Abstract:A series of natural berberine-derived nitroimidazoles as novel antibacterial agents were designed, synthesized and characterized by nuclear magnetic resonance(NMR), infrared spectra(IR), and high resolution mass spectra(HRMS) spectra. The antimicrobial evaluation showed that some target molecules exhibited moderate to good inhibitory activities against the tested bacteria and fungi including clinical drug-resistant strains isolated from infected patients. Especially, 2-fluorobenzyl derivative8 f not only gave strong activity against drug-resistant E. coli with the minimal inhibitory concentration(MIC) value of0.003 m M, 33-fold more active than norfloxacin, but also exhibited low toxicity toward RAW 264.7 cells and less propensity to trigger resistance. The aqueous solubility and Clog P values of target compounds were investigated to elucidate the structureactivity relationships. Molecular docking and quantum chemical studies for compound 8 f rationally explained its antibacterial effect. The further exploration of antibacterial mechanism revealed that the highly active compound 8 f could effectively permeabilize E. coli cell membrane and intercalate into DNA isolated from resistant E. coli to form 8 f-DNA complex that might block DNA replication to exert the powerful bioactivities. Compound 8 f could also selectively address resistant E. coli from a mixture of various strains.

Abstract:A series of novel aminopyrimidinyl benzimidazoles as potentially antimicrobial agents were designed, synthesized and characterized by IR, NMR and HRMS spectra. The biological evaluation in vitro revealed that some of the target compounds exerted good antibacterial and antifungal activity in comparison with the reference drugs. Noticeably, compound 7d could effectively inhibit the growth of A. flavus, E. coli DH52 and MRSA with MIC values of 1, 1 and 8 μg/mL, respectively. Further studies revealed that pyrimidine derivative 7d could exhibit bactericidal mode of action against both Gram positive (S. aureus and MRSA) and Gram negative (P. aeruginosa) bacteria. The active molecule 7d showed low cell toxicity and did not obviously trigger the development of resistance in bacteria even after 16 passages. Furthermore, compound 7d was able to beneficially regulate reactive oxygen species (ROS) generation for an excellent safety profile. Molecular docking study revealed that compound 7d could bind with DNA gyrase by the formation of hydrogen bonds. The preliminary exploration for antimicrobial mechanism disclosed that compound 7d could effectively intercalate into calf thymus DNA to form a steady supramolecular complex, which might further block DNA replication to exert the powerful bioactivities. The binding investigation of compound 7d with human serum albumins (HSA) revealed that this molecule could be effectively transported by HSA.
Synthesis, Antimicrobial Evaluation and Docking Study of Novel 3,5-Disubstituted-2-Isoxazoline and 1,3,5-Trisubstituted-2-Pyrazoline Derivatives

Abstract:Background: The frequent use of antibacterial agents leads to antimicrobial resistance, which is one of the biggest threats to global health today. Therefore, the discovery of novel antimicrobial agents is still urgently needed to overcome the severe infections caused by these putative pathogens resistant to currently available drugs. Objective: The present work was aimed to synthesize and investigate the preliminary structureactivity relationships (SARs) of isoxazoline and pyrazoline derivatives as antimicrobial agent. Methods: Target compounds were obtained in a multistep reaction synthesis and the antimicrobial activity was investigated in several species; two-gram negative (Escherichia coli and Pseudomonas aeruginosa), two-gram positive (Staphylococcus aureus and Bacillus subtilis) and one fungi (Candida albicans), using cup-plate agar diffusion method. The most potent compounds were docked into glucosamine-6-phosphate synthase (GlcN-6-P), the molecular target enzyme for antimicrobial agents, using Autodock 4.2 program. Results: Herein, thirteen novel target compounds were synthesized in moderate to good isolated yield. Based on the SARs, two compounds (2c and 5c) were found to be potent antimicrobial agents on all tested targets, recording potency higher than amoxicillin, the standard antimicrobial drug. Compound 2b identified as selective for gram-negative, while compound 7a found to be selective for gram-positive. The hit compounds (2c, 5a, 5c and 5d) were subjected to a docking study on glucosamine-6-phosphate synthase (GlcN-6-P). All hits were found to bind to the orthosteric (active) site of the enzyme, which might represent a competitive mechanism of inhibition. Conclusion: The newly synthesized heterocyclic compounds could serve as potent leads for the development of novel antimicrobial agents.

