Abstract:Background Soil and understory vegetation are vital components of forest ecosystems. Identifying the interaction of plantation management to vegetation and soil is crucial for developing sustainable plantation ecosystem management strategies. As one of the main measures of close-to-nature management of forest plantation, few studies have paid attention to the effect of crop tree management on the soil properties and understory vegetation. Methods A 36-year-old Pinus massoniana plantation in Huaying city, Sichuan Province was taken as the research object to analyse the changes in undergrowth plant diversity and soil physicochemical properties under three different crop tree densities (100, 150, and 200 N/ha). Results Our results showed that the contents of available phosphorus, organic matter and hydrolysable nitrogen in the topsoil increased significantly after crop tree management, while content of available potassium decreased. The composition of shrub and herb layer was richer, and the dominant species were obviously replaced after crop tree management. The Shannon–Wiener index and Richness index of shrub layer, and the diversity of herb layer increased significantly after crop tree management. Herb layer diversity indexes and Richness index of shrub layer were closely related to soil organic matter, available phosphorus, hydrolysable nitrogen, available potassium, soil moisture and bulk density. As the main limiting factors for plant growth, nitrogen, phosphorus and potassium were closely related to plant diversity and to the distribution of the dominant species. At the initial stage of crop tree management, each treatment significantly improved the soil physicochemical properties and plant diversity of Pinus massoniana plantation, and the comprehensive evaluation was 200 N/ha >100 N/ha >150 N/ha >CK. Compared with other treatments, 200 N/ha had the best effect on improving the undergrowth environment of the Pinus massoniana plantation in the initial stage of crop tree management.

The effect of phosphorus addition, soil moisture, and plant type on soil nematode abundance and community composition

Abstract:PurposeEnvironmental variables such as soil moisture and phosphorus (P) might influence above- and below-ground biodiversity. In this study, we investigated the rarely reported individual and interactive multifactor effects of soil moisture and phosphorus addition with the type of above-ground tree species (biological interactions) on the soil nematode community structure.Materials and methodsWe established a completely randomized experimental design with two plant types (N2-fixer and non-nitrogen fixer) and different combinations of water treatments and P additions (i.e., water with P addition, water only, drought with P addition, and drought only) in a greenhouse and investigated their effects on the soil chemical properties and nematode community. Soil samples were collected at the end of the experiment and were analyzed for soil moisture content (SM), available phosphorus (aP), nitrate nitrogen (NO3−–N), ammonium nitrogen (NH4+–N), dissolved organic carbon (DOC), dissolved organic nitrogen (DON), and nematode community. The following trophic groups were assigned to the nematodes: bacterivores (Ba), fungivores (Fu), omnivores–predators (Op), and plant parasites (PP). The channel index (CI), enrichment index (EI), maturity index (MI), genus richness (GR), and Simpson dominance (Ig) were adopted to indicate the indices of the nematode food web.Results and discussionPhosphorus addition and its interaction with water treatments had no statistically significant effects on the soil nematode community, but there were significant decreasing (p < 0.05) effects of P addition on the total density of nematodes of the N2-fixing tree under optimum water treatment. There were no significant interactive effects of P addition and water treatments on all the trophic groups, but plant type, water treatments, and their interactions significantly affected the density of most nematode trophic groups. The total nematode abundances of bacterivores, plant parasitic, omnivores, and enrichment index were significantly higher in the N2-fixers than in the non-nitrogen-fixing tree.ConclusionsSoil nematode abundance and community composition were more affected by the plant type than by the P addition and its interaction with water treatments. Drought exerted adverse effects on the total density of soil nematodes, the dominant genera, and the trophic groups. This study demonstrated that the rate of drought impact hinges more on the type of tree and that N2-fixing tree could still maintain the soil food web structure irrespective of the environmental changes.

