Abstract:A large quantity of orange peel waste (OPW) is generated per year, yet effective biorefinery methods are lacking. In this study, Trichosporonoides oedocephalis ATCC 16958 was employed for hydrolyzing OPW to produce soluble sugars. Glycosyl hydrolases from Paenibacillus sp. LLZ1 which can hydrolyze cellulose and hemicellulose were mined and characterized, with the highest β-mannanase activity of 39.1 U/mg at pH 6.0 and 50 °C. The enzyme was overexpressed in T. oedocephalis and the sugar production was enhanced by 16 %. The accumulated sugar contains 57 % value-added mannooligosaccharides by the hydrolysis of mannans. The process was intensified by a pretreatment combining H 2 O 2 submergence and steam explosion to remove potential inhibitors. The mannooligosaccharides yield of 6.5 g/L was achieved in flask conversion and increased to 9.7 g/L in a 5-L fermenter. This study improved the effectiveness of orange peel waste processing, and provided a hydrolysis-based methodology for the utilization of fruit wastes.

Abstract:To maximize the reuse of citrus fruit processing waste, which is generated in large amounts due to juice production, this work shows the results of a new renewable approach for the extraction of high‐added‐value compounds present in orange biomass, specifically in orange peels (OP). The method is based on the use of diluted Ca2+, acid, and base solutions for the extraction of hesperidin (HSD), pectin, lignin, and cellulose sequentially, without generating any organic solid waste at the end, and without the need for air drying the starting biomass. Good yields of hesperidin were recovered (~1.56%), with excellent purity (up to 95%), as well as lignin (yield ~2.2%) and cellulose (yield ~12%). Pectin obtained by this process showed a high amount of galacturonic acid (~88%) and a low degree of methylation (~16%). All products were thoroughly characterized by applying Fourier transform infrared (FTIR) spectroscopy and solid‐state cross‐polarization/magic angle spinning carbon‐13 nuclear magnetic resonance spectroscopy (CP/MAS 13C‐NMR). The products recovered from OP can be applied in a wide array of fields, such as food, medicine, and pharmaceuticals, among others.

Ultrasound-assisted dilute acid hydrolysis for production of essential oils, pectin and bacterial cellulose via a citrus processing waste biorefinery

Abstract:An orange peel waste biorefinery was developed employing a design of experiments approach to optimize the ultrasound-assisted dilute acid hydrolysis process applied for production of useful commodities. Central composite design-based response surface methodology was used to approximate the combined effects of process parameters in simultaneous production of essential oils, pectin and a sugar-rich hydrolyzate. Application of a desirability function determined the optimal conditions required for maximal production efficiency of essential oils, pectin and sugars as 5.75% solid loading, 1.21% acid concentration and 34.2 min duration. Maximum production yields of 0.12% w/w essential oils, 45% w/w pectin and 40% w/w sugars were achieved under optimized conditions in lab- and pilot-scale facilities. The hydrolyzate formed was applied in bacterial cellulose fermentations producing 5.82 g biopolymer per 100 g waste. Design of experiments was efficient for process analysis and optimization providing a systems platform for the study of biomass-based biorefineries.
Valorization of orange peels exploiting fungal solid-state and lacto-fermentation

