Abstract:Integrins are αβ heterodimers that mediate cell adhesion and transduce signals bidirectionally across the cell membrane. Integrins often exist in low affinity (or inactive) states for ligand binding on the cell curface, but change conformations to high affinity (or activated) states when induced by stimuli from inside or outside the cell. Crystallographic and electron microscopic studies have obtained evidence that the low affinity states correspond to bent conformations where the integrin headpiece interacts with the legs, whereas the high affinity states correspond to more extended conformations where integrins stand up, suggesting a switchblade-like unbending model for integrin activation. In this model, the key conformational changes on integrin headpiece include the swing out of the hybrid domain and the downward movement of I-like domain α7 helix. These conformational changes are suggested by crystal structures of β3 integrin headpieces. The unliganded αvβ3 headpiece displays a closed hybrid domain with the I-like α7 helix in an upper position and the pseudo-liganded αIIbβ3 headpiece shows an open hybrid domain with the I-like α7 helix in a lower position. Using molecular dynamics simulations we studied the stability of the two conformations of the I-like domain α7 helix and the hybrid domain, the transition between the two conformations in each structure, and the coupling between the conformational changes in the two structures. We observed that the β-propeller domain of the α subunit helped stabilize the hybrid domain at the close conformation. The down/upward movements of the I-like α7 helix correlated with the opening/closing of the hybrid domain. Our simulations identify key residues to the α7 helix movement and suggest the existence of intermediate conformations of the hybrid domain between the open and the close conformations.

Abstract:Integrins may undergo large conformational changes during activation, but the dynamic processes and pathways remain poorly understood. We used molecular dynamics to simulate forced unbending of a complete integrin alpha(V)beta(3) ectodomain in both unliganded and liganded forms. Pulling the head of the integrin readily induced changes in the integrin from a bent to an extended conformation. Pulling at a cyclic RGD ligand bound to the integrin head also extended the integrin, suggesting that force can activate integrins. Interactions at the interfaces between the hybrid and beta tail domains and between the hybrid and epidermal growth factor 4 domains formed the major energy barrier along the unbending pathway, which could be overcome spontaneously in similar to 1 mu s to yield a partially-extended conformation that tended to rebend. By comparison, a fully-extended conformation was stable. A newly-formed coordination between the alpha(V) Asp457 and the alpha-genu metal ion might contribute to the stability of the fully-extended conformation. These results reveal the dynamic processes and pathways of integrin conformational changes with atomic details and provide new insights into the structural mechanisms of integrin activation.

Abstract:BACKGROUND:Integrin α(L)β₂ (lymphocyte function-associated antigen, LFA-1) bears force upon binding to its ligand intercellular adhesion molecule 1 (ICAM-1) when a leukocyte adheres to vascular endothelium or an antigen presenting cell (APC) during immune responses. The ligand binding propensity of LFA-1 is related to its conformations, which can be regulated by force. Three conformations of the LFA-1 αA domain, determined by the position of its α₇-helix, have been suggested to correspond to three different affinity states for ligand binding. METHODOLOGY/PRINCIPAL FINDINGS:The kinetics of the force-driven transitions between these conformations has not been defined and dynamically coupled to the force-dependent dissociation from ligand. Here we show, by steered molecular dynamics (SMD) simulations, that the αA domain was successively transitioned through three distinct conformations upon pulling the C-terminus of its α₇-helix. Based on these sequential transitions, we have constructed a mathematical model to describe the coupling between the αA domain conformational changes of LFA-1 and its dissociation from ICAM-1 under force. Using this model to analyze the published data on the force-induced dissociation of single LFA-1/ICAM-1 bonds, we estimated the force-dependent kinetic rates of interstate transition from the short-lived to intermediate-lived and from intermediate-lived to long-lived states. Interestingly, force increased these transition rates; hence activation of LFA-1 was accelerated by pulling it via an engaged ICAM-1. CONCLUSIONS/SIGNIFICANCE:Our study defines the structural basis for mechanical regulation of the kinetics of LFA-1 αA domain conformational changes and relates these simulation results to experimental data of force-induced dissociation of single LFA-1/ICAM-1 bonds by a new mathematical model, thus provided detailed structural and kinetic characterizations for force-stabilization of LFA-1/ICAM-1 interaction.

