Abstract:Cells of the obligate halophile Halobacterium cutirubrum were grown to the stationary phase in the presence of 32P-orthophosphate, 35S-sulfate, or 14C-labelled precursors (acetate, malonate, mevalonate, glycerol). 32P was incorporated into the phosphatide components in proportion to their relative concentrations, thus: phosphatidyl glycerophosphate (diether analogue) > unidentified minor phosphatide > phosphatidyl glycerol (diether analogue). 35S-Sulfate was incorporated mostly into the glycolipid sulfate ester component, but small amounts of 35S also appeared in a more polar sulfolipid and in a minor unidentified sulfolipid.14C-Mevalonate was the most efficient precursor for the phytanyl chains, followed in decreasing order by acetate and glycerol. Abstract:Photothermal therapy (PTT) and magnetic hyperthermia have emerged as promising techniques for augmenting conventional cancer treatments, such as chemotherapy and radiotherapy. However, both treatments have their inherent drawbacks, such as PTT being limited by the depth of laser penetration while magnetothermal therapy being limited by the high concentration compensation brought by a low specific absorption rate. To address this challenge, we have successfully synthesized multifunctional nanohybrids comprising magnetic nanoparticles Mn0.5Zn0.5Fe2O4 (MZF) with an average diameter of 8.5 nm coated with a light-absorbing polymer responsive to near-infrared radiation, polypyrrole (PPy). Subsequently, the nanocomposite was functionalized with poly(ethylene glycol) (PEG) to obtain MZF/PPy-PEG nanoparticles with an average diameter of 190 nm. The core–shell MZF/PPy-PEG demonstrates an effective magnetic heating effect, a photothermal stability, and a photothermal conversion efficiency of 34.6%. In addition, the PPy shell, being an organic polymer, exhibits the capability to encapsulate anticancer drug (DOX), enabling synergistic magnetothermal–PTT and chemotherapy . Our findings offer a promising approach for synergistic magneto-PTT and chemotherapy, providing an efficient treatment for cancer.

Surface design and preparation of multi-functional magnetic nanoparticles for cancer cell targeting, therapy, and imaging

Abstract:Recently, theranostic candidates based on superparamagnetic iron oxide nanoparticles (SPIONs) providing the combination of therapy and diagnosis have become one of the most promising system in cancer research. However, poor stability, premature drug release, lack of specific tumor cell targeting, and complicated multi-step synthesis processes still hinder them for potential clinical applications. In this research, the multi-functional magnetic nanoparticles (MNPs-DOX) were prepared via a simple assembly process for targeted delivery of doxorubicin (DOX) and enhanced magnetic resonance (MR) imaging detection. Firstly, the multi-functional copolymer coating, polyamidoamine (PAMAM), was designed and synthesized by Michael addition reaction, where N,N-bis(acryloyl)cystamine served as backbone linker, and DOX, dopamine (DA), and polyethylene glycol (PEG) acted as comonomers. The PAMAM was then directly assembled to the surface of SPIONs by the ligand exchange reaction with SPIONs forming the MNPs-DOX. The hydrophilic PEG moieties provide the nanoparticles with colloidal stability and good-dispersity in aqueous solution. Comparing with the quick release of free DOX, the drug release behavior of MNPs-DOX exhibited a sustained drug release. Because the chemical cleavage of disulfide in the polymer backbone, a high cumulative drug release up to 60% in GSH within 48 h was observed, rather than only 26% in PBS (pH 7.4) without GSH. The MR imaging detection experiment showed that the MNPs-DOX had an enhanced T-2 relaxivity of 126 mM(-1) S-1 for MR imaging. The results of the cytotoxicity assays showed a remarkable killing effect of cancer cells by MNPs-DOX due to the FA tumor-targeting ligand, comparing with non-targeted drug molecules. All the results showed that the as prepared multi-functional magnetic nanoparticles may serve as a promising theranostic candidate for targeted anticancer drug delivery and efficient detection through MR imaging in medical application.
pH sensitive core-shell magnetic nanoparticles for targeted drug delivery in cancer therapy

