Abstract:Breast cancer (BC) is the most common malignant tumor in women. There are different risk characteristics and treatment strategies for different subtypes of BC. The tumor microenvironment (TME) is of great significance for understanding the occurrence, development, and metastasis of tumors. The TME plays an important role in all stages of BC metastasis, immune monitoring, immune response avoidance, and drug resistance, and also plays an important role in the diagnosis, prevention, and prognosis of BC. Smart nanosystems have broad development prospect in the regulation of the BC drug delivery based on the response of the TME. In particular, TME-responsive nanoparticles cleverly utilize the abnormal features of BC tissues and cells to achieve targeted transport, stable release, and improved efficacy. We here present a review of the mechanisms underlying the response of the TME to BC to provide potential nanostrategies for future BC treatment.
Induction of intrinsic apoptotic pathway and cell cycle arrest via baicalein loaded iron oxide nanoparticles as a competent nano-mediated system for triple negative breast cancer therapy

Abstract:Magnetic nanoparticles have shown an increasing number of applications in the field of molecular medicine. In the present study we investigated the controlled synthesis of biocompatible polymer coated iron oxide nanoparticles (Fe2O3) loaded with baicalein and evaluated these for their drug loading and release behaviour. Moreover, the loaded particles were evaluated for their anti-proliferation, cell cycle arrest and apoptosis activation properties in triple negative breast cancer cells. The structure, morphology and magnetic properties of the prepared materials were studied by using scanning electronic microscopy, transmission electronic microscopy, Fourier transform infrared spectroscopy, X-ray diffraction analysis, thermogravimetric analysis, dynamic light scattering analysis and zeta potential analysis. DLS and TEM analysis confirms that the size of the synthesized iron oxide nanoparticles are about 90–100 nm and that they are extremely crystalline and spherical in nature. FTIR analysis confirms that the baicalein molecules were conjugated with PEG-coated iron oxide nanoparticles. X-ray diffraction patterns indicate that the magnetic nanoparticles were highly pure with a spinel structure. Baicalein loaded nanoparticles show a controlled release profile in response to various pH levels. We found significant cell cycle arrest at various phases in the treated group and subsequent apoptotic cell death to be evidenced by AO/EtBr, DAPI and PI of fluorescence microscopy analysis. The TUNEL assay evidences that DNA damage occurs in the treated cells. Further baicalein loaded iron oxide nanoparticles exhibit significant down-regulation of the anti-apoptotic protein Bcl-2 and up-regulation of pro-apoptotic proteins evidenced by Western blotting. Our findings clearly demonstrate that baicalein loaded iron oxide nanoparticles could efficiently deliver the drug of interest, and initiate and execute the apoptotic process in triple negative breast cancer cells. This could open a new window in polymer surface chemistry to develop metal oxide based drug delivery systems for all kinds of cancer therapeutics.

Abstract:The aim was to develop a nanoscale drug delivery system with enzyme responsive and acid sensitive particle size and intelligent degradation aiming to research the inhibitory effect on breast cancer. The delivery system addressed the problems of tissue targeting, cellular internalization, and slow drug release at the target site, which could improve the efficiency of drug delivery and provide a feasible therapeutic approach for breast cancer. The acid sensitive functional material DSPE-PEG 2000 -dyn-PEG-R9 was synthesized by Michael addition reaction. Then, the berberine plus baicalin intelligent micelles were prepared by thin-film hydration. Subsequently, we characterized the physical and chemical properties of berberine plus baicalin intelligent micelles, evaluated its anti-tumor effects in vivo and in vitro . The target molecule was successfully synthesized, and the intelligent micelles showed excellent chemical and physical properties, delayed drug release and high encapsulation efficiency. In vitro and in vivo experiments also confirmed that the intelligent micelles could effectively target tumor sites, penetrate tumor tissues, enrich in tumor cells, inhibit tumor cell proliferation, inhibit tumor cell invasion and migration, and induce tumor cell apoptosis. Berberine plus baicalin intelligent micelles have excellent anti-tumor effects and no toxicity to normal tissues, which provides a new potential drug delivery strategy for the treatment of breast cancer.

