Abstract:To reveal the response and adaptative mechanism of plants to the organic pollutants PBDEs, physiological and transcriptomic techniques were used to study the effects of exposure to BDE47 and BDE209 on tobacco (Nicotiana tabacum L.) plant growth, physiological function and response of key genes. Exposure to both BDE47 and BDE209 inhibited the growth of tobacco plants. The number of down-regulated DEGs following exposure to BDE47 was significantly higher than that following exposure to BDE209. Enrichment analysis using the KEGG showed that BDE47 and BDE209 primarily affected tobacco leaf photosynthesis-antenna proteins, photosynthesis, plant hormone signal transduction and α-linolenic acid metabolism. BDE47 primarily inhibits the synthesis of Chl a, and BDE209 has a more significant impact on Chl b. Most photosynthesis-related DEGs were concentrated in PSII and PSI; the number of down-regulated DEGs in PSI was significantly higher than that in PSII, and the range in which the PSI activity was reduced was also higher than that of PSII, i.e., PSII and PSI (particularly PSI) were sensitive to the effects of exposure to BDE47 and BDE209 on photosynthesis. The increase of the ratio of regulatory energy dissipation played an important protective role in alleviating the photoinhibition of PSII. Exposure to BDE47 and BDE209 can lead to the accumulation of ROS in tobacco leaves, but correspondingly, the activities of antioxidant enzymes SOD, POD, CAT, APX and GPX and the up-regulated expression of their coding genes play an important role in preventing excessive oxidative damage. Exposure to BDE47 and BDE209 promoted the up-regulation of gene expression related to Pro synthesis. In particular, the Pro synthetic process of the Orn pathway was promoted. Exposure to BDE47 and BDE209 induced the up-regulated expression of genes related to the synthesis of ABA and JA, promoted the synthesis of ABA and JA, and activated ABA and JA signal transduction pathways. In conclusion, both BDE47 and BDE209 inhibit the synthesis of chlorophyll and hinder the process of light energy capture and electron transfer in tobacco leaves. BDE47 was more toxic than BDE209. However, tobacco leaves can also adapt to BDE47 and BDE209 by regulating the antioxidant system, accumulating Pro and initiating the hormone signal transduction process. The results of this study provide a theoretical basis for the phytotoxicity mechanism of PBDEs.

Abstract:To better understand the role of polyamines (PAs) and their metabolism in responses to chilling stress in tobacco plants, this study investigated changes of endogenous PA contents, PA synthetase and oxidase activities, seedling growth and membrane permeability in tobacco ( Nicotiana tabacum ) cvs. MSk326 (MK, chilling-sensitive variety) and Honghuadajinyuan (HD, chilling-tolerant variety) under chilling stress. Chilling stress significantly reduced the shoot and root dry weights but improved PA contents, especially of putrescine, in the two tobacco cultivars, with higher contents in the tolerant variety. Moreover, stepwise regression analysis indicated that the relative growth rate of tobacco seedlings under chilling stress was positively correlated with endogenous PA contents, putrescine and spermidine, particularly. The above results suggested that the resistance of tobacco seedlings to chilling stress was promoted mainly by putrescine and spermidine. And further study specified that the higher PA contents in the chilling-tolerant tobacco variety were related to higher PA synthetase activities [arginine decarboxylase (ADC) and ornithine decarboxylase (ODC)] and lower oxidase activities (diamine oxidase and polyamine oxidase) under chilling stress. Again, the study demonstrated that PA biosynthesis in tobacco seedlings was mainly dependent on the ODC pathway under chilling stress, through ADC and ODC inhibitor application and recovery treatments. Compared with the ADC inhibitor ( d -arginine), the ODC inhibitor (2-fluoromethyl ornithine) more significantly inhibited PA contents and increased membrane permeability in tobacco seedlings under chilling stress. In summary, the research findings suggested PAs play an important role in regulating the chilling tolerance of tobacco seedlings.

