Abstract:Plant sap-feeding insects of Hemiptera often form intimate symbioses with microbes to obtain nutrients. The cicada Subpsaltria yangi is the only species of the subfamily Tettigadinae known from China. Using high-throughput sequencing combined with fluorescence in situ hybridization analysis, we characterize the bacterial composition of the bacteriomes, testes, ovaries and eggs of two representative populations of this species which occur in different habitats and feed on different plant hosts. In both populations, the bacterial community diversity in the testes was significantly higher than that in other tissues. The obligate endosymbiont Candidatus Sulcia muelleri was observed in all samples and was dominant in the bacteriomes, ovaries and eggs. The usual co-resident endosymbiont Candidatus Hodgkinia cicadicola found in some other cicadas was not detected. Instead, a novel Rhizobiales bacterium which shows a ~ 81% 16S rDNA similarity to Ca. Hodgkinia cicadicola was detected. Given that the genome of Ca. Hodgkinia cicadicola exhibits rapid evolution, it is possible that this novel Rhizobiales bacterium is a related endosymbiont with beneficial trophic functions similar to that of Ca. Hodgkinia cicadicola hosted by several certain other cicadas. The presence of the novel Rhizobiales species in other cicadas and its involvement with the adaptive evolution of related cicada hosts require further investigation. Discrepancy of bacterial communities associated with testes between the two populations may be closely related to the geographic isolation and divergence of habitats and host plants. Our results are informative for further studies of evolutionary divergence of related endosymbionts hosted in cicadas.

Abstract:Cicadas form intimate symbioses with bacteria to obtain nutrients that are scarce in the xylem fluid they feed on. The obligate symbionts in cicadas are purportedly confined to specialized bacteriomes, but knowledge of bacterial communities associated with cicadas is limited. Bacterial communities in the bacteriomes and organs of reproductive, digestive and excretory systems of two cicada species (Platypleura kaempferi and Meimuna mongolica) were investigated using different methods, and the bacterial diversity and distribution patterns of dominant bacteria in different tissues were compared. Within each species, the bacterial communities of testes are significantly different from those of bacteriomes and ovaries. The dominant endosymbiont Candidatus Sulcia muelleri is found not only in the bacteriomes and reproductive organs, but also in the "filter chamber + conical segment" of both species. The transmission mode of this endosymbiont in the alimentary canal and its effect on physiological processes merits further study. A novel bacterium of Rhizobiales, showing similar to 80% similarity to Candidatus Hodgkinia cicadicola, is dominant in the bacteriomes and ovaries of P. kaempferi. Given that the genome of H. cicadicola exhibits rapid sequence evolution, it is possible that this novel bacterium is a related endosymbiont with beneficial trophic functions similar to that of H. cicadicola in some other cicadas. Failure to detect H. cicadicola in M. mongolica suggests that it has been subsequently replaced by another bacterium, a yeast or gut microbiota which compensates for the loss of H. cicadicola. The distribution of this novel Rhizobiales species in other cicadas and its identification require further investigation to help establish the definition of the bacterial genus Candidatus Hodgkinia and to provide more information on sequence divergence of related endosymbionts of cicadas. Our results highlight the complex bacterial communities of cicadas, and are informative for further studies of the interactions and co- evolution of insect- microbial symbioses in Cicadoidea.

Abstract:Bacteriocyte-associated symbionts are essential for the health of many sap-sucking insects, such as cicadas, leafhoppers and treehoppers, etc., but little is known about the bacterial community in the gut and other related organs in these insects. We characterized the bacterial communities in the salivary glands, alimentary canal and the Malpighian tubules of two populations of the cicada Subpsaltria yangi occurring in different habitats and feeding on different hosts. A high degree of similarity of core microbiota was revealed between the two populations, both with the top three bacteria belonging to Meiothermus, Candidatus Sulcia and Halomonas. The bacterial communities in various organs clustered moderately by populations possibly reflect adaptive changes in the microbiota of related S. yangi populations, which provide a better understanding of the speciation and adaptive mechanism of this species to different diets and habitats. When compared with two phylogenetically distant cicada species, Hyalessa maculaticollis and Meimuna mongolica, the core microbiota in S. yangi was significantly different to that of these species. In addition, our results confirm that Ca. Sulcia distributes in the digestive and excretory organs besides the bacteriomes and gonads, which provide potential important information onto the trophic functions of this obligate endosymbiont to the host insects.

