Abstract:We report here that a Cl(-)-dependent K+ (K:Cl) efflux, which is stimulated by N-ethylmaleimide (NEM) and by increased red cell volume, exists in young red cells of individuals with normal hemoglobin A (AA) and in those homozygous for hemoglobin S (SS). We have investigated this K:Cl efflux in several density-defined red cell fractions obtained from Percoll-Stractan continuous density gradients. We found high activity of the NEM-stimulated K:Cl transport in reticulocytes and young red cells from nine sickle cell (SS) patients (43 +/- 27 mean +/- SD mmol K+/liter of cells/hr = flux units (FU)) and in the young cell fraction of three AA individuals with high reticulocytosis recuperating from nutritional anemias (41.7 +/- 10 FU). In addition, we observed significant interindividual variation of this K:Cl efflux in the discocyte fraction of SS blood. Cell swelling markedly stimulated the K:Cl efflux, in SS whole blood (9.8 +/- 7.4 FU, in SS young cells (13 +/- 13 FU), and in AA young cells (21.4 +/- 11 FU). The activity of the Na-K-Cl cotransport, as estimated by the bumetanide sensitive K+ efflux was not found to be cell-age dependent in either AA or SS cells. Measurements of red cell density by isopycnic gradients indicated that 27% of the young cells reduce their volume by a Cl(-)-dependent process in hypotonic or low pH-induced swelling. The large volume-stimulated K:Cl efflux in AA young cells raises the possibility that these fluxes may be involved in the maturation of erythropoietic precursors.(ABSTRACT TRUNCATED AT 250 WORDS)
Cell volume, K transport, and cell density in human erythrocytes

Abstract:We report here studies on the regulation of cell volume and K transport in human erythrocytes separated according to density. When cell volume was increased (isosmotic swelling, nystatin technique), erythrocytes of the least dense but not of the densest fraction shrunk back toward their original volume. This process was due to a ouabain (0.1 mM) and bumetanide (0.01 mM) (OB)-resistant K loss. OB-resistant K+ efflux from the least dense fraction was stimulated by hypotonic swelling and had a bell-shaped dependence on pH (pH optimum 6.75-7.0). These pH and volume effects were not evident in the densest fraction. The swelling-induced K+ efflux from the least dense fraction was inhibited when chloride was substituted by nitrate, thiocyanate, and acetate, whereas it was stimulated by bromide. Increasing cell Mg2+ content also markedly inhibited K+ efflux from isosmotically swollen cells. N-ethylmaleimide (NEM, 1 mM) greatly increased OB-resistant K+ efflux from the least dense fraction but not from the densest fraction. These data reveal the presence, in the lease dense fraction of normal human erythrocytes, of a pathway for K+ transport that is dependent on volume, pH, and chloride, is inhibited by internal Mg2+, and possibly plays a role in determining the erythrocyte water and cation content.
[Interleukin-2 Reduces Calcium Handling Capacity of Rat Ventricular Myocytes During Anoxia/reoxygenation].

Abstract:AIM:To investigate the effect of interleukin-2(IL-2) on the cell contractility and calcium handling in cardiomyocytes during normoxia or anoxia/reoxygenation.METHODS:Chemical anoxia introduced by Krebs-Henseleit(K-H) solution containing 10(-3) mol/L sodium dithionite was used in the enzymatically isolated rat ventricular myocytes. The video-tracking system and spectrofluorometric method were employed to verify the cell contraction and calcium handling of the single myocyte.RESULTS:During anoxia, the cell contraction, amplitude of calcium transient induced by electrical stimulation and Ca2+ release induced by caffeine were depressed while resting calcium level was elevated, but the activity of the L-type calcium channels were not changed. All the parameters could not return to the pre-anoxia level during reoxygenation. IL-2 at 2 x 10(5) U/L administrated during anoxia aggravated the effect of reoxygenation on cell contraction and the calcium handling.CONCLUSION:Coexistence of IL-2 during anoxia aggravated the effect of reoxygenation on the cell contraction and calcium handling in the isolated rat ventricular myocytes, in which the reduced calcium release from sarcoplasmic reticulum was involved.