Abstract:Bacterial infection poses a serious threat to human life due to the rapidly growing resistance of bacteria to antibacterial drugs, which is a significant public health issue. This study was focused on the design and synthesis of a new series of 25 analogues bearing a 5-cyano-6-oxo-4-substituted phenyl-1,6-dihydropyrimidine scaffold hybridized with different substituted benzenesulfonamides through the thioacetamide linker M1–25. The antimicrobial activity of the new molecules was studied against various Gram-positive, Gram-negative, and fungal strains. All the tested compounds showed promising broad-spectrum antimicrobial efficacy, especially against K. pneumoniae and P. aeruginosa. Furthermore, the most promising compounds, 6M, 19M, 20M, and 25M, were subjected to minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) assays. In addition, the antivirulence activity of the compounds was also examined using multiple biofilm assays. The new compounds promisingly revealed the suppression of microbial biofilm formation in the examined K. pneumoniae and P. aeruginosa microbial isolates. Additionally, in silico ADMET studies were conducted to determine their oral bioavailability, drug-likeness characteristics, and human toxicity risks. It is suggested that new pyrimidine–benzenesulfonamide derivatives may serve as model compounds for the further optimization and development of new antimicrobial and antisepsis candidates.

Abstract:Due to the occurrence of antibiotic resistance, bacterial infectious diseases have become a serious threat to public health. To overcome antibiotic resistance, novel antibiotics are urgently needed. N-thiadiazole4-hydroxy-2-quinolone-3-carboxamides are a potential new class of antibacterial agents, as one of its derivatives was identified as an antibacterial agent against S. aureus. However, no potency-directed structural optimization has been performed. In this study, we designed and synthesized 37 derivatives, and evaluated their antibacterial activity against S. aureus ATCC29213, which led to the identification of ten potent antibacterial agents with minimum inhibitory concentration (MIC) values below 1 mu g/mL. Next, we performed bacterial growth inhibition assays against a panel of drug-resistant clinical isolates, including methicillin-resistant S. aureus, and cytotoxicity assays with HepG2 and HUVEC cells. One of the tested compounds named 1-ethyl-4-hydroxy-2-oxo-N-(5-(thiazol-2-yl)-1,3,4-thiadiazol-2yl)-1,2-dihydroquinoline-3-carboxamide (g37) showed 2 to 128-times improvement compared with vancomycin in term of antibacterial potency against the tested strains (MICs: 0.25-1 mu g/mL vs. 1-64 14/ mu L) and an optimal selective toxicity (HepG2/MRSA, 110.6 to 221.2; HUVEC/MRSA, 77.6-155.2). Further, comprehensive evaluation indicated that g37 did not induce resistance development of MRSA over 20 passages, and it has been confirmed as a bactericidal, metabolically stable, orally active antibacterial agent. More importantly, we have identified the S. aureus DNA gyrase B as its potential target and proposed a potential binding mode by molecular docking. Taken together, the present work reports the most potent derivative of this chemical series (g37) and uncovers its potential target, which lays a solid foundation for further lead optimization facilitated by the structure-based drug design technique. Numerical investigation of a broadband dielectric resonator antenna backed by SIW cavity

Abstract:In this paper an slot excited cylindrical Dielectric Resonator Antenna (DRA) backed by a substrate integrated waveguide (SIW) cavity is numerically investigated based on Finite Element Method (FEM). It consists of a DRA placed on the conducting back plane of the SIW cavity. A resonator slot is used to couple energy from the cavity to the DRA. By merging two resonant frequencies of the slot and the DR, a wide operating bandwidth is achieved. A parametric study of the slot size, feed line length and dimensions of the DRA on the radiation performance of the antenna is carried out. Results show that the proposed structure exhibits as much as bandwidth of 26% for S11<; -10 dB. Gain of the antenna structure is at least 5 dB over the whole operation band and its radiation efficiency is more than 95%.
Antimicrobial activity of novel 5-benzylidene-3-(3-phenylallylideneamino)imidazolidine-2,4-dione derivatives causing clinical pathogens: Synthesis and molecular docking studies

