Abstract:Escherichia coli has been one of the most highlighted pathogens isolated from clinical cases of mastitis. Unfortunately, the deficiency of appropriate antimicrobial stewardship and overuse of antimicrobial agents have increased antimicrobial resistance in animal isolates. The current study aimed to demonstrate the prevalence of extended-spectrum beta-lactamase (ESBL) production, resistance genes, and multi drug resistant (MDR) among E. coli isolates from cases of clinical mastitis. For this purpose, 40 E. coli isolates were collected from clinical mastitis milk samples from five farms in Mashhad, Iran. The antimicrobial susceptibility testing (AST) was applied to 15 antimicrobial agents of veterinary and human medicine interest. Also, ESBL production was evaluated using a doubledisc synergy test (DDST). The distribution of 20 resistance genes was sought among E. coli isolates by six multiplexPCR and three uniplex-PCR assays. The highest sensitivity was identified against imipenem and amikacin (100%). On the other hand, the highest resistance was observed for tetracycline and trimethoprim-sulfamethoxazole (70% - 72.5%), respectively. According to the AST and DDST tests, one isolate was confirmed as ESBL-producing and MDR. In addition, the most frequent resistance genes were blaTEM and AmpC (100% each). The qnrA encoding resistance to quinolones was similarly prevalent and detected in 50% of the isolates. In conclusion, at least three resistant genes were detected in 28 isolates (70%), but the majority of isolates were sensitive against most of the tested antibiotics. This fact might relate to the low expression of these genes within the isolates. The horizontal gene transfer of the present genes may confer resistance to other related bacterial species in humans or domestic animals.

Abstract:Trueperella pyogenes is an opportunistic pathogen causing important diseases including mastitis and metritis in domestic animals such as dairy cows leading to prominent economic losses in food production industry. The aim of this study was to investigate bacterial species, antimicrobial susceptibility, and presence of virulence factor genes and genotyping of T. pyogenes isolates associated with summer mastitis cases from 22 different farms around Tehran, Iran. Fifty-five percent of dairy cows with clinical mastitis symptoms was infected by T. pyogenesis indicated that this pathogen is the most important contributor to clinical mastitis in dairy cows in the present study. A significant correlation was illustrated between presence of virulence factor genes of isolated pathogen, biochemical patterns, and the utter infected types. Multidrug resistance susceptibility observed between isolates indicated the important need for prudent use of antimicrobials in treatment of mastitis caused by T. pyogenes and increased concerning of consumer health associated with recent problems of antimicrobial resistance. The categorization of isolates was implemented into seven different clonal related types by COX-PCR at 80% of similarity cutoff with significance relationship to clonal types, CAMP test result and sampling time and biochemical profile. Regarding to the results obtained at the present study, T. pyogenes can be considered as an important typically cause of purulent and acute form of clinical bovine mastitis and loss of dairy productivity. Further studies with more sample size and high-throughput omic methods in various sampling time and areas are suggested for study of this pathogen precisely.

Abstract:This study was conducted to determine genetic diversity and antimicrobial susceptibility profiles of Staphylococcus aureus recovered from bovine mastitis in Zhejiang Province, China. Out of 3178 quarter milk samples from 846 lactating cows, among which 459 cows (54.3%) were found HMT positive, 890 quarters (28%) were found having subclinical mastitis. From 75 representative S. aureus isolates, 16 distinct types were identified by pulsed-field gel electrophoresis (PFGE). Four major PFGE types (A, B, C, and D) accounted for 82.7% of all isolates, and type A (41.3%) was observed in multiple herds across the studied areas. Each region was found to have a predominant type: Hangzhou type A (64.1%), Ningbo type C (34.5%) and type B (23.1%), Jinhua type D (53.3%), and Taizhou type C (62.5%). Results of antimicrobial susceptibility tests showed that 90.7% of the isolates were resistant to at least one antimicrobial. Resistance to penicillin and ampicillin (77.3%), tetracycline (60.0%), or erythromycin (48.0%) was observed. The bacteria resistant to multiple antibiotics such as penicillin, ampicillin, tetracycline, and erythromycin were commonly found. The information obtained from this study is useful for designing specific control programs for bovine S. aureus mastitis in this region.

