Abstract:The perennial ryegrass Lolium perenne can be used in conjunction with cadmium (Cd)-tolerant bacteria such as Cdq4-2 (Enterococcus spp.) for bioremediation of Cd-contaminated soil. In this study, a theoretical basis was provided to increase the efficiency of L. perenne remediation of Cd-contaminated soil using microorganisms to maintain the stability of the soil microbiome. The experimental design involved three treatment groups: CK (soil without Cd addition) as the control, 20 mg·kg<sup>-1</sup> Cd-contaminated soil, and 20 mg·kg<sup>-1</sup> Cd-contaminated soil + Cdq4-2, all planted with L. perenne. The soil was collected on day 60 to determine the soil microbial activity and bacterial community structure and to analyze the correlation between soil variables, the bacterial community, available Cd content in the soil, Cd accumulation, and L. perenne growth. The soil microbial activity and bacterial community diversity decreased under Cd stress, and the soil microbial community composition was changed; while inoculation with Cdq4-2 significantly increased soil basal respiration and the activities of urease, invertase, and fluorescein diacetate (FDA) hydrolase by 83.65%, 79.72%, 19.88%, and 96.15% respectively; and the stability of the community structure was also enhanced. The Actinobacteriota biomass, the amount of available Cd, and the above- and belowground Cd content of L. perenne were significantly negatively correlated with the total phosphorus, total potassium, and pH. The activity of urease, invertase, and FDA hydrolase were significantly positively correlated with the biomasses of Acidobacteriota and L. perenne and significantly negatively correlated with the Chloroflexi biomass. Further, the available soil Cd content and the above- and belowground Cd levels of L. perenne were significantly positively correlated with the Actinobacteriota biomass and significantly negatively correlated with the Gemmatimonadetes biomass. Overall, inoculating Cd-tolerant bacteria improved the microbial activity, diversity, and abundance, and changed the microbial community composition, facilitating the remediation of Cd-contaminated soil by L. perenne.

Effect of Liming and Organic Matter Application on Cd Uptake by Ryegrass in a Cadmium (Cd) Contaminated Soil and Its Influencing Factors

Abstract:One acidic Cd-contaminated soil was collected near a copper smelter to study the effect of liming and application of compost and peat on Cd uptake by ryegrass. The treatments consisted of three levels of liming, i.e. 2, 4 and 6 g·kg-1 soil, and two levels of organic matter, i.e. 2.5% and 5% in a complete random design. Results showed that the application of both lime and organic matter significantly improved the growth of ryegrass. Liming remarkably increased soil pH and decreased 0.05 mol·L-1 CaCl2 extractable soil available Cd, however, the Cd content in ryegrass showed no significant decrease and even an increase in root, thus resulting in an increased R/S ratio of the Cd content in ryegrass, suggesting that liming can partly affect the translocation of Cd from root to shoot. Organic matter application could decrease Cd uptake, and such an effect was further improved with the addition of liming. Compost had better effect than peat due to the acidic property of the later, so it was important to take the pH of the organic matter into consideration when applying different organic matters to remediate Cd contaminated soils. It was highly recommended to apply acidic organic matter together with lime. Under the current experimental condition, soil pH and 0.05 mol·L-1 CaCl2 extractable available Cd were not the good index to reflect the impact of liming and organic matter application on Cd uptake by ryegrass.
Combined Effect of Ryegrass and Hyphomicrobium Sp. GHH on the Remediation of EE2-Cd Co-Contaminated Soil

