Abstract:Aging is typically accompanied by a decline in working memory (WM) capacity, even in the absence of pathology. Proficient WM requires cognitive control processes which can retain goal-relevant information for easy retrieval and resolve interference from irrelevant information. Aging has been associated with a reduced ability to resolve proactive interference (PI) in WM , leading to impaired retrieval of goal-relevant information. It remains unclear how age-related differences in the ability to resolve PI in WM are related to patterns of resting-state functional connectivity (rsFC) in the brain. Here we investigated the association between PI in WM and rsFC cross-sectionally (n = 237) and 5-year longitudinally (n = 134) across the adult life span by employing both seed-based and data-driven approaches. Results revealed that the ability to resolve PI was associated with differential patterns of rsFC in younger/middle-aged adults (25-60y) and older adults (65-80y) in two clusters centered in the vermis and caudate. Specifically, more PI was associated with stronger inferior frontal gyrus – vermis connectivity and weaker inferior frontal gyrus – caudate connectivity in older adults, while younger/middle-aged adults showed associations in the opposite directions with the identified clusters. Whole brain multivariate pattern analyses showed age-differential patterns of rsFC indicative of age-related structural decline and age-related compensation. The current results show that rsFC is associated with the ability to control PI in WM and that these associations are modulated by age.

Abstract:Previous magnetic resonance imaging (MRI) research suggests that aging is associated with a decrease in the functional interconnections within and between groups of locally organized brain regions (modules). Further, this age-related decrease in the segregation of modules appears to be more pronounced for a task, relative to a resting state, reflecting the integration of functional modules and attentional allocation necessary to support task performance. Here, using graph-theoretical analyses, we investigated age-related differences in a whole-brain measure of module connectivity, system segregation , for 68 healthy, community-dwelling individuals 18–78 years of age. We obtained resting-state, task-related (visual search), and structural (diffusion-weighted) MRI data. Using a parcellation of modules derived from the participants' resting-state functional MRI data, we demonstrated that the decrease in system segregation from rest to task (i.e., reconfiguration) increased with age, suggesting an age-related increase in the integration of modules required by the attentional demands of visual search. Structural system segregation increased with age, reflecting weaker connectivity both within and between modules. Functional and structural system segregation had qualitatively different influences on age-related decline in visual search performance. Functional system segregation (and reconfiguration) influenced age-related decline in the rate of visual evidence accumulation (drift rate), whereas structural system segregation contributed to age-related slowing of encoding and response processes (nondecision time). The age-related differences in the functional system segregation measures, however, were relatively independent of those associated with structural connectivity.

Abstract:Alterations in the default mode network (DMN) are associated with aging. We assessed age-dependent changes of DMN interactions and correlations with a battery of neuropsychological tests, to understand the differences of DMN directed connectivity between young and older subjects. Using a novel multivariate analysis method on resting-state functional MRI data from fifty young and thirty-one healthy older subjects, we calculated intra- and inter-DMN 4-nodes directed pathways. For the old subject group, we calculated the partial correlations of inter-DMN pathways with: psychomotor speed and working memory, executive function, language, long-term memory and visuospatial function. Pathways connecting the DMN with visual and limbic regions in older subjects engaged at BOLD low frequency and involved the dorsal posterior cingulate cortex (PCC), whereas in young subjects, they were at high frequency and involved the ventral PCC. Pathways combining the sensorimotor (SM) cortex and the DMN, were SM efferent in the young subjects and SM afferent in the older subjects. Most DMN efferent pathways correlated with reduced speed and working memory. We suggest that the reduced sensorimotor efferent and the increased need to control such activities, cause a higher dependency on external versus internal cues thus suggesting how physical activity might slow aging.

Abstract:Abstract Human learning and cognitive functions change with age and experience, with late-developed complex cognitive functions, particularly those served by the prefrontal cortex, showing more age-dependent variance. Reading as a complex process of constructing meaning from print uses the left prefrontal cortex and may show a similar aging pattern. In this study, we delineated the lifespan developmental changes in the neural substrates and functional connectivity for visual semantic processing from childhood (age 6) to late adulthood (age 74). Different from previous studies that reported aging as a form of activation or neuronal changes, we examined additionally how the functional connectivity networks changed with age. A cohort of 122 Chinese participants performed semantic and font-size judgment tasks during functional magnetic resonance imaging. Although a common left-lateralized neural system including the left mid-inferior prefrontal cortex was recruited across all participants, the effect of age, or reading experience, is evident as 2 contrastive developmental patterns: a declining trend in activation strength and extent and an increasing trend in functional connections of the network. This study suggests that visual semantic processing is not prone to cognitive decline, and that continuous reading until old age helps strengthen the functional connections of reading-related brain regions.