The pH-sensitive chondroitin sulphate-based nanoparticles for co-delivery of doxorubicin and berberine enhance the treatment of breast cancer

Abstract:In the tumor microenvironment (TME), cancer associated fibroblasts (CAFs) facilitate drug resistance and tumor metastasis. Therefore, more and more attention has been focused on the regulation of TME by preventing the cross-talk between tumor cells and CAFs in the treatment of breast cancer. In this study, we have combined the benefits of deep drug penetration, pH sensitivity, and tumor-targeting delivery to prepare chondroitin sulphate (CS)-based nanomicelles (BBR/CS-DOX) for the co-delivery of doxorubicin (DOX) and berberine (BBR). A unique MCF-7 + MRC-5 co-cultured cell model and 4 T1 + NIH3T3 co-implanted mice model, were established to simulate the TME of breast cancer (BC). As expected, BBR/CS-DOX could accumulate in tumor egion, be taken up by both tumor cells and CAFs, and improve drug absorption and retention. Compared with free drugs, BBR/CS-DOX demonstrated stonger pro-apoptotic and anti-metastatic effect in vitro and in vivo, respectively the histological studies showed that BBR/CS-DOX efficiently prevented the activation of fibroblasts, inhibited extracellular matrix (ECM) deposition, and decreased tumor angiogenesis, showing superior anti-tumor efficacy. In summary, BBR/CS-DOX has the potential to significantly enhance the therapeutic effect of breast cancer through inhibiting the "CAFs-tumor cells" crosstalk, and has promising clinical application prospects.

Abstract:A bismuth (Bi)‐based core‐shell nanoparticle, which can serve as both a photothermal converter and a computed tomography (CT) imaging contrast agent and loaded with a chemotherapeutic drug (doxorubicin), is modified with a cancer‐targeting ligand. It is guided by the ligand to precisely target the breast tumors and achieves the selective imaging and destruction of breast tumors. Due to the complexity and heterogeneity of cancer, the clear and accurate tumor imaging and image‐guided multimodal synergistic therapy are highly in demand. Here, bismuth (Bi)‐based mesoporous‐silica‐coated nanoparticles (BMSNs) are successfully fabricated with the silica shell thickness being lower than the Bi nanoparticle diameter. Then, an MCF‐7 breast cancer‐targeting peptide (termed AR) is modified onto the surface of BMSNs. The obtained nanoparticles (BMSN‐AR) show a significantly higher loading of doxorubicin hydrochloride (DOX). The resultant BMSN‐AR‐DOX can agglomerate in the tumor site, enabling enhanced computed tomography (CT) imaging of the whole tumor. Under 808 nm near‐infrared (NIR) laser irradiation, the BMSN‐AR‐DOX also effectively converts light energy into thermal energy to achieve synergistic chemo‐photothermal therapy in vivo. The chemo‐photothermal combination therapy is found to be more effective than chemotherapy or photothermal therapy alone. This work demonstrates that tumor‐homing peptides can guide nanoparticles to tumors and allow the nanoparticles to enhance cancer imaging and photothermal/chemo combination therapy.

Abstract:Introduction Currently, the main treatment for advanced breast cancer is still chemotherapy. Immunological and chemical combination therapy has a coordinated therapeutic effect and achieves some efficacy. However, the immunosuppressive tumor microenvironment is a major cause for the failure of immunotherapy in breast cancer. CpG oligodeoxynucleotides can activate the tumor immune microenvironment to reverse the failure of immunotherapy. Methods In this study, we designed an amphiphilic peptide micelle system (Co-LMs), which can targeted delivery of the immune adjuvant CpG and the chemotherapeutic drug doxorubicin to breast cancer tumors simultaneously. The peptide micelle system achieved tumor microenvironment pH and redox-sensitive drug release. Results and Discussion Co-LMs showed 2.3 times the antitumor efficacy of chemotherapy alone and 5.1 times the antitumor efficacy of immunotherapy alone in triple-negative breast cancer mice. Co-LMs activated cytotoxic CD8+ T lymphocytes and CD4+ T cells in mice to a greater extent than single treatments. We also found that Co-LMs inhibited the metastasis of circulating tumor cells in the bloodstream to some extent. These results indicate that the Co-LMs offer a promising therapeutic strategy against triple-negative breast cancer.

