Abstract:Ideal bone scaffolds having good biocompatibility, good biodegradability, and beneficial mechanical properties are the basis for bone tissue engineering. Specifically, cell migration within 3D scaffolds is crucial for bone regeneration of critical size defects. In this research, hydroxyapatite scaffolds with three different types of architectures (tortuous, parallel, and graded channels) are fabricated using the freeze‐casting (ice‐templating) method. While most studies promote cell migration by chemical factors, it can be greatly enhanced by introducing only graded channels as compared with tortuous or parallel channels. The results provide insights and guidance in designing novel scaffolds to enhance cell migration behavior for bone tissue regeneration.

Abstract:Bone scaffolds are widely employed for treating various bone disorders, including defects, fractures, and accidents. Gradient bone scaffolds present a promising approach by incorporating gradients in shape, porosity, density, and other properties, mimicking the natural human body structure. This design offers several advantages over traditional scaffolds. A key advantage is the enhanced matching of human tissue properties, facilitating cell adhesion and migration. Furthermore, the gradient structure fosters a smooth transition between scaffold and surrounding tissue, minimizing the risk of inflammation or rejection. Mechanical stability is also improved, providing better support for bone regeneration. Additionally, gradient bone scaffolds can integrate drug delivery systems, enabling controlled release of drugs or growth factors to promote specific cellular activities during the healing process. This comprehensive review examines the design aspects of gradient bone scaffolds, encompassing structure and drug delivery capabilities. By optimizing the scaffold's inherent advantages through gradient design, bone regeneration outcomes can be improved. The insights presented in this article contribute to the academic understanding of gradient bone scaffolds and their applications in bone tissue engineering.

Abstract:Continuous gradients present at tissue interfaces such as osteochondral systems, reflect complex tissue functions and involve changes in extracellular matrix compositions, cell types and mechanical properties. New and versatile biomaterial strategies are needed to create suitable biomimetic engineered grafts for interfacial tissue engineering. Silk protein-based composites, coupled with selective peptides with mineralization domains, were utilized to mimic the soft-to-hard transition in osteochondral interfaces. The gradient composites supported tunable mineralization and mechanical properties corresponding to the spatial concentration gradient of the mineralization domains (R5 peptide). The composite system exhibited continuous transitions in terms of composition, structure and mechanical properties, as well as cytocompatibility and biodegradability. The gradient silicified silk/R5 composites promoted and regulated osteogenic differentiation of human mesenchymal stem cells in an osteoinductive environment in vitro. The cells differentiated along the composites in a manner consistent with the R5-gradient profile. This novel biomimetic gradient biomaterial design offers a useful approach to meet a broad range of needs in regenerative medicine.

Abstract:One of the most important issues in bone tissue engineering is the search for new materials and processing techniques to create novel scaffolds with 3-D porous structures. Although many properties such as biodegradability and porosity have been considered in designing bone scaffolds, very limited attention is paid to their capillary effect. In nature, capillary effect is ubiquitously used by plants and animals to constantly transport water and nutrients based on morphological and/or chemical gradient structures at multiple length-scales. In this work, we developed a modified freeze-casting technique to prepare ceramic scaffolds with gradient channel structures. The results show that our hydroxyapatite (HA) scaffolds have interconnected gradient channels that mimic the porous network of natural bone. More importantly, we demonstrate that such a scaffold has a very unique capillary behavior that promotes the self-seeding of cells when in contact with a cell solution due to spontaneous capillary flow generated from gradient channel structures. The strategy developed here provides a new avenue for designing "smart" scaffolds with complex porous structures and biological functions that mimic natural tissues.