Abstract:Irrigation and fertilization accelerate the accumulation of environmental hormones such as 17α-Ethinyestradiol (EE2) and heavy metals such as Cd in soil, which in turn harms crops and human health. The objective of this study was to investigate the interaction of the plant ryegrass and EE2 degrading bacteria Hyphomicrobium sp. GHH on the remediation of EE2-Cd co-contaminated soil. The concentration of EE2 in soil was set as 25 mg kg−1, and the concentration of Cd was set as 0, 5, 20, and 80 mg kg−1, respectively. Pot experiments were carried out to investigate the control treatment without GHH inoculation and/or ryegrass cultivation (C), GHH inoculation alone (B), ryegrass cultivation alone (P), and combined use of ryegrass with GHH (B + P) on the EE2 and Cd removal from soil. The plant biomass, EE2 and Cd concentration in root and shoot of ryegrass, the bioconcentration factor (BCF), translocation factor (TF), phytoextraction efficiency of EE2 and Cd, and removal rate of EE2 and Cd from soil, as well as the soil urease activity and microbial biomass carbon (MBC) content, were determined to evaluate the interaction of plant and microbe on the soil remediation. After 28 days of treatment, in soil with spiked Cd at 5, 20, and 80 mg kg−1, the removal rate of EE2 from soil was: B + P > P > B > C; the effect on the Cd removal was B + P > P, while Cd cannot be removed by microbes directly. In B + P treatment, the removal rate was 89%, 80%, and 71%, respectively for EE2, and 0.81%, 0.43%, and 0.38% respectively for Cd. EE2 and Cd were mainly stored in root of ryegrass because that all the TFs were < 1. The inoculation of GHH significantly promoted plant growth and substantially increased the extraction efficiencies of EE2 and Cd by ryegrass (P < 0.05). Both cultivation of ryegrass and inoculation of GHH improved the soil environment, and the improving effect on the urease activity and MBC content was B + P > P > B > C. 5 mg kg−1 Cd increased soil urease activity and MBC content, and promoted the root growth. However, soil Cd at the concentration > 20 mg kg−1 caused irreparable harm to ryegrass and microorganisms; as the consequence, the EE2 extraction by plant significantly decreased (P < 0.05). The combined use of ryegrass with GHH was effective to remediate the EE2-Cd co-contaminated soil.

Abstract:Cadmium (Cd) pollution has been rapidly increasing due to the global rise in industries. Cd not only harms the ecological environment but also endangers human health through the food chain and drinking water. Therefore, the remediation of Cd-polluted soil is an imminent issue. In this work, ryegrass and a strain of Cd-tolerant bacterium were used to investigate the impact of inoculated bacteria on the physiology and biochemistry of ryegrass and the Cd enrichment of ryegrass in soil contaminated with different concentrations of Cd (4 and 20 mg/kg). The results showed that chlorophyll content increased by 24.7% and 41.0%, while peroxidase activity decreased by 56.7% and 3.9%. In addition, ascorbic acid content increased by 16.7% and 6.3%, whereas glutathione content decreased by 54.2% and 6.9%. The total Cd concentration in ryegrass increased by 21.5% and 10.3%, and the soil's residual Cd decreased by 86.0% and 44.1%. Thus, the inoculation of Cd-tolerant bacteria can improve the antioxidant stress ability of ryegrass in Cd-contaminated soil and change the soil's Cd form. As a result, the Cd enrichment in under-ground and above-ground parts of ryegrass, as well as the biomass of ryegrass, is increased, and the ability of ryegrass to remediate Cd-contaminated soil is significantly improved.

Abstract:Cadmium (Cd) pollution has led to a serious deterioration in soil quality, plant growth, and human health. Therefore, restoration of soil quality is imperative. Phytoremediation is inexpensive and yields acceptable outcomes. Phytoremediation involves interaction between plant physiology and microbial activity and has been widely used in the remediation of Cd-contaminated soil. In the present study, Lolium perenne L. (perennial ryegrass) was planted in Cd-spiked soil and indole-3-acetic acid (IAA) was used to explore the physiological and biochemical characteristics of ryegrass as well as soil enzyme activity to remove Cd. The present study provides a theoretical basis for the phytoremediation of Cd-contaminated soil. The study investigated the effect of 30-mg/kg Cd-spiked soil on ryegrass (C) and 30-mg/kg Cd-spiked soil on ryegrass treated with 10-mg/kg IAA (CI) compared with uncontaminated soil and ryegrass as the control. At the end of the experiment, the ryegrass biomass, total chlorophyll, superoxide dismutase (SOD) activity, and soil invertase activity in C group were decreased by 33.7%, 23.0%, 29.7%, and 18.3%, respectively, whereas the peroxidase (POD) activity and soil basal respiration increased by 17.1% and 87.9%, respectively, compared with the control. In the CI group, the biomass of ryegrass, chlorophyll content, SOD activity, sucrase activity, fluorescein diacetate (FDA) hydrolase activity, and Cd removal rates increased by 14.5%, 19.9%, 24.3%, 12.1%, 20.4%, and 15.1%, respectively, whereas the POD activity, soil basal respiration, and Cd residues in the soil declined by 8.0%, 15.0%, and 17.0%, respectively, compared with the C group. Therefore, exposure to exogenous IAA alleviated the Cd stress on ryegrass and soil microorganisms and improved Cd absorption by ryegrass from the contaminated soil.

