Abstract:Mosquitoes, one of the deadliest animals on the planet, cause millions of fatalities each year by transmitting several human illnesses. Synthetic pesticides were previously used to prevent the spread of diseases by mosquitoes, which was effective in protecting humans but caused serious human health problems, environmental damage, and developed mosquito pesticide resistance. This research focuses on exploring new, more effective, safer, and environmentally friendly compounds to improve mosquito vector management. Phytochemicals are possible biological agents for controlling pests and many are target-specific, rapidly biodegradable, and eco-friendly. The potential of extracts of Lantana camara, Melia azedarach, Nerium oleander, Ricinus communis, and Withania somnifera against 3rd instar Culex pipiens (Common house mosquito) larvae was evaluated. Methanol extracts had more toxic effects against Cx. pipiens larvae (95-100%, 24 h post-treatment) than aqueous extracts (63-91%, 24 h post-treatment). The methanol extracts of Nerium oleander (LC50 = 158.92 ppm) and Ricinus communis (LC50 = 175.04 ppm) were very effective at killing mosquito larvae, 24 h after treatment. N. oleander (LC50 = 373.29 ppm) showed high efficacy in aqueous plant extracts. Among the different extracts of the five plants screened, the methanol extract of R. communis recorded the highest ovicidal activity of 5% at 800 ppm concentration. Total developmental duration and growth index were highly affected by R. communis and M. azedarach methanol extracts. In field tests it was clear that plant extracts decreased mosquito larval density, especially when mixed with mosquito Bti briquette, with stability up to seven days for N. oleander. GC-MS results showed that the methanol extract had a higher number of chemical compounds, particularly with more terpene compounds. A high-performance liquid chromatography (HPLC) technique was used to detect the existence of non-volatile polyphenols and flavonoids. All five methanol extracts showed high concentrations of active ingredients such as gallic acid, chlorogenic acid (more than 100 μg/ml) and the rosmarinic acid was also found in all the five extracts in addition to 17 active polyphenols and flavonoids presented at moderate to low concentrations. Molecular modeling of 18 active ingredients detected by the HPLC were performed to the vicinity of one of the fatty acid binding proteins of lm-FABP (PDB code: 2FLJ). Rutin, Caffeic acid, coumaric acid and rosmarinic acid which presented densely in R. communis and N. oleander showed multiple and stable intermolecular hydrogen bonding and π-π stacking interactions. The inhibition ability of the fatty acid binding protein, FABP4, was evaluated with remarkable receptor inhibition evident, especially with R. communis and N. oleander having inhibitory concentrations of IC50 = 0.425 and 0.599 µg/mL, respectively. The active phytochemical compounds in the plants suggest promising larvicidal and ovicidal activity, and have potential as a safe and effective alternative to synthetic insecticides.
Bioactivity of select essential oil constituents against life stages of Anopheles arabiensis (Diptera: Culicidae)

Abstract:Malaria is transmitted by infected female Anopheles mosquitoes, and An. arabiensis is a main malaria vector in arid African countries. Like other anophelines, its life cycle comprises of three aquatic stages; egg, larva, and pupa, followed by a free flying adult stage. Current vector control interventions using synthetic insecticides target these stages using adulticides or less commonly, larvicides. With escalating insecticide resistance against almost all conventional insecticides, identification of agents that simultaneously act at multiple stages of Anopheles life cycle presents a cost-effective opportunity. A further cost-effective approach would be the discovery of such insecticides from natural origin. Interestingly, essential oils present as potential sources of cost-effective and eco-friendly bioinsecticides. This study aimed to identify essential oil constituents (EOCs) with potential toxic effects against multiple stages of An. arabiensis life cycle. Five EOCs were assessed for inhibition of Anopheles egg hatching and ability to kill larvae, pupae and adult mosquitoes of An. arabiensis species. One of these EOCs, namely methyleugenol, exhibited potent Anopheles egg hatchability inhibition with an IC50 value of 0.51 ± 0.03 μM compared to propoxur (IC50: 5.13 ± 0.62 μM). Structure-activity relationship study revealed that methyleugenol and propoxur share a 1,2-dimethoxybenze moiety that may be responsible for the observed egg-hatchability inhibition. On the other hand, all five EOCs exhibited potent larvicidal activity with LC50 values less than 5 μM, with four of them; cis-nerolidol, trans-nerolidol, (-)-α-bisabolol, and farnesol, also possessing potent pupicidal effects (LC50 < 5 μM). Finally, all EOCs showed only moderate lethality against adult mosquitoes. This study reports for the first time, methyleugenol, (-)-α-bisabolol and farnesol as potent bioinsecticides against early life stages of An. arabiensis. This synchronized activity against Anopheles aquatic stages shows a prospect to integrate EOCs into existing adulticide-based vector control interventions.
Screening for feeding deterrent activity of herbal extracts against the larvae of malaria vector Anopheles subpictus Grassi

