Abstract:Objective: To investigate the structural changes of aorta, and evaluate the effects of atorvastatin on the remodeling of thoracic aorta in spontaneously hypertensive rats (SHR). Methods: Twelve eight-week-old SHR were randomized into atorvastatin treated group (ATV group,n=6) and distilled water group (DW group,n=6); Wistar-Kyoto rats (WKY) were used as normal controls. Atorvastatin was administered to ATV group for 10 weeks by gavage in mixture with distilled water(1 ml); the latter two groups were given the same amount of distilled water by gavage for 10 weeks. Systolic blood pressure of caudal artery was examined before and after treatment, and serum concentrations of total cholesterol, triglycerides and HDL-C were measured. Wall thickness, media thickness, medial cross-sectional area and lumen diameter of thoracic aorta were assessed with computed video processing. Results: Systolic blood pressure in ATV group was markedly lower than that in DW group (P<0.01). Compared with DW group and WKY group, serum concentrations of total cholesterol, triglycerides and HDL-C in ATV group were significantly lower (P<0.01,P<0.05). Wall thickness, media thickness, and medial cross-sectional area to lumen ratio in DW group were significantly higher than those in WKY group and ATV group (P<0.01,P<0.05), but no such difference was found between WKY group and ATV group (P>0.05). Conclusion: Vascular structural changes of aorta are due to the alteration of the vessel wall in early stage of SHR. Atorvastatin can markedly improve vascular remodeling.
Rosuvastatin Reverses Hypertension-Induced Changes in the Aorta Structure and Endothelium-Dependent Relaxation in Rats Through Suppression of Apoptosis and Inflammation.

Abstract:Abstract: Vascular remodeling is one of the most critical complications caused by hypertension. Previous studies have demonstrated that rosuvastatin has anti-inflammatory, antioxidant, and antiplatelet effects and therefore can be used to treat cardiovascular disease. In this study, we explored the beneficial effects of rosuvastatin in reversing aortic remodeling in spontaneously hypertensive rats. After treating with different doses of rosuvastatin, its antilipid, antiapoptosis, and anti-inflammatory effects were determined. We also examined whether rosuvastatin can improve the structure and function of the aorta. We found that rosuvastatin treatment of spontaneously hypertensive rats for 2 months at 2 different doses can effectively reduce the media thickness of the aorta compared with the control group. Similarly, rosuvastatin improved the vascular relaxation function of the aortic rings at a high level of acetylcholine in vitro. Mechanistically, it was found that rosuvastatin increased the expression of endothelial nitric oxide synthase and plasma nitrite/nitrate levels. Besides, rosuvastatin suppressed the apoptosis and inflammation and upregulated the expression of gap-junction complex connexin 43 both in media and endothelium. Finally, rosuvastatin inhibited the AT1R/PKCα/HSP70 signaling transduction pathway. In summary, these findings demonstrated that rosuvastatin could improve the vascular structure and function mainly by increasing endothelial nitric oxide synthase expression and preventing apoptosis and inflammation. This study provided evidence that rosuvastatin has beneficial effects in reversing the remodeling of the aorta due to hypertension.
Effects of Xuezhikang on Ventricular Remodeling in Spontaneously Hypertensive Rats and the Possible Mechanism

Abstract:OBJECTIVE To observe the effects of Xuezhikang on left ventricular hypertrophy and myocardial interstitial fibrosis in spontaneously hypertensive rats(SHR) and the possible mechanism.METHODS Thirty male SHRs were randomly treated with Xuezhikang(300 mg?kg?1?d?1)(XZK group,n=10),simvastatin(5 mg?kg?1?d?1)(SIM group,n=10) or 0.9% normal saline(SHR group,n=10).Age-matched Wistar-Kyoto rats(WKY)(WKY group,n=10) were also gavaged by 0.9% normal saline for 12 weeks.Left ventricular weight index(LVWI) was calculated.In addition,the serum lipid concentrations,the blood pressure levels,the myocardial collagen volume fraction(CVF) and perivascular collagen area(PVCA),the serum levels of procollagen typeⅠC-terminal peptide(PⅠCP),the levels of myocardial superoxide dismutase(SOD) and malondialdehyde(MDA) were also measured in this study.RESULTS After 12 weeks of treatment,the LVWI(P<0.05),CVF(P<0.01),PVCA(P<0.01) and serum PⅠCP concentrations(P<0.01) of XZK group and SIM group were significantly lower than those of SHR group.Myocardial SOD activity and MDA concentrations had no significant changes among four groups.CONCLUSION This study demonstrats that Xuezhikang significantly inhibited left ventricular hypertrophy and myocardial interstitial fibrosis was independent of its antihypercholesterolemic role and exhibited the same effects as simvastatin.The levels of myocardial SOD activity and MDA concentrations seem to be uninvolved in the reverse effects of Xuezhikang.
[Effects of Atorvastatin on Endothelium Protection in Spontaneously Hypertensive Rats].

