Abstract:The two-spotted spider mite Tetranychus urticate is an important agricultural pest worldwide. It is extremely polyphagous and has developed resistance to many pesticides. Here, we assessed the pesticide resistance of seven field populations of T. urticae in China, their target site mutations and the activities of their detoxification enzymes. The results showed that abamectin and the traditional pesticides pyridaben, profenofos and bifenthrin had higher resistance or lower toxicity than more recently developed pesticides including chlorfenapyr, spinetoram, cyflumetofen, cyenopyrafen, bifenazate and B-azolemiteacrylic. The frequency of point mutations related to abamectin resistance, G314D in the glutamate-gated chloride channel 1 (GluCl1) and G326E in GluCl3, ranged 47%-70% and 0%-97%, respectively. The frequency of point mutations in A1215D and F1538I of the voltage-gated sodium channel gene (VGSC), which may increase resistance to pyrethroids, ranged 88%-100% and 10%-100%, respectively. For target sites related to organophosphate resistance, mutation frequencies ranged 25%-92% for G119S and 0%-23% for A201S in the acetycholinesterase gene (Ace). Mutation G126S in the bifenazate resistance-related cytochrome b gene (Cytb) was observed in three of the seven T. urticae populations. Higher activities of detoxification enzymes (P450, GST, CarEs and UGTs) were observed in two T. urticae populations, with significant difference in the XY-SX population. These results provide useful information on the status of pesticide resistance of T. urticae in China and suggest that T. urticae field populations may have multiple resistance mechanisms.

Abstract:The intensive use of pesticides is a common practice for the management of the two-spotted spider mite, Tetranychus urticae, in greenhouses and field farms of Ethiopia. However, incidence of resistance and possible resistance mechanisms in T. urticae populations from Ethiopia have not yet been studied. Here, we assessed the toxicity of various acaricides—bifenazate, abamectin, emamectin benzoate, profenofos, fenbutatin oxide, fenpyroximate, amitraz and chlorfenapyr—on T. urticae populations sampled from six flower greenhouse farms, three strawberry greenhouse farms, one field-grown vegetable farm and two wild populations. In parallel, all populations were screened for known target-site mutations. All tested populations were fully susceptible to bifenazate, abamectin, emamectin benzoate and profenofos, but resistant against fenbutatin oxide and fenpyroximate. Four populations showed considerable levels of resistance against amitraz and one population was resistant to chlorfenapyr. Several target-site mutations were identified in the tested populations, including G119S, A201S, T280A, G328A and F331W/C/Y in acetylcholinesterase and the F1538I and L1024V mutation in the voltage-gated sodium channel. The F1538I mutation was found in eight out of 12 populations, whereas the L1024V mutation was only found in two populations. The H92R mutation in the PSST subunit of complex I and the I1017F mutation in chitin synthase 1 was detected in half of the tested populations. The G326E and I321T mutations in the glutamate-gated chloride channel 3 were also detected, but more rarely, whereas mitochondrial cytochrome b mutations were not detected. The current study revealed multiple resistance patterns in Ethiopian T. urticae populations and together with the wide presence of target-site mutations, calls for the wise use of acaricides in the management of T. urticae in Ethiopia.

Abstract:The spider mite Tetranychus urticae is a polyphagous pest that establishes high levels of resistance to acaricides in industrial glasshouses. In the present paper, spider mite lines selected for resistance to acaricides were used to compare point mutations in the key resistance-associated genes. Both resistant (R-) and susceptible (S-) lines were obtained after 188 and 44 generations of selection to bifenthrin and abamectin, respectively. In bifenthrin-selected mites, para sodium ion channel of the plasma membrane (para SC) locus was analysed and one non-synonymous and one synonymous point mutations were found. The same mutations were reported previously in T. urticae that developed resistance to bifenthrin under natural conditions. As for abamectin-selected lines, glutamate-gated chloride channel (GluCl) locus was analysed, and no mutations were found. Thus, the para SC mutations in strains and lines of mites with developed resistance to pyrethroid acaricides is a widespread phenomenon in T. urticae. Experimental selection results in mutations that are also formed under natural conditions and may therefore serve as a model to study acaricide resistance mechanisms in spider mites. On the other hand, mutations in GluCl gene of avermectin-resistant mites are not so common, inferring involvement of alternative mechanisms of resistance.

