Abstract:Copper-based nanoparticles (NPs) display a strong potential to replace copper salts (e.g., CuSO4) for application in agricultures as antimicrobial agents or nutritional amendments. Yet, their effects on crop quality are still not comprehensively understood. In this study, the Cu contents in soybeans grown in soils amended with Cu NPs and CuSO4 at 100-500 mg Cu/kg and the subsequent effects on the plant physiological markers were determined. The Cu NPs induced 29-89% at the flowering stage (on Day 40) and 100-165% at maturation stage (on Day 100) more Cu accumulation in soybeans than CuSO4. The presence of particle aggregates in the root cells with deformation upon the Cu NP exposure was observed by transmission electron microscopy. The Cu NPs at 100 and 200 mg/kg significantly improved the plant height and biomass, yet significantly inhibited at 500 mg/kg, compared to the control. In leaves chlorophyll-b was more sensitive than chlorophyll-a and carotenoids to the Cu NP effect. The Cu NPs significantly decreased the root nitrogen and phosphorus contents, while they significantly increased the leaf potassium content in comparison with control. Our results imply that cautious use of Cu NPs in agriculture is warranted due to relatively high uptake of Cu and altered nutrient quality in soybeans.

Abstract:With the increased use of metal nanoparticles (NPs), their access to the food chain has become a main concern to scientists and holds controversial social implications. This research particularly sheds light on copper nanoparticles (CuNP), as they have been commonly used in several industries nowadays. In this study, we investigated the phytotoxicity of CuNP on cucumber (Cucumis sativus) plants grown hydroponically. Atomic Absorption Spectroscopy (AAS), X-Ray Fluorescence (XRF), and Scanning Electron Microscopy (SEM) analysis confirmed that C. sativus treated with CuNP accumulated CuNP in the plant tissues, with higher levels in roots, with amounts that were concentration dependent. Furthermore, genotoxicity was assessed using Random amplified polymorphic DNA (RAPD) technique, and our results showed that CuNP caused genomic alterations in C. sativus. Phenotypical, physiological, and biochemical changes were assessed by determining the CuNP treated plant's total biomass, chlorophyll, H2O2 and MDA contents, and electrolyte leakage percentage. The results revealed notable adverse phenotypical changes along with decreased biomass and decreased levels of the photosynthetic pigments (Chlorophyll a and b) in a concentration-dependent manner. Moreover, CuNP induced damage to the root plasma membrane as determined by the increased electrolyte leakage. A significant increase in H2O2 and MDA contents were detected in C. sativus CuNP treated plants. Additionally, copper-zinc superoxide dismutase (Cu-Zn SOD) gene expression was induced under CuNP treatment. Overall, our results demonstrated that CuNP of 10-30 nm size were toxic to C. sativus plants. This finding will encourage the safe production and disposal NPs. Thus, reducing nano-metallic bioaccumulation into our food chain through crop plants; that possesses a threat to the ecological system.

Abstract:Copper oxide nanoparticles (CuO NPs) have received considerable attention for their toxic effects on crops and potential application in agriculture. In order to investigate the biological effects of CuO NPs on plants, we exposed cucumber (Cucumis sativus) to two sizes of CuO NPs (510 nm, μCuO and 43 nm, nCuO). Results indicated that with concentration increased, the available Cu content in soil increased significantly. The addition of CuO NPs increased Cu content and other nutrient element (e.g., K, P, Mn, and Zn) content in plants. However, diverse particle sizes had different effects. The nCuO treatment had larger translocation factor, higher nutrient element content in fruits, and lower oxidative damage than μCuO treatment. Moreover, nCuO of 100 mg/kg could stimulate cucumber growth, while μCuO had no obvious effects on growth. Conclusively, CuO NPs could be used as copper fertilizer to supply copper to cucumber. The nCuO had better effects on improving the bioavailability of Cu and nutritional value of fruits. These results can help develop strategies for safe disposal of CuO NPs as agricultural fertilizer.
Proteomic Analysis Unravels Response and Antioxidation Defense Mechanism of Rice Plants to Copper Oxide Nanoparticles: Comparison with Bulk Particles and Dissolved Cu Ions