Abstract:Drought is one of the most severe forms of abiotic stresses that threaten the survival of plants, including crops. In turn, plants dramatically change their physiology to increase drought tolerance, including reconfiguration of proteomes. Here, we studied drought-induced proteomic changes in leaves of cultivated tobacco (Nicotiana tabacum), a solanaceous plant, using the isobaric tags for relative and absolute quantitation (iTRAQ)-based protein labeling technology. Of identified 5570 proteins totally, drought treatment increased and decreased abundance of 260 and 206 proteins, respectively, compared with control condition. Most of these differentially regulated proteins are involved in photosynthesis, metabolism, and stress and defense. Although abscisic acid (ABA) levels greatly increased in drought-treated tobacco leaves, abundance of detected ABA biosynthetic enzymes showed no obvious changes. In contrast, heat shock proteins (HSPs), thioredoxins, ascorbate-, glutathione-, and hydrogen peroxide (H2O2)-related proteins were up- or down-regulated in drought-treated tobacco leaves, suggesting that chaperones and redox signaling are important for tobacco tolerance to drought, and it is likely that redox-induced posttranslational modifications play an important role in modulating protein activity. This study not only provides a comprehensive dataset on overall protein changes in drought-treated tobacco leaves, but also shed light on the mechanism by which solanaceous plants adapt to drought stress.

Abstract:Biochemical responses of wheat (Triticum aestivum) to the stress of tetrabromobisphenol A (TBBPA) as an emerging pollutant were examined. The results indicated that reduction of the chlorophyll (CHL) content in wheat leaves could be observed. However, the changes in the CHL content with the increasing TBBPA concentration from 50 to 5000mgkg−1 were insignificant (p>0.05). Increased malondialdehyde levels were detected in wheat leaves after both 7-d and 12-d exposures. The changes in the activity of superoxide dismutases (SOD), peroxidases (POD) and catalases (CAT) in wheat leaves irregularly fluctuated with time as the TBBPA concentration increased. However, significant (p<0.05) decrease in the activity of POD and CAT treated with 500 and 5000mgkg−1 TBBPA could be observed. Our data also showed that the plant has the capacity to tolerate the oxidative stress, but the capacity would be lost with prolonged exposure and increasing TBBPA concentration. There were no dose-response effects in the changes between the activity of antioxidant enzymes (SOD, POD and CAT) and the concentration of TBBPA. The decrease in the activity of POD and CAT could be considered as good biomarkers of serious stress by TBBPA in soil environment.

Abstract:Tetrabromobisphenol A (TBBPA) and its derivatives are widely used as brominated flame retardants. Because of their high production and wide environment distribution, TBBPA derivatives have increased considerable concern. Previous studies have primarily focused on TBBPA, with limited information available on its derivative. In this study, we investigated the uptake, biotransformation and physiological response of two derivatives, Tetrabromobisphenol A bis(allyl ether) (TBBPA BAE) and Tetrabromobisphenol A bis(2,3-dibromopropylether) (TBBPA BDBPE), in Helianthus annus ( H . annus ) through a short-term hydroponic assay. The results revealed that H . annus could absorb TBBPA BAE and TBBPA BDBPE from solution, with removal efficiencies of 98.33 ± 0.5% and 98.49 ± 1.56% after 10 days, respectively, which followed first-order kinetics. TBBPA BAE was absorbed, translocated and accumulated while TBBPA BDBPE couldn't be translocated upward due to its high hydrophobicity and low solubility. The concentrations of TBBPA derivatives in plants peaked within 72 h, and then decreased. We identified twelve metabolites resulting from ether bond breakage, debromination, and hydroxylation in H . annus . The high-level TBBPA BAE suppressed the growth and increased malondialdehyde (MDA) content of H . annus , while TBBPA BDBPE didn't pose a negative effect on H . annus . TBBPA BAE and TBBPA BDBPE increased the activity of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), with higher levels of these enzymes activity found in high concentration treatments. Contrastingly, TBBPA BAE exhibited higher toxicity than TBBPA BDBPE, as indicated by greater antioxidant enzyme activity. The findings of this study develop better understanding of biotransformation mechanisms of TBBPA derivatives in plants, contributing to the assessment of the environmental and human health impacts of these contaminants.

