Abstract:Co-amorphous systems (CAMs) have shown promise in addressing the challenges associated with poorly water-soluble drugs. However, the limited selection of co-formers and the use of lab-scale techniques for their preparation present challenges in fully utilizing the advantages of CAMs. In this study, we used aspartame (a methyl ester of the aspartic acid/phenylalanine) as a model dipeptide with the BCS class II drug dipyridamole, to prepare co-amorphous systems using spray drying. The feed solutions were prepared by dissolving the drug and co-former into methanol–water mixtures. The spray drying process was evaluated and solid-state properties were compared with those of the individual amino acids, amino acid mixtures and aspartame as co-formers. Co-amorphous systems prepared with aspartame (AspPhe) exhibited better solid-state properties, including a higher glass transition temperature (T g ), compared to the individual amino acids and the mixture of amino acids. Additionally, this formulation showed improved physical stability when stored at 25 °C/60 % RH conditions. Hirshfeld Surface (HS) analysis was employed to visualize and analyse the molecular interaction sites within the crystal structures of dipyridamole and aspartame. The observed interactions were then correlated with the molecular interactions identified through FT-IR spectroscopic analysis within the CAMs. The spectroscopic analysis revealed molecular interactions between the sites found at the shortest distances in the HS analysis. The dominant hydrogen bond interactions identified in the co-amorphous DPM-AspPhe system was found to contribute significantly to its improve stability. X-ray powder diffraction in non-ambient mode reveals that both temperature and humidity play a role in the crystallization of the co-amorphous DPM-AspPhe. Crystallization rates increased notably at high temperature and humidity. To predict stability under accelerated conditions, the crystallization rates from DPM-AspPhe were fitted to a modified Arrhenius equation. However, the predictive accuracy of the resulting model was limited to a specific range of conditions.

Abstract:Co-amorphous supersaturated drug delivery systems are emerging as an alternative strategy to improve the water solubility of BCS II drugs. Typically, the supersaturation and stability of co-amorphous systems largely depend on the type of employed co-former. This study aims to assess the potential for active metabolites of drugs as co-former in drug-drug co-amorphous formulations. Toltrazuril (Tol) was chosen as the model drug, to which ponazuril (Pon) was added as co-former. Considering the importance of intermolecular interactions in co-amorphous systems, we performed highlighted investigations including molecular dynamics simulation and quantum mechanics calculations. The results indicated that Tol and Pon molecules were connected by N-H···O = C hydrogen bonds in the form of a complementary pairing of amide groups. Further, the solubility/dissolution and solid-state stability of the co-amorphous system were investigated. We found that co-amorphous Tol-Pon was stable for at least one month at 40 °C/75% RH, while amorphous materials underwent recrystallization within 10 days. Moreover, both drugs in the co-amorphous system exhibited enhanced "spring parachute effect" during the dissolution process. This could be attributed to the noticeably increased solid-state stabilization as well as inhibition of Pon on the crystallization of Tol from a supersaturated state. In general, our study provides some useful information and molecular insights to guide the development of drug-active metabolite-based co-amorphous formulations.

Abstract:To increase the performance of indomethacin (IND) by enhancing its water solubility and promoting its therapeutic efficiency, a ternary physically stable co-amorphous system of this poorly soluble drug with paracetamol and polyvinylpyrrolidone (PVP K30) was prepared. The effect of PVP on molecular interactions was also evaluated. Binary solid dispersions (IND:paracetamol) were prepared by a kneading method. Then, the PVP as a third component was added at different weight ratios (w/w). The generated systems were characterized by x-ray diffraction, Fourier transform infrared (FTIR) spectroscopy, scanning electronic microscopy, and 13 C NMR spectroscopy. As evidenced by FTIR spectroscopy and 13 C NMR investigations, the PVP interacted with paracetamol and IND through hydrogen bonds. A physical stability study under high stress conditions at relative humidity = 75% and T = 40°C for 3 months showed the ability of PVP to stabilize amorphous drug molecules. This has significantly improved the water solubility of IND at T = 37°C and pH = 7.0 in comparison with the binary solid dispersion (IND:paracetamol) or the pure state of the drug. The synergistic effect that PVP provides to the binary mixture of IND and paracetamol was the driving force behind forming a robust co-amorphous system with enhanced physical stability and water solubility.

Abstract:Recent studies have shown that strong, alternating focusing magnets can be used to greatly increase the momentum acceptance of hadron therapy gantries. With the high gradients achievable with superconducting magnets a level of momentum acceptance can be reached which may have significant implications to medical gantries and to the introduction of superconducting technology in this area. The design of such a superconducting magnet system for a proton therapy gantry will be presented. The Canted-Cosine-Theta concept is extended to a curved magnet system generating the desired bending and alternating focusing fields for the achromatic optics. Magnetic, structural, and thermal analysis of this design is presented along with preliminary efforts towards fabrication and assembly of the curved magnet.

