Abstract:Single-nanoparticle detection has received tremendous interest due to its significance in fundamental physics and biological applications. Here, we demonstrate an optical nanofibre-enabled microfluidic sensor for the detection and sizing of nanoparticles. Benefitting from the strong evanescent field outside the nanofibre, a nanoparticle close to the nanofibre can scatter a portion of the field energy to the environment, resulting in a decrease in the transmitted intensity of the nanofibre. On the other hand, the narrow and shallow microfluidic channel provides a femtoliter-scale detection region, making nanoparticles flow through the detection region one by one. By real-time monitoring of the transmitted intensity of the nanofibre, the detection of a single polystyrene (PS) nanoparticle as small as 100 nm in diameter and exosomes in solution is realised. Based on a statistical analysis, the mean scattering signal is related to the size of the nanoparticle. Experimentally, a mixture of nanoparticles of different diameters (200, 500, and 1000 nm) in solution is identified. To demonstrate its potential in biological applications, high-throughput counting of yeasts using a pair of microchannels and dual-wavelength detection of fluorescently labelled nanoparticles are realised. We believe that the developed nanoparticle sensor holds great potential for the multiplexed and rapid sensing of diverse viruses. Tunable plasmonic tweezers based on graphene nano-taper for nano-bio-particles manipulation: numerical study

Abstract:We take advantage of graphene nano-taper plasmons to design tunable plasmonic tweezers for neuroblastoma extracellular vesicles manipulation. It consists of Si/SiO2/Graphene stack topped by a microfluidic chamber. Using plasmons of isosceles-triangle-shaped graphene nano-taper with a resonance frequency of 6.25 THz, the proposed device can efficiently trap the nanoparticles. The plasmons of graphene nano-taper generate a large field intensity in the deep sub-wavelength area around the vertices of the triangle. We show that by engineering the dimensions of the graphene nano-taper and an appropriate choice of its Fermi energy, the desired near-field gradient force for trapping can be generated under relatively low-intensity illumination of the THz source when the nanoparticles are placed near the front vertex of the nano-taper. Our results show that the designed system with graphene nano-taper of L = 1200 nm length and W = 600 nm base size and THz source intensity of I = 2 mW/µm2, can trap polystyrene nanoparticles with diameters of D = 140, 73, and 54 nm, and with trap stiffnesses of ky = 9.9 fN/nm, ky = 23.77 fN/nm, and ky = 35.51 fN/nm at Fermi energies of Ef = 0.4, 0.5, and 0.6 eV, respectively. It is well known that the plasmonic tweezer as a high-precision and non-contact means of control has potential applications in biology. Our investigations demonstrate that the proposed tweezing device with L = 1200 nm, W = 600 nm, and Ef = 0.6 eV can be utilized to manipulate the nano-bio-specimens. So that, at the given source intensity, it can trap the neuroblastoma extracellular vesicles, which are released by neuroblastoma cells and play an important role in modulating the function of neuroblastoma cells and other cell populations, as small as 88 nm at the front tip of isosceles-triangle-shaped graphene nano-taper. The trap stiffness for the given neuroblastoma extracellular vesicle is obtained as ky = 17.92 fN/nm.

Abstract:Optical tweezers are key tools to trap and manipulate nanoparticles in a non-invasive way, and have been widely used in the biological and medical fields. We present an integrated multifunctional 2D plasmonic optical tweezer consisting of an array of graphene discs and the substrate circuit. The substrate circuit allows us to apply a bias voltage to configure the Fermi energy of graphene discs independently. Our work is based on numerical simulation of the finite element method. Numerical results show that the optical force is generated due to the localized surface plasmonic resonance (LSPR) mode of the graphene discs with Fermi Energy Ef = 0.6 eV under incident intensity I = 1 mW/μm2, which has a very low incident intensity compared to other plasmonic tweezers systems. The optical forces on the nanoparticles can be controlled by modulating the position of LSPR excitation. Controlling the position of LSPR excitation by bias voltage gates to configure the Fermi energy of graphene disks, the nanoparticles can be dynamically transported to arbitrary positions in the 2D plane. Our work is integrated and has multiple functions, which can be applied to trap, transport, sort, and fuse nanoparticles independently. It has potential applications in many fields, such as lab-on-a-chip, nano assembly, enhanced Raman sensing, etc.

