Abstract:Objective To visualize the topographical changes of serum albumin passing through mouse glomerular capillary loops and its reabsorption renal proximal tubules by immunohistochemistry combination with in vivo cryotechnique.Methods The in vivo cryotechnique was used to treat the left kidneys of mice under normotensive,experimentally acute hypertensive and heart-arrest conditions. The cryofixed tissues were routinely processed for freeze-substitution. Serial deparaffinized sections were stained with hematoxylin-eosine and immunostained with anti-mouse albumin antibody. Results Under the normotensive and heart-arrest conditions,albumin was clearly immunolocalized the blood vessels and slightly the apical cytoplasmic parts of some proximal tubules. Under the acute hypertensive condition,the albumin was strongly immunolocalized the apical cytoplasm of almost all proximal tubules.Conclusion The in vivo cryotechnique combined with freeze-substitution can transiently capture the reabsorption of albumin living functional renal proximal tubules and can also visualize the functional morphology living animal tissues and cells situ.

Abstract:The light‐sensitive protein Opsin 3 (Opn3) is present throughout the mammalian brain; however, the role of Opn3 in this organ remains unknown. Since Opn3 encoded mRNA is modulated in the supraoptic and paraventricular nucleus of the hypothalamus in response to osmotic stimuli, we have explored by in situ hybridization the expression of Opn3 in these nuclei. We have demonstrated that Opn3 is present in the male rat magnocellular neurones expressing either the arginine vasopressin or oxytocin neuropeptides and that Opn3 increases in both neuronal types in response to osmotic stimuli, suggesting that Opn3 functions in both cell types and that it might be involved in regulating water balance. Using rat hypothalamic organotypic cultures, we have demonstrated that the hypothalamus is sensitive to light and that the observed light sensitivity is mediated, at least in part, by Opn3. The data suggests that hypothalamic Opn3 can mediate a light‐sensitive role to regulate circadian homeostatic processes.

Abstract:Objective:To observe the pathologic changes of the retinal ganglion cells(RGCs)and capillary in rats with diabetes mellitus for seven months.Methods:Diabetic rats were induced by streptozotocin.After seven months we removed the eyes under anesthesia and made retinal slides and digest preparations.The RGCs and capillary were observed by light and electron microscopy.Results:In light microscopy the retinal tissues were indistinct disturbance and capillaries were enlarged.There were some early symptoms of diabetic retinopathy(DR),such as extensive pericyte loss,endothelial cell proliferation,pericyte ghost,acellular capillary and apoptosis of endothelial cell.Retinal nerve fiber layer was edematous,nuclear shrinkage and mitochondrion condensed.In electron microscopy endothelial cell of capillary and mitochondrial was edematous obviously,the basement membranes were thickened.RGCs were vacuolated,chromatin of nucleus aggregated marginally decreased and densification.Some of RGCs were necrosis,nucleus were dissolved,mitochondria was edematous,the rough endoplasmic reticula decreased obviously.Conclusion:In the early stage of DR in rats,accompanying the retinal capillary,the RGCs were damaged.

Abstract:The immunohistochemical localization of tyrosine hydroxylase, calbindin D-28K, calretinin, parvalbumin and olfactory marker protein in olfactory bulb of adult Sprague Dawley rat was studied. Tyrosine hydroxylase are mainly found in glomerular layer,few in external plexiform layer. Calbindin D-28K is mainly found in glomerular layer, few in external plexiform layer and internal plexiform layer. Calretinin is found in glomerular layer, external plexiform layer, mitral cell layer, internal plexiform layer and granule cell layer except olfactory nerve layer. Parvalbumin is mostly found in external plexiform layer, some in mitral cell layer, few in glomerular layer. Olfactory marker protein is found only in olfactory nerve layer and glomerular layer. Results are similar to the pioneering research in this field, but some difference exists.
Isolation of microglia from retinas of chronic ocular hypertensive rats

Abstract:Microglia are the principal glial cells involved in the processes of immune inflammation within both retina and optic nerve, especially under the context of glaucomatous neuropathy. Considering the distinguishing role of retinal microglia in glaucoma and the lack of established protocol for microglia isolation from animal glaucoma model, the present study aimed to develop and validate a method with characteristics of both simplicity and efficiency for retinal microglia isolation from chronic ocular hypertensive (COH) rats. A Percoll gradient of various concentrations was used to separate microglia from whole retinal cells of the COH rats and control group. The finally isolated microglia were identified by CD11b and Iba-1 immunofluorescence staining, and the cell viability was determined by trypan blue staining. Additionally, the proportion of microglia in the whole retina cells was identified by flow cytometry. Results showed that the survival rates of isolated retinal microglia with the Percoll gradient method were 67.2 +/- 4% and 67.6 +/- 3% in control and COH groups, respectively. The proportion of the microglia population in the whole retinal cells was about 0.4-0.93%. To conclude, the present study confirmed that the application of Percoll gradient could effectively separate microglia from retinas of COH rats, which will probably enrich the tool kit for basic researchers of glaucoma specialty and help with scientific investigations.
A comparison of neuropeptide immunocytochemistry in fluid-fixed and freeze-dried brains

