Abstract:Ninety six growing pigs (Duroc x Landrace x Jia) were used to investigate the effects of betaine on growth, nutrient utilization, carcass composition and moat quality at different levels of betaine in the diet. The pigs were randomly assigned by weight to four groups, each of which was replicated three times with eight pigs per replicate. Four groups were fed with diets supplemented with 0, 1,000, 1,500 and 2,000 mg/kg betaine. The pigs were grown from 20 kg live weight to approximately 64 kg. The 1,000 mg/kg and 1,500 mg/kg betaine treated groups increased average daily gain by 13.20% (p<0.01) and 9.28% (p<0.05) and average daily feed intake by 7.30% (p<0.05) and 7.33% (p<0.01) respectively and decreased feed conversion ratio by 7.93% (p<0.01) and 6.55% (p<0.05) respectively compared to the control group. However, these differences were not found in the other betaine treated groups. The 1,000 mg/kg betaine treated group significantly elevated carcass lean proportion by 7.49% (p<0.05) and longissimus muscle area by 19.12% (p<0.05) and contents of ether extract and myoglobin in longissimus dorsi. by 34.21% (p<0.01) and 29.56% (p<0.01) respectively, reduced carcass fat proportion and fat depth by 27.21% (p<0.05) and 14.86% (p<0.05) respectively compared to the control group. It is concluded that betaine supplementation in the diets may improve growth performance and carcass characteristics in growing pigs.

Abstract:An experiment was conducted to investigate the effect of dietary betaine supplementation on carcass characteristics, hormones, growth factor and lipid metabolism in finishing pigs. Forty-eight crossbred barrows and gilts (Seghers×Seghers×Duroc) weighing about 55 kg were divided into two groups, each with three replicates of eight pigs (four barrows and four gilts) per replicate, and fed corn–soybean meal basal diets supplemented with 0 and 0.125% betaine for 42 days. At trial termination, two pigs (one barrow and one gilt) weighing about 90 kg were selected from each replicate and slaughtered for analyses. The results showed that betaine increased carcass lean percentage and longissimus muscle area by 5.19% (P<0.01) and 17.85% (P<0.01), respectively, and decreased carcass fat percentage and average backfat thickness by 13.07% (P<0.01) and 10.30% (P<0.05), respectively. Serum growth hormone, insulin-like growth factor I, free triiodothyronine, free thyroxine and insulin levels in pigs fed betaine were elevated by 45.61% (P<0.01), 55.50% (P<0.01), 57.95% (P<0.01), 51.80% (P<0.01) and 42.34% (P<0.05), respectively. Fatty acid synthase activity in the 10th rib subcutaneous adipose tissue was decreased by 24.35% (P<0.05) with betaine supplementation, whereas hormone-sensitive lipase activity was significantly increased (P<0.05). Meanwhile, serum free fatty acids concentration in betaine-fed pigs was 25.75% higher compared to controls (P<0.01). The study suggested that betaine could induce changes in hormones and growth factor in finishing pigs, and therefore could inhibit fat synthesis through reducing lipogenic enzymes activities and promote fat degradation by increasing hormone-sensitive lipase activity, with a resultant decrease in adipose tissue mass and improvement in carcass characteristics.

