Abstract:Quantification information for nonpoint source pollution is required to develop a Total Maximum Daily Load (TMDL) at the watershed scale. All export coefficient models and some complex mechanistic models rely on the pollutant export coefficients to quantify and identify nonpoint source pollution. Typically, pollutant export coefficients are estimated by monitoring field plots or small catchments with a single land-use or by monitoring and statistically analyzing mixed land-use watersheds. However, these approaches underestimate export coefficients by neglecting in-stream pollutant retention and they also neglect spatio-temporal variability for export coefficients within a watershed. In addition, these methods do not address the uncertainty associated with estimations. This study combined the export coefficient model, a stream pollutant load model and a Bayesian statistical method to inversely estimate pollutant export coefficients for multiple land-use types from commonly available stream monitoring data. The efficacy of this inversed Bayesian modeling approach was confirmed by determining total nitrogen (TN) export coefficients for farmland, residential land and woodland for six catchments of the ChangLe River watershed in eastern China covering 72 field observation dates in 2004-2009. In-stream retention processes were considered in estimating TN export coefficients. The temporal (across 72 field observation dates) and spatial (across 6 catchments) variability of TN export coefficients for each land-use type within the watershed are discussed, as well as the uncertainties associated with TN export coefficient estimation. This inversed Bayesian modeling approach overcomes the shortcomings involved in current widely used approaches for estimating pollutant export coefficients; thus it can more efficiently support development of TMDL programs, particularly in circumstance where limited data are available. Stream Nitrogen Sources Apportionment and Pollution Control Scheme Development in an Agricultural Watershed in Eastern China

Abstract:A modeling system that couples a land-usebased export coefficient model, a stream nutrient transport equation, and Bayesian statistics was developed for stream nitrogen source apportionment. It divides a watershed into several sub-catchments, and then considers the major landuse categories as stream nitrogen sources in each subcatchment. The runoff depth and stream water depth are considered as the major factors influencing delivery of nitrogen from land to downstream stream node within each sub-catchment. The nitrogen sources and delivery processes are lumped into several constant parameters that were calibrated using Bayesian statistics from commonly available stream monitoring and land-use datasets. This modeling system was successfully applied to total nitrogen (TN) pollution control scheme development for the ChangLe River watershed containing six sub-catchments and four land-use categories. The temporal (across months and years) and spatial (across sub-catchments and land-use categories) variability of nonpoint source (NPS) TN export to stream channels and delivery to the watershed outlet were assessed. After adjustment for in-stream TNretention, the time periods and watershed areas with disproportionately high-TN contributions to the stream were identified. Aimed at a target stream TN level of 2 mg L-1, a quantitative TN pollution control scheme was further developed to determine which sub-catchments, which land-use categories in a sub-catchment, which time periods, and how large of NPS TN export reduction were required. This modeling system provides a powerful tool for stream nitrogen source apportionment and pollution control scheme development at the watershed scale and has only limited data requirements.
A Modified Load Apportionment Model for Identifying Point and Diffuse Source Nutrient Inputs to Rivers from Stream Monitoring Data

Abstract:Determining point (PS) and diffuse source (DS) nutrient inputs to rivers is essential for assessing and developing mitigation strategies to reduce excessive nutrient loads that induce eutrophication. However, application of watershed mechanistic models to assess nutrient inputs is limited by large data requirements and intensive model calibration efforts. Simple export coefficient models and statistical models also require extensive primary watershed attribute information and further they cannot address seasonal patterns of nutrient delivery. In practice, monitoring efforts to identify all PSs within a watershed are very difficult due to time and economic limitations. To overcome these issues, based on the fundamental hydrological differences between PS and DS pollution, a modified load apportionment model (LAM) was developed relating the river nutrient load to nutrient inputs from PS, DS and upstream inflow sources while adjusting for in-stream nutrient retention processes. Estimates of PS and DS inputs can be easily achieved through Bayesian calibration of the five model parameters from commonly available stream monitoring data. It considers in-stream nutrient retention processes, temporal changes of PS and DS inputs, and nutrient contributions from upstream inflow waters, as well as the uncertainty associated with load estimations. The efficacy of this modified LAM was demonstrated for total nitrogen (TN) source apportionment using a 6-year record of monthly water quality data for the ChangLe River in eastern China. Aimed at attaining the targeted river TN concentration (2 mg L-1), required input load reductions for PS, DS and upstream inflow were estimated. This modified LAM is applicable for both district-based and catchment-based water quality management strategies with limited data requirements, providing a simple, effective and economical tool for apportioning PS and DS nutrient inputs to rivers. Multiplex Analysis of Fundamental Surface Water Quality Indicators by Multichannel Electrochemical Device Combined with MFCSC Model