Abstract:Immunocytochemical staining of luteinizing hormone-releasing hormone (LHRH), somatostatin, and neurophysin was compared in rat brains fixed with 1) formalin, 2) Bouin's solution, 3) freeze-dried (FD), or 4) freeze-dried + paraformaldehyde vapor perfused (FDV). The distribution of LHRH fibers was similar in all preparations; however, beads of granular reaction product often appeared finer and more numerous in the median eminence of FD- and FDV brains. Positively stained LHRH perikarya were not observed in any of the preparations. In contrast, somatostatin-immunoreactive perikarya were present in the fluid-fixed and FD brains, although few were observed in FDV brains. Somatostatin-immunoreactive fibers were present in all preparations, but appeared most numerous in the median eminence of FD brains. Staining of neurophysin-containing perikarya and fibers was similar in all preparations. These observations suggest that the FD brain can provide a suitable tissue substrate for immunocytochemistry, demonstrating staining comparable to or surpassing that of more conventional preparations. However, staining of antigens in FD brain was not uniformly successful and may depend on stereochemical characteristics of each antigen as well as properties of the primary antisera used in the staining procedure.

Thus, the "in vivo cryotechnique" with freeze-substitution can be used for clarifying not only the functional morphology of living animal cells, but also in situ immunohistochemical localization of their components.
Regulation of Gene Expression in Magnocellular Neurons in Rat Supraoptic Nucleus During Sustained Hypoosmolality

Abstract:Hypoosmolality produces a dramatic inhibition of vasopressin (VP) and oxytocin gene expression in the supraoptic nucleus (SON). This study examines the effect of sustained hypoosmolality on global gene expression in the oxytocin and VP magnocellular neurons of the hypothalamo-neurohypophysial system, to identify genes associated with the magnocellular neuron’s adaptation to this physiological condition. Using laser microdissection of the SON, T7-based linear amplification of its RNA, and a 35,319-element cDNA microarray, we compare gene expression profiles between SONs in normoosmolar (control), 1-desamino-[8-d-arginine]-VP-treated normoosmolar, and hypoosmolar rats. We found 4959 genes with statistically significant differences in expression between normosmolar control and the hypoosmolar SONs, with 1564 of these differing in expression by more than 2-fold. These genes serve a wide variety of functions, and most were up-regulated in gene expression in hypoosmolar compared with control SONs. Of these, 90 were preferentially expressed in the SON, and 44 coded for transcription-related factors, of which 15 genes were down-regulated and 29 genes were up-regulated in the hypoosmolar rat SONs. None of these transcription-related factor genes significantly changed in expression after sustained 1-desamino-[8-d-arginine]-VP-treatment alone, indicating that these changes were associated with the hypoosmolar state and not due solely to a decreased activity in the SON. Quantitative in situ hybridization histochemistry was selectively used to confirm and extend these microarray observations. These results indicate that the hypoosmolar state is accompanied by a global, but selective, increase in expression of a wide variety of regulatory genes in the SON.
Application of Periodic Acid-Schiff Fluorescence Emission for Immunohistochemistry of Living Mouse Renal Glomeruli by an "in Vivo Cryotechnique"

Abstract:To identify the distribution of endogenous serum proteins in living mouse renal glomeruli under various hemodynamic conditions, we used the periodic acid-Schiff (PAS) and its fluorescence emission as a marker for the glomerular basement membrane (GBM). The immunostaining for collagen type IV was hardly observed without microwave treatment in specimens prepared by an "in vivo cryotechnique". However, PAS staining and its fluorescence emission could be clearly visualized at the GBM with the "in vivo cryotechnique". Under normotensive conditions, immunoreaction products of albumin and immunoglobulin G heavy and light chains (IgG(H+L)) were localized within glomerular capillary loops (GCL) but not colocalized with the PAS fluorescence emission of the GBM. Under heart-arrest conditions and with quick-freezing of resected tissues, albumin, IgG (H+L), immunoglobulin kappa light chain, and IgG1 heavy chain (IgG1) were immunolocalized within the GCL and mesangial areas, but only albumin and the kappa light chain were additionally immunolocalized in Bowman's space, indicating their passage through the GBM. Under acute hypertensive conditions, both albumin and the kappa light chain, but not IgG1, were clearly immunolocalized along the GBM and in the Bowman's space, indicating their increased passage through the GBM. The overlapping areas of PAS fluorescence emission and the albumin or kappa light chain appeared to be larger with quick-freezing and under the heart arrest or acute hypertensive conditions than under normal circulation, whereas those of PAS emission and IgG1 did not differ among these conditions. The serum proteins passing through the GBM were clearly visualized with the "in vivo cryotechnique", immunofluorescence staining, and PAS fluorescence emission.
Immunohistochemical Detection of Phosphorylated Rhodopsin in Light-exposed Retina of Living Mouse with in Vivo Cryotechnique