Abstract:OBJECTIVE:To investigate the effects of two different dosage of atorvastatin on endothelium protection in spontaneously hypertensive rats (SHR).METHODS:SHRs (n=18) were randomly divided into three groups (n=6): SHR control group, 50 mg atorvastatin group and 10 mg group. Six male Wistar-Kyoto rats were selected as normal control group (WKY group). All animals were given vehicle once a day by gavage for 10 weeks. Systolic blood pressure (SBP) was measured before and after treatment with atorvastatin every 2 weeks. Plasma NO and vWF were measured by nitrate reductase method and double antibodies ELISA, respectively. Plasma lipids were also measured.RESULTS:SBP in all SHR groups was much higher than that in WKY group before experiment (P<0.01). Compared with SHR control group, SBP significantly decreased in 50 mg atorvastatin group at 6, 8 and 10 weeks and in 10 mg atorvastatin group merely at 10 weeks (P<0.05 or P<0.01). The plasma levels of NO in SHR control group was significantly lower than those in WKY group (P<0.01). After 10 weeks, plasma NO levels in 50 mg and 10 mg atorvastatin groups were markedly higher than those in SHR control group (P<0.01 or P<0.05). The plasma levels of vWF in SHR control group was significantly higher than those in WKY group (P<0.01). After 10 weeks, plasma vWF levels in 50 mg and 10 mg atorvastatin groups were markedly lower than those in SHR control group (P<0.01). Plasma lipids in 50 mg and 10 mg atorvastatin groups were lower than those in SHR control group (P<0.05 or P<0.01).CONCLUSION:Atorvastatin can decrease blood pressure and significantly improve endothelial function in SHR by increasing plasma NO level and decreasing plasma vWF level.
Tissue-specific Effects of Atorvastatin on 3-Hydroxy-3-methylglutaryl-coenzyme A Reductase Expression and Activity in Spontaneously Hypertensive Rats

Abstract:Aim: Cardiovascular remodeling is closely associated with cholesterol and is attenuated by statins. The spontaneously hypertensive rat (SHR) has a low serum cholesterol level and evident cardiovascular remodeling. The aims of the present study were to characterize the effects of atorvastatin on tissue cholesterol content and 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase expression and activity in four tissues from SHR: liver, heart, aorta and kidney. Methods: SHR and normotensive Wistar-Kyoto rats (WKY) were treated daily with atorvastatin (50 mg/kg) for 8 weeks. Cholesterol levels of serum and tissues (liver, heart, aorta and kidney) were determined by commercial enzymatic methods. Western blot analysis and high performance liquid chromatogram (HPLC) were used to assay the expression and activity of enzyme respectively. Results: Treatment with atorvastatin decreased cholesterol content and HMG-CoA reductase expression and activity in all four tissues of SHR. However, in WKY, atorvastatin only altered HMG-CoA reductase in liver, where the protein expression was upregulated but the enzyme activity was decreased. Conclusion: The present study demonstrates that the effects of atorvastatin on tissue cholesterol content and HMG-CoA reductase are strain- and tissue-specific.