Abstract:The tissue loss or organ failure, resulting from trauma or diseases, is a major concern in human healthcare. The recent development of regenerative medicine by combing the cells and degradable scaffolds offers a unique pathway to recreate tissues and organs with high bioactivity and biomimetic structures. This article reviews the state-of-art of the ordered and gradient scaffolds in terms of their preparation methods and biological performance. In contrast with the homogeneous counterpart, the ordered scaffolds facilitate delivery of nutrients and metabolites, and can guide cell growth, distribution and new tissue formation in vitro, leading to construction of 3D-engineered tissues with ordered cells and extracellular matrix. On the other hand, the gradient cues ( e. g. physical, chemical and biological properties) incorporated into the 3D scaffolds can create a microenvironment governing the cell behaviors such as migration rate and direction as well as differentiation, and thereby guiding the tissue regeneration. The methods of phase separation, electrospinning, injection and centrifugation are usually employed to prepare the ordered and gradient 2D substrate and 3D scaffolds. The biological performance of these scaffolds is discussed. Wish such a structure mimicking the native tissue-like environment, the scaffolds are expected to show exceptional advantages in regeneration of tissues and organs of better functions and structures.

Abstract:Introduction: The bone ingrowth depth in the porous scaffolds is greatly affected by the structural design, notably the pore size, pore geometry, and the pore distribution. To enhance the bone regeneration capability of scaffolds, the bionic design can be regarded as a potential solution.Objectives: We proposed a Haversian system-like gradient structure based on the triply periodic minimal surface architectures with pore size varying from the edge to the center. And its effects in promoting bone regeneration were evaluated in the study.Methods: The gradient scaffold was designed using the triply periodic minimal surface architectures. The mechanical properties were analyzed by the finite element simulation and confirmed using the universal machine. The fluid characteristics were calculated by the computational fluid dynamics analysis. The bone regeneration process was simulated using a in silico computational model containing the main biological, physical, and chemical variation during the bone growth process. Finally, the in vitro and in vivo studies were carried out to verify the actual osteogenic effect.Results: Compared to the uniform scaffold, the biomimetic gradient scaffold demonstrated better performance in stress conduction and reduced stress shielding effects. The fluid features were appropriate for cell migration and flow diffusion, and the permeability was in the same order of magnitude with the natural bone. The bone ingrowth simulation exhibited improved angiogenesis and bone regeneration. Higher expression of the osteogenesis-related genes, higher alkaline phosphatase activity, and increased mineralization could be observed on the gradient scaffold in the in vitro study. The 12-week in vivo study proved that the gradient scaffold had deeper bone inserting depth and a more stable bone-scaffold interface.Conclusion: The Haversian system-like gradient structure can effectively promote the bone regeneration. Natural and synthetic polymers, bioresorbable inorganic and hybrid materials, and tissue decellularized were deemed biomaterials scaffolding because of their improved structural, mechanical, and biological abilities.Various biomaterials, existing treatment methodologies and emerging technologies in the field of Three-dimensional (3D) and hydrogel processing, and the unique fabric concerns for tissue engineering. A scaffold that acts as a transient matrix for cell proliferation and extracellular matrix deposition, with subsequent expansion, is needed to restore or regenerate the tissue. Abstract:Every year, millions of people around the world undergo bone graft and artificial prosthesis transplants which engenders the repair and/or replacement of native bone for treating bone defects and injury. As a result, bone has become the second most transplanted tissue globally and there is a growing need for advances in bone tissue engineering (BTE) to ensure improved quality of life. The advent of 3D bone scaffolds has led to a paradigm shift in this field. These scaffolds act as an extra cellular matrix (ECM) providing a 3D environment for cell adhesion, proliferation, and differentiation. The scaffolds so fabricated, must meet the mechanical and biological criteria for successful clinical translation. Initial iterations of these scaffolds were not constructed with precise geometries. This review looks at the various advances in scaffold designs, materials and 3D printing techniques employed, which enable us to fabricate scaffolds with tailored architecture and control its functionality. These advances allow us to optimize the parameters in the fabrication of these scaffolds, such that we can create structures with desired characteristics like controllable biodegradability using the least amount of material possible along with a precise pore size and high permeability for purposes of osteo-induction and osseointegration. This is followed by the computational analysis carried out on the samples to ensure they are suitable for a given application. Finally, this paper talks about regulatory hurdles (ethical, legal and social) faced by researchers, in addition to the various technical challenges, when it comes to the real-life implementation of this technology.