Abstract:Triple-negative breast cancer (TNBC) is an aggressive type of breast cancer for which effective therapies are lacking. Targeted remodeling of the immunosuppressive tumor microenvironment (TME) and activation of the body's immune system to fight tumors with well-designed nanoparticles have emerged as pivotal breakthroughs in tumor treatment. To simultaneously remodel the immunosuppressive TME and trigger immune responses, we designed two potential therapeutic nanodelivery systems to inhibit TNBC. First, the bromodomain-containing protein 4 (BRD4) inhibitor JQ1 and the cyclooxygenase-2 (COX-2) inhibitor celecoxib (CXB) were coloaded into chondroitin sulfate (CS) to obtain CS@JQ1/CXB nanoparticles (NPs). Then, the biomimetic nanosystem MM@P3 was prepared by coating branched polymer poly(β-amino ester) self-assembled NPs with melittin embedded macrophage membranes (MM). Both in vitro and in vivo, the CS@JQ1/CXB and MM@P3 NPs showed excellent immune activation efficiencies. Combination treatment exhibited synergistic cytotoxicity, antimigration ability, and apoptosis-inducing and immune activation effects on TNBC cells and effectively suppressed tumor growth and metastasis in TNBC tumor-bearing mice by activating the tumor immune response and inhibiting angiogenesis. In summary, this study offers a novel combinatorial immunotherapeutic strategy for the clinical TNBC treatment.

Abstract:Many metal complexes not only had excellent cytotoxic antitumor effects but also could function as a positive immunomodulatory to improve antitumor effects by modifying the local tumor microenvironment. Herein, a dual-target nanotherapeutics (MP3/ACPP/AE105@NPs) with uPAR targeting and tumor microenvironment-responsive ability was developed by using AE105 as the targeting ligand and ACPP as the tumor microenvironment-responsive peptide to locate the metal complex to cells. Assisted by the surface modification, MP3/ACPP/AE105@NPs demonstrated excellent cellular uptake of the drugs in in vitro experiments, thereby enhancing the therapeutic utility of the loaded metal complex. The nanotherapeutics induced the excessive ROS generation by inhibiting the activity of TrxR and modulated those proteins which were related to metastasis through inhibiting ERK/AKT activation mediated by FAK in MDA-MB-231 cells. This nanotherapeutics could also significantly improve the therapeutic benefits in vivo accompanied by reduced toxic side effects. Importantly, MP3/ACPP/AE105@NPs treatment stimulated immunotherapeutic response, which was indicated by the activation of tumor-infiltrated cytotoxic T cells, and improved the maturation of dendritic cells (DCs), and the proliferation-inhibitory effect of tumor-associated M2 macrophages. Taken together, the results suggested that this dual-targeted nanotherapeutics offered new opportunities for boosting the synergistic treatment of breast cancer with chemotherapy and immunotherapy.

