Abstract:Introduction Cortisol, as a steroid hormone secreted by the human body, is a typical stress marker. It plays an important role in regulating blood pressure, blood sugar, carbohydrate metabolism and other physiological processes. Among them, measuring saliva cortisol seems to be advantageous because it is a relatively easy and non-invasive method to extract saliva samples, and saliva can be conveniently processed and stored. Under normal circumstances, the body can well control the secretion of cortisol and regulate the content of cortisol in the blood. However, under greater physical and psychological stress, a high level of cortisol would be induced and kept for a long time, which will lead to the blood sugar, increased appetite, increased weight, and extreme fatigue, etc. The traditional methods for cortisol detection have biochemical analysis, radioimmunoassays, which is based on large-scale equipment and difficult to real-time monitoring. Therefore, the development of rapid, on-situ and sensitive cortisol detection products has important national defense strategic significance. In this study, the Cu-MOF catalyst integrated-antibody was used for cortisol in-situ colorimetric immunoassay based on the smartphone-based portable electric eye (E-eye) (Fig.1). As a metal-organic framework, Cu-MOF can conjugate with antibody without complicated operations, and then specifically recognize cortisol. As a catalase, Cu-MOF can not only catalyze the coloration of TMB, but also amplify the signal, and also protect the antibody from long-term high temperature. Based on the indirect competition method, the cortisol in the samples competes with the cortisol-BSA coated on the microtiter plate to bind the cortisol antibody. And then the conjugation of MOF-antibody (Cu-MOF@IgG) will bind with the antibody, which could catalyze the H2O2 and TMB substrates to blue color and amplify the signal, which could be quantitative recognition by portable E-eye. The degree of blue is inversely proportional to the cortisol content. Based on the portable E-eye, the proposed method processes the features of rapid, simple, high sensitivity and high throughout, which could achieve cortisol in-situ detection in practical applications. Preparation of Cu-MOF The Cu-MOF was fabricated by mixing 7.8mM CuCl2 aqueous and 31.2mM 4,4’-bipyridine equally with 2h gentle stirring under room temperature. The blue precipitates were obtained by centrifugation and washed by ultrapure water. Then it was re-dissolved in 500µl pure water for further use. The catalytic activity of Cu-MOF was verified by adding Cu-MOF to the H2O2 and TMB mixture. Under the successful preparation of Cu-MOF, the mixed solution will turn blue due to the catalase activity of Cu-MOF. The pH of the system was adjusted by NaAC/HAc buffer. The morphology (Fig.2(a) and (b)) and the catalase activity (Fig.2(c)) of Cu-MOF materials were characterized by transmission electron microscope and microplate reader. Preparation of Cu-MOF@IgG The Cu-MOF@IgG materials were formed via the following two steps: (1) 100µl of 20mg/L IgG was mixed with the 2mL of 7.8mM CuCl2 aqueous at 4 ℃ for 10 min. (2) Then 2mL of 31.2mM 4,4’-bipyridine was added to the above mixture with 12h continuously stirring at 4 ℃. Also, the 4,4’-bipyridine above was prepared by dissolving 0.0487g of 4,4’-bipyridine powder in 10 mL ultrapure and water mixture with equal volume. (3) The product obtained from the above reaction was thoroughly centrifuged and washed thrice with ultrapure water, and then re-dissolved in 500ul pure water. The prepared Cu-MOF@IgG are stored at 4 ℃ for further use. The verification of the combination of Cu-MOF and IgG was as follows: First, it has been verified that the material of Cu-MOF has no absorption to the microtiter plate (Fig. 2(d)). The obtained Cu-MOF@IgG solution was diluted with carbonate buffer, and then coated on a microtiter plate. After incubating for 12 h at 4℃, the microtiter plate was washed by PBST (0.01 M PBS + 0.01% Tween) four times to remove excess free Cu-MOF and IgG. Then the mixture of H2O2 and TMB was added to the coated microtiter plate. After incubating of 20 min at 37℃, the color of the solution would be reserved for the successful preparation of Cu-MOF@IgG (Fig.2(e)). Cortisol detection based on Cu-MOF@IgG The cortisol-BSA was diluted to 2.7μg/mL with carbonate buffer (pH = 9.6), and then coated on a microtiter plate with a volume of 100μL per well. After incubating overnight at 4°C, the coated plate was washed with PBST four times to remove the excess free primary antigen. Then the free binding sites were sealed with 2% BSA solution for 2h at 37℃ to prevent the nonspecific binding. After washing with PBST four times, the cortisol standard and cortisol MAb was added to the plate and incubate at 37°C for 1 h. Then 100µl of Cu-MOF@IgG was added to binding with the coated cortisol MAb. After incubating at 37℃ for 2h, the unbound Cu-MOF@IgG was removed by washing with PBST four times. Then the mixture of H2O2 and TMB was added to wells and incubate for 20 min at 37°C. Finally, the absorbance of wells at 650nm was detected by the microplate reader. Also, the color changes could be detected by the homemade portable E-eye. As shown in Fig. 2(f), the detection limit of cortisol in the saliva is 0.12µg/L with the detection range of 0.88-1000µg/L. Abstract:Cortisol is commonly used as a significant biomarker of psychological or physical stress. With the accelerated pace of life, non-invasive cortisol detection at the point of care (POC) is in high demand for personal health monitoring. In this paper, an ultrasensitive immunosensor using gold nanoparticles/molybdenum disulfide/gold nanoparticles (AuNPs/MoS2/AuNPs) as transducer was explored for non-invasive salivary cortisol monitoring at POC with the miniaturized differential pulse voltammetry (DPV) system based on a smartphone. Covalent binding of cortisol antibody (CORT-Ab) onto the AuNPs/MoS2/AuNPs transducer was achieved through the self-assembled monolayer of specially designed polyethylene glycol (PEG, SH-PEG-COOH). Non-specific binding was avoided by passivating the surface with ethanolamine. The miniaturized portable DPV system was utilized for human salivary cortisol detection. A series current response of different cortisol concentrations decreased and exhibited a linear range of 0.5–200 nM, the detection limit of 0.11 nM, and high sensitivity of 30 μA M−1 with a regression coefficient of 0.9947. Cortisol was also distinguished successfully from the other substances in saliva. The recovery ratio of spiked human salivary cortisol and the variation of salivary cortisol level during one day indicated the practicability of the immunosensor based on the portable system. The results demonstrated the excellent performance of the smartphone-based immunosensor system and its great potential application for non-invasive human salivary cortisol detection at POC.