Abstract:Cadmium (Cd) is a highly toxic and cumulative environmental pollutant. Siderophores are heavy metal chelators with high affinity to heavy metals, such as Cd. Ryegrass (Lolium perenne L.) has a potential remediation capacity for soils contaminated by heavy metals. Consequently, using ryegrass alongside beneficial soil microorganisms that produce siderophores may be an effective means to remediate soils contaminated with Cd. In this study, the Bacillus strains WL1210 and CD303, which were previously isolated from the rhizospheres of Nitraria tangutorum in Wulan and Peganum harmala L. in Dachaidan, Qinghai, China, respectively, both arid and sandy environments, were evaluated for heavy metal pollution mitigation. Our quantitative analyses have discerned that the two bacterial strains possess commendable attributes of phosphorus (P) solubilization and potassium (K) dissolution, coupled with the capacity to produce phytohormones. To assess the heavy metal stress resilience of these strains, they were subjected to a cadmium concentration gradient, revealing their incremental growth despite cadmium presence, indicative of a pronounced tolerance threshold. The subsequent phylogenetic analysis, bolstered by robust genomic data from conserved housekeeping genes, including 16S rDNA, gyr B gene sequencing, as well as dnaK and recA, delineated a species-level phylogenetic tree, thereby confirming the strains as Bacillus atrophaeus. Additionally, we identified the types of iron-carrier-producing strains as catechol (WL1210) and carboxylic acid ferrophilin (CD303). A genomic analysis uncovered functional genes in strain CD303 associated with plant growth and iron carrier biosynthesis, such as fnr and iscA. Ryegrass seed germination assays, alongside morphological and physiological evaluations under diverse heavy metal stress, underscored the strains' potential to enhance ryegrass growth under high cadmium stress when treated with bacterial suspensions. This insight probes the strains' utility in leveraging alpine microbial resources and promoting ryegrass proliferation.
Rhizospheric mechanisms of Bacillus subtilis bioaugmentation-assisted phytostabilization of cadmium-contaminated soil

Abstract:Plant growth promoting (PGP) traits of inoculation in bioaugmentation assisted phytostabilization of heavy metal-contaminated soil have been well documented. The property of inoculation to immobilize heavy metals is another major contributor to phytostabilization efficiency. This study investigated the effects of inoculation with different concentrations of rhizobacteria Bacillus subtilis on the cadmium (Cd) bioavailability and distribution, enzyme activities, and bacterial community structure in soil planted with ryegrass (Lolium multiflorum L.). Addition of a high dosage of Bacillus subtilis decreased plant malondialdehyde (MDA) amount, increased plant antioxidant enzyme and soil nutrient cycling-involved enzyme activities, and subsequently enhanced biomass by 20.9%. In particular, the inoculation reduced the Cd bioavailability in soil, bioaccumulation coefficient (BCF), translocation factors (TF), and accumulation in ryegrass by 39.1%, 36.5%, 24.2%, and 27.9%, respectively. Furthermore, 16S rRNA gene sequencing analysis of rhizosphere soil revealed microbial community structure alterations (e.g., enrichment of Proteobacteria), eight phenotype regulations, and seventeen Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway transformations accounted for the stress mitigation and Cd immobilization in the presence of inocula. Besides, intracellular accumulation and biofilm sequestration were proposed as primary immobilization mechanisms induced by bioaugmentation.

Combined Application of High-Throughput Sequencing and Metabolomics to Evaluate the Microbial Mechanisms of Plant-Growth-Promoting Bacteria in Enhancing the Remediation of Cd-Contaminated Soil by Hybrid Pennisetum