Abstract:Intermolecular interactions between drug and co-former are crucial in the formation, release and physical stability of co-amorphous system. However, the interactions remain difficult to investigate with only experimental tools. In this study, intermolecular interactions of co-amorphous curcumin-piperine (i.e., CUR-PIP CM) during formation, dissolution and storage were explored by integrating experimental and modeling techniques. The formed CUR-PIP CM exhibited the strong hydrogen bond interaction between the phenolic OH group of CUR and the C=O group of PIP as confirmed by FTIR, ss 13 C NMR and molecular dynamics (MD) simulation. In comparison to crystalline CUR, crystalline PIP and their physical mixture, CUR-PIP CM performed significantly increased dissolution accompanied by the synchronized release of CUR and PIP, which arose from the greater interaction energy of H 2 O-CUR molecules and H 2 O-PIP molecules than CUR-PIP molecules, breaking the hydrogen bond between CUR and PIP molecules, and then causing a pair-wise solvation of CUR-PIP CM at the molecular level. Furthermore, the stronger intermolecular interaction between CUR and PIP was revealed by higher binding energy of CUR-PIP molecules, which contributed to the excellent physical stability of CUR-PIP CM over amorphous CUR or PIP. The study provides a unique insight into the formation, release and stability of co-amorphous system from MD perspective. Meanwhile, this integrated technique can be used as a practical methodology for the future design of co-amorphous formulations.

Abstract:Solid dispersions (SDs) of dipyridamole (DIP) with a novel carrier copolyvidonum Plasdone S-630 (CoPVP) were developed by solvent evaporation method. The solid state of SDs of DIP with CoPVP (SDs CoPVP) was characterized by fourier transform infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC), X-ray diffraction (XRD) and polarizing microscopy, compared with that of SDs of DIP with polyvinylpyrrolidone Plasdone K-29/32 (SDs PVP). FT-IR analysis demonstrated the presence of intermolecular hydrogen bonding between DIP and CoPVP or PVP in SDs. DSC and XRD studies indicated that DIP presented in amorphous state in both SDs CoPVP and SDs PVP at higher weight ratios. The dissolution property of SDs CoPVP was significantly improved in comparison of pure DIP and physical mixtures with CoPVP (PM CoPVP). Both SDs CoPVP and SDs PVP powder showed the favorable flowability. However, SDs CoPVP showed better compressibility than SDs PVP. The lower hydroscopicity of SDs CoPVP could be advantageous to the stability to SDs. This study proves the potential of CoPVP as a carrier in the formulations of SDs for poorly soluble drugs.

Abstract:Dipyridamole (DIP) is a medication utilized to treat cardiovascular diseases, belonging to the BCS II class of drugs with low solubility and high permeability. As the effectiveness of oral preparations is influenced by the solubility and dissolution rate of the active ingredients, this study developed eight salts for solid modification of DIP to overcome poor water solubility. These salts were characterized using single crystal X-ray diffraction, powder X-ray diffraction and thermal analysis. Dipyridamole-oxalate (DIP-OLA) and dipyridamole-malonate (DIP-MNA) showed significant increase in apparent solubility and dissolution rate in buffer compared to DIP. The crystal structure analysis showed that the improved solubility is due to the change of the dipyridamole conjugate system and the molecular arrangement in the crystal after the introduction of counterions. It is noteworthy that the fluorescence emission of some salts in the solid state is significantly stronger than that of DIP. According to the crystal structure, the destruction of π-π stacking of DIP molecules after salification is the main mechanism of fluorescence enhancement, and Hirshfeld analysis and HOMO-LUMO analysis are also used to further explain the fluorescence changes.

Abstract:Co-amorphous drug delivery systems (CAMS) are characterized by the combination of two or more (initially crystalline) low molecular weight components that form a homogeneous single-phase amorphous system. Over the past decades, CAMS have been widely investigated as a promising approach to address the challenge of low water solubility of many active pharmaceutical ingredients. Most of the studies on CAMS were performed on a case-by-case basis, and only a few systematic studies are available. A quantitative analysis of the literature on CAMS under certain aspects highlights not only which aspects have been of great interest, but also which future developments are necessary to expand this research field. This review provides a comprehensive updated overview on the current published work on CAMS using a quantitative approach, focusing on three critical quality attributes of CAMS, i.e., co-formability, physical stability, and dissolution performance. Specifically, co-formability, molar ratio of drug and co-former, preparation methods, physical stability, and in vitro and in vivo performance were covered. For each aspect, a quantitative assessment on the current status was performed, allowing both recent advances and remaining research gaps to be identified. Furthermore, novel research aspects such as the design of ternary CAMS are discussed.

