Abstract:Sensory signals undergo substantial recoding when neural activity is relayed from sensors through pre-thalamic and thalamic nuclei to cortex. To explore how temporal dynamics and directional tuning are sculpted in hierarchical vestibular circuits, we compared responses of macaque otolith afferents with neurons in the vestibular and cerebellar nuclei, as well as five cortical areas, to identical three-dimensional translational motion. We demonstrate a remarkable spatio-temporal transformation: otolith afferents carry spatially aligned cosine-tuned translational acceleration and jerk signals. In contrast, brainstem and cerebellar neurons exhibit non-linear, mixed selectivity for translational velocity, acceleration, jerk and position. Furthermore, these components often show dissimilar spatial tuning. Moderate further transformation of translation signals occurs in the cortex, such that similar spatio-temporal properties are found in multiple cortical areas. These results suggest that the first synapse represents a key processing element in vestibular pathways, robustly shaping how self-motion is represented in central vestibular circuits and cortical areas.

Abstract:To maintain stable and coherent perception in an ever-changing environment, the brain needs to continuously and dynamically calibrate information from multiple sensory sources, using sensory and non-sensory information in a flexible manner. Here, we review how the vestibular and visual signals are recalibrated during self-motion perception. We illustrate two different types of recalibration: one long-term cross-modal (visual–vestibular) recalibration concerning how multisensory cues recalibrate over time in response to a constant cue discrepancy, and one rapid-term cross-modal (visual–vestibular) recalibration concerning how recent prior stimuli and choices differentially affect subsequent self-motion decisions. In addition, we highlight the neural substrates of long-term visual–vestibular recalibration, with profound differences observed in neuronal recalibration across multisensory cortical areas. We suggest that multisensory recalibration is a complex process in the brain, is modulated by many factors, and requires the coordination of many distinct cortical areas. We hope this review will shed some light on research into the neural circuits of visual–vestibular recalibration and help develop a more generalized theory for cross-modal plasticity.

Visual-Based Spatial Coordinate Dominates Probabilistic Multisensory Inference in Macaque MST-d Disparity Encoding

Abstract:Numerous studies have demonstrated that animal brains accurately infer whether multisensory stimuli are from a common source or separate sources. Previous work proposed that the multisensory neurons in the dorsal medial superior temporal area (MST-d) serve as integration or separation encoders determined by the tuning–response ratio. However, it remains unclear whether MST-d neurons mainly take a sense input as a spatial coordinate reference for carrying out multisensory integration or separation. Our experimental analysis shows that the preferred tuning response to visual input is generally larger than vestibular according to the Macaque MST-d neuronal recordings. This may be crucial to serving as the base of coordinate reference when the subject perceives moving direction information from two senses. By constructing a flexible Monte-Carlo probabilistic sampling (fMCS) model, we validate this hypothesis that the visual and vestibular cues are more likely to be integrated into a visual-based coordinate rather than vestibular. Furthermore, the property of the tuning gradient also affects decision-making regarding whether the cues should be integrated or not. To a dominant modality, an effective decision is produced by a steep response-tuning gradient of the corresponding neurons, while to a subordinate modality a steep tuning gradient produces a rigid decision with a significant bias to either integration or separation. This work proposes that the tuning response amplitude and tuning gradient jointly modulate which modality serves as the base coordinate for the reference frame and the direction change with which modality is decoded effectively.

Abstract:The perception of self-motion direction, or heading, relies on integration of multiple sensory cues, especially from the visual and vestibular systems. However, the reliability of sensory information can vary rapidly and unpredictably, and it remains unclear how the brain integrates multiple sensory signals given this dynamic uncertainty. Human psychophysical studies have shown that observers combine cues by weighting them in proportion to their reliability, consistent with statistically optimal integration schemes derived from Bayesian probability theory. Remarkably, because cue reliability is varied randomly across trials, the perceptual weight assigned to each cue must change from trial to trial. Dynamic cue reweighting has not been examined for combinations of visual and vestibular cues, nor has the Bayesian cue integration approach been applied to laboratory animals, an important step toward understanding the neural basis of cue integration. To address these issues, we tested human and monkey subjects in a heading discrimination task involving visual (optic flow) and vestibular (translational motion) cues. The cues were placed in conflict on a subset of trials, and their relative reliability was varied to assess the weights that subjects gave to each cue in their heading judgments. We found that monkeys can rapidly reweight visual and vestibular cues according to their reliability, the first such demonstration in a nonhuman species. However, some monkeys and humans tended to over-weight vestibular cues, inconsistent with simple predictions of a Bayesian model. Nonetheless, our findings establish a robust model system for studying the neural mechanisms of dynamic cue reweighting in multisensory perception.