Abstract:Introduction: Management practices are essential for maintaining forest ecological functions under increasing diseases and pest disasters. The effects of nitrogen fertilization (NF) and clear-cutting (CC) on the soil microbial community structure and greenhouse gases emission were investigated of pinewood nematodes (Bursaphelenchus xylophilus)-infected Masson pine (Pinus massoniana) plantations. Outcomes: CC increased the soil microbial biomass carbon (SMBC) and soil microbial biomass nitrogen (SMBN) contents relative to the control (CK). NF increased the SMBN but had no significant effect on the SMBC content. The total fungal and bacterial abundances increased in the CC treatment compared with the CK, but there was no significant difference between the NF and CK. The cumulative soil CO2 emission (2.35 t C·hm−2) was higher than that of CK (1.65 t C·hm−2) in summer, and the cumulative annual N2O emission (16.90 kg N·hm−2 yr−1) of NF was approximately 47 times of CK (0.36 t N·hm−2·yr−1). CC increased the CO2 flux (2.21 t C·hm−2) in summer but have no significantly effects on N2O emission. Conclusion: These results indicated that NF and CC practices changed forest soil microbial community structure and affected soil greenhouse gas emissions in pinewood nematodes -infected Masson pine plantations. The CO2 emission rates increased in the NF and CC treatments, which reduced the carbon sequestration function of forests and had a negative impact on climate change.

The Distribution pattern of carbon,nitrogen and phosphorus and the stoichiometry characteristics of Pinus massoniana plantation in different ages

Abstract:In order to understand the distribution pattern of carbon、nitrogen and phosphorus and the stoichiometry characteristics of pinus massoniana plantation ecosystem at hilly area in upper reaches of the Yangtze River, we used the space-temporal exchanging method, and selected three different forest stand age(5-year-old young growth, 14-year-old half-mature forest, 39-year-old mature forest) of masson pine plantation in similar site conditions as the objects, and analyzed the concentration of carbon, nitrogen and phosphorus and w(C):w(N):w(P) stoichiometry characteristics in the needle leaf, litter and soil. The results showed that the concentration of C, N and P were significant difference between needle leaf, litter and soil which in order of needle leaf litter soil; forest age had significant impact on C, N, P concentrations and w(C):w(N), w(C):w(P) stoichiometry ratios; The C, N, P content of soil were the highest in the mature forest, while the C content of needle leaf and litter were lowest in mature forest, and the N, P content of needle leaf and litter was highest in the mid-maturation forest; N and P use efficiency of masson pine plantation reduced, as w(C):w(N) and w(C):w(P) ratio of needle leaf, litter and soil declined with forest age increasing; The w(N):w(P) ratio of needle leaf was between 14.37 and 15.53, which indicated that the masson pine plantation was restricted by N and P together, but the influence of N and P limitation was not significantly by forest age. To increase the productivity of pinus massoniana plantation in the area, we should appropriately increase N and P fertilization in the plantation tending management. In addition, we also could interplant the leguminous nitrogen-fixing plants to improve soil fertility. This study combined masson pine needles, litter and soil to explore the distribution pattern of C, N and P nutrient elements and the change of chemical measurement characteristics with forest age increasing, will help to reveal the nutrient cycling of pinus massoniana plantation ecosystem roundly and systematically, it is very significant to guide the production of pinus massoniana plantation, adjust and improve the growth environment of masson pine, improve the nutrient use efficiency and the forest land productivity of the system.

Abstract:In forests, microbial populations in the soil can directly influence the decomposition of carbon from surface plants, promoting carbon storage and stability. However, in sustainable forest management, it is still unclear how soil microorganisms under different plantation types affect organic carbon sequestration and whether the mechanisms of influence are the same. In this research, we focused on four mixed forests and pure Pinus massoniana-planted forest in the state-owned forest farm of Dushan County. Three replicated plots were set up for each model, and soil samples were collected from different layers (0–20 cm, 20–40 cm, and 40–60 cm), totaling 45 samples. We elucidated the effects of soil microorganisms on carbon sequestration under five mixed modification models of P. massoniana and further explored the mechanisms by which microbial functional communities regulate soil carbon sequestration under different mixed models through molecular sequencing and collinear network analysis. Variance analysis indicated that the soil organic carbon (SOC) of the same soil layer varied significantly, and there were also significant differences in the composition of soil bacterial and fungal microbial communities. Moreover, the bacterial community was more sensitive to changes in the vegetation environment, while the fungal community structure was more resistant to changes in the soil environment. Correlation analysis indicated that the diversity and composition of the bacterial community had more positive effects on soil organic carbon than those of the fungal community. Linear fitting and redundancy analysis (RDA) showed that particulate organic carbon (POC) in soil had the strongest correlation with SOC content. Soil microorganisms affected the storage and stability of soil carbon mainly by regulating the conversion of litter (carbon sources) into POC. The soil environment of different mixed models had different effects on soil carbon accumulation. Both correlation and collinearity network analyses indicated that soil microbial functional groups could enhance carbon storage by regulating readily oxidizable carbon (EOC) and POC content in mixed forest plantations. The results of our study provide a sound basis for replanting a reasonable forest model structure to improve forest carbon storage.

