Abstract:Due to the increasing cases of bone damage and bone graft demand, bone-repair technology has great social and economic benefits and the manufacturing of artificial bone implants has become a focus in the domain of regenerative therapy. Considering that the traditional manufacturing process cannot effectively control the overall size of the scaffold, the diameter and shape of micropores, and the interoperability of micropores, 3D printing technology has emerged as a focal point of research within the realm of bone tissue engineering. However, the printing accuracy of extrusion-based biological 3D printing techniques is low. In this research, we utilized three-dimensional printing technology to develop high-precision magnesium-containing silicate (CSi-Mg) scaffolds. The precision of this innovative method was scrutinized and the influence of pore size on scaffold strength was systematically analyzed. Furthermore, the influence of the pore architecture on the sidewalls of these 3D-printed scaffolds was evaluated in terms of mechanical properties. The CSi-Mg scaffold, post a 3-week immersion in a simulated body of fluid, demonstrated a high modulus of elasticity (exceeding 404 MPa) and significant compressive strength (beyond 47 MPa). Furthermore, it exhibited commendable bioactivity and biodegradability. These results suggest that the high-precision 3D-printed CSi-Mg scaffolds hold great promise for addressing challenging bone defect cases.
3D Printing of Complex GelMA-based Scaffolds with Nanoclay

Abstract:Photo-crosslinkable gelatin methacrylate (GelMA) has become an attractive ink in 3D printing due to its excellent biological performance. However, limited by low viscosity and long cross-linking time, it is still a challenge to directly print GelMA by extrusion-based 3D printing. Here, to balance the printability and biocompatibility, biomaterial ink composed of GelMA and nanoclay was specially designed. Using this ink, complex scaffolds with high shape fidelity can be easily printed based on the thixotropic property of nanoclay. In this study, we tried to answer some basic printing-required questions of this ink, including the printability window, general properties (porosity, mechanical strength, et al), and biocompatibility. We found that the GelMA/Nanoclay ink enabled printing complex 3D scaffolds, such as a bionic ear and a branched vessel. Furthermore, the addition of nanoclay improved the porosity, increased the mechanical strength, reduced the degradation ratio, and maintained a good biocompatibility of the printed scaffolds. Therefore, this method offers an easy way to print complex scaffolds with good shape fidelity and biological performance, and it might open up new potential applications for the customized therapy of tissue defects.
3D Printing of Biomimetic Multi-Layered GelMA/nHA Scaffold for Osteochondral Defect Repair

Abstract:Currently, osteochondral defects frequently cause limited motion and impaired function of the joint, leading to serious healthcare problems, and it is still very challenging to realize the simultaneous regeneration of subchondral bone with cartilage. In the current study, we designed a tri-layered scaffold and fabricated it using the extrusion-based multi-nozzle 3D printing technology. The bioinks used for 3D printing included a 15% methacrylated gelatin (GelMA) hydrogel for cartilage on top layer, a combination of 20% GelMA and 3% nanohydroxyapatite (nHA) (20/3% GelMA/nHA) hydrogel for interfacial layer, and a 30/3% GelMA/nHA hydrogel for subchondral bone at bottom layer. The water absorption capacity, biodegradation, and mechanical properties of hydrogels and scaffolds were characterized, and in vitro assay with bone marrow mesenchymal stem cells (BMSCs) was performed to indicate the biocompatibility of scaffolds. Based on the results of in vivo repair of rabbit osteochondral defect, the neo-tissues in defects integrated better with the surrounding tissues, the joint surface of the defects was smoother, and more cartilage-specific extracellular matrix and collagen type II were observed using the tri-layered scaffolds. This study not only provides a potential manufacturing method for multi-layered scaffolds, but also is helpful for understanding the regeneration mechanism of cartilage-subchondral bone.
In vitro static and dynamic cell culture study of novel bone scaffolds based on 3D-printed PLA and cell-laden alginate hydrogel

