvae. This study's findings indicate the potential for developing environmentally friendly vector control products using the bioactive components of extracts from P. guajava leaves.
Efficacy of bioactive compounds from Curcuma longa L. against mosquito larvae

Abstract:To identify larvicidal compounds from the ethanolic extracts of Curcuma longa root, the active compounds were isolated using activity-guided fractionation with column chromatography and identified based on nuclear magnetic resonance (NMR) and mass spectrometry (MS) data. The dipping method was used to determine the larvicidal activities of each compound against 4th-instar larvae of Culex pipiens pallens. Two compounds were isolated and identified, ar-turmerone and 8-hydroxyl-ar-turmerone. The two compounds exhibited larvicidal activities against the 4th-instar larvae of C.pipiens pallens after 24hr of treatment with LC50 values of 138.86 and 257.68ppm, respectively. The larvicidal activities of ar-turmerone and 8-hydroxyl-ar-turmerone against C.pipiens pallens are reported herein for the first time. The elucidation of the structure of these phytochemicals and their insecticidal activities are important for assessing the potential of this plant as a botanical insecticide.

Abstract:Helicoverpa armigera , a ubiquitous polyphagous pest, poses a significant threat to global agriculture, causing substantial economic losses and demonstrating resistance to synthetic pesticides. This study investigates the potential of emamectin benzoate (EMB), an avermectin derivative, as an effective control agent against H. armigera . The larvae of the NBII-MP–NOC–01 strain of H. armigera were reared on an artificial diet. The impact of dietary EMB was examined on four midgut enzymes; alanine aminotransferase (ALT), aspartate aminotransferase (AST), acid phosphatase (ACP), and alkaline phosphatase (ALP). Results showed a dose-dependent and time-dependent reduction in ALT and AST activity, while an initial increase and subsequent decline in ACP and ALP activity at higher EMB concentrations. Computational modelling of enzyme structures and molecular docking studies revealed differential binding of EMB with the midgut enzymes. The strongest interaction was observed between EMB and ALT residues, contrasting with weakest interactions observed with AST. The study also showed that decreased activity of transaminases in H. armigera caused by EMB may be because of stability-activity trade-off, while in phosphatases reverse may be the case. This research provides crucial insights into the biochemical responses and the intricate insecticide-enzyme interactions in H. armigera caused by EMB exposure. This study lays the foundation for further research aimed at developing environmentally friendly approaches for managing H. armigera , addressing the challenges associated with conventional pesticides.

Insecticidal and larvicidal activities of medicinal plant extracts against mosquitoes

Abstract:In recent years, use of environment friendly and biodegradable natural insecticides of plant origin have received renewed attention as agents for vector control because they are rich in bioactive chemicals, active against a limited number of species including specific target insects, and biodegradable. The present study was carried out to evaluate the adulticidal, repellent, and larvicidal activity of crude hexane, ethyl acetate, and methanol extracts of eight plants, viz. Aristolochia indica L., Cassia angustifolia Vahl, Diospyros melanoxylon Roxb., Dolichos biflorus L., Gymnema sylvestre (Retz) Schult, Justicia procumbens L., Mimosa pudica L., and Zingiber zerumbet L., were tested against adult and early fourth instar larvae of Culex gelidus Theobald and Culex quinquefasciatus Say (Diptera: Culicidae). The effective adult mortality was observed in methanol extract of A. indica, ethyl acetate extract of D. biflorus, and ethyl acetate and hexane extract of Z. zerumbet against C. gelidus and C. quinquefasciatus (LD50 = 37.75, 78.56, 129.44, 86.13, 80.06, 112.42, 53.83, and 46.61; LD90 = 166.83, 379.14, 521.50, 289.83, 328.18, 455.72, 181.15, and 354.50 ppm, respectively). Complete protections for 150 min were found in hexane and methanol extract of A. indica and Z. zerumbet at 1,000 ppm against mosquito bites. The highest larval mortality was found in the hexane extract of Z. zerumbet, ethyl acetate extract of D. biflorus, and methanol extracts of A. indica against C. gelidus (LC50 = 26.48, 33.02, and 12.47 ppm; LC90 = 127.73, 128.79, and 62.33 ppm) and against C. quinquefasciatus (LC50 = 69.18, 34.76, and 25.60 ppm; LC90 = 324.40, 172.78, and 105.52 ppm), respectively, after 24 h. The plant extracts are potential to be used as an ideal eco-friendly approach for the control of the Japanese encephalitis vector, C. gelidus, and lymphatic filariasis vector, C. quinquefasciatus.