Abstract:Abstract Recognition of laminin by integrin receptors is central to the epithelial cell adhesion to basement membrane, but the structural background of this molecular interaction remained elusive. Here, we report the structures of the prototypic laminin receptor α6β1 integrin alone and in complex with three-chain laminin-511 fragment determined via crystallography and cryo-electron microscopy, respectively. The laminin-integrin interface is made up of several binding sites located on all five subunits, with the laminin γ1 chain C-terminal portion providing focal interaction using two carboxylate anchor points to bridge metal-ion dependent adhesion site of integrin β1 subunit and Asn189 of integrin α6 subunit. Laminin α5 chain also contributes to the affinity and specificity by making electrostatic interactions with large surface on the β-propeller domain of α6, part of which comprises an alternatively spliced X1 region. The propeller sheet corresponding to this region shows unusually high mobility, suggesting its unique role in ligand capture.

Abstract:Extracellular matrix (ECM) plays an integral role in different developmental stages and in multiple systems. However, due to ECM being composed of various extracellular components (growth factors, cytokines, and hormones), its innate complexity and the lack of any efficient purification techniques limit further research into the detailed mechanisms of its role in cellular activities. Laminin (LN), a synthetic recombinant basement membrane protein, can solve the above problems as it is a critical component of ECM and can be completely and reproducibly chemically defined. This article summarizes the functions and mechanisms of LN during osteogenic differentiation and stemness maintenance. LN-111 enhances osteogenic differentiation of mesenchymal stem cells (MSCs) and bone marrow progenitor cells (BMPCs) via the ECM receptor integrin-alpha 1, alpha V, alpha 6, and beta 1. LN-332 stimulates osteogenic differentiation of MSCs and bone-marrow-derived MSCs (BMSCs) by alpha 3 beta 1/alpha 3 beta 3 integrin-mediated activation of the focal adhesion kinase (FAK)/extracellular-signal-regulated kinase (ERK), Wnt5a, and bone morphogenic proteins (BMP) signaling pathways. Moreover, LN-111, LN-211, and LN-332 regulate the osteogenic differentiation of dental follicle cells (DFCs) via the integrin-alpha 2/beta 1 and FAK/ERK signaling pathways. LN-511 and LN-521 can preserve the pluripotency of pluripotent stem cells (PSCs) and human embryonic stem cells (hESCs) via the integrin-alpha 6 beta 1/alpha V beta 1 and the PI3k/Akt pathways. In addition, a variety of laminin fragments such as iMatrix-411; iMatrix-511; bioactive peptide sequences of LN-alpha 2, PPFEGCIWN, and DLTIDDSYWYRI; and LN-332 large globular 3 (LG3), were confirmed to induce osteogenic differentiation. LN511-E8, LN332-E8 fragments, and the laminin-mimicking sequence YIGSR sustain stemness. LN may have potential applications in surface gene markers, xeno-free cultures, and enhancing the expression of osteogenic factors.

Abstract:Laminins are cell-adhesive glycoproteins that are essential for basement membrane assembly and function. Integrins are important laminin receptors, but their binding site on the heterotrimeric laminins is poorly defined structurally. We report the crystal structure at 2.13 Å resolution of a minimal integrin-binding fragment of mouse laminin-111, consisting of ∼50 residues of α1β1γ1 coiled coil and the first three laminin G-like (LG) domains of the α1 chain. The LG domains adopt a triangular arrangement, with the C terminus of the coiled coil situated between LG1 and LG2. The critical integrin-binding glutamic acid residue in the γ1 chain tail is surface exposed and predicted to bind to the metal ion-dependent adhesion site in the integrin β1 subunit. Additional contacts to the integrin are likely to be made by the LG1 and LG2 surfaces adjacent to the γ1 chain tail, which are notably conserved and free of obstructing glycans.

Abstract:Excessive foot pronation has been considered to be related to anterior knee pain. We undertook a prospective study to test the hypothesis that exertional anterior knee pain is related to the static and dynamic parameters of foot pronation. Two weeks before beginning basic training lasting for 14 weeks, 473 infantry recruits were enrolled into the study and underwent two-dimensional measurement of their subtalar joint displacement angle during walking on a treadmill. Of the 405 soldiers who finished the training 61 (15%) developed exertional anterior knee pain. No consistent association was found between the incidence of anterior knee pain and any of the parameters of foot pronation. While a statistically significant association was found between anterior knee pain and pronation velocity (left foot, p = 0.05; right foot, p = 0.007), the relationship was contradictory for the right and left foot. Our study does not support the hypothesis that anterior knee pain is related to excessive foot pronation.