Abstract:Bio-scaffolds are synthetic entities widely employed in bone and soft-tissue regeneration applications. These bio-scaffolds are applied to the defect site to provide support and favor cell attachment and growth, thereby enhancing the regeneration of the defective site. The progressive research in bio-scaffold fabrication has led to identification of biocompatible and mechanically stable materials. The difficulties in obtaining grafts and expenditure incurred in the transplantation procedures have also been overcome by the implantation of bio-scaffolds. Drugs, cells, growth factors, and biomolecules can be embedded with bio-scaffolds to provide localized treatments. The right choice of materials and fabrication approaches can help in developing bio-scaffolds with required properties. This review mostly focuses on the available materials and bio-scaffold techniques for bone and soft-tissue regeneration application. The first part of this review gives insight into the various classes of biomaterials involved in bio-scaffold fabrication followed by design and simulation techniques. The latter discusses the various additive, subtractive, hybrid, and other improved techniques involved in the development of bio-scaffolds for bone regeneration applications. Techniques involving multimaterial printing and multidimensional printing have also been briefly discussed.

Abstract:Nature does nothing in vain. Through millions of years of revolution, living organisms have evolved hierarchical and anisotropic structures to maximize their survival in complex and dynamic environments. Many of these structures are intrinsically heterogeneous and often with functional gradient distributions. Understanding the convergent and divergent gradient designs in the natural material systems may lead to a new paradigm shift in the development of next-generation high-performance bio-/nano-materials and devices that are critically needed in energy, environmental remediation, and biomedical fields. Herein, we review the basic design principles and highlight some of the prominent examples of gradient biological materials/structures discovered over the past few decades. Interestingly, despite the anisotropic features in one direction (i.e., in terms of gradient compositions and properties), these natural structures retain certain levels of symmetry, including point symmetry, axial symmetry, mirror symmetry, and 3D symmetry. We further demonstrate the state-of-the-art fabrication techniques and procedures in making the biomimetic counterparts. Some prototypes showcase optimized properties surpassing those seen in the biological model systems. Finally, we summarize the latest applications of these synthetic functional gradient materials and structures in robotics, biomedical, energy, and environmental fields, along with their future perspectives. This review may stimulate scientists, engineers, and inventors to explore this emerging and disruptive research methodology and endeavors.
A biomimetic cartilage gradient hybrid scaffold for functional tissue engineering of cartilage

Abstract:<p>Osteochondral tissue has a complex layered structure that is not self-repairing after a cartilage defect. Therefore, constructing a biomimetic gradient scaffold that meets the specific structural requirements of osteochondral tissue is a major challenge in the field of cartilage tissue engineering. In this study, chitosan/Sodium β-glycerophosphate/Gelatin (Cs/GP/Gel) biomimetic gradient scaffolds were prepared by regulating the mass ratio of single layer raw materials. The same ratio of Cs/GP/Gel hybrid scaffold material was used as the control. Physical properties such as water absorption, porosity and the degradation rate of the material were compared to optimize the proportion of scaffold materials. P3 Bone Mesenchymal Stem Cells (BMSCs) were inoculated on the gradient and the control scaffolds to investigate its biocompatibility. Scanning electron microscopy (SEM) results show that 3:1:2, 6:1:3.5, 9:1:5, 12:1:6.5, 15:1:8 Cs/GP/Gel gradient scaffolds had excellent three-dimensional porous structures. Channels were also shown to have been interconnected, and the walls of the pores were folded. In the longitudinal dimension, gradient scaffolds had an obvious stratified structure and pore gradient gradualism, that effectively simulated the natural physiological stratified structure of real cartilage. The diameter of the pores in the control scaffold was uniform and without any pore gradient. Gradient scaffolds had good water absorption (584.24 ± 3.79˜677.47 ± 1.70%), porosity (86.34 ± 5.10˜95.20 ± 2.86%) and degradation (86.09 ± 2.46˜92.48 ± 3.86%). After considering the physical properties assessed, the Cs/GP/Gel gradient scaffold with a ratio of 9:1:5 was found to be the most suitable material to support osteochondral tissue. BMSCs were subsequently inoculated on the proportional gradient and hybrid scaffolds culture. These cells survived, distributed and extended well on the gradient and hybrid scaffold material. The biomimetic gradient scaffold designed and prepared in this study provides an important foundation for the development of new gradient composite biomedical materials for osteochondral repair.</p>