Abstract:A novel electrochemical sensor was developed for the detection of salivary cortisol levels. The sensor employs a combination of a molecularly imprinted polymer (MIP) and gold nanoparticles (AuNPs) that are electrodeposited onto a screen-printed electrode (SPE). The study utilised density functional theory and molecular docking techniques to determine the geometry of molecular orbitals, electrostatic potential energies, and binding energy of cortisol and the polymers. The thin film of cortisol-imprinted polymer on the SPE was created by electro-polymerizing pyrrole and thiophene-3-carboxylic acid on the electrode surface along with cortisol as the template molecule. The MIP film was characterised using scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), and electrochemical techniques. The sensor exhibited a linear response in the concentration range of 0.05 nmol L −1 to 2.5 μmol L −1 , with a limit of detection of 0.01 nmol L −1 , as determined by differential pulse voltammetry. This method offers a simple yet efficient and sensitive approach to detecting cortisol levels in human saliva samples.

Electropolymerized Molecularly Imprinted Polymer Synthesis Guided by an Integrated Data-Driven Framework for Cortisol Detection.

Abstract:Molecularly imprinted polymers (MIPs), often called "synthetic antibodies", are highly attractive as artificial receptors with tailored biomolecular recognition to construct biosensors. Electropolymerization is a fast and facile method to directly synthesize MIP sensing elements in situ on the working electrode, enabling ultra-low-cost and easy-to-manufacture electrochemical biosensors. However, due to the high dimensional design space of electropolymerized MIPs (e-MIPs), the development of e-MIPs is challenging and lengthy based on trial and error without proper guidelines. Leveraging machine learning techniques in building the quantitative relationship between synthesis parameters and corresponding sensing performance, e-MIPs' development and optimization can be facilitated. We herein demonstrate a case study on the synthesis of cortisol-imprinted polypyrrole for cortisol detection, where e-MIPs are fabricated with 72 sets of synthesis parameters with replicates. Their sensing performances are measured using a 12-channel potentiostat to construct the subsequent data-driven framework. The Gaussian process (GP) is employed as the mainstay of the integrated framework, which can account for various uncertainties in the synthesis and measurements. The Sobol index-based global sensitivity is then performed upon the GP surrogate model to elucidate the impact of e-MIPs' synthesis parameters on sensing performance and interrelations among parameters. Based on the prediction of the established GP model and local sensitivity analysis, synthesis parameters are optimized and validated by experiment, which leads to remarkable sensing performance enhancement (1.5-fold increase in sensitivity). The proposed framework is novel in biosensor development, which is expandable and also generally applicable to the development of other sensing materials.

