Abstract:Herein we report a highly specific electrochemical aptasenseor for AFB1 determination based on AFB1-controlled diffusion of redox probe (Ru(NH3)63+) through nanochannels of AFB1-specific aptamer functionalized VMSF. A high density of silanol groups on the inner surface confers VMSF with cationic permselectivity, enabling electrostatic preconcentration of Ru(NH3)63+ and producing amplified electrochemical signals. Upon the addition of AFB1, the specific interaction between the aptamer and AFB1 occurs and generates steric hindrance effect on the access of Ru(NH3)63+, finally resulting in the reduced electrochemical responses and allowing the quantitative determination of AFB1. The proposed electrochemical aptasensor shows excellent detection performance in the range of 3 pg/mL to 3 μg/mL with a low detection limit of 2.3 pg/mL for AFB1 detection. Practical analysis of AFB1 in peanut and corn samples is also accomplished with satisfactory results by our fabricated electrochemical aptasensor.

Abstract:A rapid and label free aflatoxin B1 (AFB1) microfluid sensor was proposed and tested. The device was fabricated with hollow-core photonics crystal fiber infiltrated with the AFB1 solution. The autofluorescence emitting from the AFB1 molecules was detected. The sensor length was optimized. The AFB1 concentration was tested with a 4 cm long sensor. The best limit of detection was achieved as low as 1.34 ng/ml, which meets the test requirement of the national standards for AFB1 in food. The effectiveness of this sensor being applied in beer solution was also verified to be a little more sensitive than in aqueous solution. Compared with traditional AFB1 detection methods, the proposed single-ended device perfectly satisfies the demand of process control in alcoholic beverages manufacture.

Abstract:Rapid and accurate analysis of food draws considerable attention in the modern pace of the world due to the close relationship between human health and food safety. Traditional detection technologies for food evaluation are often restricted by high cost, lengthy time, bulky instruments and trained personnel. The marriage of biosensors with smartphones enables the development of powerful analysis platforms for food evaluation including detection of food contaminants, toxins, pathogens, allergens and nutrition. Here, we provide an overview of recent developments on smartphone-based biosensors for portable food evaluation. Owing to operability, connectivity, portability and built-in sensors, smartphones have become ideal control, interaction and analysis tools in the on-site sensing systems. Fusion of smartphones with different kinds of sensitive and selective biosensors enables to develop portable and user-friendly analytical devices. In addition to introducing technical principles, detection methods and selected applications in food science, challenges and future perspectives for smartphone-based biosensors are also discussed.

Abstract:Rapid, high-frequency, and accurate identification of aflatoxin B1 (AFB1) is crucial for ensuring food safety and reducing population mortality. Herein, we constructed S martphone powered M obile m I crofluidic L ab-on-fiber d E vice ( SMILE ) comprising a compact optical system, fiber nano-bioprobe-embedded microfluidic-chip system, mini-photodetector, and software application to facilitate the rapid and sensitive point-of-need quantitative testing for AFB1. The elegant optical design of SMILE significantly improves light transmission efficiency, detection sensitivity, and portability by integrating a compacted all-fiber optical structure with a fiber nano-bioprobe-embedded microfluidic chip. Furthermore, the nanopore layer of the fiber nano-bioprobe improves detection sensitivity by increasing the biorecognition molecule number and enhancing the interaction between the evanescent field and dye. Through an indirect competitive immunoassay mechanism, SMILE achieves sensitive quantitative detection of AFB1 with a detection limit of 0.08 μg/L. Herein, SMILE was validated using several feedstuff samples tested with a simple aqueous extraction protocol, demonstrating good correlation with high-performance liquid chromatography for AFB1-contaminated feedstuffs. The immunoassay process is completed within 12 min, boasting high sensitivity, specificity, reusability, and reproducibility. Owing to its sensitivity, portability, flexibility, plug-and-play, and smartphone integration, SMILE is highly scalable for rapid and high-frequency point-of-need testing for AFB1 and other trace contaminants.

Abstract:The development of dual-mode strategies with superior sensitivity and accuracy have garnered increasing attention for researchers in Aflatoxin B1 (AFB1) analysis. Herein, a colorimetric-electrochemiluminescence (ECL) dual-mode biosensor was constructed for onsite and ultrasensitive determination of AFB1. The multi-wall carbon nanotubes (MWCNTs) were integrated with the ZnO metal organic frameworks (MOFs) to accelerate the electron transfer and boost the ECL intensity of g-C3N4 nanoemitters. Through the aptamer-based DNA sandwich assay, the CuO@CuPt nanocomposites were introduced onto the electrode and acted as the dual functional signal nanoprobes. Due to the good spectrum overlap between the CuO@CuPt nanoprobes and g-C3N4 nanosheets, ECL signal could be efficiently quenched. Additionally, the CuO@CuPt nanoprobes show superior catalytic properties towards the TMB and H2O2 colorimetric reactions, and an obvious color alteration from colorless to blue can be observed using the smartphone. Under optimized conditions, a sensitive and accurate dual-mode analysis of the AFB1 was accomplished with the colorimetric detection limit of 3.26 fg/mL and ECL detection limit of 0.971 fg/mL (S/N = 3). This study combines innovative nanomaterial properties of ZnO@MWCNTs, g-C3N4 and CuO@CuPt for ultrasensitive dual-mode detection, which offers new opportunities for the innovative engineering of the dual-mode sensors and demonstrates significant potential in food safety analysis.