Monitoring of Early Stage Resistance to Novel Classes of Insecticides in Spodoptera Exigua (Lepidoptera:Noctuidae)

Abstract:Several classes of new insecticides with novel mode of action,such as emamectin benzoate,abamectin,indoxacarb,chlorfenapy,methoxyfenozide,JS118 and tebufenozide were developed or introduced to control the beet armyworm,spodoptera exigua(Hubner)in China.Resistance of this pest to many conventional insecticides has been reported in different countries and areas,but there is few evidence of resistance to these new insecticides in beet armyworm as yet.In order to make good use of these new insecticides and implement effective resistance management strategy,susceptibility to several classes of new insecticides was determined using leaf-dipping method in field populations of Spodoptera exigua from six areas of Jiangsu and Henan Province,in China.The results showed that in all six field populations tested with abamectin,three populations from Fengxian and Fu'ning counties,Jiangsu province and Henan province were susceptible or had decreased susceptibility(2.5~4.4-fold),one population from Dafeng county,Jiangsu province developed low level resistance(5.2-fold),and two populations from Jiangning and Sheyang counties,Jiangsu province first developed moderate levels of resistance(10.4~14.2-fold).In all four populations tested,three populations from Henan province and Jiangning,Fu'ning and Fengxian of Jiangsu province were susceptible(2.3~2.9-fold)to indoxacarb except that one population from Fengxian of Jiangsu province had low level of resistance(6.8-fold)to indoxacarb.Four populations from Henan province and Jiangning,Fu'ning and Sheyang counties of Jiangsu province were susceptible to emamectin benzoate(RR0.6~2.0-fold).In case of chlorfenapy,all four populations tested were susceptible(1.5~2.7-fold).Similarly,all populations tested were susceptible to methoxyfenozide(0.4~1.7-fold)and JS118(0.3~1.3-fold).In all six populations tested with tebufenozide,four populations from Dafeng,Jiangning and Fengxian of Jiangsu province and Henan province were susceptible(1.6~2.5-fold),and two populations from Fu'ning and Sheyang,Jiangsu province had decreased susceptibility(3.2-fold).These results provide a guideline for developing a more effective resistance management program for beet armyworm.

Abstract:The presence of cellular renal casts in the newborn infant is considered evidence of renal tubular injury. A cause-and-effect relationship between anoxia and these renal lesions is suggested by this study. The histopathologic manifestations of tubular disease best demonstrated in sections stained for fat and with the periodic acid-Schiff routine are dependent not only upon the severity and duration of anoxia, but also upon the length of survival after injury. In this regard, the finding of urinary cellular casts in the newborn infant may suggest to the clinician an early and vigorous approach against the pathologic effects of anoxia.

Abstract:The two-spotted spider mite, Tetranychus urticae Koch, is a constant threat to sustainable production of numerous economically important crops globally. Management of T. urticae is heavily reliant on the application of synthetic acaricides. However, T. urticae has rapidly developed resistance to most of the acaricides available for its control due to its very broad host plant range, extremely short lifecycle, high fecundity, arrhenotokous parthenogenesis, and overwintering strategy. Despite the recent progress in identifying genetic changes/markers associated with resistance toward some commonly used acaricides for T. urticae, there is still limited studies that select and apply these markers in field populations to guide sustainable pest management strategy design. Temporal and spatial characterization of acaricide-resistant phenotypes and their underlying mechanisms are crucial for the design and implementation of successful and sustainable integrated mite management programs. This review highlights the current acaricide resistance status of field-collected T. urticae populations and the underlying molecular mechanisms of resistance. Our review found that some genetic mutations in target sites and/or overexpression of metabolic genes confer resistance in geographically exclusive populations, while some resistance markers appear to be specific to populations at biogeographical areas. Thus, there is a need for locally based coordinated efforts to understand the mechanisms of resistance present in endemic T. urticae populations. Moreover, we discuss a prospective template for designing an effective acaricide resistance management program within various agroecosystems.