Abstract:An experiment was conducted to investigate the effect of dietary betaine supplementation on growth performance, carcass characteristics, lipogenic enzyme activities in subcutaneous adipose tissue and mRNA expression of fatty acid synthase (FAS) in finishing pigs. Forty-eight crossbred barrows and gilts (Duroc×Seghers 15 line×Seghers 12/36 line) with a body weight of 55.7±0.48kg were divided into two groups, and three replicates of eight pigs (four barrows and four gilts) were assigned to each group. Pigs were fed a maize–soybean meal basal diet supplemented with either 0 or 1250mg betaine/kg feed for 42 days. At the end of the trial, two pigs (one barrow and one gilt) weighing around 90kg were selected from each replicate and slaughtered. Average daily gain increased by 5.5% (P<0.05) with betaine supplementation, but average daily feed intake and feed conversion ratio were not affected. Adding betaine to diet increased carcass lean proportion and loin muscle area (P<0.01) and decreased carcass fat proportion (P<0.01) and average backfat thickness (P<0.05). Betaine did not influence dressing proportion, proportion of skin and bone of the carcass or leaf fat weight. Betaine supplementation decreased the activities of acetyl-CoA carboxylase, FAS and malic enzyme in subcutaneous adipose tissue by 18.0% (P<0.01), 18.8% (P<0.05) and 14.5% (P<0.05), respectively, but the activities of glucose-6-phosphate dehydrogenase and isocitrate dehydrogenase were not affected (P>0.10). Semi-quantitative reverse transcription polymerase chain reaction analysis revealed a 25% decrease in FAS mRNA expression in pigs fed betaine (P<0.05). A significant correlation between mRNA expression level and potential enzyme activity for FAS was found (r=0.93; P<0.01). The study suggests that the reduction in adipose tissue observed in pigs fed additional betaine might result from diminished rates of lipogenesis that is a consequence of decreased activities and gene expression of lipogenic enzymes, as evidenced by the decrease in FAS mRNA expression.
Betaine affects muscle lipid metabolism via regulating the fatty acid uptake and oxidation in finishing pig

Abstract:BackgroundBetaine affects fat metabolism in animals, but the specific mechanism is still not clear. The purpose of this study was to investigate possible mechanisms of betaine in altering lipid metabolism in muscle tissue in finishing pigs.MethodsA total of 120 crossbred gilts (Landrace × Yorkshire × Duroc) with an average initial body weight of 70.1 kg were randomly allotted to three dietary treatments. The treatments included a corn–soybean meal basal diet supplemented with 0, 1250 or 2500 mg/kg betaine. The feeding experiment lasted 42 d.ResultsBetaine addition to the diet significantly increased the concentration of free fatty acids (FFA) in muscle (P < 0.05). Furthermore, the levels of serum cholesterol and high-density lipoprotein cholesterol were decreased (P < 0.05) and total cholesterol content was increased in muscle (P < 0.05) of betaine fed pigs. Experiments on genes involved in fatty acid transport showed that betaine increased expression of lipoprotein lipase(LPL), fatty acid translocase/cluster of differentiation (FAT/CD36), fatty acid binding protein (FABP3) and fatty acid transport protein (FATP1) (P < 0.05). The abundance of fatty acid transport protein and fatty acid binding protein were also increased by betaine (P < 0.05). As for the key factors involved in fatty acid oxidation, although betaine supplementation didn’t affect the level of carnitine and malonyl-CoA, betaine increased mRNA and protein abundance of carnitine palmitransferase-1(CPT1) and phosphorylated-AMPK (P < 0.05).ConclusionsThe results suggested that betaine may promoted muscle fatty acid uptake via up-regulating the genes related to fatty acid transporter including FAT/CD36, FATP1 and FABP3. On the other hand, betaine activated AMPK and up-regulated genes related to fatty acid oxidation including PPARα and CPT1. The underlying mechanism regulating fatty acid metabolism in pigs supplemented with betaine is associated with the up-regulation of genes involved in fatty acid transport and fatty acid oxidation.

Effects of Betaine on Fat and Protein Metabolism in Different Stages of Swine

Abstract:The feeding and detecting experiments were conducted to study the effects of 800, 1 000 ,1 750 mg/kg betaine on fat and protein metabolism in weanling piglets, growing pigs and finishing pigs. The results showed that adding betaine in the diet increased average daily weight gain by 8.71 %(P0.05), 13.20%(P0.01) and 13.32%(P0.01), decreased percentage of dissected fat by 12.97%(P0.01), 9.62%(P0.01) and 12.49(P0.01), increased percentage of dissected lean of carcass by 4.08%(P0.05), 7.15%(P0.05) and 3.31%(P0.05) respectively. The content of free carnitine in liver was enhanced by 8.98%(P0.05), 21.36%(P0.05) and 30.18 %(P0.01) respectively. The growth hormone level in serum was promoted by 46.15%(P 0.01 ), 102.11%(P0.01) and 58.33%(P0.05), the insulin-like growth factor-1 was promoted by 38.74%(P0.05), 44.74%(P0.01) and 47.95%(P0.01) respectively. Futhermore, betaine could enhance the malic dehydrogenase activities of the subcutaneous fat by 13.60%(P0.05) and 14.59%(P0.01) in growing and finishing pigs, but had no significant effect on weaned piglets(P0.05).