Abstract:Mutualistic associations between symbiotic bacteria and their insect hosts are widespread. The bacterial diversity and community composition within hosts may play an important role in shaping insect biology, ecology, and evolution. Here, we focused on the bacterial communities in bacteriomes, ovaries and testes of three representative populations of the cicada Platypleura kaempferi (Fabricius) using high-throughput 16S rRNA amplicon sequencing approach combined with light microscopy and confocal imaging approach. The obligate symbiont Sulcia was detected in all the examined samples, which showed a relatively high abundance in most bacteriomes and ovaries. The unclassified OTUs formerly identified as an unclassified Rhizobiales bacterium was demonstrated to be the co-obligate symbiont Hodgkinia, which showed 100% infection rate in all the examined samples and had an especially high abundance in most bacteriomes and ovaries. Hodgkinia and Sulcia occupy the central and peripheral bacteriocytes of each bacteriome unit, respectively. Cluster analysis revealed that the bacterial communities in bacteriomes, ovaries and testes of Zhouzhi and Ningshan populations separated strongly from each other. Significant difference was also detected between the Yangling and Ningshan populations, but no significant difference was detected between the Yangling and Zhouzhi populations. This may be related to the difference of host plants and genetic differentiation of these populations. Our findings show that the bacterial communities can be influenced by the population differentiation of the host cicadas and/or the host plants of cicadas, which improve our understanding of the associations between the bacterial community and population differentiation of sap-feeding insects.

Abstract:Insect feeding structures, such as mandibles, interact with the ingesta (food or/and substrate) and can be adapted in morphology, composition of material and mechanical properties. The foraging on abrasive ingesta, as on algae covering rocks, is particularly challenging because the mandibles will be prone to wear and structural failure, thus suggesting the presence of mandibular adaptations to accompany this feeding behavior. Adaptations to this are well studied in the mouthparts of molluscs and sea urchins, but for insects there are large gaps in our knowledge. In this study, we investigated the mandibles of a grazing insect, the larvae of the trichopteran Glossosoma boltoni . Using scanning electron microscopy, wear was documented on the mandibles. The highest degree was identified on the medial surface of the sharp mandible tip. Using nanoindentation, the mechanical properties, such as hardness and Young's modulus, of the medial and lateral mandible cuticles were tested. We found, that the medial cuticle of the tip was significantly softer and more flexible than the lateral one. These findings indicate that a self-sharpening mechanism is present in the mandibles of this species, since the softer medial cuticle is probably abraded faster than the harder lateral one, leading to sharp mandible tips. To investigate the origins of these properties, we visualized the degree of tanning by confocal laser scanning microscopy. The autofluorescence signal related to the mechanical property gradients. The presence of transition and alkaline earth metals by energy dispersive X-ray spectroscopy was also tested. We found Ca, Cl, Cu, Fe, K, Mg, Mn, P, S, Si, and Zn in the cuticle, but the content was very low and did not correlate with the mechanical property values.

