Abstract:We have had a long-standing interest in developing organic prodrugs for controlled delivery of CO for various therapeutic applications. Based on an earlier approach of taking advantage of a cheletropic extrusion reaction of norbornadienone to release CO, a new structural scaffold of CO prodrugs has been designed using an adamantane moiety instead of aryl groups for stabilizing a critical precursor structure, cyclopentadienone. This approach allows for elimination of multiple aromatic groups on the previous scaffolds and offers a handle for installing additional moieties for improving solubility. Specifically, prodrugs with different solubilizing groups have been synthesized and assessed for their water solubility and potency in inhibiting inflammation using TNF-α as a marker. In cell-culture studies, varying potency was observed in a fashion consistent with the polarity of a prodrug being a key factor affecting membrane permeability and thus the local concentration of CO intracellularly. However, these prodrugs all offer systemic availability of CO in animal models even if the prodrug does not appear to have the ability to traverse cellular membrane. Such results suggest that member permeability is not a major factor in determining the ability to deliver CO systemically. Further, caution is needed in directly extrapolating results from cell culture to animal models, especially for such a gaseous active principal.

Abstract:This study has hereby designed and synthesized MIL‐101(Cr)@CoOx nanoparticles to achieve ultrasound‐mediated synergistic tumor inhibition by combining CO gas therapy and chemodynamic therapy effectively. Carbon monoxide (CO) as an endogenous gaseous signaling molecule, is widely studied for its ability to inhibit cancer cell proliferation as gas therapy. However, the therapeutic efficiency of CO gas therapy is far from ideal due to the low availability of CO supply. Here, inspired by the band structure and Fenton‐related elements, MIL‐101(Cr)@CoOx nanoparticles are introduced for the first time to achieve ultrasound‐mediated synergistic therapy by combining CO gas therapy and chemodynamic therapy together effectively. MIL‐101(Cr)@CoOx can present substantial CO2 adsorption capacity for the BDC2− linkers and Co center, and enable to transformation of endogenous CO2 to CO efficiently under ultrasound irradiation due to the appropriate band structure and conduction band position. With the Co‐engaged Fenton‐like reaction, MIL‐101(Cr)@CoOx triggered the reactions to catalyze the over‐expressed intracellular H2O2 into cytotoxic hydroxyl radicals (·OH). The potential therapeutic outcome of chemodynamic therapy can be amplified by the ultrasonic cavitation‐induced conversion between Co3+ and Co2+ in vitro is demonstrated. The proliferation, ATP, and mitochondria functions of 4T1 mouse breast cancer cells are severely compromised by CO‐promoted ROS generation and the sono‐Fenton effect. Further, in vivo studies confirm that MIL‐101(Cr)@CoOx combined with ultrasound irradiation exhibits superior tumor suppression, and achieves the ultrasound‐mediated chemodynamic‐gas cancer therapy with higher precisions for personalized treatments. This new system offers a distinct concept for constructing smart ultrasound‐stimulus systems to realize synergetic cancer therapy.

Abstract:As one of the gaseous signals in living cells, carbon monoxide (CO) not only participates in many biological activities but also serves as a therapeutic agent for the treatment of diseases. However, the limited applicability of CO in gas therapy emerges from the inconvenience of direct administration of CO. Here we reported the construction of guanidinylated CO-releasing micelles, which are composed of poly(trimethylene carbonate) (PTMC)-based CO donors. The in vitro studies demonstrated that micelles in the presence of light irradiation can induce cancer death, whereas no obvious toxicity to normal cells was observed. Moreover, the functionalization of guanidine groups imparts improved cellular uptake efficiency to micelles owing to the specific interactions with the surface of cells, which synergistically increase the anticancer capacity of the system. The guanidine-functionalized CO-releasing micelles provide a new strategy for the construction of CO-releasing nanocarriers, which are expected to find applications in gas therapeutics.