Abstract:Objective To evaluate the effects ofatorvastatin on aortic blood pressure and vascular endothelial function in spontaneously hypertensive rats(SHR).Methods The 8-week-old SHR were randomized into atorvastatin treated group (ATV group,n=6),distilled water group (DW group,n=6),and Wistar-Kyoto (WKY) rats as normal controls.Atorvastatin [50 mg/(kg,d)] and appropriate amounts of distilled water were administered to ATV group for 10 weeks by gavage.Blood pressure of caudal artery was examined before and after treatment,and serum titres of total cholesterol (TC),triglycerides (TG),high density lipoprotein-cholesterol (HDL-C) and changes of serum endothelin-1 (ET-1),activity of nitric oxide synthase (NOS) in aorta tissue were measured.Results Blood pressure in ATV group was much lower than that in DW group (P＜0.01).Compared with DW group and WKY group,serum titres of TC,TG and HDL-C in ATV group were significantly lower (P＜0.01,P＜0.05).Serum level of ET-1 was significant higher in DW group than in WKY group (P＜0.05),and the activity of NOS in aorta tissue was remarkedly reduced (P＜0.05).However,serum level of ET-1 was decreased (P＜0.05) while the activity of NOS in aorta tissue was elevated after treatment with ATV (P＜0.01).Conclusion Long-term administration ofatorvastatin can effectively reduce blood pressure.Atorvastatin may delay pathological progress of hypertension by ameliorating vascular endothelial function.
Losartan Reduced Connexin43 Expression in Left Ventricular Myocardium of Spontaneously Hypertensive Rats

Abstract:Objective To assess the effect of angiotensin II type 1 (AT 1 ) receptor antagonist losartan on myocardium connexin43 (Cx43) gap junction (GJ) expression in spontaneously hypertensive rats (SHRs) and investigate possible mechanisms. Methods Sixteen 9-week-old male SHRs and 8 age-matched male Wistar-Kyoto (WKY) rats were included in this study. SHRs were randomly divided into two groups to receive losartan at 30 mg/(kg·d) by oral gavage once daily for 8 weeks (SHR-L) or vehicle (0.9% saline) to act as controls (SHR-V); WKY rats receiving vehicle for 8 weeks served as normotensive controls. At the end of the experiment, rats were sacrificed and the hearts were removed. Expressions of Cx43 and nuclear factor-kappaB p65 (NF-κB p65) proteins in all three groups were observed and further investigations on the effect of angiotensin II type 1 receptor antagonist losartan (30 mg/(kg·d), 8 weeks) on Cx43 expression were conducted with Western blot and immunohistochemistry. NF-κB p65 protein in nuclear extracts was determined by Western blot. Results Left ventricular (LV) hypertrophy was prominent in SHRs, Cx43 and NF-κB p65 protein expressions were obviously upregulated and Cx43 distribution was dispersed over the cell surface. Treatment with losarton reduced the over-expressions of Cx43 and NF-κB p65 in LV myocardium. The distribution of Cx43 gap junction also became much regular and confined to intercalated disk after losartan treatment. Conclusion Cx43 level was upregulated in LV myocardium of SHR during early stage of hypertrophy. Angiotensin II type 1 receptor antagonist losartan prevented Cx43 gap junction remodeling in hypertrophied left ventricles, possibly through the NF-κB pathway.
Role of Losartan and Carvedilol on in Extracellular Signal-Regulated Kinases in Myocardium of Spontaneously Hypertensive Rats

Abstract:OBJECTIVE To explore the role of losartan and carvedilol on extracellular signal-regulated kinase (ERK) and mitogen-actirated protein kinase phosphatase-1 (MKP-1) in the myocardium of spontaneously hypertensive rat (SHR). METHODS Twenty-one 14-week old SHRs were randomily divided into 3 groups, 7 rats each. One group was given with carvedilol 30 mg·kg~(-1)·d~(-1), one group was given with losartan 10 mg·kg~(-1)·d~(-1) for 8 weeks, while the SHR sham group and 7 Wistar Kyoto rats(WKY)as control group were routinely given with vehicle (0.5% carboxymethyl cellulose) only for equal time. Then the activity of ERK and the expression of MKP-1 mRNA in myocardium were studied by immuno-precipitation and RT-PCR, respectively. RESULTS ①After administration for 8 weeks, the blood pressure of the SHR group treated with carvedilol or losartan decreased significantly (P0.01). LVW/BW ratio in SHR with carvedilol or losatan treatment was lower than that in SHR sham group (carvedilol group, P0.01, losartan group, P0.05), and the reduction in hypertrophy with losartan treatment was more than with carvedilol (P0.05). ②After treatment in SHR, the activity of ERK in the group with carvedilol or losartan can treatment was lower than that in the SHR control (P0.01), while the expression of MKP-1 mRNA of them was higher (P0.05). CONCLUSION Both carvedilol and losartan can inhibit the activity of ERK, increase the expression of MKP-1 mRNA, and then relieve cardiacl hypertrophy.