Abstract:The contamination of soil with the heavy metal cadmium (Cd) is increasingly prominent and severely threatens food security in China. Owing to its low cost, suitable efficacy, and ability to address the shortcomings of plant remediation by enhancing the ability of plants to take up Cd, plant–microbe combination remediation technology has become a research hotspot in heavy metal pollution remediation. A pot experiment was performed to examine the effects of inoculation with the plant-growth-promoting bacterium Brevibacillus sp. SR-9 on the biomass, Cd accumulation, and soil nutrients of hybrid Pennisetum. The purpose of this study was to determine how Brevibacillus sp. SR-9 alleviates stress caused by heavy metal contamination. High-throughput sequencing and metabolomics were used to determine the effects of inoculation on the soil bacterial community composition and microbial metabolic functions associated with hybrid Pennisetum. The results suggest that mutation of Brevibacillus sp. SR-9 effectively alleviates Cd pollution stress, leading to increased biomass and accumulation of Cd in hybrid Pennisetum. The aboveground biomass and the root weight increased by 12.08% and 27.03%, respectively. Additionally, the accumulation of Cd in the aboveground sections and roots increased by 21.16% and 15.50%, respectively. Measurements of the physicochemical properties of the soil revealed that the strain Brevibacillus sp. SR-9 slightly increased the levels of available phosphorus, total nitrogen, total phosphorus, and available potassium. High-throughput DNA sequencing revealed that Brevibacillus sp. SR-9 implantation modified the composition of the soil bacterial community by increasing the average number of Actinobacteria and Bacillus. The total nitrogen content of the soil was positively correlated with the Actinobacteria abundance, total phosphorus level, and available phosphorus level. Metabolomic analysis revealed that inoculation affected the abundance of soil metabolites, and 59 differentially abundant metabolites were identified (p < 0.05). Among these, 14 metabolites presented increased abundance, whereas 45 metabolites presented decreased abundance. Fourteen metabolic pathways were enriched in these metabolites: the folate resistance pathway, the ABC transporter pathway, D-glutamine and D-glutamic acid metabolism, purine metabolism, and pyrimidine metabolism. The abundance of the metabolites was positively correlated with the levels of available phosphorus, total potassium, total phosphorus, and total nitrogen. According to correlation analyses, the development of hybrid Pennisetum and the accumulation of Cd are strongly associated with differentially abundant metabolites, which also impact the abundance of certain bacterial populations. This work revealed that by altering the makeup of microbial communities and their metabolic processes, bacteria that promote plant development can mitigate the stress caused by Cd. These findings reveal the microbiological mechanisms through which these bacteria increase the ability of hybrid Pennisetum to take up the Cd present in contaminated soils.

Unraveling the effects of arbuscular mycorrhizal fungi on cadmium uptake and detoxification mechanisms in perennial ryegrass (Lolium perenne)

Abstract:Cadmium (Cd) is a major environmental pollutant and one of the most toxic metals in the environment. Arbuscular mycorrhizal fungi (AMF) assisted phytoremediation can be used to remove Cd from polluted soils but the role of AMF, which mediate in Cd accumulation and tolerance, remains poorly understood.Here we inoculated Lolium perenne with two different AMF species (Glomus etunicatum and Glomus mosseae). Mycorrhizal L. perenne and non-mycorrhizal controls were exposed to Cd stress and we tested the effects of AMF mycorrhization on Cd uptake and subsequent tolerance, as well as the underlying mechanisms.Mycorrhizal infection increased root Cd2+ uptake and we observed that net Cd2+ influx was coupled with net Ca2+ influx. The inactivation of Ca2+ transporter channels decreased Cd2+ uptake in non-inoculated roots to a greater extent than in inoculated roots, indicating that AMF activates additional ion transport channels. In consequence, inoculated plants exhibited higher Cd accumulation in both roots and shoots than non-inoculated controls. However, AMF-inoculated plants showed higher chlorophyll concentrations, photosynthesis, and growth under Cd, indicating lower Cd toxicity in AMF-inoculated plants, despite the increase in Cd uptake. We observed that AMF-inoculated favored the isolation of Cd within cell walls and vacuoles, and had higher concentrations of superoxide dismutase activity and glutathione concentration in roots than non-inoculated plants, consequently experiencing less stress upon Cd exposure.Our results highlight the potential and mechanism of AMF for enhancing phytoremediation of L. perenne in heavy metal contaminated environments.

