Abstract:Abstract Benzoin, also known as semipetrified amber, is the resin secreted from the trunk of the benzoin plant. This medicinal material has blood circulation-promoting and pain-relieving properties. There are approximately 130 species of benzoin plants in the world, and the resin of many plants in the benzoin genus is used, so there is confounding of the species of origin. In this study, DNA was successfully extracted from the semipetrified amber medicinal material benzoin, and the species of benzoin in the medicinal herb market were evaluated by DNA barcoding. BLAST comparison of ITS2 primary sequences and homology prediction analysis of ITS2 secondary structures showed that the commercially available benzoin base originated from Styrax tonkinensis (Pierre) Craib ex Hart. and Styrax japonicus Sieb. et Zucc. of the genus Styrax Linn., and tissue from other genera was also detected to be mixed in with some samples, accounting for 29.6% of the samples. There is a risk that benzoin resin medical materials will be mixed with tissues from other plants. This is the first report on the subject, and the approach taken provides technical feasibility for the identification of the basis of the benzoin resin medical materials.
Benzoin Resin: An Overview on Its Production Process, Phytochemistry, Traditional Use and Quality Control

Abstract:Benzoin is a pathologic exudation produced by plants of the family Styrax. It is secreted by traumatic resin ducts after injury, which are derived from parenchymal cells in secondary xylem by schizolysigeny. Some 63 chemical constituents have been isolated and identified from this resin, including balsamic acid esters, lignans and terpenoids. It has a long history of applications, including as incense along with olibanum, a flavor enhancer in the food industry, materials in the daily chemistry industry as well as therapeutic uses. Up to now, high-performance liquid chromatography (HPLC) and gas chromatography mass spectrometry (GC-MS) have been widely used in qualitative and quantitative analysis of benzoin. Other technologies, including near-infrared reflectance spectroscopy (NIR), proton transfer reaction time-of-flight mass spectrometry (PTR-ToF-MS) and Fourier-transform infrared spectroscopy (FT-IR), have also been used to distinguish different resins. Herein, this paper provides a comprehensive overview of the production process, phytochemistry, traditional uses and quality control of benzoin and looks to the future for promoting its further research and applications.
Comparative metabolomic reveals chemotaxonomic markers of resin fossils for identification of botanical origins

Abstract:Fossil resins are often used to indicate the botanical origins of ancient plants through their secondary metabolites. However, the existing chemotaxonomy of fossil resins is limited in its ability to distinguish them at phylum, family, and genus levels using small amount of samples. Herein an advanced head-space solid phase microextraction- comprehensive two-dimensional gas chromatography- time-of-flight mass spectrometer platform was established to analyze the complex compositions of resin fossils derived from ancient gymnosperms (e.g., Eocene Cupressaceae , Cretaceous Araucariaceae ) and angiosperms (Miocene Dipterocarpaceae and Miocene Hymenaea ). The secondary metabolites including 83 monoterpenes, 186 sesquiterpenes, 84 diterpenes, and 43 n-alkanes were annotated and then chemically fingerprinted for the first time. Prior to conducting chemical composition classification, this study tested the vitrinite reflectance of resin-bearing rocks and Tmax of the ambers to demonstrate their similar thermal maturity. After that, chemotaxonomic markers were disclosed using multivariate statistical analysis to identify the botanical origins of resin fossils, and four identification rules were concluded. Gymnosperm and angiosperm resins can be distinguished by the relative content of monoterpenes and diterpenes. Eocene Cupressaceae and Cretaceous Araucariaceae resins can be identified by ten chemotaxonomic markers which were screened out by orthogonal partial least-squares regression. Miocene Hymenaea and Dipterocarpaceae resins have different combinations of chemical superclasses, with sesqui- and di-terpenes being the major compositions of Hymenaea resins, and mono- and sesqui- terpenes accounting for most of the compositions of Dipterocarpaceae resins. Specific chemotaxonomic markers can identify Hymenaea mexicana and Hymenaea protera resins. As the chemical components of fossil resins were determined by the genes present in their botanical origins, the chemotaxonomic markers revealed in this study have a genetic basis. These findings provide a comprehensive database of secondary metabolites from ancient resin plants, which can be utilized for further research on their corresponding paleoenvironment and to clarify their phylogeny.