Abstract:Carbon monoxide (CO) therapy has emerged as a hot topic under exploration in the field of gas therapy as it shows the promise of treating various diseases. Due to the gaseous property and the high affinity for human hemoglobin, the main challenges of administrating medicinal CO are the lack of target selectivity as well as the toxic profile at relatively high concentrations. Although abundant CO releasing molecules (CORMs) with the capacity to deliver CO in biological systems have been developed, several disadvantages related to CORMs, including random diffusion, poor solubility, potential toxicity, and lack of on-demand CO release in deep tissue, still confine their practical use. Recently, the advent of versatile nanomedicine has provided a promising chance for improving the properties of naked CORMs and simultaneously realizing the therapeutic applications of CO. This review presents a brief summarization of the emerging delivery strategies of CO based on nanomaterials for therapeutic application. First, an introduction covering the therapeutic roles of CO and several frequently used CORMs is provided. Then, recent advancements in the synthesis and application of versatile CO releasing nanomaterials are elaborated. Finally, the current challenges and future directions of these important delivery strategies are proposed.
A Near-Infrared Fluorescence Probe with a Large Stokes Shift for Detecting Carbon Monoxide in Living Cells and Mice

Abstract:Carbon monoxide (CO) can be endogenously produced in mammalian cells and regarded as an essential cell signaling molecule in the body. And it is closely related with a range of physiological and pathological processes. Although some fluorescence probes for tracking CO are reported, near-infrared (NIR) fluorescence probe for CO with a large Stokes shift remains scarce. Herein, a NIR fluorescence probe named DCX-CO is designed and synthesized. In the probe, the allyl formate group is chosen as recognizing moiety and benzopyranonitrile-xanthene-based dye is selected as fluorophore. Upon the addition of CO, the probe shows NIR emission at 762 nm with a large Stokes shift (164 nm). Moreover, the probe displays high sensitivity for CO with 16-fold fluorescence enhancement and excellent selectivity to CO over other potential interference in biological sample. Based on the low cytotoxicity, the probe can be used for the detection of exogenous and endogenous CO in HepG2 cells and HCT116 cells. In addition, taking advantage of NIR emission with a large Stokes shift, DCX-CO is applied for monitoring CO in vivo.
Patient satisfaction with anaesthesia care: development of a psychometric questionnaire and benchmarking among six hospitals in Switzerland and Austria.

Abstract:BACKGROUND We describe the development and comparison of a psychometric questionnaire on patient satisfaction with anaesthesia care among six hospitals. METHODS We used a rigorous protocol: generation of items, construction of the pilot questionnaire, pilot study, statistical analysis (construct validity, factor analysis, reliability analysis), compilation of the final questionnaire, main study, repeated analysis of construct validity and reliability. We compared the mean total problem score and the scores for the dimensions: 'Information/Involvement in decision-making', and 'Continuity of personal care by anaesthetist'. The influence of potential confounding variables was tested (multiple linear regression). RESULTS The average problem score from all hospitals was 18.6%. Most problems are mentioned in the dimensions 'Information/Involvement in decision-making' (mean problem score: 30.9%) and 'Continuity of personal care by anaesthetist' (mean problem score: 32.2%). The overall assessment of the quality of anaesthesia care was good to excellent in 98.7% of cases. The most important dimension was 'Information/Involvement in decision-making'. The mean total problem score was significantly lower for two hospitals than the total mean for all hospitals (significantly higher at two hospitals) (P<0.05). Amongst the confounding variables considered, age, sex, subjective state of health, type of anaesthesia and level of education had an influence on the total problem score and the two dimensions mentioned. There were only marginal differences with and without the influence of the confounding variables for the different hospitals. CONCLUSIONS A psychometric questionnaire on patient satisfaction with anaesthesia care must cover areas such as patient information, involvement in decision-making, and contact with the anaesthetist. The assessment using summed scores for dimensions is more informative than a global summed rating. There were significant differences between hospitals. Moreover, the high problem scores indicate a great potential for improvement at all hospitals.
The construction of a near-infrared fluorescent probe with dual advantages for imaging carbon monoxide in cells and in vivo

Abstract:As a kind of toxic gas, carbon monoxide (CO) can hinder uptake of oxygen in humans. However, more and more studies have shown that CO is an important gaseous messenger in the body and playing an indispensable role in intracellular signaling pathways. So, it is necessary to develop an analytical method to study CO in living organisms. Although there are many CO-responsive probes, most of them have the disadvantages of a small Stokes shift or short emission wavelength. In order to address the above issue, a novel probe (FDX-CO) with a large Stokes shift (190 nm) and long emission wavelength (770 nm) was firstly synthesized to detect CO. The probe shows high sensitivity and superior selectivity toward CO. Moreover, the probe was successfully used for visualizing exogenous and endogenous CO in cells by fluorescence imaging, 3D quantification analysis and flow cytometric analysis. More importantly, FDX-CO could excellently detect CO in mice, which suggests that this probe has the potential ability to image CO in vivo. This probe can be viewed as a useful tool in the research of CO.
X-ray-controllable release of carbon monoxide potentiates radiotherapy by ultrastable hybrid nanoreservoirs

