Abstract:Changes in intestinal flora affect the health and cause metabolic diseases of the host. The extent to which the impact of different changes in intestinal flora would have on the metabolism of an individual has not been reported. This study aims to investigate the effect of different changes in intestinal flora on the metabolism of Sprague-Dawley (SD) normal rats' individuals. Forty-eight SD rats were randomly divided into 6 groups (8 rats per group), which were treated with normal saline, probiotics, nonpathogenic Escherichia coli, Salmonella enteritidis, gentamicin, and magnesium sulfate, respectively. After 7 days, the ileum of each group of rats was collected and real-time polymerase chain reaction was used to analyze the composition of intestinal flora. And gas chromatography/mass spectrometry (GC/MS) was used to analyze plasma metabolic profile. The results revealed that the decrease in alanine content in the probiotics group was statistically significant, while the alanine content in the nonpathogenic Escherichia group increased significantly. Alanine, leucine, isoleucine, and serine decreased significantly in the Salmonella group. Proline and butyric acid decreased significantly in the gentamicin group. The principal component analysis showed significant differences in the Salmonella group compared with other test groups. Overall, the most significant metabolic changes were observed in SD rats in the Salmonella group, while a great similarity was observed in the probiotics, Escherichia group, and gentamicin groups compared with the normal group. Changes in intestinal flora had a certain impact on the metabolism in SD rats, especially on amino acid levels.
[Relaxant Effect of Astragalus Membranaceus on Smooth Muscle Cells of Rat Thoracic Aorta].

Abstract:OBJECTIVE:To investigate the effect of Astragalus membranaceus(AM) on vascular circles and the underlying mechanisms.METHODS:The study was performed with the model of isolate rat thoracic aorta rings in organ bath. When the endothelium of rat thoracic aorta was removed,the effect of accumulated AM on aorta rings in resting tension, or pre-constricted with KCl, or pre-constricted with phenylephrine (PE) was observed. And to explove the mechanism, the aorta rings were incubated with Ca(2+)-free medium alone, or Ca(2+)-free medium plus heparin, or propranolol alone before pre-contraction with PE.RESULTS:AM had no significant effects on aorta rings in resting tension or pre-constricted with KCl. When the concentration of AM was cumulated to 10(-1), 3 x 10(-1),10(0), 3 x 10(0) g/L, it caused concentration-dependent relaxation while aorta rings were pre-constricted with PE(3 x 10(-7)mol/L), compared with the control [(90.4 +/-4.2)% compared with (94.7 +/-2.4)%,(86.1 +/-5.0)% compared with (92.6 +/-3.2)%, (82.3 +/-5.9)% compared with (90.4 +/-3.6) %, (78.3 +/-6.0)% compared with (88.1 +/-4.0)%]. This effect was not inhibited by Ca(2+)-free medium or propranolol alone. However, the effect was attenuated by the co-incubation with heparin and Ca(2+)-free medium [without heparin:(76.2+/-4.3)% compared with (92.3 +/-5.9)%, with heparin: (95.3+/-0.5)% compared with (95.1+/-0.6)%].CONCLUSION:The results indicate that AM can relax the rat thoracic aorta rings without endothelium. The mechanism may include the inhibition of intracellular calcium ions release by the 1,4,5-triphosphate inositol-receptor-dependent pathway in vascular smooth muscle cells.

Abstract:Evidences suggest that the homeostasis of gut microbiota is among the most important factors for maintaining the physical and mental health of the host. Among the multiple factors affecting the homeostasis of gut microbiota, diet is one of the decisive factors. Bioactive peptides derived from protein hydrolyzed by protease or fermented by microorganism have many physiological activities that their parent proteins do not have. Currently, bioactive peptides attract more and more attention due to their bidirectional interaction with gut microbes. It has been reported that some bioactive peptides could alter the composition of gut microbiota by influencing the intestinal microenvironment. Meanwhile, quite a few bioactive peptides that are released by gut microbes or intestinal cells could resist the pathogenic bacteria to sustain the homeostasis of gut microbiota. In this review, some exogenous bioactive peptides derived from food and some endogenous bioactive peptides released from intestinal cells or microbes were discussed to summary their effects on the modulation of gut microbiota. This review is expected to provide new ideas for related research, and as well to promote the application of bioactive peptides in the fields of food and medicine.