Abstract:In dairy industry, bovine mastitis is the most prevalent disease, which reduces milk production and causes economic losses. This study was conducted to estimate the prevalence of Mycoplasma bovis and some bacteria causing mastitis in dairy farms and partial sequencing of 16SrRNA target genes and Quinolones Resistance Determining Regions (QRDRs) ( gyr A and par C) in M. bovis isolates. 370 milk samples were obtained from farms located in villages in Fayoum governorate, Egypt. The examined milk samples (8,91%) were positive for the California mastitis test (CMT). Multiplex RT-PCR was used for the recognition of microorganisms causing mastitis ( Staphylococcus (S.) aureus, Streptococcus species (spp.), Escherichia (E.) coli , and Mycoplasma (M.) bovis ) from mastitic milk. The results revealed that E. coli was the most predominant (84.8%) followed by S. aureus (81.8%) while M. bovis was the lowest one (51.5%). Mixed infection with two or more mastitic bacterial agents was also identified. All 33 examined mastitic milk samples were diagnosed with mixed infection with E. coli , S. aureus, Streptococcus spp. and M. bovis (36.36%), E. coli and S. aureus (21.21%), and rephrase E. coli , M. bovis, and Streptococcus spp. (6.06%). The sequence analysis of M. bovis 16SrRNA genes illustrated a high similarity of examined isolates to strains previously deposited in the GenBank recovered from the same locality. The gyr A amino acids showed no substitution but showed 100% similarity with M. bovis isolates worldwide. However, the amino acid sequence of par C, showed substitution at positions 2 (Gln to Arg) (CAG >>CGT), 75 (Ile to Ser) (ATT>>AGC), and 79 (Asn to Asp) (AAC>>GAT). Sequence results can lead to the creation of appropriate treatment and control measures for M. bovis , while multiplex RT-PCR, can be exploited as a standard diagnostic method for major mastitis pathogens.

Abstract:Background and Aim: Mastitis is a common and economically important disease in dairy cattle. It remains one of the most common reasons for the extensive use of antimicrobials in dairy farms leading to the emergence of antimicrobial-resistant pathogens. The aim of this study was to determine the patterns of antimicrobial resistance of Escherichia coli isolates from bovine mastitis and to identify prominent antimicrobial resistance and virulence genes among isolated strains. Materials and Methods: Antimicrobial susceptibility testing against six antibiotic groups, including tetracyclines, aminoglycosides, beta-lactams, macrolides, sulfonamides, and fluoroquinolones was performed using the disk diffusion method. PCR was performed on resistant isolates to detect resistance and virulence genes using commercially available primers. Results: Out of 216 milk samples cultured, 14 samples yielded E. coli isolates. All isolates (100%) were resistant to ampicillin, amoxicillin, procaine penicillin, streptomycin, oxytetracycline, and sulfamethoxazole-trimethoprim. Only one isolate (7%) was sensitive to gentamicin, and all isolates (100%) were sensitive to enrofloxacin and ciprofloxacin. All isolates carried at least one resistance gene against one or more of the major antibiotic groups. All isolates carried the ereA, tetG, tetE, and tetB genes, followed by tetA (93%), ampC (86%), strA (86%), sul1 (78%), tetD (71%), tetC (57%), aadA (57%), and strB (36%). The lowest percentage of isolates carried bla1 (17%) and bla2 (12%) genes, and none of the isolates carried the qnrA gene. Most of the isolates (93%) carried the Shiga toxin 1 virulence gene, followed by complement resistance protein (79%), intimin (64%), Shiga toxin 2 (36%), cytotoxic necrotizing factor (35%), aerotaxis receptor (21%), and type 1 fimbriae (15%). Conclusion: Results of this study indicate that the high percentages of E. coli isolate from bovine mastitis are resistant to two or more of the major antibiotic groups, irrespective of the presence or absence of relevant resistance or virulence genes.