Abstract:Background: This work is development of new hydantoin molecules as treatment of potential antibacterial and antifungal activity against clinical pathogens causing infectious disease. Synthesized compounds were evaluated in molecular docking studies, the most effective compound is used to dock against the targets of 1U1Z, and 1AI9 kinases, to evaluate its binding affinity, hoping to rationalize and obtain potent of antibacterial, antifungal agents. Material and method: The FTIR, 1H &13C NMR, and mass spectra were used to conform new molecules and their evaluation of antimicrobial activity. Gram-negative bacteria of Pseudomonas aeruginosa (ATCC-27853), Klebsiella pneumoniae (recultured) and Escherichia coli (ATCC-25922), and gram-positive bacteria of Enterococcus faecalis (recultured) and Staphylococcus aureus (ATCC-25923) were evaluated for all compounds. The in vitro antifungal activity was evaluated against Cryptococcus neoformans (recultured), Candida albicans (recultured), Aspergillus niger, Microsporum audouinii (recultured) and Aspergillus fumigatus (recultured) for all synthesized compounds. Result: Antibacterial screening, we identified highly active antimicrobial agents for this study for example; gram-negative bacterial screening of 3g was highly (MIC: 0.25 μg/mL) active in contradiction of P. aeruginosa, whereas bacterial screening of 3e and 3h were more active (MIC: 2 μg/mL) in contradiction of K. pneumoniae and also 3g was more (MIC: 2 μg/mL) active in contradiction of E. faecalis than standard ciprofloxacin. Antifungal activity, the 3b was more active (MIC: 0.25 μg/mL) against C. albicance,3g (MIC: 2 μg/mL) and 3h (MIC: 4 μg/mL) were more potential of A. funigatus, and the compound 3c was highly (MIC: 4 μg/mL) active on M. audouinii than clotrimazole. Molecular docking studies also supported the new finding of potent antimicrobial agents, the compound 3g, 3b, and controls Ciprofloxacin, Clotrimazole were checked again proteins 1U1Z and 1AI9 by Autodock Vina program. The compound 3g was highest binding affinity (-8.4 kcal/mol) than ciprofloxacin (-8.2 kcal/mol) in 1U1Z protein and the compound 3b was highest binding affinity (-8.8 kcal/mol) than clotrimazole (-6.8 kcal/mol) in 1AI9 protein respectively. Conclusion: A novel set of imidazolidine-2,4-dione compounds 3a-h have synthesized and characterized successfully. The screening of antimicrobial activity shows that all compounds possess antimicrobial activities. In addition, the objective of the study was succeeded with a few of the promising molecules, which are proving to be a potential treatment of bacterial infection candidates.
Development and evaluation of 2,4‐disubstituted‐5‐aryl pyrimidine derivatives as antibacterial agents

Abstract:A novel class of 5‐aryl‐N2,N4‐dibutylpyrimidine‐2,4‐diamine analogs was designed and evaluated for its antibacterial activity. Structure–activity relationship analysis led to the discovery of a hit compound (12) with optimum broad‐spectrum potency complemented with a good safety profile. The mode of action of compound 12 was preliminarily tested in a macromolecule inhibition assay. Designing novel candidates as potential antibacterial scaffolds has become crucial due to the lack of new antibiotics entering the market and the persistent rise in multidrug resistance. Here, we describe a new class of potent antibacterial agents based on a 5‐aryl‐N2,N4‐dibutylpyrimidine‐2,4‐diamine scaffold. Structural optimization focused on the 5‐aryl moiety and the bioisosteric replacement of the side chain linker atom. Screening of the synthesized compounds focused on a panel of bacterial strains, including gram‐positive Staphylococcus aureus strains (Newman MSSA, methicillin‐ and vancomycin‐resistant), and the gram‐negative Escherichia coli (ΔAcrB strain). Several compounds showed broad‐spectrum antibacterial activity with compound 12, bearing a 4‐chlorophenyl substituent, being the most potent among this series of compounds. This frontrunner compound revealed a minimum inhibitory concentration (MIC) value of 1 μg/mL against the S. aureus strain (Mu50 methicillin‐resistant S. aureus/vancomycin‐intermediate S. aureus) and an MIC of 2 μg/mL against other tested strains. The most potent derivatives were further tested against a wider panel of bacteria and evaluated for their cytotoxicity, revealing further potent activities toward Streptococcus pneumoniae, Enterococcus faecium, and Enterococcus faecalis. To explore the mode of action, compound 12 was tested in a macromolecule inhibition assay. The obtained data were supported by the safety profile of compound 12, which possessed an IC50 of 12.3 μg/mL against HepG2 cells. The current results hold good potential for a new class of extended‐spectrum antibacterial agents.