Abstract:In the last decade, nanobiotechnology has evolved rapidly with an extensive impact on biomedical area. In order to improve bioavailability and minimize adverse effects, drug delivery systems based on magnetic nanocomposites are under development mainly for cancer imaging and antitumor therapy. In this regard, pH sensitive core-shell magnetic nanoparticles (NPs) with accurate controlled size and shape are synthesized by various modern methods, such as homogeneous precipitation, coprecipitation, microemulsion or polyol approaches, high temperature and hydrothermal reactions, sol-gel reactions, aerosol÷vapor processes and sonolysis. Due to their unique combined physico-chemical and biological properties (such as higher dispensability, chemical and thermal stability, biocompatibility), pH responsive core-shell magnetic NPs are widely investigated for controlled release of cytostatic drugs into the tumor site by means of pH change: magnetite@silicon dioxide (Fe3O4@SiO2), Fe3O4@titanium dioxide (TiO2), β-thiopropionate-polyethylene glycol (PEG)-modified Fe3O4@mSiO2, Fe3O4 NPs core coated with SiO2 with an imidazole group modified PEG-polypeptide (mPEG-poly-L-Asparagine), polyacrylic acid (PAA) and folic acid (FA) coating of the iron oxide NP core, methoxy polyethylene glycol-block-polymethacrylic acid-block-polyglycerol monomethacrylate (MPEG-b-PMAA-b-PGMA) attached by a PGMA block to a Fe3O4 core, PEG-modified polyamidoamine (PAMAM) dendrimer shell with Fe3O4 core and mesoporous silica coated on Fe3O4, mostly coated with an anticancer drug. This review paper highlights the modern research directions currently employed to demonstrate the utility of the pH responsive core-shell magnetic NPs in diagnosis and treatment of oncological diseases.

Abstract:Recently, increasing attention has been paid to magneto-conjugated polymer core–shell nanoparticles (NPs) as theranostic platforms. However, the utilization of surfactants and extra oxidizing agents with potential toxicity in synthesis, the lack of general methods for the controlled synthesis of various kinds of magnetic NP (MNP)@conjugated polymer NPs, and the difficulty of obtaining balanced magneto-optical properties have greatly limited the applications of magneto-conjugated polymers in theranostics. We developed an in situ surface polymerization method free of extra surfactants and oxidizing agents to synthesize MNP@polypyrrole (PPy) NPs with balanced, prominent magneto-optical properties. MNP@PPy NPs with an adjustable size, different shapes, and a controlled shell thickness were obtained using this method. The method was extended to synthesize other MNP-conjugated polymer core–shell NPs, such as MNP@polyaniline and MNP@poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate) (PEDOT:PSS). We discuss the formation mechanism of the proposed method according to our experimental results. Finally, using the optical and magnetic properties of the obtained MNP@PEDOT:PSS NPs, in vivo multimodal imaging-guided hyperthermia was induced in mice, achieving an excellent tumor-ablation therapeutic effect. Our work is beneficial for extending the application of MNP-conjugated polymer core–shell NPs in the biomedical field.

Abstract:Although chemotherapy with magnetic nanocarriers has received a significant advancement in the field of cancer treatment, multimodal diagnosis and combitorial therapy using a single nanoplatform will have much better efficacy in achieving greater heights. Here, we have constructed a smart theranostic system by combining pH sensitive tartaric acid stabilizedFe3O4magnetic nanocarriers (TMNCs) with SPECT imaging and chemotherapeutic agents for image-guided chemo-hyperthermia therapy. The carboxyl enriched exteriors of TMNCs provide sites for conjugation of chemotherapeutic drug (doxorubicin hydrochloride, DOX) and radiolabeling (Cerium-141, 141Ce). The usage of 145.4 keV gamma made it an ideal choice for in-vivo SPECT-CT imaging which has shown retention of nanoformulation in tumor site even after 28 days. Further, the TMNCs show very high transverse relaxation rate (r2) of 171 mM-1s-1 which is higher than the clinically approved magnetic resonance imaging (MRI) contrast agents like ferumoxtran (65 mM−1 s−1) and ferumoxides (120 mM−1 s−1). Further, the developed drug loaded hybrid platform showed significantly higher cytotoxicity towards breast cancer cells and the same is augmented by in-vitro magnetic hyperthermia. Bright field microscopy and cell cycle analysis suggests that the cell death occurred through induction of G2-M arrest and subsequent apoptosis. These findings clearly suggested the potential ability of developed hybrid nanoplatform for image guided combination therapy.