Abstract:Although transovarial transmission of bacteriome-associated symbionts in hemipteran insects is extremely important for maintaining intimate host-symbiont associations, our knowledge of cellular mechanisms underlying the transmission process is quite limited. We investigated bacterial communities of salivary glands, bacteriomes, and digestive and reproductive organs and clarified the transovarial transmission of bacteriome-associated symbionts of the mountain-habitat specialist Pycna repanda using integrated methods. The bacterial communities among different gut tissues and those of bacteriomes of males and females both show similarity, whereas differences are exhibited among bacterial communities in testes and ovaries. The primary symbionts "Candidatus Sulcia muelleri" (hereafter "Ca. Sulcia") and "Candidatus Hodgkinia cicadicola" (hereafter "Ca. Hodgkinia") were not only restricted to but also dominant in the bacteriomes and ovaries. "Ca. Hodgkinia" cells in the bacteriomes of both sexes exhibited different colors by histological and electron microscopy. Also considering the results of a restriction fragment length polymorphism (RFLP)-based cloning approach, we hypothesize that "Ca. Hodgkinia" may have split into cytologically different cellular lineages within this cicada species. Regarding the dominant secondary symbionts, Rickettsia was detected in the salivary glands, digestive organs, and testes, whereas Arsenophonus was detected in the bacteriomes and ovaries. Our results show that Arsenophonus can coexist with "Ca. Sulcia" and "Ca. Hodgkinia" within bacteriomes and can be transovarially transmitted with these obligate symbionts together from mother to offspring in cicadas, but it is not harbored in the cytoplasm of "Ca. Sulcia." The change in the shape of "Ca. Sulcia" and "Ca. Hodgkinia" during the transovarial transmission process is hypothesized to be related to the limited space and novel microenvironment. IMPORTANCE Cicadas establish an intimate symbiosis with microorganisms to obtain essential nutrients that are extremely deficient in host plant sap. Previous studies on bacterial communities of cicadas mainly focused on a few widely distributed species, but knowledge about mountain-habitat species is quite poor. We initially revealed the physical distribution of the primary symbionts "Ca. Sulcia" and "Ca. Hodgkinia" and the dominant secondary symbionts Rickettsia and Arsenophonus in the mountain-habitat specialist Pycna repanda and then clarified the transovarial transmission process of bacteriome-associated symbionts in this species. Our observations suggest that "Ca. Hodgkinia" may have split into cytologically distinct lineages within this cicada species, and related cicadas might have developed complex mechanisms for the vertical transmission of the bacteriome-associated symbionts. We also revealed that Arsenophonus can be transovarially transmitted in auchenor-rhynchan insects when it is not harbored in the cytoplasm of other endosymbionts. Our results highlight transovarial transmission mechanisms of bacteriome-associated symbionts in sap-feeding insects.
Structural diversity of symbionts and related cellular mechanisms underlying vertical symbiont transmission in cicadas

Abstract:Many insects depend on symbiont(s) for survival. This is particularly the case for sap-feeding hemipteran insects. In this study, we revealed that symbionts harboured in cicadas are diverse and complex, and the yeast-like fungal symbionts (YLS) are present in most cicada species, but Hodgkinia is absent. During vertical transmission, Sulcia became swollen with the outer membrane drastically changed, while Hodgkinia became shrunken and changed from irregular to roughly spherical. Sulcia and/or Hodgkinia were exocytosed from the bacteriocytes to the intercellular space of bacteriomes, where they gathered together and were extruded to haemolymph. YLS and associated facultative symbiont(s) in the fat bodies were released to the haemolymph based on bacteriocyte disintegration. The obligate symbiont(s) were endocytosed and exocytosed successively by the epithelial plug cells of the terminal oocyte, while associated facultative symbiont(s), and possibly also YLS, may take a 'free ride' on the transmission of obligate symbiont(s) to gain entry into the oocyte. Then, the intermixed symbionts formed a characteristic 'symbiont ball' in the oocyte. Our results suggest that YLS in cicadas represent a new example of a relatively early stage of symbiogenesis in insects and contribute to a better understanding of the diversity and transmission mechanisms of symbionts in insects.
Mandible elemental composition and mechanical properties from distinct castes of the leafcutter ant Atta laevigata (Attini; Formicidae)

Abstract:Leafcutter ant colonies are divided into castes with the individuals performing different tasks, based mostly on size. With the mandibles, the small minims care for the brood or the fungus, whereas the larger minors and mediae cut and transport plant material, with the ant size positively related to the material size. The mechanical properties and composition of the mandible cuticle have been previously tested in the soldiers as the largest caste, revealing that the cutting edges contained high contents of the cross-linking transition metal zinc (Zn). With regard to the smaller castes, no data are present. To study how the mandible size and function relates to its mechanical properties, we here tested the mandibles of minims, minors and mediae by nanoindentation. We found that the hardness (H) and Young's modulus (E) values increased with increasing ant size and that the mandible cutting edges in each caste have the highest H- and E-values. To gain insight into the origins of these properties, we characterized the elemental composition by energy-dispersive X-ray analysis, revealing that minors and mediae possessed higher content of Zn in the cutting edges in contrast to the minims containing significantly less Zn. This shows, that Zn content relates to higher mechanical property values. Additionally, it shows that all of these parameters can differ within a single species.