Abstract:The aim of this paper was to design and fabricate a novel composite scaffold based on the combination of 3D-printed polylactic acid-based triply periodic minimal surfaces (TPMSs) and cell-laden alginate hydrogel. This novel scaffold improves the low mechanical properties of alginate hydrogel and can also provide a scaffold with a suitable pore size, which can be used in bone regeneration applications. In this regard, an implicit function was used to generate some gyroid TPMS scaffolds. Then the fused deposition modeling process was employed to print the scaffolds. Moreover, the micro computed tomography technique was employed to assess the microstructure of 3D-printed TPMS scaffolds and obtain the real geometries of printed scaffolds. The mechanical properties of composite scaffolds were investigated under compression tests experimentally. It was shown that different mechanical behaviors could be obtained for different implicit function parameters. In this research, to assess the mechanical behavior of printed scaffolds in terms of the strain-stress curves on, two approaches were presented: equivalent volume and finite element-based volume. Results of strain-stress curves showed that the finite-element based approach predicts a higher level of stress. Moreover, the biological response of composite scaffolds in terms of cell viability, cell proliferation, and cell attachment was investigated. In this vein, a dynamic cell culture system was designed and fabricated, which improves mass transport through the composite scaffolds and applies mechanical loading to the cells, which helps cell proliferation. Moreover, the results of the novel composite scaffolds were compared to those without alginate, and it was shown that the composite scaffold could create more viability and cell proliferation in both dynamic and static cultures. Also, it was shown that scaffolds in dynamic cell culture have a better biological response than in static culture. In addition, scanning electron microscopy was employed to study the cell adhesion on the composite scaffolds, which showed excellent attachment between the scaffolds and cells.

Abstract:Tissue engineering scaffolds are three-dimensional structures that provide an appropriate environment for cellular attachment, proliferation, and differentiation. Depending on their specific purpose, these scaffolds must possess distinct features, including appropriate mechanical properties, porosity, desired degradation rate, and cell compatibility. This investigation aimed to fabricate a new nanocomposite scaffold using a 3D printing technique composed of poly(ε-caprolactone) (PCL)/Gelatin (GEL)/ordered mesoporous calcium-magnesium silicate (om-CMS) particles. Different weight ratios of om-CMS were added and optimized, and a series of scaffolds were constructed for comparison purposes, including PCL 50%/Gel 50%, PCL 50%/Gel 45%/om-CMS%5, and PCL 50%/Gel 40%/om-CMS%10. The optimized weight ratio of om-CMS was 10% without leaving behind negative effects on the filaments' structure. The scaffolds' physical and chemical properties were assessed using various techniques, and their degradation rate, bioactivity potential, cell viability, attachment, and ALP activity were evaluated in vitro. The results demonstrated that the PCL 50%/Gel 40%/om-CMS10% scaffold had promising potential for further studies in bone tissue regeneration.

Abstract:A microfluidics system is used to create gelatin methacryloyl (GelMA) nanofibrous microspheres (GNM), which are employed as bioink for 3D printing hierarchically porous GNM scaffolds (HP‐GNMS). Bone marrow stromal cells (BMSCs) are seeded onto these scaffolds and implanted in a rat cranial defect model, promoting cell infiltration, and guiding osteogenic differentiation and extracellular matrix deposition. Current limitations in 3D printing pose significant challenges for the fabrication of hierarchical 3D scaffolds with nanofibrous structures that simulate the natural bone extracellular matrix (ECM) for enhanced bone regeneration. This study presents an innovative approach to 3D printing customized hierarchical porous scaffolds with nanofiber structures using biodegradable nanofibrous microspheres as the bio‐ink. In vitro investigations demonstrate that the hierarchical porous architecture substantially enhances cell infiltration and proliferation rates, while the nanofiber topology provides physical cues to guide osteogenic differentiation and ECM deposition. When serving as a cell carrier, the 3D‐printed nanofibrous scaffold promotes bone tissue regeneration and integration in vivo. Additionally, the facile and versatile chemical modification facilitates the precise tailoring of the scaffold's functionality. Using nanofibrous microspheres with highly biomimetic and versatile modification properties as the foundational constituent in this universal 3D printing methodology enables comprehensive manipulation of scaffolding biological properties, spanning from macroscopic external morphology to molecular‐scale biochemical kinetics, thereby addressing a diverse spectrum of clinical requisites.