Abstract:A combination of multiple therapies is conducive to the treatment efficacy of cancer. Chemotherapy combined with magnetothermal therapy provides an outstanding synergy for cancer treatment. In this study, magnetic mesoporous silica nanoparticles (MMSN) were developed as a multipurpose nanoplatform for doxorubicin (DOX) delivery and magnetothermal treatment of cancer simultaneously. The MMSN were composed of Fe 3 O 4 and mesoporous silica as core and shell. The folic acid (FA) linked chitosan (CS) was conjugated at the outlets of MMSN (MMSN-SS-FA) using disulfide bonds to avert the premature release of the drug in mesopores of MMSN, target folate receptor-overexpressed cancer cells and increase the safety and physiological stability of MMSN. The temperature of the MMSN suspension could be elevated from 37 °C to 45 °C at the concentration of 1 mg/mL under an alternating magnetic field (AMF), which exceeded the tumor ablating temperature. The in vitro drug release proved that the DOX/MMSN-SS-FA displayed the redox/pH/AMF multiple-responsive release. The cellular uptake results indicated that the MMSN-SS-FA could target folate receptor-overexpressed HeLa cells. The DOX/MSNS-SS-FA with AMF had the highest cytotoxicity at the equivalent drug concentration towards HeLa cells with a combination index (CI) of 0.276, demonstrating the synergistic treatment effect of chemotherapy and magnetic-thermal therapy. Meanwhile, the DOX/MMSN-SS-FA+AMF showed the strongest tumor inhibitory effect of 84 % on tumor growth. Therefore, the constructed MMSN-SS-FA with good biocompatibility and biosafety can act as multifunctional nano-platforms for tumor treatment.

Abstract:This paper reports a type of multifunctional hybrid nanoparticle (NP) composed of gold nanocrystals coated on and/or embedded in a magnetite-fluorescent porous carbon core-shell NP template (Fe3O4@PC-CDs-Au) for biomedical applications, including magnetic/NIR-responsive drug release, multicolor cell imaging, and enhanced photothermal therapy. The synthesis of the Fe3O4@PC-CDs-Au NPs firstly involves the preparation of core-shell template NPs with magnetite nanocrystals clustered in the cores and fluorescent carbon dots (CDs) embedded in a porous carbon shell, followed by an in situ reduction of silver ions (Ag+) loaded in the porous carbon shell and a subsequent replacement of Ag NPs with Au NPs through a galvanic replacement reaction using HAuCl4 as a precursor. The Fe3O4@PC-CDsAu NPs can enter the intracellular region and light up mouse melanoma B16F10 cells in multicolor mode. The porous carbon shell, anchored with hydrophilic hydroxyl/carboxyl groups, endows the Fe3O4@PC-CDs-Au NPs with excellent stability in the aqueous phase and a high loading capacity (719 mg g(-1)) for the anti-cancer drug doxorubicin (DOX). The superparamagnetic Fe3O4@PC-CDs-Au NPs with a saturation magnetization of 23.26 emu g(-1) produce localized heat under an alternating magnetic field, which triggers the release of the loaded drug. The combined photothermal effects of the Au nanocrystals and the CDs on/in the carbon shell can not only regulate the release rate of the loaded drug, but also efficiently kill tumor cells under NIR irradiation. Benefitting from their excellent optical properties, their magnetic field and NIR light-responsive drug release capabilities and their enhanced photothermal effect, such nanostructured Fe3O4@PC-CDs-Au hybrid NPs are very promising for simultaneous imaging diagnostics and high efficacy therapy.