Abstract:Centipedes are terrestrial and predatory arthropods that possess an evolutionary transformed pair of appendages used for venom injection—the forcipules. Many arthropods incorporate reinforcing elements into the cuticle of their piercing or biting structures to enhance hardness, elasticity or resistance to wear and structural failure. Given their frequent exposure to high mechanical stress, we hypothesise that the cuticle of the centipede forcipule might be mechanically reinforced. With a combination of imaging, analytical techniques and mechanical testing, we explore the centipede forcipule in detail to shed light on its morphology and performance. Additionally, we compare these data to characteristics of the locomotory leg to infer evolutionary processes.

Abstract:Insects process their food with their cuticle‐based mouthparts. These feeding structures reflect their diversity and can, in some cases, showcase adaptations in material composition, mechanical properties, and shape to suit their specific dietary preferences. To pave the way to deeply understand the interaction between mouthparts and food and to determine potential adaptations of the structures to the food, this study focuses on the mandibles of two praying mantis species. Gongylus gongylodes feeds mainly on Diptera, and Sphodromantis lineola forages on larger prey. Employing scanning electron microscopy, the mandibular morphologies were analyzed. The degree of the cuticle tanning was tested using confocal laser scanning microscopy. Furthermore, the contents of transition and alkaline earth metals in the mandible cuticle were studied using energy‐dispersive X‐ray spectroscopy and the mechanical properties tested by nanoindentation. We found that S. lineola mandibles show pronounced gradients of Young's modulus and hardness from the basis to the tip, which might be an adaptation against high stresses during biting and chewing. G. gongylodes, in contrast, did not show pronounced gradients, which may indicate that there is less stress involved in feeding—necessary to test in future studies. The mechanical properties of manidibles in both species are related to the degree of cuticle tanning but also positively correlate with the content of magnesium. These findings enrich our understanding of insect cuticle biology but also present new sets of data on praying mantis structures.

A multilayer micromechanical model of the cuticle of Curculio longinasus Chittenden, 1927 (Coleoptera: Curculionidae)

Abstract:Curculio longinasus Chittenden, 1927 (Coleoptera: Curculionidae), is a weevil species common throughout the southwestern United States that uses its rostrum - a very slender, curved, beak-like projection of the head - to excavate tunnels in plant organs (such as acorns) for egg laying (oviposition). Once the apical portion of the rostrum has been inserted into the preferred substrate for oviposition, the female begins rotating around the perimeter of the hole, elevating her head by extending the fore-legs, and rotating the head in place in a drilling motion. This action causes significant elastic deformation of the rostrum, which will bend until it becomes completely straight. To better understand the mechanical behavior of the cuticle as it undergoes deformation during the preparation of oviposition sites, we develop a comprehensive micro/macro model of the micromechanical structure and properties of the cuticle, spanning across all cuticular regions, and reliably mirroring the resultant macroscale properties of the cuticle. Our modeling approach relies on the use of multi-scale, hierarchical biomaterial representation, and employs various micromechanical schemata - e.g., Mori-Tanaka, effective field, and Maxwell - to calculate the homogenized properties of representative volume elements at each level in the hierarchy. We describe the configuration and behavior of this model in detail, and discuss the theoretical implications and limitations of this approach with emphasis on future biomechanical and comparative evolutionary research. Our detailed account of this approach can thereby serve as a methodological template for exploring the biomechanical behavior of new insect structures.
Breaking up the wall: metal-enrichment in Ovipositors, but not in mandibles, co-varies with substrate hardness in gall-wasps and their associates

