Abstract:Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials, but also increase the economic value. Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities. However, pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio, which seriously affected their in vivo stability and integrity when applied as tissue engineering scaffolds. Thus, simultaneously improving the mechanical strength and biological compatibility of pure chitosan (CS) scaffolds becomes very important. Here, inspired by the fiber-reinforced construction of natural extracellular matrix and the porous structure of cancellous bone, we built silk microfibers/chitosan composite scaffolds via ice-templating technique. This biomimetic strategy achieved 500% of mechanical improvement to pure chitosan, and meanwhile still maintaining high porosity (> 87%). In addition, the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation. More importantly, after subcutaneous implantation in mice for four weeks, the composite scaffold showed greater structural integrity, as well as better collagenation, angiogenesis, and osteogenesis abilities, suggesting its great potential in biomedicine.

Abstract:The collagen-chitosan complex with a three-dimensional nanofiber structure was fabricated to mimic native ECM for tissue repair and biomedical applications. Though the three-dimensional hierarchical fibrous structures of collagen-chitosan composites could provide more adequate stimulus to facilitate cell adhesion, migrate and proliferation, and thus have the potential as tissue engineering scaffolding, there are still limitations in their applications due to the insufficient mechanical properties of natural materials. Because poly (vinyl alcohol) (PVA) and thermoplastic polyurethane (TPU) as biocompatible synthetic polymers can offer excellent mechanical properties, they were introduced into the collagen-chitosan composites to fabricate the mixed collagen/chitosan/PVA fibers and a sandwich structure (collagen/chitosan-TPU-collagen/chitosan) of nanofiber in order to enhance the mechanical properties of the nanofibrous collagen-chitosan scaffold. The results showed that the tensile behavior of materials was enhanced to different degrees with the difference of collagen content in the fibers. Besides the Young's modulus had no obvious changes, both the break strength and the break elongation of materials were heightened after reinforced by PVA. For the collagen-chitosan nanofiber reinforced by TPU, both the break strength and the Young's modulus of materials were heightened in different degrees with the variety of collagen content in the fibers despite the decrease of the break elongation of materials to some extent. In vitro cell test demonstrated that the materials could provide adequate environment for cell adhesion and proliferation. All these indicated that the reinforced collagen-chitosan nanofiber could be as potential scaffold for tissue engineering according to the different mechanical requirements in clinic.

Abstract:Nanofibrous biocomposite scaffolds of chitosan (CS), PVA, and hydroxyapatite (HA) were prepared by electrospinning. The scaffolds were characterized by FTIR, SEM, TEM, and XRD techniques. Tensile testing was used for the characterization of mechanical properties. Mouse fibroblasts (L929) attachment and proliferation on the nano-fibrous scaffold were investigated by MTT assay and SEM observation. FTIR, TEM, and XRD results showed the presence of nanoHA in the scaffolds. The scaffolds have porous nanofibrous morphology with random fibers in the range of 100-700 nm diameters. The CS/PVA (90/10) fibrous matrix (Without HA) showed a tensile strength of 3.1 +/- 0.2 MPa and a tensile modulus 10 I MPa with a strain at failure of 21.1 +/- 0.6%. Increase the content of HA up to 2% increased the ultimate tensile strength and tensile modulus, but further increase HA LIP to 5-10% caused the decrease of tensile strength and tensile modulus. The attachment and growth of mouse fibroblast was on the Surface of nanofibrous structure, and cells' morphology characteristics and viability were unaffected. A combination of nanofibrous CS/PVA and HA that mimics the nanoscale features of the extra cellular matrix Could be promising for application as scaffolds for tissue regeneration, especially in low or nonload bearing areas. Abstract:Developing wound dressing that inhibits bacterial infection for treating complex wound healing processes has been a research hotspot. Here, we report the fabrication of Cu-MOFs (HKUST-1) incorporated electrospun chitosan/polyvinyl alcohol (HKUST-1/chitosan/PVA) fibers through the blending electrospinning for wound therapy. HKUST-1/chitosan/PVA fibers displayed satisfying physical properties, such as mechanical property, water uptake, water vapor transmission rate, etc. Cytotoxicity test indicated that HKUST-1/chitosan/PVA fibers were biocompatible and could support cell adhesion. Due to the HKUST-1 incorporation, HKUST-1/chitosan/PVA fibers exhibited the good antibacterial activity against Escherichia coli and Staphylococcus aureus with 99% antibacterial efficiency. Furthermore, in animal studies, compared with commercial chitosan dressings and chitosan/PVA fibers, HKUST-1/chitosan/PVA fibers were more efficient to heal the wound with less inflammation. In summary, the HKUST-1/chitosan/PVA fibers with good physicochemical property, biocompatibility and antibacterial property is an excellent wound dressing for full-thickness skin repair.