Abstract:This no-choice, laboratory study focuses on the feeding of homogeneous powdered, dried, yeast with different plant extracts on mosquito fourth-instar larvae to determine the effects on their mortality. Screening for antifeedant activity of plant extracts with some known medicinal attributes could lead to the discovery of new agents for vector control. The aim of this study was to investigate the antifeedant activity of crude leaf hexane, ethyl acetate, acetone, and methanol extracts of Andrographis lineata Wallich ex Nees. (Acanthaceae), Anisomeles malabarica (L.) Sims. (Lamiaceae), Argemone mexicana L. (Papaveraceae), Aristolochia bracteolata Lam. (Aristolochiaceae), Chrysanthemum indium L. (Asteraceae), Datura metal L. (Solanaceae), Eclipta prostrata L. (Asteraceae), and Sesbania grandiflora (L.) Pers. (Fibaceae) against the fourth-instar larvae of Anopheles subpictus Grassi (Diptera:Culicidae). All the crude extracts showed antifeedant activity in a dose-dependent manner. The plant extracts exhibited a significant antifeedant activity after 24 and 48 h of exposure; however, the highest larval mortality was found in leaf ethyl acetate extract of A. malabarica, acetone extract E. prostrata, methanol extract of A. lineata, C. indium, and S. grandiflora after 24 h (LC(50) = 2.53, 2.82, 2.31, 2.56, and 2.08 mg/mL; LC(90) = 6.40, 8.06, 7.45, 6.98, and 6.20 mg/mL), respectively. The hexane extract of A. lineata, D. metal, methanol extract of A. bracteolata and E. prostrata showed larval mortality after 48 h (LC(50) = 3.05, 2.11, 3.00, and 2.18 mg/mL; LC(90) = 9.06, 6.22, 8.23, and 5.77 mg/mL), respectively. One hundred percent larval mortality was observed in methanol extract of A. lineata, and C. indium after 24 h and the hexane extract of A. lineata and D. metal after 48 h at 10 mg/100 mL. The methanol extracts of A. lineata and C. indium significantly increased larval mortality in comparison to larvae fed with untreated diet. Bacillus thuringiensis subsp. israelensis is widely accepted as a biological pesticide because of its highly specific activity against dipteran insects without adverse effects on other organisms. The feeding deterrent activity of different herbal extracts against the larvae of malaria vector A. subpictus exhibited significantly lower toxicity compare with the bio larvicides, B. thuringiensis. These results suggest that the methanol extract of A. lineata, C. indium, the hexane extract of A. lineata and D. metal have the potential to be used as an ideal eco-friendly approach for the control of the medically important vector A. subpictus. These findings corroborate traditional insecticidal application of selected plants and the results can be extended for the control of mosquitoes.
The insecticidal activity of essential oil constituents against pyrethroid-resistant Anopheles funestus (Diptera: Culicidae)

Abstract:Malaria vector control relies on the use of insecticides for indoor residual spraying and long-lasting bed nets. However, insecticide resistance to pyrethroids among others, has escalated. Anopheles funestus, one of the major African malaria vectors, has attained significant levels of resistance to pyrethroids. Overexpressed P450 monooxygenases have been previously identified in pyrethroid resistant An. funestus. The escalating resistance against conventional insecticides signals an urgent need for identification of novel insecticides. Essential oils have gained recognition as promising sources of alternative natural insecticides. This study investigated six essential oil constituents, farnesol, (-)-α-bisabolol, cis-nerolidol, trans-nerolidol, methyleugenol, santalol (α and β isomers) and essential oil of sandalwood, for the adulticidal effects against pyrethroid-resistant An. funestus strain. The susceptibility against these terpenoids were evaluated on both pyrethroid-susceptible and resistant An. funestus. Furthermore, the presence of overexpressed monooxygenases in resistant An. funestus was confirmed. Results showed that both the pyrethroid-susceptible and resistant An. funestus were susceptible to three EOCs; cis-nerolidol, trans-nerolidol and methyleugenol. On the other hand, the pyrethroid-resistant An. funestus survived exposure to both farnesol and (-)-α-bisabolol. This study however does not show any direct association of the overexpressed Anopheles monooxygenases and the efficacy of farnesol and (-)-α-bisabolol. The enhanced activity of these terpenoids against resistant An. funestus that has been pre-exposed to a synergist, piperonyl butoxide, suggests their potential effectiveness in combination with monooxygenase inhibitors. This study proposes that cis-nerolidol, trans-nerolidol and methyleugenol are potential agents for further investigation as novel bioinsecticides against pyrethroid-resistant An. funestus strain.
Evaluation of mosquitocidal, histopathological and non-target effect of botanical pesticides from Stemodia viscosa and their mixtures against immature stages of Aedes aegypti, Anopheles stephensi and Culex quinquefasciatus