Abstract:The functional connectome changes with aging. We systematically evaluated aging related alterations in the functional connectome using a whole-brain connectome network analysis in 39,675 participants in UK Biobank project. We used adaptive dense network discovery tools to identify networks directly associated with aging from resting-state fMRI data. We replicated our findings in 499 participants from the Lifespan Human Connectome Project in Aging study. The results consistently revealed two motor-related subnetworks (both permutation test p-values <0.001) that showed a decline in resting-state functional connectivity (rsFC) with increasing age. The first network primarily comprises sensorimotor and dorsal/ventral attention regions from precentral gyrus, postcentral gyrus, superior temporal gyrus, and insular gyrus, while the second network is exclusively composed of basal ganglia regions, namely the caudate, putamen, and globus pallidus. Path analysis indicates that white matter fractional anisotropy mediates 19.6% (p<0.001, 95% CI [7.6% 36.0%]) and 11.5% (p<0.001, 95% CI [6.3% 17.0%]) of the age-related decrease in both networks, respectively. The total volume of white matter hyperintensity mediates 32.1% (p<0.001, 95% CI [16.8% 53.0%]) of the aging-related effect on rsFC in the first subnetwork.

Abstract:Cognitive task performance can be supported through multiple neural pathways, a concept referred to as brain degeneracy. We used a novel approach to consider brain degeneracy during a visual short-term memory task (VSTM) across the adult lifespan in the Cam-CAN study (n = 113, 23-87 years old). Our main goal was to identify subgroups of participants whose VSTM performance was characterized by distinct brain activation patterns. First, we identified seven brain modules that responded similarly to the VSTM task and resembled previously identified functional networks (adjusted mutual information [aMI] = 0.45). Subsequently, latent profile analysis revealed four distinct subgroups of participants. Each subgroup was characterized by different recruitment patterns of these brain modules, predominantly in the frontal control module (FCM), visual module (VM), and default mode module (DMM). Subgroups did not differ in demographics or task performance. However, associations between brain activity and performance varied across subgroups, particularly in the FCM, suggesting that individuals may use different cognitive operations to perform the VSTM task. Further analyses revealed group differences in white matter integrity, mostly in the uncinate fasciculus, suggested that individual differences in structural brain properties may shape the different brain activation patterns. Altogether, our study contributes to our understanding of how multiple neural pathways could underlie cognitive performance.

Abstract:Age-related alterations of functional brain networks contribute to cognitive decline. Current theories indicate that age-related intrinsic brain functional reorganization may be a critical marker of cognitive aging. Yet, little is known about how intrinsic interhemispheric functional connectivity changes with age in adults, and how this relates to critical executive functions. To address this, we examined voxel-mirrored homotopic connectivity (VMHC), a metric that quantifies interhemispheric communication, in 93 healthy volunteers (age range: 19–85) with executive function assessment using the Delis-Kaplan Executive Function System (D-KEFS) scales. Resting functional MRI data were analyzed to assess VMHC, and then a multiple linear regression model was employed to evaluate the relationship between age and the whole-brain VMHC. We observed age-related reductions in VMHC of ventromedial prefrontal cortex (vmPFC) and hippocampus in the medial temporal lobe subsystem, dorsal anterior cingulate cortex and insula in salience network, and inferior parietal lobule in frontoparietal control network. Performance on the color-word inhibition task was associated with VMHC of vmPFC and insula, and VMHC of vmPFC mediated the relationship between age and CWIT inhibition reaction times. The percent ratio of correct design scores in design fluency test correlated positively with VMHC of the inferior parietal lobule. The current study suggests that brain interhemispheric functional alterations may be a promising new avenue for understanding age-related cognitive decline.

Abstract:Abstract Functional brain networks have preserved architectures in rest and task; nevertheless, previous work consistently demonstrated task-related brain functional reorganization. Efficient rest-to-task functional network reconfiguration is associated with better cognition in young adults. However, aging and cognitive load effects, as well as contributions of intra- and inter-network reconfiguration, remain unclear. We assessed age-related and load-dependent effects on global and network-specific functional reconfiguration between rest and a spatial working memory (SWM) task in young and older adults, then investigated associations between functional reconfiguration and SWM across loads and age groups. Overall, global and network-level functional reconfiguration between rest and task increased with age and load. Importantly, more efficient functional reconfiguration associated with better performance across age groups. However, older adults relied more on inter-network reconfiguration of higher cognitive and task-relevant networks. These reflect the consistent importance of efficient network updating despite recruitment of additional functional networks to offset reduction in neural resources and a change in brain functional topology in older adults. Our findings generalize the association between efficient functional reconfiguration and cognition to aging and demonstrate distinct brain functional reconfiguration patterns associated with SWM in aging, highlighting the importance of combining rest and task measures to study aging cognition.