Abstract:As the combination of photothermal therapy (PTT) with immunotherapy provides an effective strategy in cancer treatment, a magnetic nanoparticle delivery system was constructed to load indocyanine green (ICG) and immunostimulator R837 hydrochloride (R837) for spatio-temporally PTT/immunotherapy synergism in cancer. This delivery system is composed of Fe3O4 magnetic nanoparticles (MPs) as the core to load ICG and polyethylene glycol polyphenols (DPA-PEG) as the coating layer to load R837, which formed R837 loaded polyphenols coating ICG loaded magnetic nanoparticles (MIRDs). After intravenous injection, the formed MIRDs resulted in long circulation, magnetic resonance imaging (MRI) guides, and magnetic targeting. Once targeting to the tumor, the MIRDs with the near-infrared (NIR) irradiation caused tumor ablation and resulted in tumor-associated antigens releasing to induce the body's immunological response, which was markedly improved it to attack the tumors with the R837 releasing from the outer DPA-PEG. In this case, the synergism of the PTT and immunotherapy inhibited tumor growth, metastasis and recurrence, which resulted in potent anticancer therapeutic effects with few side effect.

Abstract:Magnetic nanoparticles(NPs) are characterized by a rich variety of properties. Because of their excellent physical and chemical properties, they have come to the fore in biomedicine and other fields. The magnetic NPs were extensively studied in magnetic separation of cells, targeted drug delivery, tumor hyperthermia, chemo-photothermal therapy, magnetic resonance imaging (MRI) and other biomedical fields. Magnetic NPs are increasingly used in magnetic resonance imaging (MRI) based on their inherent magnetic targeting, superparamagnetic enzyme-like catalytic properties and nanoscale size. Poly(lactic-co-glycolic acid) (PLGA) is a promising biodegradable material approved by FDA and EU for drug delivery. Currently, PLGA-based magnetic nano-drug delivery systems have attracted the attention of researchers. Herein, we achieved the effective encapsulation of sized-controlled polyethylene glycol-3,4-dihydroxy benzyl-amine-coated superparamagnetic iron oxide nanoparticles (SPIO NPs) and euphorbiasteroid into PLGA nanospheres via a modified multiple emulsion solvent evaporation method (W1/O2/W2). NPs with narrow size distribution and acceptable magnetic properties were developed that are very useful for applications involving cancer therapy and MRI. Furthermore, SPIO-PLGA NPs enhanced the MRI T2 relaxation properties of tumor sites.The prepared SPIO NPs and magnetic PLGA nanospheres can be promising magnetic drug delivery systems for tumor theranostics. This study has successfully constructed a tumor-targeting and magnetic-targeting smart nanocarrier with enhanced permeability and retention, multimodal anti-cancer therapeutics and biodegradability, which could be a hopeful candidate for anti-tumor therapy in the future.

Abstract:The use of targeted chemotherapy is a promising solution to mitigate the side effects and dosage of drugs. This research focuses on the development of magnetic microspheres (MMS) based drug carriers for targeted chemotherapy, formulated with iron oxide nanoparticles (Fe3O4 NPs) and poly (l-lactic acid) (PLA) loaded with the antibiotic drug Ciprofloxacin (CIF). In this study, Fe3O4 NPs were synthesized using pomegranate peel extract as a natural reducing and stabilizing agent. The double emulsification method (W1/O/W2) was employed to produce Fe3O4@LEC-CIF-PLA-MMS, which were characterized using various spectral and microscopic techniques. The drug encapsulation efficiency for Fe3O4@LEC-CIF-PLA-MMS was found to be 80.7 %. The in vitro drug release of CIF from Fe3O4@LEC-CIF-PLA-MMS induced by H2O2 and GSH- stimuli was found to be 87.55 % and 82.32 %, respectively in acidic pH 4.5. Notably, the magnetically triggered drug release behaviour of Fe3O4@LEC-CIF-PLA-MMS (93.56 %) was assessed in acidic pH environment upon exposure to low-frequency alternating magnetic field (LF-AMF). Fe3O4@LEC-CIF-PLA-MMS demonstrated significantly enhanced in vitro cytotoxicity (IC50 = 0.8 ± 0.03 μg/mL) against the HeLa-S3 cancer cell lines. Nevertheless, these research findings highlight the potential of Fe3O4@LEC-CIF-PLA-MMS for further development as a chemotherapeutic agent and hold promise for the future of targeted cancer treatment.
Biocompatible superparamagnetic core-shell nanoparticles for potential use in hyperthermia-enabled drug release and as an enhanced contrast agent