Abstract:Currently, mosquito control programs primarily rely on the utilization of chemically based insecticides. The objective of this study was to evaluate the efficacy of the plant Stemodia viscosa Roxb. against the early life stages of Culex quinquefasciatus, Anopheles stephensi and Aedes aegypti mosquitoes. The larvae, pupae and eggs of the mosquitoes were exposed to four different concentrations (62.5 to 500 ppm). After 120 h of treatment, the hexane and ethyl acetate extracts of S. viscosa exhibited 100% ovicidal activity against the eggs of the treated mosquitoes. The hexane and ethyl acetate extracts showed significant larvicidal activity against all three mosquitoes. The LC50 values of the hexane extract were 272.5, 233.5, 217.9 ppm and the LC50 values of the ethyl acetate extract were 238.6, 230.5 and 169.1 ppm respectively, after 24 h. The hexane and ethyl acetate extracts of S. viscosa exhibited promising pupicidal activity with LC50 values of 286.6, 265.3, 257.9 and 192.2, 290.9 and 238.1 ppm, respectively, after 24 h against all three mosquitoes. Additionally, histopathological and growth disruption studies revealed severe damage in the midgut cells of the treated larvae. The suitability index (SI) or predator safety factor (PSF) values indicated that the extracts from S. viscosa could be safely utilized in Integrated Mosquito Management (IMM) without harming the predators. Furthermore, formulations based on hexane and ethyl acetate extracts demonstrated strong activity while exhibiting no toxicity towards non-target organisms. The results imply that the extract holds potential as an eco-friendly substitute for synthetic insecticides in mosquito control. However, additional research is required to identify the most potent components of the plant extract to enhance product efficacy and stability.

Abstract:The management of mosquito resistance to chemical insecticides and the biting behaviour of some species are motivating the search for complementary and/or alternative control methods. The use of plants is increasingly considered as a sustainable biological solution for vector control. The aim of this study was to evaluate the biological effects of the essential oil (EO) of Lippia alba harvested in Abidjan (Côte d'Ivoire) against Anopheles gambiae and Aedes aegypti mosquitoes. Phytochemical compounds were identified by GC–MS. Knockdown and mortality were determined according to the WHO test tube protocol. Contact irritancy was assessed by observing the movement of mosquitoes from a treated WHO tube to a second untreated tube. Non-contact repellency was assessed using a standardised high-throughput screening system (HITSS). Blood meal inhibition was assessed using a membrane feeding assay treated with EO. The EO was identified as the citral chemotype. The EO gave 100% KD60 in both species at a concentration of 1%. Mortalities of 100% were recorded with An. gambiae and Ae. aegypti at concentrations of 1% and 5% respectively. The highest proportions of females escaping during the contact irritancy test were 100% for An. gambiae at 1% concentration and 94% for Ae . aegypti at 2.5% concentration. The 1% concentration produced the highest proportions of repelled mosquitoes in the non-contact repellency tests: 76.8% ( An. gambiae ) and 68.5% ( Ae. aegypti ). The blood meal inhibition rate at a dose of 10% was 98.4% in Ae . aegypti but only 15.5% in An. gambiae . The citral chemotype of L. alba EO has promising biological effects in both species that make it a potentially good candidate for its use in mosquito control. The results obtained in this study encourage the further evaluation of L. alba EOs from other localities and of different chemotypes, under laboratory and field conditions.