Abstract:The purpose of this study is to analyze the time-dependent molecular states of rhodopsin (Rho) phosphorylation in the specimens originating from eyeballs cryoimmobilized in situ in living animals. Whole eyeballs of living mice under various dark- and light-exposure conditions were quickly frozen using the in vivo cryotechnique with isopentane-propane cryogen cooled down in liquid nitrogen (-196C). The frozen whole-mount eyeballs were freeze substituted in acetone containing paraformaldehyde and embedded in paraffin wax. Deparaffinized sections were immunostained with anti-phosphorylated (334)Ser Rho (P-Rho334) antibody. Immunoreactivity of P-Rho334 was specifically recognized in the outer segments of mouse retinas exposed to daylight. In the 12-h dark-adapted retinas, P-Rho334 immunoreactivity was completely eliminated. Moreover, in other retinas dark adapted for 12 or 36 hr and then exposed under the safety red light for 2 min, it was still barely recognized. Even in the eyeballs exposed to strong visible light for 10 sec, it was not detected. However, after 30, 60, and 180 sec of visible light exposure, P-Rho334 immunoreactivity was definitely recovered, similar to that under daylight condition. This is a new immunohistochemical approach to visualize the time-dependent Rho phosphorylation of living mice using the in vivo cryotechnique, in which changes could be detected within seconds following exposure to light.
Preparation of Newborn Rat Brain Tissue for Ultrastructural Morphometric Analysis of Synaptic Vesicle Distribution at Nerve Terminals.

Abstract:Our laboratory and many others have exploited the high resolving power of transmission electron microscopy to study the morphology and spatial organization of synaptic vesicles. In order to obtain high-quality electron micrographs that can yield the degree of morphological detail necessary for quantitative analysis of pre-synaptic vesicle distribution, optimal specimen preparation is critical. Chemical fixation is the first step in the process of specimen preparation, and of utmost importance to preserve fine ultrastructure. Vascular fixation with a glutaraldehyde-formaldehyde solution, followed by treatment of vibratome-sectioned specimens with osmium tetroxide, stabilizes the maximum number of molecules, especially proteins and lipids, and results in superior conservation of ultrastructure. Tissue is then processed with counterstaining, sequential dehydration and resin-embedding. En bloc staining with uranyl acetate (i.e., staining of vibratome-sectioned tissue before resin embedding) enhances endogenous contrast and stabilizes cell components against extraction during specimen processing. Contrast can be further increased by applying uranyl acetate as a post-stain on ultrathin sections. Double-staining of ultrathin sections with lead citrate after uranyl acetate treatment also improves image resolution, by intensifying electron-opacity of nucleic acid-containing structures through selective binding of lead to uranyl acetate. Transmission electron microscopy is a powerful tool for characterization of the morphological details of synaptic vesicles and quantification of their size and spatial organization in the terminal bouton. However, because it uses fixed tissue, transmission electron microscopy can only provide indirect information regarding living or evolving processes. Therefore, other techniques should be considered when the main objective is to study dynamic or functional aspects of synaptic vesicle trafficking and exocytosis.

Abstract:The changes of activities of enzymes relating to energy metabolism in rabbit's retina in acute ocular hypertension were observed. The activities of succinate dehydrogenase and adenosine triphosphatase were found to be reduced, while the activities of the lactatic dehydrogenase and glucose-6-phosphatase increased. The results revealed the metabolic disturbance of energy in retina after acute ocular hypertension might be the underlying factors relating to the defects of the functions and structures of the retina.
Confocal laser scanning microscopy of whole mouse ovaries: Excellent morphology, apoptosis detection, and spectroscopy