Abstract:Rapid and accurate detection of water quality is essential for environmental monitoring and management. Among them, permanganate index (CODMn), nitrate–nitrogen (NO3−-N), dissolvable inorganic phosphorus (DIP), and ammonia nitrogen (AN) are four important water quality indicators. A portable multichannel electrochemical device (MCED) combined with a sensor array composed of gold nanoparticles (AuNPs), silver nanodendritic structures (AgNDs), and reduced graphene oxide (rGO)-modified screen-printed electrodes (SPEs) was applied for facile, rapid on-site inspection. The sensor array exhibits good complementarity in voltammetric measurements of water quality indicators and has cross-sensitivity in multiplex measurement. A multiple fully connected shared convolution (MFCSC) model was constructed to process the voltammetric data, extracting unique features to resolve the overlap in voltammetric data of complex solutions for the quantitative determination of target analytes. Accurate predictions were successfully achieved for the four parameters, CODMn, NO3−-N, DIP, and AN in the mixture, where CODMn, NO3−-N, and AN have high coefficients of determination ( ${R} ^{{2}}$ ) and low mean absolute percentage errors (MAPE%). Using the migration ability of MFCSC, combined with MCED, it is further applied for real water sample testing, which proves the good prediction of the water quality indicators of real water samples in complex matrices. The results show that the sensing system developed in this work has wide application potential and can be used to rapidly analyze surface water quality and obtain water quality information in a wide range of water bodies for monitoring.
Nitrate Source Apportionment in a Subtropical Watershed Using Bayesian Model

Abstract:Nitrate (NO3-) pollution in aquatic system is a worldwide problem. The temporal distribution pattern and sources of nitrate are of great concern for water quality. The nitrogen (N) cycling processes in a subtropical watershed located in Changxing County, Zhejiang Province, China were greatly influenced by the temporal variations of precipitation and temperature during the study period (September 2011 to July 2012). The highest NO3- concentration in water was in May (wet season, mean±SD=17.45±9.50 mg L(-1)) and the lowest concentration occurred in December (dry season, mean±SD=10.54±6.28 mg L(-1)). Nevertheless, no water sample in the study area exceeds the WHO drinking water limit of 50 mg L(-1) NO3-. Four sources of NO3(-) (atmospheric deposition, AD; soil N, SN; synthetic fertilizer, SF; manure & sewage, M&S) were identified using both hydrochemical characteristics [Cl-, NO3-, HCO3-, SO42-, Ca2+, K+, Mg2+, Na+, dissolved oxygen (DO)] and dual isotope approach (δ15N-NO3- and δ(18)O-NO3-). Both chemical and isotopic characteristics indicated that denitrification was not the main N cycling process in the study area. Using a Bayesian model (stable isotope analysis in R, SIAR), the contribution of each source was apportioned. Source apportionment results showed that source contributions differed significantly between the dry and wet season, AD and M&S contributed more in December than in May. In contrast, SN and SF contributed more NO3- to water in May than that in December. M&S and SF were the major contributors in December and May, respectively. Moreover, the shortcomings and uncertainties of SIAR were discussed to provide implications for future works. With the assessment of temporal variation and sources of NO3-, better agricultural management practices and sewage disposal programs can be implemented to sustain water quality in subtropical watersheds.

Abstract:Nitrate (NO 3 – ) is the major form of bioavailable nitrogen in most rivers, and its dramatic increase may lead to a number of serious environmental issues, such as eutrophication and biodiversity decline. Quantification of NO 3 – sources and fluxes in rivers is essential for effective management of NO 3 – pollution. Isotope mixing and hydrological models are proven practical tools for quantifying the sources of NO 3 – in rivers, yet both methods have severe limitations. To improve the accuracy and reliability of riverine NO 3 – source apportionment, this study proposed a protocol that integrates the strengths of both tools, capable of solving issues such as the calculation bias caused by isotope endmember overprinting in isotope mixing model and the lack of validation data for hydrological models. We applied the framework to a typical agricultural river, the Qihe River, and illustrated the effectiveness of the method in quantifying riverine NO 3 – sources. The proportional contribution of sewage simulated by the SWAT model approximated that estimated by the isotopic mixing model, yet the SWAT-based contributions of soil organic nitrogen and chemical fertilizer were 16.3 % lower and 14.0 % higher, respectively than the MCMC-based results. After integrating both models, we adopted the proportions of NO 3 – sources in the river from chemical fertilizer, sewage, and soil organic nitrogen as 45.1 %, 30.9 %, and 23.9 %, respectively. This study showed that coupling isotopic and hydrological models provided a new dimension for the accurate quantification of N sources in rivers.