Abstract:Background: Orange peels can serve as a cost-effective raw material for the production of lactic acid. Indeed, given their high concentration of carbohydrates and low content of lignin, they represent an important source of fermentable sugars, recoverable after a hydrolytic step. Results: In the present article, the fermented solid, obtained after 5 days of Aspergillus awamori growth, was used as the only source of enzymes, mainly composed of xylanase (40.6 IU g-1 of dried washed orange peels) and exo-polygalacturonase (16.3 IU g-1 of dried washed orange peels) activities. After the hydrolysis, the highest concentration of reducing sugars (24.4 g L-1 ) was achieved with 20% fermented and 80% non-fermented orange peels. The hydrolysate was fermented with three lactic acid bacteria strains (Lacticaseibacillus casei 2246 and 2240 and Lacticaseibacillus rhamnosus 1019) which demonstrated good growth ability. The yeast extract supplementation increased the lactic acid production rate and yield. Overall, L. casei 2246 produced the highest concentration of lactic acid in mono-culture. Conclusion: To the best of our knowledge this is the first study exploiting orange peels as low-cost raw material for the production of lactic acid avoiding the employment of commercial enzymes. The enzymes necessary for the hydrolyses were directly produced during A. awamori fermentation and the reducing sugars obtained were fermented for lactic acid production. Despite this preliminary work carried out to study the feasibility of this approach, the concentrations of reducing sugars and lactic acid produced were encouraging, leaving open the possibility of other studies for the optimization of the strategy proposed here. © 2023 The Authors. Functional use of carbon dioxide for the sustainable valorization of orange peel in the pyrolysis process

Abstract:Although biomass is carbon-neutral, its use as a primary feedstock faces challenges arising from inconsistent supply chains. Therefore, it becomes crucial to explore alternatives with reliable availability. This study proposes a strategic approach for the thermochemical valorization of food processing waste, which is abundantly generated at single sites within large-scale processing plants. As a model biomass waste from the food industry, orange peel waste was particularly chosen considering its substantial consumption. To impart sustainability to the pyrolysis system, CO2, a key greenhouse gas, was introduced. As such, this study highlights elucidating the functionality of CO2 as a reactive feedstock. Specifically, CO2 has the potential to react with volatile pyrolysates evolved from orange peel waste, leading to CO formation at ≥490 °C. The formation of chemical constituents, encompassing acids, ketones, furans, phenols, and aromatics, simultaneously decreased by 15.1 area% in the presence of CO2. To activate the efficacy of CO2 at the broader temperature spectrum, supplementary measures, such as an additional heating element (700 °C) and a nickel-based catalyst (Ni/Al2O3), were implemented. These configurations promote thermal cracking of the volatiles and their reaction kinetics with CO2, representing an opportunity for enhanced carbon utilization in the form of CO. Finally, the integrated process of CO2-assisted catalytic pyrolysis and water-gas shift reaction was proposed. A potential revenue when maximizing the productivity of H2 was estimated as 2.62 billion USD, equivalent to 1.11 times higher than the results from the inert (N2) environment. Therefore, utilizing CO2 in the pyrolysis system creates a promising approach for enhancing the sustainability of the thermochemical valorization platform while maximizing carbon utilization in the form of CO.

Abstract:It is widely recognised that orange peels contain a considerable quantity of phenolics, primarily in the form of glycosides. The process of fermentation holds potential as a viable method for extracting phenolic compounds and facilitating their biotransformation into novel metabolites. The aim of this study was to assess the enhanced release of phenolic compounds through the process of solid-state fermentation of orange peels using microorganisms. Following a 6-day incubation period, the methanol extract obtained from the sample fermented with starter Banh men exhibited the highest concentration of total phenolic compounds (17.57 ± 0.34 mg GAE/g DW) and demonstrated the most significant DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity (55.03%). The Reverse Phase High Performance Liquid Chromatography (RP-HPLC) analysis revealed that the predominant phenolic compounds in all fermented samples were flavonoid aglycones, specifically naringenin, hesperetin, and nobiletin. Conversely, in the unfermented orange peels, the major compound observed was the glycoside derivative hesperidin.