In vitro and in silico analysis of the Anopheles anticholinesterase activity of terpenoids

Abstract:Anopheles gambiae, An. coluzzii, An. arabiensis, and An. funestus are major vectors in high malaria endemic African regions. Various terpenoid classes form the main chemical constituent repository of essential oils, many of which have been shown to possess insecticidal effects against Anopheles species. The current study aimed to assess the bioactivity of terpenoids including four sesquiterpene alcohols, farnesol, (-)-α-bisabolol, cis-nerolidol, and trans-nerolidol; a phenylpropanoid, methyleugenol, and a monoterpene, (R)-(+)-limonene, using the larvicidal screening assay against the four Anopheles species. The mechanism of action was investigated through in vitro acetylcholinesterase inhibition assay and in silico molecular modelling. All six terpenoids showed potent larvicidal activity against the four Anopheles species. Insights into the mechanism of action revealed that the six terpenoids are strong AChE inhibitors against An. funestus and An. arabiensis, while there was a moderate inhibitory activity against An. gambiae AChE, but very weak activity against An. coluzzii. Interestingly, in the in silico study, farnesol established a favourable hydrogen bonding interaction with a conserved amino acid residue, Cys447, at the entrance to the active site gorge. While (-)-α-bisabolol and methyleugenol displayed a strong interaction with the catalytic Ser360 and adjacent amino acid residues; but sparing the mutable Gly280 residue that confers resistance to the current anticholinesterase insecticides. As a result, this study identified farnesol, (-)-α-bisabolol, and methyleugenol as selective bioinsecticidal agents with potent Anopheles AChE inhibition. These terpenoids present as natural compounds for further development as anticholinesterase bioinsecticides.
Insecticidal and larvicidal activities of medicinal plant extracts against mosquitoes

Abstract:In recent years, use of environment friendly and biodegradable natural insecticides of plant origin have received renewed attention as agents for vector control because they are rich in bioactive chemicals, active against a limited number of species including specific target insects, and biodegradable. The present study was carried out to evaluate the adulticidal, repellent, and larvicidal activity of crude hexane, ethyl acetate, and methanol extracts of eight plants, viz. Aristolochia indica L., Cassia angustifolia Vahl, Diospyros melanoxylon Roxb., Dolichos biflorus L., Gymnema sylvestre (Retz) Schult, Justicia procumbens L., Mimosa pudica L., and Zingiber zerumbet L., were tested against adult and early fourth instar larvae of Culex gelidus Theobald and Culex quinquefasciatus Say (Diptera: Culicidae). The effective adult mortality was observed in methanol extract of A. indica, ethyl acetate extract of D. biflorus, and ethyl acetate and hexane extract of Z. zerumbet against C. gelidus and C. quinquefasciatus (LD50 = 37.75, 78.56, 129.44, 86.13, 80.06, 112.42, 53.83, and 46.61; LD90 = 166.83, 379.14, 521.50, 289.83, 328.18, 455.72, 181.15, and 354.50 ppm, respectively). Complete protections for 150 min were found in hexane and methanol extract of A. indica and Z. zerumbet at 1,000 ppm against mosquito bites. The highest larval mortality was found in the hexane extract of Z. zerumbet, ethyl acetate extract of D. biflorus, and methanol extracts of A. indica against C. gelidus (LC50 = 26.48, 33.02, and 12.47 ppm; LC90 = 127.73, 128.79, and 62.33 ppm) and against C. quinquefasciatus (LC50 = 69.18, 34.76, and 25.60 ppm; LC90 = 324.40, 172.78, and 105.52 ppm), respectively, after 24 h. The plant extracts are potential to be used as an ideal eco-friendly approach for the control of the Japanese encephalitis vector, C. gelidus, and lymphatic filariasis vector, C. quinquefasciatus.