Abstract:Benzoinum is a resin derived from Styrax tonkinensis bark that has long been used in clinical settings as a traditional Chinese medicine with cardioprotective properties. The mechanistic basis for these pharmacological properties, however, remains to be demarcated. We used a network pharmacology approach to characterise the mechanism of action that benzoinum used to treat AS. The PharmMapper database was used to find potential benzoin target proteins, and the TCMSP, CTD, TTD, GAD, drug Bank, and pharmGkb databases were used to identify AS-related target genes. Following assessments of pathway enrichment, the functional roles of these genes were discovered, allowing the creation of compound-target and target-pathway networks. HUVEC cells were then used to validate these predicted benzoinum-related mechanisms of action, revealing that an active derivative of this resin was able to modulate signaling activity in TNF-Î±-exposed HUVECs by regulating the caspase-9 and NF-ÎºB signaling pathways. Overall, our predictive network pharmacology analyses and supporting data highlighted the potential mechanisms whereby benzoinum may exert cardiovascular activity in the context of AS.
Qualitative and Quantitative Secondary Metabolite Profiles in a Large Set of Sumatra Benzoin Samples

Abstract:Sumatra benzoin, a resin produced by Styrax benzoin and Styrax paralleloneurum, is used as an aromatic agent and may have the potential to be developed as a new agricultural fungicide. In this context, we performed a comprehensive metabolite profiling of a commercial grade A resin by high-performance liquid chromatography coupled with photodiode array detection, evaporative light scattering detection, and mass spectrometry (HPLC-PDA-ELSD-MS) analysis in combination with 1H NMR. Thirteen compounds including a new cinnamic acid ester containing two p-coumaroyl residues were identified after preparative isolation. These compounds accounted for an estimated 90% of the crude resin according to 1H NMR analysis. The two major constituents, p-coumaryl cinnamate (5) and sumaresinolic acid (11), were quantified by HPLC analysis. In a next step, the chemical profiles and the content in p-coumaryl cinnamate were compared in a large set of resin samples of different quality grades that were obtained from various commercial suppliers in Sumatra. The qualitative profiles of the samples were very similar, but significant quantitative differences were observed between different quality grades and origins of the samples for the relative contents.
Three-step identification of infrared spectra of similar tree species to Pterocarpus santalinus covered with beeswax

Abstract:It is difficult to distinguish the Pterocarpus santalinus in the furniture wood market, since it is similar to other tree species such as Dalbergia louuelii and Pterocarpus tinctorius. Moreover, the features of the wood are usually covered with the beeswax on the surface, making it even harder to be identified the tree species. Extractive is important components of wood and plays a significant role in the color of wood. Benzene-alcohol extractives contain specific chemical components of wood, which is an important part to distinguish tree species. Color parameters show that the color is obviously deepened when the sample is covered with beeswax. As shown in the SEM examination, beeswax appears on the wood surface in a large area and is immersed in the vessels of wood. In this paper, with the infrared spectrum three-step identification, the differences of the chemical composition of benzene-alcohol extractive are analyzed to distinguish tree species. The results show that the functional groups of the compounds contained in beeswax are mainly composed of long-chain methyl -CH3, methylene –CH2- and ester carbonyl CO, carboxyl C–O stretching at 2956, 2917, 2850, 1740, 1468, 1378 and 721cm-1. The absorption peaks of beeswax are so strong that they weaken the peaks of benzene-alcohol extractives from three tree species. In the FT-IR and SD-IR spectra, P. santalinus and P. tinctorius have strong peaks at 1736 cm−1, 1590 cm−1, 1151 cm−1 (1149 cm−1), 1067 cm−1 (1064 cm−1), 1033 cm−1, 958 cm−1 (959 cm−1), and 835 cm-1 (833 cm-1), while D. louvelii has a weak broad peak at 1630 cm-1 and 1602 cm-1, and there is no absorption peak at other wave numbers. At 754 cm-1 and 700 cm-1, no absorption peaks are found in the other two tree species. In the Synchronous 2DCOS-IR spectra in the ranges of 900–1200cm-1,1200-1425 cm-1, and 1480-1700cm-1, the sensitivity of some functional groups in P. santalinus and D. louvelii for the temperature perturbation tends to be the same. The main difference between them is that more functional groups of P. santalinus are sensitive to temperature disturbance. For P. tinctorius, it is sensitive to temperature perturbation, in which, the ester groups C–O stretching vibration at 1151, 1168, 1202 cm-1 and aromatic ring skeleton stretching belongs to flavonoids and lignin at 1511 and 1594 cm-1. The above results indicate that the three-step identification by the infrared spectroscopy can accurately and quickly distinguish tree species.