Abstract:1. This study was conducted to investigate the effects of methionine and betaine supplementation on growth performance, carcase composition and lipid metabolism in growing broilers. 2. A total of 450 commercial broilers, 22 d of age, were randomly allocated to three groups, each of which included three replicates (50 birds per replicate). The groups received the same methionine-deficient diet supplemented with 0 or 1 g/kg methionine, or 0.5 g/kg betaine, respectively. 3. Methionine and betaine supplementation significantly improved weight gain and feed conversion. Supplemental methionine and betaine also significantly increased breast muscle yield and decreased abdominal fat content. Meanwhile, addition of methionine and betaine significantly increased the contents of creatine and free carnitine in liver, the activity of hormone-sensitive lipase in abdominal fat and the concentration of free fatty acid in serum, whereas uric acid concentration in serum was significantly decreased. 4. The results of this study suggest that betaine can spare methionine in its function as an essential amino acid and is as effective as methionine in improving performance and carcase quality of growing broilers if the diet is moderately deficient in methionine. The decrease in abdominal fat may be due to the increased carnitine synthesis in liver and hormone-sensitive lipase activity in abdominal fat.
Effects of Betaine on Performence, Carcass Characteristics and Hepatic Betaine-homocysteine Methyltransferase Activity in Finishing Barrows

Abstract:This experiment was conducted to determine the effect of dietary betaine (0, 0.125%) oil performance, carcass composition, pork quality and hepatic betaine-homocysteine methyltransferase (BHMT) activity of crossbred finishing barrows. Three replicates of ten pigs were used for each treatment, The results showed that average daily gain. feed intake and Iced conversion ere not affected by betaine. Compared with the control group. pigs treated with betaine had a 8.17% (p < 0.05) decrease ill carcass fat percentage. and a 8.84% (p < 0.05) reduction in 10th-rib backfat thickness, but dressing percentage, percentage lean. longissimus muscle area. and average backfat thickness were not affected. There were also no significant differences in musele color score, marbling score, pork pH value and water loss rate between the control and betaine-treated groups. Hepatic betaine-homocysteine methyltransferase (BHMT) activity was significantly increased by 13.97% (p < 0.05) when pigs were offered 0.125% betanie.
EFFECT OF BETAINE ON REPARTITION OF CARCASS FAT IN GROWING AND FINISHING PIGS AND APPROACH TO IT’S MECHANISM

Abstract:Two trials were conducted to investigate the effect of feeding betaine on the deposition of subcutaneous fat，carcass fat and the contents of intramusclar fat of the growing and finishing pigs and approach to mechanism of the effect by feeding experiment，determination of carcass，and liver，muscle and serum sample analysis．Trial 1 examined the effect of betaine supplementation （1000 mg／kg of diet） to the growing pigs．Forty-eight pigs（Duroc×Landrace×Yorkshire，20 kg initially） were allotted to two treatments set with three replicate pens per treatments by weight and sex．Trial 2 examined the effect of feeding betaine （1500 mg／kg of diet） to the finishing pigs．Sixty pigs （Duroc×Landrace×Yorkshire，60 kg initially） were allotted to two treatments set with three replicate pens per treatments by weight and sex．The results showed that betaine supplementation increased the ADG of growing pigs by 13．20％（P＜0．05），decreased the FCR by 7．93％（P＜0．05），decreased the mean backfat depth of the growing and finishing pigs by 14.82％（P＜0．05） and 14．93％（P＜0．05），decreased the percentage of dissected fat of carcass by 5．76％（P＞0．05） and 11．51％（P＜0．05），increased the contents of intramusclar fat in longissimus dorsi（LD） of the growing and finishing pigs by 36．00％（P＜0．01） and 17．66％（P＜0．05） respectively．The serum analysis indicated that betaine supplementation increased the lipase activity of the growing pig and finishing pigs by 10．49％（P＜0．05） and 7.50％（P＝0.06），increased the FFA contents of growing and finishing pigs by 20．16％（P＜0．05） and 13.43％（P＜0．05） respectively．In addition，the contents of free carnitine in liver of the growing and finishing pigs fed betaine increased by 20．68％（P＜0．05） and 23．53％（P＜0．05） respectively．The contents of acid-insoluble carnitine，the ratio of acid-insoluble carnitine and free carnitine in LD of finishing pigs fed betaine increased by 48．59％（P＜0．05） and 38．24％（P＜0．01） respectively．These results indicated that betaine supplementation could reduce the lipid deposition and exert the repartition effects of the carcass fat throughout donate the methyl to produce carnitine to improve the long chain fatty acid-β-oxidation．