Lifespan differences in hippocampal subregion connectivity patterns during movie watching

Abstract:Age-related episodic memory decline is attributed to functional alternations in the hippocampus. Less clear is how aging affects the functional connections of the hippocampus to the rest of the brain during episodic memory processing. We examined fMRI data from the CamCAN dataset, in which a large cohort of participants watched a movie (N = 643; 18-88 years), a proxy for naturalistic episodic memory encoding. We examined connectivity profiles across the lifespan both within the hippocampus (anterior, posterior), and between the hippocampal subregions and cortical networks. Aging was associated with reductions in contralateral (left, right) but not ipsilateral (anterior, posterior) hippocampal subregion connectivity. Aging was primarily associated with increased coupling between the anterior hippocampus and regions affiliated with Control, Dorsal Attention and Default Mode networks, yet decreased coupling between the posterior hippocampus and a selection of these regions. Differences in age-related hippocampal-cortical, but not within-hippocampus circuitry selectively predicted worse memory performance. Our findings comprehensively characterize hippocampal functional topography in relation to cognition in older age, suggesting that shifts in cortico-hippocampal connectivity may be sensitive markers of age-related episodic memory decline.

Abstract:Executive functions, essential for daily life, are known to be impaired in older age. Some executive functions, including working memory updating and value-based decision-making, are specifically sensitive to age-related deterioration. While their neural correlates in young adults are well-described, a comprehensive delineation of the underlying brain substrates in older populations, relevant to identify targets for modulation against cognitive decline, is missing. Here, we assessed letter updating and Markov decision-making task performance to operationalize these trainable functions in 48 older adults. Resting-state functional magnetic resonance imaging was acquired to quantify functional connectivity (FC) in task-relevant frontoparietal and default mode networks. Microstructure in white matter pathways mediating executive functions was assessed with diffusion tensor imaging and quantified by tract-based fractional anisotropy (FA). Superior letter updating performance correlated with higher FC between dorsolateral prefrontal cortex and left frontoparietal and hippocampal areas, while superior Markov decision-making performance correlated with decreased FC between basal ganglia and right angular gyrus. Furthermore, better working memory updating performance was related to higher FA in the cingulum bundle and the superior longitudinal fasciculus. Stepwise linear regression showed that cingulum bundle FA added significant incremental contribution to the variance explained by fronto-angular FC alone. Our findings provide a characterization of distinct functional and structural connectivity correlates associated with performance of specific executive functions. Thereby, this study contributes to the understanding of the neural correlates of updating and decision-making functions in older adults, paving the way for targeted modulation of specific networks by modulatory techniques such as behavioral interventions and non-invasive brain stimulation.

Nineteen healthy older adults and 22 young adults were recruited, and underwent two functional magnetic resonance imaging (fMRI) scanning sessions (i.e., resting-state and localizer tasks) along with a visual word perceptual processing task. We examined age-related alterations in resting-state functional connectivity (FC) within the word network, as well as between the word network and other networks, and tested their associations with behavioral performance in word and symbol-form processing. We found that compared with young adults, old adults exhibited increased FC between the two word-selective regions located in the left and right ventral occipitotemporal cortex (vOT).Additionally, the FC between these two word-selective regions and other non-word selective regions was increased in older adults compared with young adults.Moreover, the FC alterations were correlated with individuals' behavioral performance in visual word perceptual processing. These results suggest that cognitive decline in visual word perception is associated with decreased segregation both within and beyond the word network in the aging brain. Our findings support the neural dedifferentiation hypothesis for the cognitive decline in visual word processing, and advance our understanding of the interactive neural specialization theory.