Abstract:Gut health has significant implications for swine overall health status and nutrient utilization, due to its various functions including digestion and absorption of nutrients, secretion of mucins and immunoglobulins, and selective barrier protection against harmful antigens and pathogens. Both the basic anatomical structure of the gut (such as epithelial cells) and its luminal microbiota play important roles for maintaining gut health and functions. The interactions between epithelial cells and luminal microbiota have significant impact on host nutrition and health through the metabolism of dietary components. Amino acids, which are major nutrients for pigs, are not only obligatory for maintaining the intestinal mucosal mass and integrity, but also for supporting the growth of microorganisms in the gut. Dietary amino acids are the major fuel of the small intestinal mucosa. Particularly, glutamate, glutamine, and aspartate are the major oxidative fuel of the intestine. Emerging evidence shows that arginine activates the mTOR signaling pathway in the small intestine. Utilization of glycine by the small intestinal mucosa to synthesize glutathione is a very important physiological pathway, and the role of glycine as a powerful cytoprotectant has also been recognized. The major end products of methionine and cysteine metabolism are glutathione, homocysteine and taurine, which play important roles in the intestinal immune and anti-oxidative responses. Threonine is highly utilized by the gut and is particularly important for mucin synthesis and maintenance of gut barrier integrity. Moreover, either a deficiency or an excess of dietary threonine can reduce the synthesis of intestinal mucosal proteins and mucins in young pigs. Various new functions of amino acids on gut health and functions have been discovered in recent years. Thus, this review is to provide some up-to-date knowledge for industry application of dietary amino acids in order to enhance swine gut health and functions, and also it is to provide a comprehensive reference for further scientific research in this regard.

Effect of Chemical Drugs on Electrical Physiology Characteristics of the Isolated Smooth Muscle of Rabbits Small Intestine

Abstract:To observe the effective mechanism of chemical durgs on the isolated smooth muscle of rabbits small intestine,the duodenum,jejunum and ileum smooth muscle were isolated by the routine experimental method from rabbits.The automatic contracting characteristic and changing pattern were recorded before and after drug taking.The results showed that: acetylcholine and histamine phosphate of different concentrations had a strengthening effect on automatic contracting activity of isolated smooth muscle and there were significant differences in frequency and scope before and after drug taking(P0.01),and had dose-response dependent relationship.Adrenalinum and atropine of different concertration had a restraining effect on contracting frequency and scope of isolated smooth muscle of small intestine(P0.01).The function in duodenum was the strongest,the next was jejunum and ileum the last.

Abstract:Consumption of a high-protein diet increases protein entry into the colon. Colonic microbiota can ferment proteins, which results in the production of protein fermentation end-products, like polyamines. This review describes the effects of polyamines on biochemical, cellular and physiological processes, with a focus on the colon. Polyamines (mainly spermine, spermidine, putrescine and cadaverine) are involved in the regulation of protein translation and gene transcription. In this, the spermidine-derived hypusination modification of EIF5A plays an important role. In addition, polyamines regulate metabolic functions. Through hypusination of EIF5A, polyamines also regulate translation of mitochondrial proteins, thereby increasing their expression. They can also induce mitophagy through various pathways, which helps to remove damaged organelles and improves cell survival. In addition, polyamines increase mitochondrial substrate oxidation by increasing mitochondrial Ca2+-levels. Putrescine can even serve as an energy source for enterocytes in the small intestine. By regulating the formation of the mitochondrial permeability transition pore, polyamines help maintain mitochondrial membrane integrity. However, their catabolism may also reduce metabolic functions by depleting intracellular acetyl-CoA levels, or through production of toxic by-products. Lastly, polyamines support gut physiology, by supporting barrier function, inducing gut maturation and increasing longevity. Polyamines thus play many roles, and their impact is strongly tissue- and dose-dependent. However, whether diet-derived increases in colonic luminal polyamine levels also impact intestinal physiology has not been resolved yet.

Abstract:Undigested dietary and endogenous proteins, as well as unabsorbed amino acids, can move from the terminal part of the ileum into the large intestine, where they meet a dense microbial population. Exfoliated cells and mucus released from the large intestine epithelium also supply nitrogenous material to this microbial population. The bacteria in the large intestine luminal fluid release amino acids from the available proteins, and amino acids are then used for bacterial protein synthesis, energy production, and in other various catabolic pathways. The resulting metabolic intermediaries and end products can then accumulate in the colorectal fluid, and their concentrations appear to depend on different parameters, including microbiota composition and metabolic activity, substrate availability, and the capacity of absorptive colonocytes to absorb these metabolites. The aim of the present review is to present how amino acid-derived bacterial metabolites can affect microbial communication between both commensal and pathogenic microorganisms, as well as their metabolism, physiology, and growth.