Abstract:We examined the effects of Cd and Zn exposure on the photosynthetic function and the tolerant mechanisms of ROS metabolism in tobacco leaves. The results showed that the photosynthesis inhibition caused by Cd exposure was due to the limitation of both stomatal and non-stomatal factors, while Zn exposure only showed a significant effect on the Gs of tobacco leaves. Cd increased the generation rate of O2•– and the content of H2O2, but Zn did not lead to the ROS burst. Cd enhanced the POD activity, but inhibited SOD and CAT activities. The activities of SOD and POD significantly increased under Zn exposure. Cd inhibited the activity and expression of APX. The up regulation of GPX, GR and GST expression and the increase of their activities also play a positive role in the adaptation to Cd. Although Zn inhibited the activities of APX and GPX, it had little effect on other enzyme activities and protein expression in AsA-GSH cycle. TPX and TrxR activities, Trx-Prx pathway-related proteins are very sensitive to Cd. However, only PrxQ and 2-Cys Prx BAS1, which are located in chloroplast, and Trx-HFC164 were significantly down-regulated in Trx-Prx pathway under Zn exposure.

Abstract:Stomatal oscillation has been described as an efficient mechanism to prevent water loss, decrease the transpiration rate and to improve water use efficiency under severe drought conditions. Former researches demonstrated that ABA-induced H2O2 production and H2O2activated Ca 2+ channels were important mechanism for ABA-induced stomatal oscillation. In this study, how sensitivity to abscisic acid (ABA) regulates stomatal oscillation was investigated with Arabidopsis thaliana mutants (abi1-1, insensitivity to ABA and era1-2, hypersensitive to ABA) and wild types. The results showed that (1) hypersensitivity to ABA strengthens stomatal oscillation and closure induced by ABA; (2) insensitivity to ABA abolishes stomatal oscillation and closure induced by ABA; and (3) there is a positive relationship between stomatal oscillation, closure and sensitivity to abscisic acid. The data indicates that sensitivity to ABA may regulate stomatal oscillation and closure in Arabidopsis thaliana.

Abstract:Abiotic stresses trigger premature leaf senescence by affecting some endogenous factors, which is an important limitation for plant growth and grain yield. Among these endogenous factors that regulate leaf senescence, abscisic acid (ABA) works as a link between the oxidase damage of cellular structure and signal molecules responding to abiotic stress during leaf senescence. Considering the importance of ABA, we collect the latest findings related to ABA biosynthesis, ABA signaling, and its inhibitory effect on chloroplast structure destruction, chlorophyll (Chl) degradation, and photosynthesis reduction. Post-translational changes in leaf senescence end with the exhaustion of nutrients, yellowing of leaves, and death of senescent tissues. In this article, we review the literature on the ABA-inducing leaf senescence mechanism in rice and Arabidopsis starting from ABA synthesis, transport, signaling receptors, and catabolism. We also predict the future outcomes of investigations related to other plants. Before changes in translation occur, ABA signaling that mediates the expression of NYC, bZIP, and WRKY transcription factors (TFs) has been investigated to explain the inducing effect on senescence-associated genes. Various factors related to calcium signaling, reactive oxygen species (ROS) production, and protein degradation are elaborated, and research gaps and potential prospects are presented. Examples of gene mutation conferring the delay or induction of leaf senescence are also described, and they may be helpful in understanding the inhibitory effect of abiotic stresses and effective measures to tolerate, minimize, or resist their inducing effect on leaf senescence.
Exogenous 1′,4′-trans-Diol-ABA Induces Stress Tolerance by Affecting the Level of Gene Expression in Tobacco (Nicotiana tabacum L.)