Abstract:AIM: To investigate the effects of atorvastatin on blood pressure and expression of cytochrome P450 hydroaylase (CYP) 4A1 in spontaneously hypertensive rats (SHR). METHODS: SHRs (n=18) were randomly divided into three groups: SHR control group, 50 mg atorvastatin (HATV) group and 10 mg (LATV) group. Six male Wistar-Kyoto rats were selected as normal control group (WKY group). All rats were given vehicle once a day by gavage for 10 weeks. Systolic blood pressure (SBP) was measured before and after treatment every 2 weeks. The expressions of CYP 4A1 mRNA and protein in heart, liver, kidney, and aorta were detected by RT-PCR and Western blotting analysis, respectively. Plasma lipids were also measured.RESULTS: SBP in all SHR groups was much higher than that in WKY group before experiment (P0.01). Compared with SHR control group, SBP significantly decreased in HATV group at 6, 8, 10 weeks and in LATV group merely at 10 weeks (P0.01 or P0.05). The Effects of Atorvastatin on Left Ventricular Remodeling in Spontaneously Hypertensive Rats

Abstract:Background Abnormal cardiomyocyte apoptosis and myocardial connective tissue growth factor(CTGF) expression were involved in the development of left ventricular remodeling in hypertension.Statins administration improved left ventricular remodeling in spontaneously hypertensive rats(SHR),but the mechanisms are not clear.Objective To investigate the effects of atorvastatin administered in the early stage on left ventricular remodeling in SHR,and to explore the underlying mechanisms involved.Methods Sixteen male SHR were randomized to receive distilled water(placebo) or atorvastatin [50 mg/(kg·d) SHR-ATV] for 12 weeks.The age matched male Wistar-Kyoto rats(WKY) were gavaged with distilled water as control.Left ventricular remodeling was evaluated by determination of the ratio of left ventricular weight to body weight,cardiomyocyte diameter and interstitial fibrosis.Myocardium CTGF expression level was examined with Western blot,and cardiomyocyte apoptosis with TUNEL method.Results Compared with WKY and placebo,atorvastatin treatment significantly decreased serum TC,TG,LDL-C levels and systolic blood pressure in SHR;atorvastatin significantly inhibited left ventricular remodeling,decreased the ratio of left ventricular weight to body weight [(4.0±0.4) vs(4.7±0.4) mg/g,P<0.05],cardiomyocyte diameter [(16.2±2.8) vs(19.0±1.0) μm,P<0.05] and interstitial fibrosis [(3.3±2.1)% vs(4.5±1.8)%,P<0.05].Atorvastatin seems to have modulating effect in myocardial CTGF and apoptosis manifested by preventing the increases in CTGF,while accentuating the cells apoptosis ratio in placebo SHR[SHR-ATV:(5.2±0.6)% vs placebo:(1.9±0.3)%,P<0.05].ConclusionAtorvastatin administered in the early stage effectively pervented left ventricular remodeling in SHR by inhibiting myocardium CTGF expression and inducing cardiomyocyte apoptosis.
Atorvastatin Improves Cardiac Lipid Metabolism in Spontaneous Hypertensive Rats and Ameliorates Cardiac Hypertrophy

Abstract:Objective To investigate the effect of atorvastatin on cardiac lipid metabolism and cardiac hypertrophy. Methods SHR(n=6) and WKY(n=6) were given atorvastatin 50 mg/kg·d body weight by gavage. After 10 weeks, LV/BW ratio, serum free fatty acid(FFA), peroxisome proliferator-activated receptor α(PPARα) and carnitime palmitoyltransferase(CPT-Ⅰ) mRNA in myocardium were investigated. Results Serum and myocardial free fatty acid in SHR were higher than that in WKY(seurm FFA:SHR:798.2±38.0 vs WKY:354.7±27.7 nmol/L, P0.01); (myocardium:635.0±77.4 vs WKY:245.3±47.3 nmol/L, P0.01). The expression of PPARα and CPT-Ⅰ mRNA in myocardium in SHR was decreased as compared with those in WKY group(0.285±0.062 vs WKY: 0.478±0.093, P0.01; 0.795±0.139 vs WKY: 1.115±0.109, P0.01 respectively), VWI and TDM in SHR were greater than those in WKY (3.16±0.08 vs WKY: 2.99±0.10 g/kg, P0.05; 21.32±1.25 vs WKY: 18.18±0.75 μm, P0.01). Atorvastatin decreased serum fatty acid (571.0±40.8 vs control:798.2±38.0 nmol/L, P0.01) and free fatty acid in cardiac myocytes (287.7±57.7 vs control:635.0±77.4 nmol/L, P0.01). Meanwhile, the expression of PPARα and CPT-Ⅰ mRNA in myocardium in atorvastatin group were increased (PPARα mRNA: 0.457±0.075 vs control:0.285±0.062, P0.01; CPT-Ⅰ mRNA: 1.037±0.101 vs control: 0.795±0.139, P0.05) in associated with lower VWI and TDM(2.98±0.05 vs control:3.16±0.08 g/kg, P0.01; 18.91±1.05 vs control:21.32±1.25 μm, P0.05). Conclusion Atorvastatin may ameliorate cardiac hypertrophy by improves cardiac lipid metabolism in the SHR.
Rosuvastatin-attenuated heart failure in aged spontaneously hypertensive rats via PKCα/β2 signal pathway.