Abstract:Abstract Superparamagnetic iron oxide nanoparticles (SPIONs) and core-shell type nanoparticles, consisting of SPIONs coated with mesoporous silica and/or lipid, were synthesised and tested for their potential theranostic applications in drug delivery, magnetic hyperthermia and as a contrast agent. Transmission Electron Microscopy (TEM) confirmed the size of bare and coated SPIONs was in the range of 5–20 nm and 100–200 nm respectively. The superparamagnetic nature of all the prepared nanomaterials as indicated by Vibrating Sample Magnetometry (VSM) and their heating properties under an AC field confirm their potential for hyperthermia applications. Scanning Column Magnetometry (SCM) data showed that extrusion of bare-SPION (b-SPION) dispersions through a 100 nm polycarbonate membrane significantly improved the dispersion stability of the sample. No sedimentation was apparent after 18 h compared to a pre-extrusion estimate of 43% settled at the bottom of the tube over the same time. Lipid coating also enhanced dispersion stability. Transversal relaxation time (T2) measurements for the nanoparticles, using a bench-top relaxometer, displayed a significantly lower value of 46 ms, with a narrow relaxation time distribution, for lipid silica coated SPIONs (Lip-SiSPIONs) as compared to that of 1316 ms for the b-SPIONs. Entrapment efficiency of the anticancer drug, Doxorubicin (DOX) for Lip-SPIONs was observed to be 35% which increased to 58% for Lip-SiSPIONs. Moreover, initial in-vitro cytotoxicity studies against human breast adenocarcinoma, MCF-7 cells showed that % cell viability increased from 57% for bSPIONs to 82% for Lip-SPIONs and to 87% for Lip-SiSPIONs. This suggests that silica and lipid coatings improve the biocompatibility of bSPIONs significantly and enhance the suitability of these particles as drug carriers. Hence, the magnetic nanomaterials prepared in this work have potential theranostic properties as a drug carrier for hyperthermia cancer therapy and also offer enhancement of contrast agent efficacy and a route to a significant increase in dispersion stability.

Herein, we have developed a biomimetic drug delivery system consisting of superparamagnetic iron oxide nanoparticles (SPIONs) coated by platelet membranes (PM) for carrying Paclitaxel (PTX) to exploit the synergism effect of chemotherapy and magnetic hyperthermia. Controlled-release PTX nanoparticles exhibited consistent behavior over time, indicating no significant difference in release between SPION/PTX and SPION/PTX/PM at pH 7.4. However, at pH 5.5, improved release was observed, specifically a 1.4-fold increase for SPION/PTX/PM. The confocal and flow cytometry results show an enhancement in the cellular uptake of SPION/PTX/PM nanoparticles, with an average fluorescence intensity of 142 ±12.5. MTT results indicated that SPION/PTX/PM demonstrates superior cytotoxic effects compared to SPION/PTX and PTX alone, showing an IC50 value of 5 micrograms per milliliter after 48 hours of treatment. Furthermore, the IC50 decreased to 1 microgram per milliliter when an alternating magnetic field was applied. Hence, the in vivo results and histopathological staining showed that the SPION/PTX/PM-AFM treatment group exhibited the highest rate of tumor growth inhibition, reaching nearly 92.14 %. These findings highlight the potential of using platelet membrane-coated nanoparticles for targeted delivery, combining magnetic hyperthermia and chemotherapy to minimize chemotherapy's undesirable effects while maximizing therapeutic outcomes.