Abstract:Aging is accompanied by a decline of working memory, an important cognitive capacity that involves stimulus-selective neural activity that persists after stimulus presentation. Here, we unraveled working memory dynamics in older human adults (male and female) including those diagnosed with mild cognitive impairment (MCI) using a combination of behavioral modeling, neuropsychological assessment, and MEG recordings of brain activity. Younger adults (male and female) were studied with behavioral modeling only. Participants performed a visuospatial delayed match-to-sample task under systematic manipulation of the delay and distance between sample and test stimuli. Their behavior (match/nonmatch decisions) was fit with a computational model permitting the dissociation of noise in the internal operations underlying the working memory performance from a strategic decision threshold. Task accuracy decreased with delay duration and sample/test proximity. When sample/test distances were small, older adults committed more false alarms than younger adults. The computational model explained the participants' behavior well. The model parameters reflecting internal noise (not decision threshold) correlated with the precision of stimulus-selective cortical activity measured with MEG during the delay interval. The model uncovered an increase specifically in working memory noise in older compared with younger participants. Furthermore, in the MCI group, but not in the older healthy controls, internal noise correlated with the participants' clinically assessed cognitive integrity. Our results are consistent with the idea that the stability of working memory contents deteriorates in aging, in a manner that is specifically linked to the overall cognitive integrity of individuals diagnosed with MCI.

Abstract:In the era of functional brain networks, our understanding of how they evolve across life in a healthy population remains limited. Here, we investigate functional connectivity across the human lifespan using magnetoencephalography in a cohort of 792 healthy individuals, categorized into young (13 to 30 yr), middle (31 to 54 yr), and late adulthood (55 to 80 yr). Employing corrected imaginary phase-locking value, we map the evolving landscapes of connectivity within delta, theta, alpha, beta, and gamma classical frequency bands among brain areas. Our findings reveal significant shifts in functional connectivity patterns across all frequency bands, with certain networks exhibiting increased connectivity and others decreased, dependent on the frequency band and specific age groups, showcasing the dynamic reorganization of neural networks as age increases. This detailed exploration provides, to our knowledge, the first all-encompassing view of how electrophysiological functional connectivity evolves at different life stages, offering new insights into the brain's adaptability and the intricate interplay of cognitive aging and network connectivity. This work not only contributes to the body of knowledge on cognitive aging and neurological health but also emphasizes the need for further research to develop targeted interventions for maintaining cognitive function in the aging population.

Abstract:GOAL: Working memory (WM) is a memory system with a limited capacity that can process and store information temporarily in the performing of cognitive tasks. Despite WM is known to be influenced by age, the difficulty of tasks and trained or not from behavior studies, little is known about their relationships from the aspect of the brain functional network. Our goal was to explore the factor of aging-related changes of WM with brain functional networks.METHODS: In this study, 25 healthy elderly and 23 healthy young volunteers were recruited for electroencephalogram (EEG) recording during the visual WM task with four difficulty levels (1-4 backs). In each back, we repeat the experiment with four sessions, and we add training sections between session one and session two as well as between session two and session three. However, we remove any training section between session three and session four in order to evaluate the impact of forgetting on WM in different age groups. After the experiment, we utilized graph theoretical analysis to characterize the brain functional network in three frequency bands (alpha, beta, and theta).RESULTS: From the well-designed experiment, we found that physiological aging influences brain network connectivity and makes the functional brain network less differentiated. Moreover, there is an inverse relationship between alpha activity and WM load for the elderly group, which is absent in the young group. At the same time, theta band activity will be correlated with behavioral performance for the elderly group with WM training between sessions, which is also absent in the young group. To further study the influence of difficulty of tasks and training on the WM, we distinguish the tasks with quantified topological characteristics, and the classification results manifest that the training is more effective for the young group. Finally, through the establishment of a brain map before and after training, we find that the right parietal lobe plays an important role in the training of WM for the elderly group whereas the beta band plays an important role in WM for both the elderly group and the young group.CONCLUSION: Taken together, our findings clarify the underlying mechanism of WM under different frequency bands in terms of physiological aging, the influence of training, and task difficulty.SIGNIFICANCE: the working memory capacities can be uncovered in terms of the combination of three-way ANOVA and EEG-based graph theoretical analysis.

Abstract:During aging the inter-individual variability in both the neural and behavioral functions is likely to be emphasized. Decreased competence particularly in working memory and general executive control compromises many aspects of the quality of life also within the nonclinical population. We aimed, first, to clarify the brain basis of visual working memory and inhibition during multi-stage natural-like task performance, and second, to identify associations between variation in task-related neural activity and relevant cognitive skills, namely inhibition and general working memory capacity. We recorded, using magnetoencephalography (MEG), the neural modulations associated with encoding, maintenance, and retrieval, as well as interference suppression during a visual working memory task in older adults. We quantified the neural correlates of these cognitive processes through two complementary approaches: evoked responses and oscillatory activity. Neural activity during memory retrieval and interference suppression were correlated with behavioral measures of task switching and general executive functions. Our results show that general inhibitory control induced frontocentral neural modulation across a broad range of frequencies whereas domain-specific inhibition was limited to right posterior areas. Our findings also suggest that modulations particularly in phase-locked evoked neural activity can be reliably associated with explicit measures of cognitive skills, with better inhibitory control linked with an early neural effect of distractor inhibition during retrieval. In general, we show that exploiting the inherent inter-individual variability in neural measures and behavioral markers of cognition in aging populations can help establish reliable links between specific brain functions and their behavioral manifestations.