Abstract:Mastitis is considered one of the most widespread infectious disease of cattle and buffaloes, affecting dairy herds. The current study aimed to characterize the Staphylococcus aureus isolates recovered from subclinical mastitis animals in Pothohar region of the country. A total of 278 milk samples from 17 different dairy farms around two districts of the Pothohar region, Islamabad and Rawalpindi, were collected and screened for sub clinical mastitis using California Mastitis Test. Positive milk samples were processed for isolation of Staphylococcus aureus using mannitol salt agar. The recovered isolates were analyzed for their antimicrobial susceptibility and virulence genes using disc diffusion and PCR respectively. 62.2% samples were positive for subclinical mastitis and in total 70 Staphylococcus aureus isolates were recovered. 21% of these isolates were determined to be methicillin resistant, carrying the mecA gene. S. aureus isolates recovered during the study were resistant to all first line therapeutic antibiotics and in total 52% isolates were multidrug resistant. SCCmec typing revealed MRSA SCCmec types IV and V, indicating potential community-acquired MRSA (CA-MRSA) transmission. Virulence profiling revealed high prevalence of key genes associated with adhesion, toxin production, and immune evasion, such as hla, hlb, clfA, clfB and cap5 . Furthermore, the Panton-Valentine leukocidin (PVL) toxin, that is often associated with recurrent skin and soft tissue infections, was present in 5.7% of isolates. In conclusion, the increased prevalence of MRSA in bovine mastitis is highlighted by this study, which also reveals a variety of virulence factors in S. aureus and emphasizes the significance of appropriate antibiotic therapy in combating this economically burdensome disease.

Unravelling the complex mechanisms of multidrug resistance in bovine mastitis pathogens: Insights into antimicrobial resistance genes, biofilm dynamics, and efflux systems

Abstract:Mastitis remains a paramount economic threat to dairy livestock, with antibiotic resistance severely compromising treatment efficacy. This study provides an in-depth investigation into the multidrug resistance (MDR) mechanisms in bacterial isolates from bovine mastitis, emphasizing the roles of antimicrobial resistance genes (ARGs), biofilm formation, and active efflux systems. A total of 162 Staphylococci, eight Escherichia coli, and seven Klebsiella spp. isolates were obtained from 215 milk samples of clinical and subclinical mastitis cases. Antibiotic susceptibility testing identified Twenty Staphylococci (12.35 %), six E. coli (75 %) and seven Klebsiella (100 %) identified as MDR displaying significant resistance to β-lactams and tetracyclines The Multiple Antibiotic Resistance (MAR) index of these isolates ranged from 0.375 to 1.0, highlighting extensive resistance. Notably, 29 of the 33 MDR isolates produced biofilms on Congo red agar, while all exhibited biofilm formation in the Microtitre Plate assay. Critical ARGs (blaZ, blaTEM, blaCTX-M, tetM, tetA, tetB, tetC, strA/B, aadA) and efflux pump genes (acrB, acrE, acrF, emrB, norB) regulating active efflux were identified. This pioneering study elucidates the synergistic contribution of ARGs, biofilm production, and efflux pump activity to MDR in bovine mastitis pathogens. To our knowledge, this comprehensive study is the first of its kind, offering novel insights into the complex resistance mechanisms. The findings underscore the imperative need for advanced antibiotic stewardship and strategic interventions in dairy farming to curb the rise of antibiotic-resistant infections, thereby protecting both animal and public health.
Genetic Diversity, Biofilm Formation, and Antibiotic Resistance of Pseudomonas aeruginosa Isolated from Cow, Camel, and Mare with Clinical Endometritis