Abstract:3D bioprinting uses biomaterials combined with cells to develop living constructs. This study explores the optimization of natural polymers, including silk fibroin, gelatin, and alginate, as bioink for 3D bioprinting in bone tissue engineering. The physicochemical properties of the bioink were thoroughly examined, revealing high water uptake and reduced degradation rate due to the addition of silk fibroin. The compressive modulus increased with higher alginate concentration. Rheological analysis confirmed shear-thinning properties and viscoelasticity of the ink. Through meticulous parameter optimization, the ink achieved the highest print accuracy with 4 % w/v alginate content. The printed scaffolds exhibited both macro and micro porosity, making them suitable for bone tissue regeneration. Furthermore, the scaffolds remained stable in culture medium for 36 days. The optimal composition for the hydrogel was determined to be a blend of 5 % fibroin, 7 % gelatin, and 4 % alginate in equal ratios. The bioink demonstrated excellent biocompatibility and, when supplemented with alendronate, enhanced alkaline phosphatase activity in MG-63 osteoblast-like cells. This finding indicates the commitment of cells toward the osteoblastic phenotype. Overall, this study successfully optimized the bioink and bioprinting process for bone tissue engineering applications, highlighting its promising potential for future advancements in the field.

Abstract:Three-dimensional (3D) printing allows precise manufacturing of bone scaffolds for patient-specific applications and is one of the most recently developed and implemented technologies. In this study, bilayer and multimaterial alginate dialdehyde-gelatin (ADA-GEL) scaffolds incorporating polydopamine (PDA)/SiO 2 -CaO nanoparticle complexes were 3D printed using a pneumatic extrusion-based 3D printing technology and further modified on the surface with bovine serum albumin (BSA) for application in bone regeneration. The morphology, chemistry, and in vitro bioactivity of PDA/SiO 2 -CaO nanoparticle complexes were characterized ( n = 3) and compared with those of mesoporous SiO 2 -CaO nanoparticles. Successful deposition of the PDA layer on the surface of the SiO 2 -CaO nanoparticles allowed better dispersion in a liquid medium and showed enhanced bioactivity. Rheological studies ( n = 3) of ADA-GEL inks consisting of PDA/SiO 2 -CaO nanoparticle complexes showed results that may indicate better injectability and printability behavior compared to ADA-GEL inks incorporating unmodified nanoparticles. Microscopic observations of 3D printed scaffolds revealed that PDA/SiO 2 -CaO nanoparticle complexes introduced additional topography onto the surface of 3D printed scaffolds. Additionally, the modified scaffolds were mechanically stable and elastic, closely mimicking the properties of natural bone. Furthermore, protein-coated bilayer scaffolds displayed controllable absorption and biodegradation, enhanced bioactivity, MC3T3-E1 cell adhesion, proliferation, and higher alkaline phosphatase (ALP) activity ( n = 3) compared to unmodified scaffolds. Consequently, the present results confirm that ADA-GEL scaffolds incorporating PDA/SiO 2 -CaO nanoparticle complexes modified with BSA offer a promising approach for bone regeneration applications.

Abstract:"Lean" (continuous improvement) organizations make use of daily management systems (DMS) that are designed so that problems can be quickly identified, front-line staff are empowered to fix the problems that they can, and problems that the front-line staff cannot fix are escalated and countermeasures created quickly. Key components of a DMS include leadership standard work, visual controls, and a daily accountability process, as well as discipline involving each of these three components. The author's organization recently had the opportunity to open a new, nonreplacement hospital, allowing the incorporation of continuous improvement principles into the hospital's design and operations. One high-priority task was the creation of a DMS, which was structured as a tiered "huddle" system. All of the front-line clinical areas, as well as all clinical and nonclinical ancillary support areas, conduct morning huddles. Problems identified at these huddles and needing escalation are then brought to a patient flow huddle and an integrated huddle. All of these huddles occur daily and have a standard format with three clearly defined components: metrics-goal review, daily readiness assessment, and problem accountability reporting. The huddles also provide a daily opportunity to see and converse with the people with whom one needs to discuss certain issues. The process of bringing people together for these huddles can contribute significantly to team formation, coordination of efforts, and development of a culture of trust.
Fabrication and Characterization of 3D Nanostructured Polycaprolactone-Gelatin/Nanohydroxyapatite-Nanoclay Scaffolds for Bone Tissue Regeneration