Baicalein sensitizes triple negative breast cancer MDA-MB-231 cells to doxorubicin via autophagy-mediated down-regulation of CDK1

Abstract:Triple negative breast cancer (TNBC) is a kind of refractory cancer with poor response to conventional chemotherapy. Recently, the combination of baicalein and doxorubicin was reported to exert a synergistic antitumor effect on breast cancer. However, the underlying mechanism how baicalein sensitizes breast cancer cells to doxorubicin remains to be elucidated. Here, it was found that 20 μM baicalein increased the autophagy markers including the ratio of LC3B II/I, GFP-LC3 punctate aggregates and down-regulation of p62 expression, and up-regulated mitophagy marker PINK1 and Parkin in TNBC MDA-MB-231 cells as well. In contrast, doxorubicin decreased the levels of autophagy markers, and significantly up-regulated CDK1 in MDA-MB-231 cells. Pretreatment with baicalein markedly inhibited the doxorubicin-induced decrease in autophagy markers and up-regulation of CDK1, which was reversed by the autophagy inhibitor 3-Methyladenine. Moreover, baicalein alleviated the doxorubicin-induced expression and phosphorylation (at Ser616) of mitochondrial fission protein Drp1. Intriguingly, the autophagy inhibitor 3-Methyladenine also significantly weakened the effect of baicalein on doxorubicin-induced viability decrease and apoptosis in MDA-MB-231 cells. Taken together, our data indicate that baicalein improves the chemosensitivity of TNBC cells to doxorubicin through promoting the autophagy-mediated down-regulation of CDK1, also suggest a novel strategy for prevention of TNBC in the future.

Abstract:Problems, such as toxic side effects and drug resistance of chemoradiotherapy, target therapy and immunotherapy accompanying the current anti-cancer treatments, have become bottlenecks limiting the clinical benefit for patients. Therefore, it is urgent to find promising anti-cancer strategies with higher efficacy and lesser side effects. Baicalein, a flavonoid component derived from the Chinese medicine scutellaria baicalensis, has been widely studied for its remarkable anti-cancer activity in multiple types of malignancies both at the molecular and cellular levels. Baicalein exerts its anti-tumor effects by inhibiting angiogenesis, invasion and migration, inducing cell apoptosis and cell cycle arrest, as well as regulating cell autophagy, metabolism, the tumor microenvironment and cancer stem cells with no obvious toxic side effects. The role of classic signaling pathways, such as PI3K/AKT/mTOR, MAPK, AMPK, Wnt/β-catenin, JAK/STAT3, MMP-2/-9, have been highlighted as the major targets for baicalein exerting its anti-malignant potential. Besides, baicalein can regulate the relevant non-coding RNAs, such as lncRNAs, miRNAs and circ-RNAs, to inhibit tumorigenesis and progression. In addition to the mentioned commonalities, baicalein shows some specific anti-tumor characteristics in some specific cancer types. Moreover, the preclinical studies of the combination of baicalein and chemoradiotherapy pave the way ahead for developing baicalein as an adjunct treatment with chemoradiotherapy. Our aim is to summary the role of baicalein in different types of cancer with its mechanisms based on in vitro and in vivo experiments, hoping providing proof for baicalein serving as an effective and safe compound for cancer treatment in clinic in the future.

Preparation and evaluation of folate-modified albumin baicalin-loaded nanoparticles for the targeted treatment of breast cancer

Abstract:Baicalin (BA) has been shown to be one of the natural compounds with anti-proliferative activity against numerous cancer cell lines, but its application is restricted due to its rapid metabolism and non-targeting to specific tissues. This study exploited folic acid (FA)-conjugated bovine serum albumin (BSA) nanoparticles (NPs) loaded with baicalin (FA-BSANPs/BA) by desolvation crosslinking method to improve its bioavailability and therapeutic efficacy. The optimized FA-BSANPs/BA showed spherical shape and uniform distribution. The average particle size, zeta potential, encapsulation efficiency (EE) and drug loading (DL) of FA-BSANPs/BA were 228.41±2.36 nm, -32.70±1.27 mV, 76.88±0.59% and 7.41±0.23%, respectively. X-ray diffraction (XRD) and thermogravimetric (TG) analysis exhibited that the drug was amorphous in the particles. The results of drug release behavior in vitro demonstrated that FA-BSANPs/BA releases BA slowly and continuously. In vitro cytotoxicity test results suggested that the IC50 of FA-BSANPs/BA and BA on MCF-7 cells was 16.7 and 75.96 μg/mL, respectively. The uptake efficiency of FA-BSANPs was significantly higher than that of BSANPs, which leads to a stronger potential for apoptosis. In vivo antitumor studies showed that FA-BSANPs/BA can greatly inhibit tumor growth compared with BA and exhibited the ability to target tumors in MCF-7 xenograft tumor-bearing nude mice. Biomimetic Cucurbitacin B-Polydopamine Nanoparticles for Synergistic Chemo-Photothermal Therapy of Breast Cancer