Abstract:We consider the matrix completion problem under a form of row/column weighted entrywise sampling, including the case of uniformentrywise sampling as a special case. We analyze the associated random observation operator, and prove that with high probability, it satisfies a form of restricted strong convexity with respect to weighted Frobenius norm. Using this property, we obtain as corollaries a number of error bounds on matrix completion in the weighted Frobenius norm under noisy sampling and for both exact and near low-rank matrices. Our results are based on measures of the 'spikiness' and 'low-rankness' of matrices that are less restrictive than the incoherence conditions imposed in previous work. Our technique involves an M-estimator that includes controls on both the rank and spikiness of the solution, and we establish non-asymptotic error bounds in weighted Frobenius norm for recovering matrices lying with lq-"balls" of bounded spikiness. Using information-theoretic methods, we show that no algorithm can achieve better estimates (up to a logarithmic factor) over these same sets, showing that our conditions on matrices and associated rates are essentially optimal.

Abstract:We investigated the role of Na(+)-K(+)-Cl(-) cotransporter (NKCC1) in conjunction with Na(+)/Ca(2+) exchanger (NCX) in disruption of endoplasmic reticulum (ER) Ca(2+) homeostasis and ER stress development in primary cortical neurons following in vitro ischemia. Oxygen-glucose deprivation (OGD) and reoxygenation (REOX) caused a rise in [Na(+)](cyt) which was accompanied by an elevation in [Ca(2+)](cyt). Inhibition of NKCC1 with its potent inhibitor bumetanide abolished the OGD/REOX-induced rise in [Na(+)](cyt) and [Ca(2+)](cyt). Moreover, OGD significantly increased Ca(ER)(2+) accumulation. Following REOX, a biphasic change in Ca(ER)(2+) occurred with an initial release of Ca(ER)(2+) which was sensitive to inositol 1,4,5-trisphosphate receptor (IP(3)R) inhibition and a subsequent refilling of Ca(ER)(2+) stores. Inhibition of NKCC1 activity with its inhibitor or genetic ablation prevented the release of Ca(ER)(2+). A similar result was obtained with inhibition of reversed mode operation of NCX (NCX(rev)). OGD/REOX also triggered a transient increase of glucose regulated protein 78 (GRP78), phospho-form of the alpha subunit of eukaryotic initiation factor 2 (p-eIF2 alpha), and cleaved caspase 12 proteins. Pre-treatment of neurons with NKCC1 inhibitor bumetanide inhibited upregulation of GRP78 and attenuated the level of cleaved caspase 12 and p-eIF2 alpha. Inhibition of NKCC1 reduced cytochrome C release and neuronal death. Taken together, these results suggest that NKCC1 and NCX(rev) may be involved in ischemic cell damage in part via disrupting ER Ca(2+) homeostasis and ER function.
Effects of Diazoxide on the Mitochondrial Ultrastructure and Permeability in Donor Rat Myocardium

Abstract:OBJECTIVE:To investigate the effect of diazoxide (DE) on the myocardial ultrastructure and opening of maitochondrial permeability transition pore (MPTP) in donor rat heart suffered from long-term hypothermic preservation.METHODS:The Langendorff model of isolated rat heart was used. The hearts were stored in 4 degrees C Celsior solution containing different concentration of DE (15, 30, or 45 micromol/L) for 9 h followed by 60 min of reperfusion. The recovery of rate-pressure product (RPP) was observed. The opening of MPTP and myocardial mitochondria ultrastructure were also evaluated.RESULTS:(1) As compared with the celsior solution preserved group, DE (30 micromol/L) increased recovery of RPP during reperfusion and inhibited the opening of MPTP. DE also alleviated the myocardial mitochondrial ultrastucture damage induced by long-term hypothermic preservation. (2) The above effects of DE were attenuated by a mitoK(ATP) channel inhibitor 5-hydroxydecanoate and a MPTP opener atractyloside.CONCLUSION:In the donor rat heart, DE protects myocardial mitochondria ultrastructure against long-term hypothermic preservation injury via inhibiting the opening of MPIP.
Activation of K+/Cl- cotransport in human erythrocytes exposed to oxidative agents