Abstract:Visual-vestibular conflicts can induce motion sickness and further postural instability. Visual-vestibular habituation is recommended to reduce the symptoms of motion sickness and improve postural stability with an altered multisensory reweighting progress. However, it is unclear how the human brain reweights multisensory information after repeated exposure to visual-vestibular conflicts. Therefore, we synchronized a rotating platform and a virtual scene to present visual-vestibular congruent (natural visual stimulation) and incongruent (conflicted visual stimulation) conditions and collected EEG and center of pressure (COP) data. We constructed the effective brain connectivity of region of interest (ROI) derived from source-space EEG in theta-band activity, and quantified the postural stability and the inflow and outflow of each ROI. We found repeated exposure to congruent and incongruent conditions both decreased COP path length and increased COP complexity. Besides, we found that repeated exposure to the incongruent environment decreased the inflow into visual cortex, suggesting the brain down-weighted the less reliable visual information for postural stability. In contrast, repeated exposure to the congruent environment increased the inflow into posterior parietal cortex and the outflow from visual cortex and S1, suggesting an increase in efficiency of multisensory integration. We concluded that repeated exposure to congruent and incongruent conditions both improved postural stability with different multisensory reweighting patterns as revealed by different dynamic changes of brain networks.

Abstract:The different physiological roles of interleukin-1alpha (IL-1alpha) and interleukin-1beta (IL-1beta) are not well understood, especially when considering the apparent overlap and redundancy of the two IL-1 molecules. Characterization of IL-1alpha and IL-1beta expression was performed in this study in organs from young and old mice, using immunohistochemistry and ELISA (enzyme-linked immunosorbent assay). The results indicate that organ IL-1alpha and IL-1beta display different patterns of expression: IL-1alpha is manifested more prominently in lymphoreticular organs (lungs, small intestine, spleen, liver), while IL-1beta is more evident in highly specialized and more vulnerable organs, which do not play a leading role in defense against infections and intoxication (heart, brain, skeletal muscle, kidney). This differential expression is more accentuated in old mice, possibly pointing to the special relevance of these cytokines to organ homeostasis in old age. These findings may shed new light on the physiological functions of IL-1alpha and IL-1beta, and may also lead to the development of improved therapeutic approaches, based on the specific manipulation of these cytokines.

Abstract:Abstract Physiological studies have demonstrated that inputs from different sensory modalities converge on, and are integrated by, individual superior colliculus neurons and that this integration is governed by specific spatial rules. The present experiments were an attempt to relate these neural processes to overt behavior by determining if behaviors believed to involve the circuitry of the superior colliculus would show similar multisensory dependencies and be subject to the same rules of integration. The neurophysiological-behavioral parallels proved to be striking. The effectiveness of a stimulus of one modality in eliciting attentive and orientation behaviors was dramatically affected by the presence of a stimulus from another modality in each of the three behavioral paradigms used here. Animals trained to approach a low intensity visual cue had their performance significantly enhanced when a brief, low intensity auditory stimulus was presented at the same location as the visual cue, but their performance was significantly depressed when the auditory stimulus was disparate to it. These effects were independent of the animals' experience with the modifying (i.e. auditory) stimulus and exceeded what might have been predicted statistically based on the animals' performance with each single-modality cue. The multiplicative nature of these multisensory interactions and their dependence on the relative positions and intensities of the two stimuli were all very similar to those observed physiologically for single cells. The few differences that were observed appeared to reflect the fact that understanding integration at the level of the single cell requires reference to the individual cell's multisensory receptive field properties, while at the behavioral level populations of receptive fields must be evaluated. These data illustrate that the rules governing multisensory integration at the level of the single cell also predict responses to these stimuli in the intact behaving organism.