Abstract:Tissue engineering (TE) approaches often employ polymer-based scaffolds to provide support with a view to the improved regeneration of damaged tissues. The aim of this research was to develop a surface modification method for introducing chitosan as an antibacterial agent in both electrospun membranes and 3D printed poly-ε-caprolactone (PCL) scaffolds. The scaffolds were functionalized by grafting methacrylic acid N-hydroxysuccinimide ester (NHSMA) onto the surface after Ar-plasma/air activation. Subsequently, the newly-introduced NHS groups were used to couple with chitosan of various molecular weights (Mw). High Mw chitosan exhibited a better coverage of the surface as indicated by the higher N% detected by X-ray photoelectron spectroscopy (XPS) and the observations with either scanning electron microscopy (SEM)(for fibers) or Coomassie blue staining (for 3D-printed scaffolds). A lactate dehydrogenase assay (LDH) using L929 fibroblasts demonstrated the cell-adhesion and cell-viability capacity of the modified samples. The antibacterial properties against S. aureus ATCC 6538 and S. epidermidis ET13 revealed a slower bacterial growth rate on the surface of the chitosan modified scaffolds, regardless the chitosan Mw.

Abstract:Silk fibroin (SF) is increasingly needed in tissue engineering for its superior biocompatibility. However, the practical applications of pure SF biomaterials confront bacterial infection problems. In this study, chitosan (CS) and polydopamine (PDA) were introduced into electrospun nanofibrous SF mats through layer-by-layer self-assembly (LBL) to obtain enhanced antibacterial ability and cytocompatibility. The surface morphology and composition analysis confirmed the successful deposition. After depositing 15 bilayers, the tensile modulus of the mats in wet condition increased from 2.16 MPa (pristine SF mats) to 4.89 MPa. A trend towards better hydrophilicity performance was also recorded with more bilayers coating on the mats. Besides, LBL structured mats showed improved antibacterial ability of more than 98 % against E. coli and S. aureus. In addition, advancement in biocompatibility was observed during the proliferation experiment of L929 cells. Overall, the deposition of CS and PDA may further expand the use of SF in biomedical field.

Abstract:AIM: Electrospunning is a process that the charged polymer solution or melt jet efflux in the electrostatic field to prepare the polymer nanofibers. This study aimed to prepare electrospun chitosan/collagen composite nanofibers and evaluate their cell biocompatibility. METHODS: The experiment was carried out in the Laboratory of Biomaterials and Tissue Engineering, College of Chemistry, Chemical Engineering and Biotechnology at Donghua University between November 2006 and October 2007. ① Taking hexafluoroisopropanol/trifluoroacetic acid as solvent, the composite nanofibers were prepared by electrospunning according to the mass ratio of chitosan/collagen as 100:0, 80:20, 50:50, 20:80 and 0:100.②The composite nanofibers were inoculated with porcine iliac artery endothelial cells in vitro. The cell adhesion and proliferation as well as cell morphologies were evaluated by MTT method and HE stain, respectively. RESULTS: The porcine iliac artery endothelial cells adhered tightly on surface of electrospun chitosan/collagen composite nanofibers, exhibited plump and fusiform shape, as well as good growth. MTT results showed that nanofibers could obviously enhance the adhesion and proliferation of endothelial cells on the surface of materials, and the cells had the best spreading and attachment on the composite nanofibers with a ratio of 20:80, then 50:50. CONCLUSION: The electrospun chitosan/collagen composite nanofibers hold great potential for tissue engineering application due to their good cell biocompatibility.