Identification of insecticidal molecule aucubin from Metarhizium anisopliae ethyl acetate crude extract against disease mosquito vector

Abstract:Identification of insecticidal compounds from the entomopathogenic fungus Metarhizium anisopliae crude extract was performed by TLC and column fractionation. From the fractionation four fractions were separated and tested for larvicidal, pupicidal and adulticidal potential against Aedes aegypti, Anopheles stephensi and Culex quinquefasciatus mosquitoes. Among the four fractions the fraction-3 was most effective against larvae with LC50 values, 1.238, 1.300 and 2.899 for pupae, 1.440, 1.657, 2.593 and adult, 1.988, 2.037 and 4.086 μg/ml against Ae. aegypti, An. stephensi and Cx. quinquefasciatus at 24 h post treatment respectively. The effective fraction-3 were charectrized through FT-IR, UV–Vis spectroscopy, 13C-NMR and 1H-NMR, HPLC, and LC–MS analysis allowed to identifying the effective compound in fraction-3. Overall, the results confirmed the use of the fungus derived molecules as an active green pesticide against mosquito vectors of public importance and led to the identification of highly effective mosquitocidal products. In particular the toxic activity of aucubin far encompassed that of the large majority of earlier studied microbial insecticides, making this molecule an ideal candidate for the development of insecticides in the "real world".

Abstract:Mosquito-borne diseases, such as malaria, dengue, and Zika, pose major public health challenges globally, affecting millions of people. The growing resistance of mosquito populations to synthetic insecticides underscores the critical need for effective and environmentally friendly larvicides. Although chemical pesticides can initially be effective, they often lead to negative environmental consequences and health hazards for non-target species, including humans. This study aimed to evaluate the larvicidal effects of Trachyspermum ammi essential oil and Delphinium speciosum extract on the larvae of three major mosquito species: Aedes aegypti, Anopheles stephensi, and Culex quinquefasciatus. Mosquito larvae of Ae. aegypti, An. stephensi, and Cx. quinquefasciatus were reared under controlled laboratory conditions. The larvicidal activity of T. ammi essential oil and D. speciosum extract was evaluated through standard bioassays, using various concentrations of essential oils (10, 20, 40, 80, and 160 ppm) and extracts (160, 320, 640, 1280, and 2560 ppm) to determine the lethal concentration (LC50) values after 24 h of exposure. Fresh plant materials were collected, with the essential oil extracted via hydro-distillation, and the extract prepared using methanol solvent extraction. The chemical composition of T. ammi essential oil was examined using gas chromatography-mass spectrometry (GC-MS). Additionally, the preliminary analysis of the chemical compounds in D. speciosum extract was carried out using thin layer chromatography (TLC) and nuclear magnetic resonance spectroscopy (NMR) techniques. The results indicated that the essential oil of T. ammi exhibited more effective larvicidal activity compared to the D. speciosum extract. Specifically, the essential oil demonstrated LC50 values of 18 ppm for Cx. quinquefasciatus and 19 ppm for Ae. aegypti. In contrast, the D. speciosum extract showed the strongest larvicidal effect against An. stephensi, with an LC50 of 517 ppm. Concentrations of 40 ppm of the essential oil and 1280 ppm of the extract resulted in 100% mortality across all three species. Both the essential oil of T. ammi and the D. speciosum extract exhibited concentration-dependent larvicidal activity, and these results were statistically significant (p < 0.001) compared to the no-treatment group. GC-MS analysis revealed thymol (88.95%), o-cymen-5-ol (4.11%), and γ-terpinene (2.10%) as the major constituents of the T. ammi essential oil. Additionally, TLC verified the presence of alkaloids in both chloroform and methanolic extracts. Proton NMR identified a diterpene structure for these alkaloids. These findings suggest that T. ammi essential oil is a promising candidate for natural mosquito control strategies. Given its efficacy, further research is warranted to explore its potential in integrated vector management programs.
Eco-friendly bio-larvicidal and antimicrobial activity of isolated bioactive compound from Kurthia gibsonii (Bacillales)