Abstract:Owing to their potential to transform traditional medical diagnostics and health monitoring, wearable biosensors have become an alternative evolutionary technology in the field of medical care. However, it is still necessary to overcome some key technique challenges, such as the selectivity, sensitivity, and stability of biometric identification. Herein, a novel, wearable electrochemical sensor based on a molecularly imprinted polymer (MIP) integrated with a copper benzene-1,3,5-tricarboxylate metal-organic framework (MOF) was designed for the detection of stress through the on-body monitoring of cortisol in sweat. The MOF was used as the substrate for MIP deposition to enhance the stability and sensitivity of the sensor. The sensor consisted of two layers, with a microfluidic layer as the top layer for spontaneous sweating and a modified electrode as the bottom layer for sensing. The sensor measured cortisol levels by detecting the current change that occurred when the target molecules bound to the imprinted cavities, using Prussian blue nanoparticles embedded in the MIP framework as the REDOX probe. The proposed sensor exhibited a linear detection range of 0.01-1000 nM with a detection limit of 0.0027 nM, and favorable specificity over other analogies. This facile anti-body free sensor showed excellent stability, and can be successfully applied for in situ cortisol monitoring.

Abstract:Abstract. Knowledge of the impact of potential sources of error in spatial frequency domain imaging (SFDI) is essential for the quantitative characterization of absorption and scattering in tissue and other turbid media. We theoretically investigate the error in the derived absorption and scattering parameter, subject to typical experimental and theoretical sources of errors. This provides a guideline to properly assess the significance of various parameters related to the measurement and the theoretical evaluation of spatial frequency domain reflectance data. At the same time, this study serves as a reference to estimate the overall precision of derived optical parameters of semi-infinite scattering media using SFDI.

Abstract:The cortisol in human body is a crucial biomarker in terms of wellness management, mental state monitoring and stress-related disorder diagnosis. Therefore, the rapid, reliable and facile measurement of cortisol concentration has attracted extensive research interest. However, traditional cortisol detection such as immunosensing requires demanding laboratory layout, lengthy procedures and high costs, which means, consequently, it is incompatible with the current goal of cortisol sensing. Given the contradiction, an electrochemical sensor based on molecularly imprinted polymer (MIP) for simple, efficient, non-invasive cortisol detection was proposed. The two-step approach employed is simple enough and allows for the mass production of devices. And the embedding of Prussian Blue (PB) within the MIP layer eliminates the need for complex external probes, thereby making the resultant sensors more suitable for integration into wearable devices. We firstly demonstrated the feasibility of the proposed strategy and characterized the successful formation of cavities specific to cortisol molecules. Thereafter, we measured the dependence of the current response on cortisol concentration in Phosphate Buffered Saline (PBS) buffer, which revealed a near-linear relationship between the logarithm of the cortisol concentration and the redox current from 10−9 mol/L to 10−5 mol/L, covering the optimal range of cortisol concentration in sweat. Subsequently, sensors with the same specifications were prepared and tested in PBS buffer, exhibiting good consistency. In artificial sweat, we further demonstrated that they have benign selectivity, interference immunity and great potential in practical applications.
Quinoline antifolate thymidylate synthase inhibitors: variation of the C2- and C4-substituents.

Abstract:Modifications to the bicyclic ring system of the potent thymidylate synthase (TS) inhibitor N-[4-[N-[(2-amino-3,4-dihydro-4-oxo-6- quinazolinyl)methyl]-N-prop-2-ynylamino]benzoyl]-L-glutamic acid (1, CB3717) have led to the synthesis of a series of quinoline antifolates bearing a variety of substituents at the C2 and C4 positions. In general the synthetic route involved the coupling of the appropriate diethyl N-[4-(prop-2-ynylamino)benzoyl]-L-glutamate with a disubstituted 6-(bromomethyl)quinoline followed by deprotection using mild alkali. The compounds were tested as inhibitors of partially purified L1210 TS. As a measure of cytotoxicity, the compounds were tested for their inhibition of the growth of L1210 cells in culture. Good enzyme inhibition and cytotoxicity were found for compounds containing chloro, amino, or methyl substituents at the C2 position with chloro or bromo substituents at C4. The effect on enzyme inhibition of varying the N10 substituent of 2h was similar to that observed in the quinazolinone-containing antifolates, indicating that the quinoline compounds may be interacting with the enzyme in a similar way to the quinazolinones. Also, the introduction of a 2'-fluoro substituent into the benzoyl ring of several of the quinoline antifolates led to an increase in both TS inhibition and the inhibition of L1210 cell growth. These data demonstrate that the N3-H of the pyrimidine ring of the quinazolinone antifolates is not required for binding to TS if appropriate substituents are placed at the C2 and C4 positions of the bicyclic ring system.