Abstract:The increasing application of nanomaterials in agriculture raises environmental risks, but the information on how nanomaterials impact plant proteins is still limited. Here, we exposed the roots of rice seedlings to CuO nanoparticles (NPs, 100 mg/L), CuO bulk particles (BPs, 100 mg/L), and dissolved Cu ions (0.5 mg/L) for 14 days. The results show that CuO NPs caused more Cu accumulation, higher ascorbate peroxidase activity, and a higher ratio of GSH and GSSG but lower activity of Mn-SOD than BPs and Cu ions. Isotopic multilabel relative quantitative proteomics results indicate that NPs induced double differentially expressed proteins compared to BPs and Cu ions, especially phosphoglycerate kinase (fold change (FC) > 10) and glycosyltransferase (FC < 0.1). Unlike BPs and Cu ions, the proteins that responded to NPs were primarily connected with the C2 domain and glutathione S-transferase and significantly enriched for drug metabolism and monovalent inorganic cation transport processes and oxidative phosphorylation and phagosome pathways. Furthermore, almost 20% of proteins in the protein-protein interaction belonged to the ribosomal protein family. The parallel reaction monitoring further proved the defense response to NPs involving in redox, signal transduction, and heavy metal detoxification. Thus, proteomic analysis suggests that CuO NPs caused higher phytotoxicity mainly due to the nanospecific effect but elevated cysteine synthesis and GSSG reduction for detoxification.
Phytotoxic impact of copper oxide nanoparticles fabricated from rose petals ( Rosa indica ) on germination, biological growth, and phytochemicals of tomato ( Solanum lycopersicum )

Abstract:Global exploration of nanoparticles as nano-fertilizers/nano-fungicides to optimize crop production has attracted greater attention. Among metal oxide nano-formulations, the increasing application and discharge of copper oxide nanoparticles (CuO-NPs) into the environment requires urgent attention due to their toxic and stimulatory impact on plants, including vegetables. This study aimed at the bio-fabrication of CuO-NPs from Rosa indica petal extracts and to assess their nanotoxicity on germination and biological and biochemical growth of tomato plants. The morphological characterization of phytofabricated CuO-NPs by microscopic (SEM/TEM) and spectroscopic (XRD/FTIR) techniques revealed its nanostructure, spherical shape ranging in size between 23 and 47 nm exhibiting different functional groups. Furthermore, tomato-CuO-NPs interactions resulted in reduced germination percentage and plant biomass. At the biochemical level, the chlorophyll content decreased consistently, while malonaldehyde and proline content in tomato foliage enhanced substantially with increasing concentrations of CuO-NPs. Size, lycopene content (71 % reduction), and fruit quality (metabolites like sugars, amino acids, and organic acids) assessed by 1 H NMR declined after CuO-NPs exposure in a dose-dependent manner. Further in situ experiments revealed deformation in a stomatal aperture in leaves and accumulation of CuO-NPs in roots, suggesting toxicity under SEM. In summation, this study highlights the toxicity of biosynthesized CuO-NPs to tomato plants paving the way to develop novel strategies to mitigate its toxicity to vegetable crops, particularly tomato under real field conditions.

Abstract:The speedy growth of copper oxide nanoparticle (CuO NP) manufacturing due to their wide application in industries has caused concerns due to their increased discharge into the environment from both purposeful and accidental sources. Their presence at an elevated concentration in the environment can cause potential hazards to the plant kingdom, specifically to staple food crops. However, limited research is available to determine the consequences of CuO NPs. The present study aimed to assess the morphological and cytological changes induced by CuO NPs on Pisum sativum L., a key staple food crop. Seeds of Pisum sativum were exposed to various concentrations of CuO NPs (0, 25, 50, 75, 100, and 125 ppm) for 2 h, and their effects on seed germination (SG), radicle length (RL), cell proliferation kinetics (CPK), mitotic index (MI), cell death (CD), micronucleus frequency (MNF), and chromosomal aberration frequency (CAF) were studied. The results indicate a significant reduction in SG, RL, CPK, and MI and a significant dose-dependent increase in CD, MNF, and CAF. CuO NP treatment has led to abnormal meiotic cell division, increased incidence of micronucleus frequency, and chromosomal aberration frequency. Additionally, the CuO NP-treated groups showed an increase in the percentage of aberrant meiotic cells such as laggard (LG), double bridge (DB), stickiness (STC), clumped nuclei (CNi), precocious separation (PS), single bridge (SB), and secondary association (SA). CuO NP treatment led to reductions in SG as follows: 55% at 24 h, 60.10% at 48 h, and 65% at 72 h; reductions in RL as follows: 0.55 ± 0.021 cm at 24 h, 0.67 ± 0.01 cm at 48 h, and 0.99 ± 0.02 cm at 72 h; reductions in CPK as follows: 34.98% at prophase, 7.90% at metaphase, 3.5% at anaphase, and 0.97% at telophase. It also led to a 57.45% increase in CD, a 39.87% reduction in MI, and a 60.77% increase in MNF at a higher concentration of 125 ppm. The findings of this study clearly show that CuO NPs have a genotoxic effect on the food crop plant Pisum sativum.