Abstract:Pinus massoniana is the major afforestation and vegetation restoration tree in southern China, and it plays an important role in the sustainable development of plantations. However, long-term single planting of P. massoniana has resulted in the decline of soil quality and forest productivity, and a soil fertility assessment is urgently needed. We selected P. massoniana plantations of four age stages for plot investigation and sampling to determine the soil physicochemical properties, microbial diversity and composition, and enzyme activities at different soil depths. The results showed that soil total phosphorus (TP) and available phosphorus (AP) decreased with the increase of age, especially low C/N ratio and high C/P and N/P ratio in the 30-year and 36-year stands, leading to P limitation. Meanwhile, the bacterial Shannon index also decreased with the increase of age and was positively correlated with AP, NO3−-N, and pH. However, the fungal Shannon index decreased first and then increased with the increase of age; soil acid phosphatase (S-ACP) and urease activities showed a similar trend. Correlation analysis demonstrated that the increase of total organic carbon (TOC) and total nitrogen (TN) promoted the increase of fungal Shannon index, which was beneficial to the secretion of more enzymes. We found that soil physicochemical properties, microbial diversity, and enzyme activity decreased simultaneously when soil depths increased. Moreover, Acidobacteria and Basidiomycota were the most abundant bacterial and fungal communities, respectively, followed by Proteobacteria and Actinobacteria for bacteria and Ascomycota for fungi, and these microbial taxa were significantly affected by soil water content (SWC), TOC, AP, and C/P. In conclusion, this work reveals the potential correlation among soil physicochemical properties, microbial diversity and composition, and enzyme activities, and revealed potential correlations among them which will help to improve understanding of soil conditions and provide a reference for rational management of soil resources.

Abstract:To address the ecological challenges arising from pure forest plantations and the wood supply–demand imbalance, implementing sustainable forest management is paramount. Accordingly, we studied crop trees at three densities (100, 150, and 200 N/ha) in a subtropical Pinus massoniana plantation. Our study revealed that the dominant phyla and genera within the fungal community remained largely consistent, with Basidiomycota and Ascomycota occupying prominent positions. Notably, the β diversity of the fungal community exhibited significant changes. Ectomycorrhizal and saprophytic fungi emerged as crucial functional guilds, and crop-tree thinning contributed to increased complexity within the fungal network, with a prevalence of positive rather than negative correlations among genera. The significant roles played by Camphor plants and ferns were evident in the fungal networks. Additionally, under crop-tree thinning, plant diversity experienced a significant boost, fostering interactions with the fungal community. Herb diversity played a vital role in the fungal community, affecting it either directly or indirectly, by altering the content of total phosphorus or organic matter in the soil. This study underscores the relationship between undergrowth plants and soil fungal communities, offering a scientific basis for evaluating the sustainability of restoring inefficient forest-plantation ecosystems.

Abstract:Declining soil quality and microecological imbalances were evaluated in larch plantations in this study. One potential solution to this problem is the cultivation of mixed coniferous and broad-leaved plantations. However, it is unclear whether and how soil microbial community structure and nutrient cycling function would be affected by mixed plantations and soil depths. In this study, we used high-throughput sequencing technology to investigate bacterial 16S and fungal ITS regions for comparisons of soil microbial diversity among plantation types (a Larix gmelinii pure plantation, a Fraxinus mandshurica pure plantation, a Larix – Fraxinus mixed plantation within the Larix row, the Fraxinus row, and between the Larix and Fraxinus rows) and soil depths (0–10, 10–20, and 20–40 cm). These data were used to evaluate variations in microbial communities and nutrient cycling function with the determining environmental factors. Our results indicated that bacteria had a stronger spatial dependence than did fungi, while plantation types significantly affected the fungal community. The relative abundance of Gaiellaceae , as well as bacterial ligninolysis, nitrate ammonification, and nitrite ammonification functions significantly increased with increasing soil depth. Compared with other plantations, the relative abundance of Inocybaceae was significantly higher in the Larix plantation. Distance-based redundancy analysis (db-RDA) showed that Gaiellaceae and Inocybaceae abundances were positively correlated with ammonium nitrogen content, available phosphorus content, and phosphatase activity. Our findings indicate that variations in soil available phosphorus are closely related to the relative abundances of Gaiellaceae at different soil depths and Inocybaceae in different plantation types. Mixed plantations might change the availability of soil phosphorus by controlling the relative abundance of Inocybaceae . We recommend that fungal community changes be considered in the sustainable management of mixed plantations.