Enhancing the effect of novel cd mobilization bacteria on phytoremediation and microecology of cadmium contaminated soil

Abstract:The efficacy of phytoextraction for remediating heavy-metal contaminated soil depends on the bioavailability of the heavy metals and plant growth. In this study, we employed a synergistic system comprising water-soluble chitosan and the novel Cd mobilization bacteria, Serratia sp . K6 (hereafter K6), to enhance cadmium (Cd) extraction by Lolium perenne L. (ryegrass). The application of chitosan and K6 resulted in an increase in the biomass of ryegrass by 11.81% and Cd accumulation by 73.99% and effective-state Cd by 43.69% and pH decreased by 4.67%, compared to the control group. Microbiome and metabolomics analyses revealed significant alterations in the inter-root microbial ommunity, with rhizobacteria such as Sphingomonas , Nocardioides , and Bacillus likely contributing to enhanced plant growth and Cd accumulation in response to chitosan and K6 addition. Additionally, the contents of various organic acids, amino acids, lipids, and other metabolites exhibited significant changes under different additive treatments, suggesting that ryegrass can regulate its own metabolites to resist Cd stress. This study provides valuable insights into the effects of additives on phytoextraction efficiency and the soil bacterial community, offering a promising approach for phytoremediation of Cd-contaminated soils. Our aim was to uncover the mechanisms by which chitosan affects ryegrass growth and its uptake of HMs in the presence of added exogenous bacterial agents. The findings of our study contribute to a deeper understanding of the interaction mechanisms between exogenous microorganisms and chitosan, thereby improving extraction efficiency in soil-plant systems.

Effects of intercropping ryegrass with hollyhock and inoculation with Bacillus thuringiensis on enhancing phytoremediation of Cd-contaminated soils

Abstract:Cd is a heavy metal that contaminates soils. These kinds of heavy metals pose a serious threat to food security, ecosystems, and human health. To improve the phytoremediation efficiency of moderately Cd-contaminated cropland soils and achieve simultaneous production and remediation, intercropping ryegrass (Lolium perenne L.) with hollyhock (Althaea rosea) was investigated using pot experiments, and Bacillus thuringiensis (B. thuringiensis) inoculation was used as a booster to strengthen the absorption and accumulation of Cd in plants. The results showed that intercropping (Int treatment) decreased the Cd concentration in plants compared to hollyhock and ryegrass monocropping. However, the Cd accumulation in ryegrass and hollyhock was promoted by B. thuringiensis addition to intercropping (Int-B treatment), as the biomass of ryegrass and hollyhock was 2.33 and 1.13 times that of the Int treatment, respectively. Compared with the Int treatment, the total Cd concentration in soils of the Int-B treatment decreased by 8.1%, while diethylenetriaminepentaacetic acid solution extracted Cd (DTPA-Cd) increased by 18.2%, indicating that B. thuringiensis increases the available Cd concentration in soils to promote Cd adsorption by hollyhock enrichment plants. High-throughput sequencing results further revealed that the dominant microflora in the soils of the Int and Int-B treatments were consistent with the control, although their abundance and diversity decreased slightly. Overall, intercropping with B. thuringiensis addition effectively increased the hollyhock remediation efficiency in moderately Cd-contaminated soils, and the concentration of Cd in forage crops of ryegrass was lower than the limit value of “Hygienic standards for feeds’ (GB 13078–2017) in China.

Abstract:The green and efficient remediation of soil cadmium (Cd) is an urgent task, and plant-microbial joint remediation has become a research hotspot due to its advantages. VY-1 effectively mitigated Cd stress on Hybrid pennisetum, resulting in increased plant biomass, Cd accumulation, and translocation factor, thereby enhancing phytoremediation efficiency. Analysis of soil physical-chemical properties revealed that strain VY-1 could increase soil total nitrogen, total phosphorus, available phosphorus, and available potassium content. Principal coordinate analysis (PCoA) indicated that strain VY-1 significantly influenced bacterial community composition, with Proteobacteria, Firmicutes, Chloroflexi, among others, being the main differential taxa. Redundancy analysis (RDA) revealed that available phosphorus, available potassium, and pH were the primary factors affecting bacterial communities. Partial Least Squares Discriminant Analysis (PLS-DA) demonstrated that strain VY-1 modulated the metabolite profile of Hybrid pennisetum rhizosphere soil, with 27 differential metabolites showing significant differences, including 19 up-regulated and eight down-regulated expressions. These differentially expressed metabolites were primarily involved in metabolism and environmental information processing, encompassing pathways such as glutamine and glutamate metabolism, α-linolenic acid metabolism, pyrimidine metabolism, and purine metabolism. This study utilized 16S rRNA high-throughput sequencing and metabolomics technology to investigate the impact of the plant growth-promoting bacterium Enterobacter sp. VY-1 on the growth and Cd enrichment of Hybrid pennisetum, providing insights into the regulatory role of plant growth-promoting bacteria in microbial community structure and metabolic function, thereby improving the microbiological mechanisms of phytoremediation.