Abstract:The residual microbes and uncertain volatile fatty acids (VFAs) components in anaerobic fermentation hydrolysate cause microbial contamination and multi-type polyhydroxyalkanoates (PHA) generation in pure microbial synthesis systems. Meanwhile, the regulation of biosynthesis of poly(3-hydroxybuturate- co -3-hydroxyvalerate) (PHBV), one type of excellent PHA, by re-utilization of waste-organics as carbon substrates is still elusive. In this study, four pretreatment methods for sludge hydrolysate containing 4746 mg-COD·L −1 of VFAs were established to assess the accumulation of PHBV by using Paracoccus sp. TOH as the chassis. Results showed that the autoclaving system exhibited an optimal PHBV concentration of 202.9 mg L −1 with the highest relative abundance of Paracoccus sp. TOH (92.4%). Valeric acid was identified as the primary substrate for 3-hydroxyvalerate accumulation, and the highest PHBV production of 3.76 g L −1 was achieved using 3 g L −1 valeric acid and 5 g L −1 glycerol as co-substrates. The increased carbon metabolism flows through the sequential action of acyl-coenzyme A dehydrogenase and enoyl-coenzyme A hydratase in the fatty acid β-oxidation pathway facilitated PHBV accumulation. This study provides a blueprint for the microbial synthesis of PHBV with oriented acid production from organic wastes.

Waste and Biomass Valorization

Abstract:Fruit peels, in general, are especially rich in sugar and bioactive compounds, including pigments, polyphenols, enzymes, vitamins, antioxidants, and essential oils. Often, the full potential of these peels is not used, and they end up being discarded, generating a large volume of waste that could cause environmental problems. Thinking about taking advantage of all the potential that fruit peels can offer, this study aims to extract D-limonene and also hydrolyze the sugars present in the waste mix of fruit peels (orange, banana, mango, pineapple, and lemon) for the production of bioethanol. D-limonene extraction was done through Soxhlet, an added-value compound with diverse environmental and chemical applications. Using the biomass hydrolysis methodology from the fruit mix, it is possible to obtain fermentable sugars to obtain bioethanol. Enzymatic hydrolysis requires optimization to get a high yield of sugars; for this, we use a Central Composite Rotational Design 2 2 (CCRD), with variables being the mass of the fruit peel mix and the enzymatic concentration, followed by fermentation with the yeast Wickerhamomyces sp. UFFSCE-3.1.2 for the production of bioethanol. The results demonstrated that optimizing enzymatic hydrolysis makes obtaining a satisfactory amount of sugars and bioethanol possible. Through experimental planning, we received a maximum of 63.90 g/L of total reducing sugars in enzymatic hydrolysis, followed by 7.14 g/L of bioethanol in the fermentation stage. D-limonene recovery from the fruit mix yielded 6.41 mg/g of fruit waste, the expectation that global demand for essential oils will rise 7.5% by 2027, which adds even more value to this compound.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Biotechnological processes are part of modern industry as well as stricter environmental requirements. The need to reduce production costs and pollution demands for alternatives that involve the integral use of agro-industrial waste to produce bioactive compounds. The citrus industry generates large amounts of wastes due to the destruction of the fruits by microorganisms and insects together with the large amounts of orange waste generated during the production of juice and for sale fresh. The aim of this study was used orange wastes rich in polyphenolic compounds can be used as source carbon of <em>Aspergillus fumigatus</em> MUM 1603 to generate high added value compounds, for example, ellagic acid and other molecules of polyphenolic origin through submerged fermentation system.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The orange peel waste had a high concentration of polyphenols, 28% being condensed, 27% ellagitannins, 25% flavonoids and 20% gallotannins. The major polyphenolic compounds were catechin, EA and quercetin. The conditions, using an experimental design of central compounds, that allow the production of the maximum concentration of EA (18.68 mg/g) were found to be: temperature 30°C, inoculum 2 × 10<sup>7</sup> (spores/g) and orange peel polyphenols 6.2 (g/L).</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>The submerged fermentation process is an effective methodology for the biotransformation of molecules present in orange waste to obtain high value-added as ellagic acid that can be used as powerful antioxidants, antibacterial and other applications.</p>