Abstract:Numerous clinical bone disorders, such as infections and bone loss from cancer or trauma, increase the need for bone regeneration. Due to the difficulty of self-repairing large bone defects, bone tissue engineering has gained popularity. In this study, polycaprolactone(PCL)-gelatin(GEL) scaffolds with varying concentrations of nanohydroxyapatite (NHA) and nanoclay (NC) particles were fabricated using 3D printing technology, and their physiochemical and biological properties were assessed. PCL has excellent mechanical properties, but its hydrophobicity and long-term degradation limit its utility in scaffold fabrication. Thus, GEL, NHA and NC have been used to improve the overall performance of the polymer such as hydrophilicity, strength, adhesiveness, biocompatibility, biodegradability, and osteoconductivity. The morphological analysis revealed 3D printed structures with rectangular interconnected pores and well-distributed nanoparticles. The highest porosity belonged to PCL-GEL/NHA-NC (30/70) at 69.49%, which may directly contributed to the increase in the compressive modulus and degradation rate. The wettability, compressive strength, water uptake rate, biodegradability, and bioactivity of PCL-GEL scaffolds improved significantly as the NC concentration increased. The behavior of the seeded MG-63 cells on the 3D printed nanocomposite scaffolds was evaluated using the MTT assay, DAPI staining, and SEM micro images. It was discovered that the inclusion of NHA and NC nanoparticles can promote cell proliferation, viability, and adherence. Through the obtained in vitro results, the fabricated 3D printed PCL-GEL/NHA scaffold with higher NC concentration can be regarded as a promising scaffold for expediting the repair of bone defects.

Abstract:Three-dimensional grafts/scaffolds with hierarchically biomimetic features from nano to macro scale and high porosity are required for bone tissue engineering. In this study, biomimetic organic/inorganic composite scaffolds with high porosity (78.7 ± 3.2%) and features from nano (nano apatite coatings) to macro (macro pores and hollow channels) scale were fabricated based on highly concentrated alginate/GelMA bioinks via co-axial 3D printing and in situ mineralization under mild conditions. Nano apatites were coated on both inner and outer surfaces of the hollow fiber scaffolds, homogeneously. Proteins were directly loaded in the bioinks achieving sustained release from the scaffolds over 28 days. The in vitro cell experiments showed that the scaffolds with good biocompatibility could support cells adhesion and proliferation. The nano apatite coatings presented remarkable osteogenic capability. The in vivo study indicated that the hollow fiber scaffolds with biomimetic nano apatite coatings showed the capability to enhance bone formation after 12 weeks of implantation. In conclusion, the prepared biomimetic organic/inorganic scaffolds with homogeneous nano apatite coatings and hollow channels structures might be potential candidates for bone tissue engineering.

Abstract:New gelatin methacryloyl (GelMA)-strontium-doped nanosize hydroxyapatite (SrHA) composite hydrogel scaffolds were developed using UV photo-crosslinking and 3D printing for bone tissue regeneration, with the controlled delivery capacity of strontium (Sr). While Sr is an effective anti-osteoporotic agent with both anti-resorptive and anabolic properties, it has several important side effects when systemic administration is applied. Multi-layer composite scaffolds for bone tissue regeneration were developed based on the digital light processing (DLP) 3D printing technique through the photopolymerization of GelMA. The chemical, morphological, and biocompatibility properties of these scaffolds were investigated. The composite gels were shown to be suitable for 3D printing. In vitro cell culture showed that osteoblasts can adhere and proliferate on the surface of the hydrogel, indicating that the GelMA-SrHA hydrogel has good cell viability and biocompatibility. The GelMA-SrHA composites are promising 3D-printed scaffolds for bone repair.
Biomimetic surface modification of Three-dimensional printed Polylactic acid scaffolds with custom mechanical properties for bone reconstruction

