Abstract:This experiment was conducted to investigate the effects of glutamine(Gln) and Clostridium butyricum(CB) on growth performance,immune function,small intestinal morphology and cecal microflora in weanling piglets.Two hundred and eighty-eight piglets weaned at the age of 21 d were randomly divided into four groups with three replicates in every group and twenty-four piglets in every replicate.The four groups were basal diet group(CT),basal diet+1% glutamine group(GT),basal diet+500 mg/kg Clostridium butyricum group(CBT),and basal diet+1% glutamine+500 mg/kg Clostridium butyricum group(GCT),respectively.The results were showed as follows: 1) during 14 to 21 d and 0 to 21 d after weaned,the average daily gain(ADG) of GT and GCT was significantly higher than that of CT,the ADG of GCT was significantly higher than that of CBT,and the feed to gain ratio of GCT was significantly lower than that of CT.2) On the 7th day after weaned,the concentration of complement 4(C4) in GCT and SOD activity in GT,CBT and GCT were significantly higher than those in CT,meanwhile,the T-AOC in CBT and GCT were significantly higher than that in CT.On the 14th day after weaned,the lysozyme activity,the concentration of complement 3(C3) and T-AOC in GT,CBT and GCT were significantly higher than those in CT;the concentration of C4 and SOD activity in GT and GCT were significantly higher than those in CT,respectively.3) On the 7th day after weaned,the villus height of duodenum in GT,CBT and GCT were significantly increased compared with that in CT(P0.05);the villus height of ileum in CBT was increased significantly compared with that in CT(P0.05);the crypt depth of jejunum and ileum in GCT were significantly reduced compared with those in CT(P0.05).On the 14th day after weaned,the villus height of duodenum and ileum in GCT were significantly increased compared with those in CT(P0.05),while the crypt depth of duodenum and ileum in GCT were significantly reduced compared with those in CT(P0.05).4) On the 14th day after weaned,the number of E.coli in GCT was significantly less than that in CT(P0.05),while the number of Bifidobacterium and Lactobacillus were significantly larger than those in CT(P0.05).

Abstract:Abstract The gastrointestinal tract has a central role in production efficiency due to its involvement in the digestive function as well as in immunological, nervous, and metabolic functions. In ruminants, a great deal of attention is placed on ruminal functionality and safety. However, it is now recognized that the hindgut is involved in the onset of some metabolic issues, normally associated with ruminal imbalances, as well as in the digestion efficiency. Thus, the application of nutritional strategies that target the hindgut are gaining interest in cattle nutrition. The study evaluated the effects of a protected source of calcium gluconate on performance, digestibility, and health in beef cattle. Charolaise bulls (n = 241) were randomly divided into two groups: i) Control (n = 120) basal diets + 10g/head/day of a placebo of wheat bran; ii) Treatment (n = 121) basal diets + 10 g·bull−1·d−1of a protected source of calcium gluconate (Lactibute, Selko Trouw Nutrition). The population was further divided into two subgroups: i) “Overall population” (n = 201) housed in pens of 7to 8 animals each; ii) “Beefmonitor population” (n = 40). The latter was housed in two pens (Control and Treatment) with 20 animals each equipped with two automatic scales for BW located at the drinkers, to evaluate the daily growth trends. Growth performance, feed intake (FI), feed conversion ratio (FCR), were evaluated with specific approaches in the two populations. The incidence of diseases was evaluated equally in the two populations. The apparent total tract digestibility (aTTD) was evaluated on 10 representative pens per group using a portable near-infrared system that compares the composition of the diets and the correlated feces. All the data were analyzed statistically using the SAS software and applying specific models. The average daily gain (ADG) was improved by the Treatment (+40 and +43 g·bull−1·d−1compared with the Control group, in the “Overall” and “Beefmonitor” populations; Table 1 and Figure 1; P = 0.0084 and < 0.0001, respectively), and the final BW were also heavier (706.82 and 705.83 vs 699.75 and 697.33 kg in the “Overall” and “Beefmonitor” populations; P = 0.0084 and 0.037, respectively). The treatment improved both, FCR (6.90 and 7.03 vs 7.51 and 7.28 in the “Overall” and “Beefmonitor” populations; P = 0.0008 and < 0.0001 respectively; Table 1 and Figure 1) and the aTTDs of starch (97.05 vs 95.71%; P < 0.0001), cellulose (57.91 vs 52.95%; P < 0.0001) and NDF (52.02 vs 50.02%; P = 0.020). The incidence of lameness was reduced in the Treatment group (0.99 vs 7%; P = 0.0282). Including protected sources of calcium gluconate can be functional to improve production efficiency and animal health, as a consequence of its proactive role on intestinal health, functionality and epithelial integrity.