Metabolomics Reveals How Cucumber (cucumis Sativus) Reprograms Metabolites to Cope with Silver Ions and Silver Nanoparticle-Induced Oxidative Stress

Abstract:Due to their well-known antifungal activity, the intentional use of silver nanoparticles (AgNPs) as sustainable nanofungicides is expected to increase in agriculture. However, the impacts of AgNPs on plants must be critically evaluated to guarantee their safe use in food production. In this study, 4-week-old cucumber (Cucumis sativus) plants received a foliar application of AgNPs (4 or 40 mg/plant) or Ag+ (0.04 or 0.4 mg/plant) for 7 days. Gas chromatography-mass spectrometry (GCMS)=based nontarget metabolomics enabled the identification and quantification of 268 metabolites in cucumber leaves. Multivariate analysis revealed that all the treatments significantly altered the metabolite profile. Exposure to AgNPs resulted in metabolic reprogramming, including activation of antioxidant defense systems (upregulation of phenolic compounds) and downregulation of photosynthesis (upregulation of phytol). Additionally, AgNPs enhanced respiration (upregulation of tricarboxylic acid cycle intermediates), inhibited photorespiration (downregulation of glycine/serine ratio), altered membrane properties (upregulation of pentadecanoic and arachidonic acids, downregulation of linoleic and linolenic acids), and reduced inorganic nitrogen fixation (downregulation of glutamine and asparagine). Although Ag ions induced some of the same metabolic changes, alterations in the levels of carbazole, lactulose, raffinose, citraconic acid, lactamide, acetanilide, and pbenzoquinone were AgNP-specific. The results of this study offer new insight into the molecular mechanisms by which cucumber responds to AgNP exposure and provide important information to support the sustainable use of AgNPs in agriculture.

Abstract:Because copper nanoparticles are being increasingly used in agriculture as pesticides, it is important to assess their potential implications for agriculture. Concerns have been raised about the bioaccumulation of nano-Cu and their toxicity to crop plants. Here, the response of cucumber plants in hydroponic culture at early development stages to two concentrations of nano-Cu (10 and 20 mg/L) was evaluated by proton nuclear magnetic resonance spectroscopy ((1)H NMR) and gas chromatography-mass spectrometry (GC-MS) based metabolomics. Changes in mineral nutrient metabolism induced by nano-Cu were determined by inductively coupled plasma-mass spectrometry (ICP-MS). Results showed that nano-Cu at both concentrations interferes with the uptake of a number of micro- and macro-nutrients, such as Na, P, S, Mo, Zn, and Fe. Metabolomics data revealed that nano-Cu at both levels triggered significant metabolic changes in cucumber leaves and root exudates. The root exudate metabolic changes revealed an active defense mechanism against nano-Cu stress: up-regulation of amino acids to sequester/exclude Cu/nano-Cu; down-regulation of citric acid to reduce the mobilization of Cu ions; ascorbic acid up-regulation to combat reactive oxygen species; and up-regulation of phenolic compounds to improve antioxidant system. Thus, we demonstrate that nontargeted (1)H NMR and GC-MS based metabolomics can successfully identify physiological responses induced by nanoparticles. Root exudates metabolomics revealed important detoxification mechanisms.

Abstract:Arthrobacter ramosus S34, which produces trehalose from maltooligosaccharide, was isolated. A trehalose-producing operon, treYZ, was cloned from the genome. Expression experiments with treY and treZ confirmed that they coded malto-oligosyltrehalose synthase and malto-oligosyltrehalose trehalohydrolase, respectively. The amino acid sequence of TreY from A. ramosus S34 and that from Arthrobacter sp. Q36 did not show high identity, nor did those to TreZ.
Copper oxide nanoparticle toxicity in mung bean (Vigna radiata L.) seedlings: physiological and molecular level responses of in vitro grown plants