Abstract:Since the discovery of graphene, its excellent physical properties have greatly improved the performance of optoelectronic devices and brought important technological advances to optical research and its applications. Here, graphene is introduced to the field of optical-tweezer technology and demonstrate a new graphene-based opto-thermoelectric tweezer. This technology not only reduces the incident light energy required by two orders of magnitude (compared with traditional optical tweezers), it also brings new advantages such as a much broader working bandwidth and a larger working area compared to those of widely researched gold-film-based opto-thermoelectric tweezers. Compared with gold film, graphene exhibits higher thermal conductivity and higher uniformity and is easier to process. Thus, it is found that even monolayer graphene provides stable trapping for particles in a broad bandwidth and that performance is enhanced as the number of graphene layers increases. Furthermore, parallel trap multiple particles as desired shapes can be easily generated with structured graphene patterns. This work demonstrates the enormous application potential of graphene in optical-tweezer technology and will promote their application to the trapping or concentration of cells and biomolecules as well as to microfluidics and biosensors.

Here, we introduce graphene into the field of optical tweezers technology and demonstrate a new thermoelectric optical tweezers technology based on graphene. This technology can not only reduce the incident light energy by 2 orders of magnitude (compared with traditional optical tweezers), but also bring new advantages such as much broader working bandwidth and larger working area than the thermoelectric optical tweezers based on gold film widely studied before. Compared with gold film, graphene has more novel characteristics like high thermal conductivity, high uniformity and easy process. Furthermore, structured graphene patterns can be easily generated to holographically trap multiple particles as desired shapes. This work verifies the great application potential of two-dimensional materials in op-tical tweezers technology, and it will promote more promising applications in cell trapping, tapping or concentration of biomolecules, microfluidics and biosensors.

Abstract:In this Letter, we report a nano-optical conveyor belt containing a couple of graphene nano-ribbon pairs (GNRPs) with graded lengths for the realization of trapping and transport of nanoparticles through manipulating the Fermi energy of graphene. In the mid-infrared waveband, graphene behaves like metals, and even more importantly, it supports tunable surface plasmon resonance (SPR) via manipulating its Fermi energy. The spectral responses of these graded graphene nano-ribbons with individual dimensions are dominated by their geometric parameters and surface conductivity, which is tunable and connected with Fermi energy. Optically trapped nano-particles can be transferred from one GNR to another under a single-wavelength polarized excitation beam by sequentially switching the Fermi energy via electric gating. The feasibility of our design to deliver target nano-particles or biomolecules over a long distance with nanometer accuracy is confirmed by the three-dimensional finite-difference time-domain technique. With the capacity to trap and transfer, our design offers a versatile scheme for conducting mechanical sample manipulation in many on-chip optofluidic applications.

Far-field position-tunable trapping of dielectric particles using a graphene-based plasmonic lens

Abstract:In this report, a graphene-based plasmonic lens is designed for far-field position-tunable trapping of dielectric particles at a wavelength of 1550 nm, in which target particles can be floated at a variable z-position, using a variable gate voltage applied to the graphene ribbons. Preventing proximity of the trapped particle and the metallic lens structure, we can diminish general thermal issues in plasmonic tweezers, while realizing higher degrees of freedom in studying target characteristics of the particles by achieving position-tunable 3D trapping. These advantageous aspects are impossible in conventional plasmonic tweezers, because of the highly evanescent nature of the plasmonic field at the metal interface. The proposed structure is comprised of two concentric circular slit-sets (S1, S2), each capable of sending a directive beam, which can lead to a constructive interference, and forming a subwavelength focal spot in the far-field. Taking advantage of the epsilon-near-zero (ENZ) behavior of graphene, each of the radiating slit-sets can be switched ON/OFF, with a radiation switching ratio of about 49, by applying a small electric pulse of 80 meV to change the Fermi energy of the corresponding graphene ribbon from 0.535 eV to 0.615 eV. Hence, inverting the radiation state of the designed lens, from (S1:ON, S2:OFF) to (S1:OFF, S2:ON), we can change the z-position of the focal trapping site from 5000 nm to 9800 nm. This configuration can be proposed as a new generation of long-range, electrostatically tunable 3D plasmonic tweezing, without the need for any external bulky optomechanical equipment.
Thermophoresis suppression by graphene layer in tunable plasmonic tweezers based on hexagonal arrays of gold triangles: numerical study