Abstract:The cuticle of certain insect body parts can be hardened by the addition of metals, and because niche separation may require morphological adaptations, inclusion of such metals may be linked to life history traits. Here, we analysed the distribution and enrichment of metals in the mandibles and ovipositors of a large family of gall-inducing wasps (Cynipidae, or Gall-Wasps) (plus one gall-inducing Chalcidoidea), and their associated wasps (gall-parasitoids and gall-inquilines) (Cynipidae, Chalcidoidea and Ichneumonoidea). Both plant types/organs where galls are induced, as well as galls themselves, vary considerably in hardness, thus making this group of wasps an ideal model to test if substrate hardness can predict metal enrichment. Non-galler, parasitic Cynipoidea attacking unconcealed hosts were used as ecological "outgroup". With varying occurrence and concentration, Zn, Mn and Cu were detected in mandibles and ovipositors of the studied species. Zn tends be exclusively concentrated at the distal parts of the organs, while Mn and Cu showed a linear increase from the proximal to the distal parts of the organs. In general, we found that most of species having metal-enriched ovipositors (independently of metal type and concentration) were gall-invaders. Among gall-inducers, metals in the ovipositors were more likely to be found in species inducing galls in woody plants. Overall, a clear positive effect of substrate hardness on metal concentration was detected for all the three metals. Phylogenetic relationships among species, as suggested by the most recent estimates, seemed to have a weak role in explaining metal variation. On the other hand, no relationships were found between substrate hardness or gall-association type and concentration of metals in mandibles. We suggest that ecological pressures related to oviposition were sufficiently strong to drive changes in ovipositor elemental structure in these gall-associated Hymenoptera.
Structure, mechanics and material properties of claw cuticle from mole cricket Gryllotalpaorientalis

Abstract:Powerful shovel-like forelimbs with special shape, structure and biological materials enable mole cricket to digging efficiently. During digging, the tip of the claw needs to wedge into the soil, and the base needs to withstand considerable anti-shear force. In this study, we analysed the structural characteristics, material composition and mechanical properties of the claw teeth using scanning electron microscopy, plasma atomic emission spectroscopy, nanoindentation and finite element analysis. The results show that the tips of claw teeth have a dense and homogeneous structure and a higher hardness and contents of Mn and Zn compared with the base. The structure of the base of claw teeth has an obvious laminar structure and higher fracture resistance. Moreover, it is speculated from the simulation results that basal position of the claw teeth is tough enough to withstand high stress, and the presence of the ribs effectively improves the mechanical stability and load-bearing capacity of the teeth during excavation. The results of this study can provide inspiration for the design of efficient mechanical components and agricultural implements.

Adult diet strongly affects cuticle thickness and its injury resistance in the insect (Hemiptera: Coreidae)

Abstract:Arthropods are the most diverse phylum on earth, accounting for up to 90% of animal species. The cuticular exoskeleton has played a vital role in their evolutionary success, but we know surprisingly little about the factors influencing its development, structure, and biomechanical properties. In this study, we examined whether and how nutrition affects the cuticle after an insect has completed its final molt into the adult stage. We found that a high-quality adult diet provided to the leaf-footed cactus bug, (Hemiptera: Coreidae), over three weeks led to 4.1-times thicker cuticle with 3.7-times greater injury resistance relative to those that consumed a poor, but still ecologically relevant, diet. We further discovered that a high-quality adult diet allowed compensation for a sub-optimal juvenile diet. The cuticle of males was more injury resistant than that of females, as expected due to the aggressive male competition in this species. Altogether, our results show that, even after the final molt, nutrition can strongly influence the development and properties of the arthropod cuticle, a phenomenon that likely has drastic fitness-related consequences for locomotion, predator-prey interactions, and resource acquisition.