Abstract:Soil microbes play key roles that support forest ecosystem functioning, while their community characteristics are strongly determined by tree species identity. However, the majority studies primarily focus on soil microorganisms in the topsoil, resulting in limited understanding of the linkages between tree species identity and the microbial communities that inhabit deep soils. Here we investigated the diversity, structure, function, and co-occurrence networks of soil bacterial and fungal communities, as well as related soil physicochemical properties, to a depth of two meters in dryland forests dominated by either Pinus tabuliformis, a native coniferous species, Robinia pseudoacacia, an exotic broadleaf and nitrogen-fixing species, or both. Tree species identity had stronger effects on soil multifunctionality and microbial community structure in the deep layers (80-200 cm) than in the top layers (0-60 cm). In addition, fungal communities were more responsive to tree species identity, whereas bacteria were more sensitive to soil depth. Tree species identity strongly influenced microbial network stability and complexity, with higher quantities in R. pseudoacacia than the other plantations, by affecting microbial composition and their associations. The increased in microbial network complexity and the relative abundance of keystone taxa enhance the soil multifunctionality of microbial productivity, sugar and chitin degradation, and nutrient availability and cycling. Meanwhile, the relative abundance of keystone taxa was more representative of soil multifunctionality than microbial diversity. Our study highlights that tree species identity significantly influences soil microbial community characteristics and multifunctionality, especially in deep soils, which will help us understand soil nutrients processed in plantation forest ecosystem and provide a reference for tree species selection in ecological restoration.
Responses of Plant Species Diversity and Biomass to Forest Management Practices after Pine Wilt Disease

Abstract:Pine wilt disease (PWD), which is caused by the Bursaphelenchus xylophilus, is one of the most serious biological invasions in East Asia. Removal of infected pine trees is generally used to prevent the spread of PWD. However, how this strategy affects plant species diversity and ecosystem functions needs to be clarified. We compared alpha diversity, species composition, and biomass for all woody plant species, adults, saplings, and seedlings in infected Masson pine forests where removal of infected trees occurred (CTD) and where infected trees were retained (UTD), uninfected Masson pine forests (PMF), and evergreen broad-leaved forests (EBF). UTD had the highest alpha diversity of all species and saplings, and UTD and PMF had the lowest alpha diversity of seedlings. CTD and EBF had a similar composition of seedlings, and CTD and UTD had similar species composition of all plant species and saplings. UTD had the lowest biomass for all species and adults but had the highest saplings biomass. Soil properties were significantly related to plant biomass. The retention of infected trees likely maintained soil fertility which increased species alpha diversity and the biomass of saplings, and unchanged species composition compared to the removal of infected trees, indicating that the retention strategy could help to maintain ecosystem functions.
Nitrogen and phosphorous dynamics with stand development of Pinus massoniana plantations in Southeast China

Abstract:Introduction: Nutrient resorption is a key mechanism to conserve nutrients and overcome nutrient limitation in perennial plants. As an important afforested tree species in subtropical regions, Pinus massoniana grows well in nutrient-poor environments, however, the age-related pattern of nutrient acquisition strategy and the underlying mechanisms in P. massoniana plantations remain unclear. Methods: In this study, concentrations of nitrogen (N) and phosphorus (P) were measured in green and senesced needles, roots and soil samples collected from P. massoniana plantations with different stand ages (9-, 17-, 26-, 34- and 43-year-old) in south China. From these samples, nutrient resorption efficiency (RE) and stoichiometry were calculated. Results: Needle PRE significantly decreased with stand age, while there was no clear pattern of NRE along the stand development. Green needle N:P in older stands was significantly lower than in younger ones. Senesced needle C:P and N:P significantly decreased with stand age. Root and soil available P concentrations were significantly higher in older stands than in younger ones, and PRE was negatively correlated with soil available P concentration. Discussion: There was a shift from "conservative consumption" to "resource spending" P-use strategy, and P limitation decreased with stand development of P. massoniana plantations. The results provide information of changes in nutrients dynamics, which is relevant for the sustainable management of subtropical forest plantations.