Abstract:Agroindustrial raw materials (AIRM) especially fruit peels are rich as nutrient source which can be recycled using bioprocesses yielding important value added products. The recycling has an added benefit of ecofriendly decomposition of the waste material which otherwise generate environmental hazards. Solid state fermentations (SSF) are gaining importance as a method for valorization of the AIRM for enzyme production. The presence of lignocellulosic content requires hydrolysis which offer resistance and pose a difficulty in its easy fermentative biotransformations. The present study aims to study the physiochemical properties of the AIRM wastes (Pomegranate, Orange, Pineapple, Sweet lime and Banana peels) which play a critical role in the valorization susceptibility and can enhance the rate of growth accompanied by increased yield. The Water Absorption Index (WAI) and apparent density (AD) were analyzed by determining the moist weight of the AIRM. The growth rate was determined spectrophotometrically accompanied by xylanase production in SSF. One way ANOVA and Correlation analysis was performed between the investigated parameters. Amongst the ten isolates screened, isolate seven identified as Aspergillus flavus NFCCI4565 exhibited highest xylanase production (46.61 ± 0.008) IU/ml on day 5 of fermentation. Growth kinetics studies of the mold revealed maximum growth on pomegranate peel followed by orange peel. The pomegranate peel revealed minimum WAI (3.04 gel/g dry wt) and maximum apparent density 4.86 cm3 complemented with highest xylanase production of 80.2 IU/ml amongst all the invested peels. The features also supported maximum biomass of the Aspergillus flavus NFCCI4565. A positive correlation was found between the AD and Biomass, AD and Xylanase production. A negative correlation was detected between WAI and Biomass, WAI and Xylanase production. The selection of an appropriate solid substrate plays an important role in the development of an efficient SSF process. The results provided important knowledge about the characteristics of these materials revealing their potential for use in fermentation processes.Graphical Abstract

Abstract:Citrus peels are an ideal feedstock for their use in integrated, resource focused biorefinery activity for the production of high-value bio-chemicals, bio-fuels and bio-materials. Pectin and D-limonene, can be produced together with a rare form of mesoporous cellulose in a single step, without use of added acid, using a low temperature microwave hydrothermal processing of orange peel. A process temperature change enables the conversion of D-limonene to alpha-terpineol, showing that this process not only enables the separation of the major components but also adds further value to the citrus industry through the production of high-value marketable products.
Effect of the concentration of essential oil on orange peel waste biomethanization: Preliminary batch results

Abstract:The cultivation of orange (Citrus×sinensis) and its transformation is a major industry in many countries in the world, it leads to the production of about 25-30Mt of orange peel waste (OPW) per year. Until now many options have been proposed for the management of OPW but although they are technically feasible, in many cases their economic/environmental sustainability is questionable. This paper analyse at lab scale the possibility of using OPW as a substrate for anaerobic digestion. Specific objectives are testing the possible codigestion with municipal biowaste, verifying the effect on methane production of increasingly high concentration of orange essential oil (EO, that is well known to have antioxidant properties that can slower or either inhibit biomass activity) and obtaining information on the behaviour of d-limonene, the main EO component, during anaerobic digestion. The results indicate that OPW can produce up to about 370LnCH4/kgVS in mesophilic conditions and up to about 300LnCH4/kgVS in thermophilic conditions. The presence of increasingly high concentrations of EO temporary inhibits methanogenesis, but according to the results of batch tests, methane production restarts while d-limonene is partially degraded through a pathway that requires its conversion into p-cymene as the main intermediate.

Abstract:In-depth proteome discovery analysis represents new strategy in an effort to identify novel reliable specific protein markers for hypertrophic cardiomyopathy and other life threatening cardiovascular diseases. To systematically identify novel protein biomarkers of cardiovascular diseases with high mortality we employed an isobaric tag for relative and absolute quantitation (iTRAQ) proteome technology to make comparative analysis of plasma samples obtained from patients suffering from non-obstructive hypertrophic cardiomyopathy, stable dilated cardiomyopathy, aortic valve stenosis, chronic stable coronary artery disease and stable arterial hypertension. We found 128 plasma proteins whose abundances were uniquely regulated among the analyzed cardiovascular pathologies. 49 of them have not been described yet. Additionally, application of statistical exploratory analyses of the measured protein profiles indicated the relationship in pathophysiology of the examined cardiovascular pathologies.