Abstract:1′,4′-trans-diol-ABA is a key precursor of the biosynthesis of abscisic acid (ABA) biosynthesis in fungi. We successfully obtained the pure compound from a mutant of Botrytis cinerea and explored its function and possible mechanism on plants by spraying 2 mg/L 1′,4′-trans-diol-ABA on tobacco leaves. Our results showed that this compound enhanced the drought tolerance of tobacco seedlings. A comparative transcriptome analysis showed that a large number of genes responded to the compound, exhibiting 1523 genes that were differentially expressed at 12 h, which increased to 1993 at 24 h and 3074 at 48 h, respectively. The enrichment analysis demonstrated that the differentially expressed genes (DEGs) were primarily enriched in pathways related to hormones and resistance. The DEGs of transcription factors were generally up-regulated and included the bHLH, bZIP, ERF, MYB, NAC, WRKY and HSF families. Moreover, the levels of expression of PYL/PYR, PP2C, SnRK2, and ABF at the ABA signaling pathway responded positively to exogenous 1′,4′-trans-diol-ABA. Among them, seven ABF transcripts that were detected were significantly up-regulated. In addition, the genes involved in salicylic acid, ethylene and jasmonic acid pathways, reactive oxygen species scavenging system, and other resistance related genes were primarily induced by 1′,4′-trans-diol-ABA. These findings indicated that treatment with 1′,4′-trans-diol-ABA could improve tolerance to plant abiotic stress and potential biotic resistance by regulating gene expression, similar to the effects of exogenous ABA.

Abstract:The effect of brassinosteroid (BR) on relative water content (RWC), stomatal conductance (g(s)), net photosynthetic rate (PN), intercellular CO2 concentration (Ci), lipid peroxidation level, activities of antioxidant enzymes and abscisic acid concentration (ABA) in tomato (Lycopersicon esculentum)seedlings under water stress was investigated. Two tomato genotypes, Mill. cv. Ailsa Craig (AC) and its ABA-deficient mutant notabilis (not), were used. Water stress was achieved by withholding water and both the AC and not plants were treated with 1 mu M 24-epibrassinolide (EBR) or distilled water as a control. The RWC, g(s), Ci and PN were significantly decreased under water stress. However, EBR treatment significantly alleviated water stress and increased the RWC and PN. EBR application also markedly increased the activities of antioxidant enzymes (catalase, ascorbate peroxidase and superoxide dismutase) while it decreased g(s). Ci and the contents of H2O2 and malondialdehyde (MDA). Interestingly, ABA concentration in AC and not plants was markedly elevated after EBR treatment although the increasing rate and amplitude of ABA in not plants treated by EBR was significantly lower than those in AC plants. Our study suggested that amelioration of the drought stress of tomato seedlings may be caused by EBR-induced elevation of endogenous ABA concentration and/or the activities of antioxidant enzymes. Brachypodium distachyon BdPP2CA6 Interacts with BdPYLs and BdSnRK2 and Positively Regulates Salt Tolerance in Transgenic Arabidopsis .

Abstract:The phytohormone abscisic acid (ABA) is essential in plant responding to biotic and abiotic stresses. Although ABA signaling model is well established in Arabidopsis, ABA receptor PYL family and clade A PP2C subfamily are not yet characterized in monocot model plant Brachypodium distachyon. In this study, we identified 12 PYLs and 8 clade A PP2Cs from B. distachyon genome and successfully cloned 12 PYLs and 7 clade A PP2Cs. Bioinformatic and expression analyses showed that most of the identified genes respond to several signal molecules and abiotic stresses. Protein-protein interaction analysis revealed that many BdPYLs and BdPP2CAs participate in the classic ABA-PYL-PP2C-SnRK2 signaling pathway. A clade A PP2C, designated BdPP2CA6, interacted with BdPYL11 in the absence of ABA and localized in nucleus. Most clade A PP2C members from Arabidopsis showed negatively regulation in ABA signaling pathway, whereas BdPP2CA6-overexpression transgenic Arabidopsis showed ABA hypersensitive phenotype, resulting in enhanced stomatal closure and salinity tolerance. Our results indicate that BdPP2CA6 positively regulates ABA and stress signal pathway in transgenic Arabidopsis plant seedlings.