A harmonic selective unified power quality conditioner using MVR with Kalman filters

Abstract:This paper employs the unified power quality conditioner (UPQC), an integration of series and shunt active filters, to perform harmonic compensation and power factor correction. Selective harmonic compensation can be implemented too. In addition, the UPQC can be used to perform load sharing with the supply. The UPQC is controlled by the multi-variable regulator-based (MVR) UPQC with Kalman filters. This control design employs Kalman filters to extract the fundamental and the harmonic components of the supply voltage and the load current in a power distribution system. Kalman filter is also used to estimate the variables of the UPQC. Simulation results on a single-phase power distribution system are presented to verify the performance.
Design, synthesis and biological evaluation of amphiphilic benzopyran derivatives as potent antibacterial agents against multidrug-resistant bacteria

Abstract:Antimicrobial resistance has emerged as a significant threat to global public health. To develop novel, high efficiency antibacterial alternatives to combat multidrug-resistant bacteria, A total of thirty-two novel amphiphilic benzopyran derivatives by mimicking the structure and function of antimicrobial peptides were designed and synthesized. Among them, the most promising compounds 4h and 17e displayed excellent antibacterial activity against Gram-positive bacteria (MICs = 1-4 μg/mL) with weak hemolytic activity and good membrane selectivity. Additionally, compounds 4h and 17e had rapid bactericidal properties, low resistance frequency, good plasma stability, and strong capabilities of inhibiting and eliminating bacterial biofilms. Mechanistic studies revealed that compounds 4h and 17e could effectively disrupt the integrity of bacterial cell membranes, and accompanied by an increase in intracellular reactive oxygen species and the leakage of proteins and DNA, ultimately leading to bacterial death. Notably, compound 4h exhibited comparable in vivo antibacterial potency in a mouse septicemia model infected by Staphylococcus aureus ATCC43300, as compared to vancomycin. These findings indicated that 4h might be a promising antibacterial candidate to combat antimicrobial resistance.
Novel sulfonylamido benzoxazole derivatives: synthesis, characterisation, molecular docking, DFT, and antimicrobial activity investigations

Abstract:In this work, numerous novel 2‐(p‐substitutedphenyl)‐5‐[(p‐substitutedphenyl)sulfonylamido]benzoxazole derivatives were designed, synthesized, and structurally characterized using mass spectroscopy, 1H‐NMR, 13C‐NMR, and elemental analysis approaches. The antimicrobial activity against several Gram (+), Gram (‐), and fungal species was determined using the in vitro microdilution technique. A molecular docking analysis was performed on all produced compounds utilizing the S. aureus Gyrase complex with Ciprofloxacin and DNA. Two of the most effective compounds against S. aureus, N4 and N9, have binding energies of ‐8.7 kcal/mol and ‐8.6 kcal/mol, respectively, and their interactions have been demonstrated in 2D and 3D. Furthermore, utilizing the 6‐311G(d,p) base set and DFT/B3LYP theory, MEP analysis, geometric optimization, and molecular reactivity analysis (HOMO‐LUMO) of N4 and N9 were performed, and the results were presented. All compounds' theoretical ADMET profiles were computed as well, and they met Lipinski and other strict criteria. With all of this knowledge, this study could be a pioneer in the development of novel anti‐S. aureus compounds.