Abstract:Orange peel waste (OPW), a discarded part of orange fruit, is a rich source of essential constituents that can be transformed into highly value-added bioproducts. OPW is being generated in million tonnes globally and returns to the environment without complete benefit. Thus, a high volume of annually produced OPW in the industry requires effective valorization. In this regard, limited data is available that summarizes the broader spectrum for the sustainable fate of OPW to produce value-added bioproducts. The main objective of this treatise is to explore the sustainable production of bioproducts from OPW. Therefore, this review covers all the aspects of OPW, from its production to complete valorization. The review encompasses the extraction technologies employed for extracting different valuable bioactive compounds, such as: essential oil (EO), pectin, and carotenoids, from OPW. Furthermore, the suitability of bioconversion technologies (digestion/fermentation) in transforming OPW to other useful bioproducts, such as: biochemicals (lactic acid and succinic acid), biopolysaccharides (xanthan and curdlan gum), and bioenergy (biomethane and bioethanol) is discussed. Also, it includes the concept of OPW-based biorefineries and their development that shall play a definite role in future to cover demands for: food, chemicals, materials, fuels, power, and heat. Lastly, this review focuses on OPW-supplemented functional food products such as: beverages, yogurts, and extruded products. In conclusion, insights provided in this review maximize the potential of OPW for commercial purposes, leading to a safe, and waste-free environment.

Integrated biorefinery for xylooligosaccharides, pectin, and bioenergy production from orange waste

Abstract:Orange waste is one of the most underutilized biowastes. It accounts for 50% of the fruit weight after juice extraction, presenting an environmental problem due to inappropriate disposal. Industrial waste exploitation is a sustainable way to obtain bioproducts and bioenergy. However, the efficiency of chemical treatments and enzymatic cocktails is still a bottleneck in these processes. One potential route to reduce costs is to design consecutive extraction and hydrolysis stages to yield more products from different fractions of the same feedstock, reducing the use of time and bulk materials. This study assessed the economic performance of an orange processing waste (OPW) biorefinery for xylooligosaccharides (XOS), pectin, and bioenergy, involving three aspects: (1) experimental procedures, (2) a simulation approach, and (3) optimization tools. The optimization of hydrothermal treatment for pectin extraction decreased material costs by more than 60% and preserved the xylan content for xylooligosaccharide production by enzymatic hydrolysis. This study analyzed different biorefinery capacities, and solid waste exploitation for heat and steam generation, improving sustainability. The production cost per kg of high‐purity pectin and xylooligosaccharides is 3.67 $ with an efficient solvent recovery and self‐sufficiency for standard power usage. These results proved the economic feasibility and low environmental impact of the OPW biorefinery.

Nevertheless, the step towards real process implementation must overcome a series of stages based on process sustainability in the early design stages. Orange peel (OP) has been profiled as a potential raw material for producing different products. Few studies have estimated the sustainability of OP-based biorefineries considering the upstream influence on the final process performance. This research aims to perform the sustainability assessment of several OP valorization pathways based on experimental data applying the biorefinery concept. Steam distillation and polyphenolic compound extraction prior to saccharification and anaerobic digestion increase the process performance. A glucose concentration and biogas yield of 21.43 g/L (0.44 g/g OP, db) and 415 mL/g SV were obtained, respectively. An essential oil extraction yield of 1.17 g/100 g OP (db) with a d-limonene content of 91.62% was produced. Moreover, hesperidin, apigenin, and naringenin yields of 7.88 mg/g, 0.475 mg/g, and 0.675 mg/g were obtained. An OP-based biorefinery addressed to produce essential oil, polyphenolic compounds, and biogas with a processing 25 tons/day (wb) has a sustainability index of 66.88%, higher than the values obtained with lesser upstream stages. In conclusion, an integral OP upgrading leads to better enzymatic and anaerobic digestion performances, as well as, a high process sustainability.