Abstract:High entropy oxides (HEOs) are a class of materials that have garnered significant attention in recent years due to their potential use as catalysts in energy-related applications[1]. These materials are derived from high entropy compounds, which consist of equimolar or near equimolar ratios of multiple primary elements and have configurational entropy in the formation of a single-phase structure. The cocktail effect of HEOs gives rise to various catalytically active sites due to the synergistic interaction of the different elements, and lattice distortion during the formation of a single-phase solid solution results in the generation of abundant oxygen vacancies, which enhances the adsorption activity of HEOs and improves their catalytic performance[2]. HEOs can also be cost-efficient by utilizing low-cost transition metal elements as their active centers. Despite their widespread use as catalysts, the application of HEOs in electrocatalysis for molecule monitoring and early diagnosis remains largely unexplored. Cortisol, a glucocorticoid hormone that regulates many aspects of metabolism, stress, and immune response, is closely related to various diseases and disorders[3]. Current methods for noninvasive monitoring of cortisol relies upon immunological assays and typically limited to labor-intensive and time-consuming laboratory techniques. As a consequence, detecting cortisol in a label-free manner that is low cost and offers significant sensor stability and high sensitivity is desirable for many emerging applications, particularly for on-body monitoring. Here, we present work that examines the catalytic activities of HEOs as nanozymes for the electrocatalysis of cortisol. The electrochemical cortisol sensor is based on a novel one-pot microwave-assisted reaction synthesized (Fe, Co, Mn, Ni, Cu) Ox. The catalytic mechanism of electrochemical sensors depends on the formation of various valence metal redox species pairs, where the higher valence state oxidizes cortisol species present on the electrode surface, accompanied by a reduced lower valence state. The one-pot microwave-assisted synthesized HEOs exhibited ultra-high catalytic activity towards cortisol species as measured by cyclic voltammetry and chronoamperometry. Experimental results show a superior sensing performance including a detection limit as low as 0.5 μM (s/N=3), a good linear detection range between 0.01 ~ 4.01 μM, and a sensitivity of 46.5 μA μM-1 cm-2. Factors leading to the U . S . housing bubble : a structural equation modeling approach