Abstract:This study focuses on the synthesis of novel 2,3‐disubstituted quinazolin‐4(3H)‐one derivatives and evaluates their antibacterial properties against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus. The antibacterial activity GA4A1 exhibited notable efficacy against E. coli, which was further supported by the highest molecular docking score and stable binding interactions in molecular dynamics simulations. The findings highlight that Compound GA4A1 is a promising candidate for further development as an antimicrobial agent against E. coli. The rise of multidrug‐resistant organisms (MDROs) represents a significant challenge to healthcare, underscoring the need for novel antimicrobial agents. Quinazolinone compounds, noted for their diverse biological activities, particularly at the 2nd and 3rd positions, in conjunction with sulphanilamide and isatin derivatives, present a promising avenue for antibacterial development. This study focuses on the synthesis of novel 2,3‐disubstituted quinazolin‐4(3H)‐one derivatives from Schiff base intermediates GA3A and GA3B. The synthesis involved the reaction of 2‐(substituted phenyl)‐4H‐benzo[d][1,3]‐oxazin‐4‐one with sulphanilamide, and the benzoxazinone intermediates were prepared by reacting anthranilic acid with benzoyl chloride. The antibacterial activities of the Schiff base intermediates and the final Mannich base compounds were evaluated against Staphylococcus aureus, Bacillus cereus, Escherichia coli, and Pseudomonas aeruginosa at concentrations of 50 μg/mL and 100 μg/mL using the agar well diffusion method, with Norfloxacin (50 μg/mL) as the reference standard. While all tested compounds exhibited lower antibacterial activity compared to the standard, GA4A1 showed enhanced efficacy against E. coli, achieving the highest docking score of 78.0352 against the E. coli protein (PDB ID: 1KZN). Molecular dynamics simulations revealed that the GA4A1‐E. coli complex stabilized after 40,000 ps, with root mean square deviation (RMSD) values ranging from 2.5 Å to 5 Å and low root mean square fluctuation (RMSF) values between 0.05 Å and 0.2 Å, indicating the stability of the complex. These findings underscore GA4A1's potential as a potent antimicrobial agent against E. coli.

Abstract:Quinoline and its derivatives are a family with unique medicinal properties, including antibacterial effects. It was assumed that the four quinoline derivatives 1, 2, 3 and 4 had significant activity against pathogenic bacteria. These compounds were synthesized and characterized by TLC, IR, 1H NMR, and 13C NMR analyses. The biological activity of compounds was determined as inhibition zone (IZ) in mm. For compound 1 IZ was 19 ± 0.22 against Klebsiella pneumoniae, IZ was 18 ± 0.22 against Bacillus subtilis, and IZ was 17 ± 0.22 against Staphylococcus aureus. 2 displayed IZ of 18 ± 0.22 against both Klebsiella pneumoniae and Bacillus subtilis. 3 showed IZ of 17 ± 0.22 against Staphylococcus aureus. 4 displayed IZ of 21 ± 0.22, thus showing a higher inhibitory activity against Escherichia coli, than ciprofloxacin. These results demonstrate the potential of the synthesized compounds to work as antibacterial drugs against these strains by inhibiting or deactivating the target proteins.

Abstract:In response to the challenge of antibiotic-resistant microorganisms, oxazolone analogs are frequently used for bacterial, antiviral, and anti-inflammatory treatments. However, few studies have shown bis-oxazolone analogs possess antibacterial activities. In this study, we modified bis-oxazolone molecules with various aromatic amines to create new bis-benzamide and bis-imidazolone derivatives. These derivatives were analyzed using FT-IR, 1H-NMR, and 13C-NMR spectroscopy. Molecular docking revealed favorable interactions with DNA gyrase, with compounds 3, 4a, and 5e showing higher binding affinities than penicillin G and ampicillin. These findings suggest their potential as future antimicrobial agents. The tested compounds demonstrated efficacy against bacterial strains, particularly E. coli and S. aureus, with significant activity observed in compounds 4a, 4e, 5d, and 5e. Antioxidant activity, assessed using the DPPH method, showed bis-compounds with excellent results comparable to ascorbic acid. This encourages further studies to explore their potential. Overall, the synthesized bis-oxazolone derivatives demonstrated increased medicinal activity and high potential as future antimicrobial and antioxidant agents. Abstract:In an attempt to develop effective and safe antibacterial agents, we synthesized novel thiazinanones by combining the quinolone scaffold and the 1,3-thiazinan-4-one group by reaction between ((4-hydroxy-2-oxo-1,2-dihydroquinolin-3-yl)methylene)hydrazinecarbothioamides and 2,3-diphenylcycloprop-2-enone in refluxing ethanol in the presence of triethyl amine as a catalyst. The structure of the synthesized compounds was characterized by spectral data and elemental analysis, IR, MS, 1 H and 13 C NMR spectroscopy which showed two doublet signals for CH-5 and CH-6 and four sharp singlets for the protons of thiazinane NH, CHN, quinolone NH and OH, respectively. Also, the 13 C NMR spectrum clearly showed the presence of two quaternary carbon atoms which were assigned to thiazinanone-C-5 and C-6. All the 1,3-thiazinan-4-one/quinolone hybrids were screened for antibacterial activity. Compounds 7a , 7e and 7g showed broad spectrum antibacterial activity against most of the tested strains either G +ve or G −ve. Compound 7e is the most potent antibacterial agent against MRSA with the minimum inhibitory concentration against MRSA found to be 48 μg mL −1 compared to the drug ciprofloxacin (96 μg mL −1 ). Additionally, a molecular docking study was performed to understand the molecular interaction and binding mode of the compounds on the active site of S. aureus Murb protein. In silico docking assisted data strongly correlated with the experimental approach of antibacterial activity against MRSA.