Abstract:Taking advantage of highly confined evanescent fields to overcome the free-space diffraction limit, we show plasmonic tweezers enable efficient trapping and manipulation of nanometric particles by low optical powers. In typical plasmonic tweezers, trapping/releasing particles is carried out by turning the laser power on and off, which cannot be achieved quickly and repeatedly during the experiment. We introduce hybrid gold-graphene plasmonic tweezers in which the trap stiffness is varied electrostatically by applying suitable voltages to a graphene layer. We show how the graphene layer absorbs the plasmonic field around the gold nanostructures in particular chemical potentials, allowing us to modulate the plasmonic force components and the trapping potential. We show graphene monolayer (bilayer) with excellent thermal properties enables more efficient heat transfer throughout the plasmonic tweezers, reducing the magnitude of thermophoretic force by about 23 (36) times. This thermophoresis suppression eliminates the risk of photothermal damage to the target sample. Our proposed plasmonic tweezers open up possibilities to develop tunable plasmonic tweezers with high-speed and versatile force-switching functionality and more efficient thermal performance.

Abstract:We numerically demonstrate a scheme of optofluidic sorting of nanoparticles on a silicon-based lab-on-a-chip system, with reconfigurable and multilevel sorting size thresholds. Size-dependent sorting of nanoparticles originates from the size-dependent distribution of trapping potential wells, which determines whether a nanoparticle can jump back and forth between parallel waveguides in weakly coupling condition. In numerical modeling, we find that it is easier for larger ones to transfer and the size threshold is power ratio dependent. By setting a thermal tuning ring-assisted Mach-Zehnder interferometer ahead, we can adjust effectively the input power ratio of the parallel waveguides as well as the potential well distribution across them, working as a thermal tuning reconfigurable sorting unit. Consequently, a tree cascaded unit with different thresholds is also presented to be a reconfigurable multilevel sorting unit. The proposed design offers a simple and ultracompact scheme for the reconfigurable and multilevel optofluidic sorting of nanoparticles on thermo-optofluidics chip for the first time.

Abstract:Developing kernels of the maize (Zea mays) hybrid W64A x W182E germinated precociously following fluridone treatment. Likewise, following premature drying, the kernels germinated upon subsequent rehydration. Tolerance of the aleurone layer to premature desiccation considerably preceded that of the embryo. The increase in alpha-amylase activity following premature drying was substantial and was equal to, or exceeded, the increase which occurred following normal maturation drying. In contrast, there was only a small increase in enzyme activity, regardless of the concentration of the supplied gibberellic acid, following fluridone treatment. Both fluridone and drying cause a decrease in abscisic acid content within the developing kernels. While this decline in growth regulator may permit kernels to germinate, alone this is not sufficient to permit an increase in alpha-amylase activity. Thus drying is necessary to sensitize the aleurone layer to gibberellin, and thereby elicit enzyme synthesis. For this tissue to achieve its full potential to produce alpha-amylase, it must not only be free of the inhibitory effects of abscisic acid, but it must also be competent to respond to gibberellin.
Exploring an In-Plane Graphene and Hexagonal Boron Nitride Array for Separation of Single Nucleotides

Abstract:Regular nanofluidic sieving structures are emerging as rapid and compatible on-chip techniques for biomolecular separation. Although the current nanofluidic sieving devices, mostly based on three-dimensional nanostructures, have achieved a separation resolution of similar to 20 nm, it is still far away from single-nucleotide resolution. Using all-atom molecular dynamics simulations, here we demonstrate a two-dimensional (2D) nanofluidic sieve consisting of an in-plane graphene (GRA)/hexagonal boron nitride (h-BN) nanoarray, which enables ultrahigh resolution in the successful separation of four types of single nucleotides. The alternating GRA and h-BN stripes can create size-dependent energy barriers for adsorbed nucleotides, which provide a strong modulation for their mobility, thus causing distinct band separations on the 2D surface. We further show that this 2D sieve is particularly sensitive when the sample dimensions are within the range from a half period to one period of the nanoarray. This 2D sieving structure may shed light on the development of lab-on-a-chip sequencing in the future.
Disruptive Forces in Metamaterial Tweezers for Trapping 20 nm Nanoparticles Based on Molecular Graphene Quantum Dots