Abstract:Carbon monoxide (CO) exhibits unique abilities in sensitizing cancer radiotherapy (RT). However, the development of a highly stable CO-delivery nanosystem with sustained CO release in tumor tissues and the prevention of CO leakage into normal tissues remains a challenge. Herein, an organic-inorganic hybrid strategy is proposed to create ultrastable CO nanoreservoirs by locking an unstable iron carbonyl (FeCO) prodrug in a stable mesoporous silica matrix. Different from traditional FeCO-loading nanoplatforms, FeCO-bridged nanoreservoirs not only tethered labile FeCO in the framework to prevent unwanted FeCO leakage, but also achieved sustained CO release in response to X-ray and endogenous H 2 O 2 . Importantly, FeCO-bridged nanoreservoirs exhibited the sequential release of CO and Fe 2+ , thereby performing highly efficient chemodynamic therapy. Such a powerful combination of RT, gas therapy, and chemodynamic therapy boosts robust immunogenic cell death, thus enabling the elimination of deeply metastatic colon tumors with minimal side effects. The proposed organic-inorganic hybrid strategy opens a new window for the development of stable nanoreservoirs for the on-demand delivery of unstable gases and provides a feasible approach for the sequential release of CO and metal ions from metal carbonyl complexes.

Abstract:The important role of gasotransmitters in physiology and pathophysiology suggest employing gasotransmitters for biomedical treatment. Unfortunately, the difficulty in storage and controlled delivery of these gaseous molecules hindered the development of effective gasotransmitters-based therapies. The design of a safe, facile, and wide-scale method to delivery multiple gasotransmitters is a great challenge. Herein, we use an ultrasonic assisted preparation γ-cyclodextrin metal organic framework (γ-CD-MOF) as a broad-spectrum delivery vehicle for various gasotransmitters, such as SO2, NO, and H2S. The release rate of gasotransmitters could be tuned by modifying the γ-CD-MOF with different Pluronics. The biological relevance of the exogenous gasotransmitters produced by this method is evidenced by the DNA cleavage ability and the anti-inflammatory effects. Furthermore, the γ-CD-MOF composed of food-grade γ-CD and nontoxic metal salts shows good biocompatibility and particle size (180 nm). Therefore, γ-CD-MOF is expected to be an excellent tool for the study of co-delivery and cooperative therapy of gasotransmitters.

Interfacial delivery of carbon monoxide via smart titanium implant coating for enhanced soft tissue integration with switchable antibacterial and immunomodulatory properties

Abstract:Soft tissue integration around titanium (Ti) implants is weaker than that around natural teeth, compromising long-term success of Ti implants. Carbon monoxide (CO) possesses distinctive therapeutic properties, rendering it as a highly promising candidate for enhancing STI. However, achieving controlled CO generation at the STI interface remains challenging. Herein, a controlled CO-releasing dual-function coating was constructed on Ti surfaces. Under near-infrared (NIR) irradiation, the designed surface could actively accelerate CO generation for antibiosis against both aerobic and anaerobic bacteria. More importantly, in the absence of NIR, the slow release of CO induces macrophage polarization from pro-inflammatory phenotype towards pro-regenerative phenotype. In a rat implantation model with induced infection, the designed surface effectively controlled the bacterial infection, alleviates accompanying inflammation and modulated immune microenvironment, leading to enhanced STI. Single-cell sequencing revealed that the coating alters the cytokine profile within the soft tissue, thereby influencing cellular functions. Differentially expressed genes in macrophages are highly enriched in the PIK3-Akt pathway. Furthermore, the cellular communication between fibroblasts and macrophages was significantly enhanced through the CXCL12/CXCL14/CXCR4 and CSF1-CSF1R ligand-receptor pair. These findings indicate that our coating showed an appealing prospect for enhancing STI around Ti implants, which would ultimately contribute to the improved long-term success of Ti implants.