Abstract:In this paper we extract and validate a small-signal neural model for FinFETs. Namely, artificial neural networks are used for modeling the dependence of the small-signal admittance parameters for a FinFET on bias voltages and frequency. The model is developed for the actual transistor, which is obtained after de-embedding the effects of the probe pads, transmission lines, and substrate. The extracted model is validated in a wide range of operating bias conditions up to 50 GHz.
Ovicidal toxicity of plant essential oils and their major constituents against two mosquito vectors and their non-target aquatic predators

Abstract:Plant essential oil (EO) is a natural alternative to synthetic chemical insecticides for mosquito control. EOs from Citrus aurantium L., Cymbopogon citratus (Stapf.), and Cinnamomum verum (J. Presl.) were selected for topical assay of their ovicidal activity against Aedes aegypti (Linnaeus) and Aedes albopictus (Skuse). Their efficacy was compared to that of 1% ( w/w ) temephos. In addition, their non-toxicity against aquatic mosquito predators, Poecilia latipinna and Poecilia reticulata , was tested. Found by GC–MS analysis, the major constituent of C . verum EO was trans -cinnamaldehyde, of C . aurantium EO was d -limonene, and of C . citratus EO was geranial. Both C . verum EO and trans -cinnamaldehyde at a high concentration (30,000 ppm) exhibited high ovicidal activity against Ae . aegypti and Ae . albopictus eggs after 48 h of incubation with an inhibition rate of 91.0–93.0% for C . verum EO and 96.7–95.2% for trans -cinnamaldehyde. The combination of C . verum EO + geranial exhibited the strongest synergistic inhibition activity (100%) against the two mosquito vectors and was five times more effective than temephos. Moreover, they were not toxic to the non-target fishes. As a safe ovicidal agent for mosquito egg control, the combination of C . verum EO + geranial has excellent potential.

Abstract:Background: With insecticide resistance in malaria vectors spreading in geographical range and intensity, there is a need for compounds with novel modes of action to maintain the successes achieved to date by long-lasting insecticidal nets and indoor residual sprays, used as part of an insecticide resistance management strategy. Screening existing registered pesticides, predominantly those developed for use in agriculture, may provide a more rapid and less logistically challenging route to identifying active ingredients of value to public health than screening and chemical synthesis programmes for novel compounds. Methods: Insecticides and acaricides from all IRAC classes, including those with unclassified modes of action, were assessed for inclusion in a laboratory bioassay testing cascade against adult female Anopheles gambiae mosquitoes. A longlist of representative candidate compounds was selected, excluding those with safety concerns, unsuitable physiochemical properties, and likely hurdles to registration for public health use. An initial screen using topical application eliminated compounds with insufficient intrinsic activity, and a tarsal contact assay identified those with activity at an appropriate concentration. Compounds of interest were ranked by relative potency using dose response assays and discriminating dose calculations. Results: Inclusion of an adjuvant enhanced the tarsal efficacy of several compounds, facilitating the promotion of chemistries with great potential, given suitable formulation, which would not progress based on activity of compound alone. Comparison of data between stages in the testing cascade suggest that a more streamlined approach, topical application to test for intrinsic activity and determining the discriminating dose to compare relative potency of compounds, may be sufficient to identify compounds with potential value for use in long lasting insecticidal nets and indoor residual spray products. Conclusions: Identified were 11 compounds of interest as vector control agents (in descending order of potency): clothianidin, spinetoram, metaflumizone, dinotefuran, indoxacarb, abamectin, sulfoxaflor, oxazosulfyl, triflumezopyrim, fenpyroximate, and tolfenpyrad.

Abstract:Mosquito-borne diseases, such as malaria, dengue, and Zika, pose major public health challenges globally, affecting millions of people. The growing resistance of mosquito populations to synthetic insecticides underscores the critical need for effective and environmentally friendly larvicides. Although chemical pesticides can initially be effective, they often lead to negative environmental consequences and health hazards for non-target species, including humans. This study aimed to evaluate the larvicidal effects of Trachyspermum ammi essential oil and Delphinium speciosum extract on the larvae of three major mosquito species: Aedes aegypti, Anopheles stephensi, and Culex quinquefasciatus. Mosquito larvae of Ae. aegypti, An. stephensi, and Cx. quinquefasciatus were reared under controlled laboratory conditions. The larvicidal activity of T. ammi essential oil and D. speciosum extract was evaluated through standard bioassays, using various concentrations of essential oils (10, 20, 40, 80, and 160 ppm) and extracts (160, 320, 640, 1280, and 2560 ppm) to determine the lethal concentration (LC50) values after 24 h of exposure. Fresh plant materials were collected, with the essential oil extracted via hydro-distillation, and the extract prepared using methanol solvent extraction. The chemical composition of T. ammi essential oil was examined using gas chromatography-mass spectrometry (GC-MS). Additionally, the preliminary analysis of the chemical compounds in D. speciosum extract was carried out using thin layer chromatography (TLC) and nuclear magnetic resonance spectroscopy (NMR) techniques. The results indicated that the essential oil of T. ammi exhibited more effective larvicidal activity compared to the D. speciosum extract. Specifically, the essential oil demonstrated LC50 values of 18 ppm for Cx. quinquefasciatus and 19 ppm for Ae. aegypti. In contrast, the D. speciosum extract showed the strongest larvicidal effect against An. stephensi, with an LC50 of 517 ppm. Concentrations of 40 ppm of the essential oil and 1280 ppm of the extract resulted in 100% mortality across all three species. Both the essential oil of T. ammi and the D. speciosum extract exhibited concentration-dependent larvicidal activity, and these results were statistically significant (p < 0.001) compared to the no-treatment group. GC-MS analysis revealed thymol (88.95%), o-cymen-5-ol (4.11%), and γ-terpinene (2.10%) as the major constituents of the T. ammi essential oil. Additionally, TLC verified the presence of alkaloids in both chloroform and methanolic extracts. Proton NMR identified a diterpene structure for these alkaloids. These findings suggest that T. ammi essential oil is a promising candidate for natural mosquito control strategies. Given its efficacy, further research is warranted to explore its potential in integrated vector management programs.