Abstract:Activation of K+/Cl- cotransport was studied after exposure of normal human erythrocytes to the oxidative action of acetylphenylhydrazine (APH), menadione sodium bisulfite (MSB), hydrogen peroxide (H2O2) or phenazine metasulfate (PMS). In order to better define the relative contributions of K+/Cl- cotransport on ouabain and bumetanide-resistant (OBR) K+ efflux induced by oxidation, we used (dihydroindenyl)oxyalkanoic acid (DIOA) and carbocyanine as specific inhibitors, respectively, of cotransport system and Ca(2+)-activated K+ channel. APH, MSB and - to much less extent - H2O2 promoted a K+ efflux pathway with features corresponding to those of K+/Cl- cotransport. This pathway showed: (i) kinetics of efflux compatible with a specific cation transport system; (ii) requirement for chloride anion; (iii) resistance to ouabain, bumetanide and carbocyanine inhibition; (iv) stimulation by hypotonic challenge; (v) susceptibility to inhibition by DIOA. Dithiothreitol (DTT) or 2-mercaptoethanol (2-ME) decreased K+/Cl- cotransport activation, suggesting that oxidative mechanisms affected crucial SH groups of the transporter. These data suggest that oxidation represents a factor capable of modulating activation of K+/Cl- cotransport. Its possible contribution in situations with high oxidative risk, such as sickle-cell anaemia or beta thalassemia, is discussed.
Cation transport and cell volume changes in maturing rat reticulocytes

Abstract:During maturation, reticulocytes lose membrane material, including transporters, and this is accompanied by a loss of cell water and volume. Here we determined a possible role of ion transport in adjusting cell volume during maturation. Reticulocytes and red blood cells of different ages were prepared from erythropoietin-treated rats by density gradient fractionation. Cell volume and ion transport were measured in freshly prepared cells and in reticulocytes during in vitro maturation. Reticulocytes had an increased K content and cell volume, whereas intracellular Na was decreased. All parameters approached whole blood values after 2 days in culture. Na-K pump was elevated in reticulocytes and decreased during maturation. Na-K-2Cl cotransport (NKCC) activity was lower in reticulocytes and was activated 8- and 20-fold by shrinkage and okadaic acid, respectively, whereas stimulation was barely detectable in high-buoyant density red blood cells. The ouabain- and bumetanide-insensitive Na flux in reticulocytes decreased on maturation. Most of it was inhibited by amiloride, indicating the presence of Na/proton exchange. Our results show that, although the Na-K-pump activity in reticulocytes is very much increased, the enhanced capacity of NKCC is essentially cryptic until stimulated. Both types of capacities (activities) decrease during maturation, indicating a possible loss of transport protein. The decrease was constrained to the period of reticulocyte maturation. Loss of transport capacity appears to exceed the loss of membrane surface area. Reticulocyte age-related changes in the net electrochemical driving force indicate that the increasing NKCC activity might contribute to the reduction in cell water.
Role of Nitrite, a Nitric Oxide Derivative, in K-Cl Cotransport Activation of Low-Potassium Sheep Red Blood Cells

Abstract:Abstract. K-Cl cotransport (COT) is the coupled movement of K and Cl, present in most cells, associated with regulatory volume decrease, susceptible to oxidation and functionally overexpressed in sickle cell anemia. The aim of this study was to characterize the effect of the oxidant nitrite (NO2−) on K-Cl COT. NO2− is a stable metabolic end product of the short-lived highly reactive free radical nitric oxide (NO), an oxidant and modulator of ion channels, and a vasodilator. In some systems, the response to NO2− is identical to that of NO. We hypothesized that NO2− activates K-Cl COT. Low potassium (LK) sheep red blood cells (SRBCs) were used as a model. The effect of various concentrations (10−6 to 10−1m) of NaNO2 was studied on K efflux in hypotonic Cl and NO3 media, Cl-dependent K efflux (K-Cl COT), glutathione (GSH), and methemoglobin (MetHb) formation. In support of our hypothesis, K efflux and K-Cl COT were stimulated by increasing concentrations of NaNO2. Stimulation of K efflux was dependent upon external Cl and exhibited a lag phase, consistent with activation of K-Cl COT through a regulatory mechanism. Exposure of LK SRBCs to NaNO2 decreased GSH, an effect characteristic of a thiol-oxidizing agent, and induced MetHb formation. K-Cl COT activity was positively correlated with Methb formation. N-ethyl-maleimide (NEM), a potent activator of K-Cl COT, was used to assess the mechanism of NO2− action. The results suggest that NEM and NO2− utilize at least one common pathway for K-Cl COT activation. Since NaNO2 is also a well known vasodilator, the present findings suggest a role of K-Cl COT in vasodilation.