Abstract:For the past decade, academics and practitioners have debated the existence of a housing bubble. Given the sharp declines in the housing market and the financial crisis, there is little doubt that a bubble occurred and then burst. Nevertheless, an important research question remains, what factors contributed to the creation of the bubble? This research addresses this issue by selecting well understood factors that traditionally drive the housing market and constructing a regression model to investigate the nature of the relationships. Because of the co-dependence of many of the factors, structural equation modeling (SEM) is used rather than traditional regression analysis. Using this technique addresses the difficulties presented by the high levels of multi-co-linearity present in many of the factors. Because all the variables used in our models are observable rather than latent, measurement model issues typical in most SEM analyses are not a concern. Further, there is no question that values dropped anywhere from a modest ten percent in sTable markets to fifty percent in markets that were overheated. With large numbers of home owners still experiencing economic distress, more homes will undoubtedly be liquidated at below purchase prices, putting further downward pressure on housing prices. With hindsight, it is easy to conclude that housing prices have generally plummeted from lofty values, and therefore, a housing bubble must have occurred. However, the problem is more complex than that. Questions still remain as what factors created the housing bubble. Further, the answers to these questions lend insight into dynamics of one the most important consumer sectors of our economy. The authors of this paper will attempt to shed some light on these issues, using empirical data and a sophisticated methodology. Many factors have been suggested as contributing to the housing bubble, which began in approximately 1998, lasting until 2006. Consumer buying behavior was driven by forces, such as greed, the desire to live in a larger house, the need to build retirement assets, and desire to avoid “ineviTable” higher prices in the future. Market conditions also contributed to higher prices, because of pressure from increases in population, shifts in demographics, availability of easy credit, and the relaxation of lending standards. Economic factors of low inflation, rising salaries, and low interest rates also have been suggested as playing a significant role in driving up housing prices. This research addresses these issues by selecting well understood factors that traditionally drive the housing market and constructing a regression model to investigate the nature of the relationships. Because of the co-dependence of many of the factors, structural equation modeling (SEM) is used rather than traditional regression analysis. This technique deals with the high levels of correlation among the many of the factors driving the housing market. Because all of the variables used in our models are observable, measurement model issues typical in most SEM analyses are not a concern. REVIEW OF THE LITERATURE Behavior of the housing market has been the subject of a substantial research stream over the past decade. Kindleberger (1987) provided a definition of a housing bubble based on buyer’s expectation that many researchers have used as a starting point for their research. Some studies questioned the existence of the bubble, such as Himmelberg, Meyer, and Sinai (2005). Other studies, such as Mints (2007), Baker (2007), and Chambers, Carriga, and Schagenhauf (2008) and Chomsisengphet and Pennington-Cross (2006) focused on factors that drove the housing market. Case and Shiller (2003) studied what factors might cause a housing bubble and studied several diverse housing markets to validate their hypotheses. Mayer and Quigley (2003) added insights to the results of Case and Shiller (2003) and took issue with their over emphasis on the investment motive of buyers. For their research, Smith and Smith (2006) defined a bubble in financial terms rather than using Kindleberger’s approach, which focused on buyer’s expectations. A more extensive review of the literature can be found in Kohn and Bryant (2010). As discussed in Kohn and Bryant (2010), there has been considerable debate concerning the definition of a bubble, methods of detection of the bubble, and root causes of the bubble, if, in fact, it did exist. Using standard regression analysis, the authors determine that a bubble did Research in Business and Economics Journal Factors leading to the U.S. housing bubble, Page 3 occur, with significant differences between two examined periods, pre-bubble, 1988 to 1996, and bubble, 1997 to 2007. Also, there were important findings from this previous work. Using median asking prices as the dependant variable, seven independent variables were included in regression analysis. These variables were the consumer price index, housing inventory, 30-year conventional mortgage rates, personal income, population, vacancy rates, and median asking rents. Results show only two variables were retained in the pre-bubble model, personal income and vacancy rates. By comparison, the bubble-period analysis revealed only two of the seven were removed, the 30-year conventional mortgage rates and personal income. Both models exhibited high values of coefficients of determination. The authors note, however, that co-linearity was very high among independent variables in the bubble model, but not significant in the pre-bubble model. Results were clear and useable in that the research reveals that a bubble did occur and variables were significant in their effects on the housing market. As the authors state, “for research with a forecasting orientation, the strong co-linearity effects would be problematic. Since we are primarily interested in identifying indications of a housing bubble, the issue of co-linearity is not a consideration.”[1] Kohn and Bryant go on to point out that more sophisticated research techniques should be used to reduce the effects of co-linearity on model results. The current research takes that next step and uses SEM to resolve problems caused by co-linearity and to be able to confirm earlier findings. The current research takes a structural approach by modeling a set of commonly accepted factors that affect the housing market and attempt to determine what role, if any, they played in driving the housing market. By using SEM rather than traditional regression analysis, the complex nature of inter-dependencies of these variables can be more accurately analyzed. There are several indicators that can be used to reflect housing market behavior. The median asking price was used as a proxy for the house price boom, since it reflects the seller’s subjective expectations of the home’s value. In some sense, this variable also captures the element of greed that exists in all bubble situations, namely, sellers in any overheated market are driven by the prospect of substantial gains to demand even higher prices for their assets. This research will investigate the behavior of median asking prices to determine what factors did or did not play a significant role in explaining the behavior of housing prices. SEM models will also help determine if the substantial shift in the behavior of housing prices that occurred over the past two decades was reflective of a bubble. The collapse of the housing market and sharp declines in housing values may not necessarily be indicative of a housing bubble, since values of assets decline during deflationary periods. Case and Shiller (2003) suggest that a bubble “referred to a situation in which excessive public expectations of future price increases cause prices to be temporarily elevated.” Our definition of a housing bubble is based on a variation of Case and Shiller’s definition. A bubble occurs when the market price of any asset rises substantially above traditionally accepted values, as determined by historical behavior. By modeling a pre-bubble period and comparing it to a bubble period, differences between the two models can be studied to determine if they are structurally different. The pre-bubble period should reflect a more sTable market in which traditional factors contribute to a rise or fall of median asking prices. During the bubble period in which housing prices have been rising substantially, a different set of factors should influence housing prices. This structural approach may shed more light on the behavior of the housing market, and hence, whether a bubble did occur. Abstract:Based on the heterojunction of V 2 CT x MXene and bimetallic CoNi-layered double hydroxide (CoNi-MLDH), a novel dual-modal electrochemical aptasensor (denoted as CoNi-MLDH@V 2 CT x ) was developed for the noninvasive point-of-care detection of cortisol in different body fluids (such as sweat, urine, and blood). Herein, V 2 CT x nanosheets (NSs) were used as scaffolds for the in-situ growth of cobalt/nickel zeolitic imidazolate framework (CoNi-ZIF) polyhedrons, which was then converted to CoNi-MLDH NSs through an in situ etching–coprecipitating process. Benefiting from the merits of V 2 CT x NSs (excellent electrochemical activity) and CoNi-ZIF (large specific surface and pore size, rich functionality, excellent oxygen reduction, and strong bioaffinity toward aptamer), and from the synergistic effect, the CoNi-LDH@V 2 CT x junction showed great potential as a platform for the constructing the electrochemical aptasensor for detecting cortisol. Within a wide cortisol range 10 fg mL −1 to 100 ng mL −1 , the dual-modal impedimetric and zinc-air battery-driven self-powered aptasensor based on CoNi-MLDH@V 2 CT x illustrated the ultralow detection limit of cortisol, 0.26 and 0.17 fg mL −1 , respectively. They demonstrated high selectivity, superior stability, excellent reproducibility, and promising regeneration ability, and showed potential applications for cortisol analysis in different environments (sweat, urine, and blood). The present work provides a promising option for maintaining people's mental health through the sensitive and selective detection of cortisol in human fluids.