Abstract:Background: The development of multidrug resistant strains of extended-spectrum β-lactamase-producing Escherichia coli has become a global problem; therefore, the discovery of new antibacterial agents is the only available solution. Objective: To improve and propose new compounds with antibacterial activity, the three-dimensional quantitative structure-activity relationship and molecular docking studies were carried out on Aztreonam analogs as E. coli inhibitors in DNA gyrase B. Method: This study's 3D-Quantitative structure-activity relationship model was created using on the Comparative Molecular Field Analysis and the Comparative Molecular Similarity Indices Analysis. Using the Comparative Molecular Field Analysis (Q 2 = 0.73; R 2 = 0.82), excellent predictability was achieved, and the best Comparative Molecular Similarity Indices Analysis model (Q 2 = 0.88; R 2 = 0.9). The generated model's ability to predict outcomes was assessed through external validation using a test set compound and an applicability domain technique. In this study, the steric, electrostatic, and hydrogen bond acceptor fields played a key role in antibacterial activity. Results: The results of the molecular docking revealed that the newly generated compound A6 has the highest binding affinity with DNA gyrase B. It forms 10 hydrogen bonds with amino acid residues of Asn104, Asn274, Asn132, Ser70, Ser237, Thr105, Glu273, and 2 salt bridges with amino acid residues of Ser70 and Glu273 and one pi-pi interacting with Gys271 amino acid residue in the binding site of 5G1, and this result was validated by a new assessment method. We created some novel, highly effective DNA gyrase B inhibitors based on the earlier findings, and the most accurate model predicted their inhibitory actions. The ADMET characteristics and pharmacological similarity of these novel inhibitors were also examined. Conclusion: These findings would be very beneficial in guiding the optimization process for the identification of novel drugs that can address the issue of multiple drug resistance.

Abstract:A series of new 2‐(4‐(3‐(4‐formylphenoxy)propyl)‐1H‐1,2,3‐triazol‐1‐yl)‐N‐phenylacetamides was successfully designed and synthesized via the click reaction. The alkyne agent 4‐(pent‐4‐yn‐1‐yloxy) benzaldehyde was prepared from 4‐hydroxybenzaldehyde and 5‐chloropent‐1‐yne. Meanwhile, the 2‐azido‐N‐phenylacetamides were obtained by the azidation of 2‐chloro‐N‐phenylacetamides which were formed from aromatic amine and chloroacetyl chloride. The novel compounds were characterized by FT‐IR, 1H/ 13C NMR, HRMS, MS, and elemental analysis, and evaluated for their in vitro antimicrobial activity against Acinetobacter baumannii, Carbapenem resistant pseudomonas aeruginosa, Klebsiella pneumoniae, Enterobacter aerogenes, Vancomycin resistant enterococcus faecium and Methicillin resistant staphylococcus aureus bacterial strains, as well as Candida albicans and Aspergillus niger fungal strains. Some of the compounds revealed significant potential activity compared to the reference drugs. Compound 4h showed the highest activity (MIC = 133 μM, 106 μM, 133 μM, 106 μM) against Ac. Baumannii, Klebsiella pneumoniae, Enterobacter aerogenes, and Methicillin‐resistant Staphylococcus aureus (MRSA), respectively, as antibacterial and against Candida albicans (MIC = 534 μM) and Aspergillus niger (MIC = 1,068 μM) as antifungal. Using in silico molecular docking and ADMET studies, the compounds with the highest potential activity were examined. The results showed strong receptor site binding affinity and indicated promising antimicrobial potential.