Abstract:In this study, the toxic effect of copper oxide nanoparticles (CuONPs) at the physiological and molecular level was investigated in mung bean (Vigna radiata L.) plants. The seedlings were grown in half strength Murashige and Skoog medium supplemented with different concentrations of CuONPs (0, 20, 50, 100, 200 and 500 mg l−1) for 21 days under controlled growth conditions. Exposure to 200 and 500 mg l−1 of CuONPs significantly reduced shoot length and biomass. Significant reduction in root length and biomass was observed upon exposure to all concentrations of CuONPs. Retardation of primary and lateral root growth was observed upon exposure to different concentrations of CuONPs. At 100, 200 and 500 mg l−1 of CuONPs exposure, the total chlorophyll contents reduced significantly. Exposure to different concentrations of CuONPs has not resulted in any significant change in carotenoid contents. The proline content significantly increased upon exposure to 100, 200 and 500 mg l−1 of CuONPs. Significant increase in hydrogen peroxide content and lipid peroxidation was observed in roots upon exposure to 20, 50, 100, 200 and 500 mg l−1 of CuONPs. Histochemical staining with nitroblue tetrazolium and treatment with 3′-(p-hydroxyphenyl) fluorescein indicated a concentration-dependent increase in reactive oxygen species generation in roots. Exposure to CuONPs has resulted in excess lignification of roots cells as revealed by phloroglucionol-HCl staining. Gene expression analysis using real-time polymerase chain reaction showed modulations in the expression of CuZn superoxide dismutase, catalase and ascorbate peroxidase genes in roots of CuONPs exposed plants.

Metabolic Profile and Physiological Response of Cucumber Foliar Exposed to Engineered MoS2 and TiO2 Nanoparticles

Abstract:Nanoparticle (NPs) use is widespread across a wide range of sectors and accurate assessment of risk and/or benefits posed to important receptors such as plant species is necessary to ensure the sustainable development and application of nanotechnology. Here, 4-week old cucumber plants were foliar exposed to different doses (0-27 mg/plant) of MoS2 or TiO2 NPs for 16 d. Foliar NPs exposure had no impact on plant biomass or photosynthetic pigment production. However, the higher dose of TiO2 NPs exposure decreased lipid peroxidation by 34.7%, and the higher dose of MoS2 and TiO2 NPs exposure significantly increased total phenolics content by 27.4%-28.2%. The photosynthetic rate was increased by 23.6-30.8% with exposure to the higher dose of MoS2 NPs, as well as both lower and higher dose of TiO2 NPs. A metabolomic analysis revealed that 33 metabolites were upregulated or downregulated by MoS2 and TiO2 NPs exposure, including 8 amino acids, 5 organic acids, 3 fatty acids, 5 sugar and sugar derivatives, 4 alcohols, and 3 aromatic compounds. The fluctuation in metabolite content suggests that when plants are exposed to NPs, their metabolic processes are altered, and subsequent responses such as modulated defense, adaptation, and photosynthesis are displayed. With regard to dose, the higher concentration of TiO2 NPs altered several metabolic pathways, including carbohydrate and nitrogen metabolism, whereas the lower dose of TiO2 NPs and both doses of MoS2 had minimal impact on cucumber metabolism. These findings suggest that appropriate doses of MoS2 and TiO2 NPs have the potential to increase plant production by increasing plant photosynthetic rate without inducing excessive stress or toxicity.

Abstract:Increased impetus on the application of nano-fertilizers to improve sustainable food production warrants understanding of nanophytotoxicity and its underlying mechanisms before its application could be fully realized. In this study, we evaluated the potential particle size-dependent effects of soil-applied copper oxide nanoparticles ( n CuO) on crop yield and quality attributes (photosynthetic pigments, seed yield and nutrient quality, seed protein, and seed oil), including root and seed Cu bioaccumulation and a suite of oxidative stress biomarkers, in soybean ( Glycine max L.) grown in field environment. We synthesized three distinct sized (25 nm = S [small], 50 nm = M [medium], and 250 nm = L [large]) n CuO with same surface charge and compared with soluble Cu 2+ ions (CuCl 2 ) and water-only controls. Results showed particle size-dependent effects of n CuO on the photosynthetic pigments (Chla and Chlb), seed yield, potassium and phosphorus accumulation in seed, and protein and oil yields, with n CuO-S showing higher inhibitory effects. Further, increased root and seed Cu bioaccumulation led to concomitant increase in oxidative stress (H 2 O 2 , MDA), and as a response, several antioxidants (SOD, CAT, POX, and APX) increased proportionally, with n CuO treatments including Cu 2+ ion treatment. These results are corroborated with TEM ultrastructure analysis showing altered seed oil bodies and protein storage vacuoles with n CuO-S treatment compared to control. Taken together, we propose particle size-dependent Cu bioaccumulation-mediated oxidative stress as a mechanism of nCuO toxicity. Future research investigating the potential fate of varied size n CuO, with a focus on speciation at the soil-root interface, within the root, and edible parts such as seed, will guide health risk assessment of n CuO.