Abstract:Computational models are useful tools to help understand signalling pathways in plant cells. A systems biology approach where models and experimental data are combined can provide experimentally verifiable predictions and novel insights. The brassinosteroid insensitive 1 (BRI1) receptor is one of the best-understood receptor systems in Arabidopsis with clearly described ligands, mutants and associated phenotypes. Therefore, BRI1-mediated signalling is attractive for mathematical modelling approaches to understand and interpret the spatial and temporal dynamics of signal transduction cascades in planta. To establish such a model, quantitative data sets incorporating local protein concentration, binding affinity and phosphorylation state of the different pathway components are essential. Computational modelling is increasingly employed in studies of plant growth and development. In this section, we have focused on the use of quantitative imaging of fluorescently labelled proteins as an entry point in modelling studies.

Abstract:It has been shown in a previous work that sodium influx is trebled, whereas potassium efflux is slightly reduced, when erythrocytes are suspended in a 142 mM bicarbonate medium containing ouabain. The present work deals with the active transport of sodium and potassium occurring in the presence of 142 mM bicarbonate (38° C, pH 7.40). Sodium influx and potassium efflux were unaffected by the removal of ouabain, whereas sodium efflux, potassium influx, and glucose metabolism were all strongly stimulated. The relation between ouabain sensitive ion transport and the calculated extra formation of ATP was 3.1 moles Na/mole ATP and 2.3 moles K/mole ATP, in agreement with values previously found on red cells transporting Na and K in plasma. Red cells incubated in the bicarbonate medium accumulated potassium until extracellular potassium concentration had been reduced to 1.8–2 mM. The normal volume regulation of the cells was severely interfered with, because accumulation of sodium and potassium was accompanied by a net flux of water of 6 g per meq cation. All effects observed could be interpreted to be secondary to the increased sodium influx, leading to an increase of intracellular sodium concentration, which in turn stimulated active cation transport, thereby accelerating the rate of ATP turn-over. The properties of the ouabain sensitive transport mechanism were not affected by the presence of bicarbonate. Both potassium efflux and sodium influx are increased by 200–300 per cent in the presence of 120 mM thiocyanate. It was found, however, that active cation transport is not stimulated in a 120 mM thiocyanate medium. On the contrary the ouabain sensitive ion transport was steadily decreasing during incubation in spite of conditions otherwise favouring transport of Na and K, i.e. increasing extracellular potassium and intracellular sodium concentrations.

The Chloride Conductance Inhibitor NS3623 Enhances the Activity of a Non-selective Cation Channel in Hyperpolarizing Conditions

Abstract:Handbooks of physiology state that the strategy adopted by red blood cells (RBCs) to preserve cell volume is to maintain membrane permeability for cations at its minimum. However, enhanced cation permeability can be measured and observed in specific physiological and pathophysiological situations such as in vivo senescence, storage at low temperature, sickle cell anemia and many other genetic defects affecting transporters, membrane or cytoskeletal proteins. Among cation pathways, cation channels are able to dissipate rapidly the gradients that are built and maintained by the sodium and calcium pumps. These situations are very well-documented but a mechanistic understanding of complex electrophysiological events underlying ion transports is still lacking. In addition, non-selective cation (NSC) channels present in the RBC membrane have proven difficult to molecular identification and functional characterization. For instance, NSC channel activity can be elicited by Low Ionic Strength conditions (LIS): the associated change in membrane potential triggers its opening in a voltage dependent manner. But, whereas this depolarizing media produces a spectacular activation of NSC channel, Gárdos channel-evoked hyperpolarization's have been shown to induce sodium entry through a pathway thought to be conductive and termed P cat . Using the CCCP method, which allows to follow fast changes in membrane potential, we show here (i) that hyperpolarization elicited by Gárdos channel activation triggers sodium entry through a conductive pathway, (ii) that chloride conductance inhibition unveils such conductive cationic conductance, (iii) that the use of the specific chloride conductance inhibitor NS3623 (a derivative of Neurosearch compound NS1652), at concentrations above what is needed for full anion channel block, potentiates the non-selective cation conductance. These results indicate that a non-selective cation channel is likely activated by the changes in the driving force for cations rather than a voltage dependence mechanism per se .