The results indicated:1000mg/kg betaine increased growing and finishing stage average daily gain(ADG) by 13.20%(P<0.01) and 5.68%(P<0.05) respectively,the whole stage ADG were increased by 12.74%(P<0.01); Betaine enhanced betaine-homocysteine(BHMT) methyltransferase activity in liver, in 1000mg/kg group, total and special activity were enhanced by 22.38%(P<0.01)和18.18%(P<0.05) in growing stage, and 16.33%(P<0.05) and 15.63%(P<0.05) was increased in finishing stage; Free-carnitine was improved significantly in liver and LD by adding betaine during the growing stage, but only the free-carnitine in liver and acid-insoluble carnitine in LD were changed obviously during the finishing stage.
Effects of Methyl-Donor on the Performances and Mechanisms of Growth-Promoting Hormone in Piglets.

Abstract:Seventy-two piglets were allotted to three groups by weight. Every group included three replicates of eight each. The groups received the same basal diet supplemented with 0,800mg/kg betaine and 2000mg/kg methionine for 36d. The results indicated that betainegroup obviously increased ADG by 11.82%(P0.05), FI by 8.71%(P0.05) and BHMT activity in liver by 61.41%(P0.05), decreased F/G by 2.79%(P0.05) compared with Metunadded group. Compared with Met-added group, betaine group promoted BHMT activity by 55.36%(p0.05). In addition, betaine group apparently reduced SUN level by 35.70%(P0.01) and increased total iron binding ability and GH and IGF-1 levels(P0.05) in serum respectively compared with Met-unadded group. These results suggested that betaine might exert the following functions:(1)sparing methionine by providing effective labile methyl for methylation products synthesis instead of methionine under control of BHMT.(2)promoting growth by increasing GH and IGF-1 levels in serum.

Abstract:Pigs supplemented with 1 000mg/kg betaine grew fastest, whose ADG was increased by 13.20% (P<0.01), F/G was decreased by 7.93% (P<0.01), and data obtained from samples analysis of this group showed that levels of GH, IGF-I, T_(3), T_(4) in serum were elevated by 101.76% (P<0.01), 44.75% (P<0.01), 26.53% (P<0.01) and 16.83% (P<0.05) respectively; concentration of free Ser in serum was elevated by 14.28% (P<0.05); concentration of TP was increased by 21.69% (P<0.01), while SUN was decreased by 47.67% (P<0.01); content of RNA in LD and liver, and RNA/RNA ratio in LD were enhanced by 12.60% (P<0.05), 17.80% (P<0.02) and 19.79% (P<0.02) respectively; concentrations of cAMP in liver and anterior pituitary were increased obviously by 47.53% (P<0.01) and 65.21% (P<0.01).
95 Betaine Improves Growth Performance by Increasing Digestive Enzymes Activities, and Ameliorating Intestinal Structure of Piglets