Abstract:There are controversies concerning the capacity of Rosuvastatin to attenuate heart failure in end-stage hypertension. The aim of the study was to show whether the Rosuvastatin might be effective or not for the heart failure treatment. Twenty-one spontaneously hypertensive rats (SHRs) aged 52 weeks with heart failure were randomly divided into three groups: two receiving Rosuvastatin at 20 and 40 mg/kg/day, respectively, and the third, placebo for comparison with seven Wistar-Kyoto rats (WKYs) as controls. After an 8-week treatment, the systolic blood pressure (SBP) and echocardiographic features were evaluated; mRNA level of B-type natriuretic peptide (BNP) and plasma NT-proBNP concentration were measured; the heart tissues were observed under electron microscope (EM); myocardial sarcoplasmic reticulum Ca2+ pump (SERCA-2) activity and mitochondria cytochrome C oxidase (CCO) activity were measured; the expressions of SERCA-2a, phospholamban (PLB), ryanodine receptor2 (RyR2), sodiumcalcium exchanger 1 (NCX1), Ca2+/calmodulin-dependent protein kinase II (CaMKII) and protein phosphatase inhibitor-1 (PPI-1) were detected by Western blot and RT-qPCR; and the total and phosphorylation of protein kinase Ca/beta (PKCa/beta) were measured. Aged SHRs with heart failure was characterized by significantly decreased left ventricular ejection fraction and left ventricular fraction shortening, enhanced left ventricular end-diastolic diameter and LV Volume, accompanied by increased plasma NT-proBNP and elevated BNP gene expression. Damaged myofibrils, vacuolated mitochondria and swollen sarcoplasmic reticulum were observed by EM. Myocardium mitochondria CCO and SERCA-2 activity decreased. The expressions of PLB and NCX1 increased significantly with up-regulation of PPI-1 and down-regulation of CaMKII, whereas that of RyR2 decreased. Rosuvastatin was found to ameliorate the heart failure in aged SHRs and to improve changes in SERCA-2a, PLB, RyR2, NCX1, CaMKII and PPI-1; PKCa/beta 2 signal pathway to be suppressed; the protective effect of Rosuvastatin to be dose dependent. In conclusion, the heart failure of aged SHRs that was developed during the end stage of hypertension could be ameliorated by Rosuvastatin.

Abstract:Background Farnesyltransferase (FT), an essential enzyme at the downstream of mevalonate pathway, was reported to be upregulated in hypertrophic cardiomyocytes of spontaneously hypertensive rats (SHRs) compared with myocardium of Wistar-Kyoto rats (WKYs). This upregulation was accompanied with cardiac remodeling. This study was designed to determine whether FT inhibition can alter cardiac remodeling in SHRs. Methods Twelve-week-old SHRs were randomized to receive infusion of either NS or FTI-276 (307μg/kg/d i.v. each n=10). WKY rats served as normal controls (n=6). Echocardiography was performed before and after intervention. SHR hearts were perfused ex vivo for the evaluation of cardiac performance, collagen deposition and biochemical changes (activation of Ras, extracellular-signal regulated kinases/ERK1/2, procollagen type І/Ш, TGF-β1, connective tissue growth factor/CTGF, and bone morphogenetic protein-7/BMP-7 expression). Results FTI-276 intervention decreased interventricular septum wall thickness at end- diastole (IVSd) and relative wall thickness (RWT) of SHRs (P<0.05). Three week intervention with FTI-276 attenuated hydroxyproline content (P<0.05), collagen deposition (P<0.01), Ras activation, ERK1/2 phosphorylation (P<0.01) and mRNA expression of procollagen type I, TGF-β1 and CTGF and elevated mRNA expression of BMP-7 (P<0.05) in left ventricle of SHRs. Conclusion The present study indicated that FT inhibition could attenuate myocardial fibrosis and partly improve cardiac remodeling in SHRs. The beneficial effects might be mediated through suppression of the activation of Ras and ERK1/2 phosphorylation pathway. The enhanced mRNA expression of BMP-7 with inhibition of TGF-β1 and CTGF mRNA expression might be an important mechanism.
Effects of atorvastatin on expression of cytochrome P450 epoxygenase 2C11 in spontaneously hypertensive rats