Abstract:Mosquitoes (Diptera: Culicidae) are arthropod vectors involved in transmitting deadly infectious diseases to humans and animals, causing several million human deaths every year worldwide. Plant‐based products provide environment‐friendly mosquitocidal properties and are harmless to humans, non‐target organisms, and the environment. This review covers up‐to‐date information about plant products (plant compounds, extracts, and essential oils), which are utilized to control all the life cycle stages (egg, larva, pupa, and adult) of the mosquito genera Aedes, Anopheles, and Culex. Mosquitoes pose a severe threat to the environment as vectors of numerous harmful diseases affecting humans and animals. Mosquitoes transmit pathogens that cause dengue, chikungunya, malaria, zika, yellow fever, Japanese encephalitis, and filariasis. Today's mosquito control strategies heavily rely on the use of chemical insecticides such as N,N‐diethyl‐meta‐toluamide (DEET), N,N‐diethyl mandelic acid amide (DEM), and dimethyl phthalate (DMP). However, the widespread use of chemical insecticides has resulted in pollution, bio‐magnification, and other health and environmental issues. It has also become ineffective because of the mosquitoes' aptitude to develop resistance, emphasizing the urgent need for safe, effective, and long‐lasting strategies. An alternative and promising approach to circumventing these obstacles involves the implementation of insecticides made from natural compounds found in plants. Therefore, the scientific community has shifted its focus towards plant‐based phytochemicals, oils, and extracts, as these are eco‐friendly, safe, and cost‐effective alternatives to conventional chemical insecticides. This review aims to provide details on current advances in plant‐based products (plant compounds, extracts, and essential oils), which are used to control all the life cycle stages (egg, larva, pupa, and adult) of the mosquito genera Aedes, Anopheles, and Culex. Hopefully, this review will pave the way to devise control strategies against these challenging pests.

Abstract:Mosquitoes are one of the deadliest and most hazardous animals on Earth, where they transmit several diseases that kill millions of people annually. There is an ongoing search almost everywhere in the world for more effective and contemporary ways to control mosquitoes other than pesticides. Phytochemicals are affordable, biodegradable biological agents that specialize in eliminating pests that represent a risk to public health. The effectiveness of Acacia nilotica methanol and aqueous leaf extracts against 4th instar larvae was evaluated. The results revealed that the methanol extract of A. nilotica had a noticeable influence on the mortality rate of mosquito larvae, especially at high concentrations. Not only did the mortality rate rise significantly, but the hatching of the mosquito eggs was potentially suppressed.Terpenes, fatty acids, esters, glycosides, pyrrolidine alkane, piperazine, and phenols were the most prevalent components in the methanol extract, while the aqueous extract of A. nilotica exclusively showed the presence of fatty acids. The insecticidal susceptibility tests of both aqueous and alcoholic extract of A. nilotica confirmed that the Acacia plant could serves as a secure and efficient substitute for chemical pesticides because of its promising effect on killing larvae and egg hatching delaying addition to their safety as one of the natural pesticides. Molecular docking study was performed using one of the crucial and life-controlling protein targets, fatty acid binding protein (FABP) and the most active ingredients as testing ligands to describe their binding ability. Most of the structurally related compounds to the co-crystallized ligand, OLA, like hexadecanoic acid furnished high binding affinity to the target protein with very strong and stable intermolecular hydrogen bonding and this is quite similar to OLA itself. Some other structural non-related compounds revealed extraordinarily strong binding abilities like Methoxy phenyl piperazine. Most of the binding reactivities of the majortested structures are due to high structure similarity between the positive control, OLA, and tested compounds. Such structure similarity reinforced with the binding abilities of some detected compounds in the A. nilotica extract could present a reasonable interpretation for its insecticidal activity via deactivating the FABP protein. The FABP4 enzyme inhibition activity was assessed for of both methanolic and aqueous of acacia plant extract and the inhibition results of methanol extract depicted noticeable potency if compared to orlistat, with half-maximal inhibitory concentration (IC 50 ) of 0.681, and 0.535 μg/ml, respectively.