Abstract:A series of nanofibrous scaffolds were prepared by electrospinning of poly(vinyl alcohol) (PVA)/gelatin aqueous solution. PVA and gelatin was dissolved in pure water and blended in full range, then being electrospun to prepared nanofibers, followed by being crosslinked with glutaraldehyde vapor and heat treatment to form nanofibrous scaffold. Field emission scanning electron microscope (FESEM) images of the nanofibers manifested that the fiber average diameters decreased from 290 to 90 nm with the increasing of gelatin. I n vitro degradation rates of the nanofibers were also correlated with the composition and physical properties of electrospinning solutions. Cytocompatibility of the scaffolds was evaluated by cells morphology and MTT assay. The FESEM images revealed that NIH 3T3 fibroblasts spread and elongated actively on the scaffolds with spindle‐like and star‐type shape. The results of cell attachment and proliferation on the nanofibrous scaffolds suggested that the cytotoxicity of all samples are grade 1 or grade 0, indicating that the material had sound biosafety as biomaterials. Compared with pure PVA and gelatin scaffolds, the hybrid ones possess improved biocompatibility and controllability. Abstract:Hydrogels have great potential for biomedical applications. However, there are still significant challenges in preparing biomass-based hydrogels with excellent adhesive, antibacterial and biocompatible properties. In this study, pure physically cross-linked tannic acid (TA)/polyvinyl alcohol (PVA)/2-hydroxypropyltrimethyl ammonium chloride chitosan (HACC) double-network (PVA/HACC-TA) hydrogels are prepared by a combination approach of cyclic freeze-thaw and soaking method. The prepared PVA/HACC-TA hydrogels are formed by hydrogen bond cross-linking and do not use any toxic and harmful cross-linking agents. Meanwhile, the prepared PVA/HACC-TA hydrogels have excellent mechanical properties, the maximum tensile strength can reach 4.13 MPa and the maximum tensile strain can attain 830%. In addition, PVA/HACC-TA hydrogels display good adhesion to different materials. At the same time, the PVA/HACC-TA hydrogels also show favorable antibacterial activities. Therefore, the excellent mechanical, adhesive, antibacterial and cytocompatible biomass hydrogels showed promising potential applications in the biomedicine field, especially as the skin wound dressings.

Abstract:In this study, orthogonal experiments were designed to explore the optimal process parameters for preparing polycaprolactone(PCL) scaffolds by the near-field direct-writing melt electro-spinning(NFDWMES) technology. Based on the optimal process parameters, the PCL scaffolds with different thicknesses, gaps and structures were manufactured and the corresponding hydrophilicities were characterized. The PCL scaffolds were modified by chitosan (CS) and hyaluronic acid(HA) to improve biocompatibility and hydrophilicity. Both Fourier transform infrared spectroscopy(FTIR) analysis and antibacterial experimental results show that the chitosan and hyaluronic acid adhere to the surface of PCL scaffolds, suggesting that the modification plays a positive role in biocompatibility and antibacterial effect. The PCL scaffolds were then employed as a carrier to culture cells. The morphology and distribution of the cells observed by a fluorescence microscope demonstrate that the modified PCL scaffolds have good biocompatibility, and the porous structure of the scaffolds is conducive to adhesion and deep growth of cells.