Abstract:Covalent organic frameworks (COFs) as drug-delivery carriers have been mostly evaluated in vitro due to the lack of COFs nanocarriers that are suitable for in vivo studies. Here we develop a series of water-dispersible polymer-COF nanocomposites through the assembly of polyethylene-glycol-modified monofunctional curcumin derivatives (PEG-CCM) and amine-functionalized COFs (APTES-COF-1) for in vitro and in vivo drug delivery. The real-time fluorescence response shows efficient tracking of the COF-based materials upon cellular uptake and anticancer drug (doxorubicin (DOX)) release. Notably, in vitro and in vivo studies demonstrate that PEG-CCM@APTES-COF-1 is a smart carrier for drug delivery with superior stability, intrinsic biodegradability, high DOX loading capacity, strong and stable fluorescence, prolonged circulation time and improved drug accumulation in tumors. More intriguingly, PEG350-CCM@APTES-COF-1 presents an effective targeting strategy for brain research. We envisage that PEG-CCM@APTES-COF-1 nanocomposites represent a great promise toward the development of a multifunctional platform for cancer-targeted in vivo drug delivery.

Abstract:As nanomaterials have gained prominence in drug delivery technology, exploring their feasibility through computational methods is beneficial before practical tests. In this study, we aim to evaluate the capability of the porphyrin-based covalent organic framework COF-366 as a nanocarrier for two anticancer drugs, irinotecan (IRI) and doxorubicin (DOX). The optimal binding conformation of the drug molecules on the COF surface was predicted by using molecular docking. Subsequently, molecular dynamic simulation (MD) was performed to assess the adsorption mechanism of drug molecules on the COF in the aqueous environment. The free energy of adsorption for DOX and IRI was estimated to be -20.07 and -23.89 kcal/mol, respectively. The adsorption of both drugs on the COF surface is mainly influenced by the π-π interaction. Furthermore, density functional theory (DFT) calculation, natural bond orbital (NBO), and quantum theory of atoms in molecules (QTAIM) analyses were employed to investigate the structural stability of Drug@COF complexes and gain a detailed understanding of the interaction between them at the molecular level. Based on DFT results, it was found that in addition to π-π interaction, the bis-piperidine-phenylene interaction affects the adsorption of IRI on the COF surface. Moreover, the diffusion behavior of the drug molecule inside the COF pore was simulated using a ten-layer COF. Based on the mean square displacement analysis, the diffusion coefficients of DOX and IRI within the COF pore were calculated to be 108 and 97 um2/s, respectively. This computational study sheds light on how different types of interactions between the drug molecule and COF affect the adsorption and diffusion process. Our findings validated that the porphyrin-based COF-366 can serve as a nanobased platform for delivering DOX and IRI.

Abstract:As one of the most valuable endogenous gas signaling molecules, carbon monoxide (CO) has been demonstrated in numerous studies to show excellent promise in the treatment of diseases, such as cancer. However, for many years, the inherent high affinity of CO for hemoglobin severely impeded the clinical transformation of CO-based treatments. Therefore, the controlled delivery of CO to target tissues has become a common challenge. Herein, an efficient ultrasonic-triggered and targeted CO release strategy was constructed based on a novel targeted acoustic release carrier of carbon monoxide (TARC-CO) that we synthesized in this study. The designed TARC-COs could afford a safe, stable, and ultrasoundguided delivery of CO in vivo by loading a specified dose of CO inside microbubbles, resulting in breast tumor suppression. Taking advantage of the high loading capacity of microbubbles, the unit volume of TARC-CO suspension could encapsulate up to 337.1 +/- 8.0 (x10(3) ppm) of CO. In addition, the satisfactory ultrasound contrast-enhanced ability of TARC-COs achieved real-time interactive guidance and visual policing of CO delivery. For the in vitro antitumor study, TARC-COs with ultrasonic irradiation were demonstrated to effectively induce mitochondrial dysfunction by reducing mitochondrial membrane potential, leading to the apoptosis of 4T1 cells. In addition, we realized that TARC-CO-based treatment could significantly slow the growth rate of tumors by inducing apoptosis, inhibiting the proliferation of cancer cells, and limiting tumor angiogenesis. In summary, this proof-of-concept study demonstrates the feasibility and tremendous potential of TARC-COs for controlled release of CO, which can be expected to provide new inspirations and a promising perspective for therapy based on active gases.
Strain-Promoted, Visible Light Triggerable CO Releasing Micelles