Abstract:ABSTRACT Various studies have demonstrated a role for E2F proteins in the control of transcription of genes involved in DNA replication, cell cycle progression, and cell fate determination. Although it is clear that the functions of the E2F proteins overlap, there is also evidence for specific roles for individual E2F proteins in the control of apoptosis and cell proliferation. Investigating protein interactions that might provide a mechanistic basis for the specificity of E2F function, we identified the E-box binding factor TFE3 as an E2F3-specific partner. We also show that this interaction is dependent on the marked box domain of E2F3. We provide evidence for a role for TFE3 in the synergistic activation of the p68 subunit gene of DNA polymerase α together with E2F3, again dependent on the E2F3 marked box domain. Chromatin immunoprecipitation assays showed that TFE3 and E2F3 were bound to the p68 promoter in vivo and that the interaction of either E2F3 or TFE3 with the promoter was facilitated by the presence of both proteins. In contrast, neither E2F1 nor E2F2 interacted with the p68 promoter under these conditions. We propose that the physical interaction of TFE3 and E2F3 facilitates transcriptional activation of the p68 gene and provides strong evidence for the specificity of E2F function.
[Integrin Alpha6 Beta4 and Experimental Allergic Neuritis].

Abstract:OBJECTIVE:To explore the relationship between integrins alpha6,beta4 and the process of the injury and repair of the myelin, their expressions in experimental allergic neuritis were analyzed.METHODS:Experimental allergic neuritis (EAN) models were induced in Lewis rats. The expressions of integrins alpha6 and beta 4 and their receptor laminin, both in the acute phase and recovery phase of EANs, were analyzed by in situ hybridization and immunohistochemistry.RESULTS:The expression of beta4 decreased in the acute phase P<0.05 and showed no significant changes in the recovery phase(P=0.6840). There were no significant changes of the expression of alpha6 and laminin both in the acute phase and recovery phase of EANs by immunohistochemistry (alpha6 P=0.0751;laminin P=0.2047). The similar results were obtained by in situ hybridization (beta4 in acute phase P<0.05;beta4 in recovery phase P=0.823;alpha6 P= 0.81).CONCLUSION:The expressions of integrins of Schwann cells were affected by inflammation injury, and their patterns of expression were similar to those of integrins alpha6, beta1 and beta4 during embryogenesis. It is inferred that integrin beta4 and the reformation of myelin have a more imitate relationship. The expression changes of alpha6 and beta4 correlate with the injury and repair of myelin during EAN.
In situ-RT-PCR and immunohistochemistry for the localisation of the mRNA of the alpha 3 chain of laminin and laminin-5 during human organogenesis

Abstract:Abstract. Laminin-5 is known to be an integral part of the hemidesmosome and therefore responsible for the integrity of the connection of the epithelium to the basement membrane. This is also an important mechanism during embryonic development, as documented by studies in mice. In an attempt to elucidate its implication for human development we localised the mRNA of the α3 chain of laminin with the help of in situ RT-PCR, and the laminin-5 protein immunohistochemically. We systematically investigated kidney, lung, skin and intestinal tissue of consecutive developmental stages during human embryogenesis. From gw 6.5 onwards, the mRNA of the α3 chain of laminin was found exclusively in the cytoplasm of epithelial cells of the developing kidney, lung, skin and intestine. Interestingly, in the skin and intestine from gw 8 onwards, the superficial cell layers also stained positive for the mRNA, while the protein was still only found in the dermal-epidermal and enteric basement membrane zones. In all developing organs investigated, the mRNA of the α3 chain of laminin is strictly of epithelial origin and the corresponding protein localised in the underlying basement membrane zones. Due to this discrepancy, we postulate a broader role for laminin-5 during human embryogenesis, for example, for epithelial cell development, beyond its involvement in hemidesmosome formation and cell adhesion.