Abstract:Journal of Biomaterials Applications, Ahead of Print. 3D printing has recently emerged as an innovative fabrication method to construct critical-sized and patient-specific bone scaffolds. The ability to control the bulk geometry of scaffolds in both macro and micro-scales distinguishes this technology from other fabrication methods. In this study, bone tissue-specific scaffolds with different pore geometries were printed from polylactic acid (PLA) filaments at three given infill densities ranging from 20 to 30%. A hybrid hydrogel made of synthetic biphasic calcium phosphate (BCP) and collagen was applied to coat 3D printed well-structured triangular samples with 30% infill density. The coating process changed the surface texture, increased the average strand diameter and average pore size, and decreased the open porosity of samples, all of which increased the mechanical strength of biomimetic-coated scaffolds. According to matrix mineralization staining and osteo-related gene expression, the coating of scaffolds significantly facilitates metabolic activity and osteogenic differentiation of dental pulp-derived mesenchymal stem cells (DPSCs). Taken together, these results indicated that the biomimetic coating is a highly promising approach that could be taken into consideration in the design of a porous scaffold for bone tissue engineering.

Mussel-inspired polydopamine decorated alginate dialdehyde-gelatin 3D printed scaffolds for bone tissue engineering application

Abstract:This study utilized extrusion-based 3D printing technology to fabricate calcium-cross-linked alginate dialdehyde-gelatin scaffolds for bone regeneration. The surface of polymeric constructs was modified with mussel-derived polydopamine (PDA) in order to induce biomineralization, increase hydrophilicity, and enhance cell interactions. Microscopic observations revealed that the PDA layer homogeneously coated the surface and did not appear to induce any distinct change in the microstructure of the scaffolds. The PDA-functionalized scaffolds were more mechanically stable (compression strength of 0.69 ± 0.02 MPa) and hydrophilic (contact angle of 26) than non-modified scaffolds. PDA-decorated ADA-GEL scaffolds demonstrated greater durability. As result of the 18-days immersion in simulated body fluid solution, the PDA-coated scaffolds showed satisfactory biomineralization. Based on theoretical energy analysis, it was shown that the scaffolds coated with PDA interact spontaneously with osteocalcin and osteomodulin (binding energy values of −35.95 kJ mol −1 and −46.39 kJ mol −1 , respectively), resulting in the formation of a protein layer on the surface, suggesting applications in bone repair. PDA-coated ADA-GEL scaffolds are capable of supporting osteosarcoma MG-63 cell adhesion, viability (140.18% after 7 days), and proliferation. In addition to increased alkaline phosphatase secretion, osteoimage intensity also increased, indicating that the scaffolds could potentially induce bone regeneration. As a consequence, the present results confirm that 3D printed PDA-coated scaffolds constitute an intriguing novel approach for bone tissue engineering.

Fabrication and finite element simulation of antibacterial 3D printed Poly L-lactic acid scaffolds coated with alginate/magnesium oxide for bone tissue regeneration

Abstract:This study proposes 3D-printed Poly L-lactic acid (PLA) scaffolds coated with alginate/MgO, and includes three different cellular topologies. Three unique scaffold models were considered: Perovskite type 1 (P1), Perovskite type 2 (P2), and IWP. Each scaffold was coated with alginate/MgO at the concentrations of 0 wt%, 5 wt%, 10 wt%, 15 wt%, and 20 wt%. For morphological and phase study, the microstructure of fabricated scaffolds was characterized using a Field Emission Scanning Electron Microscope (FESEM), energy dispersive spectroscopy (EDS), and X-ray diffraction (XRD) analysis. Besides, the biological characteristics of scaffolds, such as biocompatibility, antibacterial activity, and cell survival were studied after 21 days of soaking in the simulated body fluid (SBF). The results of biological studies indicate that the apatite layer covered the majority of composite scaffold's surface and sealed the pores' surface. The material properties of Alginate/MgO RVEs were evaluated under PBC, and it described that the elastic modulus enhanced from 100 (pure Alginate) to 130 MPA by adding 20 wt% MgO nanoparticles. The presented findings were compared to the results obtained by the experimental procedure and revealed satisfactory agreement. RVE-achieved material properties were used in the additional studies on the scaffolds to find the best candidate due to the material properties and architectures. Furthermore, experiment and finite element simulation were used to evaluate the mechanical properties of scaffolds under the compressive deformation. According to the results, the compressive strength of structures follows the order σPerovskite type 1>σPerovskite type 2 >σIWP. The results indicate that increasing MgO content from 0 wt% to 20 wt% enhances each structure's compressive strength and elastic modulus. In conclusion, based on the biological findings and simulation results, PLA scaffold with Perovskite type 1 (P1) architecture coated with Alginate/ 20 wt% MgO had the best response which is the final research candidate.