Abstract:Pseudomonas aeruginosa is a ubiquitous opportunistic bacterium that causes diseases in animals and humans. This study aimed to investigate the genetic diversity, antimicrobial resistance, biofilm formation, and virulence and antibiotic resistance genes of P. aeruginosa isolated from the uterus of cow, camel, and mare with clinical endometritis and their drinking water. Among the 180 uterine swabs and 90 drinking water samples analysed, 54 (20%) P. aeruginosa isolates were recovered. Isolates were identified biochemically to the genus level by the automated Vitek 2 system and genetically by the amplification of the gyrB gene and the sequencing of the 16S rRNA gene. Multilocus sequence typing identified ten different sequence types for the P. aeruginosa isolates. The identification of ST2012 was significantly (p ≤ 0.05) higher than that of ST296, ST308, ST111, and ST241. The isolates exhibited significantly (p ≤ 0.05) increased resistance to piperacillin (77.8%), ciprofloxacin (59.3%), gentamicin (50%), and ceftazidime (38.9%). Eight (14.8%) isolates showed resistance to imipenem; however, none of the isolates showed resistance to colistin. Multidrug resistance (MDR) was observed in 24 isolates (44.4%) with a multiple antibiotic resistance index ranging from 0.44 to 0.77. MDR was identified in 30 (33.3%) isolates. Furthermore, 38.8% and 9.2% of the isolates exhibited a positive extended-spectrum-β-lactamase (ESBL) and metallo-β-lactamase (MBL) phenotype, respectively. The most prevalent β-lactamase encoding genes were blaTEM and blaCTX-M, however, the blaIPM gene was not detected in any of the isolates. Biofilm formation was observed in 49 (90.7%) isolates classified as: 11.1% weak biofilm producers; 38.9% moderate biofilm producers; 40.7% strong biofilm producers. A positive correlation was observed between the MAR index and biofilm formation. In conclusion, the results highlighted that farm animals with clinical endometritis could act as a reservoir for MDR and virulent P. aeruginosa. The emergence of ESBLs and MBLs producing P. aeruginosa in different farm animals is a public health concern. Therefore, surveillance programs to monitor and control MDR P. aeruginosa in animals are required.

Abstract:To investigate the drug resistance and the correlation between phenotypes and genotypes of bovine mastitis pathogens,70 Staphylococcus aureus were isolated from mastitic cases in the major dairy regions of Zhejiang Province,and they were studied for their susceptibility to 13 commonly used antimicrobial agents,and the genes of mecA,aac(6′)/aph(2″),erm and tetM in connection with penicillin G,gentamicin,erythromycin and tetracycline were detected by PCR.The results indicated that the S.aureus isolates had serious resistance to antibacterial agents,and the phenotypes and genotypes were closely associated.Of the 70 S.aureus,the bacteria mainly resistant to penicillin,gentamicin,erythromycin and tetracycline were 90.0%,67.1%,50.0% and 60.0%,respectively;92.9%,77.1%,48.6% and 61.4% of the bacteria were detected to have the genes of mecA,aac(6′)/aph(2″),erm and tetM.The compatibility between the phenotypes and genotypes were 97.1%,70.0%,87.1% and 81.4%,respectively.
Phenotypic and Genotypic Characterization of the Virulence Factors and Antimicrobial Resistance of Enterobacteriaceae Isolates Associated with Clinical Mastitis in Dairy Cattle

Abstract:Mastitis is a prevalent disease in dairy cattle. One of its important etiological agents is the species belonging to the Enterobacteriaceae family. Thus, this study aims to characterize the virulence and multi-drug resistance (MDR) profiles of Enterobacteriaceae strains previously isolated from dairy cattle with clinical mastitis in Region 4-A, Philippines. Results showed that 60% of Klebsiella pneumoniae and 100% of the Proteeae tribe (Proteus spp., Providencia spp., and Morganella spp.) exhibited hemolytic activity. Hemolysin-coding gene, viz. hpmA, was suspected to contribute to the hemolytic activity of all Proteus spp. Biofilm formation was observed in several isolates and mrkD, ireA, ucaA, atfA, and ureG genes were expected to be accountable for this virulence trait. All Enterobacteriaceae strains were classified as MDR pathogens. All isolates exhibited resistance to erythromycin, penicillin, clindamycin, and lincomycin. Resistance to streptomycin and tetracycline was also exhibited by a significant number of isolates and the resistance genes (rrs and tetK) responsible for this resistance were most frequently detected. Only one isolate of M. morganii harbored integron-related gene intI2. The characterization of these strains has significant health and economic implications. Severe virulence and drug resistance of these strains pose a challenge in the management and treatment of intramammary infections in dairy farms. Being reservoirs of antimicrobial resistance-associated genes, these strains pose a threat to the food chain.
Molecular Epidemiology and Antimicrobial Resistance of Outbreaks of Klebsiella Pneumoniae Clinical Mastitis in Chinese Dairy Farms