Abstract:The successful integration of visual and auditory stimuli requires information about whether visual and auditory signals originate from corresponding places in the external world. Here we report crossmodal effects of spatially congruent and incongruent audio-visual (AV) stimulation. Visual and auditory stimuli were presented from one of four horizontal locations in external space. Seven healthy human subjects had to assess the spatial fit of a visual stimulus (i.e. a gray-scaled picture of a cartoon dog) and a simultaneously presented auditory stimulus (i.e. a barking sound). Functional magnetic resonance imaging (fMRI) revealed two distinct networks of cortical regions that processed preferentially either spatially congruent or spatially incongruent AV stimuli. Whereas earlier visual areas responded preferentially to incongruent AV stimulation, higher visual areas of the temporal and parietal cortex (left inferior temporal gyrus [ITG], right posterior superior temporal gyrus/sulcus [pSTG/STS], left intra-parietal sulcus [IPS]) and frontal regions (left pre-central gyrus [PreCG], left dorsolateral pre-frontal cortex [DLPFC]) responded preferentially to congruent AV stimulation. A position-resolved analysis revealed three robust cortical representations for each of the four visual stimulus locations in retinotopic visual regions corresponding to the representation of the horizontal meridian in area V1 and at the dorsal and ventral borders between areas V2 and V3. While these regions of interest (ROIs) did not show any significant effect of spatial congruency, we found subregions within ROIs in the right hemisphere that showed an incongruency effect (i.e. an increased fMRI signal during spatially incongruent compared to congruent AV stimulation). We interpret this finding as a correlate of spatially distributed recurrent feedback during mismatch processing: whenever a spatial mismatch is detected in multisensory regions (such as the IPS), processing resources are re-directed to low-level visual areas.
Aversive Conditioning of Spatial Position Sharpens Neural Population-Level Tuning in Visual Cortex and Selectively Alters Alpha-Band Activity

Abstract:Processing capabilities for many low-level visual features are experientially malleable, aiding sighted organisms in adapting to dynamic environments. Explicit instructions to attend a specific visual field location influence retinotopic visuocortical activity, amplifying responses to stimuli appearing at cued spatial positions. It remains undetermined both how such prioritization affects surrounding nonprioritized locations, and if a given retinotopic spatial position can attain enhanced cortical representation through experience rather than instruction. The current report examined visuocortical response changes as human observers (N = 51, 19 male) learned, through differential classical conditioning, to associate specific screen locations with aversive outcomes. Using dense-array EEG and pupillometry, we tested the preregistered hypotheses of either sharpening or generalization around an aversively associated location following a single conditioning session. Competing hypotheses tested whether mean response changes would take the form of a Gaussian (generalization) or difference-of-Gaussian (sharpening) distribution over spatial positions, peaking at the viewing location paired with a noxious noise. Occipital 15 Hz steady-state visual evoked potential responses were selectively heightened when viewing aversively paired locations and displayed a nonlinear, difference-of-Gaussian profile across neighboring locations, consistent with suppressive surround modulation of nonprioritized positions. Measures of alpha-band (8-12 Hz) activity were differentially altered in anterior versus posterior locations, while pupil diameter exhibited selectively heightened responses to noise-paired locations but did not evince differences across the nonpaired locations. These results indicate that visuocortical spatial representations are sharpened in response to location-specific aversive conditioning, while top-down influences indexed by alpha-power reduction exhibit posterior generalization and anterior sharpening.SIGNIFICANCE STATEMENT It is increasingly recognized that early visual cortex is not a static processor of physical features, but is instead constantly shaped by perceptual experience. It remains unclear, however, to what extent the cortical representation of many fundamental features, including visual field location, is malleable by experience. Using EEG and an aversive classical conditioning paradigm, we observed sharpening of visuocortical responses to stimuli appearing at aversively associated locations along with location-selective facilitation of response systems indexed by pupil diameter and EEG alpha power. These findings highlight the experience-dependent flexibility of retinotopic spatial representations in visual cortex, opening avenues toward novel treatment targets in disorders of attention and spatial cognition.
The brain can develop conflicting multisensory principles to guide behavior