Abstract:Cortisol, a famous stress biomarker, can be considered a potential predictor of cardiac diseases in humans. The presence of cortisol in saliva has encouraged researchers to design point-of-care devices for cortisol concentration in biological fluids. Here, human salivary cortisol was analyzed through a new non-invasive voltammetric aptasensor. Although cortisol is an electroactive compound, generally, the reduction in the current peak has been considered; however, this does not show a strong signal on a bare electrode surface, especially at low concentration levels. Hence, in this study, cortisol concentration was measured electrochemically and indirectly by monitoring the difference between electrochemical probe signals in the presence and absence of cortisol. A new polymeric nanocomposite of samarium molybdate flower-like nanoparticles decorated in poly(pyrrole) was electro-synthesized on the surface of a glassy carbon electrode. Then, reduced graphene oxide was cast on the surface. Finally, the cortisol aptamer was immobilized covalently on the reduced graphene oxide. This platform was used to increase the oxidation current peak of the ferricyanide solution as a probe as well as its electrocatalyst. The novel designed polymeric has the potential ability for effective immobilization of aptamers on the electrode surface without decreasing their biological activities. Additionally, it can enhance the probe electrochemical signal. The differential pulse voltammetric method (DPV) was applied as the detection technique. By optimizing the effective parameters, a determination range of 5.0 × 10−14–1.5 × 10−11 mol/L and a limit of detection of 4.5 × 10−14 mol/L were obtained. Selectivity of the proposed aptasensor relative to β-estradiol, progesterone and also prednisolone was studied as well. Finally, cortisol in a healthy human saliva sample was successfully analyzed by the proposed biosensors.

Abstract:Herein, we describe a label-free transition metal oxide based biosensor platform to detect both saliva and sweat cortisol levels at a point-of-care testing (POCT) perspective. The immobilization of ACAs on GCE/beta-MnO2 CNs electrode is conducted by electrostatic interaction approach via isoelectric-point (IEP) of beta-MnO2 (7.30) and cortisol antibodies (4.50), which establish a suitable atmosphere for effective immobilization. Bovine serum albumin (BSA) was served as blocking-agent for non-specific adsorption on the surface of MnO2 nanostructured electrode. Under optimized conditions, ACAs/beta-MnO2 cacti-like nanostructured (beta-MnO2 CNs) electrode specifically interacted with cortisol molecules, which enabled rapid and sensitive electrochemical impedance detection of cortisol via 3D analyte diffusion profile at the electrode interface and also enhanced cortisol capture achieved by large specific surface of MnO2 nanostructures. This ACAs/beta-MnO2 nanostructured electrode shows wide-linear EIS signals from 0.1 pM to 1500 pM with a limit of detection (LOD) of 0.023 pM (S/N = 3) and displays rapid binding kinetics with cortisol versus other glucocorticoids in terms of selectivity. Finally, the cortisol detection in human sweat, and saliva samples has been demonstrated for practical applicability and obtained results were compared with ELISA method.