Abstract:Combination chemo-photothermal therapy with nanomaterials can reduce the dose of chemotherapeutic drugs required for effective cancer treatment by minimizing toxic side effects while improving survival times. Toward this end, we prepare hyaluronic acid (HA)-modified poly(lactic-co-glycolic acid) (PLGA) magnetic nanoparticles (MNP) for the CD44 receptor-mediated and magnetic field-guided dual-targeted delivery of cisplatin (CDDP). By co-encapsulating the CDDP and oleic acid-coated iron oxide MNP (IOMNP) in PLGA, the PMNPc was first prepared in a single emulsification/solvent evaporation step and successively surface modified with chitosan and HA to prepare the HA/PMNPc. Spherical HA/PMNPc nanoparticles of ~300 nm diameter can be prepared with 18 and 10% (w/w) loading content of CDDP and IOMNP and a pH-sensitive drug release to facilitate the endosomal release of the CDDP after intracellular uptake. This leads to the higher cytotoxicity of the HA/PMNPc toward the U87 glioblastoma cells than free CDDP with reduced IC50, a higher cell apoptosis rate, and the enhanced expression of cell apoptosis marker proteins. Furthermore, the nanoparticles show the hyperthermia effect toward U87 after short-term near-infrared (NIR) light exposure, which can further elevate the cell apoptosis/necrosis rate and upregulate the HSP70 protein expression due to the photothermal effects. The combined cancer therapeutic efficacy was studied in vivo using subcutaneously implanted U87 cells in nude mice. By using dual-targeted chemo-photothermal combination cancer therapy, the intravenously injected HA/PMNPc under magnetic field guidance and followed by NIR laser irradiation was demonstrated to be the most effective treatment modality by inhibiting the tumor growth and prolonging the survival time of the tumor-bearing nude mice.

Abstract:Photothermal therapy (PTT) has become one of the most promising therapies in cancer treatment as its noninvasiveness, high selectivity, and favorable compliance in clinic. However, tumor thermotolerance and distal metastasis reduce its efficacy, becoming the bottleneck of applying PTT in clinic. In this study, a chidamide-loaded magnetic polypyrrole nanocomposite (CMPP) has been fabricated as a visualized cancer photothermal agent (PTA) to counter tumor thermotolerance and metastasis. The efficacy of CMPP was characterized by in vitro and in vivo assays. As a result, this kind of magnetic polypyrrole nanocomposites were black spherical nanoparticles, possessing a favorable photothermal effect and the suitable particle size of 176.97 ± 1.45 nm with a chidamide loading rate of 12.92 ± 0.45%. Besides, comparing with PTT, CMPP exhibited significantly higher cytotoxicity and cellular apoptosis rate in two tumor cell lines (B16-F10 and HepG2). In vivo study, the mice showed obvious near-infrared (NIR) and magnetic resonance imaging (MRI) dual-modal imaging at tumor sites and sentinel lymph nodes (SLNs); on the other hand, magnetic targeting guided CMPP achieved a cure level on melanoma-bearing mice through preventing metastasis and thermotolerance. Overall, with high loading efficiency of chidamide and strong magnetic targeting to tumor sites and SLNs, CMPP could significantly raise efficiency of PTT by targeting tumor thermotolerance and metastasis, and this strategy may be exploited therapeutically to upgrade PTT with MPP as one of appropriate carriers for histone deacetylase inhibitors (HDACis).

Abstract:Magnetic nanoparticles (MNPs) are of great scientific interest for the researchers from a wide range of biomedicine applications, including clinical magnetic resonance imaging (MRI), cancer magnetic hyperthermia, triggered drug release, and catalysis. Especially, the high performance of these MNPs, such as controllable size and morphology, superior magnetism, high magnetically induced heating effects, favorable biocompatibility, accurate targeting ability, and long circulation, is crucial for their effective application in clinical diagnosis and therapy of disease. In a brief overview, the controlled chemical design and synthesis, the formation mechanism, as well as the surface functional strategy of MNPs are firstly presented. High-quality MNPs are typically prepared through a thermal decomposition of organometallic compounds in high-boiling organic solvent containing surfactants, in which the shape, size, composition, surface states, magnetic characteristics, biocompatibility, and some other important features are synthetically controlled. Understanding the interplay of these parameters is critical for the effective imaging and therapeutics, which include MRI relaxivity, heat emission, and attractive forces. We then discuss approaches to constructing versatile MNP probes as MRI contrast agents with high sensitivity, with which the sensitive and target-specific observation of biological imaging at the molecular and cellular levels is possible. Finally, we present an overview of recent breakthroughs in the development of MNP-based magnetic hyperthermia. The purpose of this section is to address different factors that affect the targeting, heating, and biodistribution to safely control the therapeutic efficacy of targeted magnetic hyperthermia (TMH).