Abstract:Partial inhibition of creatine kinase (OK) has been shown to be associated with cardiomyopathy and heart failure. However, the link between CK inhibition and contractile dysfunction has not been elucidation. We have examined the hypothesis that partial inhibition of CK act iv i ty associated with sarcoplasmic reticulum (SR) by iodoacetamide (IAA) may deplete Ca 2. store and thus attenuates Ca transient and contraction in singte cardiac myocytes. IAA (1 I.tM) inhibited about 30% of the CK activity in isolated cells. This concentration of IAA did not alter [Ca2q~ transient or contraction of single cells electrically stimulated at 0.2 Hz. However, when the rate of stimulation was increased to 1 Hz, IAA inhibited the amplitudes of [Ca2*]i and contraction by about 25%. When myooytes stimulated at 0.2 Hz were treated with 10 nM isoproterenol, the amplitudes of [Ca2÷]~ and contraction were increased by about 350%. IAA attenuated the amplitudes of [Ca~*]~ transient and contraction by about 60%. IAA also attenuated the amplitude of caffeine-induced [Ca2+l~ transient by about 40% and contraction by about 40% in the single ventricular myocytes following stimulation at 1 Hz. IAA did not alter the rate of Ca 2÷ uptake into isolated SR vesicles supported by exogenous ATP, but reduced the rate by 23% when ATP was supplied endogenously by CK from phosphocreatine (PCr) + ADP. IAA did not affect L-type Ca ~* current in whole-cell patch clamped single myocytes indicating that its inhibitory effect on [Ca2÷]~ and contraction is not clue to inhibition of Ca 2÷ channel. The data demonstrate that partial inhibition of CK activity causes reduction in Ca 2÷ uptake into SR, decreases SR Ca 2. store, and consequently decreases [Ca2*]~ transient and contraction in stimulated single myocytes. Inhibition of [Ca2+]~ transient and contraction by IAA in stimulated cells [s probably due to inhibition of ATP supply by CK and development of energy deprivation during higher rate of ATP breakdown. Therefore, partial inhibition of CK may contribute to development of cardiomyopethy or heart failure during increased workload. 002
Effect of irradiation on red cell cation content and transport

Abstract:The effect of irradiation on red cell (RBC) cation content and membrane transport was studied. When units of RBCs collected in AS-1 (n = 8) were exposed to radiation (2000 cGy) and then stored at 4 degrees C, there was a significant increase in external K and internal Na and a decrease in internal K relative to control units (n = 3). Similar changes in internal cation contents were noted for RBCs collected in CPD; because of the higher hematocrit, the external K concentration was higher than that in AS-1-collected RBCs. Cellular K loss and Na gain were balanced, and the total cation content remained constant; however, both control and irradiated cells swelled over time as a consequence of the decreased pH and 2.3 DPG content. These changes in volume and cation content were reversed when the cells were incubated at 37 degrees C. A specific effect of radiation on the Na-K pump and Na-K-Cl and K-Cl cotransport systems was ruled out, because no significant differences between irradiated cells and controls were found. These data indicate that radiation induces an increased permeability to Na and K on the part of the RBC membrane lipid bilayer and that this increase is balanced and reversible.

Anion-dependent cation transport in erythrocytes

Abstract:A selective survey of the literature reveals at least three major anion-dependent cation transport systems, defined as Na+ + Cl-, K+ + Cl- and Na+ + K+ + Cl- respectively. In human red cells, kinetic data on the fraction of K+ and Na+ influx inhibitable by bumetanide are presented to indicate an Na+:K+ stoichiometry of 1:2. For LK sheep red cells the large Cl- -dependent K+ leak induced by swelling is shown to share many characteristics with that induced by N-ethylmaleimide (NEM) treatment. NEM has complex effects, both inhibiting and then activating Cl- -dependent K+ fluxes dependent on NEM concentration. The alloantibody anti-L can prevent the action of NEM. In human red cells NEM induces a large Cl- -dependent specific K+ flux, which shows saturation kinetics. Its anion preference is Cl- greater than Br- greater than SCN- greater than I- greater than NO3- greater than MeSO4-. This transport pathway is not inhibited by oligomycin or SITS, although phloretin and high concentrations of furosemide and bumetanide (over 0.3 mM) do inhibit. Quinine (0.5 mM) is also an inhibitor. It is concluded that at least two distinct Cl- -dependent transport pathways for K+ are inducible in mammalian red cells, although the evidence for their separation is not absolute.

