Abstract:Listeria monocytogenes ( L. monocytogenes ) is a ubiquitous foodborne pathogens responsible for listeriosis, which is associated with high morbidity and mortality. Development of simple and fast methods for L. monocytogenes detection is of great needed. In this study, we present a simple method for L. monocytogenes detection combined aptamers decorated magnetic beads for selective capture and gold nanoparticles (AuNPs) mediated signal amplication. A NH 2 labeled aptamer specific to L. monocytogenes was decorated on magnetic beads for selective capture and isolation of L. monocytogenes from food sample. AuNPs was functionalized with sulfhydryl modified primers specific to hly gene of L. monocytogenes in acidic environments. The AuNP@primers could catalyze TMB-H 2 O 2 to TMB 2+ and generate a significant blue color, this reaction was found to be enhanced by DNA. AuNP@primers was used as a signal indicator for the presence of L. monocytogenes DNA by enhancing the catalytic reaction of TMB-H 2 O 2 . The colour change was observed by naked eye and the optical density of TMB 2 + was analyzed to quantify the result. The developed assay was highly specific and the limit of detection was 10 1 CFU/mL of L. monocytogenes . For the detection of L. monocytogenes in spiked food samples, the recoveries were ranging from 73.63 to 84.21% with the coefficient of variation less than 2.41% in meat, and ranged from 94.13 to 100.71% with the coefficient of variation less than 2.82% in milk, respectively. Therefore, the developed colorimetric assay is suitable for food sample testing, the detection takes less than 1.5 h, which enables on-site screening. This study offers a promising approach for rapid, sensitive and accurate detection of L. monocytogenes in food.

Abstract:Pathogenic bacteria contamination poses a major threat to human health. The detection of low-abundance bacteria in complex samples has always been a knotty problem, and high-sensitivity bacterial detection remains challenging. In this work, a novel magnetic platform with high enrichment efficiency for L. monocytogenes was developed. The magnetic platform was designed by branched polyglutamic acid-mediated indirect coupling of cefepime on magnetic nanoparticles (Cefe-PGA-MNPs), and the specific enrichment of low-abundance L. monocytogenes in real samples was achieved by an external magnet, with a capture efficiency over 90%. A controllable and highly active platinum-palladium nanozyme was synthesized and further introduced in the magnetic nanoplatform for the construction of enzymatic colorimetric biosensor. The total detection time for L. monocytogenes was within 100 min. The colorimetric signals generated by labelled nanozyme were corresponding to different concentrations of L. monocytogenes , with a limit of detection (LOD) of 3.1 × 10 1 CFU/mL, and high reliability and accuracy (with a recovery rate ranging from 96.5% to 116.4%) in the test of real samples. The concept of the developed method is applicable to various fields of biosensing that rely on magnetic separation platforms.

Abstract:Listeria monocytogenes (L. monocytogenes) is one of the most common food-borne pathogens. The authors describe a sensitive and reliable multicolorimetric assay for L. monocytogenes using a sensing system based on TMB2+ etching of gold nanorods. Apt-MNP was used as the capture probe, and IgY-BSA-MnO2 NPs was chosen as an oxidase-like nano-artificial enzyme to oxidize TMB to generate TMB2+. Under the optimized conditions, the longitudinal shift of localized surface plasmon resonances had a linear correlation with the L. monocytogenes concentration in the range between 10 to 10(6) cfu mL(-1). Meanwhile, the sensing system can generate vivid color responses as colorful as a rainbow, and the limit of detection is as low as 10 cfu mL(-1) at a glance. Recoveries ranging from 97.4 to 101.3% are found when analyzing spiked food samples without pre-enrichment. In our perception, it shows promise in rapid instrumental and on-site visual detection of L. monocytogenes. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:Abstract A host–guest colorimetric strategy is described for the detection of Listeria monocytogenes (L. monocytogenes). The optical probes were self-assembled based on the supramolecular interactions between the carbonyl groups of cucurbit[7]uril portals and gold nanoparticles (CB[7]-AuNPs). Aptamer and urease modified magnetic nanoparticles were used to specifically recognize and binding to L. monocytogenes, simultaneously hydrolyzing urea to produce ammonium ion (NH4+) that can reverse CB[7] induced AuNPs aggregation. In the presence of L. monocytogenes, the above-mentioned magnetic conjugates preferentially bind to the bacterial surface, which results in blocking the catalytic active sites, thus inhibiting the production of ammonium ions. The normalized absorbance ratio of A700 nm/A525 nm was proportional to the L. monocytogenes concentration ranging from 10 to 106 cfu·mL−1, and the visual determination can be done down to 10 cfu·mL−1. For spiked food samples analyzed without pre-enrichment, recoveries of 98.4% to 99.3% were achieved could be verified and RSD were less than 10%. This work may offer a broad prospect for sensitive and specific determination of pathogens.Graphical abstract