Abstract:Abstract The objective of this study was to evaluate effects of feeding supplemental isoacids in a growing diet on parameters of rumen fermentation and in situ DM disappearance. Eight mature ruminally cannulated beef heifers (Angus crossbred; mean BW 618 ± 31 kg) were used in a replicated 4 x 4 Latin square design. Heifers were housed in individual pens and fed the following treatment diets: no supplemental isoacids (ISO) in the basal diet (Control); 7.2 g of 2-methylbutyric acid· animal-1· day-1 (ISO1); 7.2 g of 2-methylbutyric + 7.2 g isobutyric acids· animal-1· day-1 (ISO2); and 7.2 g 2-methylbutyric + 7.2 g isobutyric + 7.2 g isovaleric acid· animal-1· day-1 (ISO3). Basal diet consisted of (DM basis) 26.9% wheat hay, 35.5% steam flaked corn, 22.6% wet corn gluten feed, 12.3% DDG, and 2.7% mineral/vitamin supplement. Each experimental period lasted 28 d, with 21 d for diet adaptation and 7 d for data and sample collection. On day 1 of each sampling period ruminal fluid was collected 2 hours before and 1, 3, 7 and 10 hours after morning feeding. The pH of ruminal contents was immediately measured with an electrode pH meter. Urine samples (approximately 200 mL) were collected on day 2 of each sampling period 4 h after the feeding and filtered to remove particles. Urine was analyzed for creatinine, uric acid, and allantoin. Bags were placed into rumen for 72, 48, 36, 24, 12, 8, 4 and 0 hours and all bags were removed at same time on the end of incubation (hour 0), rinsed with tap water and dried at 55°C. Data were analyzed using the MIXED procedure of SAS. A contrast was observed (P = 0.029) for overall rumen pH between control versus isoacid treatments (6.44 vs 6.31). Uric acid, allantoin, total purines and intestinal flow of microbial nitrogen were not affected by treatments (P ≥ 0.10) with overall means of 10.47, 328.80, 339.27 mmol/d and 246.64 g N/d, respectively. Overall, no effects were observed for in situ dry matter disappearance (P ≥ 0.10). However, at hour 4 differences between ISO2 and ISO3 (P = 0.02; 44.89% vs 38.45%) and ISO2 and ISO4 (P = 0.05; 44.89% vs 39.75%) were observed. No effects of in situ DM disappearance was observed (P ≥ 0.10). Supplementing beef heifers with isoacids may reduce rumen pH as a result of increased ruminal volatile fatty acid production; however, supplemental ISO in this study did not affect microbial nitrogen synthesis or dry matter in situ disappearance. Further analyses are currently in progress.

Abstract:Twenty-one beef heifers were used in a randomized complete block design lasting 206 ± 8 d (mean ± SD) to evaluate DMI, growth, and gastrointestinal tract and carcass characteristics with increasing dose of dietary hydrogenated fat-embedded calcium gluconate (HFCG). Dietary treatments included HFCG included at: 0.0 (CON); 0.09% (CG09), or 0.18% DM (CG18). Heifers were fed individually and fecal samples were collected on d 171 and 192. Gastrointestinal tissue, digesta, carcass merit, and meat quality and sensory characteristics were assessed after slaughter. Data were analyzed to determine the linear and quadratic effects of HFCG dose. Dry matter intake, ADG, and G:F were not affected (P ≥ 0.08) by dose of HFCG. Gastrointestinal fermentation was minimally affected but quadratic responses (P = 0.03 and P = 0.01, respectively) were observed for jejunal digesta pH and for the molar proportion of isovalerate in colon digesta where both increased and then decreased as HFCG dose increased. Carcass (P ≥ 0.30), meat quality (P ≥ 0.06), and sensory characteristics (P ≥ 0.21) were not affected by dose of HFCG. We conclude that feeding HFCG to finishing beef heifers diet did not affect DMI, growth, gastrointestinal fermentation parameters, or carcass merit, meat quality and palatability.

Effect of composition of ruminally-infused short-chain fatty acids on net fluxes of nutrients across portal-drained viscera in underfed ewes

Abstract:Four ewes, each fitted with a rumen cannula and with catheters in the mesenteric artery and portal and mesenteric veins, received continuous intrarumen infusions of water or of short-chain fatty acids (SCFA). SCFA infusions were isoenergetic (83 kJ/h) and provided rumen molar proportions (acetate : propionate : butyrate) of 70 : 20 : 10, 50 : 40 : 10 or 50 : 20 : 30. The rumen SCFA production rate with the basal diet was 90·0, 23·1 and 8·8 mmol/h for acetate, propionate and butyrate respectively. Portal net fluxes indicated that 74, 67 and 22–30 % of infused acetate, propionate and butyrate respectively, reached the portal vein. Portal net release of β-hydroxybutyrate increased with SCFA infusions, irrespective of the amount of butyrate infused. Portal net release of lactate decreased with high-butyrate infusion. Portal net uptake of glucose increased with the SCFA infusions. In ewes infused with water, a portal net uptake of total amino acids (AA) was observed. SCFA infusions decreased the uptake of nonessential AA (glutamate, glycine, but not glutamine) and increased the net release of tyrosine and essential AA (isoleucine, leucine). Portal net fluxes of AA were similar with both high-acetate and high-propionate infusions. Lower net uptake of glutamine and net release of most essential AA and some nonessential AA were observed with the high-butyrate infusion. Energetic summation of portal net release was not significantly different between the three SCFA infusions, although it tended to be lower with high-butyrate infusion. This may be related to the higher trophic effect of butyrate on the digestive mucosa.