Abstract:Breast cancer is the most common malignant tumor in women. Researchers have found that the combined use of multiple methods to treat tumors is a promising strategy. Here, we have developed a biomimetic nano-platform PDA@MB for tumor targeted photothermal therapy (PTT) combined with chemotherapy. The 4T1 cell membrane loaded with cucurbitacin B (CuB) was used to coat polydopamine (PDA) nanoparticles, which gave PDA@MB nanoparticles the ability to target tumors and escape immune cells from phagocytosis. PDA@MB showed excellent photothermal performance including high photothermal conversion efficiency and photostability, and exhibited outstanding in vitro PTT effect under NIR laser irradiation. The high temperature ruptured the PDA@MB membrane to release CuB, which changed the tumor hypoxic environment, down-regulated the FAK/MMP signaling pathway, and significantly inhibited the metastasis and proliferation of tumor cells. The results of in vivo experiments indicated that the tumor growth of the 4T1 mouse tumor model was significantly inhibited. Additionally, toxicity studies showed that PDA@MB had good biocompatibility and safety. In conclusion, this study provides a promising chemo-photothermal therapy (CPT) nano-platform for precise and effective breast cancer therapy.

Abstract:Polymeric nanoparticles are developed, that deliver chemotherapeutics and scavenge damage‐associated molecular patterns simultaneously to inhibit chemotherapy‐induced breast cancer metastasis. The cationic nanoparticles can adsorb cell‐free nucleic acids based on charge–charge interaction, which downregulates the expression of Toll‐like receptors and then reduces the secretion of inflammatory cytokines. Chemotherapy, although effective against primary tumors, may promote metastasis by causing the release of proinflammatory factors from damaged cells. Here, polymeric nanoparticles that deliver chemotherapeutics and scavenge proinflammatory factors simultaneously to inhibit chemotherapy‐induced breast cancer metastasis are developed. The cationic nanoparticles can adsorb cell‐free nucleic acids (cfNAs) based on charge–charge interaction, which downregulates the expression of Toll‐like receptors and then reduces the secretion of inflammatory cytokines. Through in vitro structural optimization, cationic polyamidoamine (PAMAM) dendrimers modified with drug‐binding dodecyl groups and diethylethanolamine surface groups (PAMAM‐G3‐C125‐DEEA20) exhibit the most desirable combination of nanoparticle size (≈140 nm), drug loading, cytotoxicity, cfNA binding, and anti‐inflammatory activity. In the mouse models of breast cancer metastasis, paclitaxel‐loaded nanoparticles reduce serum levels of cfNAs and inflammatory cytokines compared with paclitaxel treatment alone and inhibit both primary tumor growth and tumor metastasis. Additionally, no significant side effects are detected in the serum or major organs. These results provide a strategy to deliver chemotherapeutics to primary tumors while reducing the prometastatic effects of chemotherapy.