Recent research focuses on aging processes determined by resting state (RS) functional connectivity (FC), but little is known about differences in FC during RS and cognitive task conditions in elderly participants. The purpose of this study is to compare FC within and between the cognitive control (CCN) and dorsal attention network (DAN) at RS and during a cognitive task using functional near-infrared spectroscopy (fNIRS). In a matched, neurodegenerative high-risk cohort comprising early (n = 98; 50–65 y) and late (n = 98; 65–85 y) elder subjects, FC was measured at RS and during performance of the Trail Making Test (TMT) via fNIRS. Both, under RS and task conditions our results revealed a main effect for age, characterized by reduced FC for late elder subjects within the left inferior frontal gyrus. During performance of the TMT, negative correlations of age and FC were confirmed in various regions of the CCN and DAN. For the whole sample, FC of within-region connections was elevated, while FC between regions was decreased at RS. The results confirm a reorganization of functional brain connectivity with increasing age and cognitive demands.

Abstract:Magnetoencephalography recordings of functional brain activity reveal large-scale cortical networks that are associated with cognition. The correlation of individualised networks with age and cognitive performance (age effects and cognitive performance effects respectively) was studied using a large cross-sectional healthy cohort (N=612, 18-88 years old) and accounting for a comprehensive set of confounds. Age effects were found in time-averaged functional networks in five canonical frequency bands (delta, theta, alpha, beta, gamma) that are consistent with the posterior-anterior shift with age observed in functional magnetic resonance imaging. Evidence from cognitive performance effects in time-averaged networks suggested the importance of maintaining alpha-band activity for cognitive health. A more detailed description of the functional activity was obtained by adopting an established machine learning approach (the Hidden Markov Model). Ten transient large-scale cortical networks with fast dynamics (~100 ms) were identified, which provided insight into age and cognitive performance effects that were not observed in the time-averaged analyses. The time spent in most networks increased with age, whereas the time spent in frontal networks decreased. The cognitive performance effects for the transient networks suggested that age effects in the frontal networks are compensatory. Thus, our study suggests both the maintenance of functional activity (lesser age effects) and the recruitment of compensatory functional activity can co-occur to produce good cognitive performance in older individuals. The time-averaged and transient functional networks have been made publicly available as a resource.

Abstract:Sustained attention is important for maintaining cognitive function and autonomy during ageing, yet older people often show reductions in this domain. The role of the underlying neurobiology is not yet well understood, with most neuroimaging studies primarily focused on fMRI. Here, we utilise sMRI to investigate the relationships between age, structural brain volumes and sustained attention performance. Eighty-nine healthy older adults (50-84 years, Mage 65.5 (SD=8.4) years, 74 f) underwent MRI brain scanning and completed two sustained attention tasks: a rapid visual information processing (RVP) task and sustained attention to response task (SART). Independent hierarchical linear regressions demonstrated that greater volumes of white matter hyperintensities (WMH) were associated with worse RVP_A' performance, whereas greater grey matter volumes were associated with better RVP_A' performance. Further, greater cerebral white matter volumes were associated with better SART_d' performance. Importantly, mediation analyses revealed that both grey and white matter volumes completely mediated the relationship between ageing and sustained attention. These results explain disparate attentional findings in older adults, highlighting the intervening role of brain structure.

Abstract:Aging is associated with changes in brain functional patterns as well as cognition. The present research sought to investigate longitudinal changes in whole brain functional connectivity strength (FCS) and cognitive performance scores in very old cognitively unimpaired individuals. We studied 34 cognitively normal elderly individuals at both baseline and 4-year follow-up (baseline age = 78 +/- 3.14 years) with resting-state functional magnetic resonance imaging (r-fMRI), structural MRI scans, and neuropsychological assessments conducted. Voxel-based whole brain FCS was calculated and we found that bilateral superior parietal and medial frontal regions showed decreased FCS, while the supplementary motor area (SMA) and insula showed increased FCS with age, along with a decrease in bilateral prefrontal cortical thickness. The changes of FCS in left precuneus were associated with an aging-related decline in global cognition. Taken together, our results suggest changes in FCS with aging with the precuneus as a hub and this may underlie changes in global cognition that accompany aging. These findings help better understand the normal aging mechanism.