Abstract:Carbon monoxide (CO), along with nitric oxide and hydrogen sulfide, is one of a trinity of known gasotransmitters, or endogenously produced gaseous molecules that signal and regulate a panoply of physiological functions. CO releasing molecules (CORMs) are chemical tools that enable the study and application of this ephemeral gas, that, ideally, release CO on-demand when externally stimulated. Surveying the available triggers, photolysis is optimal: it is contactless and grants practitioners unparalleled spatial and temporal control. However, current photo-triggered CORMs are capricious and do not meet current needs. Presented here is a highly efficient platform for the visible light triggered release of CO gas. This platform is built on a unique CO containing functionality, the cyclopropenone, which undergoes facile decar-bonylation through visible light (470 nm) mediated photoredox catalysis. Due to the exothermic strain-release that oc-curs upon formation of CO, this photoreaction is rapid, quantitative, and has tunable release rates. To render this photo-CORM water soluble, deliverable, and to keep reactants in proximity, necessary components were polymerized into block copolymers that self-assemble into CO releasing micelles (CORMIs). This platform was compared directly to other state-of-the-art CORMs, showing significantly improved CO production efficiency, lower toxicity, tunable release rates, and con-sistent efficacy in ex vivo and in vitro settings.

Abstract:The purpose of this study was to investigate the delivery of poorly water-soluble non-steroidal anti-inflammatory drugs (NSAIDs) by carboxyl-functionalized mesoporous silica nanoparticles (MSN-COOH) with high specific surface area (SBET). In this study, MSN-COOH was prepared by collaborative self-assembly using cetyltrimethylammonium bromide (CTAB) as template and hydrolysis (3-triethoxyl-propyl)succinic anhydride (TESPSA) as co-structure auxiliary directing agent (CSDA). The drug delivery systems were constructed with NSAIDs including Nimesulide (NMS) and Indomethacin (IMC) as model drugs. Moreover, the characterization techniques, hemolysis and bio-adsorption testes, in vitro drug release and in vivo biological studies of MSN-COOH were also carried out. The characterization results showed that MSN-COOH is spheres with clearly visible irregular honeycomb nanopores and rough surface (SBET: 1257 m2/g, pore volume (VP): 1.17 cm3/g). After loading NMS/IMC into MSN-COOH with high drug loading efficiency (NMS: 98.7% and IMC: 98.2%), most crystalline NMS and IMC converted to amorphous phase confirmed using differential scanning calorimeter (DSC) and X-ray power diffraction (XRD) analysis. Meanwhile, MSN-COOH significantly increased the dissolution of NMS and IMC compared with non-functionalized mesoporous silica nanoparticles (MSN), which was also confirmed by wettability experiments. The results of in vivo biological effects showed that MSN-COOH had higher bioavailability of NMS and IMC than MSN, and exerted strong anti-inflammatory effects by delivering more NMS and IMC in vivo.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Statement of Significance</h3>: This study successfully prepared MSNs-COOH (mesoporous silica nanoparticles modified with negatively charged carboxyl groups on the surface and in the pores) with high specific surface area and pore volume by using the negatively charged carboxyl group (hyd-TESPSA) and the positively charged CTAB self-assembled through electrostatic attraction under alkaline conditions. The drug delivery systems were constructed with Nimesulide (NMS) and Indomethacin (IMC) as model drugs. The results showed MSNs-COOH had high drug loading capacity and also exhibited good in vitro drug release properties. Interestingly, NMS loaded MSNs-COOH also had a potential pH responsive release effect. In vivo biological studies revealed that NMS/IMC loaded MSNs-COOH could evidently improve the bioavailability and played the strong anti-inflammatory effects.

Abstract:Highlights • The recent advances in CO releasing nanomaterials for biological delivery of CO are reviewed. • The synthesis and properties of CORNMs, especially the release mechanisms and detection methods are discussed and listed. • A brief outlook on the future research of employing nanomaterials in CO biological release are explored and discussed. As an endogenous signaling molecule, carbon monoxide (CO) has emerged as an increasingly promising option regarding as gas therapy due to its positive pharmacological effects in various diseases. Owing to the gaseous nature and potential toxicity, it is particularly important to modulate the CO release dosages and targeted locations to elucidate the biological mechanisms of CO and facilitate its clinical applications. Based on these, diverse CO-releasing molecules (CORMs) have been developed for controlled release of CO in biological systems. However, practical applications of these CORMs are limited by several disadvantages including low stability, poor solubility, weak releasing controllability, random diffusion, and potential toxicity. In light of rapid developments and diverse advantages of nanomedicine, abundant nanomaterials releasing CO in controlled ways have been developed for therapeutic purposes across various diseases. Due to their nanoscale sizes, diversified compositions and modified surfaces, vast CO-releasing nanomaterials (CORNMs) have been constructed and exhibited controlled CO release in specific locations under various stimuli with better pharmacokinetics and pharmacodynamics. In this review, we present the recent progress in CORNMs according to their compositions. Following a concise introduction to CO therapy, CORMs and CORNMs, the representative research progress of CORNMs constructed from organic nanostructures, hybrid nanomaterials, inorganic nanomaterials, and nanocomposites is elaborated. The basic properties of these CORNMs, such as active components, CO releasing mechanisms, detection methods, and therapeutic applications, are discussed in detail and listed in a table. Finally, we explore and discuss the prospects and challenges associated with utilizing nanomaterials for biological CO release. Loading cinnamon oil into pH-sensitive chitosan grafted mesoporous silica nanoparticles via supercritical carbon dioxide