Abstract:Coamorphous drug delivery systems have emerged as a promising formulation technique for improving the solubility and oral bioavailability of poorly soluble drugs. The selection of a suitable coformer is the key to obtaining a successful coamorphous formulation. This study aims to investigate the impacts of coformers with similar chemical structures but different physical properties on the crystallization behavior and molecular dynamics of binary amorphous systems. The addition of three profen analogs, ibuprofen (IBU), ketoprofen (KETO) and indoprofen (INDO) leads to significantly different effects on the crystallization kinetics of amorphous nimesulide (NIME). The crystal growth rates for amorphous NIME are substantially accelerated in the presence of IBU, but drastically reduced in the presence of INDO, while the incorporation of KETO results in a negligible effect. Broadband dielectric spectroscopy is employed to characterize the molecular dynamics of neat amorphous NIME and coamorphous systems. The addition of three structural analogs alters the molecular mobility of amorphous NIME in different ways, which is consistent with the trend observed for their impacts on the crystallization kinetics, suggesting that the relative mobility between the components of coamorphous mixtures governs the physical stability. In addition, it is found that the temperature dependence of the α-relaxation times for NIME with and without coformers is superimposed once the temperature is scaled by Tg/T, whereas the crystallization kinetics do not overlap on a Tg/T scale. This deviation can result from a complex interplay of thermodynamic and kinetic factors involved in multicomponent amorphous systems. This study provides insights into the crystallization kinetics and molecular dynamics of coamorphous systems containing drug analogs, which can potentially offer more flexibility for the control of physical stability without sacrificing therapeutic efficacy.

Abstract:Despite the potential advantages of amorphism-induced supersaturation, the merit of new amorphization formation methods on the properties of the amorphous drug including the stability of the amorphous state, dissolution/solubility, supersaturation, and “spring-parachute” process is still poorly understood, particularly for certain amorphous supersaturating drug delivery systems (aSDDS). The present work aimed to explore the detailed merit of current attractive amorphization manufacturing methods (i. g., hot-melt extrusion (HME) technique) on the property improvement of aSDDS in form of amorphous solid dispersion microparticles by employing a model BCS II drug nitrendipine and a polyvinylpyrrolidone-based model polymer copovidone. Many aSDDS systems were developed by various methods, and their physicochemical properties were characterized by SEM, PXRD and DSC. HME-triggered amorphization induced superior supersaturation by the observation of the highest dissolution and solubility. HME induced the optimal supersaturation duration by the observed greatest extension of “spring-parachute” process (e. g., maximum AUCspring-parachute). HME technique is comparable with other techniques for the stabilization of amorphous state during storage. All aSDDS systems by HME and other methods showed improved long-term stability of the amorphous state in comparison to the pure amorphous drug. Fourier transformation infrared spectroscopy, Noyes-Whitney equation, nucleation theory and Gibbs free energy of transfer (ΔGto) were used to analyze the underlying mechanisms. Molecular mechanism studies indicated that HME caused a stronger crystallization inhibition effect in the aSDDS systems than other methods, but molecular interaction is not a dominant mechanism for property enhancement caused by HME. For the mechanism associated with the polymer itself (PVPVA64), it could inhibit the drug recrystallization, solubilize the drug spontaneously and cause the improved molecular interactions in all aSDDS systems. This study provided a deep insight into detailed advantage of HME-triggered supersaturation/amorphization and facilitated the applications of the technique both in the field of particuology and in pharmaceutical industry.
Poly (amino acid)s as new co-formers in amorphous solid dispersion

Abstract:The drug-amino acid co-amorphous systems and amorphous solid dispersions (ASDs) are promising methods to address the poor water solubility of poorly water-soluble drugs. However, some amino acids might not be perfect co-formers for co-amorphous systems, and the relatively low drug-loading of many ASDs is one of the main disadvantages of ASDs. Thus, poly-l-lysine and polyglutamic acid were selected as the co-formers, ball milled with basic mebendazole, neutral tadalafil and acidic valsartan at different weight ratios (from 3:1 to 1:3) to prepare poly (amino acid)-based ASDs, aiming to combine the advantages of co-amorphous systems (high drug-loading) and ASDs (relatively high Tg and high physical stability). All the mixtures were converted into amorphous after milling. The powder dissolution studies showed that drug-poly (amino acid) ASDs improved the dissolution rate of the drug in different ways and to different degrees. Moreover, the two poly (amino acid)s enhanced the physical stability of amorphous drugs. It is worthy to mention that the salt formation between the drug and the poly (amino acid) does not necessarily mean better performance compared to non-salt forming systems, and salt formation is also not a prerequisite for the formation of promising drug-poly (amino acid) ASDs.
Effect of Drug–Polymer Interaction in Amorphous Solid Dispersion on the Physical Stability and Dissolution of Drugs: The Case of Alpha-Mangostin