Abstract:Tetrabromobisphenol A (TBBPA) is one of the most widely used brominated flame retardants. In the present study, the accumulation of TBBPA and its consequent biological responses were examined in coontail (Ceratophyllum demersum L.) over 14 days' exposure. Most of the TBBPA was accumulated after 4d exposure and TBBPA concentration in plant increased with increasing TBBPA concentration in growth solution (R(2)=0.99). By using electron paramagnetic resonance (EPR), we found that the TBBPA exposure significantly increased total free radicals generation in the plants. A good positive relationship (R(2)=0.99) was found between the free radicals formation and accumulation level of TBBPA in plant. Lipid peroxidation was enhanced and chlorophyll content showed declined after TBBPA exposure. The activities of superoxide dismutase (SOD), peroxidase (POD), and the contents of glutathione were also detected. The results suggest that TBBPA accumulation in C. demersum induces oxidative stress and the level of tolerance depends on the antioxidative capacity of the plants.

Abstract:Tetrabromobisphenol A (TBBPA), Tetrachlorobisphenol A (TCBPA), Tetrabromobisphenol S (TBBPS) and their derivatives as the most widely used halogenated flame retardants (HFR), had been employed in the manufacturing industry to raise fire safety. HFRs have been shown to be developmentally toxic to animals and also affect plant growth. However, little was known about the molecular mechanism responded by when plants were treated with these compounds. In this study, when Arabidopsis was exposed to four HFRs (TBBPA, TCBPA, TBBPS-MDHP, TBBPS), the stress of these compounds had different inhibitory effects on seed germination and plant growth. Transcriptome and metabolome analysis showed that all four HFRs could influence the expression of transmembrane transporters to affect ion transport, Phenylpropanoid biosynthesis, Plant-pathogen interaction, MAPK signalling pathway and other pathways. In addition, the effects of different kinds of HFR on plants also have variant characteristics. It is very fascinating that Arabidopsis shows the response of biotic stress after exposure to these kinds of compounds, including the immune mechanism. Overall, the findings of the mechanism recovered by methods of transcriptome and metabolome analysis supplied a vital insight into the molecular perspective for Arabidopsis response to HFRs stress.

Abstract:The presence of the ozone hole increases the amount of UV radiation reaching a plant's surface, and UV-B radiation is an abiotic stress capable of affecting plant growth. Rhododendron chrysanthum Pall. (R. chrysanthum) grows in alpine regions, where strong UV-B radiation is present, and has been able to adapt to strong UV-B radiation over a long period of evolution. We investigated the response of R. chrysanthum leaves to UV-B radiation using widely targeted metabolomics and transcriptomics. Although phytohormones have been studied for many years in plant growth and development and adaptation to environmental stresses, this paper is innovative in terms of the species studied and the methods used. Using unique species and the latest research methods, this paper was able to add information to this topic for the species R. chrysanthum. We treated R. chrysanthum grown in a simulated alpine environment, with group M receiving no UV-B radiation and groups N and Q (externally applied abscisic acid treatment) receiving UV-B radiation for 2 days (8 h per day). The results of the MN group showed significant changes in phenolic acid accumulation and differential expression of genes related to phenolic acid synthesis in leaves of R. chrysanthum after UV-B radiation. We combined transcriptomics and metabolomics data to map the metabolic regulatory network of phenolic acids under UV-B stress in order to investigate the response of such secondary metabolites to stress. L-phenylalanine, L-tyrosine and phenylpyruvic acid contents in R. chrysanthum were significantly increased after UV-B radiation. Simultaneously, the levels of 3-hydroxyphenylacetic acid, 2-phenylethanol, anthranilate, 2-hydroxycinnamic acid, 3-hydroxycinnamic acid, α-hydroxycinnamic acid and 2-hydroxy-3-phenylpropanoic acid in this pathway were elevated in response to UV-B stress. In contrast, the study in the NQ group found that externally applied abscisic acid (ABA) in R. chrysanthum had greater tolerance to UV-B radiation, and phenolic acid accumulation under the influence of ABA also showed greater differences. The contents of 2-phenylethanol, 1-o-p-coumaroyl-β-d-glucose, 2-hydroxy-3-phenylpropanoic acid, 3-(4-hydroxyphenyl)-propionic acid and 3-o-feruloylquinic ac-id-o-glucoside were significantly elevated in R. chrysanthum after external application of ABA to protect against UV-B stress. Taken together, these studies of the three groups indicated that ABA can influence phenolic acid production to promote the response of R. chrysanthum to UV-B stress, which provided a theoretical reference for the study of its complex molecular regulatory mechanism.