Influence of blood meal and age of mosquitoes on susceptibility to pyrethroids in Anopheles gambiae from Western Kenya

Abstract:Background: Physiological characteristics (age and blood feeding status) of malaria vectors can influence their susceptibility to the current vector control tools that target their feeding and resting behaviour. To ensure the sustainability of the current and future vector control tools an understanding of how physiological characteristics may contribute to insecticide tolerance in the field is fundamental for shaping resistance management strategies and vector control tools. The aim of this study was to determine whether blood meal and mosquito age affect pyrethroid tolerance in field-collected Anopheles gambiae from western Kenya. Methods: Wild mosquito larvae were reared to adulthood alongside the pyrethroid-susceptible Kisumu strain. Adult females from the two populations were monitored for deltamethrin resistance when they were young at 2-5 days old and older 14-16 days old and whether fed or unfed for each age group. Metabolic assays were also performed to determine the level of detoxification enzymes. Mosquito specimens were further identified to species level using the polymerase chain reaction (PCR) method. Results: Anopheles gambiae sensu stricto was the predominant species comprising 96% of specimens and 2.75% Anopheles arabiensis. Bioassay results showed reduced pyrethroid induced mortality with younger mosquitoes compared to older ones (mortality rates 83% vs. 98%), independently of their feeding status. Reduced mortality was recorded with younger females of which were fed compared to their unfed counterparts of the same age with a mortality rate of 35.5% vs. 83%. Older blood-fed females showed reduced susceptibility after exposure when compared to unfed females of the same age (mortality rates 86% vs. 98%). For the Kisumu susceptible population, mortality was straight 100% regardless of age and blood feeding status. Blood feeding status and mosquito age had an effect on enzyme levels in both populations, with blood fed individuals showing higher enzyme elevations compared to their unfed counterparts (P < 0.0001). The interaction between mosquito age and blood fed status had significant effect on mosquito mortality. Conclusion: The results showed that mosquito age and blood feeding status confers increased tolerance to insecticides as blood feeding may be playing an important role in the toxicity of deltamethrin, allowing mosquitoes to rest on insecticide-treated materials despite treatment. These may have implications for the sustained efficacy of indoor residual spraying and insecticide-treated nets based control programmes that target indoor resting female mosquitoes of various gonotrophic status.
Resistance of mosquitoes to Lambda-Cyhalothrin and DDT in a Niger Delta Region of Nigeria

Abstract:Background: The escalation of insecticide resistance across the World Health Organization (WHO) African region calls for the routine monitoring of insecticide resistance in mosquitoes. While pyrethroids are recommended for use and dichlorodiphenyltrichloroethane (DDT) has been prescribed, it is imperative to reevaluate their efficacy across diverse geographical settings. The extent of resistance among mosquitoes to DDT and lambda-cyhalothrin remains understudied in Ika North East, Delta State, Nigeria, where the yellow fever virus was reported in 2019. Materials and methods: We investigated the susceptibility of female mosquitoes to DDT (4%) and lambda-cyhalothrin (0.05%) in this specific study area. Adult female mosquitoes of Anopheles gambiae, Culex quinquefasciatus, and Aedes albopictus, aged between 2 and 4 days, were subjected to these insecticides utilizing the WHO bioassay method. The assessment of knockdown was done between 10 and 60 min, then mortality after 24 h. Results: C. quinquefasciatus and A. albopictus mosquitoes exposed to DDT and lambda-cyhalothrin exhibited high susceptibility, resulting in complete mortality (100%); however, A. gambiae displayed resistance, with mortality rates of 19% and 76%, respectively . Notably, A. gambiae mosquitoes exposed to piperonyl butoxide (PBO)-DDT and PBO-lambda-cyhalothrin exhibited enhanced mortality, reaching 95% (indicating suspected resistance) and 100% (indicating susceptibility), respectively. Knockdown time (KDT) for 50% in mosquitoes exposed to lambda-cyhalothrin ranged from 3.94 to 33.51 min. Similarly, KDT model for 95% ranged from 19.04 to 84.15 min. Among the tested mosquito species, Culex mosquitoes exhibited the shortest knockdown resistance time for lambda-cyhalothrin, recorded at 3.94 min. Similarly, the KDT for DDT ranged from 24.97 to 187.06 min for 50% mortality and from 61.04 to 431.03 min for 95% mortality, respectively. Anopheles mosquitoes exposed to PBO + DDT recorded the lowest KDT. Conclusion: Our study underscores the potential of exercising caution in the use of DDT and lambda-cyhalothrin insecticides for the control of mosquitoes due to emerging resistance.