Abstract:Cortisol is a well-known stress biomarker; this study focuses on using electrochemical immuno-sensing to measure the concentration of cortisol selectively and sensitively in artificial samples. Anti-cortisol antibodies have been immobilised on polycrystalline Au electrodes via strong covalent thiol bonds, fabricating an electrochemical bio-immunosensor for cortisol detection. IrOx was then anodically electrodeposited as a reference electrode on a commercial screen-printed electrode and electrochemical impedance spectrometry (EIS) studies were used to correlate the electrochemical response to cortisol concentration and the induced changes in charge transfer resistance (Rct). A linear relationship between the Rct and the logarithm of cortisol concentration was found in concentrations ranging from 1 ng/mL to 1 mg/mL with limit of detection at 11.85 pg/mL (32.69 pM). The modification of the reference electrode with iridium oxide has greatly improved the reproducibility of the screen-printed electrode. The sensing system can provide a reliable and sensitive detection approach for cortisol measurements.

Abstract:Cortisol, also known as the "stress hormone," is secreted under the control of the hypothalamic-pituitary-adrenocortical (HPA) axis in response to psychobiological stress. Real-time and continuous monitoring of the cortisol levels throughout the day can provide the information necessary to identify any abnormalities in cortisol's circadian rhythm that may disrupt the several processes that cortisol is involved in in the body. This review presents a systematic search of the literature on electrochemical cortisol sensing techniques that allow real-time measurement of cortisol in human biofluids. Several structural and performance-related parameters of sensors are being discussed, including the sensor stack layers, the limit of detection (LoD), dynamic range, sensitivity, selectivity, reusability, redox probe usage, and the electrochemical detection technique used. The sensors here are primarily categorized based on the type of bioreceptors used: antibodies, molecularly imprinted polymers (MIPs), and aptamers. According to this review, cortisol aptasensors and the MIP-based sensors present, in general, superior stability and sensitivity over immunosensors. They also promise reversible binding, albeit limited research exists on sensors deploying such bioreceptors. Additionally, notable advancements in the field and their impact on the development of point-of-care (PoC) and wearable devices are discussed.

Abstract:In this study, we examined the influence of functionalized poly(3,4-ethylenedioxythiophene) (PEDOT) nanostructures decorated on the channel layer of an organic electrochemical transistor (OECT) for the detection of sweat cortisol, an adrenocorticosteroid stress hormone. The OECT device featured a bilayer channel confined by a PEDOT:polystyrenesulfonate (PSS) underlayer and a nanostructure-decorated upper layer engineered from the monomers EDOT-COOH and EDOT-EG3 through template-free electrochemical polymerization. This molecular design allowed antibody conjugation using 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide/N-hydroxysulfosuccinimide coupling through the carboxylic acid side chain, with EDOT-EG3 known to minimize nonspecific binding of biomolecules. We also engineered an OECT device having a channel area without any nanostructures to gain insight into the effect of the nanostructures on cortisol sensing. Our new nanostructure-embedded OECT device facilitated real-time detection of cortisol at concentrations ranging from 1 fg/mL to 1 μg/mL with a detection limit of 0.0088 fg/mL with good linearity (R2 = 0.9566), in addition to excellent selectivity toward cortisol among other structurally similar interfering compounds and high stability and reproducibility. With its rapid response for the detection of 100 ng/mL cortisol-spiked artificial sweat, this nanostructure-decorated OECT device has potential clinical practicality and utility in wearable sensors for future healthcare applications.

Abstract:Cortisol is established as a reliable biomarker for stress prompting intensified research in developing wearable sensors to detect it via eccrine sweat. Since cortisol is present in sweat in trace quantities, typically 8–140 ng/mL, developing such biosensors necessitates the design of bioreceptors with appropriate sensitivity and selectivity. In this work, we present a systematic biomimetic methodology and a semi-automated high-throughput screening tool which enables rapid selection of bioreceptors as compared to ab initio design of peptides via computational peptidology. Candidate proteins from databases are selected via molecular docking and ranked according to their binding affinities by conducting automated AutoDock Vina scoring simulations. These candidate proteins are then validated via full atomistic steered molecular dynamics computations including umbrella sampling to estimate the potential of mean force using GROMACS version 2022.6. These explicit molecular dynamic calculations are carried out in an eccrine sweat environment taking into consideration the protein dynamics and solvent effects. Subsequently, we present a candidate baseline peptide bioreceptor selected as a contiguous sequence of amino acids from the selected protein binding pocket favourably interacting with the target ligand (i.e., cortisol) from the active binding site of the proteins and maintaining its tertiary structure. A unique cysteine residue introduced at the N-terminus allows orientation-specific surface immobilization of the peptide onto the gold electrodes and to ensure exposure of the binding site. Comparative binding affinity simulations of this peptide with the target ligand along with commonly interfering species e.g., progesterone, testosterone and glucose are also presented to demonstrate the validity of this proposed peptide as a candidate baseline bioreceptor for future cortisol biosensor development.