Abstract:Cell migration participates in a variety of physiological and pathological processes such as embryonic development, cancer metastasis, blood vessel formation and remoulding, tissue regeneration, immune surveillance and inflammation. The cells specifically migrate to destiny sites induced by the gradually varying concentration (gradient) of soluble signal factors and the ligands bound with the extracellular matrix in the body during a wound healing process. Therefore, regulation of the cell migration behaviours is of paramount importance in regenerative medicine. One important way is to create a microenvironment that mimics the in vivo cellular and tissue complexity by incorporating physical, chemical and biological signal gradients into engineered biomaterials. In this review, the gradients existing in vivo and their influences on cell migration are briefly described. Recent developments in the fabrication of gradient biomaterials for controlling cellular behaviours, especially the cell migration, are summarized, highlighting the importance of the intrinsic driving mechanism for tissue regeneration and the design principle of complicated and advanced tissue regenerative materials. The potential uses of the gradient biomaterials in regenerative medicine are introduced. The current and future trends in gradient biomaterials and programmed cell migration in terms of the long-term goals of tissue regeneration are prospected.

Abstract:Osteochondral defects are the main cause of joint morbidity and disability in elderly patients, and osteochondral tissue engineering is one of the methods to repair osteochondral defects. The method of osteochondral tissue engineering involves the manufacture of osteochondral biomimetic gradient scaffolds that should mimic the physiological properties of natural osteochondral tissue (e. g. , the gradient transition between cartilage surface and subchondral bone). The osteochondral biomimetic gradient scaffolds exhibit discrete gradients or continuous gradients to establish the characteristics of osteochondral tissue in many studies, such as biochemical composition, structure and mechanical properties. An advantage of the continuous osteochondral biomimetic gradient scaffold is that there is no obvious interface between each layer, therefore it more closely mimics the natural osteochondral tissue. Although promising results have been achieved so far on the regeneration of the osteochondral biomimetic gradient scaffold, there are still differences between the osteochondral biomimetic gradient scaffold and natural osteochondral tissue. Due to these differences, the current clinical treatment of osteochondral biomimetic gradient scaffolds to repair osteochondral defects needs further research. Firstly, the research progress on discrete and continuous gradient scaffolds from the background of osteochondral defects, the micro-scale structure and mechanical properties of osteochondral to the materials and methods related to the manufacture of osteochondral biomimetic gradient scaffolds was summarized in this article. Secondly, due to the 3D printing method of the osteochondral biomimetic gradient scaffold having the ability to precisely control the geometry of the scaffold hole and the mechanical properties of the scaffold,the application of computational simulation models in osteochondral tissue engineering was further introduced, for example, optimizing scaffold structure and mechanical properties are considered to predict tissue regeneration. Finally, the challenges related to the repair of osteochondral defects and prospects for the future research of osteochondral tissue regeneration were presented. For example, continuous osteochondral bionic gradient scaffolds need to more similarly simulate the structure of natural osteochondral tissue units, that is, the transition of mechanical properties and biochemical properties is more smooth naturally. At the same time, although most osteochondral biomimetic gradient scaffolds have achieved good results in in vivo and in vitro experiments, clinical research and application still need to be further studied.