Abstract:Infections from invasive Listeria monocytogenes (L. monocytogenes) frequently occur in food and can cause high morbidity and death. Thus, the sensitive, specific, and rapid detection of L. monocytogenes is critical for ensuring food safety and public health. Herein, a fluorescence immunoassay for trace L. monocytogenes detection was designed based on guinea pig antibody-functionalized magnetic nanoparticles (Fe3O4 NPs/pAb1) and rabbit antibody-anchored CdZnTe quantum dots (CdZnTe QDs/pAb2). Because of the antibody-directed magnetic separation and long-wave fluorescent emission for CdZnTe QD indication, the constructed immunoassay strategy presented excellent anti-interference performance toward a biological matrix. The immunosensor exhibited a wide detection range of 1 to 109 CFU mL-1 for L. monocytogenes and a low limit of detection (LOD) of 1 CFU mL-1, achieving an exceptionally sensitive detection of trace L. monocytogenes. Meanwhile, the immunosensor showed good specificity and had a short time-consumption of 60 min to realize the accurate determination of trace Listeria monocytogenes in spiked tap water and pasteurized milk samples.
A Novel Fluorescence Method for the Rapid and Effective Detection of Listeria Monocytogenes Using Aptamer-Conjugated Magnetic Nanoparticles and Aggregation-Induced Emission Dots.

Abstract:Fluorescence-based assays are efficient tools for the detection of Listeria monocytogenes (L. monocytogenes). However, they are always restricted by the phenomenon of aggregation-caused quenching (ACQ). The emergence of aggregation-induced emission (AIE) materials perfectly overcomes this shortcoming. Through harnessing the AIE characteristic with magnetic enrichment, we propose an approach to achieve a promising detection method that combines an aptamer and antibody-based dual recognition units. Aptamer-coupled magnetic beads were used for the specific capture of L. monocytogenes. IgG-TPE-OH@BSA NPs were facilely synthesized through encapsulating 1-(4-hydroxyphenyl)-1,2,2-triphenylethene (TPE-OH) in bovine serum albumin (BSA) microspheres, and possessed a bright fluorescence signal due to the aggregation of TPE-OH in BSA NPs. Rabbit immunoglobulin G (IgG) antibodies were labelled on the surface. In the detection system, the fluorescence intensity of the IgG-TPE-OH@BSA NPs in the supernatant was monitored to avoid the signal interference resulting from the deposit after magnetic separation. Using our strategy, the range of detection for L. monocytogenes is 10-106 cfu mL-1, and the detection limit is as low as 10 cfu mL-1 with a good selectivity. Upon analysis of spiked samples, the recoveries ranged from 95.37% to 101.90% without any pre-enrichment.