Identification and characterization of striking multiple-insecticide resistance in a Tetranychus urticae field population from Greece

Abstract:Background: Tetranychus urticae is a notorious crop pest with a worldwide distribution that has developed resistance to a wide range of acaricides. Here, we investigated the resistance levels of a T. urticae population collected from an ornamental greenhouse in Peloponnese, Greece, and analyzed its resistance mechanisms at the molecular level. Results: Toxicological assays showed resistance against compounds with different modes of action, with resistance ratios of: 89-fold for abamectin; > 1000-fold for clofentezine; > 5000-fold for etoxazole; 27-fold for fenpyroximate and pyridaben; 20- and 36-fold for spirodiclofen and spirotetramat, respectively; and 116- and > 500-fold for cyenopyrafen and cyflumetofen, respectively. Bioassays with synergists indicated the involvement of detoxification enzymes in resistance to abamectin, but not to cyflumetofen and spirodiclofen. RNA sequencing (RNA-seq) analysis showed significant over-expression of several genes encoding detoxification enzymes such as cytochrome P450 monooxygenases and UDP-glycosyltransferases, which have been previously associated with acaricide resistance. Known target-site resistance mutations were identified in acetyl-choline esterase, chitin synthase 1 and NDUFS7/psst, but putative novel resistance mutations were also discovered in targets such as glutamate-gated chloride channel subunit 3. Interestingly, target-site resistance mutations against pyrethroids or bifenazate were not identified, possibly indicating a recent reduced selection pressure in Greece, as well as a possible opportunity to rotate these chemistries. Conclusion: We identified and characterized a striking case of multiple acaricide resistance in a field population of T. urticae. Exceptionally strong resistance phenotypes, with accumulation of multiple resistance mutations and over-expression of P450s and other detoxification genes in the same field population are reported.

Target-Site Mutations and Glutathione S-Transferases Are Associated with Acequinocyl and Pyridaben Resistance in the Two-Spotted Spider Mite Tetranychus urticae (Acari: Tetranychidae)

Abstract:The two-spotted spider mite Tetranychus urticae is a difficult-to-control pest due to its short life cycle and rapid resistance development. In this study, we characterized field strains collected in 2001 and 2003 that were selected for acequinocyl resistance (AR) and pyridaben resistance (PR), respectively. These strains displayed resistance ratios of 1798.6 (susceptible vs. AR) and 5555.6 (susceptible vs. PR), respectively, and were screened for cross-resistance against several currently used acaricides. The AR strain exhibited pyridaben cross-resistance, but the PR strain showed no cross-resistance. The AR strain exhibited point mutations in cytb (I256V, N321S) and PSST (H92R). In contrast, the PR strain exhibited the H92R but not the I256V and N321S point mutations. In some cases increased glutathione S-transferase (GST) activity has previously been linked to enhanced detoxification. The AR strain exhibited approximately 2.3-, 1.8-, and 2.2-fold increased GST activity against 1-chloro-2,4-dinitrobenzene (CDNB), 1,2-dichloro-4-nitrobenzene (DCNB), and 4-nitrobenzyl chloride (NBC), respectively. Among the five GST subclass genes (delta, omega, mu, zeta, and kappa), the relative expression of delta class GSTs in the AR strain were significantly higher than the PR and susceptible strain. These results suggest that the I256V and N321S mutations and the increased GST metabolism and GST delta overexpression might be related to acequinocyl resistance in T. urticae.

Abstract:The mechanisms underlying insecticide and acaricide resistance in insects and mites are often complex, including additive effects of target-site insensitivity, increased metabolism and transport. The extent to which target-site resistance mutations contribute to the resistance phenotype is, however, not well studied. Here, we used marker-assisted backcrossing to create 30 congenic lines carrying nine mutations (alone, or in combination in a few cases) associated with resistance to avermectins, pyrethroids, mite growth inhibitors and mitochondrial complex III inhibitors (QoI) in a polyphagous arthropod pest, the spider mite Tetranychus urticae. Toxicity tests revealed that mutations in the voltage-gated sodium channel, chitin synthase 1 and cytochrome b confer high levels of resistance and, when fixed in a population, these mutations alone can result in field failure of acaricide treatment. In contrast, although we confirmed the implication of mutations in glutamate-gated chloride channels in abamectin and milbemectin insensitivity, these mutations do not lead to the high resistance levels that are often reported in abamectin resistant strains of T. urticae. Overall, this study functionally validates reported target-site resistance mutations in T. urticae, by uncoupling them from additional mechanisms, allowing to finally investigate the strength of the conferred phenotype in vivo.