Cinnamic Acid in Frankincense Sap as a Criterion for Determining the Best Mother Plant for Vegetative Propagation of Styrax benzoin (Sumatra Benzoin) in Sumatra, Indonesia

Abstract:Cinnamic acid, contained in frankincense sap produced by Styrax benzoin (Sumatra benzoin tree), is an important compound that is used for various purposes, such as preservatives, fragrances, cosmetics, and pharmaceutical products. The production of frankincense sap as a forest product can be increased through the inclusion of S. benzoin seeds, which are propagated from the best mother plants. This study aims to use the content of cinnamic acid contained in the sap of S. benzoin as a criterion for determining the best mother plant for propagation of seeds. The research was conducted using healthy plants, taking sap samples, identifying and confirmation of cinnamic acid levels, and determining the best mother tree based on the content of cinnamic acid. The results of this study have identified six individual S. benzoin trees of very good quality based on their phenotypic advantages and the quantity of sap production. Isolation and identification of cinnamic acid from frankincense samples showed that the composition of cinnamic acid was high (12 to 21%). Three good quality S. benzoin trees, with high cinnamic acid composition, were SBN-7 (21%), SBN-3 (18%), and SBN-10 (17%). The SBN-7 tree was then chosen as the best S. benzoin, producing 2.70 kg year−1 sap, containing cc. 21% cinnamic acid. A selected mother plant will be used as a source of plant material for vegetative propagation to produce good quality seeds similar to the properties of the parent plant forforest conservation and to increase the production of nontimber forest raw materials for medicine and other purposes. The finding of this study is the first to use the composition of cinnamic acid as a criterion for determining the best mother plant. The composition of cinnamic acid in the sap is an important parameter in determining the superiority of S. benzoin plants.
Rapid Identification of Commercial Frankincense Products by MALDITOF Mass Spectrometry

Abstract:Background: Frankincense is a resin secreted by the Boswellia tree. It is used in perfumery, aromatherapy, skincare, and traditional Chinese medicine. However, all Boswellia species are under threat owing to habitat loss and overexploitation. As a result, the market is getting flooded with counterfeit frankincense products. Objective: This study aims to establish a high-throughput method to screen and identify the authenticity of commercial frankincense products. We report, for the first time, a matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS)-based method for rapid and high-throughput screening of frankincense samples. Methods: MALDI-TOF MS, HPLC, thin-layer chromatography (TLC), and in vitro antiinflammatory activity assay were used to examine the frankincense samples. Results: Well-resolved peaks of frankincense triterpenoids in the spectra were observed in the crude extract of commercial samples, including α-boswellic acids (αBAs), β-boswellic acids (βBAs), 11-keto-β-boswellic acids (KBAs), acetyl-11-keto-β-boswellic acids (AKBAs), and their esters. These compounds can be used as indicators for determining the authenticity of frankincense. Conclusion: Unlike LC-MS, which is a time-consuming and expensive method, and TLC, which requires a reference sample, our inexpensive, rapid high-throughput identification method based on MALDI-TOF MS is ideal for large-scale screening of frankincense samples sold in the market.
Rapid identification and on-site analysis by miniature mass spectrometry of chemical markers for fragrant rosewood authentication