Here, we prepared mixed nano-sized formulations composed of pH-sensitive liposomes (Ber/Ru486@CLPs) and small-sized nano-micelles (Dox@CLGs). These liposomes and nano-micelles were modified by chondroitin sulfate (CS) to selectively target breast cancer cells. Methods: Ber/Ru486@CLPs and Dox@CLGs were prepared by thin-film dispersion and ethanol injection, respectively. To mimic actual TME, the in vitro "condition medium of fibroblasts + MCF-7" cell model and in vivo " 4T1/NIH-3T3" co-implantation mice model were established to evaluate the anti-tumor effect of drugs. Results: The physicochemical properties showed that Dox@CLGs and Ber/Ru486@CLPs were 28 nm and 100 nm in particle size, respectively. In vitro experiments showed that the mixed formulations significantly improved drug uptake and inhibited cell proliferation and migration. The in vivo anti-tumor studies further confirmed the enhanced anti-tumor capabilities of Dox@CLGs + Ber/Ru486@CLPs, including smaller tumor volumes, weak collagen deposition, and low expression levels of α-SMA and CD31 proteins, leading to a superior anti-tumor effect. Conclusion: In brief, this combination therapy based on Dox@CLGs and Ber/Ru486@CLPs could effectively inhibit tumor development, which provides a promising approach for the treatment of breast cancer. Keywords: drug delivery, cancer-associated fibroblasts, glucocorticoids, combination therapy Breast cancer is one of the most prevalent malignant tumors worldwide, ranking as the second leading cause of cancer-related fatalities among female subjects. 1,2 Elevated rates of cancer progression, metastasis, and chemotherapy resistance are pivotal factors associated with the diminished survival of breast cancer patients. 3,4 Research has proven that the tumor microenvironment (TME) significantly contributes to these outcomes. 5,6 TME consists of the extracellular matrix (ECM), cytokines and various tumor stromal cells, including fibroblasts, immune cells, inflammatory cells and endothelial cells. 7 The combined strategy of TME-regulating the pro-apoptotic activities has attracted more and more attention in anti-tumor therapy. Cancer-associated fibroblasts (CAFs), as the most abundant stromal cells in TME, are activated from normal fibroblasts, 8 and play an important role in fostering tumor growth. 9,10 Researchers have shown that CAFs could induce epithelial-mesenchymal transitions (EMT) of tumor cells by secreting TGF-β, 11 ultimately leading to tumor metastasis. 12 In addition, CAFs are directly implicated in immunosuppression and drug resistance. They possess the ability to reprogram tumor cell metabolism, and protecting them from apoptosis triggered through treatment. 13–17 Due to the specific distribution and important role of CAFs within tumors, they are increasingly becoming a therapeutic target for the treatment of breast cancer. Berberine (Ber), an isoquinoline quaternary alkaloid extracted from rhizoma coptidis, cortex phellodendri and other medicinal plants, has been proven to possess multiple pharmacological characteristics, including anti-inflammation, antioxidative, anti-diabetic and antitumor activities. Recent research showed that Ber could inhibit CAFs activation, 18,19 collagen deposition, and TGF-β secretion, 20 resulting in the effective suppression of cancer cell metastasis. Membrane Fusion Liposomes Deliver Antifibrotic and Chemotherapeutic Drugs Sequentially to Enhance Tumor Treatment Efficacy by Reshaping Tumor Microenvironment.