Supplementing sodium butyrate to limit-fed heifers: Effects on growth, coccidiosis, urinary purine derivatives, and apparent total-tract nutrient digestibility

Abstract:The objective of this study was to assess the growth, apparent total-tract digestibility of nutrients, the prevalence of coccidia, and purine derivatives in postweaning heifers when limit-fed a diet supplemented with sodium butyrate (SB). A 12 wk randomized complete block experiment was conducted using 24 Holstein heifers (92.8 d ± 1.9 d of age and initial body weight [BW] of 99.6 ± 15.2 kg [mean ± standard deviation]). Treatments were 100 g soybean meal (control; CON) and 0.75 g of SB/kg of BW + 100 g soybean meal (SB). Diets were formulated to contain 16.4% crude protein, 2.27 Mcal/kg metabolizable energy (ME), and fed at a feed out rate of 2.15% of BW on a dry matter basis. Intakes were recorded daily while growth measurements and BW were recorded weekly. Urine and fecal samples were taken every 2 wk. On d 42 through d 49 an apparent total-tract digestibility phase took place using acid detergent insoluble ash as a marker. Growth measurements were similar among treatments except CON heifers grew longer and tended to be taller at the withers. A trend was observed for CON animals to have lower levels of coccidian oocytes by week. Heifers fed SB had lower blood glucose levels and higher levels of ketones in their blood. Urinary volume was greater for heifers fed SB throughout the 12 wk study. Total purine derivatives were greater in CON heifers. Dry matter, organic matter and acid detergent fiber digestibilities were greater for heifers fed SB compared with CON heifers. Crude protein, neutral detergent fiber, and ash digestibilities tended to be greater in heifers fed SB than in CON heifers. These results suggested no growth benefit of supplementing SB to limit-fed heifers; however, apparent total-tract fiber, ash, and crude protein digestibilities were improved in the SB fed heifers likely due to improved ruminal and intestinal development.
Effects of a multispecies direct-fed microbial on gastrointestinal permeability during feed restriction in a growing heifer model

Abstract:The objectives were to evaluate the effects of a 4-strain direct-fed microbial (DFM) on gastrointestinal tract (GIT) permeability and inflammation during feed restriction (FR) in heifers. Holstein heifers (n = 32; mean ± standard deviation; 295 ± 25 kg body weight; 287 ± 17 d of age) were used in an experiment conducted in 2 replicates (16/replicate). Heifers were randomly assigned to 1 of 2 top-dressed dietary treatments: (1) control (CON; 10 g/d dried lactose; n = 16) or (2) DFM containing a commercial blend of Lactobacillus animalis, Propionibacterium freudenreichii, Bacillus licheniformis, and Bacillus subtilis at 11.8 × 109 cfu/d (PRO; 10 g/d 4-strain DFM; n = 16). The trial consisted of 2 experimental periods (P): P1 (14 d) served as baseline for P2 (5 d), when all heifers were restricted to 40% of their P1 dry matter intake (DMI). On P1 d 12 and P2 d 2 and 5, GIT permeability was evaluated using oral chromium (Cr)-EDTA. By design, FR decreased DMI (60%) and body weight (∼18 kg) in all heifers. Regardless of treatment, during FR, all heifers had decreased circulating glucose, β-hydroxybutyrate, insulin, and l-lactate (4, 14, 45, and 19%, respectively), but increased nonesterified fatty acids, serum amyloid A, and haptoglobin (3.0-, 1.7-, and 5.0-fold, respectively). Circulating white blood cells, neutrophils, lymphocytes, and basophils decreased (4, 7, 5, and 6%, respectively), whereas eosinophils increased (41%) during P2 irrespective of dietary treatment. Circulating IFN-γ inducible protein-10 increased (23%) during FR compared with P1 regardless of treatment. Plasma Cr area under the curve increased in all heifers on d 2 and 5 (10 and 14%, respectively) of P2 relative to P1, but this was unaltered by dietary treatment. In summary, FR compromised GIT barrier function and stimulated an inflammatory response, but this did not appear to be ameliorated by PRO.
Effects of bacterial direct-fed microbial combinations on beef cattle growth performance, feeding behavior, nutrient digestibility, ruminal morphology, and carcass characteristics