Abstract:Klebsiella pneumoniae is an opportunistic pathogen that causes serious infections in humans and animals. However, the availability of epidemiological information on clinical mastitis due to K. pneumoniae is limited. To acquire new information regarding K. pneumoniae mastitis, data were mined about K. pneumoniae strains on dairy cattle farms (farms A to H) in 7 Chinese provinces in 2021. Hypermucoviscous strains of K. pneumoniae were obtained by the string test. MICs of antimicrobial agents were determined via the broth microdilution method. Ten antimicrobial resistance genes and virulence genes were identified by PCR. The prevalence of K. pneumoniae was 35.91% (65/181), and 100% of the bacteria were sensitive to enrofloxacin. Nine antimicrobial resistance genes and virulence genes were identified and compared among farms. The hypermucoviscous phenotype was present in 94.44% of isolates from farm B, which may be a function of the rmpA virulence gene. Based on these data, the multidrug-resistant strains SD-14 and HB-21 were chosen and sequenced. Genotypes were assayed for K. pneumoniae isolates from different countries and different hosts using multilocus sequence typing (MLST). Ninety-four sequence types (STs) were found, and 6 STs present a risk for spreading in specific regions. Interestingly, ST43 was observed in bovine isolates for the first time. Our study partially reveals the current distribution characteristics of bovine K. pneumoniae in China and may provide a theoretical basis for the prevention and treatment of bovine K. pneumoniae mastitis. IMPORTANCE K. pneumonia is ubiquitous in nature and infects a wide range of hosts, including animals, and humans. It is one of the leading inducements of clinical mastitis (CM) in dairy cows, a prevalent and costly disease that is predominantly associated with bacterial infection. In general, CM caused by Gram-negative bacteria is more difficult to cure than that associated with Gram-positive pathogens, with an average cost per case of 211.03 U.S. dollars (USD) for Gram-negative bacterial infections compared with 133.73 USD for Gram-positive bacterial CM cases. After Escherichia coli, K. pneumoniae is the second most common Gram-negative cause of bovine CM, but it is the most detrimental in terms of decreased milk yield, discarded milk, treatment costs, death, and culling. In view of the economic implications of K. pneumoniae infection in dairy farming, research into population structure and antibiotic resistance is particularly important.
A culture-, amplification-independent, and rapid method for identification of pathogens and antibiotic resistance profile in bovine mastitis milk

Abstract:Introduction Rapid and accurate diagnosis of causative pathogens in mastitis would minimize the imprudent use of antibiotics and, therefore, reduce the spread of antimicrobial resistance. Whole genome sequencing offers a unique opportunity to study the microbial community and antimicrobial resistance (AMR) in mastitis. However, the complexity of milk samples and the presence of a high amount of host DNA in milk from infected udders often make this very challenging. Methods Here, we tested 24 bovine milk samples (18 mastitis and six non-mastitis) using four different commercial kits (Qiagens’ DNeasy ® PowerFood ® Microbial, Norgens’ Milk Bacterial DNA Isolation, and Molzyms’ MolYsisTM Plus and Complete5) in combination with filtration, low-speed centrifugation, nuclease, and 10% bile extract of male bovine (Ox bile). Isolated DNA was quantified, checked for the presence/absence of host and pathogen using PCR and sequenced using MinION nanopore sequencing. Bioinformatics analysis was performed for taxonomic classification and antimicrobial resistance gene detection. Results The results showed that kits designed explicitly for bacterial DNA isolation from food and dairy matrices could not deplete/minimize host DNA. Following using MolYsisTM Complete 5 + 10% Ox bile + micrococcal nuclease combination, on average, 17% and 66.5% of reads were classified as bovine and Staphylococcus aureus reads, respectively. This combination also effectively enriched other mastitis pathogens, including Escherichia coli and Streptococcus dysgalactiae . Furthermore, using this approach, we identified important AMR genes such as Tet (A), Tet (38 ), fosB-Saur, and blaZ. We showed that even 40 min of the MinION run was enough for bacterial identification and detecting the first AMR gene. Conclusion We implemented an effective method (sensitivity of 100% and specificity of 92.3%) for host DNA removal and bacterial DNA enrichment (both gram-negative and positive) directly from bovine mastitis milk. To the best of our knowledge, this is the first culture- and amplification-independent study using nanopore-based metagenomic sequencing for real-time detection of the pathogen (within 5 hours) and the AMR profile (within 5–9 hours), in mastitis milk samples. These results provide a promising and potential future on-farm adaptable approach for better clinical management of mastitis.