Abstract:The intricate tumor microenvironment in triple-negative breast cancer (TNBC) hampers chemotherapy and immunotherapy efficacy due to dense extracellular matrix (ECM) by tumor-associated fibroblasts (TAFs). Nanoparticle-based therapies, especially "all in one" nanoparticles, have shown great potential in combined drug delivery strategies to reshape the tumor microenvironment and enhance therapeutic efficiency. However, these "all in one" nanoparticles suffer from limitations in targeting different target cells, uncontrollable dosing ratio, and disregarding the impact of delivery schedules. This study prepared cell membrane fusion liposomes (TAFsomes and CCMsomes) to load FDA-approved anti-fibrotic drug pirfenidone (PFD/TAFsomes) and anti-tumor drug doxorubicin (DOX/CCMsomes). These liposomes can specifically target TAFs cells and tumor cells, and combined administration can effectively inhibit TAF activity, reshape the TME, and significantly enhance the tumor chemotherapy efficacy. Combined drug delivery defeats "all in one" liposomes (DOX/PFD/Liposomes, DOX/PFD/TAFsomes, and DOX/PFD/CCMsomes) by flexibly adjusting the drug delivery ratio. Moreover, an asynchronous delivery strategy that optimizes the administration schedule not only further improves the therapeutic effect, but also amplifies the effectiveness of α-PD-L1 immunotherapy by modulating the tumor immune microenvironment. This delivery strategy provides a personalized treatment approach with clinical translation potential, providing new ideas for enhancing the therapeutic effect against solid tumors such as TNBC. Abstract:Abstract Background Poor targeting and penetration of chemotherapy drugs in solid tumors, and the development of resistance to chemotherapeutic agents are currently hindering the therapy of breast cancer; meanwhile, breast cancer metastasis is one of the leading causes of death in breast cancer patients. With the development of nanotechnology, nanomaterials have been widely used in tumor therapy. Results A multi-functional nano-platform containing gambogic acid (GA) and paclitaxel (PTX) was characterized by a small size, high encapsulation efficiency, slow release, long systemic circulation time in vivo, showed good targeting and penetrability to tumor tissues and tumor cells, and exhibited higher anti-tumor effect and lower systemic toxicity in BALB/c mice bearing 4T1 tumor. GA not only overcame the multidrug resistance of PTX by inhibiting P-glycoprotein (P-gp) activity in MCF-7/ADR cells, but also inhibited MDA-MB-231 cells migration and invasion, playing a crucial role in preventing and treating the lung metastasis of breast cancer caused by PTX; meanwhile, the synergistic anti-tumor effect of GA and PTX has also been verified in vitro and in vivo experiments. Conclusion Our data described the better recognition and penetration of tumor cells of R9dGR-modified versatile nanosystems containing GA and PTX, which exerted one stone three birds clinical therapeutic efficacy of multifunctionality.

Abstract:Breast-to-brain metastatic cells can interact with the surrounding cells, including astrocytes and microglia, to generate a pro-tumorigenic niche. Breast-to-brain metastasis can be treated using a dual strategy of eliminating metastatic tumor cells and normalizing their localized microenvironment. The effective accumulation of drugs at the action site of metastasis is crucial to realizing the above strategy, especially when dealing with the bloodbrain barrier (BBB)-penetrating and tumor-targeting tactics. Here, we establish an in-situ microenvironmenttailored micelle (T-M/siRNA) to co-deliver therapeutic siRNA and paclitaxel (PTX) into the breast-to-brain metastasis. Anchored with a D-type cyclic peptide, T-M/siRNA can penetrate the BBB and subsequently target the brain metastases. Upon internalization by metastatic tumor cells, T-M/siRNA can release PTX in the highlevel glutathione (GSH), resulting in killing cancer cells. Meanwhile, the micellar structure is dissociated, resulting in lowering the charge density to release the loaded siRNA that can targeted downregulate the expression of protocadherin 7 (PCDH7). Treatment of model mice revealed that T-M/siRNA can inhibit the abnormal activation of astrocytes and immunosuppressive activation of microglia, resulting in significantly enhanced synergistic anti-tumor efficacy. This study indicates that the micelle system can serve as a hopeful strategy to treat breast-to-brain metastasis.