Abstract:3D-printed biofunctional scaffolds have promising applications in bone tissue regeneration. However, the development of bioinks with rapid internal vascularization capabilities and relatively sustained osteoinductive bioactivity is the primary technical challenge. In this work, we added rat platelet-rich plasma (PRP) to a methacrylated gelatin (GelMA)/methacrylated alginate (AlgMA) system, which was further modified by a nanoclay, laponite (Lap). We found that Lap was effective in retarding the release of multiple growth factors from the PRP-GelMA/AlgMA (PRP-GA) hydrogel and sustained the release for up to 2 weeks. Our in vitro studies showed that the PRP-GA@Lap hydrogel significantly promoted the proliferation, migration, and osteogenic differentiation of rat bone marrow mesenchymal stem cells, accelerated the formation of endothelial cell vascular patterns, and promoted macrophage M2 polarization. Furthermore, we printed hydrogel bioink with polycaprolactone (PCL) layer-by-layer to form active bone repair scaffolds and implanted them in subcutaneous and femoral condyle defects in rats. In vivo experiments showed that the PRP-GA@Lap/PCL scaffolds significantly promoted vascular inward growth and enhanced bone regeneration at the defect site. This work suggests that PRP-based 3D-bioprinted vascularized scaffolds will have great potential for clinical translation in the treatment of bone defects.

Synergetic effect of bioglass and nano montmorillonite on 3D printed nanocomposite of polycaprolactone/gelatin in the fabrication of bone scaffolds

Abstract:Nowadays, bone injuries and disorders have increased all over the world and can reduce the quality of human life. Bone tissue engineering repair approaches require new biomaterials and methods to construct scaffolds with the required structural properties as well as improved performance. As potential therapeutic strategies in bone tissue engineering, 3D printed scaffolds have been developed. Polycaprolactone/Ceramic composites have attracted considerable attention due to their cytocompatibility, biodegradability, and physical properties. In this study, a 3D printing process was used to create polycaprolactone (PCL)-Gelatin (GEL) scaffolds containing varying concentrations of Bioglass (BG) and Nano Montmorillonite (MMT). This mixture was then loaded into a 3D printer, and the scaffolds were printed layer by layer. After constructing the scaffolds, they were then examined for their physical, chemical, and biological characteristics. Surface appearance was analyzed with a scanning electron microscope (SEM), which revealed that NC increased the diameter of pores from 465 to 480 μm. The elements in the scaffolds were evaluated by EDX analysis, and a uniform dispersion of nano montmorillonite particles was observed. The compressive strength reached 76.43 MPa for PCL/G/35 %MMT/15 %BG scaffold. Also, the rate of water absorption, biodegradability and bioactivity of PCL-GEL scaffolds increased significantly in the presence of NC. According to the MTT cell test results, adding BG and NC increased cell proliferation, adhesion and cell viability to 127.7 %. These findings indicated that the 3D printed PCL/G/35 %MMT/15 %BG scaffold has promising strategies for bone repair applications. Also, polynomial curve fitting shows that scaffold degradability after soaking in PBS can be predicted using the initial weight and soaking time. Adding more variables and data could improve prediction accuracy, reducing the need for experiments and conserving resources.