Abstract:The effects of the dietary inclusion of a mixture of bacterial direct-fed microbial (DFM) on feedlot beef cattle growth performance, carcass characteristics, nutrient digestibility, feeding behavior, and ruminal papillae morphology were evaluated. Crossbred-Angus steers (n = 192; initial body weight (BW) = 409 kg ± 8 kg) were blocked by BW and randomly assigned into 48 pens (4 steers/pen and 16 pens/treatment) following a randomized complete block design. A steam-flaked corn-based fishing diet was offered to ad libitum intake once daily for 153 d containing the following treatments: (1) Control (no DFM, lactose carrier only); (2) treat-A (Lactobacillus animalis, Propionibacterium freudenreichii, Bacillus subtilis, and Bacillus licheniformis), at 1:1:1:3 ratio, respectively; totaling 6 × 109 CFU (50 mg)/animal-daily minimum; and (3) treat-B, the same DFM combination, but with doses at 1:1:3:1 ratio. Bacterial counts were ~30% greater than the minimum expected. Data were analyzed using the GLIMMIX procedure of SAS, with pen as the experimental unit, the fixed effect of treatment, and the random effect of BW-block, while preplanned contrasts comparing Control × treat-A or treat-B were used. Steers offered treat-A had increased carcass-adjusted average daily gain (P = 0.03) by 6.7%, gain efficiency (P < 0.01) by 6%, tended (P = 0.07) to have increased carcass-adjusted final BW by 15 kg, and hot carcass weight (P = 0.07) by 10 kg, while treat-B did not differ (P ≥ 0.17) from control. Overall dry matter (DM) intake (P = 0.36) and other carcass traits (P ≥ 0.13) were not affected by treatments. Steers offered treat-A tended to have increased digestibility of DM (P = 0.07) by 3%, neutral detergent fiber (P = 0.10), and hemicellulose (P = 0.08) by 9% compared with control, while treat-B did not differ (P ≥ 0.10) from control. No treatment × period interactions (P ≥ 0.21) or main effects of treatment (P ≥ 0.12) were observed during 24-h feeding behavior. Steers ruminated, ate, chewed, and were more active (P ≤ 0.01) during the second behavioral assessment (day 113), while drinking behavior was not affected (P ≥ 0.88). Ruminal papillae morphology and ruminal ammonia concentration (ruminal fluid collected at slaughter facility) were not affected by treatment (P ≥ 0.39). Steers offered the DFM treat-A had improved growth performance and it positively affected carcass weight and nutrient digestion. The DFM combinations did not seem to affect feedlot cattle feeding behavior or ruminal papillae morphology.

Abstract:Three dairy heifers received intraperitoneal infusions (one liter) of the following solutions in a 3×3 Latin-square-designed trial: (I) 0.9% sodium chloride (control); (II) 5.4% glucose; (III) 25% glucose. The heifers were fed a completely pelleted ration (60% alfalfa meal, 40% ground corn) ad libitum once daily for a 3-hr period, and the infusions were given immediately before food was offered. The food consumption for Treatments I, II, and III was 62.6, 71.6, and 72.1 g/BWkg.73, and jugular blood glucose concentration 1 hr after feeding was 50.5, 57.2, and 69.2mg per 100ml, respectively. It was concluded that the glucostatic mechanism for food intake regulation is not important in ruminants.Isocaloric amounts of acetate, propionate, butyrate, and a A7FA mixture (60, 20, and 20% of the calories from acetate, propionate, and butyrate, respectively) were infused intraruminally into two cows fed a pelleted ration (75% alfalfa meal, 25% ground corn) and two cows fed a good-quality alfalfa hay. The rations were fed ad libitum from 10 am to 1 pm and from 4:30 pm to 8 am. Three consecutive days of water infusion (control) preceded three days of metabolite infusion. The infusions were given from 8 am to 1 pm each day, and the level of metabolite infused was equivalent to 15% of the estimated digestible energy requirement of the animals. The infusion of acetate, propionate, and the VFA mixture reduced (P<.05) the voluntary consumption of the pelleted ration during the 10 am to 1 pm feeding interval. Propionate and butyrate infusion caused a significant (P<.05) reduction in the consumption of hay in the 10 am to 1 pm feeding interval. It was concluded that acetate, propionate, and butyrate can act as satiety signal compounds in the regulation of food intake. Blood and rumen metabolites were examined, and the possible mechanism of action of these metabolites in causing satiation was discussed.