Abstract:Midbrain multisensory neurons undergo a significant postnatal transition in how they process cross-modal (e.g. visual-auditory) signals. In early stages, signals derived from common events are processed competitively; however, at later stages they are processed cooperatively such that their salience is enhanced. This transition reflects adaptation to cross-modal configurations that are consistently experienced and become informative about which correspond to common events. Tested here was the assumption that overt behaviors follow a similar maturation. Cats were reared in omnidirectional sound thereby compromising the experience needed for this developmental process. Animals were then repeatedly exposed to different configurations of visual and auditory stimuli (e.g. spatiotemporally congruent or spatially disparate) that varied on each side of space and their behavior was assessed using a detection/localization task. Animals showed enhanced performance to stimuli consistent with the experience provided: congruent stimuli elicited enhanced behaviors where spatially congruent cross-modal experience was provided, and spatially disparate stimuli elicited enhanced behaviors where spatially disparate cross-modal experience was provided. Cross-modal configurations not consistent with experience did not enhance responses. The presumptive benefit of such flexibility in the multisensory developmental process is to sensitize neural circuits (and the behaviors they control) to the features of the environment in which they will function. These experiments reveal that these processes have a high degree of flexibility, such that two (conflicting) multisensory principles can be implemented by cross-modal experience on opposite sides of space even within the same animal.
"Congruent" and "opposite" Neurons: Sisters for Multisensory Integration and Segregation.

Abstract:Experiments reveal that in the dorsal medial superior temporal (MSTd) and the ventral intraparietal (VIP) areas, where visual and vestibular cues are integrated to infer heading direction, there are two types of neurons with roughly the same number. One is "congruent" cells, whose preferred heading directions are similar in response to visual and vestibular cues; and the other is "opposite" cells, whose preferred heading directions are nearly "opposite" (with an offset of 180 degrees) in response to visual vs. vestibular cues. Congruent neurons are known to be responsible for cue integration, but the computational role of opposite neurons remains largely unknown. Here, we propose that opposite neurons may serve to encode the disparity information between cues necessary for multisensory segregation. We build a computational model composed of two reciprocally coupled modules, MSTd and VIP, and each module consists of groups of congruent and opposite neurons. In the model, congruent neurons in two modules are reciprocally connected with each other in the congruent manner, whereas opposite neurons are reciprocally connected in the opposite manner. Mimicking the experimental protocol, our model reproduces the characteristics of congruent and opposite neurons, and demonstrates that in each module, the sisters of congruent and opposite neurons can jointly achieve optimal multisensory information integration and segregation. This study sheds light on our understanding of how the brain implements optimal multisensory integration and segregation concurrently in a distributed manner.
No effect of spatial congruence on rapid temporal recalibration to audiovisual asynchrony

Abstract:The brain integrates multisensory information to construct coherent perceptual representations based on spatial and temporal congruence. Intriguingly, multisensory timing perception can be flexibly calibrated. Repeated exposure to audiovisual asynchrony induces shifts in subjective simultaneity (temporal recalibration). Spatial congruence is known to serve as a grouping cue for recalibration when the audiovisual temporal relationship is ambiguous during exposure. A single exposure to audiovisual asynchrony can also trigger temporal recalibration (rapid recalibration). However, it has been suggested that the underlying mechanisms of these temporal recalibrations differ. Here, we examined whether spatial congruence can be a grouping cue for rapid recalibration when audiovisual pairs are not defined by temporal relationships. Participants made a simultaneity judgment for a pair of audiovisual stimuli after adapting three consecutive stimuli once in a "light–sound–light" or "sound–light–sound" order with an equal temporal interval. The spatial positions of the adapting stimuli were manipulated as an audiovisual pair from the same position (e.g., left) and the remaining stimulus from another position (e.g., right). In three experiments, the spatial congruence of the audiovisual adapting stimuli did not show a modulatory effect, while we replicated the rapid recalibration effects. Rather, rapid recalibration occurred according to the temporal order of the first light and sound. Our findings suggest that, in contrast to temporal recalibration with repeated exposure, the perceptual systems underlying rapid recalibration simply combine individual visual and auditory inputs based on the order in which they arrive.

The Responses of Single Neurons in the Temporal Visual Cortical Areas of the Macaque when More Than One Stimulus is Present in the Receptive Field.