Abstract:Suspensions of ovine luteal cells were used to study the effects of inhibitors of transglutaminase on the internalization of [125I]hCG. Ovine corpora lutea were collected from ewes during the midluteal phase of the estrous cycle, dissociated into cell suspensions with collagenase, incubated with [125I]hCG for 5–7 min, and washed five times to remove radioactivity which was not bound to receptor. The cells were then incubated for 6– 12 h in the presence of bacitracin (1 mM), ethylamine (1–5 DIM), methylamine (5–15 mM), or dansylcadaverine (100–500 μM) to determine the effects of these inhibitors of transglutaminase on the rates of internalization and degradation of the receptorbound [125I]hCG and/or progesterone secretion. Characterization of Organellar-Specific ABA Responses during Environmental Stresses in Tobacco Cells and Arabidopsis Plants

Abstract:Abscisic acid (ABA) is a critical phytohormone involved in multifaceted processes in plant metabolism and growth under both stressed and nonstressed conditions. Its accumulation in various tissues and cells has long been established as a biomarker for plant stress responses. To date, a comprehensive understanding of ABA distribution and dynamics at subcellular resolution in response to environmental cues is still lacking. Here, we modified the previously developed ABA sensor ABAleon2.1_Tao3 (Tao3) and targeted it to different organelles including the endoplasmic reticulum (ER), chloroplast/plastid, and nucleus through the addition of corresponding signal peptides. Together with the cytosolic Tao3, we show distinct ABA distribution patterns in different tobacco cells with the chloroplast showing a lower level of ABA in both cell types. In a tobacco mesophyll cell, organellar ABA displayed specific alterations depending on osmotic stimulus, with ABA levels being generally enhanced under a lower and higher concentration of salt and mannitol treatment, respectively. In Arabidopsis roots, cells from both the meristem and elongation zone accumulated ABA considerably in the cytoplasm upon mannitol treatment, while the plastid and nuclear ABA was generally reduced dependent upon specific cell types. In Arabidopsis leaf tissue, subcellular ABA seemed to be less responsive when stressed, with notable increases of ER ABA in epidermal cells and a reduction of nuclear ABA in guard cells. Together, our results present a detailed characterization of stimulus-dependent cell type-specific organellar ABA responses in tobacco and Arabidopsis plants, supporting a highly coordinated regulatory network for mediating subcellular ABA homeostasis during plant adaptation processes.