Abstract:This study was conducted to examine the effects of dietary isobutyrate supplementation on gene ex-pressions related to rumen and small intestine development of pre-weaning and post-weaning calves. Thirty-six 30-day-old Chinese Hostein dairy calves with similar body weight [(40.0±0.2) kg] were divided into four groups at random. Isobutyrate was added to a basal diet with 0 ( control) , 3, 6 and 9 g/d, respectively. Three calves from each group were slaughtered at the age of 45, 60 ( weaning age) and 90 days, samples of ruminal and small intestinal mucosa were collected to detect the mRNA expression levels of growth hormone receptor ( GHR) , insulin receptor ( INSR) , 3-hydroxy-3-methylglutaryl-CoA synthase 1 ( HMGCS1) and sodium-glu-cose cotransporter 1 ( SGLT1 ) by the real-time quantitative PCR. The results showed that body weight and mRNA expression levels of GHR, INSR and HMGCS1 increased significantly with age ( P<0.05) , but SGLT1 mRNA expression level firstly decreased then increased before and after weaning, and the changes were signifi-cant ( P<0.05) . At the age of 45 and 60 days, body weight in 9 g/d isobutyrate group was significantly higher than that in control group (P<0.05), but was not significant different from that in 6 g/d isobutyrate group ( P>0.05) . At all days of age and all parts of rumen and small intestine, the mRNA expression levels of GHR, INSR and HMGCS1 in 6 and 9 g/d isobutyrate groups were higher than those in control group, and there were no significant differences between the two groups ( P>0.05) . At all parts of small intestine, the supplementa-tion of isobutyrate did not affect the mRNA expression level of SGLT1 of 45-day-old and 90-day-old calves ( P>0.05) . At the age of 60 days, mRNA expression level of SGLT1 of duodenum in 6 and 9 g/d isobutyrate groups was significantly higher than that in control group and 3 g/d isobutyrate group, while mRNA expression level of SGLT1 of proximal jejunum, distal jejunum and ileum in 9 g/d isobutyrate group was significantly higher than that in control group (P<0.05), but had no significant difference with that in 6 g/d isobutyrate group ( P>0.05) . The results indicate that the growth of calves and the development of ruminal and small intes-tinal mucosa can be promoted by the supplementation of isobutyrate, the optimal dose of isobutyrate is 6 g/d under conditions in the present study.

Abstract:The objective of this study was to investigate the effects of non-protected and rumen-protected gamma-aminobutyric acid (GABA) supplementation on dry matter intake (DMI), energy balance, milk performance, and serum metabolites in transition cows. Forty cows were blocked based on previous milk production, parity, estimated calving date and body weight and were randomly assigned to one of four treatments and received either no GABA (control) or were supplemented with non-protected GABA (0.6g/day) or rumen-protected GABA (0.6 and 1.2g/day), respectively. The experiment lasted from week 2 before calving to week 4 after calving. Milk yield and milk composition were recorded weekly. Serum concentrations of GABA, neuropeptide Y, cholecystokinin, leptin and biochemical and antioxidative metabolites were analyzed weekly. In week 3 and 4 after calving, DMI was higher (P<0.05) in the cows fed 1.2g/day rumen-protected GABA, compared with that of the control. In week 4 after calving, milk protein yield was higher in the cows fed 1.2g/day rumen-protected GABA than that of the control cows. No differences were observed in the serum concentrations of GABA, neuropeptide Y and leptin among all the treatments, while non-esterified fatty acids and cholecystokinin concentrations decreased (P<0.01) in cows fed rumen-protected GABA on day 15 and 22 after calving, respectively. Serum total antioxidative capacity and concentrations of glutathione peroxidase and superoxide dismutase were higher (P<0.05) at several sampling points in cows fed rumen-protected GABA. In conclusion, supplementation of rumen-protected GABA at 1.2g/day increased feed intake, improved milk protein yield, and decreased non-esterified fatty acids suggesting supplementation may be beneficial to postpartum dairy cows.
Effect of increasing ruminal butyrate on portal and hepatic nutrient flux in steers

Abstract:Six Holstein steers (mean +/- SE BW = 344 +/- 10 kg) fitted with hepatic, portal, and mesenteric vein and mesenteric artery catheters and a ruminal cannula were used in a 6 x 6 Latin square design to evaluate the effects of increasing ruminal butyrate on net portal-drained visceral and hepatic nutrient flux. Steers were fed a 40% brome hay, 60% concentrate diet in 12 portions daily at 1.25 x NEm. Water (control) or butyrate at 50, 100, 150, 200, or 250 mmol/h was supplied continuously via the ruminal cannula. Simultaneous arterial, portal, and hepatic blood samples were taken at hourly intervals from 15 to 20 h of ruminal infusion. Portal and hepatic blood flow was determined by continuous infusion of P-aminohippurate, and net nutrient flux was calculated as the difference between venous and arterial concentrations times blood flow. Ruminal and arterial concentrations and total splanchnic flux of butyrate increased (P less than .01) with increased butyrate infusion. Arterial concentrations of acetate (P less than .10), alpha-amino-N (P less than .05), and glucose (P less than .01) decreased with increased butyrate, whereas arterial beta-hydroxybutyrate (P less than .01) and acetoacetate (P less than .05) increased. Increased butyrate produced an increased portal-drained visceral flux of acetoacetate and an increased net hepatic flux of beta-hydroxybutyrate. Urea N and glucose net portal and hepatic fluxes were not affected by ruminal butyrate. Alpha-amino-N uptake by the liver decreased with increased butyrate (P less than .10). Simple linear regression (r2 = .985) indicated that 25.8% of ruminally infused butyrate appeared in portal blood as butyrate. Only 14% could be accounted for as net portal-drained visceral flux of acetoacetate plus beta-hydroxybutyrate.