Abstract:In recent years, plasmonic optical tweezers have been used to trap nanoparticles and study interactions with their environment. An unavoidable challenge is the plasmonic heating due to resonant excitation and the resulting temperature rise in the surrounding environment. In this work, we demonstrate trapping of custom-synthesized 20 nm nanoparticles based on molecular graphene quantum dots using metamaterial plasmonic tweezers. Superior trap stiffness values as high as 8.8 (fN/nm)/(mW/$\mu\mbox{m}^2$) were achieved with optical intensities lower than 1 mW/$\mu\mbox{m}^2$. By gradually increasing the laser intensity we identified a critical value beyond which the stiffness values dropped significantly. This value corresponded to a temperature rise of about 16$^o$C, evidently sufficient to create thermal flows and disrupt the trapping performance. We, therefore, identified a safe intensity regime for trapping nanoparticles without unwanted heat. Our platform can be used for efficient nanopositioning of fluorescent particles and quantum emitters in an array configuration, potentially acting as a single-photon source configuration.

Abstract:Optical tweezers use the force from a light beam to implement a precise gripping tool. Based purely on an optical principle, it works without any bodily contact with the object. In this paper we describe an optical tweezers that targets an application within the framework of nuclear magnetic resonance (NMR) spectroscopy of small objects, which are embedded inside a microfluidic channel that will be integrated in a micro-NMR detector. In the project's final stages, the whole system will be installed within the wide bore of a superconducting magnet. The aim is to precisely maintain the position of the object to be measured, without the use of susceptibility disturbing materials or geometries. In this contribution we focus on the design and construction of the tweezers. For the optical force simulation of the system we used a geometrical optics approach, which we combined with a ray fan description of the output beam of an optical system. By embedding both techniques within an iterative design process, we were able to design efficient optical tweezers that met the numerous constraints. Based on details of the constraints and requirements given by the application, different system concepts were derived and studied. Next, a highly adapted and efficient optical trapping system was designed and manufactured. After the components were characterized using vertical scanning interferometry, the system was assembled to achieve a monolithic optical component. The proper function of the optical tweezers was successfully tested by optical trapping of fused silica particles.

Abstract:In advanced biomedicine and microfluidics, there is a strong desire to sort and manipulate various cells and bacteria based on miniaturized microfluidic chips. Here, by integrating fiber tweezers into a T-type microfluidic channel, we report an optofluidic chip to selectively trap Escherichia coli in human blood solution based on different sizes and shapes. Furthermore, we simulate the trapping and pushing regions of other cells and bacteria, including rod-shaped bacteria, sphere-shaped bacteria, and cancer cells based on finite-difference analysis. With the advantages of controllability, low optical power, and compact construction, the strategy may be possibly applied in the fields of optical separation, cell transportation, and water quality analysis. Abstract:Bridging optical tweezers and microfluidics can form a multifunctional platform, which can overcome the difficulties of precise manipulation in hydrodynamic flow in a noninvasive method. However, when integrated into a microfluidic chip, the fiber optic tweezer loses its flexibility. Here, we propose a compact single fiber optical tweezer–micropipette system. It can sort particles by differences in shape and refractive index in a completely noninvasive way while retaining the flexibility, high selectivity, and precision of a fiber optical tweezer. Compact microfluidic channels are formed by combining two different-diameter micropipettes. The internal diameter of the circular microfluidic channel is less than 30 μm. Furthermore, we calculated the trapping and pushing regions of Yeast and Chlorella and achieved the separation of Yeast from Chlorella in the experiments. We did 90 sets of tests on the sorting accuracy of single fiber optical tweezer (SFOT). SFOT was able to distinguish between two types of particles in each test. With the advantages of high selectivity, high accuracy, low optical power consumption, and compact structure, such methods can be used in the fields of optical separation, cell transportation, cell sorting, and single-cell analysis.