Resistance to pyridaben in Canadian greenhouse populations of two-spotted spider mites, Tetranychus urticae (Koch)

Abstract:Two-spotted spider mite (TSSM) Tetranychus urticae (Koch) is an important agricultural pest that causes considerable yield losses to over 150 field and greenhouse crops. Mitochondrial electron transport inhibitors (METI) acaricides are commonly used to control mite species in commercial Canadian greenhouses. Development of resistance to METIs in TSSM populations have been reported worldwide, but not until recently in Canada. The objectives of this study were to: 1) monitor the acaricide-susceptibility in greenhouse TSSM populations, and 2) investigate the resistance to pyridaben, a METI acaricide, in greenhouse resistant and pyridaben-selected (SRS) mite strains. The increased mortality to the pyridaben sub-lethal concentration (LC30) when SRS mites were exposed to piperonyl butoxide (PBO), a general cytochrome P450 monooxygenase inhibitor, and higher P450 activity compared to the greenhouse strain (RS) mites, indicated that P450s may be at least partially responsible for the resistance. The molecular mechanisms of target site insensitivity-mediated resistance in the pyridaben resistant strain of TSSM were investigated by comparing the DNA sequence of NADH dehydrogenase subunits TYKY and PSST, NADH-ubiquinone oxidoreductase chain 1 and 5 (ND1, ND5) and the NADH-ubiquinone oxidoreductase subunit 49 kDa from SRS to the reference strain (SS) and RS. Despite a number of nucleotide substitutions, none correlated with the pyridaben resistance. Understanding the underlying mechanisms of TSSM adaptation to acaricides is an essential part of resistance management strategy in any IPM program. The findings of this study will encourage growers to apply acaricides with different modes of action to reduce the rate at which acaricide resistance will occur in greenhouse TSSM populations.
Molecular diagnostics of insecticide resistance in Australian Tetranychus urticae Koch (Acari: Tetranychidae) quarantine intercepts

Abstract:Australian ports are continuously exposed to exotic and invasive mite and insect species associated with imported fresh produce. Much effort is focused on preventing the exotic species from entering Australia by product fumigation or destruction in a timely manner. However, quarantine intercepts of invasive species may contain unwanted resistance to pesticides or carry viruses that can undermine Australian agricultural sustainability. We examined pesticide resistance status in 1275 Tetranychus urticae samples from quarantine intercepts from the Department of Agriculture, Fisheries and Forestry originating from some 29 countries spread over 6 main continents (Africa [4], Asia [12], Europe [3], North America [2], Oceania [4] and South America [4]). We used a high‐throughput multiamplicon sequencing platform to screen major target‐site mutations that cause acaricide resistance in T. urticae. These included G119S, A201S, T280A, G328A and F331Y (Ace‐1) associated with mode of action (MoA) 1 insecticide (organophosphates and carbamates); M918, L925I, L1014F and F1538I (VGSC) associated with MoA 3 insecticide (pyrethroids); G314D (GluCl1) associated with MoA 6 insecticide (avermectins and milbemycins); and finally, H92R (PSST) associated with MoA 21 insecticide (fenpyroximate, pyridaben and tebufenpyrad). The results revealed that four known mutations (G119S, T280A, G328A and F331W) were abundant in the quarantine samples including those from Norfolk Island and New Zealand. The mutations L1024V, F1538I and L925V (VGSC) associated with pyrethroid resistance were widespread through many samples, but the super Kdr mutation M918 was not detected. Similarly, H92R (PSST) that causes resistance to fenpyroximate, pyridaben and tebufenpyrad was found in most of the T. urticae intercept samples. We conclude that resistance alleles entering Australia through quarantine intercepts are common, suggesting that unwanted or rare alleles could enter Australia via this route. However, exotic quarantine breaches carrying such alleles remain of most serious concern.