Abstract:The authors describe a rapid colorimetric assay for Listeria monocytogenes (L. monocytogenes) based on the o-phenylenediamine-mediated deaggregation of gold nanoparticles. Silver nanoclusters are used as an artificial enzyme that can oxidize o-phenylenediamine to form o-benzoquinone diamine. Aptamer and IgY antibodies were chosen to conjugate with magnetic beads and silver nanoclusters, respectively, which can recognize and bind L. monocytogenes at different specific binding sites. This results in the disassembly of colloidal gold nanoparticles which is accompanied by a color change from blue to red, with peaks at 730 and 525 nm, respectively. The method allows L. monocytogenes to be colorimetrically determined in the 10 to 106 cfu·mL−1 concentration range without pre-enrichment, and the limit of detection is as low as 10 cfu·mL−1. Recoveries ranging from 97.4 to 101.3% are found when analyzing spiked food samples. Abstract:Listeria monocytogenes is one of the major foodborne pathogens, which may cause serious food safety problems and illnesses in humans and animals. Consequently, sensitive, fast and reliable detection methods, as well as effective sample preparation methods are of great demanding. This study established a magnetic separation method based on a aptamer functionalized positively charged magnetic beads (Fe3O4@aptamer) and constructed a fluorescent test strip sensor for rapid, sensitive and specific detection of Listeria monocytogenes. Benefit from the dual recognition and signal amplification process of Fe3O4@aptamer enrichment and PCR of hly gene, the fluorescent strip sensor for detection of Listeria monocytogenes was reliable and sensitive, a linear curve was obtained in the range of 1.0×102-1.9×108 CFU mL-1, and the detection limit was 1.0×102 CFU mL−1. The detection can be done in 3 h without culture enrichment. Furthermore, the developed method was successfully applied for the detection of Listeria monocytogenes in pork tenderloin, the recoveries ranged from 91.1% to 97.1%, with the coefficient of variation less than 23.4%, revealing the feasible and reliable application of this method in practical sample. The proposed fluorescent strip sensor is rapid, sensitive and specific, which has great application prospects in the field of pathogenic bacterium detection.

Abstract:Background: Listeria monocytogenes is a pathogenic bacterium that can lead to severe illnesses, especially among vulnerable populations. Therefore, the development of rapid and sensitive detection methods is vital to prevent and manage foodborne diseases. In this study, we used tetraethylenepentamine (TEPA)-functionalized magnetic nanoparticles (MNPs) and a loop-mediated isothermal amplification (LAMP)-based CRISPR/Cas12a-based biosensor to concentrate and detect, respectively, L. monocytogenes. LAMP enables DNA amplification at a constant temperature, providing a highly suitable approach for point-of-care testing (POCT). The ability of CRISPR/Cas12a to cleave ssDNA reporter, coupled with TEPA-functionalized MNPs effective attachment to negatively charged bacteria, forms a promising biosensor. Results: The LAMP assay was meticulously developed by selecting specific primers and designing crRNA sequences targeting a specific region within the hly gene of L. monocytogenes. We selected primer and refined the amplification conditions by systematically exploring a temperature range from 59 °C to 69 °C, ensuring the attainment of optimal performance. This process was complemented by systematic optimization of LAMP-CRISPR/Cas12a system parameters. In particular, we successfully established the optimal ssDNA reporter concentrations (0-1.2 μM) and Cas12a-mediated trans-cleavage times (0-20 min), crucial components that underpin the effectiveness of the LAMP-CRISPR/Cas12a-based biosensor. For optimizing parameters in capturing L. monocytogenes using TEPA-functionalized MNPs, capture efficiency was significantly enhanced through adjustments in TEPA-functionalized MNPs concentration, incubation times, and magnetic separation duration. Large-volume (20 mL) magnetic separation exhibited a 10-fold sensitivity improvement over conventional methods. Utilizing TEPA-functionalized MNPs, the LAMP-CRISPR/Cas12a-based biosensor achieved detection limits of 100 CFU mL-1 in pure cultures and 100 CFU g-1 in enoki mushrooms. Significance: The integration of this novel technique with the LAMP-CRISPR/Cas12a-based biosensor enhances the accuracy and sensitivity of L. monocytogenes detection in foods, and it can be a promising biosensor for POCT. The 10-fold increase in sensitivity compared to conventional methods makes this approach a groundbreaking advancement in pathogenic bacteria detection for food safety and public health.
Rapid on-site detection of viable Escherichia coli O157: H7 in lettuce using immunomagnetic separation combined with PMAxx-LAMP and nucleic acid lateral flow strip