Detection is possible via a self-made poly(methyl methacrylate) pipette, combined with a smartphone to further increase the biosensor’s portability. The combination of the two greatly simplifies experimental operations, reduces testing costs, and can minimize matrix interferences through sample pre-treatment operations. And the smartphone-APP signal readout strategy can avoid the need for a reading instrument, making it more suitable for field testing. The FSCC was developed by fully utilizing the properties of a Fe-Zr-metal–organic framework (Fe-Zr-MOF), which exhibits high catalytic activity and enables the encapsulation of abundant horseradish peroxidase in its pores, the in-situ reduction and enrichment of Pt nanoparticles on its surface, and the coordination of biotinylated DNA to achieve a high adsorption capacity of streptavidin-horseradish peroxidase. This ensured detection sensitivity, with a limit of detection = 1.57 pg/mL. The developed biosensor displays notable application potential, especially in resource-poor areas due to the advantages of low costs, simple operation, ultra-high sensitivity, portability, etc.

Abstract:This study focuses on the integration of a custom-built and optimally trained YOLO v5 model into a smartphone app developed with Java language. A dual-modal immunochromatographic rapid detection system based on a deep learning strategy for smartphones was developed for grade determination and predicting the concentration of aflatoxin B1 (AFB1). Innovative distance-type quantum dot microsphere fluorescent immunochromatographic chips enable semi-quantitative analysis by naked eye, and conventional colloidal gold nanoparticle colorimetric strips were also prepared. The compact and versatile hardware device making it easily integrable into smartphones of varying dimensions. Moreover, the wireless charging functionality of smartphones was to tackle power supply challenges. After optimized training, the accuracy, , precision, and recall metrics of the YOLO v5 model all soared to 98 %. For the dual-modal immunochromatographic chips, the R 2 values for the standard curve fits were as high as 0.993, with a broad linear range of 0.05–40 ng/mL and a standard deviation lower than 0.03 at each concentration. Finally, this system determined the grade of the AFB1 concentration with an accuracy of up to 98 % and it exhibited an ultra-sensitive quantitative detection capability with a limit of detection as low as 2.2 pg/mL, showcasing the reliability of the deep learning strategy for practical applications in smartphones. This robust technological foundation paves the way for potentially community-based, family-oriented, and personalized applications.

Abstract:Aflatoxin B1 (AFB1) is one of the most toxic mycotoxins. In view of the great harm of AFB1, it has attracted much attention to explore a super-sensitive and low-cost rapid detection method for aflatoxin B1. Herein, a dual-readout colorimetric immunoassay was established for the detection of AFB1. Under the catalysis of alkaline phosphatase (ALP), phenyl phosphate (PP) can produce phenol by dephosphorylation. Then phenol can react with 2,6-dibromoquinone-4-chloroimide (DBQC) under alkaline conditions to produce blue halogenated indophenol (HIP). Through this reaction, the color of the solution became blue, which can be observed by the naked eye. In addition, this color signal can be converted into digital signal via smartphone, enabling quantitative detection of AFB1. Under the optimal condition, the detection limit of this colorimetric immunoassay was 0.70 ng/mL by naked eye and 0.013 ng/mL by smartphone. The application results showed that the proposed daul-readout colorimetric immunoassay was feasible and real-time for AFB1 detection in actual samples.

Abstract:The fabrication of bioreceptor-free method for accurate and sensitive detection of ochratoxin A (OTA) in cereal is critical, but still a significant challenge to mitigate risks to food industries and public health. In this study, a smartphone-ratiometric fluorescence sensor for the ultrasensitive detection of OTA is developed based on a porphyrinic metal-organic framework and silica nanoparticle composite (Zr-MOF/SiNPs) away from the use of antibodies and aptamers. Due to the excellent recognition ability of Zr-MOF and good storage stability of SiNPs, OTA is detected by Zr-MOF/SiNPs with a wide linear range of 0.05–1000 ng/mL and low detection limit of 0.016 ng/mL. Moreover, the red–blue ratio values of the fluorescence images are extracted through the smartphone color recognizer application with a limit of detection of 1.74 ng/mL, lower than the permissible content of OTA in cereal prescribed by World Health Organization. This sensing platform has been successfully applied in maize samples with superior repeatability and satisfactory recoveries, providing a novel way for simple and label-free analysis of OTA in cereal.