Abstract:Bone defects caused by diseases and trauma are considered serious clinical challenges. Autologous and allogeneic transplantations are the most widely used methods to mitigate bone defects. However, transplantation poses risks such as secondary trauma, immune rejection, and disease transmission to patients. Preparing a biologically active bone tissue engineering scaffold as a bone substitute can overcome this problem. In the current study, a PLGA/gelatin (Gel) short fiber-reinforced composite three-dimensional (3D) scaffold was fabricated by electrospinning for bone tissue defect repair. A hybrid scaffold adding inorganic materials hydrotalcite (CaAl-LDH) and osteogenic factors deferoxamine (DFO) based on PLGA and Gel composite filaments was prepared. The structure, swelling, drug release, and compressive resilience performance of the 3D scaffolds in a wet state were characterized and the osteogenic effect of the crosslinked scaffold (C-DLPG) was also investigated. The scaffold has shown the optimum physicochemical attributes which still has 380 kPa stress after a 60% compression cycle and sustainedly released the drug for about twenty days. Moreover, a promisingIn vivoosteogenic performance was noted with better tissue organization. At 8 weeks after implantation, the C-DLPG scaffold could fill the bone defect site, and the new bone area reached 19 mm2. The 3D microfiber scaffold, in this study, is expected to be a promising candidate for the treatment of bone defects in the future.

Abstract:The repair of large load-bearing bone defects requires superior mechanical strength, a feat that a single hydrogel scaffold cannot achieve. The objective is to seamlessly integrate optimal microarchitecture, mechanical robustness, vascularisation, and osteoinductive biological responses to effectively address these critical load-bearing bone defects. To confront this challenge, three-dimensional (3D) printing technology was employed to prepare a polycaprolactone (PCL)-based integrated scaffold. Within the voids of 3D printed PCL scaffold, a methacrylate gelatin (GelMA)/methacrylated silk fibroin (SFMA) composite hydrogel incorporated with parathyroid hormone (PTH) peptide-loaded mesoporous silica nanoparticles (PTH@MSNs) was embedded, evolving into a porous PTH@MSNs/GelMA/SFMA/PCL (PM@GS/PCL) scaffold. The feasibility of fabricating this functional scaffold with a customised hierarchical structure was confirmed through meticulous chemical and physical characterisation. Compression testing unveiled an impressive strength of 17.81 ± 0.83 MPa for the composite scaffold. Additionally, in vitro angiogenesis potential of PM@GS/PCL scaffold was evaluated through Transwell and tube formation assays using human umbilical vein endothelium, revealing the superior cell migration and tube network formation. The alizarin red and alkaline phosphatase staining assays using bone marrow-derived mesenchymal stem cells clearly illustrated robust osteogenic differentiation properties within this scaffold. Furthermore, the bone repair potential of the scaffold was investigated on a rat femoral defect model using micro-computed tomography and histological examination, demonstrating enhanced osteogenic and angiogenic performance. This study presents a promising strategy for fabricating a microenvironment-matched composite scaffold for bone tissue engineering, providing a potential solution for effective bone defect repair.

Methods: In this study, sodium alginate (SA)/gelatin (Gel) hydrogel scaffolds doped with different contents of nano-attapulgite were fabricated via 3D printing. The surface microstructure, hydrophilicity and mechanical properties were fully evaluated. Furthermore, mouse bone marrow-derived mesenchymal stem cells (BMSCs) were cultured with the composite hydrogels in vitro, and proliferation and osteoblastic differentiation were assessed. A rabbit tibia plateau defect model was used to evaluate the osteogenic potential of the composite hydrogel in vivo. Results: When increasing nano-ATP content, the Gel/SA/nano-ATP composite hydrogels showed better mechanical property and printability. Moreover, Gel/SA/nano-ATP composite hydrogels showed excellent bioactivity, and a significant mineralization effect was observed on the surface after being incubated in simulated body fluid (SBF) for 14 days. The Gel/SA/nano-ATP composite hydrogel also showed good biocompatibility and promoted the osteogenesis of BMSCs. Finally, histological analysis demonstrates that the Gel/SA/nano-ATP composite hydrogels could effectively enhance bone regeneration in vivo. Conclusion: These properties render the Gel/SA/nano-ATP composite hydrogel scaffolds an ideal bone tissue engineering material for the repair of bone defects.