Abstract:In this paper, polyvinyl alcohol/Ag-Metal-organic framework (PVA/Ag@MOF) and polyvinyl alcohol/chitosan (PVA/CS) were used as the inner and outer layers to successfully prepare a bilayer composite hydrogel for tissue engineering scaffold. The performance of bilayer hydrogels was evaluated. The outer layer (PVA/CS) has a uniform pore size distribution, good water retention, biocompatibility and cell adhesion ability. The inner layer (PVA/Ag@MOF) has good antibacterial activity and poor biocompatibility. PVA, PVA/0.1%Ag@MOF, PVA/0.5%Ag@MOF, and PVA/1.0%Ag@MOF show anti-microbial activity in ascending order. However, its use as an inner layer avoids direct contact with cells and prevents infection. The cell viability of all samples was above 90%, indicating that the bilayer hydrogel was non-toxic to A549 cells. The bilayer hydrogel scaffold combines the advantages of the inner and outer layers. In summary, this new bilayer composite is an ideal lung scaffold for tissue engineering.

Novel Biodegradable Films and Scaffolds of Chitosan Blended with Poly(3-Hydroxybutyrate).

Abstract:In order to develop a novel biomaterial, films of chitosan blended with poly(3-hydroxybutyrate) (PHB) were prepared by an emulsion blending technique and their properties were characterized. Scanning electron microscopy (SEM) showed that PHB microspheres were formed and were entrapped in chitosan matrices, which made the film surface rough. With increasing PHB content, the roughness of the film surface increased, while the swelling capability of the films decreased. In a wet state, the blended films exhibited a lower elastic modulus, a higher elongation-at-break and a higher tensile strength compared with chitosan films. Cell-culture experiments revealed that the blended films had better cytocompatibility than chitosan films. To explore the potential application of the blended material in tissue engineering, the porous blended scaffolds were fabricated and their pore morphology was observed by SEM. The results revealed that not only pore structure but also pore wall morphology of the blended scaffolds could be controlled by selecting the parameters of the fabrication process. These advantageous properties indicate that the blended chitosan/PHB material is promising for tissue engineering applications.

Abstract:Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV)/chitin nanocrystals (CNC) composite scaffolds were synthesized by the salt leaching and thermally induced phase separation (TIPS) technique. The scaffolds have porous structures with macro-pores (100-300 mu m in diameters) and micro-pores (10 mu m). The surface characteristics of the scaffolds were characterized by X-ray photoelectron spectroscopy (XPS) and static water contact angle measurement, and the mechanical properties were investigated by a compression test. Human adipose-derived stem cells (hADSCs) were seeded onto the PHBV/CNC scaffolds and in vitro cell culture results showed that the composite scaffolds enhanced the hADSCs adhesion, which implies that the material may have potential application in tissue engineering. Abstract:In the title compound, C14H10F3N7·2CH4O, the heterocyclic ring system is essentially planar (r.m.s. deviation = 0.009 Å) and makes a dihedral angle of 6.91 (8)° with the attached benzene ring. In the crystal, the main molecules form centrosymmetric R 2 2(8) dimers via pairs of N—H⋯N hydrogen bonds between the amino groups and pyrimidine N atoms. One of the independent methanol molecules and its inversion equivalent are linked to the dimers via O—H⋯N and N—H⋯O hydrogen bonds, forming R 4 4(16) graph-set motifs. The dimers along with the hydrogen-bonded methanol molecules are stacked along the a axis, with π–π interactions between the pyrazole and triazole rings [centroid–centroid distance = 3.4953 (10) Å].

Abstract:Chitosan, a cationic polysaccharide, exhibits promising potential for tissue engineering applications. However, the poor mechanical properties and rapid biodegradation have been the major limitations for its applications. In this work, an effective strategy was proposed to optimize the mechanical performance and degradation rate of chitosan gel scaffolds by regulating the water content. Physical chitosan hydrogel (HG, with 93.57 % water) was prepared by temperature-controlled cross-linking, followed by dehydration to obtain xerogel (XG, with 2.84 % water) and rehydration to produce wet gel (WG, with 56.06 % water). During this process, changes of water content significantly influenced the water existence state, hydrogen bonding, and the chain entanglements of chitosan in the gel network. The mechanical compression results showed that the chitosan gel scaffolds exhibited tunable compressive strength (0.3128–139 MPa) and compressive modulus (0.2408–1094 MPa). XG could support weights exceeding 65,000 times its own mass while maintaining structural stability. Furthermore, in vitro and in vivo experiments demonstrated that XG and WG exhibited better biocompatibility and resistance to biodegradation compared with HG. Overall, this work contributes to the design and optimization of chitosan scaffolds without additional chemical crosslinkers, which has potential in tissue engineering and further clinical translation.