Abstract:Mycotoxin contaminations in food and environment seriously harms human health. Constructing sensitive and point-of-test early-warning tools for mycotoxin determination is in high demand. In this study, a CuCo@PDA nanozyme-based aptamer-mediated lateral flow assay (Apt-LFA) has been elaborately designed for on-site and sensitive determination of mycotoxin Aflatoxin B1 (AFB1). Benefiting from the rich functional groups and excellent peroxidase-like activity, the CuCo@PDA with original dark color can be conjugated with the specific recognition probe (i.e., aptamer), generating colorimetric signal on the test lines of Apt-LFA via a competitive sensing strategy. The signal can further be amplified in-situ by catalytic chromogenic reaction. Therefore, a visual and dual-readout detection of AFB1 has been realized. The developed Apt-LFA provides a flexible detection mode for qualitative and quantitative analysis of AFB1 by naked-eyes observation or smartphone readout. The smartphone-based LFA platform shows a reliable and ultrasensitive determination of AFB1 with the limit of detection (LOD) of 2.2 pg/mL. The recoveries in the real samples are in the range of 95.11-113.77% with coefficients of variations less than 9.84%. This study provides a new approach to realize point-of-test and sensitive detection of mycotoxins in food and environment using nanozyme-based Apt-LFAs.

Abstract:Aflatoxin B 1 (AFB 1 ) is one of the most harmful mycotoxins. Convenient and reliable determination of trace AFB 1 is highly desired. Herein, dual hemin labeled DNA hairpin was designed as a chemiluminescence (CL) switch and combined with aptamer of AFB 1 and its partial complementary strand for homogenous assay of AFB 1 . Due to the judicious design of three DNA sequences and the help of nicking enzyme, the AFB 1 recognition, signal transduction and recycling amplification can be implemented during one-step reaction. The whole protocol only involved the simple mixing of samples with homogeneous solution, avoiding the preparation of sophisticated solid-phase sensor and eliminating the multi-step washing and resulting errors, which provided a unique tool for on-site rapid testing. Importantly, when the operation process was simplified, the detection sensitivity can be enhanced facilely due to the enzyme recycling. Under optimal conditions, the intelligent CL system for sensing AFB 1 exhibited a wide dynamic range (1.0 × 10 -2 ∼ 100 ng mL −1 ) with a low detection limit (down to 1.5 pg mL −1 ), suitable for detecting trace AFB 1 in complex matrix. The methodology was validated in coix seed and decoction pieces. The superiority including free of washing and separation, high sensitivity, acceptable accuracy, and greenness, showed good applicability of the proposed CL system in safety evaluation of food and medicine.

Abstract:Aflatoxin B1 (AFB1) contamination severely threatens human and animal health, it is thus critical to construct a strategy for its rapid, accurate, and visual detection. Herein, a multimodal biosensor was proposed based on CRISPR/Cas12a cleaved G-quadruplex (G4) for AFB1 detection. Briefly, specific binding of AFB1 to the aptamer occupied the binding site of the complementary DNA (cDNA), and cDNA then activated Cas12a to cleave G4 into fragments. Meanwhile, the intact G4-DNAzyme could catalyze 3, 3', 5, 5'-tetramethylbenzidine (TMB) to form colourimetric/SERS/fluorescent signal-enhanced TMBox, and the yellow solution produced by TMBox under acidic conditions could be integrated with a smartphone application for visual detection. The colourimetric/SERS/fluorescent biosensor yielded detection limits of 0.85, 0.79, and 1.65 pg·mL<sup>-1</sup>, respectively, and was applied for detecting AFB1 in peanut, maize, and badam samples. The method is suitable for visual detection in naturally contaminated peanut samples and has prospective applications in the food industry.

Abstract:The deficiency in rapid and in-field detection methods and portable devices that are reliable, easy-to-use, and low cost, results in the difficulties to uphold the high safety standards in China. In this study, we introduce a rapid and cost-effective smartphone-based method for point-of-need food safety inspection, which employs aptamer-conjugated AuNPs as the colorimetric indicator, and a battery-powered optosensing accessory attached to the camera of a smartphone for transmission images capture. A user-friendly and easy-to-use Android application is developed for automatic digital image processing and result reporting. Streptomycin (STR) is selected as the proof-of-concept target, and its specific quantitation can be realized with a LOD of 12.3 nM (8.97 μg kg-1) using the reported smartphone-based method. The quantitation of STR in honey, milk and tap water confirm the reliability and applicability of the reported method. The extremely high acceptance of smartphone in remote and metropolitan areas of China and ease-of-use of the reported method facilitate active food contaminant and toxicant screening, thus making the implementation of the whole food supply chain monitoring and surveillance possible and hence significantly improving the current Chinese food safety control system.