Abstract:Introduction: In the ongoing fight against bacterial resistance to antibiotics, this study focuses on synthesizing and evaluating 1,2,4-triazole derivatives to explore their potential as new antibacterial agents. 1,2,4-Triazole compounds are promising drug candidates with a wide range of therapeutic effects, including pain relief, antiseptic, antimicrobial, antioxidant, antiurease, anti-inflammatory, diuretic, anticancer, anticonvulsant, antidiabetic, and antimigraine properties. Method: The structures of all the synthesized compounds were identified using their physicochemical properties and spectral techniques, such as IR and NMR. These compounds were then evaluated in molecular docking studies against antimicrobial activity in vitro and further supported by molecular dynamics studies. Result: Compound 7, featuring a 6-chloro group on the phenyl ring, emerged as the most effective against Gram-positive S. aureus compared to the standard antibiotic ciprofloxacin. Docking studies revealed high and comparable affinities for all ten ligands, with compounds 4 and 6 showing the best-docked activity against Penicillin Acylase mutants. Further, compounds 6 and 10 displayed significant affinity against D-alanine-D-alanine ligase (DDL) from Yersinia pestis during 100 ns MD simulation. Conclusion: Notably, compound 7 demonstrated the highest binding score to the 5C1P protein, suggesting its potential as a lead molecule for the development of potent and safer antimicrobial agents. This research contributes valuable insights into addressing the escalating challenge of bacterial resistance.

Abstract:The novel 4‐quinolone‐2‐carboxylic acid derivatives are synthesized and characterized, demonstrating significant antibacterial activity against Gram‐negative bacteria, particularly P. aeruginosa and E. coli. The compound 2‐(1‐(4‐chlorobenzyl)‐4‐oxo‐1,4‐dihydroquinolin‐2‐yl)acetic acid (5a) shows strong binding energy in molecular docking studies against LpxC and PBP3 enzymes, suggesting effective mechanisms of action. Additionally, the derivatives indicate favorable for ADMET profiles, supporting their potential for oral formulations with reduced toxicity. Quinolones represent a class of potent antibacterial agents that primarily function by inhibiting bacterial topoisomerases and DNA gyrase. A crucial feature of their structure is a carboxylic acid group at the C‐3 position, which facilitates interaction with the active sites of these enzymes. Since the mid‐20th century, the emergence and proliferation of antimicrobial resistance (AMR) have posed significant challenges in society, necessitating the development of novel therapeutic agents. In this context, a new series of quinolone derivatives has been designed and synthesized based on transtorine. All synthesized compounds were tested against both Gram‐positive and Gram‐negative bacteria, as well as three fungal strains. Among the new compounds, 5a, 5d, and 5f demonstrated superior activity against Gram‐negative bacteria, with 5a showing notable effectiveness against P. aeruginosa compared to ciprofloxacin and norfloxacin. However, none of the compounds exhibited activity against fungal strains. Molecular docking studies revealed that compound 5a inhibited five bacterial proteins, primarily targeting bacterial cell wall synthesis. Additionally, ADMET parameters indicated that the promising compounds are suitable for oral administration with limited penetration across the blood–brain barrier compared to existing standards. This research indicates a potential pathway for developing new quinolone‐based antibiotics in the fight against AMR.

Abstract:The limit ability of traditional antibiotics to treat drug resistant bacteria calls for new therapeutic alternatives. A class of unique sulfanilamide aminophosphonates as new potential agents against microbes was synthesized by one‐pot three‐component reaction. Noticeably, fluorobenzyl derivative 5d (MIC = 2 μg/mL) was active against drug resistant E. coli infection and exerted no obvious toxicity towards human mammalian cells. Compound 5d also displayed good anti‐biofilm activity and low possibility to induce drug resistance. Mechanism investigation elucidated that molecule 5d could disrupt E. coli membrane through generation of reactive oxygen (ROS) and then intercalate into deoxyribonucleic acid (DNA) to form a steady 5d‐DNA complex, which led to bacterial death. These results indicated that sulfanilamide aminophosphonates would shed light on developing novel potential antibacterial agents.This article is protected by copyright. All rights reserved.