Abstract:Improving drug solubility is necessary for formulations of poorly water-soluble drugs, especially for oral administration. Amorphous solid dispersions (ASDs) are widely used in the pharmaceutical industry to improve the physical stability and solubility of drugs. Therefore, this study aims to characterize interaction between a drug and polymer in ASD, as well as evaluate the impact on the physical stability and dissolution of alpha-mangostin (AM). AM was used as a model of a poorly water-soluble drug, while polyvinylpyrrolidone (PVP) and eudragit were used as polymers. The amorphization of AM-eudragit and AM-PVP was confirmed as having a halo pattern with powder X-ray diffraction measurements and the absence of an AM melting peak in the differential scanning calorimetry (DSC) curve. The solubility of amorphous AM increased in the presence of either eudragit or PVP due to amorphization and interactions of AM-polymer. Furthermore, FT-IR spectroscopy and in silico studies revealed hydrogen bond interactions between the carbonyl group of AM and the proton of eudragit as well as PVP. AM-eudragit with a ratio of 1:1 recrystallized after 7 days of storage at 25 °C and 90% RH, while the AM-PVP 1:4 and 1:10 samples retained the X-ray halo patterns, even under humid conditions. In a dissolution test, the presence of polymer in ASD significantly improved the dissolution profile due to the intermolecular interaction of AM-polymer. AM-eudragit 1:4 maintained AM supersaturation for a longer time compared to the 1:1 sample. However, a high supersaturation was not achieved in AM-PVP 1:10 due to the formation of large agglomerations, leading to a slow dissolution rate. Based on the results, interaction of AM-polymer in ASD can significantly improve the pharmaceutical properties of AM including the physical stability and dissolution.

Abstract:The co-amorphous system is an efficacious approach in enhancing the solubility and dissolution of insoluble drugs. Several small molecules, such as organic acids, amino acids, and organic amines, are commonly used ligands to prepare co-amorphous systems. In this study, a co-amorphous system of posaconazole (POS), a weakly alkaline insoluble drug, with citric acid (CA) was prepared by the solvent evaporation method. An amorphous solid dispersion (ASD) of POS-VA64 was also prepared for comparison. Powder X-ray diffraction pattern and scanning electron microscopy indicated that POS exists in an amorphous form in the POS-CA and POS-VA64 groups. Fourier-transform infrared spectroscopy showed that an intermolecular hydrogen bonding was formed between POS and CA in the co-amorphous system. The amorphous state of POS exhibited increased solubility and dissolution, and the POS-CA form showed the best solubility and dissolution, which may be attributed to the amorphization and acidic environment provided by CA. In addition, the pharmacokinetic study and stability test of the POS-CA form showed superior performance. Therefore, the rational use of small-molecule ligands to prepare co-amorphous compounds can significantly improve the properties of drugs. Abstract:Abstract.Amorphization is one of the most effective pharmaceutical approaches to enhance the dissolution and oral bioavailability of poorly water-soluble drugs. In recent years, amorphous formulations have been experiencing rapid development both in theoretical and practical application. Based on using different types of stabilizing agents, amorphous formulations can be mainly classified as polymer-based amorphous solid dispersion, coamorphous formulation, mesoporous silica-based amorphous formulation, etc. This paper summarizes recent advances in the dissolution and supersaturation of these amorphous formulations. Moreover, we also highlight the roles of stabilizing agents such as polymers, low molecular weight co-formers, and mesoporous silica. Maintaining supersaturation in solution is a key factor for the enhancement of dissolution profile and oral bioavailability, and thus, the strategies and challenges for maintaining supersaturation are also discussed. With an in-depth understanding of the inherent mechanisms of dissolution behaviors, the design of amorphous pharmaceutical formulations will become more scientific and reasonable, leading to vigorous development of commercial amorphous drug products.