Abstract:The targeted organisms include mosquito vectors, bacterial pathogens and non-targeted organisms. Preliminary mosquito larvicidal activity was conducted using cell-free supernatants (CFSs) from 11 gut bacteria. Among them, the bacterium SS11 exhibited promising results and was identified as Kurthia gibsonii based on its 16S rRNA sequence (1350 bp). The diethyl ether extract (DEE) of K. gibsonii demonstrated significant larvicidal effects, with LC50 values of 5.59 μL/mL and 8.59 μL/mL for 3rd instar larvae of Aedes aegypti and 2nd instar larvae of Anopheles stephensi , respectively. Analysis of the DEE using FT-IR, and GC-MS revealed the presence of 16 functional groups, and 7 bioactive compounds, respectively. A molecular docking study identified GC-MS compounds against odorant receptors from A. aegypti and odorant-binding proteins from A. stephensi was performed to assess the interaction and binding affinity. Overall, these findings suggest that the bioactive compounds 2, 4, 6-tribromoaniline from the DEE of K. gibsonii hold potential as an environmentally compatible alternative for biocontrol purposes, and compounds 9-tricosene and didecyl phthalate can be used for mosquito traps.

Abstract:The plant essential oils are intricate blends of semi-volatile and volatile substances. They primarily consist of monoterpenes and sesquiterpenes; however, very trace amounts of diterpenes, triterpenes, and tetraterpenes are also present. In view of environmental concerns, highly aromatic essential oils, exhibit a variety of bioactivities against the targeted species as a novel pest control alternative to synthetic insecticides. As a result, the study aims to evaluate the impact of essential oils (EO) extracted from various plant species: Azadirachta indica, Citrus limon, Eucalyptus globulus, Laurus nobilis, Mentha piperita , and Ocimum tenuiflorum . Gas chromatography and Mass spectrometry (GC–MS) were used to determine the chemical components of these essential oils. Furthermore, the extracted oils were subjected to individual and combination formulations of bio-insecticides, which were assessed on houseflies and cockroaches in comparison to a blank control and the positive control dichlorvos (dimethyl 2, 2-dichlorovinyl phosphate). The findings showed that the maximum essential oil yields from the leaves of A. indica, E. globulus , and O. tenuiflorum were 11.38, 9.64 and 10.41 %, respectively. Compared to DDVP, which killed all insects within the 12-hour period, the combined form of leaf essential oils at 100 % concentration proved to be more effective in controlling insect mortality. It accounted for 98 % of the deaths in houseflies and 76.4 % of the deaths in cockroaches, according to the contact toxicity evaluation. The active insecticidal components in the formulation include eucalyptol, undecane, tetradecane, estragole, pentadecane, and caryophyllene.

Abstract:Developed mosquitocidal resistance and associated health problems caused by synthetic insecticides, pave the way to control the mosquito population by using botanicals. Artemisia pallens / Davana, is renowned for its fragrance properties and thus well exploited in perfumery industries. The present study aimed to assess the larvicidal activity of Artemisia pallens leaf extracts against Culex quinquefasciatus. The preliminary qualitative analysis of the phytoconstituents and larvicidal activity against the fourth instar larvae of Culex quinquefasciatus was performed to analyse the efficacy of the chloroform, methanolic, and aqueous extracts of Artemisia pallens leaves. The chemical profiling of two extracts (Chloroform, Methanol) with better larvicidal activity was done by GC-MS. 3 compounds with insecticidal activity was docked against Acetylcholinesterase enzyme. All the extracts contain variable quantities of important secondary metabolites. The chloroform and methanolic extracts of Artemisia pallens leaves exhibit better larvicidal effect on Culex quinquefasciatus with a low LC50 value. The GC-MS analysis revealed the presence of 21 and 36 phytocomponents in chloroform and methanolic extracts of Artemisia pallens leaves respectively. Amongst the chosen metabolites valproic acid, pseudo solasodine diacetate, and alpha-terpineol, pseudo solasodine diacetate binds with lowest energy (-9. 86 Kcal/Mol) against Acetylcholinesterase. Thus Artemisia pallens leaves could serve as a natural weapon against Culex quinquefasciatus in the larval stage and could also control the adult form by blocking their nervous activity. As plant-based mosquitocides, are safe to humans and environment, Artemisia pallens leaves could be used to control mosquito borne diseases.