Abstract:Kidney proximal tubules develop a severe but highly reversible energetic deficit due to nonesterified fatty acid (NEFA)-induced dissipation of mitochondrial membrane potential (DeltaPsi(m)) during reoxygenation after severe hypoxia. To assess the mechanism for this behavior, we have compared the efficacies of different NEFA for inducing mitochondrial deenergization in permeabilized tubules measured using safranin O uptake and studied the modification of NEFA-induced deenergization by inhibitors of the ADP/ATP carrier and glutamate using both normoxic tubules treated with exogenous NEFA and tubules deenergized during hypoxia-reoxygenation (H/R). Among the long-chain NEFA that accumulate during H/R of isolated tubules and ischemia-reperfusion of the kidney in vivo, oleate, linoleate, and arachidonate had strong effects to dissipate DeltaPsi(m) that were slightly greater than palmitate, while stearate was inactive at concentrations reached in the cells. This behavior correlates well with the protonophoric effects of each NEFA. Inhibition of the ADP/ATP carrier with either carboxyatractyloside or bongkrekic acid or addition of glutamate to compete for the aspartate/glutamate carrier improved DeltaPsi(m) in the presence of exogenous oleate and after H/R. Effects on the two carriers were additive and restored safranin O uptake to as much as 80% of normal under both conditions. The data strongly support NEFA cycling across the inner mitochondrial membrane using anion carriers as the main mechanism for NEFA-induced deenergization in this system and provide the first evidence for a contribution of this process to pathophysiological events that impact importantly on energetics of intact cells.
Significance of two transmembrane ion gradients for human erythrocyte volume stabilization

Abstract:Functional effectiveness of erythrocytes depends on their high deformability that allows them to pass through narrow tissue capillaries. The erythrocytes can deform easily due to discoid shape provided by the stabilization of an optimal cell volume at a given cell surface area. We used mathematical simulation to study the role of transport Na/K-ATPase and transmembrane Na + and K + gradients in human erythrocyte volume stabilization at non-selective increase in cell membrane permeability to cations. The model included Na/K-ATPase activated by intracellular Na + , Na + and K + transmembrane gradients, and took into account contribution of glycolytic metabolites and adenine nucleotides to cytoplasm osmotic pressure. We found that this model provides the best stabilization of the erythrocyte volume at non-selective increase in the permeability of the cell membrane, which can be caused by an oxidation of the membrane components or mechanical stress during circulation. The volume of the erythrocyte deviates from the optimal value by no more than 10% with a change in the non-selective permeability of the cell membrane to cations from 50 to 200% of the normal value. If only one transmembrane ion gradient is present (Na + ), the cell loses the ability to stabilize volume and even small changes in membrane permeability cause dramatic changes in the cell volume. Our results reveal that the presence of two oppositely directed transmembrane ion gradients is fundamentally important for robust stabilization of cellular volume in human erythrocytes.