Abstract:This study was aimed to investigate the effects of betaine on intestinal structures and digestive enzyme activities of piglets. A total of 150 crossbred piglets (Duroc × Landrace × Yorkshire), with an average initial body weight of 8.52 ± 0.26 kg and age of 39-day, were randomly divided into three dietary treatments, each of which was replicated five times with ten pigs per replicate. The dietary treatments were basal diet supplemented with 0, 1250, 2500 mg/kg betaine, respectively. The experiments lasted for 30 days. The results showed that supplementation of betaine in the diet significantly increased average daily gain (P < 0.05), decreased diarrhea rate (P < 0.05), and improved feed conversion rate (P < 0.05) of piglets. The villus height and the ratio of villus height to crypt depth of small intestine were significantly increased with betaine supplementation (P < 0.05). The addition of betaine in diet also increased the expression of tight junction proteins in small intestine (P < 0.05) and decreased the diamine oxidase in plasma (P < 0.05). The apparent total tract digestibility of crude protein and ether extract (P < 0.05) and the activities of amylase, trypsin, and lipase (P < 0.05) in small intestinal were increased with betaine addition. Enzyme kinetic experiments in vitro indicated that hypertonic environment formed by NaCl could inhibit the activities of amylase and trypsin. While betaine addition improved the affinity of amylase and trypsin with their corresponding substrates, thereby counteracted the inhibitory effect of hyperosmolarity. In conclusion, the supplementation of betaine in diet could improve growth performance of piglets by ameliorating intestinal structure, increasing digestive enzymes activities, promoting the digestion of nutrients and decreasing the diarrhea rate.
Leucine Improves the Growth Performance, Carcass Traits, and Lipid Nutritional Quality of Pork in Shaziling Pigs

Abstract:Leucine is involved in promoting fatty acid oxidation and lipolysis, mediating lipid metabolism and energy homeostasis, thus it has been widely used in livestock production. However, the effects of leucine on fat deposition and nutrition in Shaziling pigs remain unclear. A total of 72 Shaziling pigs (150 days old, weight 35.00 ± 1.00 kg) were randomly divided into 2 groups and fed with basal diet (control group) or basal diet containing 1% leucine (leucine group) for 60 days. The results showed that leucine significantly increased the average daily feed intake but decreased the ratio of feed to gain (P < 0.05), increased the loin muscle area and serum glucose content (P < 0.05) of Shaziling pigs. Besides, leucine regulated the re-distribution of fatty acids from adipose tissue to muscle as it significantly increased the contents of C18:1n-9 and C22:6n-3 (DHA) in the longissimus thoracis while decreased the contents of C22:5n-3 (DPA), C20:5n-3 (EPA), and DHA in the adipose tissue of Shaziling pigs (P < 0.05). Lipidomic analysis showed that the contents of phosphatidylethanolamines (PEs), cardiolipins (CLs), and phosphatidylglycerols (PGs) in the longissimus thoracis and the contents of lysophosphatidylethanolamines (LPEs), ceramides (Cers), phosphatidylinositols (PIs) in adipose tissue of Shaziling pigs were decreased in leucine group (P < 0.05). Collectively, this study clarified that dietary addition of 1% leucine have a better effect on growth performance and the deposition of beneficial fatty acids in the muscle of Shaziling pigs, which is conductive to the production of high quality and healthy pork. In addition, leucine altered the lipid composition of muscle and fat in Shaziling pigs. The related results provide a theoretical basis and application guidance for regulating fat deposition in Shaziling pigs, which is important for the healthy breeding of Shaziling pigs.
Effect of betaine on muscle,liver and serum amino acid composition in finishing swine

Abstract:One hundred fifty finishing pigs (Duroc× Landrace×yorkshire) wereallotted to five groups which received the same basal diet, supplemented with 0, 1 000, 1 250, 1 500, 1 750 mg/kg betaine. On finishing40 days feeding experiment, 60 pigs were slaughted, the longissimus dorsi, liver and serum samples were collected and amino acid composition were analysed. The results indicated that different level betaine hereof did not affect the amino acid composition of longissimus dorsi and liver, but some of the free amino acid contents were improved in serum especially the serine and valine which were increased by 14.28％ (P<0.05) and 31.75％ (P<0.05), respectively.
Betaine suppresses carnitine palmitoyltransferase I in skeletal muscle but not in liver of finishing pigs