Antibiotic resistance of microorganisms isolated from cows with subclinical mastitis

Abstract:Cow mastitis is a polyetiological disease involving pathogenic microorganisms. Antibiotics are used to treat sick animals, which contributes to the development of resistant clones of bacteria, and consumption of the corresponding milk leads to the spread of antibiotic-resistant microorganisms. The purpose of the work is to determine strains of antibiotic-resistant bacteria in the milk of cows suffering from subclinical mastitis. Bacteriological studies of mammary gland secretion samples were performed by inoculating the sediment obtained after centrifugation of the examined material on an agarised nutrient medium (with 5% ram erythrocytes). The study of antimicrobial sensitivity was performed by the disk diffusion method. It was established that pathogens with multiple resistance to antibiotics circulate among cows suffering from mastitis on the farm, which indicates their indiscriminate use in animal therapy schemes, in particular mastitis. It was found that the overall level of phenotypic resistance to antimicrobial agents of the examined isolates was within 19-33% (in Staphylococcus spp. – 19.9%, in E. coli – 33.2%, in Enterococcus spp. – 32.5%). The analysis of the results allowed assigning the culture study to the category of multiresistant: 31.6% of cultures of Staphylococcus spp. demonstrated resistance to five groups of antibiotics; 54.5% of Escherichia cultures – up to five groups of antibiotics; 37.5% of cultures of Enterococcus spp. resistant to three groups of antibiotics. It was established that 100% of the studied cultures of Staphylococcus spp. are sensitive to netilmicin, sparfloxacin, gatifloxacin and clarithromycin. Therewith, 100% of the examined cultures of Enterococcus spp. sensitive to gentamicin, norfloxacin, and nitrofurantoin, and 100% of the examined E. coli cultures were sensitive to cephalothin, meropenem, gentamicin, tobramycin, amikacin, nitroxoline, and gatifloxacin. Thus, dairy cows suffering from subclinical mastitis are a source of bacteria with multiple resistance to antibiotics and carry risks for consumers due to the consumption of dairy products contaminated with pathogens of zoonotic diseases, which requires proper control by veterinary service specialists
Molecular characteristics and antibiotic resistance of Staphylococcus aureus and Staphylococcus haemolyticus isolated from bovine mastitis

Abstract:Staphylococci are classified as one of the pathogens causing bovine mastitis that can pose not only an economic loss to the dairy farms, but a serious public-health threat based on their zoonotic potential. We focused to monitor phenotypes of the isolated strains of Staphylococcus aureus (S. aureus) and Staphylococcus haemolyticus (S. haemolyticus) from milk of cows with clinical mastitis, including antibiotic resistance, biofilm forming ability and the presence of biofilm- and toxin- related genes. From a total of 191 milk samples were identified as S. aureus - 12% (22 isolates) and S. haemolyticus - 6% (12 isolates). Automatic interpreted reading of the antibiogram evaluated potentially 12 isolates as methicillin-resistant S. aureus and methicillin-resistant coagulase-negative Staphylococci. Genotypically, the isolates were positive for blaZ and negative for mecA and mecC. Others important mechanisms were inducible macrolide-lincosamide-streptogramin B (iMLSB) resistance with presence of msrA, ermC, vgaA. The most detected biofilm-associated and toxins genes were clfA, sdrD, sdrE, fnbpB, bbp, isdA, isdB, hla and see. S. aureus isolates were subjected to spa typing. It turned out that despite the strains coming from different farms, they were either resistant or sensitive to antibiotic, were all of the same spa-type t 10035. Our findings revealed the presence iMLSB, which, to our best knowledge, were described in Slovakian bovine staphylococci rarely. The majority of isolates were multidrug-resistant and carried multiple virulence genes, posing a potential public-health risk.
Seasonal Prevalence and Antibiogram Profile of Bacterial Isolates from Bovine Mastitis