Abstract:Eight rumen-fistulated Holstein cows, averaging 77 d postpartum, were used in a replicated 4 X 4 Latin square design with 28-d periods to investigate the effect of ammonium salts of isobutyrate, 2-methylbutyrate, isovalerate, and n-valerate on animal performance and their possible mechanism(s) and site(s) of action. Each cow was fed ad libitum a complete mixed diet of 55% corn silage and 45% concentrate on a dry basis that was supplemented with 1.8 kg of premix daily. Treatments were 1) control, 2) ammonium salts of volatile fatty acids in premix, 3) ammonium salts of volatile fatty acids ruminally infused, or 4) ammonium salts of volatile fatty acids abomasally infused. Mean ruminal fluid pH and concentrations of ammonia and volatile fatty acids for treatment comparisons were not different. Plasma concentrations of isobutyrate, 2-methylbutyrate, and valerate differed among treatments, but there was no significant effect on dry matter intake, milk production, milk composition, or efficiency of feed utilization. Apparent nutrient digestibility; disappearance of dry matter, cellulose, and nitrogen from polyester bags suspended in the rumen; and plasma concentrations of glucose, free fatty acids, and growth hormone also were not significantly affected by treatment.

Abstract:A 48 year old man with normal hepatic function presented with abnormally prolonged bleeding following orally administered warfarin. Warfarin and diphenylhydantoin are known to be hydroxylated in the liver. To investigate the possibility of the patient having a metabolic (hydroxylating) defect, his ability to metabolize diphenylhydantoin was studied, as it would have been unethical to administer warfarin again. Parallel studies were carried out on two healthy subjects as a basis for comparison. On three separate occasions the elimination half-life for diphenylhydantoin in the patient (158, 87, 72 hours) was significantly longer than those obtained in the normal subjects (31 and 32 hours). This difference was most probably due to decreased hydroxylation of diphenylhydantoin to 5-(p-hydroxy-phenyl)-5-phenylhydantoin since his urinary elimination of this substance was demonstrated to be significantly less than that of the normal subjects. The results suggest that this patient had a reduced capacity to hydroxylate diphenylhydantoin, and this defect in liver hydroxylation may explain his increased sensitivity to warfarin.

Abstract:Introduction: Excessive intake of lactate caused by improper use of silage in animal husbandry has adverse effects on rumen fermentation, such as rumen acidosis. The speed of absorption and metabolism of D -lactate in rumen epithelial cells was slower than that of L -lactate, making D -lactate more prone to accumulate and induce rumen acidosis. Therefore, this study was conducted to explore the effects of dietary D -lactate levels on rumen fermentation of beef cattle and its mechanism in an in vitro system. Methods: This experiment was adopted in single-factor random trial design, with 5 days for adaptation and 3 days for sample collection. Three treatments ( n = 8/treatment) were used: (1) D-LA (0.3%), basal fermentation substrate with 0.3% (dry matter, DM basis) D -lactate; (2) D-LA (0.75%), basal fermentation substrate with 0.75% (DM basis) D -lactate; and (3) D-LA (1.2%), basal fermentation substrate with 1.2% (DM basis) D -lactate. Results: With the dietary D -lactate levels increased, the daily production of total gas, hydrogen and methane, as well as the ruminal concentrations of acetate, propionate, butyrate, isobutyrate, valerate, isovalerate, total volatile fatty acid and D -lactate increased ( p < 0.05), but the ruminal pH and acetate/propionate ratios decreased ( p < 0.05). Principle coordinate analysis based on Bray-Curtis distance showed that increasing dietary D -lactate levels could significantly affect the structure of rumen bacterial community ( p < 0.05), but had no significant effect on the structure of rumen eukaryotic community ( p > 0.05). NK4A214_group , Ruminococcus_gauvreauii_group , Eubacterium_oxidoreducens_group , Escherichia-Shigella , Marvinbryantia and Entodinium were enriched in D-LA (1.2%) group ( p < 0.05), as well as WCHB1-41 , vadinBE97 , Clostridium_sensu_stricto_1 , Anaeroplasma and Ruminococcus were enriched in D-LA (0.3%) group ( p < 0.05). Changes in the composition of ruminal microorganisms affected rumen metabolism, mainly focus on the biosynthesis of glycosaminoglycans ( p < 0.05). Discussion: Overall, feeding whole-plant corn silage with high D -lactate content could not induce rumen acidosis, and the metabolization of dietary D -lactate into volatile fatty acids increased the energy supply of beef cattle. However, it also increased the ruminal CH 4 emissions and the relative abundance of opportunistic pathogen Escherichia-Shigella in beef cattle. The relative abundance of Verrucomicrobiota and Escherichia-Shigella may be influenced by glycosaminoglycans, reflecting the interaction between rumen microorganisms and metabolites.