Abstract:Certain chemotherapeutics can induce tumor cells' immunogenic cell death (ICD), release tumor antigens, and thereby trigger personalized antitumor immune responses. Co-delivery of adjuvants using nanocarriers could amplify the ICD-induced tumor-specific immunity achieving a synergistic chemo-immunotherapeutic effect. However, complicated preparation, low drug loading efficiency, and potential carrier-associated toxicity are the major challenges that limited its clinical applications. Herein, a carrier-free core–shell nanoparticle (MPLA-CpG-sMMP9-DOX, MCMD NPs) was constructed by facile self-assembly of spherical nucleic acids (SNA) with two adjuvants of CpG ODN and monophosphoryl lipid A (MPLA) as a core and doxorubicin (DOX) radially around the dual-adjuvants SNA as a shell. The results demonstrated that MCMD NPs could enhance drugs accumulation in tumors, and release DOX upon enzymatic degradation of matrix metalloproteinase-9 (MMP-9) peptide in the tumor microenvironment (TME), which enhanced the direct-killing effect of DOX on tumor cells. The core of MPLA-CpG SNA efficiently boosted the ICD-induced antitumor immune response to further attack tumor cells. Thus, MCMD NPs achieved a synergistic therapeutic effect of chemo-immunotherapy with reduced off-target toxicity. This study provided an efficient strategy for the development of a carrier-free nano-delivery system for enhanced cancer chemo-immunotherapy.

Abstract:Cancer is a complex disease that displays physiomorphological transformation in different surrounding microenvironments. Therefore, the single treatment modalities are relatively less effective, and their efficiency varies with tumor cell physiology, leading to the development of tumor resistance. Combinatorial therapeutic approaches, such as chemo-photothermal therapy, are promising for efficiently mitigating tumor progression irrespective of cancer physiology. Nanotechnology has played a significant role in this regard. Therefore, the present study reports the synthesis of poly(acrylic acid)-tetraethylene glycol (PAA-TEG)-coated BaSO4 nanoparticles (NPs) with enhanced solubility, dispersibility, and X-ray attenuation. Next, nanocomposites (NCs) are synthesized by loading BaSO4 NPs with the therapeutic drug triiodobenzoic acid (Tiba) and the photosensitizer IR780 using a lipid coating. These fabricated NCs are analyzed for dual-modal imaging (fluorescence and X-ray-based imaging) properties and chemo-phototherapeutic ability against two-dimensional (2D) and three-dimensional (3D) cultures of A549 cells. Furthermore, A549 cells are morphologically and physiologically aggravated into potent malignant cells using tobacco leaf extract (TE), and the variation in the therapeutic effect of NCs compared to cisplatin is determined. The synthesized NCs display enhanced encapsulation and excellent synergistic anticancer activity through the generation of reactive oxygen species (ROS), mitochondrial damage, and genotoxicity. Also, the NCs are more potent in inhibiting cancer cell growth than cisplatin, and their impact is unaltered in the presence or absence of TE pretreatment of A549 cells. The present study holds significant potential for various theranostic applications, which are highly desired for laparoscopic image-guided lung cancer therapy.
Coordinated regulation of BACH1 and mitochondrial metabolism through tumor-targeted self-assembled nanoparticles for effective triple negative breast cancer combination therapy

Abstract:The poor prognosis of triple negative breast cancer (TNBC) results from a lack of approved targeted therapies coupled with aggressive proliferation and metastasis, which is associated with high recurrence and short overall survival. Here we developed a strategy by employing tumor-targeted self-assembled nanoparticles to coordinately regulate BACH1 (BTB domain and CNC homology 1) and mitochondrial metabolism. The BACH1 inhibitor hemin and mitochondria function inhibitor berberine derivative (BD) were used to prepare nanoparticles (BH NPs) followed by the modification of chondroitin sulfate (CS) on the surface of BH NPs to achieve tumor targeting (CS/BH NPs). CS/BH NPs were found to be able to inhibit tumor migration and invasion by significantly decreasing the amounts of tumor cell metabolites, glycolysis and metastasis-associated proteins, which were related to the inhibition of BACH1 function. Meanwhile, decreased mitochondrial membrane potential, activated caspase 3/9 and increased ROS production demonstrated coordinated regulation of BACH1 and mitochondrial metabolism. In a xenograft mice model of breast cancer, CS/BH NPs significantly inhibited tumor growth and metastasis due to the synergetic effect of hemin and BD without showing obvious toxicities for major organs. In sum, the results of efficacy and safety experiments suggest potential clinical significance of the prepared self-assembled CS/BH nanoparticles for the treatment of TNBC.