A novel mutation in mitochondrial cytochrome b conferring resistance to bifenazate in two-spotted spider mite Tetranychus urticae Koch (Acarina: Tetranychidae)

Abstract:Background: The two-spotted spider mite Tetranychus urticae causes significant damage to ornamental, cotton, sugarcane and horticultural crops in Australia. It has a long history of developing resistance to many acaricides including bifenazate. A mutation in the conserved cd1- and ef-helices of the Qo pocket of cytochrome b is recognized as the primary mechanism of bifenazate resistance. To investigate the resistance mechanisms against bifenazate in Australian two-spotted spider mite, we sequenced the complete mitochondrion genome of five mite strains including a susceptible and bifenazate-resistant strain. Results: We identified a novel mutation D252N in the G126S background at cytochrome b being the cause of bifenazate resistance in a bifenazate-resistant strain, Bram. We validated the role of this mutation combination by reciprocal crosses between a bifenazate resistant and susceptible strain. By doing these crosses we confirmed the pattern of inheritance was maternal. Additionally, mitochondrial heteroplasmy was not observed by single mite genotyping of the mutations in cytb in a known bifenazate-resistant strain Bram. The phylogenetic analysis with the complete mitochondrion genome sequences revealed that Australian two-spotted spider mite strains are closely related to the green form of T. urticae found in China. Conclusions: The novel mutation D252N found in the cytochrome b in the G126S background was revealed to be the main cause of bifenazate resistance in the Australian T. urticae strain Bram. © 2024 Society of Chemical Industry.

Abstract:The polyphagous pest Tetranychus urticae feeds on over 1100 plant species including highly valued economic crops such as hops (Humulus lupulus). In the key hop production region of the Pacific Northwest of the USA, T. urticae is one of the major arthropod pests. Over the years, T. urticae control has been dominated by the application of various acaricides. However, T. urticae quickly adapts to these acaricides by developing resistance. Here, we determined resistance ratios of T. urticae populations in hops to three acaricides: etoxazole, fenpyroximate, and spirodiclofen. The mechanisms underlying resistance to these and three other acaricides were investigated in 37 field-collected T. urticae populations using a comprehensive diagnostic approach. Our data showed that T. urticae populations exhibited complex adaptation patterns to acaricides. Resistance to abamectin, fenpyroximate, and spirodiclofen by enhanced target metabolic detoxification gene(s) was identified in 100%, 50%, and 20% of populations tested, respectively. Resistance to bifenthrin, bifenazate, and etoxazole by target site insensitivity was pervasive among tested populations. Our study provides new information in understanding the complexity of T. urticae adaptation to multiple acaricides, which will help in designing sustainable pest control strategies for T. urticae on hops and other economically valuable crops.

Geographical distribution and origin of acetylcholinesterase mutations conferring acaricide resistance in Tetranychus urticae populations from Turkey

Abstract:Two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), is a cosmopolitan pest species that can feed on more than 1000 host plant species. Historically, organophosphate (OP) and carbamate insecticides have been used to control this extremely polyphagous pest. However, its ability to develop acaricide resistance rapidly has led to failure in control. Mutations in acetylcholinesterase gene (ace), the target-site of OP and carbamate insecticides, have been reported to be one of the major mechanisms underlying this developing resistance. In this study, mutations previously associated with resistance (G119S, A201S, T280A, G328A, F331W/Y) in ace have been screened in 37 T. urticae populations collected across Turkey. All mutations were found in various populations, except G119S. Almost all populations had F331W/Y mutation (being fixed in 32 populations), whereas only two populations harboured A201S mutation, but not fixed. On the other hand, more than half of the populations contained T280A and G328A mutations. In addition, the presence of same haplotypes in populations originating from distinct geographic locations and a wide variety of ace haplotypes might indicate multiple origins of F331W and F331Y mutations; however, this needs further investigation. The results of area-wide screening showed that ace mutations are widely distributed among T. urticae populations. Therefore, the use of this group of insecticides should be limited or only rotational use might be regarded as a resistance management tool due to its different mode of action from other main acaricide groups in T. urticae control across Turkey.
The cytochrome P450 subfamilies CYP392A and CYP392D are key players in acaricide metabolism in Tetranychusurticae