Abstract:A rumen protected fat product composed of Ca salts of fatty acids (CAF) was evaluated in beef cattle diets. Dietary treatments in a metabolism trial (six steers, 251 +/- 15 kg) and Feedlot Trial 1 (15 individually fed heifers, 355 +/- 11 kg) consisted of 62.1% corn and 3.2% soybean meal (C), 32.6% corn and 33.1% Kline barley (CB), and 28.2% corn, 33.1% barley, and 4.5% CAF (CBF) on a DM basis. Apparent DM and OM digestibilities were higher (P less than .05) and NDF digestibility was lower (P less than .05) for C than for CB and CBF. Apparent ether extract digestibility was higher (P less than .01) for CBF than for CB. Retention of N was higher (P less than .05) for C than for CB and CBF. On d 42, Feedlot Trial 1 ruminal fluid acetate:propionate ratio was lower (P less than .01) for C than for CB and CBF, and the ratio was lower (P less than .01) for CB than for CBF. Treatments did not affect (P less than .10) ADG and DMI of heifers, but feed/gain was lower (P less than .05) for C than for CB and CBF, and feed/gain was higher (P less than .05) for CBF than for CB. In Feedlot Trial 2, steers and heifers were fed the C diet or a 57% corn, 21.4% peanut hull, 4.1% soybean meal, and 4.5% CAF diet (CF) on an ad libitum basis for 50 d. Ruminal fluid acetate and propionate were unaffected (P greater than .10) by dietary treatments. Cattle ADG tended to be lower (P greater than .10), DMI was lower (P less than .05), and feed/gain tended to be improved (P greater than .10) for CF compared with C. Kline barley has potential as a feed grain for cattle, but CAF may not improve beef cattle performance, especially when diets contain barley.

Abstract:The objective of this study was to investigate effects of rumen-protected γ-aminobutyric acid (GABA) on dry matter intake, milk performance, and serum metabolites in Chinese Holstein lactating cows. Thirty-nine multiparous cows were blocked based on days in milk (60 ± 6.3 d; mean ± SD) and milk production (30.9 ± 4.17 kg; mean ± SD), and were randomly assigned to 1 of 4 treatments, with rumen-protected GABA added at levels of 0, 0.8, 1.6, or 2.4 g/d, the actual predicted available amounts being 0, 0.30, 0.61, or 0.91 g of GABA/d, respectively. The experiment lasted for 8 wk, with the first week for adaptation. Milk yield and milk compositions were recorded weekly, and serum concentrations of GABA, neuropeptide Y, and biochemical and antioxidant variables were analyzed in the first, fourth, and seventh weeks of the study. Dry matter intake linearly increased in cows receiving added GABA compared with that for the control. Addition of 0.8 g of GABA/d was associated with higher milk yield than the other treatments, but contents of milk protein and fat did not differ across the treatments. Dietary GABA tended to quadratically enhance the serum content of GABA (23.6, 30.2, 29.8, or 28.3 mmol/L for 0, 0.8, 1.6, or 2.4 g/d, respectively), and increased neuropeptide Y, with the highest value (3.07 ng/L) for 0.8 g of GABA/d. Nonesterified fatty acid quadratically decreased with GABA addition, with the lowest value (218.1 μmol/L) for 0.8 g of GABA/d. Serum glutathione peroxidase and superoxide dismutase quadratically increased in cows fed GABA, whereas serum malondialdehyde was quadratically reduced for all GABA groups. Rumen-protected GABA quadratically improved N efficiency across all treatments, contributing to the enhanced production of milk and milk protein and reduced N emission to the environment. In conclusion, addition of rumen-protected GABA is beneficial for early lactation dairy cows in terms of feed intake, lactation performance, and animal health.