Abstract:Escherichia coli (E. coli) O157: H7 is one of the most common foodborne pathogens that can be transmitted through contaminated food and water sources, posing a serious threat to food safety and public health worldwide. The rapid and accurate diagnosis of E. coli O157: H7 infection is essential for effective control and management of disease epidemics originating from food and water sources. In this study, we used immunomagnetic separation (IMS) technology to magnetically enrich E. coli O157: H7, further amplify the target gene rfbE using the method combining improved propidium monoazide (PMAxx) and loop-mediated isothermal amplification (LAMP) (PMAxx-LAMP), and detect viable E. coli O157: H7 in lettuce by the nucleic acid lateral flow strip (NALFS) method. We used IMS to capture and enrich the target bacteria, effectively eliminating the interference of the food matrix and improving the detection limit in lettuce samples. PMAxx treatment eliminated the false-positive results from dead bacteria and, in combination with LAMP-NALFS, detected viable E. coli O157: H7 accurately. The IMS-PMAxx-LAMP-NALFS assay allowed us to detect viable E. coli O157: H7 at concentrations as low as 81 CFU/g in artificially contaminated lettuce samples within 2 h without any heavy and costly instruments. Thus, this assay provides a simple, rapid, and low-cost tool for the on-site detection of pathogenic bacteria, especially with limited resources.

Abstract:Listeria monocytogenes (LM) is a foodborne pathogen, and it can pose a risk of serious diseases to the human health. Hence, the development of an effective method for the detection of LM is very important. In this study, by selecting LM as the template and 3-thiopheneacetic acid as the functional monomer, an LM-imprinted polymer (LIP)-based sensor was proposed for the first time to detect LM by electropolymerizing TPA on the glassy carbon electrode (GCE) surface in the presence of LM. After the removal of the LM template from the electrode surface, the obtained sensor was denoted as LIP/GCE, which could effectively recognize and capture LM cells. By using [Fe(CN)6]4-/3- as the probe, its peak current at LIP/GCE could be restricted when the LM cells were captured into the imprinted cavity of LIP/GCE, and the current value decreased with an increase in the LM concentration. Serious conditions were optimized for achieving highly sensitive detection, and a low detection limit (6 CFU mL-1) coupled with a wide linear range (10 to 106 CFU mL-1) was obtained for LM. Finally, the inter-electrode reproducibility, stability, selectivity, and applicability of LIP/GCE were also investigated, and the obtained results were acceptable.
Rapid Detection of Escherichia coli O157:H7 in Fresh Lettuce Based on Localized Surface Plasmon Resonance Combined with Immunomagnetic Separation.

Abstract:This study presents a method for rapid detection of Escherichia coli O157:H7 in fresh lettuce based on the properties of target separation and localized surface plasmon resonance of immunomagnetic nanoparticles. The multifunctional immunomagnetic nanoparticles enabling simultaneous separation and detection were prepared by synthesizing magnetic nanoparticles (ca. 10 nm in diameter) composed of an iron oxide (Fe3O4) core and gold shell and then conjugating these nanoparticles with the anti- E. coli O157:H7 antibodies. The application of multifunctional immunomagnetic nanoparticles for detecting E. coli O157:H7 in a lettuce matrix allowed detection of the presence of <1 log CFU mL-1 without prior enrichment. In contrast, the detection limit of the conventional plating method was 2.74 log CFU mL-1. The method, which requires no preenrichment, provides an alternative to conventional microbiological detection methods and can be used as a rapid screening tool for a large number of food samples.