Abstract:The current study examines the prevalence and seasonal occurrence of major bacterial pathogens and effect of different antibiotics on bacteria isolated from 935 bovine mastitic milk samples in northern region of India for characterization and culture sensitivity against different antibiotics. A major proportion of mastitis samples- 412 (44%) were recorded during rainy season, while, samples in equal shares, 276 (28%) and 247 (27%) were documented in winter and summer seasons, respectively. Out of total 935 mastitic milk samples, 889 (95%) samples showed microbial growth, while, 46 (5%) samples were found negative for any bacterial growth. Among total isolated bacteria, Gram positive- 471 (53%) shared a major proportion, followed by Gram negative 341 (38%), while a small part of 53 (6%) and 24 (2%) samples yielded mixed unidentified cultures and Candida species, respectively. In antibiogram study, gentamicin (91.21%), ciprofloxacin (89.60%), enrofloxacin (88.28%) and tetracycline (71.30%) were found to be highly effective antibiotics, while, penicillin (86%), colistin (83.30%), cloxacillin (78.62%), amoxycillin (70.71%) and ampicillin (62.51%) showed least effect against both Gram positive and negative bacteria. The present study showed that there was close association between season, bacterial pathogens and occurrence of of bovine mastitis. Overall, Staphylococcus spp., Streptococcus spp. and E. coli contributed as major mastitis dweller bacteria.

Genomic diversity and nutritional analysis of multi-drug resistant extended spectrum β-lactamase Producing- Klebsiella pneumoniae genes isolated from mastitic cattle milk in district peshawar, Pakistan

Abstract:The increasing incidence of resistance extended spectrum-beta lactamase (ESBL) producing Klebsiella pneumonia become worldwide issue. The current study aimed to determine the genomic diversity of ESBL-producing K. pneumoniae in milk samples collected from cows with mastitis as well as their antibiotic sensitivity profiles and genetic identification in Peshawar, Pakistan. The california mastitis test (CMT) was initially used to verify the presence for mastitis in 700 collected milk samples. The molecular identification of the 16SrRNA gene confirmed 120/700 (17.14 %) propagation of K. pneumonia. Out of these isolates MDR ESBL-producing isolates were 60/120 (50 %). The lactose were found (M = 3.96 ± 0.28, SD = 2.19), followed by fats (M = 3.12 ± 0.11, SD = 0.90), protein (M = 5.97 ± 0.24, SD = 1.84), sodium (M = 55.74 ± 2.07, SD = 15.81), potassium (M = 138.5 ± 1.53, SD = 11.71), chloride (M = 0.74 ± 0.03, SD = 0.24), calcium (M = 10.27 ± 0.31, SD = 2.42), and chlorine (M = 2.80 ± 0.22, SD = 1.70), respectively. Amikacin (80 %), ceftazidime (71 %), and tetracycline (71 %) were shown to be the most effective antimicrobials against all of the isolates. The occurrence of the blaSHV gene was observed at 56.00 % whereas the blaTEM gene and blaCTX-M gene were 36.00 %, and 30.00 %. The distribution of blaCTX-M subgroup genes was followed by blaCTX-M-1 (38.00 %), blaCTX-M-9 (22.20 %), and blaCTX-M-15 (61.10 %). Co-occurrence of blaCTX-M+ blaSHV was (15.00 %), blaCTX-M+ blaTEM were (6.60 %), and blaSHV + blaTEM were (10.00 %), respectively. The inappropriate, prolonged and common use of antibiotics may apply selective pressure for propagation and the occurrence of resistant isolates.