Abstract:In this study, poly- l -cysteine (P( l -Cys)), used as the multi-identification unit to sensitively sense multiple steroid hormones, was proposed. The mechanisms of the electropolymerization and electrochemical behavior of P( l -Cys) were revealed. Based on the experimental data and Faraday's law, the P( l -Cys) obtained by electropolymerization possessed abundant sulfonic acid groups. As steroid hormones (STHs) can bind to sulfuric acid for metabolism, the proposed P( l -Cys) system was used as synthetic receptors to detect STHs in serum. The results showed that the reduction peaks obtained from the STHs with different parent nucleus were different and did not interfere with each other when detected simultaneously. The interactions between P( l -Cys) and STHs as well as the reduction mechanisms of hormones were explored by using density functional theory and Faraday's law. To verify the generalizability of this method, thirty-eight different substances (including 30 steroid hormones and 8 nonsteroid hormones) were studied, and it was verified that this method could directly detect three classes of steroid hormones: progestogens, corticosteroids and androgens. The recognition site of this sensor for STHs was the Δ 4 -3-ketone in the parent nucleus. The P( l -Cys)/GCE sensor allows for the rapid detection of all three types of hormones simultaneously in less than 1 min. The limit of detection for progesterone, dexamethasone and hydrocortisone were 0.0008 μM b , 0.0055 μM and 0.011 μM, respectively. In addition to the applications presented in this study, P( l -Cys)/GCE is a low-cost method that can be applied to detect STHs in other samples, such as environmental and food products.

Abstract:Monitoring cortisol, a hormone released by the adrenal cortex in response to stress, is essential to evaluate the endocrine response to stress stimuli. While the current cortisol sensing methods require large laboratory settings, complex assay, and professional personnel. Herein, a novel flexible and wearable electrochemical aptasensor based on a Ni-Co metal-organic frameworks (MOF) nanosheet-decorated carbon nanotubes (CNTs)/polyurethane (PU) film is developed for rapid and reliable detection of cortisol in sweat. First, the CNTs/PU (CP) film was prepared by a modified wet spinning technology, and the CNTs/polyvinyl alcohol (PVA) solution was thermally deposited on the surface of CP film to form the highly flexible CNTs/PVA/CP (CCP) film with excellent conductivity. Then aminated Ni-Co MOF nanosheet prepared by a facile solvothermal method was conjugated with streptavidin and modified on the CCP film. Biofunctional MOF can effectively capture cortisol aptamer due to its excellent specific surface area. In addition, the MOF with peroxidase activity can catalytic oxidization of hydroquinone (HQ) by hydrogen peroxide (H2O2), which could amplify the peak current signal. The catalytic activity of Ni-Co MOF was substantially suppressed in the HQ/H2O2 system due to the formation of the aptamer-cortisol complex, which reduced the current signal, thereby realizing highly sensitive and selective detection of cortisol. The sensor has a linear range of 0.1-100 ng/mL and a detection limit of 0.032 ng/mL. Meanwhile, the sensor showed high accuracy for cortisol detection under mechanical deformation conditions. More importantly, the prepared MOF/CCP film based three-electrode was assembled with the polydimethylsiloxane (PDMS) substrate, and the sweat-cloth was used as the sweat collection channel to fabricate a wearable sensor patch for monitoring of cortisol in volunteers' sweat in the morning and evening. This flexible and non-invasive sweat cortisol aptasensor shows great potential for quantitative stress monitoring and management.

Abstract:Wearable sweat sensors are highly desirable for noninvasive monitoring of personal health. Herein, we developed a flexible electrochemical sensor based on nanofiber-involved microfluidic technique and molecularly imprinted polymer (MIP) for in situ and real-time sweat analysis. The sensor with polyethylene terephthalate (PET) as the substrate consisted of two layers, a bottom MIP-modified electrode layer for sensing and an upper nanofiber-based microfluidic layer for spontaneous sweat pumping. To enhance stability and sensitivity of the sensor, porous and interconnected 3D carbon nanofiber membrane decorated with gold nanoparticles () served as a matrix for MIP deposition. Prussian blue nanoparticles (PBnPs) as a "built-in" redox probe were embedded into MIP skeleton, enabling in situ monitoring of sweat substance. On the microfluidic layer, the super-hydrophilic and porous nanofibers allowed sweat to be automatically and rapidly pumped to the bottom sensing chamber. On the basis of the above, the sensor demonstrated a wide detection range of 1 nM∼1 μM as well as excellent selectivity and stability with cortisol as the model analyte. On-body applicability of the sensor was also validated by real-time monitoring of cortisol in healthy volunteers' sweat.