Abstract:A high concentration of nitrate (NO3−) in surface water threatens aquatic systems and human health. Revealing nitrate characteristics and identifying its sources are fundamental to making effective water management strategies. However, nitrate sources in multi-tributaries and mix land use watersheds remain unclear. In this study, based on 20 surface water sampling sites for more than two years’ monitoring from April 2012 to December 2014, water chemical and dual isotopic approaches (δ15N-NO3− and δ18O-NO3−) were integrated for the first time to evaluate nitrate characteristics and sources in the Huashan watershed, Jianghuai hilly region, China. Nitrate-nitrogen concentrations (ranging from 0.02 to 8.57 mg/L) were spatially heterogeneous that were influenced by hydrogeological and land use conditions. Proportional contributions of five potential nitrate sources (i.e., precipitation; manure and sewage, M & S; soil nitrogen, NS; nitrate fertilizer; nitrate derived from ammonia fertilizer and rainfall) were estimated by using a Bayesian isotope mixing model. The results showed that nitrate sources contributions varied significantly among different rainfall conditions and land use types. As for the whole watershed, M & S (manure and sewage) and NS (soil nitrogen) were major nitrate sources in both wet and dry seasons (from 28% to 36% for manure and sewage and from 24% to 27% for soil nitrogen, respectively). Overall, combining a dual isotopes method with a Bayesian isotope mixing model offered a useful and practical way to qualitatively analyze nitrate sources and transformations as well as quantitatively estimate the contributions of potential nitrate sources in drinking water source watersheds, Jianghuai hilly region, eastern China.

Abstract:The traceability of groundwater nitrate pollution is crucial for controlling and managing polluted groundwater. This study integrates hydrochemistry, nitrate isotope (δ15N-NO3- and δ18O-NO3-), and self-organizing map (SOM) and end-member mixing (EMMTE) models to identify the sources and quantify the contributions of nitrate pollution to groundwater in an intensive agricultural region in the Sha River Basin in southwestern Henan Province. The results indicate that the NO3--N concentration in 74% (n = 39) of the groundwater samples exceeded the WHO standard of 10 mg/L. According to the results of EMMTE modeling, soil nitrogen (68.4%) was the main source of nitrate in Cluster-1, followed by manure and sewage (16.5%), chemical fertilizer (11.9%) and atmospheric deposition (3.3%). In Cluster-2, soil nitrogen (60.1%) was the main source of nitrate, with a significant increase in the contribution of manure and sewage (35.5%). The considerable contributions of soil nitrogen may be attributed to the high nitrogen fertilizer usage that accumulated in the soil in this traditional agricultural area. Moreover, it is apparent that most Cluster-2 sampling sites with high contributions of manure and sewage are located around residential land. Therefore, the arbitrary discharge and leaching of domestic sewage may be responsible for these results. Therefore, this study provides useful assistance for the continuous management and pollution control of groundwater in the Sha River Basin.
Riverine Nitrate Source Identification Combining Δ15n/δ18o-No3- with Δ17O-NO3- and a Nitrification 15N-Enrichment Factor in a Drinking Water Source Region