Abstract:Neurons in the temporal visual cortical areas of primates have large receptive fields, which can show considerable selectivity for what the stimulus is irrespective of exactly where it is in the visual field. This is called translation invariance. However, such results have been found when there is only one stimulus in the visual field. The question arises of how the visual system operates in a cluttered environment. To investigate this we measured the responses of neurons with face-selective responses in the cortex in the anterior part of the superior temporal sulcus of rhesus macaques performing a visual fixation task. We found that the response of neurons to an effective face centred 8.5° from the fovea was decreased to 71% if an ineffective face stimulus for that cell was present at the fovea. In a similar way, introduction of a parafoveal ineffective face stimulus decreased the responses of these neurons to an effective face stimulus at the fovea to 75%. In addition to these interactions, it was found that an effective stimulus object at the fovea produced a larger response than when it was parafoveal, and that this weighting towards an object at the fovea was also seen when more than one object was present in the visual field. The implication of this weighting of the responses of neurons towards objects at the fovea, even in an environment with more than one object present, is that the output of the visual system provides information to subsequent systems particularly about objects at the fovea, so that learning about these objects (and less about other objects elsewhere in the visual field) is facilitated.

Abstract:Abstract Sensory systems must continuously adapt to optimally encode stimuli encountered within the natural environment. The prevailing view is that such optimal coding comes at the cost of increased ambiguity, yet to date, prior studies have focused on artificial stimuli. Accordingly, here we investigated whether such a trade-off between optimality and ambiguity exists in the encoding of natural stimuli in the vestibular system. We recorded vestibular nuclei and their target vestibular thalamocortical neurons during naturalistic and artificial self-motion stimulation. Surprisingly, we found no trade-off between optimality and ambiguity. Using computational methods, we demonstrate that thalamocortical neural adaptation in the form of contrast gain control actually reduces coding ambiguity without compromising the optimality of coding under naturalistic but not artificial stimulation. Thus, taken together, our results challenge the common wisdom that adaptation leads to ambiguity and instead suggest an essential role in underlying unambiguous optimized encoding of natural stimuli.

The role of binocular disparity and attention in the neural representation of multiple moving stimuli in the visual cortex

Abstract:Segmenting visual scenes into distinct objects and surfaces is a fundamental visual process, with stereoscopic depth and motion serving as crucial cues. However, how the visual system uses these cues to segment multiple objects is not fully understood. We investigated how neurons in the middle-temporal (MT) cortex of macaque monkeys represent overlapping surfaces at different depths, moving in different directions. Neuronal activity was recorded from three male monkeys during discrimination tasks under varying attention conditions. We found that neuronal responses to overlapping surfaces showed a robust bias toward the binocular disparity of one surface over the other. The disparity bias of a neuron was positively correlated with the neuron’s disparity preference for a single surface. In two animals, neurons preferring near disparities of single surfaces (near neurons) showed a near bias for overlapping stimuli, while neurons preferring far disparities (far neurons) showed a far bias. In the third animal, both near and far neurons displayed a near bias, though the near neurons showed a stronger near bias. All three animals exhibited an initial near bias across neurons relative to the average of the responses to the individual surfaces. Although attention modulated neuronal responses, the disparity bias was not caused by attention. We also found that the effect of attention was consistent with object-based, rather than feature-based attention. We proposed a model in which the pool size of the neuron population that weighs the responses to individual stimulus components can be variable. This model is a novel extension of the standard normalization model and provides a unified explanation for the disparity bias across animals. Our results reveal how MT neurons encode multiple stimuli moving at different depths and present new evidence of response modulation by object-based attention. The disparity bias allows subgroups of neurons to preferentially represent individual surfaces of multiple stimuli at different depths, thereby facilitating segmentation.