Abstract:Background: The average daily gain (ADG) of preweaning calves significantly influences their adult productivity and reproductive performance. Gastrointestinal microbes are known to exert an impact on host phenotypes, including ADG. The aim of this study was to investigate the mechanisms by which gastrointestinal microbiome regulate ADG in preweaning calves and to further validate them by isolating ADG-associated rumen microbes in vitro. Results: Sixteen Holstein heifer calves were selected from a cohort with 106 calves and divided into higher ADG (HADG; n = 8) and lower ADG (LADG; n = 8) groups. On the day of weaning, samples of rumen contents, hindgut contents, and plasma were collected for rumen metagenomics, rumen metabolomics, hindgut metagenomics, hindgut metabolomics, and plasma metabolomics analyses. Subsequently, rumen contents of preweaning Holstein heifer calves from the same dairy farm were collected to isolate ADG-associated rumen microbes. The results showed that the rumen microbes, including Pyramidobacter sp. C12-8, Pyramidobacter sp. CG50-2, Pyramidobacter porci, unclassified_g_Pyramidobacter, Pyramidobacter piscolens, and Acidaminococcus fermentans, were enriched in the rumen of HADG calves (LDA > 2, P < 0.05). Enrichment of these microbes in HADG calves' rumen promoted carbohydrate degradation and volatile fatty acid production, increasing proportion of butyrate in the rumen and ultimately contributing to higher preweaning ADG in calves (P < 0.05). The presence of active carbohydrate degradation in the rumen was further suggested by the negative correlation of the rumen microbes P. piscolens, P. sp. C12-8 and unclassified_g_Pyramidobacter with the rumen metabolites D-fructose (R < - 0.50, P < 0.05). Widespread positive correlations were observed between rumen microbes (such as P. piscolens, P. porci, and A. fermentans) and beneficial plasma metabolites (such as 1-pyrroline-5-carboxylic acid and 4-fluoro-L-phenylalanine), which were subsequently positively associated with the growth rate of HADG calves (R > 0.50, P < 0.05). We succeeded in isolating a strain of A. fermentans from the rumen contents of preweaning calves and named it Acidaminococcus fermentans P41. The in vitro cultivation revealed its capability to produce butyrate. In vitro fermentation experiments demonstrated that the addition of A. fermentans P41 significantly increased the proportion of butyrate in the rumen fluid (P < 0.05). These results further validated our findings. The relative abundance of Bifidobacterium pseudolongum in the hindgut of HADG calves was negatively correlated with hindgut 4-hydroxyglucobrassicin levels, which were positively correlated with plasma 4-hydroxyglucobrassicin levels, and plasma 4-hydroxyglucobrassicin levels were positively correlated with ADG (P < 0.05). Conclusions: This study's findings unveil that rumen and hindgut microbes play distinctive roles in regulating the preweaning ADG of Holstein heifer calves. Additionally, the successful isolation of A. fermentans P41 not only validated our findings but also provided a valuable strain resource for modulating rumen microbes in preweaning calves. Comparison of rumen and abomasal infusions of an exogenous emulsifier on fatty acid digestibility of lactating dairy cows

Abstract:We evaluated the effects of infusing an exogenous emulsifier (polysorbates-C18:1) either into the rumen or abomasum on fatty acid (FA) digestibility and production responses of lactating dairy cows. Nine ruminally cannulated multiparous Holstein cows (170 ± 13.6 d in milk) were assigned to a treatment sequence in replicated 3 × 3 Latin squares with 18-d periods consisting of 7 d of washout and 11 d of infusion. Treatments were abomasal infusions of water carrier only into the rumen and abomasum (control, CON), 30 g/d polysorbate-C18:1 (T80) infused into the rumen (RUM), or 30 g/d T80 infused into the abomasum (ABO). Emulsifiers were dissolved in water and delivered at 6-h intervals (total daily infusion was divided into 4 equal infusions per day). Cows were fed the same diet that contained [% diet dry matter (DM)] 32.2% neutral detergent fiber (NDF), 16.1% crude protein, 26.5% starch, and 3.41% FA (including 1.96% FA from a saturated FA supplement containing 28.0% C16:0 and 54.6% C18:0). Two orthogonal contrasts were evaluated: (1) the overall effect of T80 {CON vs. average of the T80 infusions [1/2 (ABO + RUM)]}, and (2) the effect of ABO versus RUM infusion. Compared with CON, infusing T80 increased the digestibilities of NDF (2.85 percentage units), total (4.35 percentage units), 16-carbon (3.25 percentage units), and 18-carbon FA (4.60 percentage units), and tended to increase DM digestibility and total and 18-carbon FA absorption. Compared with RUM, ABO decreased the intakes of total (28 g/d), 16-carbon (7 g/d), and 18-carbon FA (19 g/d); tended to increase the digestibility of total and 18-carbon FA; and had no effect on the absorption of total, 16-carbon, or 18-carbon FA. Production responses did not change among our treatments. In conclusion, infusing 30 g/d polysorbates-C18:1 increased NDF and total, 16-carbon, and 18-carbon FA digestibility. Compared with RUM, ABO tended to increase the digestibilities of total and 18-carbon FA; however, this may be related to the fact that ABO reduced the intakes of total, 16-carbon, and 18-carbon FA, not necessarily due to better emulsifying action per se. In summary, ABO and RUM both improved FA absorption.