Abstract:Stimulus-responsive mesoporous silica nanoparticles (MSNs) have displayed great potentiality for controlled-release and targeted drug delivery. In the current work, a supercritical fluid method was utilized to successfully prepare cinnamon oil loaded into chitosan grafted MSNs (CO@CS-MSNs). The influencing factors of drug loads, such as pressure, temperature, impregnation time and depressure time, were investigated. The structure of CO@CS-MSNs was demonstrated with Fourier-transform infrared (FT-IR) spectroscopy, transmission electron microscope (TEM), scanning electron microscopy (SEM), thermogravimetry (TG) as well as X-ray diffraction (XRD). The drug release assays in vitro at various pH conditions displayed that CO@CS-MSNs had an excellent pH-responsive release behavior, which confirmed that CO was loaded successfully into the CO@CS-MSNs. The findings indicated that the supercritical fluid approach is a non-destructive and efficient approach for stimulus-responsive MSNs, which is expected to further expand its application range.
Phospholipid-inspired alkoxylation induces crystallization and cellular uptake of luminescent COF nanocarriers

Abstract:The porous nature and structural variability of covalent organic frameworks (COFs) make them preferred for drug loading and delivery applications. However, most COF materials suffer from poor luminescent properties and inefficiency for cell uptake. Herein, we experimentally demonstrate the crucial role of long alkoxy chains in the synthesis of crystalline COF nanostructures with high cellular uptake efficiency. After luminescence integration through band engineering, the semiconducting COF exhibits an optical bandgap of 2.05 eV, an emission wavelength of 632 nm, a high quantum yield of 37 %, and excellent fluorescence stability (100 % at 3 h). Such excellent optical properties of the designed COF nanocarriers enable quantitative evaluations of cellular uptake and visual tracking of drug delivery. It was demonstrated that the cellular uptake efficiency was enhanced by orders of magnitude higher for the COF after the introduction of long n-octyloxy chains, which firstly delivered the anticancer camptothecin (CPT) to cell lysosomes, and then underwent "endo/lysosomal escape" to induce cell apoptosis. In vivo assay evidenced a significant enhancement in the therapeutic effect with a 96 % inhibition of tumor growth after 14 days of treatment. This progress sheds light on designing cutting-edge drug delivery nanosystems based on COF materials with integrated diagnostic and therapeutic functions.

Abstract:Carbon monoxide (CO) has been studied on the treatment of numerous diseases such as cancers, rheumatoid arthritis, and so on. However, real-time tracking and controlled release of CO gas for a safe and efficient therapy are till a challenging task. Although many CO-releasing molecules (CORMs) have been widely developed and used because of the release of CO by external stimuli, CORMs still have some disadvantages, such as fast CO release, potential toxicity and poor solubility. Herein, light-triggered CO-releasing hybrid material (UIO-66-NH2/CORM@FA-CS-5-FAM) based on UIO-66-NH2 nanoparticles and [Mn(CO)3(CH3CN)(HPYDC)]·CH3CN (H2PYDC = 2, 5-pyridinedicarboxylic acid, abbreviated as CORM) coated with FA-CS-5-FAM was constructed. The composition characteristics of the hybrid material include: a chitosan (CS) increases the residence time of drugs on the application/absorption sites, folic acid (FA) targets the cancer cells and 5-carboxyfluorescein (5-FAM) is used as an imaging agent. Comparison with the free CORM, UIO-66-NH2/CORM@FA-CS-5-FAM nanocomposite exhibits a long-time CO release and possesses the excellent characteristics of targeted cancer cells and fluorescence imaging. Cell viability assays indicate that UIO-66-NH2/CORM@FA-CS-5-FAM shows good biocompatibility and anticancer activity.