The Microscopic Morphology of Mouthparts and Their Sensilla in the Mycophagous Ladybeetle Illeis chinensis (Coleoptera: Coccinellidae)

Abstract:The morphological diversity of insect mouthparts is closely related to changes in food sources and diets. Research into the structures of insect mouthparts may help to establish a fundamental basis for a better understanding of insect feeding mechanisms. In this study, we examined the fine morphology of the mouthparts of Illeis chinensis using scanning electron microscopy. We paid particular attention to the types, quantities, and distribution of sensilla on the mouthparts. Our results showed that the basic components of the mouthparts of I. chinensis are the same as those in other lady beetles, i.e., the labrum, mandible, maxillae, labium, and hypopharynx. We also found structural specialization indicating adaptation to fungal feeding. On the mouthparts, there are eight kinds of sensilla and two kinds of glandular structures, including sensilla chaetica, sensilla basiconica, sensilla styloconica, sensilla coeloconica, sensilla campaniformia, sensilla placodea, sensilla digitiformia, Böhm bristles, perforated plates, and cuticular pores. This is the first time that sensilla digitiformia has been reported in ladybirds. Finally, variations in mouthparts among ladybirds with differing diets, as well as the putative functions of each of the mouthparts and sensilla, were discussed. This research can provide a reference for understanding the functions of the mouthparts in ladybird feeding behavior and thereby contribute to the development of precise insect behavior regulation and management strategies.

Abstract:This investigation explored chemical changes in cicadas during their developmental stages (nymph, late nymph, and adult). Tocopherols (α, δ, γ) were found at a total content of 13.7 mg/g, while γ-oryzanol was observed at 2.6 mg/g, with nymphs having the highest levels, followed by late nymphs and adults. Essential amino acids increased progressively with maturation, with methionine being the predominant amino acid in all samples. The index of essential amino acids in each tissue was as follows: adult (0.36), late nymph (0.33), and nymph (0.12). Eicosapentaenoic acid concentrations varied from 230 mg/100 g in adults to 880 mg/100 g in nymphs. Protein analysis using the Protein Simple Jess system revealed a molecular weight distribution ranging from 10 to 75 kDa, accounting for approximately 70 % of the total protein content. These findings offer valuable insights for incorporating cicadas as functional food ingredients, diversifying food product formulations.

Abstract:Insect cuticle can reach a wide range of material properties, which is thought to be the result of adaptations to applied mechanical stresses. Biomechanical mechanisms behind these property variations remain largely unknown. To fill this gap, here we performed a comprehensive study by simultaneous investigation of the microstructure, sclerotization and the elasticity modulus of the specialized cuticle of the femora of desert locusts. We hypothesized that, considering their different roles in jumping, the femora of fore-, mid- and hind legs should be equipped with cuticles that have different mechanical properties. Surprisingly, our results showed that the hind femur, which typically bears higher stresses, has a lower elasticity modulus than the fore and mid femora in the longitudinal direction. This is likely due to the lower sclerotization and different microstructure of the hind femur cuticle. This allows for some deformability in the femur wall and is likely to reduce the risk of mechanical failure. In contrast to both other femora, the hind femur is also equipped with a set of sclerotized ridges that are likely to provide it with the required stiffness to withstand the mechanical loads during walking and jumping. This paper is one of only a few comprehensive studies on insect cuticle, which advances the current understanding of the relationship between the structure, material property and function in this complex biological composite. STATEMENT OF SIGNIFICANCE: Insect cuticle is a biological composite with strong anisotropy and wide ranges of material properties. Using an example of the femoral cuticle of desert locusts, we measured the elasticity modulus, microstructure and sclerotization level of the cuticle. Our results show that, although the hind femur withstands most of the stress during locomotion, it has a lower elasticity modulus than the fore and mid femora. This is likely to be a functional adaption to jumping, in order to allow small deformations of the femur wall and reduce the risk of material failure. Our results deepen the current understanding of the structure-material-function relationship in the complex insect cuticle.
Study of the influence of macro-structure and micro-structure on the mechanical properties of stag beetle upper jaw