Abstract:In recent years, heavy metal pollution has become a global environmental problem and poses a great threat to the health of people and ecosystems. Therefore, strategies for the effective remediation of Cd from contaminated soil are urgently needed. In this study, ryegrass was utilized as a remediation plant, and its remediation potential was enhanced through the application of Citric Acid (CA) in conjunction with Bacillus megaterium (B. megaterium). The P3 treatment (CA + Bacillus megaterium) exhibited a significantly higher efficiency in promoting cadmium extraction by ryegrass, resulting in a 1.79-fold increase in shoot cadmium accumulation compared to the control group (CK) with no Bacillus megaterium or CA. Moreover, the P3 treatment led to an increased abundance of Actinobacteriota , Acidobacteriota , and Patescibacteria in the rhizosphere. The concentration of amino derivatives (such as betaine, sulfolithocholylglycine, N-alpha-acetyl-lysine, glycocholic acid, arginyl-threonine) showed significant upregulation following the P3 treatment. In summary, this study proposes a viable approach for phytoremediation of soil contaminated with cadmium by harnessing the mobilizing abilities of soil bacteria. Our aim was to gain a comprehensive understanding of the mechanisms involved in phytoremediation. These findings contribute to the existing knowledge by providing insights into the mechanism of phytoremediation in Cd-contaminated soil. They are expected to serve as a theoretical foundation for further elucidation of the phytoremediation mechanisms employed in Cd-contaminated soil.

Abstract:The objective of this study was to investigate combined effects of Cd and elevated CO(2) on growth, physiological and physiochemical characteristics, elemental compositions in Lolium mutiforum and Lolium perenne grown in soils amended with three Cd concentrations (0, 25, 100 mg kg(-1)) under two CO(2) levels (375, 810 microLL(-1)). Elevated CO(2) increased net assimilation rate and internal CO(2) concentration, and consequently increased total plant biomass by 51 to 31%. At same spiked Cd level, malondialdehyde content in leaves was lower under elevated than under ambient CO(2), whereas superoxide dismutase activity was higher. Elevated CO(2) decreased Cd, S, and phytochelatin concentrations in roots and shoots to a various degree, depending on plant species and element, but the PC-Cd ratio was not affected. It was concluded that elevated CO(2) ameliorated Cd toxicity in both Lolium species under Cd stress, and that the increase of plant biomass and the alleviation of Cd toxicity with elevated CO(2) for the Lolium species may be more dependent on increased photosynthesis and enhanced antioxidant capacity. Results of the study may provide insights into the interaction between soil Cd contamination and atmospheric CO(2) concentration with regard to plant ability to grow and remove the Cd from soils.

Abstract:Cadmium (Cd) pollution is becoming increasingly prevalent, posing a global environmental hazard due to its negative effects on plants growth and human health. Phytoremediation is a green technology that involves uptake of Cd from the soil by a combination of plants and associated microbes. The objective of this study was to investigate the role of Aspergillus aculeatus in perennial ryegrass Cd tolerance. This fungus produced indole-3-acetic acid, siderophores, and 1-aminocyclopropane-1-carboxylate deaminase. Physiological traits including growth rate, turf quality and chlorophyll content were measured to evaluate the physiological responses of perennial ryegrass to Cd stress. These physiological traits were improved after inoculated with A. aculeatus. Inoculation of A. aculeatus actively reduced DTPA-Cd concentration in the soil and Cd translocation to plant shoots. Chlorophyll a fluorescence transient and the C/N ratio in shoots were elevated by A. aculeatus, which implied that the fungus could protect the photosystem II against Cd stress and increase the photosynthetic efficiency. These results suggested that A. aculeatus is beneficial in improving Cd tolerance of perennial ryegrass and reducing Cd-induced injuries, thus, it has promising potential for application of phytostabilization in Cd contaminated soil.

Abstract:Abstract The chlorophyll, malondialdehyde and relative conductivity of Lolium perenne had different degrees of physiological response to Cd stress. Lolium perenne had certain physiological adaptability to Cd stress, which could reduce the damage of Cd to Lolium perenne cells. Low concentration of Cd stress could also promote the synthesis of chlorophyll and increase the photosynthetic products. Once the concentration of Cd was too high, the growth of Lolium perenne would be affected. The accumulation of Cd in plants exceeds a certain limit, which damages the physiological function of plants and affects the normal growth of plants. It is suggested that Lolium perenne has a certain remediation ability to cadmium contaminated soil. The experiment can provide a certain reference for the screening of cadmium tolerant plants and the remediation of cadmium contaminated soil.