Abstract:BACKGROUND: Ovaries consist of numerous follicles, oocytes, and granulosa cells in different stages of development. Many of these follicles will undergo an apoptotic process during the lifetime of the animal. By using proper tissue preparation methods, the events within the whole ovary can be observed by using 3D confocal microscopy.METHODS: Whole ovaries were stained with LysoTracker Red (LT), fixed with 4% paraformaldehyde (PF) and 1% glutaraldehyde (Glut), dehydrated with methanol (MEOH), and cleared with benzyl alcohol and benzyl benzoate (BABB). Using this tissue preparation technique, the ovary becomes relatively transparent, allowing its morphology to be observed with confocal microscopes. A spectral imaging system (PARISS) located on a conventional microscope was used to interpret the LT dye spectra and fixation products in the tissues with different excitation wavelengths.RESULTS: Apoptosis in the follicle was detected as clusters of intensely stained granulosa cells located in close proximity to the oocytes. The fixation with Glut and PF preserved morphological details, increased tissue fluorescence, thus increased the signal to noise of the background image.CONCLUSIONS: Thick tissues can be imaged after they are properly stained, aldehyde fixed, and BABB cleared. LT intensely stained single cells or clusters of apoptotic cells in the follicles and the nucleolus. Spectral differences between LT as an indicator of apoptosis and Glut-PF fixation was used to visualize ovarian morphology and apoptosis. The PARISS spectrophotometer revealed spectral peaks for LT at 609.6 nm and for Glut-PF at 471.3 nm. The proper use of the spectra from these fluorescence molecules is the foundation for high quality morphological images of apoptosis. By sequentially imaging the two probes with a 488 nm laser and a 543/568 nm laser, there was a reduction in fluorescent cross talk and an increase in image quality.

Abstract:The chamber angle of the eye of pigmented laboratory rats was studied in normal structure and experimentally with the electron microscope. Their exists a circular, flattened canal which corresponds in its topographical position and in its ultrastructural construction to the canal of Schlemm in humans and primates. A trabecular meshwork, comparable to the trabeculum cribriforme in primate eyes, lies directly below the inner wall of Schlemm's canal. The endothelial cells of the trabeculae show phagocytotic activity. The trabecular meshwork is, however, far less intensive in the rat eye than in that of man and it extends up to the spaces of Fontana. Beside a few differences there exist great similarities in structure and function of the chamber angle of the rat eye compared to the corresponding area of eye of man and primates.

Abstract:The effect of ovariotomy on virgin female rat central nervous systems was investigated in female rat hypothalamus by locating the distribution of c-fos expression protein labelled neurons using the immunocytochemistry technique. Two hours later after termination of ovariotomy, the c-fos expression neurons were detected bilaterally in periventricular nucleus of the hypothalamus (PVNH), medial amygdala nucleus (MAN), anterior nucleus of the hypothalamus (ANH), lateral preoptic nucleus (LPN), supra-chiasmatic nucleus (SCN), and especially in bilateral medical preoptic nucleus (MPN), arcuate nucleus (AR), ventromedial nucleus of the hypothalamus (VNH) and paraventricular nucleus of the hypothalamus (PANH). These results suggest that the above structures constituted a main part of the feedback regulation system of hypothalamus-pituitary-gonadal axis and the activation of these nuclei, induced by ovariotomy which produced deficiency of estrogen in blood, was involved in modulating the function of that axis.

Abstract:Yang et al. 1 have recently shown that the hypothalamus preoptic area (POA) can be stimulated via a remote source of ultrasound applied to the skull of mice or rats. The effect of this stimulation was a decrease in oxygen consumption ( O 2 ), resulting in hypothermia and torpor in the mice, and a reduction in O 2 , without torpor, in the rats. This acute and fast decline in metabolic rate replicates the long-established metabolic response in small mammals to various physiological or pharmacological stimulations of the POA, such as those produced by, but not limited to, moderate hypoxia 2,3,4,5,6 .

Abstract:Neurophil within the terminal nuclei of the accessory optic tract of the rat showed intense to moderate μ-opioid receptor-like immunoreactivity (MOR-LI). After unilateral enucleation, MOR-LI within the terminal nuclei almost disappeared or was markedly reduced on the side contralateral to the operation. Electron microscopy revealed that MOR-LI axon terminals within the terminal nuclei were filled with round synaptic vesicles and in asymmetric synaptic contact mainly with dendritic profiles, and occassionally with somatic profiles.

Abstract:Torpor is a state used by several mammals to survive harsh winters and avoid predation, characterized by a drastic reduction in metabolic rate followed by a decrease in body temperature, heart rate, and many physiological variables. During torpor, all organs and systems must adapt to the new low-energy expenditure conditions to preserve physiological homeostasis. These adaptations may be exploited in a translational perspective in several fields. Recently, many features of torpor were shown to be mimicked in non-hibernators by the inhibition of neurons within the brainstem region of the Raphe Pallidus. The physiological resemblance of this artificial state, called synthetic torpor, with natural torpor has so far been described only in physiological terms, but no data have been shown regarding the induced morphological changes.