Abstract:Dalbergia odorifera T. Chen (Dalbergiae Odoriferae Lignum, DOL), renowned for its therapeutic applications in traditional Chinese medicine and its value in crafting luxury items, faces challenges due to the slow heartwood formation process and subsequent market adulteration. The proliferation of counterfeit products necessitates reliable methods for rapid, on-site authentication. This study aims to establish a rapid, green, and on-site method to identify the authenticity of DOL. A comprehensive analysis was conducted on 10 batches of DOL samples sourced from diverse locations, utilizing a miniature mass spectrometer (mini MS) equipped with a paper capillary spray (PCS) technique. Multivariate statistical approaches were employed to classify the samples and pinpoint chemical markers indicative of authenticity. Subsequent MS-guided separation and thin-layer chromatography (TLC) verified the markers' validity and assessed the greenness profile by Analytical Eco-scale (AES), Green Analytical Procedure Index (GAPI), and Analytical GREEnness (AGREE). A total of 10 batches of DOL samples detected by PCS-mini MS were classified into authentic and counterfeit, by unsupervised cluster analysis. Sativanone (m/z 301.1, VIP=6.0, p=0.000001) and 3'-O-methylviolanone (m/z 331.1, VIP=3.2, p=0.000382) were regarded as the chemical markers for the rapid identification of DOL. The results of the TLC method were consistent with this method, and the new method is greener. The application of mini MS for on-site authentication of DOL via specific chemical markers offers significant advantages, including operational simplicity, high efficiency, greenness, and accuracy. The deployment of this strategy promises to facilitate the effective regulation of DOL, ensuring authenticity and quality.
Molecular Authentication of the Medicinal Species of Ligusticum (Ligustici Rhizoma et Radix, “Gao-ben”) by Integrating Non-coding Internal Transcribed Spacer 2 (ITS2) and Its Secondary Structure

Abstract:Ligustici Rhizoma et Radix (LReR), an important Chinese medicine known as "Gao-ben," refers to Ligusticum sinense Oliv. or Ligusticum jeholense Nakai et Kitag. However, a number of other species are commonly sold as "Gao-ben" in the herbal medicine market, which may result in a series of quality control problems and inconsistent therapeutic effects. The "Gao-ben" is commonly sold sliced and dried, making traditional identification methods difficult. Here, the mini barcode ITS2 region was examined on 68 samples representing LReR and 7 potential adulterant or substitute species. The results showed 100% success rates of PCR and sequencing and the existence of a barcoding gap. The neighbor-joining (NJ) tree indicated that all the tested samples could be exactly identified. The ITS2 secondary structure revealed a clear difference between true "Gao-ben" and three adulterant species. We therefore recommend the use of ITS2 as a mini barcode for distinguishing between closely or distantly related plant species that may be used in Chinese medicine.

Abstract:Traditional herbal patent medicine typically consists of multiple ingredients, making it challenging to supervise contamination by impurities and the improper use of raw materials. This study employed shotgun metabarcoding for the species identification of biological ingredients in traditional herbal patent medicine, Wuhu San. The five prescribed herbal materials found in Wuhu San were collected, and their reference sequences were obtained by traditional DNA barcoding using Sanger sequencing. Two lab-made and three commercial Wuhu San samples were collected, and a total of 37.14 Gb of shotgun sequencing data was obtained for these five samples using the Illumina sequencing platform. A total of 1,421,013 paired-end reads were enriched for the Internal Transcribed Spacer 2 (ITS2), psbA and trnH intergenic spacer region ( psbA-trnH ), maturase k ( matK ), and ribulose-1, 5-bisphosphate carboxylase ( rbcL ) regions. Furthermore, 80, 11, 9, and 8 operational taxonomic units were obtained for the ITS2, psbA-trnH , matK , and rbcL regions, respectively, after metagenomic assembly, annotation, and chimeric detection. In the two lab-made mock samples, all labeled ingredients in the Wuhu San prescription were successfully detected, and the positive control, Panax quinquefolius L., was detected in the HSZY172 mock sample. Three species, namely Angelica sinensis (Oliv.) Diels, Saposhnikovia divaricata (Turcz. ex Ledeb.) Schischk., and Carthamus tinctorius L., belonging to three labeled ingredients, Angelicae Sinensis Radix (Danggui), Saposhnikoviae Radix (Fangfeng), and Carthami Flos (Honghua), were detected in the three commercial samples. Angelica dahurica (Hoffm.) Benth. & Hook. f. ex Franch. & Sav., the original Angelicae Dahuricae Radix (Baizhi) species, was only detected in WHS003. Arisaema erubescens (Wall.) Schott, Arisaema heterophyllum Blume, or Arisaema amurense Maxim., the original Arisaematis Rhizoma (Tiannanxing) species, were not detected in any of the commercial samples, which could be attributed to the fact that this medicinal material underwent extensive processing. In addition, the Saposhnikovia divaricata adulterant was detected in all the commercial samples , while 24 fungal genera, including Aspergillus , were identified in both the lab-made and commercial samples. This study showed that shotgun metabarcoding provided alternative strategy and technical means for identifying prescribed ingredients in traditional herbal patent medicine and displayed the potential to effectively complement traditional methods.