Abstract:Common amino acid nutrients mainly contain glutamine, arginine, leucine, methionine and cysteine, which are not only the components participating in body protein synthesis, but also regulate the patients' immune system and metabolism. Glutamine can improve the intestinal barrier, reduce inflammatory reaction, and promote immunity recovery, but the clinical effects of different patients with different diseases are still lack of clear conclusions. The catabolism of arginine can produce NO, promoting the inflammatory reaction, and can also generate ornithine, alleviating inflammatory reaction and promoting wound healing. Two competing ways coexist, but the specific effects on different diseases have no clear conclusions yet. Leucine promotes muscle protein synthesis mainly through mTOR pathway, however, the influence on metabolism is still debating. Sulfur-containing amino acids methionine and cysteine can promote the synthesis of connective tissue and collagen conducive to wound healing, and their beneficial effects on lipid metabolism are of value. The purpose of this review is to cover potential beneficial physiological mechanisms of amino acid nutrients, to describe their results of clinical applications and to evaluate the interactions among individual nutrients or between individual nutrients and body.

Abstract:Objective To investigate the effects of Puerarin on constractive activity of small intestine smooth muscle of alcoholism rabbit and to explore it's alleviating alcoholism mechanisms.Methods The routine experimental method of small intestinal smooth muscle isolated from rabbits was used.We recorded the automatic contracting characteristic and changing law before and after taking drugs.Results Ethanol decreased the amplitude and frequency of smooth muscle.Compared with ethanol,the amplitude and frequency of smooth muscle were reduced by injected into Puerarin in dose-dependent manner.Conclusion Puerarin could enhance the inhibitory effects of ethanol on small intestine smooth muscle,so it cannt alleviate alcoholism by the above effects.

Abstract:Many researchers consider gut microbiota (trillions of microorganisms) an endogenous organ of its animal host, which confers a vast genetic diversity in providing the host with essential biological functions. Particularly, the gut microbiota regulates not only gut tissue structure but also gut health and gut functionality. This paper first summarized those common bacterial species (dominated by the Firmicutes, Bacteroidota, and Proteobacteria phyla) in swine gut and then briefly discussed their roles in swine nutrition and health, which include roles in nutrient metabolism, pathogen exclusion, and immunity modulation. Secondly, the current knowledge on how dietary nutrients and feed additives affect the gut bacterial composition and nutrient metabolism in pigs was discussed. Finally, how dietary amino acids affect the relative abundances and metabolism of bacteria in the swine gut was reviewed. Tryptophan supplementation promotes the growth of beneficial bacteria and suppresses pathogens, while arginine metabolism affects nitrogen recycling, impacting gut immune response and health. Glutamate and glutamine supplementations elevate the levels of beneficial bacteria and mitigate pathogenic ones. It was concluded that nutritional strategies to manipulate gut microbial ecosystems are useful measures to optimize gut health and gut functions. For example, providing pigs with nutrients that promote the growth of Lactobacillus and Bifidobacterium can lead to better gut health and growth performance, especially when dietary protein is limited. Further research to establish the mechanistic cause-and-effect relationships between amino acids and the dynamics of gut microbiota will allow swine producers to reap the greatest return on their feed investment.

Transport of Neutral Amino Acids in the Jejunum of Pigs with Special Consideration of L-Methionine

Abstract:Background: Methionine (Met) is a popular nutritional supplement in humans and animals. It is routinely supplemented to pigs as L-Met, DL-Met, or DL-2-hydroxy-4-(methylthio) butanoic acid (DL-HMTBA). Methods: We investigated the effect of these Met supplements on jejunal amino acid (AA) transport in male castrated Piétrain × Danbred pigs, also including a non-supplemented group. The mucosal-to-serosal flux of ten [14C]-labeled AAs (L-glutamine, glycine, L-leucine, L-lysine, L-Met, L-serine, L-threonine, L-tryptophan, L-tyrosine and L-valine) was investigated at two concentrations (50 μM and 5 mM). Inhibition of apical uptake by mucosal L-Met was also measured for these AAs. The intestinal expression of apical AA transporters, angiotensin-converting enzyme II and inflammation-related genes were compared with those of a previous study. Results: Except for tryptophan and lysine at 5 mM, all AA fluxes were Na+-dependent (p ≤ 0.05), and the uptake of most AAs, except glycine and lysine, was inhibited by L-Met (p < 0.001). A correlation network existed between Na+-dependent fluxes of most AAs (except tryptophan and partly glycine). We observed the upregulation of B0AT1 (SLC6A19) (p < 0.001), the downregulation of ATB0,+ (SLC6A14) (p < 0.001) and a lower expression of CASP1, IL1β, IL8, TGFβ and TNFα in the present vs. the previous study (p < 0.001). Conclusions: The correlating AAs likely share the same Na+-dependent transporter(s). A varying effect of the Met supplement type on AA transport in the two studies might be related to a different level of supplementation or a different inflammatory status of the small intestine.