Abstract:AIM To investigate if the effect of atorvastatin on blood pressure is related to the expression of cytochrome P450 epoxygenase 2C11 (CYP2C11) in spontaneously hypertensive rats(SHR).METHODS SHR(n=18) were randomly divided into SHR model group,atorvastatin(10 and 50 mg·kg~(-1),ig,once daily for 10 weeks) treated groups.Six male Wistar-Kyoto(WKY) rats were selected as normal control group.Systolic blood pressure(SBP) was measured before and every 2 weeks after treatment.The expression of CYP2C11 mRNA and protein in heart,liver,kidney,and aorta were detected by RT-PCR and Western blot analysis,respectively.Urinary 14,15-dihydroxyeicosatrienoic acid(DHET) excretion was quantified by ELISA method.Plasma lipids were also measured.RESULTS SBP in all SHR groups was much higher than that in WKY group before experiment.Compared with SHR model group,SBP significantly decreased after treatment with 50 mg·kg~(-1) atorvastatin for 6,8 and 10 weeks and after treatment with 10 mg·kg~(-1) for 10 weeks.After 10-week treatment,compared with SHR model group,the levels of CYP2C11 mRNA and protein in heart,liver,kidney and aorta of 50 mg·kg~(-1) atorvastatin group and in heart,kidney and aorta of 10 mg·kg~(-1) atorvastatin group were markedly higher,but those in heart,kidney and aorta of WKY group were significantly lower.Urinary DHET excretion in all SHR groups was much higher than that in WKY group before experiment,(after) 10-week treatment,that in SHR model group was lower than that in 50 mg·kg~(-1) atorvastatin group.Plasma lipids in 50 and 10 mg·kg~(-1) atorvastatin groups were markedly lower than those in SHR model group.CONCLUSION Atorvastatin can significantly up-regulate the expression of CYP2C11 mRNA and protein and increase the production of epoxyeicosatrienoic acids in SHR,which may be one of the mechanisms of statins regulating hypertension.
Alteration of RhoA Prenylation Ameliorates Cardiac and Vascular Remodeling in Spontaneously Hypertensive Rats.

Abstract:BACKGROUND:In our previous study, farnesyl pyrophosphate synthase (FPPS) was shown to be increased in spontaneously hypertensive rats (SHR) and in mice with angiotensin-II induced cardiac hypertrophy. Overexpression of FPPS induced cardiac hypertrophy and fibrosis in mice, accompanied by an increase in the synthesis of farnesyl pyrophosphate (FPP) and geranylgeranyl pyrophosphate (GGPP). In the present study, we investigated the mechanisms of reversing cardiovascular remodeling in SHR by inhibiting FPPS.METHODS AND RESULTS:Six-week-old rats were given vehicle or an FPPS inhibitor (alendronate, 100 ug/kg/d) daily for twelve weeks by osmotic mini-pump. The results demonstrated that FPPS inhibition attenuated cardiac hypertrophy and fibrosis in SHR as shown by the heart weight to body weight ratio, echocardiographic parameters, and histological examination. In addition, FPPS inhibition attenuated aortic remodeling as shown by reduced media thickness, media cross-sectional area and collagen of the aorta as well as SBP, DBP, MBP. Furthermore, 12 weeks of alendronate treatment significantly decreased FPP and GGPP levels, RhoA activation and geranylgeranylation in the heart and aorta, all of which were significantly upregulated in SHR compared with normotensive Wistar-Kyoto rats.CONCLUSION:Taken together, these results indicate that chronic treatment with alendronate decreases the development of cardiac and aortic remodeling, by a pathway which involves inhibition of the geranylgeranylation and activation of RhoA.