Abstract:Dual nitrate isotopes (delta N-15/delta O-18-NO3-) are an effective tool for tracing nitrate sources in freshwater systems worldwide. However, the initial delta N-15/delta O-18 values of different nitrate sources might be altered by isotopic fractionation during nitrification, thereby limiting the efficiency of source apportionment results. This study integrated hydrochemical parameters, site-specific isotopic compositions of potential nitrate sources, multiple stable isotopes (delta D/delta O-18-H2O, delta N-15/delta O-18-NO3- and Delta O-17-NO3-), soil incubation experiments assessing the nitrification N-15-enrichment factor (epsilon(N)), and a Bayesian mixing model (MixSIAR) to reduce/eliminate the influence of N-15/O-18-fractionations on nitrate source apportionment. Surface water samples from a typical drinking water source region were collected quarterly (June 2021 to March 2022). Nitrate concentrations ranged from 0.35 to 3.06 mg/L (mean = 0.78 +/- 0.46 mg/L), constituting similar to 70 % of total nitrogen. A MixSIAR model was developed based on delta N-15/delta O-18-NO3- values of surface waters and the incorporation of a nitrification epsilon(N) (-6.9 +/- 1.8 parts per thousand). Model source apportionment followed: manure/sewage (46.2 +/- 10.7 %) > soil organic nitrogen (32.3 +/- 18.5 %) > nitrogen fertilizer (19.7 +/- 13.1 %) > atmospheric deposition (1.8 +/- 1.6 %). An additional MixSIAR model coupling delta N-15/delta O-18-NO3- with Delta O-17-NO3- and epsilon(N) was constructed to estimate the potential nitrate source contributions for the June 2021 water samples. Results revealed similar nitrate source contributions (manure/sewage = 43.4 +/- 14.1 %, soil organic nitrogen = 29.3 +/- 19.4 %, nitrogen fertilizer = 19.8 +/- 13.8 %, atmospheric deposition = 7.5 +/- 1.6 %) to the original MixSIAR model based on epsilon(N) and delta N-15/delta O-18-NO3-. Finally, an uncertainty analysis indicated the MixSIAR model coupling delta N-15/delta O-18-NO3- with Delta O-17-NO3- and epsilon(N) performed better as it generated lower uncertainties with uncertainty index (UI90) of 0.435 compared with the MixSIAR model based on delta N-15/delta O-18-NO3- (UI90 = 0.522) and the MixSIAR model based on delta N-15/delta O-18-NO3- and epsilon(N) (UI90 = 0.442).

Abstract:The pollution source identification methods based on traditional water quality monitoring and pollutant discharge loading typically require a high frequency of monitoring and generate a level of uncertainty in the identification results, owing to their limitations on the accurate and quantitative assessment of pollution source identification, migration, and transformation. This study combined multivariate statistical analysis and stable isotope technology to identify groundwater pollution sources in a typical multiple land-use area of the Chengdu Plain. A positive matrix factorization (PMF) model was adopted to reduce the interference of mass environmental factors on source identification and to determine the main factors influencing groundwater quality. Subsequently, a Bayesian stable isotope mixing model was developed to quantify the apportionment of each pollution source to groundwater nitrate (NO3-) with the consideration of hydro-chemical and land-use information. The results showed that the concentrations of NO3-, NO2-, NH4+, Mn, Fe, SO42-, and Cl- in groundwater of the study area exceeded the standard to different extents, presenting spatial variation. The main form of inorganic nitrogen in groundwater was NO3-. In general, concentrations of groundwater NO3- were the highest in vegetable fields (9.29 mg·L-1 on average), followed by livestock and poultry breeding farms (7.66 mg·L-1) and arable land (7.09 mg·L-1), whereas concentrations of groundwater NO3- in industrial areas were the lowest (2.20 mg·L-1). Groundwater quality in the study area was affected by geological processes, agricultural activities, hydrogeochemical evolution, and domestic and industrial discharges. Agricultural activities were the main contributor to the increase in groundwater NO3- in the study area. Chemical fertilizer (32%) and soil nitrogen (25%) contributed greatly to groundwater NO3- in agricultural areas, whereas sewage (28%) and atmospheric precipitation (27%) contributed most groundwater NO3- in industrial areas. Thus, the combination of multivariate statistical analysis and stable isotope technology could identify groundwater pollution sources and their apportionment effectively, providing scientific support for the prevention and control of groundwater pollution.