Abstract:With the spread of resistance to long-established insecticides targeting Anopheles malaria vectors, understanding the actions of compounds newly identified for vector control is essential. With new commercial vector-control products containing neonicotinoids under development, we investigate the actions of 6 neonicotinoids (imidacloprid, thiacloprid, clothianidin, dinotefuran, nitenpyram and acetamiprid) on 13 Anopheles gambiae nicotinic acetylcholine receptor (nAChR) subtypes produced by expression of combinations of the Agα1, Agα2, Agα3, Agα8 and Agβ1 subunits in Xenopus laevis oocytes, the Drosophila melanogaster orthologues of which we have previously shown to be important in neonicotinoid actions. The presence of the Agα2 subunit reduces neonicotinoid affinity for the mosquito nAChRs, whereas the Agα3 subunit increases it. Crystal structures of the acetylcholine binding protein (AChBP), an established surrogate for the ligand-binding domain, with dinotefuran bound, shows a unique target site interaction through hydrogen bond formation and CH-N interaction at the tetrahydrofuran ring. This is of interest as dinotefuran is also under trial as the toxic element in baited traps. Multiple regression analyses show a correlation between the efficacy of neonicotinoids for the Agα1/Agα2/Agα8/Agβ1 nAChR, their hydrophobicity and their rate of knockdown of adult female An. gambiae, providing new insights into neonicotinoid features important for malaria vector control.

Abstract:Mosquitoes represent one of the most important vectors and are responsible for the transmission of many arboviruses that affect human and animal health. The chemical method using synthetic insecticides disturbs the environmental system and promotes the appearance of resistant insect species. Therefore, this study investigated the insecticidal effect of some binary monoterpene combinations (1,8 cineole + α-pinene and carvone + R (+)-pulegone) using a mixture design approach. The fumigant toxicity was evaluated against Culex pipiens female adults using glass jars. The results show that the toxicity varies according to the proportions of each compound. Indeed, Mixture 1 (1,8-cineole + α-pinene) displayed a strong toxic effect (51.00 ± 0.86% after 24 h and 100.00 ± 0.70% after 48 h) when the pure compounds were tested at 0.25/0.75 proportions of 1,8-cineole and α-pinene, respectively. Nevertheless, the equal proportion (0.5/0.5) of carvone and R (+)-pulegone in Mixture 2 exhibited a toxic effect of 54.35 ± 0.75% after 24 h and 89.96 ± 0.14% after 48 h, respectively. For Mixture 1, the maximum area of mortality that the proposed model indicated was obtained between 0/1 and 0.25/0.75, while the maximum area of mortality in the case of Mixture 2 was obtained between 0.25/0.75 and 0.75/0.25. Moreover, the maximum possible values of mortality that could be achieved by the validated model were found to be 51.44% (after 24 h) and 100.24% (after 48 h) for Mixture 1 and 54.67% (after 24 h) and 89.99% (after 48 h) for Mixture 2. It can be said that all purev molecules tested through the binary mixtures acted together, which enhanced the insecticide's effectiveness. These findings are very promising, as the chemical insecticide (deltamethrin) killed only 19.29 ± 0.01% and 34.05 ± 1.01% of the female adults after 24 h and 48 h, respectively. Thus, the findings of our research could help with the development of botanical insecticides that might contribute to management programs for controlling vectors of important diseases.