Abstract:Tree species composition in forest ecosystems is an important biological factor affecting the diversity of ectomycorrhizal fungi (EMF). However, little is known about the composition and diversity of EMF communities associated with Pinus massoniana in different P. massoniana association habitats (MpAHs) in subtropical mountains. This study investigated the EMF community characteristics of P. massoniana in different MpAHs using plant community surveys, soil property analyses, and mycorrhizal identification. A total of 56 operational taxonomic units (OTUs), belonging to 20 families and 22 genera, were identified. OTU richness of Basidiomycota (58.93%) was higher than that of Ascomycota (41.07%). Unclassified Helotiales, Russula, Lactarius, and Tomentella were the dominant groups. Different stand types significantly altered the EMF communities of P. massoniana (p < 0.05, for Shannon index) and the associations of P. massoniana + Populus adenopoda (Mp_Pa) had the highest diversity of EMF, while P. massoniana + Cunninghamia lanceolata (Mp_Cl) had the lowest diversity. The number of specific OTUs was higher than shared OTUs. Similarity index and principal coordinate analysis indicated that the EMF communities of P. massoniana varied significantly in different MpAHs (R2 = 0.21, p = 0.001). The linear regression model showed that the EMF diversity of P. massoniana was positively related to tree species diversity, indicating that the EMF diversity of P. massoniana is influenced by tree species diversity. The findings provide a reasonable reference for tree species configuration in the process of mixed transformation or near-natural management of plantations.

[Effects of undergrowth vegetation management measures on the soil bacterial community structure of large diameter timber plantation of Cunninghamia lanceolata]

Abstract:Given the importance of undergrowth vegetation to plantation ecosystem, this study analyzed the effects of three kinds of understory management measures, including understory preservation, understory removal, and interplanting, on the soil bacterial diversity, community structure and relative abundance under large diameter timber plantation of Cunninghamia lanceolata using high-throughput sequencing technology. The relationship between soil physical and chemical properties and bacterial community diversity were analyzed. The results showed that Chao1, Ace and Shannon indices of soil bacterial communities of understory preservation were higher than those of understory removal and interplanting. Actinobacteria, Acidobacteria and Chloroflexi were the dominant bacteria groups in the soil of C. lanceolata plantation. Compared with understory removal and interplanting, the relative abundance of Proteobacteria, Planctomycetes, Firmicutes and Verrucomicrobia in the soil of understory preservation was relatively high, while that of Actinobacteria, Acidobacteria and Chloroflexi was relatively low. There were significant differences in the relative abundance of Firmicutes, Planctomycetes, Verrucomicrobia, Parcubacteria and Actinobacteria among three understory management measures. The contents of moisture, total nitrogen, total phosphorus, hydrolyzed nitrogen and available phosphorus in the soil were important factors affecting soil bacterial community structure. Soil bacterial diversity indices had significant positive correlation with the contents of total nitrogen, total phosphorus, total potassium, hydrolyzed nitrogen and available potassium in the soil.

Abstract:Investigating the response of soil microbial biomass and ecological stoichiometry to tree species transition is of great significance for understanding soil nutrient cycling and availability in forest ecosystems. We measured soil microbial biomass carbon (MBC), nitrogen (MBN), phosphorus (MBP) and their stoichiometry across 0-40 cm soil depth between Mytilaria laosensis and Cunninghamia lanceolata plantations by the chloroform fumigation extraction method, which were replanted after the harvest of C. lanceolata plantation. The results showed that soil MBC in the 0-10 cm layer and soil MBN and MBP in the 0-20 cm layer under the M. laosensis were significantly higher than those under the C. lanceolata. The MBC/MBP in the 0-20 cm layer and MBN/MBP in the 10-20 cm layer were significantly lower under the M. laosensis plantation. The MBC/MBN showed no significant differences between the two forests. Soil moisture, organic carbon, total nitrogen, total phosphorus, available phosphorus were positively correlated with MBC, MBN and MBP, but negatively correlated with MBC/MBP and MBN/MBP. Results of stepwise linear regression analysis showed that MBN and MBP were mainly affected by soil total nitrogen and available phosphorus, while MBC/MBP and MBN/MBP were mainly driven by available phosphorus and organic carbon, respectively. Our results indicated that tree species transition from C. lanceolata to M. laosensis could increase soil microbial biomass in the surface layers, accelerate soil nutrients turnover and enhance soil nutrient supply. The increases of MBP under M. laosensis indicate alleviation of soil phosphorus limitation for tree growth.
Long-term nitrogen addition alters the community and energy channel but not diversity of soil nematodes in a subtropical forest