Abstract:We have examined the mechanism of signal transduction by the hemidesmosomal integrin alpha 6 beta 4, a laminin receptor involved in morphogenesis and tumor progression. Immunoprecipitation and immune complex kinase assays indicated that antibody- or laminin-induced ligation of alpha 6 beta 4 causes tyrosine phosphorylation of the beta 4 subunit in intact cells and that this event is mediated by a protein kinase(s) physically associated with the integrin. Co-immunoprecipitation and GST fusion protein binding experiments showed that the adaptor protein Shc forms a complex with the tyrosine-phosphorylated beta 4 subunit. Shc is then phosphorylated on tyrosine residues and recruits the adaptor Grb2, thereby potentially linking alpha 6 beta 4 to the ras pathway. The beta 4 subunit was found to be phosphorylated at multiple tyrosine residues in vivo, including a tyrosine-based activation motif (TAM) resembling those found in T and B cell receptors. Phenylalanine substitutions at the beta 4 TAM disrupted association of alpha 6 beta 4 with hemidesmosomes, but did not interfere with tyrosine phosphorylation of Shc and recruitment of Grb2. These results indicate that signal transduction by the alpha 6 beta 4 integrin is mediated by an associated tyrosine kinase and that phosphorylation of distinct sites in the beta 4 tail mediates assembly of the hemidesmosomal cytoskeleton and recruitment of Shc/Grb2.
Structural insights into the molecular recognition of integrin αVβ3 by RGD-containing ligands: The role of the specificity-determining loop (SDL)

Abstract:Integrin aVb3 is a prominent member of the "RGD-recognizing" integrin family of cell surface receptors. aVb3 binds to various extracellular matrix (ECM) proteins and oxysterols such as 25-hydroxycholesterol, is implicated in several diseases, including cancer metastasis, lung fibrosis, inflammation, and autoimmune diseases, and is pursued as a valuable therapeutic target. Despite enormous efforts to seek a pure antagonist, to date, no single drug candidate has successfully reached clinics due to associated partial agonism and toxicity issues. Developing effective and safe inhibitors require a thorough understanding of the molecular interactions and structural changes related to the receptor's activation and inhibition mechanisms. This study offers a comprehensive residue-residue contact and network analyses of the ligand-binding beta-propeller alpha-I domains (headpiece) based on all available experimental structures of integrin aVb3 in unliganded, agonist-, antagonist-, and antibody-bound states. The analyses reveal many critical interactions that were not reported before and show that specific orientation and interactions of residues from the specificity-determining loop (SDL) are critical in molecular recognition and regulation. Also, the network analysis reveals that residues from the nearby allosteric site (site II) connect to the primary RGD-binding site via SDL, which likely acts as an interface between the two sites. Our results provide valuable insights into molecular interactions, structural changes, distinct features of the active and inactive headpiece conformations, the role of SDL in ligand recognition, and SDL-mediated allostery. Thus, the insights from this study may facilitate the designing of pure antagonists or site II-mediated allosteric modulators to integrin aVb3 to treat various diseases.

Distinct Functions of Integrin Alpha and Beta Subunit Cytoplasmic Domains in Cell Spreading and Formation of Focal Adhesions.

Abstract:Integrin-mediated cell adhesion often results in cell spreading and the formation of focal adhesions. We exploited the capacity of recombinant human alpha(IIb)beta3 integrin to endow heterologous cells with the ability to adhere and spread on fibrinogen to study the role of integrin cytoplasmic domains in initiation of cell spreading and focal adhesions. The same constructs were also used to analyze the role of the cytoplasmic domains in maintenance of the fidelity of the integrin repertoire at focal adhesions. Truncation mutants of the cytoplasmic domain of alpha(IIb) did not interfere with the ability Of alpha(IIb)beta3 to initiate cell spreading and form focal adhesions. Nevertheless, deletion of the alpha(IIb) cytoplasmic domain allowed indiscriminate recruitment of alpha(IIb)beta3 to focal adhesions formed by other integrins. Truncation of the beta3 subunit cytoplasmic domain abolished cell spreading mediated by alpha(IIb)beta3 and also abrogated recruitment of alpha(IIb)beta3 to focal adhesions. This truncation also dramatically impaired the ability of alpha(IIb)beta3 to mediate the contraction of fibrin gels. In contrast, the beta3 subunit cytoplasmic truncation did not reduce the fibrinogen binding affinity of alpha(IIb)beta3. Thus, the integrin beta3 subunit cytoplasmic domain is necessary and sufficient for initiation of cell spreading and focal adhesion formation. Further, the beta3 cytoplasmic domain is required for the transmission of intracellular contractile forces to fibrin gels. The alpha subunit cytoplasmic domain maintains the fidelity of recruitment of the integrins to focal adhesions and thus regulates their repertoire of integrins.
Laminin Receptor Characterization in Acute Myeloid Leukemia: Integrin α7β1 Defines non-Leukemic Stem Cells with Migratory Potential