Abstract:Soil heavy metal(loid) (HM) source apportionment is the prerequisite to develop suitable mitigation measures and make pollution control and prevention regulations. The selection of appropriate tools (models) for source analysis is crucial, that is especially true for large-scale regions, as the Pearl River Delta (PRD), due to the high spatial variability in soil parent materials, soil topographical feature, and wide range of anthropogenic activities. The objective of this study is to evaluate the potential applications of receptor models (positive matrix factorization [PMF] and Unmix) which have been widely used in air pollution research to quantitatively apportion sources of heavy metal(loid)s in the soils. To assist the interpretation of the derived factors (sources) of the receptor models, enrichment factors and GIS mapping were used to identify the potential relationships between the factor contributions and human activities in the study area. As the models are built on completely different algorithms, a comparative approach was adopted in addition to evaluate the impact of sample size on the model results. Factor profiles generated by different receptor models were quite similar as well as their corresponding factor contributions spatial distribution. Though the stability of their results decreases with a reduced sample size, the results of PMF were less significantly influenced by the sample size than those of Unmix. Due to the difficulty (time consuming and expensive) of soil sample collection in large-scale regions, the PMF model appears to be practically more effective than Unmix. In addition, further investigation is needed for Unmix model to understand the reason for its high sensitivity and determine an appropriate sample size.

Abstract:Rapid urbanization intensifies nitrate (NO3−) pollution in groundwater. Based on hydrochemical analysis and stable isotopes (δD-H2O, δ18O–H2O, δ15N–NO3- and δ18O–NO3-, a principal component analysis-absolute principal component score-multiple linear regression (PCA-APCS-MLR) model and a Bayesian isotopic mixing model (SIAR model) were used to quantify groundwater pollution sources, especially NO3− sources, in Zhuji, East China. The results showed that the main chemical types in Zhuji were HCO3–Ca in shallow groundwater, HCO3–Ca and HCO3–Ca·Na in deep groundwater. The δD-H2O and δ18O–H2O values in groundwater ranged from −43.4‰ to −32.1% and from −7.0‰ to −4.8%, respectively, and atmospheric precipitation was the main recharge source of groundwater. The calculation of the PAC-APCS-MLR model showed that the potential pollution sources in shallow and deep groundwater were hydrogeological conditions, agricultural activities and domestic sewage/manure, with the total contributions of 77.62% and 71.90%, respectively. The contributions from agricultural activities and sewage/manure to TN, NO3− and NH4+ in shallow groundwater (>82.00%) were higher than those in deep groundwater (>74.00%). The δ15N–NO3-(6.0‰–33.0‰) and δ18O–NO3-(7‰–29.9‰) values of groundwater indicated the existence of clear denitrification in groundwater in Zhuji. The results highlighted that an accurate isotopic fractionation enrichment factor could improve NO3− source apportionment by the SIAR model. When the denitrification isotopic fractionation enrichment factors were εN = −5.3‰ to −8.7‰ and εO = −8.0‰ in the SIAR model, the NO3− contributions from sewage/manure accounted for 51.7%–40.9% in shallow groundwater and 49.3%–40.3% in deep groundwater. The NO3− contributions from soil nitrogen and chemical fertilizer were relatively low, those in shallow groundwater were 25.6%–28.4% and 22.7%–30.7%, respectively; and those in deep groundwater were 27.2%–29.8% and 23.5%–29.9%, respectively. It was demonstrated that PAC-APCS-MLR model coupled with SIAR model was an effective method for source identification and apportionment in groundwater.

Abstract:Water pollution from diffuse sources is a problem of increasing concern. Efforts to control diffuse pollution have been confined mainly to agricultural land and forests. Little attention has been paid to sources of diffuse pollution from urban areas. A diffuse nitrate modelling tool (DNMT) has been developed for modelling the fate of nitrate in urban areas. This tool works at the catchment scale and has a modular structure that consists of three components: the hydrological module, the nitrogen cycle module and the nitrate transport module. The hydrological model describes the possible flow pathways. The nitrogen cycle model accounts for the mass balance of nitrate and calculates the amount of nitrate for potential loss. The nitrate transport module simulates the movement of nitrate within and from the soil to the receiving water. This paper demonstrates the development of the tool and its application in the White Cart Water catchment. Spatial variability of source contributions to nitrate in regional groundwater based on the positive matrix factorization and Bayesian model