Abstract:Jussiaea repens (L.) leaf ethanol extract (LEE) and its major phyto-compound (MPC) have effects against larvae and adults of dengue, malarial, and filarial vectors. Total larval death rates were recorded from LEE and MPC has significant larval killing activity with LC50/LC90 values of Ae. albopictus, An. stephensi, and Cx. quinquefasciatus that were 118.3/229.9, 116.1/216.8, 114.4/213.5 and 17.7/27.5, 15.6/25.4 and 12.3/21.1 μg/ml, respectively. A best repellent activity was ascertained at 3.057 mg/cm2 concentration of LEE and MPC provided 100% protection upto 240 min against selected human vector mosquitoes (HVMs). The functional groups were confirmed by FT-IR analysis and their presence in ethanol extract and mass spectral analysis: 4-piperidineacetic acid, 1-acetyl-5-ethyl-2-[3-(2-hydroxyethyl]-1H-indol-2-yl]-á-methyl-, methyl ester compound was identified in the LEE. The results obtained suggest that J. repens LEE and its MPC were important and responsible for health protection and control of HVMs.

Abstract:Currently, mosquito control programs primarily rely on the utilization of chemically based insecticides. The objective of this study was to evaluate the efficacy of the plant Stemodia viscosa Roxb. against the early life stages of Culex quinquefasciatus, Anopheles stephensi and Aedes aegypti mosquitoes. The larvae, pupae and eggs of the mosquitoes were exposed to four different concentrations (62.5 to 500 ppm). After 120 h of treatment, the hexane and ethyl acetate extracts of S. viscosa exhibited 100% ovicidal activity against the eggs of the treated mosquitoes. The hexane and ethyl acetate extracts showed significant larvicidal activity against all three mosquitoes. The LC50 values of the hexane extract were 272.5, 233.5, 217.9 ppm and the LC50 values of the ethyl acetate extract were 238.6, 230.5 and 169.1 ppm respectively, after 24 h. The hexane and ethyl acetate extracts of S. viscosa exhibited promising pupicidal activity with LC50 values of 286.6, 265.3, 257.9 and 192.2, 290.9 and 238.1 ppm, respectively, after 24 h against all three mosquitoes. Additionally, histopathological and growth disruption studies revealed severe damage in the midgut cells of the treated larvae. The suitability index (SI) or predator safety factor (PSF) values indicated that the extracts from S. viscosa could be safely utilized in Integrated Mosquito Management (IMM) without harming the predators. Furthermore, formulations based on hexane and ethyl acetate extracts demonstrated strong activity while exhibiting no toxicity towards non-target organisms. The results imply that the extract holds potential as an eco-friendly substitute for synthetic insecticides in mosquito control. However, additional research is required to identify the most potent components of the plant extract to enhance product efficacy and stability.

Abstract:The development of insecticide resistance in mosquitoes of public health importance has encouraged extensive research into innovative vector control methods. Terpenes are the largest among Plants Secondary Metabolites and have been increasingly studied for their potential as insecticidal control agents. Although promising, terpenes are insoluble in water, and they show low residual life which limits their application for vector control. In this study, we developed and evaluated the performances of terpenoid-based nanoemulsions (TNEs) containing myrcene and p-cymene against the dengue vector Aedes aegypti and investigated their potential toxicity against non-target organisms. Our results showed that myrcene and p-cymene showed moderate larvicidal activity against mosquito larvae compared to temephos an organophosphate widely used for mosquito control. However, we showed similar efficacy of TNEs against both susceptible and highly insecticide-resistant mosquitoes from French Guyana, hence suggesting an absence of cross-resistance with conventional insecticides. We also showed that TNEs remained effective for up to 45 days in laboratory conditions. The exposure of zebrafish to TNEs triggered behavioral changes in the fish at high doses but they did not alter the normal functioning of zebrafish organs, suggesting a good tolerability of non-target organisms to these molecules. Overall, this study provides new insights into the insecticidal properties and toxicity of terpenes and terpenoid-based formulations and confirms that TNE may offer interesting prospects for mosquito control as part of integrated vector management.