Abstract:Cadmium (Cd) pollution has attracted global attention because it not only jeopardizes soil microbial ecology and crop production, but also threatens human health. As of now, microbe-assisted phytoremediation has proven to be a promising approach for the revegetation of Cd-contaminated soil. Therefore, it is important to find such tolerant microorganisms. In the present study, we inoculated a bacteria strain tolerant to Cd, Cdb8-1, to Cd-contaminated soils and then explored the effects of Cdb8-1 inoculation on the performance of the Chinese milk vetch. The results showed plant height, root length, and fresh and dry weight of Chinese milk vetch grown in Cdb8-1-inoculated soils increased compared to the non-inoculated control group. The inoculation of Cd-contaminated soils with Cdb8-1 also enhanced their antioxidant defense system and decreased the H2O2 and malondialdehyde (MDA) contents, which alleviated the phytotoxicity of Cd. The inoculation of Cdb8-1 in Cd-contaminated soils attenuated the contents of total and available Cd in the soil and augmented the BCF and TF of Chinese milk vetch, indicating that the combined application of Cd-tolerant bacteria Cdb8-1 and Chinese milk vetch is a potential solution to Cd-contaminated soils.

Abstract:Combined bioaugmentation inoculants composed of two or more plant growth-promoting bacteria (PGPB) were more effective than single inoculants for plant growth and cadmium (Cd) removal in contaminated soils. However, the principles of consortia construction still need to be discovered. Here, a pot experiment with Cd natural polluted soil was conducted and PGPB consortia with different ecological niches from hyperaccumulator Sedum alfredii Hance were used to compare their effects and mechanisms on plant growth condition, Cd phytoextraction efficiency, soil enzymatic activities, and rhizospheric bacterial community of Brassica juncea L. The results showed that both rhizospheric and endophytic PGPB consortia inoculants promoted plant growth (6.9%- 22.1%), facilitated Cd uptake (230.0%-350.0%) of oilseed rape, increased Cd phytoextraction efficiency (343.0%-441.0%), and enhanced soil Cd removal rates (92.0%-144.0%). PGPB consortia inoculants also enhanced soil microbial carbon by 22.2%-50.5%, activated the activities of soil urease and sucrase by 74.7%158.4% and 8.4%-61.3%, respectively. Simultaneously, PGPB consortia inoculants increased the relative abundance of Flavobacterium, Rhodanobacter, Kosakonia, Pseudomonas and Paraburkholderia at the genus level, which may be beneficial to plant growth promotion and bacterial phytopathogen biocontrol. Although the four PGPB consortia inoculants promoted oilseed growth, amplified Cd phytoextraction, and changed bacterial community structure in rhizosphere soil, their original ecological niches were not a decisive factor for the efficiency of PGPB consortia. therefore, the results enriched the present knowledge regarding the significant roles of PGPB consortia as bioaugmentation agents and preliminarily explored construction principles of effective bioaugmentation inoculants, which will provide insights into the microbial responses to combined inoculation in the Cd-contaminated soils.

Abstract:Contamination with cadmium poses a serious problem in paddy soils because it is a significant health hazard through soil-food chains transport. The stabilization of Cd in soil and reduction of Cd in rice plants have been investigated in some reports, but the effects on soil microorganisms, especially the comparison of microbial species and composition between contaminated soil and uncontaminated soil after restorationÂ has not been elucidated. Hence, our study aims to develop FeMg-LDH@Bentonite mixed with compost as an effective stabilizer for Cd mobilization in paddy soil, with the assistance of the community bureau of reference (BCR) sequential extraction procedure as the remediation efficiency evaluation. Such a material introduction induces the change in several factors including the soil pH, water-soluble organic carbon (WSOC), enzyme activity, soil microbial community abundance and specific microbial community composition, finally affecting the solubility and speciation of Cd. Besides, Cd availability in soil, Cd concentration in rice grains, and restoration of soil microbial community diversity and composition are considered comprehensively. This study will provide information on efficient approaches to controlling Cd bioavailability in Cd-contaminated paddy soils.