Abstract:Purpose – The purpose of this paper is to investigate the possibility to construct tissue-engineered bone repair scaffolds with pore size distributions using rapid prototyping techniques. Design/methodology/approach – The fabrication of porous scaffolds with complex porous architectures represents a major challenge in tissue engineering and the design aspects to mimic complex pore shape as well as spatial distribution of pore sizes of natural hard tissue remain unexplored. In this context, this work aims to evaluate the three-dimensional printing process to study its potential for scaffold fabrication as well as some innovative design of homogeneously porous or gradient porous scaffolds is described and such design has wider implication in the field of bone tissue engineering. Findings – The present work discusses biomedically relevant various design strategies with spatial/radial gradient in pore sizes as well as with different pore sizes and with different pore geometries. Originality/value – One of the important implications of the proposed novel design scheme would be the development of porous bioactive/biodegradable composites with gradient pore size, porosity, composition and with spatially distributed biochemical stimuli so that stem cells loaded into scaffolds would develop into complex tissues such as those at the bone–cartilage interface.

Abstract:A facile method is described herein for generating a mineral gradient in a biodegradable polymer scaffold. The gradient is achieved by swelling a composite film made of polycaprolactone (PCL) and hydroxyapatite (HAp) nanoparticles with a PCL solution. During the swelling process, the solvent and PCL polymer chains diffuse into the composite film, generating a gradient in HAp density at their interface. The thickness of the mineral gradient can be tuned by varying the extent of swelling to match the length scale of the natural tendon-to-bone attachment (20−60 µm). When patterned with an array of funnel-shaped channels, the mineral gradient presents stem cells with spatial gradations in both biochemical cues (e.g., osteoinductivity and conductivity associated with the HAp nanoparticles) and mechanical cues (e.g., substrate stiffness) to stimulate their differentiation into a graded distribution of cell phenotypes. This new class of biomimetic scaffolds holds great promise for facilitating the regeneration of the injured tendon-to-bone attachment by stimulating the formation of a functionally graded interface.

Abstract:IMPACT STATEMENT: Challenges in musculoskeletal tissue regeneration affect millions of patients globally. Scaffolds for tissue engineering bone and cartilage provide promising solutions that increase healing and decrease need for complicated surgical procedures. Porous scaffolds have emerged as an attractive alternative to traditional scaffolds. However, the success of advanced materials, use of biological factors, and manufacturing techniques can vary depending on use case. This review provides perspective on porous scaffold manufacturing, characterization and application, and can be used to inform future scaffold design.

Abstract:Large bone defect regeneration has always been recognized as a challenging clinical problem due to the difficulty of revascu-larization.Conventional treatments exhibit certain inherent disadvantages(e.g.,secondary injury,immunization,and potential infections).However,three-dimensional(3D)printing technology as an emerging field can serve as an effective approach to achieve satisfactory revascularization while making up for the above limitations.A wide variety of methods can be used to facilitate blood supply during the design of a 3D-printed scaffold.Importantly,the scaffold structure lays a foundation for the entire printing object;any method to promote angiogenesis can be effective only if it is based on well-designed scaffolds.In this review,different designs related to angiogenesis are summarized by collecting the literature from recent years.The 3D-printed scaffolds are classified into four major categories and discussed in detail,from elementary porous scaffolds to the most advanced bone-like scaffolds.Finally,structural design suggestions to achieve rapid angiogenesis are proposed by analyzing the above architectures.This review can provide a reference for organizations or individual academics to achieve improved bone defect repair and regeneration using 3D printing.

Abstract:The construction of gradient scaffolds to mimic the in vivo microenvironment for promoting tissue regeneration is attractive, especially by incorporating biochemical cues with physical guidance to manipulate cell behavior. Here, we report a general method to generate dual concentration-gradient of two types of bioactive components on scaffolds composed of uniaxially aligned electrospun nanofibers through layer-by-layer self-assembly. Different layers of collagen (COL) and chondroitin sulfate (CS) are sequentially coated onto the specific positions of uniaxially aligned electrospun polycaprolactone (PCL) nanofibers upon electrostatic attraction. In this case, dual stepwise gradients of COL and CS are generated along the fiber alignment. In demonstrating the potential application, the dual gradients can effectively promote the migration and proliferation of bone marrow stem cells (BMSCs) and spatially control their osteogenic differentiation. This simple strategy for generating dual gradients can be easily extended to different types of gradient scaffolds for bone regeneration and related applications involving cell migration, stem cell differentiation, and tissue interface.