Abstract:Previous clinical and experimental studies have demonstrated that statins, the inhibitors of 3-hydroxy-3-methylglutaryl-coenzyme A reductase, can improve left ventricular function in damaged hearts. Also, the normal expression of Ca 2+ regulatory proteins is critical for efficient myocardial function. However, it is still unclear whether the beneficial effect of statins on cardiac function is associated with alterations of Ca 2+ regulatory proteins. In this study, we investigated the effect of atorvastatin on cardiac function in spontaneously hypertensive rats (SHRs), focusing in particular on its impact on the expression of sarcoplasmic reticulum Ca 2+ -adenosine triphosphatase (SERCA2a), phospholamban (PLB) and its phosphorylated form (phosphorylated PLB), all of which are Ca 2+ regulatory proteins in myocardium. SHRs showed decreases in gene expression of SERCA2a and phosphorylated PLB, and reduction in SERCA activity in the left ventricular myocardium, as well as reduced cardiac function, compared to age-matched Wistar Kyoto rats (WKYs). Furthermore, we showed that in SHRs atorvastatin preserved cardiac dysfunction accompanied by positive alterations in calcium regulatory proteins, with up-regulation in expression of SERCA2a and phosphorylated PLB, and with improvement of SERCA activity. Thus, atorvastatin has positive effects on calcium regulatory proteins, which may be one of the mechanisms of the beneficial effect of statins on cardiac function in spontaneously hypertensive rats.
Prevention of myocardial fibrosis by using nitrendipine to the spontaneously hypertensive rats

Abstract:Aim: To determine the effects of nitrendipine on blood pressure, left ventricular hypertrophy (LVH) and myocardial fibrosis of both ventricles of spontaneously hypertensive rats (SHR). Method: Antihypertensive treatment with calcium channel blocker nitrendipine (30 mg �� kg-1/d) was given to 14-week old male SHRs (n = 8) for 12 weeks with age matched SHRs and Wistar-Kyoto rats (WKY) (n = 8) received no treatment as controls. Systolic blood pressure was measured by tail-cuff method. LVH was defined according to the ratio of left to right ventricular weights or to body weights. Myocardial fibrosis was evaluated by collagen content and concentration mg/mg dry wt. in both ventricles. Hydroxyprolin content was measured spectrophotometrically by its reaction with Ehrich's reagent (Stegemann method). And collagen content was calculate by multiplying the hydroxyprolin content with the constant 8.2. Results: Nitrendipine significantly decreased systolic blood pressure in SHRs (P < 0.05) although the value remained significantly higher than that of WKY controls. The ratio of left ventricular weight to body weight was significantly decreased in nitrendipine treated compared with untreated SHRs (P < 0.05). Right ventricular weight of the experimental group showed no significant change versus that of the controls. Collagen content and concentration of the left ventricle and right ventricle were lower in the nitrendipine treated group compared with the untreated groups. Conclusions: Nitrendipine could attenuate hypertension and decrease collagen content and concentration of both ventricles as well as left ventricular mass in SHRs. So nitrendipine might have the potential to improve the myocardial pathology in essential hypertension.
Effects and Mechanisms of Losartan on the Structure of Resistant Vessel in Spontaneously Hypertensive Rat

Abstract:Objective To evaluate the effects of Losartan on systolic blood pressure,the structure of resistant vessel and the changes of plasma angiotensinⅡ in spontaneously hypertensive rats (SHR). Methods six weeks male SHR and WKY were divided into three groups at random: SHR losartan group、SHR control group、WKY normal control group. Losartan 30 mg.kg-1.d-1 was adm inistered by gavage to rats in SHR Losartan group. After 17 weeks treatment, Wall-to-lumen ar ea ratios (W/L)of mesenteric arteries (3 rd grade branch ) were calculated by computer image analysis system. Systolic blood pressure(SBP) were measured. The level of vessel hypertrophy was assessed by means of transmission electron microscope. The plasma renin activity and AngⅡlevel was determined by RIA method. Results (1) After 17 weeks treatment, the blood pressure of SHR losartan group was decreased compared with SHR control group (P<0.05). (2) Th e W/L in SHR losartan group was significantly decreased compared with SHR control group (P<0.01). There was no remarkable difference in W/L between SHR losa rtan and WKY group(P>0.05).While the W/L in SHR losar tan group is still remain in a higher level than that of WKY rats(P<0.05). The ultrastructures of mesenteric arteries in SHR losartan group were similar with those of WKY normal group; however, ultrast ructure change in mesenteric arteries in SHR control group were pronouncedly. (3) The plasma renin activities had no significant differences (P>0.05) be tween WKY normal group and SHR control group. The plasma renin activities in SHR losartan group increased compared with SHR control group (P<0.05). AngⅡ level in SHR losartan group significantly increased compared with SHR control group (P<0.01). The AngⅡ level in SHR losartan group was hig her than that in WKY normal group (P<0.05). Conclusion These results indicated that long-term treatment with Losartan effectively reduced blood pressure、attenuated and regressed the resist ant vessel hypertrophy during the development of hypertension in SHR.
The Therapeutic Effect of Rosuvastatin on Cardiac Remodelling from Hypertrophy to Fibrosis During the End-Stage Hypertension in Rats