Abstract:BACKGROUND In order to address the challenges of food security for the ever-increasing population, the emergent of nanotechnology is an alternate technology of choice which recently accounts for the production of safer pesticides and as a substitute for conventional fertilizer. The antidrug resistance of Xanthomonas oryzae pv. oryzae (Xoo) and build-up of chemicals in the environment has made it necessary to find alternate safe measures that are effective in diseases management. Hence, in this study, copper oxide nanoparticles (CuONPs) was green synthesized by Hibiscus rosa-sinensis L. flower extract. RESULTS The characterization of CuONPs using UV - Vis spectrophotometer, Scanning electron microscopy (SEM) with Energy dispersive spectrum (EDS) profile, Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD) ascertained the presence of CuONPs which were nanorods of 28.1 nm. CuONPs significantly obstructed growth and biofilm development of Xoo by 79.65 and 79.17 % respectively. Antibacterial mechanism of CuONPs was found to be as a result of wounding cell membrane, hence, giving rise to exodus of intracellular content and generation of oxidative reactive oxygen species which invariably, inhibited Xoo respiration and growth. The toxicity study under greenhouse conditions revealed that CuONPs significantly increased growth variables and biomass of rice and reduced bacterial leaf blight. Application of CuONPs on Arabidopsis improved the chlorophyll fluorescence parameters; the ΦPSII was significantly increased by 152.05 % in comparison to control. CONCLUSION Altogether, this result suggests that CuONPs in low concentration (200.0 μg/ml) are not toxic to plants and can serve as nano-fertilizers and nano-pesticides. It was found that ZnO or CuO NPs had a stronger inhibitory effect on wheat growth than Zn 2+ or Cu 2+ . After exposure to ZnO or CuO NPs, wheat seedlings accumulated significantly higher levels of Zn or Cu than the corresponding Zn 2+ or Cu 2+ treatments, indicating the active uptake of NPs via wheat root. TEM analysis further confirmed the intake of NPs. Moreover, ZnO or CuO NPs exposure altered micronutrients (Fe, Mn, Cu, and Zn) accumulation in the tissues and decreased the activities of antioxidant enzymes. The metabolomics analysis identified 312, 357, 145, and 188 significantly changed metabolites (SCMs) in wheat root exposed to ZnO NPs, CuO NPs, Zn 2+ , and Cu 2+ , respectively. Most SCMs were nano-specific to ZnO (80%) and CuO NPs (58%), suggesting greater metabolic reprogramming by NPs than metal ions. Overall, nanospecific toxicity dominated the phytotoxicity of ZnO and CuO NPs, and our results provide a molecular perspective on the phytotoxicity of metal oxide NPs.

Abstract:The release of nanoparticles (NPs) to the environment poses an increasing potential threat to biological systems. This study investigated the phytotoxicity and accumulation of copper oxide (CuO) NPs to Elsholtzia splendens (a Cu-tolerant plant) under hydroponic conditions. The 50% effective concentration (EC50) of CuO NPs to E. splendens was about 480 mg/L, implying the tolerance of E. splendens to CuO NPs. The Cu content in the shoots treated with 1000 mg/L CuO NPs was much higher than those exposed to the comparable 0.5 mg/L soluble Cu and CuO bulk particles. CuO NPs-like deposits were found in the root cells and leaf cells. Cu K-edge X-ray absorption near-edge structure analysis further revealed that the accumulated Cu species existed predominantly as CuO NPs in the plant tissues. All these results suggested that CuO NPs can be absorbed by the roots and translocated to the shoots in E. splendens.