Abstract:Mosquito-borne diseases are a major public-health concern worldwide. The development of resistance in mosquitoes to the existing larvicides and the associated-risks necessitates the exploration of novel molecules to control vector-borne pathogens. This study demonstrates that N-alkylamides [2 E ,4 E -N-isobutyl-undeca-2,4-diene-8,10-diynamide ( 1 ) and 2 E ,4 E ,8 Z ,10 E -N-isobutyl-dodeca-2,4,8,10-tetraenamide ( 2 )] of Acmella ciliata exhibit biocidal action against Aedes aegypti and Culex quinquefasciatus larvae. The LC 50 for 1 and 2 were 44.19 and 18.28 ppm against Aedes larvae, and 30.89 and 11.75 ppm against Culex larvae, respectively. We further observed that the chain length, degree of unsaturation, and terminal methylation influence the larvicidal efficacy of N-alkylamides. HPLC-assisted quantification revealed that the content of both 1 (19.01 mg/g) and 2 (68.55 mg/g) was maximum in the flowers of A. ciliata . Thus, this study concludes that N-alkylamides are promising alternatives to control mosquito larvae and can be used as standard for quantitation in test samples.

Abstract:The larvicidal potential of crude leaf extracts of Rhizophora mucronata, the red mangrove, using diverse solvent extracts of the plant against the early fourth instar larvae of Anopheles stephensi, Culex quinquefasciatus and Aedes aegypti mosquito vectors was analyzed. The acetone extract of R. mucronata showed the greatest efficacy: for Cx. quinquefasciatus (LC50 = 0.13 mg/mL; LC90 = 2.84 mg/mL), An. stephensi (LC50 = 0.34 mg/mL; LC90 = 6.03 mg/mL), and Ae. aegypti (LC50 = 0.11 mg/mL; LC90 = 1.35 mg/mL). The acetone extract was further fractionated into four fractions and tested for its larvicidal activity. Fraction 3 showed stronger larvicidal activity against all the three mosquito larvae. Chemical characterization of the acetone extract displayed the existence of several identifiable compounds like phytol, 3,7,11,15-tetramethyl-2-hexadecen-1-ol, 1-hexyl-2-nitrocyclohexane, eicosanoic acid etc. Enzyme assay displayed that R. mucronata active F3-fractions exert divergent effects on all three mosquitos’ biochemical defensive mechanisms. The plant fractions displayed significant repellent activity against all the three mosquito vectors up to the maximum repellent time of 210 min. Thus, the bioactive molecules in the acetone extract of R. murconata leaves showed significant larvicidal and enzyme inhibitory activity and displayed novel eco-friendly tool for mosquito control.

Abstract:Common household mosquitoes are a major public nuisance, and mosquito-borne diseases are the greatest threat to public health. In the current situation, it would be ideal to control mosquito populations without harming the environment. The present study aims to evaluate the mosquitocidal efficacy of fractions and the active principle niloticin isolated from the leaves of Limonia acidissima Linn. against the life stages of Culex quinquefasciatus Say mosquitoes. The concentrations used to screen the fractions were 10.0, 7.5, 5.0, and 2.5 ppm and the active compound niloticin was tested at concentrations of 2.0, 1.5, 1.0, and 0.5 ppm on the larval (L3) and pupal stages of Cx. quinquefasciatus . The results showed that fraction-8 recorded the highest mosquitocidal activity, and the lethality results (LC 50 ) after 24 hours were 3.44 ppm for larvae and 5.14 ppm for pupae of Cx. quinquefasciatus , respectively. Similarly, LC 50 of niloticin at 24 hours were 0.42 ppm for larvae and 0.51 ppm for pupae of Cx. quinquefasciatus , respectively. In addition, at a concentration of 2.0 ppm, niloticin showed an ovicidal activity of 94.4% and a deterrent effect on oviposition of 100% and exhibited considerable growth disruption. In conclusion, the results indicate that niloticin can be used for vector mosquito control.