Abstract:Summary 1. Research has indicated that increases in nitrogen (N) deposition can greatly affect ecosystem processes and functions. There is limited information about the effects of long-term N addition on soil nematodes and their functional composition, although nematodes are the most abundant multicellular animals on Earth. 2. We conducted a field experiment in 2004 with four levels of N addition (0, 60, 120, and 240 kg N ha-1 yr-1) in a subtropical Cunninghamia lanceolata forest. Soil samples with three depths (0-20, 20-40 and 40-60 cm) were collected and the community structure, diversity and trophic groups of soil nematodes were determined in 2014. 3. N addition significantly increased the abundance of bacterial- and fungal-feeding nematodes, but decreased the abundance of plant-feeding nematodes at the 0-20 cm soil layer. Accordingly, the plant parasite index and enrichment index decreased but the basal index and channel index increased, which weaken the importance of the plant-based energy channel, but enhance the importance of the fungal-based energy channel. N addition had no effects on the diversity of soil nematodes in three soil depths. Structural equation modeling analysis indicated that N loading directly changed plant-feeding (total r2=0.42) nematodes, or indirectly affected bacterial- (r2=0.43), fungal- (r2=0.31) and plant-feeding nematodes via change soil nutrients, soil water content and pH. 4. These findings suggest that N addition can change the community structure and energy channels soil nematodes, which would affect soil processes and food web functions in forest soils under future environmental change scenarios.
Molecular Ecological Network Structure and Potential Function of the Bacterial Community in the Soil Profile under Indigenous Tree Plantations in Subtropical China

Abstract:The soil profile is a strong and complex physicochemical gradient that greatly affects bacterial community structure and function between soil layers. However, little is known about molecular ecological network structure and bacterial community function under differing soil profiles in planted forests. Four typical native tree species (Pinus massoniana Lamb., Castanopsis hystrix Miq., Mytilaria laosensis Lec., and Michelia macclurei Dandy) plantations were selected from subtropical China as the research object. We evaluated molecular ecological network structure as well as potential function of the soil bacterial community at different soil depths (0–20, 20–40, and 40–60 cm) within native tree plantations. Our results showed that (1) compared to the topsoil (0–20 cm), the bacterial molecular ecological network scale increased within the middle layer (20–40 cm) and the subsoil (40–60 cm), and the interaction between species was stronger; (2) module hubs and connectors were the key bacterial groups in each soil layer and increased with increasing soil depth; (3) the dominant functional groups of the bacterial communities in each soil layer were chemoheterotrophy, aerobic chemoheterotrophy, cellulolysis, ureolysis, nitrogen fixation, and nitrate reduction, and they were related to soil carbon and nitrogen cycling; and (4) the different molecular ecological network structures along with relative bacterial functional group abundances among diverse soil layers were mainly affected by soil organic carbon (SOC), NO3−-N, NH4+-N, available phosphorus (AP), and total phosphorus (TP). Our study provides a theoretical foundation for bacterial community structure together with function within soil profiles of native tree plantations in subtropical regions.