Abstract:The spider mite Tetranychus urticae is a major agricultural pest with a global distribution, extremely diverse host range and a remarkable ability to develop resistance to a wide variety of acaricides. P450 mono-oxygenases have been frequently associated with resistance development in this species. In particular enzymes of the CYP392A-subfamily were shown to metabolize a number of key acaricides, including abamectin, amitraz, fenpyroximate and the active metabolite of pyflubumide. However, transcriptomic studies comparing highly resistant and susceptible populations have often revealed high expression of members of the CYP392D-subfamily, but these have been only poorly studied. Here, we conducted a meta-analysis of gene expression data of 20 populations and identified two key enzymes of this family, CYP392D2 and CYP392D8, whose expression is associated with resistance. We subsequently functionally expressed these enzymes, together with CYP392A11 and CYP392A16 as known metabolizers, and compared their potential to accept a wide diversity of acaricides as substrate. This study overall confirms previous discovered substrates for CYP392A11 and CYP392A16, but also reveals unreported metabolic activity towards new acaricides. These include carbaryl, chlorpyrifos and etoxazole for CYP392A16 and carbaryl, chlorpyrifos and NNI-0711-NH pyflubumide for CYP392A11. For the newly studied CYP392D-family, we show that CYP392D2 metabolizes pyridaben, fenpyroximate, etoxazole and chlorpyrifos, while CYP392D8 metabolizes carbaryl, fenazaquin and tebufenpyrad. Last, we observed that both CYP392A- and CYP392D-subfamily enzymes activate chlorpyrifos to its corresponding oxon. Our study indicates that there is both overlap and specificity in the activity of A- and D-subfamily enzymes against acaricides and model substrates. With the recent advent of highly efficient CRISPR/Cas9 gene editing protocols in T. urticae, the way is now paved to conduct further genetic experiments revealing and quantifying the role of these enzymes in the resistance phenotype in field populations.
High-Resolution Genetic Mapping Combined with Transcriptome Profiling Reveals That Both Target-Site Resistance and Increased Detoxification Confer Resistance to the Pyrethroid Bifenthrin in the Spider Mite Tetranychus urticae

Abstract:Pyrethroids are widely applied insecticides in agriculture, but their frequent use has provoked many cases of resistance, in which mutations in the voltage-gated sodium channel (VGSC), the pyrethroid target-site, were shown to play a major role. However, for the spider mite Tetranychus urticae, it has also been shown that increased detoxification contributes to resistance against the pyrethroid bifenthrin. Here, we performed QTL-mapping to identify the genomic loci underlying bifenthrin resistance in T. urticae. Two loci on chromosome 1 were identified, with the VGSC gene being located near the second QTL and harboring the well-known L1024V mutation. In addition, the presence of an L925M mutation in the VGSC of a highly bifenthrin-resistant strain and its loss in its derived, susceptible, inbred line indicated the importance of target-site mutations in bifenthrin resistance. Further, RNAseq experiments revealed that genes encoding detoxification enzymes, including carboxyl/choline esterases (CCEs), cytochrome P450 monooxygenases and UDP-glycosyl transferases (UGTs), were overexpressed in resistant strains. Toxicity bioassays with bifenthrin (ester pyrethroid) and etofenprox (non-ester pyrethroid) also indicated a possible role for CCEs in bifenthrin resistance. A selection of CCEs and UGTs were therefore functionally expressed, and CCEinc18 was shown to metabolize bifenthrin, while teturUGT10 could glycosylate bifenthrin-alcohol. To conclude, our findings suggest that both target-site and metabolic mechanisms underlie bifenthrin resistance in T. urticae, and these might synergize high levels of resistance.

Abstract:Two spotted spider mite Tetranychus urticae Koch is an economically serious pest posing threat to major vegetable crops. Roving survey in and around Coimbatore region revealed that farmers do not target mites with acaricides instead they use higher dose of insecticides at frequent intervals which results in development of resistance. The bioassay results revealed that fenpropathrin (2.07 to 6.86-folds) and fenazaquin (2.74 to 7.13-folds) exhibit higher susceptibility, whereas diafenthiuron (5.35 to 12.25-folds) revealed a low to moderate level of resistance. The propargite (43.80 to 60.63-folds) and chlorfenapyr (61.01 to 75.10-folds) exhibited high resistance, followed by spiromesifen (222.28 to 300.26-folds) and buprofezin (382.60 to 417.87-folds), with extremely high level of resistance. The higher specific activity of GST (4.54-folds), MFO (10.06-folds) and CarE (15.06-folds) in Puthupalayam population suggested the role of biochemical resistance. A significant positive correlation was observed between diafenthiuron and CarE activity (r = 0.981*), fenpropathrin and MFO activity (r = 0.964*).