Abstract:The analgesic effect of the resin of Boswellia carterii (BC) is well known; however, the constituents that contribute to the analgesic effect remain elusive. The current study integrates ultrasonic-assisted extraction, quantitative determination, analgesic evaluation in rats, and gray relationship analysis for tracing analgesic constituents from the resin of BC. First, a robust and precise ultra-performance liquid chromatography tandem mass spectrometry approach with multiple reaction monitoring mode was developed for the simultaneous quantification of seven major constituents in crude and vinegar-processed resin of BC. Glycyrrhetinic acid was chosen as the internal standard. The approach showed good linearity. The intra- and inter-day precisions of each constituent were within 3.0%. The recoveries of each constituent were in the range of 96.4-102.7%. The approach was then applied to determine the seven constituents in 10 batches of crude and vinegar-processed resin of BC. Second, the analgesic effects of crude and vinegar-processed resin of BC were assessed in mice. Third, chemometrics methods, gray relationship analysis, and partial least squares regression were employed for determining the relationship between the contents of seven constituents and their analgesic effects. 11-Keto-β-boswellic acid, 3-acetyl-β-boswellic acid, 3-acetyl-α-boswellic acid, 3-acetyl-11-keto-β-boswellic acid, and β-sitosterol were identified as the key analgesic constituents of BC.
Comprehensively feature-based molecular networking and metabolomics approaches to reveal the differences components in Cinnamomum cassia and Cinnamomum verum

Abstract:Cinnamon was been a widely used plant in medicinal and spices for a long time and has spread all over the world. However, the differences in the components of the bark from Cinnamomum cassia and Cinnamomum verum, the two most common types of cinnamon, have not been thoroughly investigated. In the present experiment, ultra-high-performance liquid chromatography LTQ-Orbitrap Velos Pro hybrid mass spectrometer-based metabolomics coupled with chemometrics and feature-based molecular networking were employed to dramatically distinguish and annotate Cinnamomum cassia Bark and Cinnamomum verum bark. As a consequence, principal component analysis, orthogonal projection to latent structures discriminates analysis, and heat map analysis demonstrated clear discrimination between the profiles of metabolites in cinnamon. Besides, as the known compounds, proanthocyanidins (cinnamtannin B1 and procyanidin B2) and alkaloids (norboldine, norisoboldine) with variable importance in the projection scores >6, and an unknown alkaloid (formula C24H33NO6) were selected as the best markers to discriminate cinnamon. Furthermore, large numbers of proanthocyanidins and alkaloids components were identified through feature-based molecular networking for the first time. Our investigation provides new ideas for the discovery of quality markers and identification of unknown components in natural products.
On-Site Analysis and Rapid Authentication of Valuable Agarwood Samples by Paper Capillary Spray Miniature Mass Spectrometry

Abstract:Agarwood is a resinous wood produced by plants of the genus Aquilaria in the Thymelaeaceae family after being injured. The wide distribution of origins and the complexity of formation have led to various species of agarwood. The preciousness, universality of application and scarcity of resources bring difficulties to the quality control of agarwood, the in-situ ionization technique shows great potential in the rapid and accurate identification of valuable herbal medicines. In this paper, based on the combination of paper capillary spray and miniature mass spectrometry, different kinds of agarwood and counterfeits were directly added onto the chromatography paper, the active ingredients were extracted by solvent and then carried out by the portable miniature mass spectrometry, which could be implemented to perform an effective and on-site analysis, and the whole process only took about 1 min. Characteristic highly oxidized 2-(2-phenylethyl)chromones and flindersia-type 2-(2-phenylethyl)chromones were detected in authentic agarwood, and other characteristic flindersia-type 2-(2-phenylethyl)chromones were detected in authentic Kynam agarwood (special variety of agarwood), while the above characteristic components were not detected in counterfeits. Furthermore, an artificial intelligence model based on random forest algorithm was established for automatic identification. Phytochemical characterization of Styrax benzoin resin extract, molecular docking, ADME, and antibacterial activity study