H2O2 Mediates the Crosstalk of Brassinosteroid and Abscisic Acid in Tomato Responses to Heat and Oxidative Stresses

Abstract:The production of H2O2 is critical for brassinosteroid (BR)- and abscisic acid (ABA)-induced stress tolerance in plants. In this study, the relationship between BR and ABA in the induction of H2O2 production and their roles in response to heat and paraquat (PQ) oxidative stresses were studied in tomato. Both BR and ABA induced increases in RBOH1 gene expression, NADPH oxidase activity, apoplastic H2O2 accumulation, and heat and PQ stress tolerance in wild-type plants. BR could only induced transient increases in these responses in the ABA biosynthetic mutant notabilis (not), whereas ABA induced strong and prolonged increases in these responses in the BR biosynthetic mutant d (^im) compared with wild-type plants. ABA levels were reduced in the BR biosynthetic mutant but could be elevated by exogenous BR. Silencing of RBOH1 compromised BR-induced apoplastic H2O2 production, ABA accumulation, and PQ stress responses; however, ABA-induced PQ stress responses were largely unchanged in the RBOH1-silenced plants. BR induces stress tolerance involving a positive feedback mechanism in which BR induces a rapid and transient H2O2 production by NADPH oxidase. The process in turn triggers increased ABA biosynthesis, leading to further increases in H2O2 production and prolonged stress tolerance. ABA induces H2O2 production in both the apoplastic and chloroplastic compartments.

Abstract:Abscisic acid (ABA) has been implicated as a mediator in plant responses to various environmental stresses. To evaluate the transcriptional and metabolic events downstream of ABA perception, Arabidopsis thaliana seedlings were analyzed by transcript and metabolite profiling, and results were integrated, using the recently developed BioPathAt tool, in the context of the biochemical pathways affected by this treatment. Besides the up-regulation of pathways related to the biosynthesis of compatible solutes (raffinose family oligosaccharides and certain amino acids) as a response to ABA treatment, we also observed a down-regulation of numerous genes putatively localized to and possibly involved in the reorganization of cell walls, an association that had not been recognized previously. Metabolite profiling indicated that specific antioxidants, particularly α-tocopherol and l-ascorbic acid, were accumulated at higher levels in ABA-treated seedlings compared to appropriate controls. The transcription of genes involved in α-tocopherol biosynthesis were coordinately up-regulated and appeared to be integrated into a network of reactions controlling the levels of reactive oxygen species. Based upon the observed gene expression patterns, these redox control mechanisms might involve an ABA-mediated transition of mitochondrial respiration to the alternative, non-phosphorylating respiratory chain mode. The presented data herein provide indirect evidence for crosstalk between metabolic pathways and pathways regulating redox homeostasis as a response to ABA treatment, and allowed us to identify candidate genes for follow-up studies to dissect this interaction at the biochemical and molecular level. Our results also indicate an intricate relationship, at the transcriptional and possibly post-transcriptional levels, between ABA biosynthesis, the xanthophyll cycle, and ascorbic acid recycling.
Phytochrome alleviates cadmium toxicity by regulating gibberellic acid and brassinolide in Nicotiana tabacum L.

Abstract:Soil cadmium (Cd) pollution has emerged as a substantial environmental challenge globally, hampering crop production and endangering human health. Here, we found that photoreceptor phytochromes (PHYs) were involved in regulating Cd tolerance in tobacco. Compared to wildtype (WT) plants, phytochrome-defective mutants ( phyA , phyB , p hyAB ) displayed Cd sensitive phenotype, and had a higher reactive oxygen species (ROS) accumulation and malondialdehyde content. However, differences in Cd concentration among phyA mutants, phyB mutants, phyAB mutants, and WT plants were not observed. Consequently, the higher tolerance promoted the biomass of WT plants, thereby increasing the Cd accumulation. Furthermore, Cd stress altered the levels of gibberellin (GA) and brassinosteroid (BR), and these phytohormones were higher in WT plants. GA3 application induced the transcription of genes encoding antioxidant enzyme and suppressed the expression of genes associated with chlorophyll degradation, inhibiting chlorophyll breakdown and decreasing ROS levels in plants under Cd stress conditions. Additionally, epibrassinolide spraying promoted the expression of genes related to chlorophyll synthesis, thereby increasing chlorophyll content and maintaining plant acquisition ability. Our results suggested that phytochromes enhanced the tolerance of Nicotiana tabacum to Cd stress through regulating BR and GA.