Abstract:Soil microorganisms are the key driver of the geochemical cycle in forest ecosystem. Changes in litter and roots can affect soil microbial activities and nutrient cycling; however, the impact of this change on soil microbial community composition and function remain unclear. Here, we explored the effects of litter and root manipulations [control (CK), doubled litter input (DL), litter removal (NL), root exclusion (NR), and a combination of litter removal and root exclusion (NI)] on soil bacterial and fungal communities and functional groups during a 2-year field experiment, using illumina HiSeq sequencing coupled with the function prediction platform of PICRUSt and FUNGuild. Our results showed that litter and root removal decreased the diversity of soil bacteria and fungi (AEC, Shannon, and Chao1). The bacterial communities under different treatments were dominated by the phyla Proteobacteria, Acidobacteria, and Actinomycetes, and NL and NR reduced the relative abundance of the first two phyla. For the fungal communities, Basidiomycetes, Ascomycota, and Mortierellomycota were the dominant phyla. DL increased the relative abundance of Basidiomycetes, while NL and NR decreased the relative abundance of Ascomycota. We also found that litter and root manipulations altered the functional groups related to the metabolism of cofactors and vitamins, lipid metabolism, biosynthesis of other secondary metabolites, environmental adaptation, cell growth, and death. The functional groups including ectomycorrhizal, ectomycorrhizal-orchid mycorrhizal root-associated biotrophs and soil saprotrophs in the fungal community were also different among the different treatments. Soil organic carbon (SOC), pH, and soil water content are important factors driving changes in bacterial and fungal communities, respectively. Our results demonstrate that the changes in plant detritus altered the soil microbial community structure and function by affecting soil physicochemical factors, which provides important data for understanding the material cycle of forest ecosystems under global change.
Effects of Understory or Overstory Removal on the Abundances of Soil Nematode Genera in a Eucalyptus Plantation

Abstract:In south China, eucalyptus plantations typically consist of a single-species overstory (a eucalyptus monoculture) and a dense understory of a dominant fern species. In the current study, we assessed the effects of four treatments [control (CK), understory removal (UR), tree removal (TR), and all-plant removal (PR)] on the abundances of soil nematode genera, which can provide insight into the ecological functions of understory plants and trees. Soil nematodes were sampled six times (once before and five times after treatments were implemented) at 0–5 and 5–10 cm soil depths. The temporal dynamics of nematode genera were analyzed by the principle response curves (PRC) method. At 0–5 cm depth, the abundances of most nematode genera rapidly increased shortly after vegetation removal but then gradually decreased; the effects of UR were stronger than the effects of TR. The results might be explained by the pulsed input of plant debris to soil and its subsequent depletion. At 5–10 cm depth, the nematode communities were relatively unaffected by vegetation removal within the first 162 days, but the abundances of most genera sharply decreased on day 258 and then sharply increased on day 379 (the last sampling time). The results indicated that most nematode genera, even r -selected genera, were sensitive to vegetation removal in the upper soil layer and that understory vegetation can greatly affect soil nematode communities and presumably soil food webs. The nematode genera Prismatolaimus and Diphtherophora may be good indicators of the effects of vegetation removal. The results increase our understanding of the relationships between soil nematode genera and forest plant communities and of how soil biota is affected by forest management practices.

Abstract:Pinewood nematode, Bursaphelenchus xylophilus, is one of the greatest threats to pine trees and is spreading all over the world. During the nematode's pathogenesis, plant microorganisms play important roles. However, many microbial communities, such as that in Pinus massoniana, a major host of B. xylophilus that is widely distributed in China, are not well studied, especially the fungal communities. Here, the endophytic and rhizospheric bacterial and fungal communities associated with healthy and B. xylophilus–infected P. massoniana were analyzed. The results showed that 7639 bacterial and 3108 fungal OTUs were annotated from samples of P. massoniana, the rhizosphere, and B. xylophilus. There were significant diversity differences of endophytic microbes between healthy and infected P. massoniana. The abundances of endophytic bacteria Paenibacillus, unidentified_Burkholderiaceae, Serratia, Erwinia, and Pseudoxanthomonas and fungi Penicillifer, Zygoascus, Kirschsteiniothelia, Cyberlindnera, and Sporothrix in infected pines were greater than those in healthy pines, suggesting an association of particular microbial abundances with the pathogenesis of B. xylophilus in pines. Meanwhile, the abundances of microbes of unidentified_Burkholderiaceae, Saitozyma, and Pestalotiopsis were greater and Acidothermus and Trichoderma were lower in the rhizosphere under infected pines than those under healthy pines and the differences might be caused by B. xylophilus–induced weakening of the health of pines. Our study explored the endophytic and rhizospheric microbial community changes potentially caused by B. xylophilus infection of pines.