Abstract:Groundwater nitrate (NO 3 - ) pollution has attracted widespread attention; however, accurately evaluating the sources of NO 3 - and their contribution patterns in regional groundwater is difficult in areas with multiple sources and complex hydrogeological conditions. In this study, 161 groundwater samples were collected from the Poyang Lake Basin for hydrochemical and dual NO 3 - isotope analyses to explore the sources of NO 3 - and their spatial contribution using the Positive Matrix Factorization (PMF) and Bayesian stable isotope mixing (MixSIAR) models. The results revealed that the enrichment of NO 3 - in groundwater was primarily attributed to sewage/manure (SM), which accounted for more than 50 %. The contributions of nitrogen fertilizer and soil organic nitrogen should also be considered. Groundwater NO 3 - sources showed obvious spatial differences in contributions. Regions with large contributions of SM (>90 %) were located in the southeastern part of the study area and downstream of Nanchang, which are areas with relatively high population density. Nitrogen fertilizer and soil organic nitrogen showed concentrated contributions in paddy soil in the lower reaches of the Gan and Rao Rivers, and these accumulations were mainly driven by the soil type, land use type, and topography. This study provides insight into groundwater NO 3 - contamination on a regional scale.

Abstract:Identification and quantification of sources of nitrate (NO3–) in freshwater lakes provide useful information for management of eutrophication and improving water quality in lakes. Dual δ15N- and δ18O-NO3– isotopes and a Bayesian isotope mixing model were applied to identify sources of NO3– and estimate their proportional contributions to concentrations of NO3– in Tai Lake, China. In waters of Tai Lake, values for δ15N-NO3– ranged from 3.8 to 10.1‰, while values of δ18O ranged from 2.2 to 12.0‰. These results indicated that NO3– was derived primarily from agricultural and industrial sources. Stable isotope analysis in R called SIAR model was used to estimate proportional contributions from four potential NO3– sources (agricultural, industrial effluents, domestic sewage, and rainwater). SIAR output revealed that agricultural runoff provided the greatest proportion (50.8%) of NO3– to the lake, followed by industrial effluents (33.9%), rainwater (8.4%), and domestic sewage (6.8%). Contributions of those primary sources of NO3– to sub-regions of Tai Lake varied significantly (p < 0.05). For the northern region of the lake, industrial source (35.4%) contributed the greatest proportion of NO3–, followed by agricultural runoff (27.4%), domestic sewage (21.3%), and rainwater (15.9%). Whereas for the southern region, the proportion of NO3– contributed from agriculture (38.6%) was slightly greater than that contributed by industry (30.8%), which was similar to results for nearby inflow tributaries. Thus, to improve water quality by addressing eutrophication and reduce primary production of phytoplankton, NO3– from both nonpoint agricultural sources and industrial point sources should be mitigated.Graphical abstractᅟ

Abstract:Nitrate pollution has become an environmental problem of global concern. One effective way for controlling the nitrate pollution of water is to identify the pollution source and reduce the input of nitrate. This study traces and quantifies the sources of nitrate contamination to groundwater and surface water in the northeastern suburbs of Beijing, where an emergency groundwater source zone is located. Nitrogen and oxygen stable isotope analysis, geospatial analysis techniques, principal component analysis, correlation analysis, and a Bayesian isotope mixing model were used to achieve our goals. The results show that the main sources of nitrate pollution in groundwater were manure and sewage (M&S) (42.6 %) > soil nitrogen (SN) (26.6 %) > NH4+ in fertilizer and rain (NHF&R) (24.5 %) > NO3− fertilizer (NOF) (5.0 %) > NO3− in atmospheric deposition (NAD) (1.3 %), and main sources of nitrate in surface water were M&S (28.8 %) > SN (20.4 %) > NAD (19.8%) > NOF (16.5%) > NHF&R (14.5 %). Due to the high permeability of the aquifer in the study area, there was a strong hydraulic connection between groundwater and surface water. The discharge of treated wastewater (reclaimed water) into the mostly dried river channel in the study area might aggravate nitrate pollution in the groundwater.