Abstract:The increased production and use of nanoparticles (NPs) has generated concerns about their impact on living organisms. In this study, nCu, bulk Cu, nCuO, bulk CuO, Cu(OH)(2) (CuPRO 2005, Kocide 3000), and CuCl2 were exposed for 15 days to 10 days-old hydroponically grown lettuce (Lactuca sativa) and alfalfa (Medicago sativa). Each compound was applied at 0, 5, 10, and 20 mg L-1. At harvest, we measured the size of the plants and determined the concentration of Cu, macro and microelements by using ICP-OES. Catalase and ascorbate peroxidase activity was also determined. Results showed that all Cu NPs/compounds reduced the root length by 49% in both plant species. All Cu NPs/compounds increased Cu, P, and S (>100%, >50%, and >20%, respectively) in alfalfa shoots and decreased P and Fe in lettuce shoot (>50% and >50%, respectively, excluding Fe in CuCl2 treatment). Biochemical assays showed reduced catalase activity in alfalfa (root and shoot) and increased ascorbate peroxidase activity in roots of both plant species. Results suggest that Cu NPs/compounds not only reduced the size of the plants but altered nutrient content and enzyme activity in both plant species.

Abstract:Excessive use of pesticides and mineral fertilizers poses a serious threat to ecoenvironment sustainability and human health. Nano pesticides or Nano fungicides have attained great attention in the field of agriculture due to their unique characteristics, by improving crop growth with enhancing pathogenesis-related defense system. However, there is a need to develop a sustainable mechanism for the synthesis of fungicides which replace the chemical pesticides to avoid their hazardous impact. Here in, Tamarix aphylla mediated CuO-Nanoparticles (NPs) were synthesized, characterized and their activity was evaluated under in-vitro and in-vivo conditions. The structural and elemental analysis of NPs were carried out by using X-ray powder diffraction (XRD), Fourier transformed infrared spectroscopy (FTIR), UV–visible spectrophotometer, Scanning electron microscope (SEM) and Transmission electron microscope (TEM). In the greenhouse, at an optimum concentration of 50 mg/L reduced disease severity very effectively and enhanced plant growth. Application of NPs also assisted in the induction of systemic response of defense-related genes in melon. Under In vitro condition at 100 mg/L significantly reduced mycelial growth (84.5%) by directly acting on the pathogenic cell wall. Our work confirmed that dosedependent concentration of T. aphylla extract based biological CuO-NPs enhance plant growth and help to effectively resist against F. oxysporum infection.

Abstract:Rice (Oryza sativa L.), a major staple food for billions of people, was assessed for its phytotoxicity of copper oxide nanoparticle (CuO NPs, size < 50 nm). Under hydroponic condition, seven days of exposure to 62.5, 125, and 250 mg/L CuO NPs significantly suppressed the growth rate of rice seedlings compared to both the control and the treatment of supernatant from 250 mg/L CuO NP suspensions. In addition, physiological indexes associated with antioxidants, including membrane damage and antioxidant enzyme activity, were also detected. Treatment with 250 mg/L CuO NPs significantly increased malondialdehyde (MDA) content and electrical conductivity of rice shoots by 83.4% and 67.0%, respectively. The activity of both catalase and superoxide dismutase decreased in rice leaves treated with CuO NPs at the concentration of 250 mg/L, while the activity of the superoxide dismutase significantly increased by 1.66 times in rice roots exposed to 125 mg/L CuO NPs. The chlorophyll, including chlorophyll a and chlorophyll b, and carotenoid content in rice leaves decreased with CuO NP exposure. Finally, to explain potential molecular mechanisms of chlorophyll variations, the expression of four related genes, namely, Magnesium chelatase D subunit, Chlorophyll synthase, Magnesium-protoporphyrin IX methyltransferase, and Chlorophyllide a oxygenase, were quantified by qRT-PCR. Overall, CuO NPs, especially at 250 mg/L concentration, could affect the growth and development of rice seedlings, probably through oxidative damage and disturbance of chlorophyll and carotenoid synthesis.

Abstract:The effects of CuO NPs and bulk Cu at 0–1000 mg L−1 on the growth, photosynthesis and biochemical parameters were investigated in 30-day-old rice plants grown hydroponically. ICP-OES measurements showed that CuO NPs released ≤ 1 mg L−1 of Cu2+ ions compared with ≤ 81 mg L−1 by bulk Cu at their highest concentration. Both treatments showed that growth, photo-phosphorylation and carbon dioxide assimilation declined considerably. Bulk particles caused oxidative stress whereas NP showed no such effect. Electromicrographs showed that CuO NPs accumulated in chloroplasts resulting in destacking and distortions of thylakoid membranes while bulk Cu showed no such behaviour. Results suggest that NP affected the growth by accumulation in non-ionic form in chloroplasts causing damage to thylakoid membrane without oxidative damage, whereas the bulk Cu affected the growth by causing oxidative damage as a result of release of Cu2+ ions without affecting the ultrastructure of the chloroplasts.