Abstract:Abstract Optical manipulation of various kinds of nanoparticles is vital in biomedical engineering. However, classical optical approaches demand higher laser power and are constrained by diffraction limits, necessitating tailored trapping schemes for specific nanoparticles. They lack a universal and biocompatible tool to manipulate nanoparticles of diverse sizes, charges, and materials. Through precise modulation of diffusiophoresis and thermo‐osmotic flows in the boundary layer of an optothermal‐responsive gold film, we designed highly‐adaptable optothermal nanotweezers (HAONT) capable of manipulating single nanoparticle as small as sub‐10 nm. Additionally, we introduced a novel optothermal doughnut‐shaped vortex (DSV) trapping strategy, enabling a new mode of physical interaction between cells and nanoparticles. Furthermore, this versatile approach allows for the manipulation of nanoparticles in organic, inorganic, and biological forms. It also offers versatile function modes such as trapping, sorting, and assembling of nanoparticles. We believe that this approach holds the potential to be a valuable tool in fields such as synthetic biology, optofluidics, nanophotonics, and colloidal science. Abstract:Plasmonic tweezers are an emerging research topic because of their low input power and wide operating range from homogeneous particles to complex biological objects. But it is still challenging for plasmonic tweezers to trap or manipulate objects of tens of microns, especially in biological science. This study introduces a new 3D biocompatible plasmonic tweezer for single living cell manipulation in solution. The key design is a tapered tip whose three-layer surface structure consists of nanoprobe, gold nanofilm, and thermosensitive hydrogel, thiolated poly(N-isopropylacrylamide). Incident light excites the surface plasmon polaritons on gold film and generates heat to induce thermally driven phase transition of the thermosensitive hydrogel, which enables reversible binding between functionalized surface and cell membrane and avoids both thermal and mechanical stresses in the meanwhile. The 3D biocompatible plasmonic tweezer realizes selective capture, 3D pathway free transport, and position-controlled release of target cells, and it displays excellent biocompatibility, low energy consumption, and high operational flexibility.

Abstract:A nanofiber-based optical tweezer is demonstrated. Trapping is achieved by combining attractive near-field optical gradient forces with repulsive electrostatic forces. Silica-coated Fe2O3 nanospheres of 300 diameter are trapped as close as 50 nm away from the surface with 810 μW of optical power, with a maximum trap stiffness of 2.7 pN μm−1. Electrostatic trapping forces up to 0.5 pN are achieved, a factor of 50 larger than those achievable for the same optical power in conventional optical tweezers. Efficient collection of the optical field directly into the nanofiber enables ultra-sensitive tracking of nanoparticle motion and extraction of its characteristic Brownian motion spectrum, with a minimum position sensitivity of 3.4 Å/Hz.

Abstract:Optical tweezers (OTs) can transfer light momentum to particles, achieving the precise manipulation of particles through optical forces. Due to the properties of non-contact and precise control, OTs have provided a gateway for exploring the mysteries behind nonlinear optics, soft-condensed-matter physics, molecular biology, and analytical chemistry. In recent years, OTs have been combined with microfluidic chips to overcome their limitations in, for instance, speed and efficiency, creating a technology known as "optofluidic tweezers." This paper describes static OTs briefly first. Next, we overview recent developments in optofluidic tweezers, summarizing advancements in capture, manipulation, sorting, and measurement based on different technologies. The focus is on various kinds of optofluidic tweezers, such as holographic optical tweezers, photonic-crystal optical tweezers, and waveguide optical tweezers. Moreover, there is a continuing trend of combining optofluidic tweezers with other techniques to achieve greater functionality, such as antigen-antibody interactions and Raman tweezers. We conclude by summarizing the main challenges and future directions in this research field.
Optically assisted diffusophoretic tweezers using resonant plasmonic bowtie nano-antennas

Abstract:Plasmonic antennas, leveraging localized surface plasmon resonance (LSPR), hold significant promise for efficiently trapping nanoscale particles at low power levels. However, their effectiveness is hindered by photothermal effects in metallic nanoparticles, leading to repulsive thermophoretic forces. To address this limitation, we propose a novel hybrid approach that combines depletion attraction and photothermal effects inherent in plasmonic structures, capitalizing on thermally induced concentration gradients. Through the thermophoretic depletion of polyethylene glycol (PEG) molecules around plasmonic hotspots, we create sharp concentration gradients, enabling precise localization of nanoscopic particles through a synergistic effect with diffusophoretic forces. In our experiments, we successfully demonstrate the trapping and dynamic 2D manipulation of 100 nm polystyrene beads, showcasing the platform's potential for colloidal assembly at the nanoscale. Remarkably, this method maintains highly stable trapping performance even at an incredibly low 2.5 mW laser power, rendering it particularly appealing for applications involving biological species. Our study introduces a promising avenue for the precise and efficient manipulation of sub-nanoscale particles, with wide-ranging implications in nanotechnology, biophysics, and nanomedicine. This research opens up new opportunities for advancing nanoscale particle studies and applications, ushering in a new era of nanoscale manipulation techniques.