Abstract:The current developments with the production and utility of new generation conjugated typhoid vaccines have been meticulously reviewed by Szu [1]. Nevertheless, the efficacy of different vaccines conjugated to different carrier proteins like rEPA, diphtheria or tetanus toxoid, recombinant diphtheria toxin (CRM197), might be far from ideal during infection of vaccine recipients by Vi-negative Salmonella typhi strains. The existence of naturally occurring Vi-negative strains has been well established now during molecular studies though Vi-negative isolates would not be agglutinated by anti-Vi sera have been known for several decades. During the 1960s, they were identified during phage typing in various geographical locations globally [2]. Genetic control of Vi-expression has been determined, and it is possible to confirm Vi-negative S. typhi. The synthesis and transportation proteins of the Vi capsular polysaccharide of Salmonella enterica serovar Typhi (serovar Typhi) are encoded by the viaB operon, which resides on a 134-kb pathogenicity island known as SPI-7. During analysis of 60 stored strains of serovar Typhi from the Faisalabad region of Pakistan by PCR for the presence of SPI-7 and two genes essential for Vi production: tviA and tviB, 9 (15%) were tested negative for both tviA and tviB, while only two of such strains lacked SPI-7 [3]. Furthermore, the 42 multiplex PCR-positive isolates from the Faisalabad region of Pakistan included 9 Vi negatives [4]. The expanded usage of conjugated Vi vaccines [1] in areas with frequent circulation of Vi-negative isolates [3,4] could aid in selection and dissemination of the Vi-negative strains in the community. The role of circulating Vi-negative strains could be tremendous following extended usage of Vi vaccines. After mass vaccinations, they could be selected with extensive dissemination and disease morbidity and mortality. Such events could be avoided by the use of live attenuated vaccines. The clinical picture associated with such Vi-negative isolates would be indistinguishable from the one caused by isolates carrying Vi antigen [5]. The role of circulating Vi-negative isolates in the community for failures associated with Vi vaccines has not been examined. The reported failures encountered during different randomized trials with typhoid fever vaccines [6] might well have been associated with circulating Vi-negative strains.

Abstract:Groundwater and surface water pollution from nonpoint sources are problems of increasing concern. Nitrate transport is a crucial part in the whole process of nitrate loss. Previous methods for simulating the nitrate transport process usually involved solving complex advection–dispersion equations and required large amount of parameters and input data, which makes a model application expensive or invalid in terms of computation and data availability. The aim of this research is to develop a simple conceptual method to simulate the nitrate transport. In this paper, a simple conceptual method utilizing the distribution of nitrate loss against cumulative drainage to simulate the nitrate transport process is introduced. The concept and construction of this method are presented, and its applicability at different spatial scales is demonstrated. Results of model application at laboratory, pilot, and catchment scales show that this method has a good capability to simulate the transport of nitrate. Impact of spatial scales on parameters’ values is also discussed. Compared to conventional methods, this method requires fewer parameters, less computing time, and less input data.

Abstract:Dissolved organic matter (DOM) in river watersheds dynamically changes based on its source during a monsoon period with storm event. However, the variations in DOM in urban and rural river watersheds that are dominated by point and non-point sources have not been adequately explored to date. We developed an innovative approach to reveal DOM sources in complex river watershed systems during pre-monsoon, monsoon, and post-monsoon periods using end-member mixing analysis (EMMA) by combining multi-isotope values (δ13C-DOC, δ15N-NO3 and δ18O-NO3) and spectroscopic indices (fluorescence index [FI], biological index [BIX], humification index [HIX], and specific UV absorbance [SUVA]). Several potential end-members of DOM sources were collected from watersheds, including top-soils, groundwater, plant group (fallen leaves, riparian plants, suspended algae), and different effluents (cattle and pig livestock, agricultural land, urban, industry facility, swine treatment facility and wastewater treatment facility). Concentrations of dissolved organic carbon, dissolved organic nitrogen, NO3-N, and NH4-N increased during the monsoon period with an increase in the input of anthropogenic DOM, which have higher HIX values owing to the flushing effect. The results of EMMA indicate that soil and agricultural effluents accounted for a substantial contribution of anthropogenic DOM at varying rates based on seasons. We also found that results of EMMA based on combining spectroscopic indices and δ13C-DOC isotope values were more accurate in tracing DOM sources with respect to land-use characteristics compared to applying only spectroscopic indices. The positive relationship between FI, BIX and δ15N-NO3 were revealed that nitrate would be decomposed from DOM affected by intensive agricultural activities. In addition, consistent with the EMMA results, the molecular composition of the DOM was clearly evidenced by a large number of CHON formulas, accounting for over 50% of the total characterized compounds, including pesticides and pharmaceuticals used in agriculture farmland and livestock. Our results clearly demonstrated that EMMA based on combing multi-stable isotopes and spectroscopic indices could be trace the DOM source, which is important for understanding changes in the DOM quality, and application of nitrate isotopes and molecular analysis supports in-depth interpretation. This study provides easy and intuitive techniques for the estimation of the relative impacts of DOM sources in complex river watersheds, which can be verified in various ways rather than relying on a single technique approach.