Abstract:An experiment was conducted to investigate the effect of dietary betaine supplementation on the enzyme activity and mRNA abundance for carnitine palmitoyltransferase I (CPT I) in liver and skeletal muscle of finishing pigs. Forty-eight crossbred barrows and gilts [Duroc × (Seghers × Seghers)] weighing about 55kg were divided into two dietary treatments, each with three replicates of eight pigs (four barrows and four gilts) per replicate. Pigs were fed a corn–soybean meal basal diet supplemented with betaine at 0 or 1250mg/kg feed for 42days. At trial termination, two pigs (one barrow and one gilt) weighing about 90kg were selected from each replicate (six pigs per dietary treatment) and slaughtered for analyses. The results showed that intramuscular fat content in the longissimus muscle of pigs fed betaine was 23.6% higher than that of controls (P<0.05), whereas hepatic fat content was not affected with dietary betaine treatment. Muscle-type CPT I (M-CPT I) activity, but not liver-type CPT I (L-CPT I) activity was decreased by betaine supplementation. Furthermore, betaine supplementation reduced M-CPT I mRNA abundance by 14.6% (P<0.05) but did not affect L-CPT I mRNA abundance. There was a positive correlation between enzyme activity and mRNA abundance for both L-CPT I and M-CPT I (r=0.67 and r=0.72 for L-CPT I and M-CPT I, respectively; P<0.05). The study suggests that betaine may be involved in fat partitioning in pigs by reducing the activity and mRNA abundance of M-CPT I, with a resultant increase in intramuscular fat content.
Effects of "Shiquan Dabu" Chinese herb residues on growth, carcass composition and meat quality in finishing pigs

Abstract:Ninety-six finishing pigs were selected to study the effects of different doses of "Shiquan Dabu" Chinese herb residues on growth, carcass composition and meat quality. For the pigs treated with 0.5%, 1.0% and 1.5% of Chinese herb residue in their diet, daily gains are increased by 7.40%(P < 0.05), 12.88%(P < 0.05) and 9.75% (P < 0.05), feed gain ratios are decreased by 5.20% (P < 0.05), 7.23% (P < 0.05) and 6.07% (P < 0.05), lean meat percentages are increased by 3.86% (P > 0.05), 6.27% (P < 0.05) and 5.80% (P < 0.05), carcass fat percentages are decreased by 6.42%(P < 0.05), 9.63%(P < 0.05) and 9.30% (P < 0.05), back-fat depths are reduced by 7.20% (P < 0.05), 13.26% (P < 0.05) and 12. 12%(P < 0.05), longissimus dorsi areas are increased by 4.46%(P > 0.05), 9.24% (P < 0.05) and 6. 30% (P < 0.05), meat red color degrees are increased by 2.75% (P > 0.05), 6.67% (P<0. 05) and 3. 92% (P>0. 05), drip loss rates are decreased by 9.32% (P < 0.05), 12.42% (P < 0.05) and 8.07(P < 0.05), contents of intramuscular fat are increased by 20.80% (P < 0.05), 28.47% (P < 0.01) and 25.55% (P < 0.01) compared with the control group fed with basal diet, respectively. The results indicated that "Shiquan Dabu" Chinese herb residues could make a novel alternative feed additive and improve growth performance, carcass composition and meat quality of finishing pigs.

Abstract:This paper studied the effects of50of100mg/kg L-carnitine supplementation on activities of digestive enzymes of duodenal contents in three-breed growing-finishing pigs(Duroc×Yorkshire×White,initially weight35kg,finality weight90kg).At the end of feeding trial18pigs were selected to slaughter,the samples of serum and liver were taken to test.The results showed supplementation of L-carnitine could enhance the activities of total protein enzyme,amylase and lipase of duodenal contents in different degree.The100mg/kg L-carnitine significantly increased the activity of total protein enzyme,amylase and lipase by63.49%(P0.05),285.85%(P0.05)and360.86%(P0.01),respectively.In addition,the100mg/kg L-carnitine increased the content ofβ-hydroxybutyrate by27.80%(P0.05)in serum,16.95%(P0.05)in liver.The50mg/kg L-carnitine gained the approximate results in serum or liver.The results indicated that the supplementation of L-carnitine could facilitated the fatty acids utilization and protein deposition,enhance the activities of digestive enzymes in the duodenum.