Abstract:Macrostructural control of stress distribution and microstructural influence on crack propagation is one of the strategies for obtaining high mechanical properties in stag beetle upper jaws. The maximum bending fracture force of the stag beetle upper jaw is approximately 154, 000 times the weight of the upper jaw. Here, we explore the macro and micro-structural characteristics of two stag beetle upper jaws and reveal the resulting differences in mechanical properties and enhancement mechanisms. At the macroscopic level, the elliptic and triangular cross-sections of the upper jaw of the two species of stag beetles have significant effects on the formation of cracks. The crack generated by the upper jaws with a triangular section grows slowly and deflects easily. At the microscopic level, the upper jaw of the two species is a chitin cross-layered structure, but the difference between the two adjacent fiber layers at 45° and 50° leads to different deflection paths of the cracks on the exoskeleton. The mechanical properties of the upper jaw of the two species of stag beetle were significantly different due to the interaction of macro-structure and micro-structure. In addition, a series of bionic samples with different cross-section geometries and different fiber cross angles were designed, and mechanical tests were carried out according to the macro-structure and micro-structure characteristics of the stag beetle upper jaw. The effects of cross-section geometry and fiber cross angle on the mechanical properties of bionic samples are compared and analyzed. This study provides new ideas for designing and optimizing highly loaded components in engineering. STATEMENT OF SIGNIFICANCE: The upper jaw of the stag beetle is composed of a complex arrangement of chitin and protein fibers, providing both rigidity and flexibility. This structure is designed to withstand various mechanical stresses, including impacts and bending forces, encountered during its burrowing activities and interactions with its environment. The study of the upper jaw of the stag beetle can provide an efficient structural design for engineering components that are subjected to high loads. Understanding the relationship between structure and mechanical properties in the stag beetle upper jaw holds significant implications for biomimetic design and engineering.
Enhanced stress-tolerance of transgenic tobacco plants expressing a human dehydroascorbate reductase gene.

Abstract:To analyze the physiological role of dehydroascorbate reductase (DHAR, EC 1.8.5.1) catalyzing the reduction of DHA to ascorbate in environmental stress adaptation, T1 transgenic tobacco (Nicotiana tabacum cv. Xanthi) plants expressing a human DHAR gene in chloroplasts were biochemically characterized and tested for responses to various stresses. Fully expanded leaves of transgenic plants had about 2.29 times higher DHAR activity (units/g fresh wt) than non-transgenic (NT) plants. Interestingly, transgenic plants also showed a 1.43 times higher glutathione reductase activity than NT plants. As a result, the ratio of AsA/DHA was changed from 0.21 to 0.48, even though total ascorbate content was not significantly changed. When tobacco leaf discs were subjected to methyl viologen (MV) at 5 mumol/L and hydrogen peroxide (H2O2) at 200 mmol/L, transgenic plants showed about a 40% and 25% reduction in membrane damage relative to NT plants, respectively. Furthermore, transgenic seedlings showed enhanced tolerance to low temperature (15 degrees C) and NaCl (100 mmol/L) compared to NT plants. These results suggest that a human derived DHAR properly works for the protection against oxidative stress in plants.

Abstract:The gastric mill of Decapoda is a unique feature, which comprises teeth, stabilizing ossicles, and particle sorting setae. Involved in the fragmentation and sorting of the food, this structure serves as interface between the organism and its environment. As material properties complement morphology and hold information about function and trophic preferences, we here provide a basis for more comparative research on gastric mills. For gastric mill components of the adult red swamp crayfish Procambarus clarkii , we studied (a) the micro-structure via scanning electron microscopy, (b) the elemental composition by energy-dispersive X-ray spectroscopy, (c) the heterogeneities in material properties and degree of tanning (autofluorescence) by confocal laser scanning microscopy, and (d) the mechanical properties hardness and elasticity by nanoindentation technique. The morphology and micro-structure were previously described for this species, but the mechanical properties and the autofluorescence were not studied before. As epicuticle and exocuticle could be analyzed individually, material property gradients, with values decreasing from the interacting surface towards interior, could be determined. Finally, we were able to relate the mechanical property data with the elemental composition and the degree of tanning. We found that the epicuticle of the teeth is among the hardest and stiffest biological materials in invertebrates due to the incorporations of high proportions of silicon.