Abstract:It is necessary to develop accurate and sensitive AFB1 detection method due to its high toxicity of pathogenicity and carcinogenicity. The results of traditional single-mode AFB1 detection methods are easily affected by the test environment and human operation. Herein, an electrochemical and fluorescent dual-mode separated aptasensor was developed for detecting AFB1 with triple-signal readout. The AFB1 aptasensor was constructed by combining the Fc-labeled AFB1 aptamer with its complementary DNA (cDNA). Subsequently, the methylene blue (MB) was incorporated into dsDNA as another signal tag. In the presence of AFB1, the Fc tag approached to electrode surface, both of the cDNA and MB molecule released due to the interaction between AFB1 and its aptamer, resulting in "signal-on" and "signal-off" dual-signal strategy. There was a good linear relationship between the SWV electrochemical response signal and the logarithm of AFB1 concentration at 0.01–200 ng/mL (LOD 2.5 pg/mL, S/N = 3). Meanwhile, the released cDNA was designed to trigger hybridization chain reaction (HCR), and the HCR product was served as template for the formation of fluorescent copper nanoparticles (CuNPs). There was a good linear relationship between the fluorescent intensity of CuNPs and the logarithm of AFB1 concentration at 0.001–200 ng/mL (LOD 0.28 pg/mL, S/N = 3). The separated fluorescent method showed broadened linear range and lower LOD compared with SWV method, both up to 10-folds, which improved the detection sensitivity. The constructed dual-mode AFB1 aptasensor showed satisfactory selectivity, stability and reproducibility, and the good recoveries of real peanut samples indicated its practical application in food detection.

Specifically, aminated mesoporous silica nanoparticles (MSNs) were used as nanocontainers to load the Rhodamine 6G (R6G) with fluorescence and Raman effects, aptamers and polydopamine (PDA) were sequentially assembled on the surface of the MSNs as dual-gated molecules, and MXenes and Au nanorods were used as SERS substrates. After adding AFB1 under acidic conditions, the degradation of PDA and specific bringing of the aptamer with AFB1 resulted in the release of R6G. Results showed that AFB1 could be quantified by Raman and fluorescence spectroscopic with detection limits of 0.133 pg mL −1 and 0.214 pg mL −1 , respectively. Moreover, the biosensor could be applied to the detection of AFB1 in real peanut, maize and badam samples, providing a possible alternative solution for the detection of other analytes.

Abstract:This work describes a novel "off-on" biosensor for super-sensitive and selective detection of aflatoxin B1 (AFB1). In the biosensor, electrochemiluminescence (ECL) and aggregation-induced emission (AIE) work together to enhance ECL performance by a dibenzo-[b,d]thiophene sulfone-based conjugated microporous polymer, and hairpin DNA1 (H1) and ferrocene (Fc)-apt act as the switches for the "off-on" mode. In the absence of target AFB1, the ECL signal is inhibited because of the steric-hindrance effect, and the biosensor remains in the "off" state. In the presence of AFB1, ECL signal recovery is observed caused by the specific recognition between the AFB1 and Fc-apt, i.e., the biosensor changes to the "on" state. Under optimal experimental conditions, AFB1 can be quantified from 1 pg·mL −1 to 1 μg·mL −1 with an ultralow detection limit of 0.46 pg·mL −1 by the "off-on" biosensor. The biosensor exhibited good practicability for AFB1 assay in corn samples, thus providing a simple and sensitive method for monitoring food safety.

Abstract:Here, based on the characteristics of Graphene oxide(GO) and SYBR Green I(SGI) dye, an enzyme-free and label-free fluorescent biosensor with signal amplification through DNA strand reaction is proposed for the detection of Aflatoxin B1(AFB1) in food safety. Firstly, without the addition of AFB1, the substrate in the system includes a double stranded Apt-S with a long sticky end and two hairpins H1 and H2. Although the complementary pairing of bases may exhibit fluorescence due to the insertion of SGI dyes, the use of GO, which is highly capable of adsorbing single stranded parts and quenching fluorescence, cleverly reduces the background fluorescence. Adding the target AFB1 triggers DNA inter chain reactions, generating a large amount of long double stranded DNA H1-H2, thereby generating strong fluorescence signals under the action of SGI. More importantly, logical theory verification and computer simulation were conducted before biological experiments, providing a theoretical basis for the implementation of the biosensor. After analysis, the fluorescence biosensor exhibits a good linear relationship with AFB1 concentration in the range of 5-50nM, with a detection limit of 0.76nM. It also has good specificity, anti-interference ability, and practical application ability, and has broad application prospects in the field of food safety.