Abstract:O 2 diffusion across red blood cell (RBC) membranes is a critically important physiological process for the maintenance of life. Our previous work on murine RBCs ( bioRxiv. doi: 10.1101/2020.08.28.265066 ) showed that (a) the genetic deletion of aquaporin-1 (AQP1) and the Rh complex (mainly RhAG) together reduce O 2 permeability of RBC membranes ( P M, O2 ) by ~55%, and (b) the double knockout (dKO) of AQP1 and RhAG plus pCMBS (sulfhydryl reagent and nonspecific inhibitor of membrane proteins that is nonetheless excluded from the RBC interior) reduces P M, O2 by ~91%. In order to identify the unknown membrane protein(s) that may function as O 2 channels and contribute to the missing 91% – 55% = ~36% of P M, O2 , we use comparative physiology. Using stopped-flow absorbance spectroscopy to monitor the hemoglobin (Hb) absorbance spectrum, we studied the oxygen-offloading rate from hemoglobin ( k HbO2 ) of RBCs from genetically diverse mouse strains. We compared two different mouse strains purchased from Jackson Labs—C57BL/6J and BALB/cJ with our standard mouse strain that ultimately derives from mice received from UCSF—C57BL/6Case. We found that, compared with C57BL/6Case, C57BL/6J mice have a k HbO2 for intact RBCs that is ~22% higher (N=18, 9 male plus 9 female, p<0.01), and a k HbO2 for pure hemolysate that is ~7% lower (N=12, 6 male plus 6 female, p<0.01). Comparing with C57BL/6Case, BALB/cJ mice have a k HbO2 for intact RBCs that is ~9% lower (N=18, 9 male plus 9 female, p<0.01), and a k HbO2 of pure hemolysate that is ~10% higher (N=12, 6 male plus 6 female, p<0.01). Because changes in k HbO2 of pure hemolysate are opposite in direction to changes in k HbO2 of intact RBCs, differences in Hb cannot contribute to the observed differences in O 2 -offloading rates in the two Jackson strains. Likewise, changes of mean corpuscular volume (MCV) and mean corpuscular hemoglobin concentration (MCHC), together, would only produce minor percent changes of k HbO2 in either C57BL/6J or BALB/cJ mice. Thus, it is likely that the differences in k HbO2 for intact RBCs among the three mouse strains reflect major differences in the O 2 permeabilities of the RBC membranes. Planned comparative analyses of morphology (e.g., major diameter of biconcave discs), proteomics, lipidomics, and genomics (for deduced amino-acid sequences of proteins) will provide valuable insight into the contribution of specific membrane proteins and lipids to the relatively large differences in k HbO2 of intact RBCs between C57BL/6J and BALB/cJ mice. Abstract:Background: Hypoxia plays a crucial role in regulating various cellular functions, including ion-transport mechanisms in the kidney. The sodium-chloride co-transporter (NCC) is essential for sodium reabsorption in the distal convoluted tubule (DCT). However, the effects of hypoxia on NCC expression and its regulatory pathways remain unclear. We aimed to explore the effects and potential mechanisms of hypoxia on NCC in vitro. Methods: mDCT15 cells were treated with cobalt chloride (CoCl2) at a concentration of 300 μmol/L to induce hypoxia. The cells were harvested at different time points, namely 30 min, 1 h, 6 h, and 24 h, and the expression of NCC and CaMKII-β was analyzed using Western blot. Results: A time-dependent upregulation of NCC and CaMKII-β expression in response to CoCl2-induced hypoxia. KN93 reversed the effect of CoCl2 on NCC and phosphorylated NCC expression. Conclusions: Hypoxia, mediated through cobalt chloride treatment, upregulates NCC expression via the CaMKII-β pathway in mDCT15 cells.

Abstract:Abstract Background The use of dialysis fluids (DFs) during haemodialysis has been associated with increased oxidative stress and reduced serum magnesium (Mg) levels, contributing to chronic inflammation. Since the role of Mg in modulating immune function and reducing oxidative stress has been demonstrated, the aim of this study was to characterize in vitro whether increasing the Mg concentration in DFs could protect immune cells from oxidative stress and damage. Methods The effect of citrate [citrate dialysis fluid (CDF), 1 mM] or acetate [acetate dialysis fluid (ADF), 3 mM] dialysates with low (0.5 mM; routinely used) or high (1 mM, 1.25 mM and 2 mM) Mg concentrations was assessed in THP-1 human monocytes. The levels of reactive oxygen species (ROS), malondialdehyde (MDA) and oxidized/reduced (GSSG/GSH) glutathione were quantified under basal and inflammatory conditions (stimulation with lipopolysaccharide, LPS). Results The increase of Mg in CDF resulted in a significant reduction of ROS production under basal and inflammatory conditions (extremely marked in 2 mM Mg; P < 0.001). These effects were not observed in ADF. Interestingly, in a dose-dependent manner, high Mg doses in CDF reduced oxidative stress in monocytes under both basal and inflammatory conditions. In fact, 2 mM Mg significantly decreased the levels of GSH, GSSG and MDA and the GSSG/GSH ratio in relation to 0.5 mM Mg. Conclusions CDF produces lower oxidative stress than ADF. The increase of Mg content in DFs, especially in CDF, could have a positive and protective effect in reducing oxidative stress and damage in immune cells, especially under inflammatory conditions.