Abstract:Styrax benzoin fumes have a spiritual aspect from ancient times, magical essence like a pleasant perfume, and are employed in religious ceremonies in India. This study aims to identify the volatile compounds in S. benzoin extract, their binding affinity to the bacterial target proteins, and study the antibacterial activity of the potential extract. The compounds obtained from GC-MS analysis of S. benzoin extract were subjected to molecular docking studies against DHFR of Staphylococcus aureus, tRNA synthetase of Escherichia coli , DHPS of Mycobacterium tuberculosis . Molecular docking studies revealed that seventeen compounds out of 20 compounds exhibited higher binding affinity than co-ligand (-7.00 kcal/mol) against the Staphylococcus aureus enzyme DHFR. Consequently, the crude extracts were evaluated for antibacterial activity against S. aureus, and the acetone extract showed promising findings. S. benzoin fumes might replace synthetic room fresheners, and promising compounds could be exploited in the cosmetics industry. Graphical

Abstract:The Chinese term Anxi xiang 安息香 is now usually applied to benzoin, an aromatic resin obtained from species of the genus Styrax native to Southeast Asia. However, it appears from Chinese records that this aromatic originally was imported into China from western Asia. What this early Anxi xiang was has never been ascertained. It is here suggested that, like benzoin, it was produced from a species of Styrax , and was the storax of the ancient Mediterranean region. A complication is that storax was obtained from more than one plant source. Another issue is that a different substance, known to the Chinese as suhe 蘇合, has often been identified with storax. The available evidence is assessed and probable solutions are proposed.
Identification of Veratrum Species in Pimacao Based on ITS2 Sequences and Steroidal Alkaloids by a Pseudo-Targeted Metabolomics Method

Abstract:Pimacao is a traditional Chinese folk medicine and is the main component of the famous Chinese herbal remedy “Yunnan Baiyao” for its significant analgesic activity in the treatment of wounds. Due to increases in consumption, its wild population is now difficult to find, and adulterant from the same genus has occurred. However, this is challenging to distinguish the species of Veratrum in Pimacao using dried roots and rhizomes or medicinal powder. ITS2 sequences and steroidal alkaloids by the non-targeted and pseudo-targeted metabolomics methods were taken advantage of establishing an effective identification method. Based on the ITS2 sequence, metabolite profiling of steroidal alkaloids and morphological characteristics, the classification of two distinct subspecies in V. mengzeanum has been reinforced. In addition, the new subspecies V. mengzeanum subsp. phuwae was collected in China for the first time. The ITS2 sequence could be used in the identification of V. taliense , V. mengtzeanum , V. stenophyllum , and V. nigrum , but is insufficient for intraspecific identification. Simultaneously, 147 variables were labeled by non-targeted analysis accomplished utilizing an ultra-high-performance liquid chromatography electrospray ionization orbitrap tandem mass spectrometry (UPLC-ESI-QE-Orbitrap-MS) system consisting of an Orbitrap QE HF-X. Followed by a pseudo-targeted analysis method developed for the Qtrap 6500-plus mass spectrometry system coupled with an ESI source, 29 labeled steroidal alkaloids detected by the MRM mode could distinguish between four species. Notably, 25 labeled steroidal alkaloids could distinguish between three closely related species. These have the potential to be used as markers for identification. Furthermore, there were several variables with statistical differences between two subspecies of V. mengtzeanum and populations of V. taliense , V. mengtzeanum , and V. stenophyllum .