Abstract:A novel strip test system combining immunomagnetic separation with lateral flow immunoassay (LFIA) was established for the accurate detection of Listeria monocytogenes. In this system, a pair of matched monoclonal antibodies was used to construct a sandwich immunoassay, in which superparamagnetic particles were coupled with one of the antibodies as a labeled antibody to capture the target bacteria, while the other antibody was immobilized on the detection zone. After a 20-min reaction, the strips were analyzed by a novel instrument which could detect the magnetic signal of the immunocomplex in a magnetic field. Sensitivity evaluation showed that the limit of detection (LOD) of the superparamagnetic LFIA system for L. monocytogenes was 10(4) CFU/mL, which was at least one log lower than conventional LFIA. No cross-reaction was observed when Salmonella, Escherichia coli O157:H7, or three types of harmless Listeria strains were tested. Further evaluation with actual food samples indicated that the superparamagnetic LFIA system showed 100 % concordance with real-time PCR. Therefore, this novel superparamagnetic LFIA system could be used as a rapid, sensitive, and specific method for the detection of L. monocytogenes.
POD-like nanozyme constructed from perspective of charge transfer engineering for biosensing of magnetic separation treated Listeria monocytogenes

Abstract:For foodborne pathogens pose a serious threat to public health, a magnetic separation strategy and a nanozyme-based biosensor are proposed for biosensing of Listeria monocytogenes (L. monocytogenes). In this work, doripenem is selected as a recognized molecule for the modification of magnetic beads to capture L.monocytogenes in food and environmental samples. Furthermore, the POD-like MXene-Hemin-Au is constructed from perspective of charge transfer engineering which provides a vivid example to rational design of nanozymes. Finally, the captured L.monocytogenes is labeled with MXene-Hemin-Au@mAb, forming the sandwich complexes for quantitative determination. The current signals that generated by the complexes exhibit a good linear relationship with a limit of detection of 2.3 × 101 CFU/mL. The biosensor shows a satisfactory applicability in real samples with recoveries of 91.19% to 102.98%. Overall, the biosensor with integrated magnetic separation strategy presents a potential approach for high sensitivity biosensing of foodborne pathogens.

Abstract:Healthcare-associated infections (HAIs) have increasingly become a threat to global health. Rapid, accurate, and reliable detection of HAI-related bacteria is urgently needed. Here, we presented a novel antimicrobial peptide-based surface-enhanced Raman scattering lateral flow assay (SERS-LFA) for the detection of multiple HAI-related bacteria. Antimicrobial peptide cecropin 1(CP1) was immobilized on the surface of Fe 3 O 4 @Au by the streptavidin-biotin system to capture Pseudomonas aeruginosa ( P. aeruginosa ) and Escherichia coli O157:H7 ( E. coli O157:H7) simultaneously. The synthesized Fe 3 O 4 @Au-CP1 exhibits excellent broad-spectrum capturing ability, rapid magnetic enrichment function, and superior antibacterial activity. LFA strips enable the specific detection of two target bacteria in a quick and simple manner, with bacterial type differentiation via corresponding monoclonal antibodies sprayed. The highly sensitive SERS signals and the amplification effect of streptavidin-biotin significantly improved the sensitivity of detection. The LODs of P. aeruginosa and E. coli O157:H7 were 12 and 16 cells/mL, respectively. This assay also exhibited good specificity and reproducibility when applied to urine samples. Therefore, Fe 3 O 4 @Au-CP1-based SERS-LFA has great potential for detecting multiple bacteria after infection as well as regularly monitoring therapeutic efficacy.

Detection of Listeria monocytogenes based on combined aptamers magnetic capture and loop-mediated isothermal amplification

Abstract:Rapid detection of Listeria monocytogenes (L. monocytogenes) is important to ensure timely treatment and to prevent outbreaks of listeriosis. Here, we reported an efficient and accurate system using aptamers magnetic capture and loop-mediated isothermal amplification (AMC-LAMP) for sensitive detection of L. monocytogenes. A set of combined aptamers (including four individual aptamers with high binding affinity to L. monocytogenes) was conjugated to magnetic beads, used for capture L. monocytogenes. After incubation at room temperature for 45 min, the captured L. monocytogenes cells were directly used for DNA extraction (1 h). Subsequently, the LAMP assays were carried out at 63 °C for 40 min, and the amplification products were visualized via SYBR Green® I staining. The AMC-LAMP had a detection limit of 5 CFU/mL, and the total assay time was approximate 3 h. By the protocols developed in this study, 17 food samples was test for L. monocytogenes without prior culture enrichment. The accuracy of AMC-LAMP was shown to be 100% when compared with culture-biotechnical examination, while PCR failed to detect L. monocytogenes in one of five positive samples. These results indicate the developed AMC-LAMP is a potential useful method for rapid screening and on-site detection of L. monocytogenes in food samples.