Abstract:End-stage hypertensive heart disease is an increasing cause of cardiac mortality. Therefore, the current study focused on the cardiac remodelling from hypertrophy to fibrosis in old-aged spontaneously hypertensive rats (SHRs), and explored the therapeutic effects of Rosuvastatin and its possible mechanism(s) of action. Spontaneously hypertensive rats at age 52 weeks were randomly divided into three groups, the first two to receive Rosuvastatin at a dose of 20 mg/kg/day and 40 mg/kg/day, respectively, and the third to receive placebo, which was to be compared with Wistar-Kyoto as controls. After 2-month treatment, SBP, heart to body weight ratio (HW/BW%) and echocardiographic features were evaluated, followed by haematoxylin and eosin and Masson trichrome staining in conjunction with qPCR of foetal gene expressions. Transferase-mediated dUTP nick-end labelling assay and immunofluorescent labelling for active caspase-3 were used to detect the apoptotic cardiomyocytes. Signaling pathways involved were examined by using western blot. Old-aged SHR developed end-stage hypertensive heart disease characterized by significant enhancement of HW/BW%, LVAWd and LVPWd, and decreased LVEF and LVFS, accompanied by cardiomyocytes enlargement and fibrosis along with activation of foetal gene programme. Cardiac apoptosis increased significantly during the transition process. Rosuvastatin reduced hypertrophy significantly via AT(1) Receptor-PKCβ2/α-ERK-c-fos pathway; protected myocardium against apoptosis via Akt-FOXO1, Bcl-2 family and survivin pathways and consequently suppressed the caspase-3 activity. The present study revealed that old-aged SHRs developed cardiac remodelling from hypertrophy to fibrosis via cardiac apoptosis during the end stage of hypertensive heart disease. These pathological changes might be the consequence of activation of AT(1) Receptor-PKCβ2/α-ERK-c-fos and AKT-FOXO1/Bcl-2/survivin/Caspase3 signaling. Rosuvastatin effectively attenuated the structural changes by reversing the signaling transductions involved.

Abstract:There are in vitro data linking all-trans retinoic acid (atRA) with inhibition of hypertrophy and hyperplasia in cardiomyocytes, vascular smooth muscle cells, and fibroblasts. In the present study, we tested the hypothesis that chronic treatment with atRA may blunt the process of myocardial remodeling in spontanously hypertensive rats (SHR). Four-week-old male SHR were treated with atRA (5 or 10 mg.kg(-1).day(-1)) given daily for 3 mo by gavage; age- and sex-matched Wistar-Kyoto rats (WKY) and placebo-treated SHR served as controls. At the end of the treatment period, cardiac geometry and function were assessed by Doppler echocardiography. Histological examination and RIA were performed to evaluate medial thickening of intramyocardial and renal arteries, perivascular and interstitial collagen content, and atrial natriuretic peptide (ANP) and IGF-I in the heart, respectively. The novel finding of the present study is that atRA prevented hypertrophy of intramyocardial and intrarenal arteries and ventricular fibrosis. However, atRA treatment did not lower blood pressure or left ventricular weight and left ventricular weight-to-body weight ratio in SHR. atRA did not change cardiac geometry and function as assessed by Doppler echocardiography. atRA showed no influence on either ANP or IGF-I levels. In conclusion, the present study suggests that chronic atRA treatment prevents medial thickening of intramyocardial and intrarenal arteries and ventricular fibrosis during the development of hypertension. Left ventricular hypertrophy and cardiac geometry and function are not changed by atRA treatment.