Abstract:To investigate the effect of betaine supplementation on growth performance, muscle protein deposition, muscle nucleic acid and amino acid contents, and muscle proteome of broilers, 160 one-day-old male partridge shank broiler chickens were randomly divided into 2 groups with 8 replicates of 10 broilers each. Broilers were fed a basal diet alone, or a basal diet supplemented with 1000 mg/kg betaine. Compared with the control group, the betaine group significantly increased (p < 0.05) the broilers average daily gain, the levels of serum insulin-like growth factor-1 (IGF-1), growth hormone (GH), total protein (TP), the contents of muscle absolute protein deposition, RNA, Ser, Glu, Met, and Phe, and the ratio of RNA/DNA, and decreased (p < 0.05) the feed conversion ratio and serum blood urea nitrogen content. Moreover, proteomic analysis revealed 35 differentially abundant proteins (DAPs) in the betaine group compared with the control group, including 27 upregulated proteins and 8 downregulated proteins (p < 0.05). These DAPs were mainly related to cell differentiation, small molecule metabolic process, and tissue development. In conclusion, diets supplemented with 1000 mg/kg betaine improved growth performance and muscle protein deposition of broilers. Increased serum GH, IGF-1, and TP contents, and alterations in muscle nucleic acids, amino acids, and protein abundance levels were involved in this process.

Abstract:Rumen-protected betaine (RPB) can enhance betaine absorption in the small intestine of ruminants, while betaine can alter fat distribution and has the potential to affect the meat quality of livestock. Hence, we hypothesized that RPB might also affect the meat quality of lambs. Sixty male Hu sheep of similar weight (30.47 ± 2.04 kg) were selected and randomly subjected to five different treatments. The sheep were fed a control diet (control treatment, CTL); 1.1 g/day unprotected-betaine supplemented diet (UPB); or doses of 1.1 g/day (low RPB treatment; L-PB), 2.2 g/day (middle RPB treatment; M-PB) or 3.3 g/day (high RPB treatment; H-PB) RPB-supplemented diet for 70 days. Slaughter performance, meat quality, fatty acid and amino acid content in the longissimus dorsi (LD) muscle, shoulder muscle (SM) and gluteus muscle (GM) were measured. Compared with CTL, betaine (including UPB and RPB) supplementation increased the average daily weight gain (ADG) (P < 0.05) and average daily feed intake (P < 0.01) of lambs. Rumen-protected betaine increased ADG (P < 0.05) compared with UPB. With increasing RPB doses, the eye muscle area of the lambs linearly increased (P < 0.05). Compared with CTL, betaine supplementation decreased water loss (P < 0.05) in SM and increased pH24 in the SM (P < 0.05) and GM (P < 0.05). Compared with UPB, RPB decreased water loss in the GM (P < 0.01), decreased shear force (P < 0.05) in the LD and SM and increased the pH of the meat 24 h after slaughter (pH24). With increasing RPB doses, the shear force and b* value in the LD linearly decreased (P < 0.05), and the pH24 of the meat quadratically increased (P < 0.05). Compared with CTL, betaine supplementation increased the polyunsaturated fatty acid in the GM (P < 0.05). Compared with UPB, RPB supplementation decreased the saturated fatty acid (SFA) content in the LD (P < 0.05) and increased the unsaturated fatty acids (UFA), mono-unsaturated fatty acids and UFA/SFA ratio in the LD (P < 0.05). Compared with CTL, the content of histidine in the LD increased with betaine supplementation. Compared with UPB, RPB supplementation increased the content of total free amino acids and flavor amino acids in the LD of lambs (P < 0.05). With increasing RPB, the isoleucine and phenylalanine contents in the LD linearly increased (P < 0.05). Overall, the data collected indicated that the meat quality of lambs (especially in the LD) improved as a result of betaine supplementation, and RPB showed better effects than those of UPB.