Abstract:Following the extraction of essential oil, citrus (Mousambi, Kinnow, and Orange) peel wastes were used to produce pectin. The yield of essential oil and pectin was maximum in orange. Pectin was characterized by Fourier-transform infrared spectroscopy (FT-IR) and X-ray diffraction (XRD) spectroscopy. The degree of esterification (DE) and methoxyl content (MC) was maximum in orange whereas, the equivalent weight was maximum in Mousambi. A significant increase (61.8%) in the Lactobacillus fermentum population was observed with pectin as compared with sugar. Three sources followed the Orange > Kinnow > Mousambi trend as a prebiotic source. It was attributed to higher DE as well as higher MC. Enhancement in the bacterial population was in the range of 79.16-87.50%. The present work confirms the potential of pectin as a probiotic source for the enhancement of the bacterial population. Thus, it has a large scope for use in the food industry targeting a circular economy.
Sustainable biosynthesis of lycopene by using evolutionary adaptive recombinant Escherichia coli from orange peel waste

Abstract:This study aimed to evaluate the hydrolysates from orange peel waste (OPW) as the low-cost carbon source for lycopene production. Initially, the dilute acid pretreatment combined with enzymatic hydrolysis of OPW resulted in a total sugar concentration of 62.18 g/L. Meanwhile, a four-month adaptive laboratory evolution (ALE) experiment using a d-galacturonic acid minimal medium resulted in an improvement in the growth rate of our previously engineered Escherichia coli strain for lycopene production. After evolutionary adaptation, response surface methodology (RSM) was adapted to optimize the medium composition in fermentation. The results obtained from RSM analysis revealed that the 5.53 % carbon source of orange peel hydrolysate (OPH), 6.57 g/L nitrogen source, and 30 °C temperature boosted lycopene production in the final strain. Subsequently, the optimized treatment for lycopene fermentation was then conducted in a 5 L batch fermenter under the surveillance of a kinetic model that uses the Logistic equation for strain growth (μm = 0.441 h-1), and Luedeking-Piret equations for lycopene production (Pm = 1043 mgL-1) with growth rate constant (α = 0.1491). At last, lycopene biosynthesized from OPH was extracted and analyzed for qualitative validation. Likewise, its data on phytic acid (between 1.01 % and 0.86 %) and DPPH radical scavenging (between 38.06 % and 29.08 %) highlighted the better antioxidant capacity of lycopene. In conclusion, the OPH can be used as a fermentation feedstock which opens new possibilities of exploiting fruit crop residues for food and pharmaceutical applications.

Abstract:The use of waste as a carbon source can significantly reduce the cost of production of Polyhydroxyalkanoates (PHAs). In this study, an acidified hydrolysate solution derived from food waste (FW) was used as a carbon source for the synthesis of PHAs by Rhodopseudomonas palustris ( R. palustris ) and optimized the process parameters. The results showed that the PHAs yield reached 48.62% under optimal conditions (an incubation time of 30 days, volatile fatty acids (VFAs) in substrate concentration of 2202.21 mg⋅L -1 , an initial pH of 8.0, and inoculum concentration of 15%). The fraction of VFAs affects the composition of PHAs, R. palustris first uses VFAs with an even number of carbons to synthesize poly(3-hydroxybutyrate) (3HB), and later uses VFAs with an odd number of carbons to synthesize poly-3-hydroxyvalerate (3HV). Pathways for the synthesis of PHAs by R. palustris were inferred. R. palustris is a strain with the potential to synthesize PHAs.