Abstract:Balancing biocompatibility and drug-loading efficiency in nanoparticles presents a significant challenge. In this study, we describe the facile fabrication of poly (acrylic acid)-mesoporous zinc phosphate/polydopamine (PAA-mZnP/PDA) Janus nanoparticles (JNPs). The PDA half-shell itself can serve as a photothermal agent for photothermal therapy (PTT), as well as to offers sites for polyethylene glycol (PEG) to enhance biocompatibility. Concurrently, the mesoporous ZnP core allows high loading of doxorubicin (DOX) for chemotherapy and the Cy5.5 dye for fluorescence imaging. The resultant PAA-mZnP/PDA-PEG JNPs exhibit exceptional biocompatibility, efficient drug loading (0.5 mg DOX/1 mg JNPs), and dual pH/NIR-responsive drug release properties. We demonstrate the JNPs' satisfactory anti-cancer efficacy, highlighting the synergistic effects of chemotherapy and PTT. Furthermore, the potential for synergistic fluorescence imaging-guided chemo-phototherapy in cancer treatment is illustrated. Thus, this work exemplifies the development of biosafe, multifunctional JNPs for advanced applications in cancer theranostics. Statement of Significance : Facile fabrication of monodispersed nanomedicine with multi-cancer killing modalities organically integrated is nontrivial and becomes more challenging under the biocompatibility requirement that is necessary for the practical applications of nanomedicines. In this study, we creatively designed PAA-mZnP/PDA JNPs and fabricated them under mild conditions. Our method reliably yields uniform JNPs with excellent monodispersity. To maximize functionalities, we achieve fourfold advantages including efficient drug/fluorescent dye loading, PTT, pH/NIR dual-responsive properties, and optimal biocompatibility. The as-fabricated JNPs exhibit satisfactory anti-cancer performance both in vitro and in vivo , and demonstrate the potential of JNPs in fluorescence imaging-guided synergistic cancer chemo-phototherapy. Overall, our research establishes a pathway in versatile inorganic/polymer JNPs for enhanced cancer diagnosis and therapy.

Abstract:We developed an intelligent device capable of controlled release of anti-cancer drugs. The nano-carrier consists of two key components: firstly, magnetic nanoparticles were synthesized through co-precipitation and then functionalized for attaching anti-cancer drugs; secondly, a polymer (poly( N -isopropylacrylamide- co -acrylic acid)) sensitive to pH and temperature was synthesized and employed to encapsulate the drug-loaded magnetic nanoparticles. This responsive polymer exhibits a lower critical solution temperature (LCST) of 38.5 °C, indicating a phase transition behavior. Numerous techniques and analyses, including Fourier transform infrared spectroscopy, thermogravimetric analysis, zeta potentials, scanning electron microscope, and transmission electron microscopy, were employed to confirm the successful execution of the aforementioned process. In vitro release assessments of the anti-cancer drug, doxorubicin, were conducted across various media (pH 5–8 and temperatures ranging from 20 to 40 °C). The outcomes revealed higher drug releases at temperatures exceeding the LCST (40 °C) and at slightly acidic pH levels (5–5.3). Notably, compared to the effectiveness of the unloaded drug, the magnetic smart polymer loaded with DOX exhibited a more potent cytotoxic effect. Considering these results, this nano-carrier emerges as a promising candidate for targeted therapeutic delivery to cancerous tissues.

Abstract:Few-layer, carbon-coated, iron (C/Fe) magnetic nanoparticles (MNPs) were synthesized with controlled sizes ranging from 7 to 9 nm. The additional loading of two anti-cancer drugs, doxorubicin and erlotinib, was achieved through - stacking onto the carbon shells. Controlled release of the drugs was successfully triggered by radio frequency (RF) heating or pH variation. Based on the experimental results, C/Fe MNPs act as heat-inducing agents and are able to thermally destroy cancer cells when RF is applied. It was found that the combination of anti-cancer drugs (in particular a low dose of doxorubicin) and RF treatment demonstrates a synergistic effect in inducing cell death in pancreatic cancer cells. Our findings demonstrate that MNPs can be used as highly efficient multimodal nanocarrier agents for an integrated approach to cancer treatment involving triggered delivery of antineoplastic drugs and RF-induced thermal therapy.