Abstract:Food safety represents a critical global public health issue, with safety challenges posed by foodborne pathogens garnering extensive attention. Therefore, we introduce a co-recognition, enrichment and sensing (CES) all-in-one strategy for analysis of bacteria with low background and high specificity. This method employs antimicrobial peptide (AMP) functionalized magnetic nanoparticles (MNPs) to enrich bacteria and uses aptamer@Au@PBA (K x MFe(CN) 6 (M = Pb and Ni)) NPs as silent SERS tags. When both S. aureus and E. coli O157:H7 are present, the silent SERS probes could specifically label the target bacteria, forming a sandwich-like structure. This binding induces silent Raman shifts (2139 cm −1 and 2197 cm −1 ), enabling quantification of two bacteria. Coupling with the modular flexible microfluidics and magnetic control slider device, this platform facilitates rapid switching between magnetic loading and elution. The CES SERS method demonstrated linear relationships for both S. aureus and E. coli O157:H7 at 50–1600 cfu mL −1 , with detection limits of 14 and 18 cfu mL −1 , respectively. The method achieved recovery rates of 85.6–112% and relative standard deviations of 1.5–8.6%. Validation using the ELISA method revealed relative errors between −7.5 and 4.3%. The CES approach has potential applications in food safety, environmental monitoring, and biomedical diagnosis.

Abstract:L. monocytogenes (L. m) is one of the most common causes of foodborne diseases. To enable timely monitoring of L. m contamination, a highly efficient and sensitive electrochemical biosensor was developed for the precise determination of L.m . The biosensor was prepared by sequentially modifying glassy carbon electrode (GCE) with silicon methylene blue (Si@MB), gold nanoparticles (AuNPs) and aptamer (Apt). Differential pulse voltammetry (DPV) was used to quantify L.m . This biosensor could be finished in 1.5 h, which significantly reduced the detection time. The resulting electrochemical biosensor achieved good linear (10 2 -10 7 CFU/mL) and excellent detection limit of 2.6 CFU/mL. Moreover, the novel strategy has been successfully applied to actual samples, with the recoveries ranging from 84.7% to116.0% in lettuce and from 80.0% to 110% in fresh-cut fruit. This biosensor shows great potential application in monitoring bacterial food contamination and infectious diseases.

Abstract:Accurate and timely diagnosis of Listeria monocytogenes (L. monocytogenes), a well-known foodborne pathogen with a serious threat to human life and health, is critical for managing bacterial infections and food quality. Here, we report a genosensor platform based on DNA biofunctionalized superparamagnetic nickel colloidal nanocrystal clusters (SNCNCs) coupling with "IS-primer" amplification reaction (ISAR) for rapid, ultrasensitive and specific L. monocytogenes detection. Specifically, the catalytic activity of SNCNCs was identified. After three-step modification, the signal unit was formed by immobilizing AuNPs and hairpin probes on the surface of SNCNCs, achieving enhanced catalytic activity, leading to the reduction of a colorimetric substrate 4-nitrophenol (4-NP). The in-situ signal amplification of target RNA on the surface of SNCNCs coupling with ISAR showed high sensitivity and a detection limit of 1.50 × 10-3 ng mL-1 under optimized conditions As SNCNCs can bind and lyse bacteria, this genosensor can detect live L. monocytogenes without RNA extraction, with a detection limit of 3.80 × 102 CFU mL-1. Furthermore, the system could also detect L. monocytogenes in artificially contaminated milk samples within 6 h. These results indicate the great potential of our method for sensitive, specific, convenient and low-cost bacterial detection in food, environment and clinics.