Abstract:Chitosan-based porous scaffolds are of great interest in biomedical applications because of their biodegradability and biocompatibility. However, the poor mechanical properties of these scaffolds hinder their broad utility. In the present study, a novel compression method was developed to fabricate chitosan scaffolds with high mechanical strength and tuneable topography, based on the ionic strength and pH-dependent solubility of chitosan. When the compressive ratio increases from 1 to 8, the compressive elastic modulus of the scaffold increases from 5.2kPa to 520kPa and the porosity decreases from 94.1% to 82.5%. Furthermore, the number of human adipose-derived stem cells adhering to the scaffolds increases as the compressive ratio increases, owing to the high density of the chitosan fibres. This method does not require external cross-linker agent, sophisticated instrumentation and/or technical proficiency and could be extended to other polysaccharides.
Fabrication of Antimicrobial Multilayered Nanofibrous Scaffolds-Loaded Drug via Electrospinning for Biomedical Application

Abstract:Nanofibers prepared by biobased materials are widely used in the field of biomedicine, owing to outstanding biocompatibility, biodegradable characters, and excellent mechanical behavior. Herein, we fabricated multilayered nanofibrous scaffolds in order to improve the performance of drug delivery. The composite layer-by-layer scaffolds were incorporated by hydrophobic poly( l -lactic acid) (PLA): polycaprolactone (PCL) and hydrophilic poly(vinyl alcohol) (PVA) nanofibers via multilayer electrospinning. Morphological and structural characteristics of the developed scaffolds measured by scanning electron microscopy (SEM), and transmission electron microscopy (TEM) confirmed smooth and uniform fibers ranging in nanometer scale. The differences in contact angles and Fourier transform infrared spectrum (FTIR) between single-layered PVA nanofibers and multilayered scaffolds verified the existence of PLA: PCL surface. In vitro biodegradable and drug release analysis depicted multilayered scaffolds had good biodegradability and potential for medical application. Due to the model drug incorporation, scaffolds exhibited good antibacterial activity against Escherichia coli and Staphylococcus aureus by the zone of inhibition test. These results revealed that the multilayered scaffolds were proved to be desirable antibacterial materials for biomedical application.

Abstract:Synthesis and characterization of nanofiber scaffold for biomedical applications. The present study aims to fabricate cellulose nanocrystals (CNCs) doped poly(vinyl alcohol) polyethylene glycol (PVA/PEG) nanocomposite films for biomedical applications. The nanocomposite films were prepared by using electrospinning techniques with varying percentages of CNCs (10% and 20%) mixed with PVA and PEG (50:50). The properties of the produced nanocomposites were evaluated using a battery of tests. The results of surface morphology and structural miscibility obtained from field emission‐scanning electron microscopy (FE‐SEM), atomic force microscopy, and Fourier transformer infra‐red studies revealed appreciable compatibility among the three components (CNCs, PVA, and PEG). FE‐SEM micrographs revealed that the nanomaterials underwent a dramatic transition from discontinuous to continuous fibers, with interconnected or network‐like fibers. The constructed scaffold material's stability went up from three to four phases, according to thermal and degradation experiments. In addition, research on biodegradation and water absorption found that CNCs outperformed pure PVA and PVA/PEG in terms of swelling percentage time and enzyme degradation rate. The hemolysis assay of the PVA/PEG/CNC scaffold showed good compatibility (below 5%) and according to the MTT assay, both NIH 3T3 and L929 cells survived well, indicating that the CNCs could serve as a useful scaffold for tissue engineering applications such skin tissue regeneration. In sum, the findings proved that biological domains including tissue engineering and wound healing can benefit from produced nanofiber scaffolds.