The potential of supramolecular synthon to develop coamorphous systems with tailored physical stability: Mechanistic insights integrating kinetics and thermodynamics

Abstract:Coamorphous drug delivery systems have received increasing interest owing to their potential to improve the solubility, dissolution and bioavailability of poorly water-soluble drugs. However, the crystallization risk is one of major limitations in their application. It has been widely recognized that the coformer plays a vital role in physical stability of coamorphous formulation. Unfortunately, the screen of optimal coformer still adopts a trial-and-error method, which is time-consuming and expensive. Herein, a supramolecular synthon approach based on the interaction between functional groups, was exploited to design coamorphous systems (CMs) consisting of lurasidone hydrochloride (LH) and three coformers, saccharin (SAC), L-tryptophan (TRP), and L-cysteine hydrochloride (CYS). X-ray powder diffraction suggested the order of physical stability of the coamorphous systems was ranked as LH-CYS CM > LH-TRP CM > LH-SAC CM. The charge-assisted hydrogen bond between LH and coformer was confirmed by infrared spectroscopy and solid-state 13C NMR. Moreover, structural, electronic, and molecular interaction information, especially hydrogen bonding interactions, were quantified by theoretical calculations, including miscibility calculations, molecular dynamics simulations and quantum chemical calculations. It was revealed that LH-CYS CM exhibited the best miscibility, strongest binding energy and strongest H-bond with partially covalent character, demonstrating the significant role of supramolecular synthon in stabilizing coamorphous formulations. Interestingly, LH-TRP CM, not LH-CYS CM, exhibited the lowest molecular mobility among three coamorphous systems, which was inconsistent with their physical stability. But from thermodynamic perspective, the order of configurational entropy and physical stability of coamorphous systems was completely consistent. We shed light on the comprehensive effects of molecular mobility and configurational entropy on physical stability of coamorphous systems. Importantly, the relationship between supramolecular synthon and kinetic/thermodynamic mechanisms was also discussed, and the positive correlation between configurational entropy and intermolecular interactions was proposed in this paper. Our findings demonstrated the great potential of supramolecular synthon in designing coamorphous systems with tailored physical stability, and further provided a deeper insight into the mechanisms of physical stability of coamorphous systems.
Amorphous Solid Dispersions: Implication of Method of Preparation and Physicochemical Properties of API and Excipients

Abstract:The present study investigated the effect of different polymers and manufacturing methods (hot melt extrusion, HME, and spray drying, SD) on the solid state, stability and pharmaceutical performance of amorphous solid dispersions. In the present manuscript, a combination of different binary amorphous solid dispersions containing 20% and 30% of drug loadings were prepared using SD and HME. The developed solid-state properties of the dispersions were evaluated using small- and wide-angle X-ray scattering (WAXS) and modulated differential scanning calorimetry (mDSC). The molecular interaction between the active pharmaceutical ingredients (APIs) and polymers were investigated via infrared (IR) and Raman spectroscopy. The in vitro release profile of the solid dispersions was also evaluated to compare the rate and extend of drug dissolution as a function of method of preparation. Thereafter, the effect of accelerated stability conditions on the physicochemical properties of the solid dispersions were also evaluated. The results demonstrated higher stability of Soluplus® (SOL) polymer-based solid dispersions as compared to hydroxypropyl methylcellulose (HPMC)-based solid dispersions. Moreover, the stability of the solid dispersions was found to be higher in the case of API having high glass transition temperature (Tg) and demonstrated higher interaction with the polymeric groups. Interestingly, the stability of the melt-extruded dispersions was found to be slightly higher as compared to the SD formulations. However, the down-processing of melt-extruded strands plays critical role in inducing the API crystal nuclei formation. In summary, the findings strongly indicate that the particulate properties significantly influence the performance of the product.
Cerec3D endocrowns--two-year clinical examination of CAD/CAM crowns for restoring endodontically treated molars.

Abstract:The present clinical study evaluates the clinical prognosis of Cerec3D endocrowns over an observation period of two years. Twenty Cerec endocrowns (VITABLOCS Mark II, Vita Zahnfabirk, Bad Säckingen, Germany) were placed with PanaviaTM F 2.0, a dual-curing bonding composite. The control examinations took place semi-annually and the restorations were evaluated based on modified US Public Health Service criteria (USPHS). The two-year survival rate of the Cerec endocrowns was 90%. Two out of 20 endocrowns were assessed as failure because of fractures. The first fracture was observed after 12 months and the second fracture after 18 months. No recurrent caries was diagnosed during the entire examination period. Slight percussion symptoms were found on three restorations in the first weeks after placement. The results of the present study show that good esthetic and functional results similar to those of other restoration types can be achieved with endocrowns and that CAD/CAM-fabricated crowns represent a very promising treatment alternative for endodontically treated molars.