Abstract:Interactions with the bone marrow (BM) niche are crucial for promoting self-renewal and survival of acute myeloid leukemia (AML) cells. Consequently, AML cells express a variety of surface receptors to engage with BM niche cells and extracellular matrix proteins, including laminins. Despite the association of laminin receptor expression with stemness in healthy hematopoiesis, the role of laminin receptors in AML remains poorly understood. In this study, we present a comprehensive examination of the laminin receptors integrin α3β1, α6β1, α7β1 and basal cell adhesion molecule (BCAM) in AML. We demonstrate that high mRNA expression of all four laminin receptors correlates with poor overall survival. Notably, integrin α6 and α7 display the highest cell surface presentation among the examined laminin receptors and are higher expressed on AML cells compared to healthy controls. Moreover, our results indicate that integrin α7 expression allows to distinguish between leukemic stem cells (LSC) and non-LSC populations. Specifically, integrin α7 appears to mark non-LSC with enhanced migratory potential. Together, our results confirm the association of high laminin receptor expression with poor prognosis and establish integrin α7 as marker of high migratory non-LSC.

Abstract:Integrin trafficking to and from membrane adhesions is a crucial mechanism that dictates many aspects of a cell's behaviour, including motility, polarisation, and invasion. In endothelial cells (ECs), the intracellular traffic of α5 integrin is regulated by both neuropilin 1 (NRP1) and neuropilin 2 (NRP2), yet the redundancies in function between these co-receptors remain unclear. Moreover, the endocytic complexes that participate in NRP-directed traffic remain poorly annotated. Here we identify an important role for the GTPase-activating protein p120RasGAP in ECs, promoting the recycling of α5 integrin from early endosomes. Mechanistically, p120RasGAP enables transit of endocytosed α5 integrin-NRP1-NRP2 complexes to Rab11 + recycling endosomes, promoting cell polarisation and fibronectin (FN) fibrillogenesis. Silencing of both NRP receptors, or p120RasGAP, resulted in the accumulation of α5 integrin in early endosomes, a loss of α5 integrin from surface adhesions, and attenuated EC polarisation. Endothelial-specific deletion of both NRP1 and NRP2 in the postnatal retina recapitulated our in vitro findings, severely impairing FN fibrillogenesis and polarised sprouting. Our data assign an essential role for p120RasGAP during integrin traffic in ECs and support a hypothesis that NRP receptors co-traffic internalised cargoes. Importantly, we utilise comparative proteomics analyses to isolate a comprehensive map of NRP1-dependent and NRP2-dependent α5 integrin interactions in ECs.

Abstract:Neutrophils are the most abundant leukocytes in human blood. Neutrophil malfunction may cause various immunological diseases. Beta2 integrins are critical for neutrophil recruitment. Kindlin-3 and talin-1 are important regulators for beta2 integrin activation. We have confirmed this using conformation-specific antibodies KIM127 and mAb24 that recognize activation epitopes in beta2 integrin. Besides activation, clustering is also critical for beta2 integrin avidity and adhesion of neutrophils. Whether kindlin-3 and talin-1 are involved in beta2 integrin clustering remains unclear. Here we used super-resolution stochastic optical reconstruction microscopy (STORM) to study their roles in beta2 integrin clustering on human neutrophil-like HL60 cells. We find that lymphocyte function-associated antigen 1 (LFA-1) and macrophage-1 antigen (Mac-1) are significantly clustered in wild-type cells after IL-8 or fMLP stimulation. Using CRISPR-Cas9-mediated kindlin-3 and talin-1 knockout HL60 cells, we found that kindlin-3 but not talin-1 are responsible for beta2 integrin clustering. Our study provides new insights into the regulation of beta2 integrin clustering and leukocyte recruitment. Abstract:The expected number of real zeros of polynomials a 0 + a 1 x + a 2 x 2 +…+a n−1 x n−1 with random coefficients is well studied. For n large and for the normal zero mean independent coefficients, irrespective of the distribution of coefficients, this expected number is known to be asymptotic to (2/π)log n. For the dependent cases studied so far it is shown that this asymptotic value remains O(log n). In this article, we show that when cov(a i , a j ) = 1 − |i − j|/n, for i = 0,…, n − 1 and j = 0,…, n − 1, the above expected number of real zeros reduces significantly to O(log n)1/2.