Abstract:Arnebia decumbens (Vent.) Coss. et Kralik , A. euchroma (Royle) Johnst and A. guttata Bunge, three commonly used traditional Chinese medicinal plants have been widely used for the clinical treatment of inflammatory diseases caused by fungal, bacterial, oxidation, and other related pathogens. However, precise identification at the similar species level is usually challenging due to the influence of the source of medicinal materials, traditional ethnic medicine and medicinal habits. Here we developed a comprehensive and efficient identification system for three source spices of Arnebiae Radix based on DNA barcoding and HPLC fingerprinting. A total of 599 samples from thirty-five wild populations were collected and identified by using DNA barcodes of ITS2 regions, and the chemotypes of seven naphthoquinoneswere revealed by HPLC quantitative analysis including principal component analysis and hierarchical clustering analysis. Our results showed that the ITS2 sequences can distinguish three source spices of Arnebiae Radix from adulterants. However, it was difficult to identify them by HPLC-specific chromatograms combined with chemometric analysis. These results indicated that DNA barcoding was a more powerful method than HPLC fingerprinting for the identification of related species that were genetically similar. DNA barcoding analysis could be a promising and reliable tool to accurately confirm the identities of medicinal materials, especially for those whose sources are multiple and difficult to be identified by conventional chromatography.

Abstract:Herbal materials have both medicinal and commercial values. As such, accurate species and content identification and verification are necessary to ensure the safe and effective use for medical and commodity purposes. Herein, we introduce a two-step approach for systematic identification and quality evaluation of wild and introduced Anemone flaccida Fr. Schmidt (aka Di Wu) using DNA barcode and ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS/MS). To begin, a precise and rapid identification method based on internal transcribed spacer 2 (ITS2) sequence was developed to ensure the authenticity of 'Di Wu' species. Next, the major active components were fully characterized utilizing a targeted profile of oleanane-type triterpenoid saponins, which was established via UPLC-QTOF-MS/MS. As a result, 34 oleanane-type triterpenoid saponins were identified or characterized in 'Di Wu.' The qualitative and relative quantitative analysis showed obvious differences between wild and introduced 'Di Wu.' Furthermore, dynamic changes in the contents of triterpenoid saponins throughout various harvesting periods were clearly explained and mid-April was identified as the appropriate harvest time. Moreover, results indicate that the contents of five main saponins (anhuienoside E, glycosideSt-I4a, hemsgiganoside B, flaccidoside II, and hederasaponin B) are more appropriate as a quality evaluation indicator than the current quality standard. The two-step approach provides a suitable strategy to evaluate the genuine quality of wild and introduced 'Di Wu,' and can be applied to the targeted analysis of other triterpenoid saponin analogues for quality evaluation.

Abstract:Agarwood is a valuable traditional medicine and fragrance. The production process is a typical injury-induced defense response. Currently, there are approximately 22 known species in the genus Aquilaria Lam., all of which can produce agarwood, whereas there are only two legal species of traditional Chinese medicinal agarwood, Aquilaria sinensis (Lour.) Spreng. and Aquilaria agallocha (Lour.) Roxb. The Taiwan herbal Pharmacopoeia of China stipulates that the medicinal agarwood species are A. sinensis and its relatives in the same genus. Moreover, there are five species of agarwood available for clinical medicinal use in Japan, including A. agallocha and A. sinensis, which are often confused with each other or used in a mixed way in the trade process. Therefore, accurate identification of traditional Chinese medicinal agarwood species is important to ensure the authenticity of traditional medicines and to guide the safety of clinical medication. In this study, 59 specific single-nucleotide polymorphism loci were screened and obtained from the chloroplast genomes of 12 species of the genus Aquilaria Lam. We established an identification method for traditional Chinese medicinal agarwood using mini-barcoding combined with high-resolution melting (HRM) and designed and validated 10 pairs of primers from the psbM-trnD, psbA, rps16, petN, ndhE-psaC, rps4, atpE, ycf1, rps15-trnN, and matK regions. The amplification products were all less than 200 bp, with a high success rate of amplification. The method was applied to successfully identify traditional Chinese medicinal agarwood species from commercial agarwood samples. Overall, the sensitivity of this method was sufficient to detect 1% of adulterants in medicinal agarwood products, proving that mini-barcoding HRM is a powerful and flexible tool. This method can be used as a fast and effective high-throughput method for authenticity testing of traditional Chinese medicinal agarwood and its raw materials containing agarwood-containing proprietary Chinese medicines and is recommended for industrial applications.