Abstract:Binocular mechanisms for visual processing are thought to enhance spatial acuity by combining matched input from the two eyes. Studies in the primary visual cortex of carnivores and primates have confirmed that eye-specific neuronal response properties are largely matched. In recent years, the mouse has emerged as a prominent model for binocular visual processing, yet little is known about the spatial frequency tuning of binocular responses in mouse visual cortex. Using calcium imaging in awake mice of both sexes, we show that the spatial frequency preference of cortical responses to the contralateral eye is ∼35% higher than responses to the ipsilateral eye. Furthermore, we find that neurons in binocular visual cortex that respond only to the contralateral eye are tuned to higher spatial frequencies. Binocular neurons that are well matched in spatial frequency preference are also matched in orientation preference. In contrast, we observe that binocularly mismatched cells are more mismatched in orientation tuning. Furthermore, we find that contralateral responses are more direction-selective than ipsilateral responses and are strongly biased to the cardinal directions. The contralateral bias of high spatial frequency tuning was found in both awake and anesthetized recordings. The distinct properties of contralateral cortical responses may reflect the functional segregation of direction-selective, high spatial frequency-preferring neurons in earlier stages of the central visual pathway. Moreover, these results suggest that the development of binocularity and visual acuity may engage distinct circuits in the mouse visual system.<b>SIGNIFICANCE STATEMENT</b> Seeing through two eyes is thought to improve visual acuity by enhancing sensitivity to fine edges. Using calcium imaging of cellular responses in awake mice, we find surprising asymmetries in the spatial processing of eye-specific visual input in binocular primary visual cortex. The contralateral visual pathway is tuned to higher spatial frequencies than the ipsilateral pathway. At the highest spatial frequencies, the contralateral pathway strongly prefers to respond to visual stimuli along the cardinal (horizontal and vertical) axes. These results suggest that monocular, and not binocular, mechanisms set the limit of spatial acuity in mice. Furthermore, they suggest that the development of visual acuity and binocularity in mice involves different circuits.
Functional Differentiation of Mouse Visual Cortical Areas Depends upon Early Binocular Experience

Abstract:The mammalian visual cortex contains multiple retinotopically defined areas that process distinct features of the visual scene. Little is known about what guides the functional differentiation of visual cortical areas during development. Recent studies in mice have revealed that visual input from the two eyes provides spatiotemporally distinct signals to primary visual cortex (V1), such that contralateral eye-dominated V1 neurons respond to higher spatial frequencies than ipsilateral eye-dominated neurons. To test whether binocular visual input drives the differentiation of visual cortical areas, we used two-photon calcium imaging to characterize the effects of juvenile monocular deprivation (MD) on the responses of neurons in V1 and two higher visual areas, LM (lateromedial) and PM (posteromedial). In adult mice of either sex, we find that MD prevents the emergence of distinct spatiotemporal tuning in V1, LM, and PM. We also find that, within each of these areas, MD reorganizes the distinct spatiotemporal tuning properties driven by the two eyes. Moreover, we find a relationship between speed tuning and ocular dominance in all three areas that MD preferentially disrupts in V1, but not in LM or PM. Together, these results reveal that balanced binocular vision during development is essential for driving the functional differentiation of visual cortical areas. The higher visual areas of mouse visual cortex may provide a useful platform for investigating the experience-dependent mechanisms that set up the specialized processing within neocortical areas during postnatal development. SIGNIFICANCE STATEMENT Little is known about the factors guiding the emergence of functionally distinct areas in the brain. Using in vivo Ca 2+ imaging, we recorded visually evoked activity from cells in V1 and higher visual areas LM (lateromedial) and PM (posteromedial) of mice. Neurons in these areas normally display distinct spatiotemporal tuning properties. We found that depriving one eye of normal input during development prevents the functional differentiation of visual areas. Deprivation did not disrupt the degree of speed tuning, a property thought to emerge in higher visual areas. Thus, some properties of visual cortical neurons are shaped by binocular experience, while others are resistant. Our study uncovers the fundamental role of binocular experience in the formation of distinct areas in visual cortex.

Abstract:The neural substrates of multisensory perception and integration are still obscure, especially at the cortical level. Alternative viewpoints emphasize (1) 'bottom-up' processes, where different modalities converge in higher order multisensory areas, or (2) 'top-down' projections from multimodal to unimodal areas. In this anatomic study, we use anterograde tracer injections in parietal (8 monkeys) and auditory (3 monkeys) association areas, and demonstrate direct projections to areas V1 and V2 in the calcarine fissure (i.e. the peripheral visual field representation). The laminar signature, with terminations in layers 1 and/or 6, could be consistent with feedback-type connections. A subset of connections from parietal areas, however, branch to both V1 and a ventral extrastriate area (TEO or TEp). Thus, the direct connections to early visual areas V1 and V2 may well operate in conjunction with polysynaptic pathways in a densely parallel network.