Abstract:AIM: Some amino acids been known to influence gastric emptying. Thus we have evaluated the effects of straight alkyl chain, extra hydroxylated alkyl chain and branched chain amino acids on gastric emptying.MATERIALS AND METHODS: Gastric emptying was evaluated in rats after feeding with Racol (nutrient formulae) containing [1-(13)C] acetic acid. Using a breath test, the content of (13)CO2 in their expired air was measured by infrared analyzers. Rats were orally administered with test amino acids, while control rats were administered orally with distilled water.RESULTS: The expired (13)CO2 content in the expired air increased with time, peaked after about 30 min and decreased thereafter. Among the amino acids having an alkyl chain, L-serine, L-alanine and L-glycine, significantly decreased the (13)CO2 content and Cmax, and delayed Tmax, suggesting inhibition and delay of gastric emptying. AUC(120min) values of L-alanine and L-glycine also decreased significantly. L-Threonine significantly decreased (13)CO2 content and delayed Tmax, but had no influence on Cmax and AUC(120min) values, suggesting a delay of gastric emptying. L-Isoleucine and L-leucine and L-valine significantly decreased (13)CO2 content, suggesting inhibition of the gastric emptying, but Cmax, Tmax and AUC(120min) values were not significantly affected.CONCLUSION: The results show that the amino acids used in the present study had different effects on gastric emptying. Moreover, it was found that inhibition and delay of gastric emptying were clearly classifiable by analyzing the change in (13)CO2 content of the expired air and the Cmax, Tmax and AUC(120min) values.
Mutation of ameloblastin gene in calcifying epithelial odontogenic tumor.

Abstract:BACKGROUND Ameloblastin (AMBN) gene expresses an important protein that acts as a cell adhesion molecule. This protein plays an important role in maintaining the ameloblast secretory stage of differentiation by binding to them and inhibiting their proliferation. Due to the relationship of this protein in the differentiation and proliferation of odontogenic cells, here, we investigated this gene in different types of odontogenic tumors. MATERIALS AND METHODS Sequencing of the all encoding region of AMBN gene was carried out in four frozen cases of odontogenic tumors: one case of calcifying epithelial odontogenic tumor (CEOT), two calcifying odontogenic cysts (COC) and one ameloblastic fibroma (AF). RESULTS DNA sequencing was modified in an important domain of the AMBN only in the CEOT. CONCLUSION The present study suggests that AMBN gene alterations may be relevant to the pathogenesis of CEOT.
Expression and regulation of the neutral amino acid transporter B0AT1 in rat small intestine

Abstract:Absorption of neutral amino acids across the luminal membrane of intestinal enterocytes is mediated by the broad neutral amino acid transporter B0AT1 (SLC6A19). Its intestinal expression depends on co-expression of the membrane-anchored peptidase angiotensin converting enzyme 2 (ACE2) and is additionally enhanced by aminopeptidase N (CD13). We investigated in this study the expression of B0AT1 and its auxiliary peptidases as well as its transport function along the rat small intestine. Additionally, we tested its possible short- and long-term regulation by dietary proteins and amino acids. We showed by immunofluorescence that B0AT1, ACE2 and CD13 co-localize on the luminal membrane of small intestinal villi and by Western blotting that their protein expression increases in distal direction. Furthermore, we observed an elevated transport activity of the neutral amino acid L-isoleucine during the nocturnal active phase compared to the inactive one. Gastric emptying was delayed by intragastric application of an amino acid cocktail but we observed no acute dietary regulation of B0AT1 protein expression and L-isoleucine transport. Investigation of the chronic dietary regulation of B0AT1, ACE2 and CD13 by different diets revealed an increased B0AT1 protein expression under amino acid-supplemented diet in the proximal section but not in the distal one and for ACE2 protein expression a reverse localization of the effect. Dietary regulation for CD13 protein expression was not as distinct as for the two other proteins. Ring uptake experiments showed a tendency for increased L-isoleucine uptake under amino acid-supplemented diet and in vivo L-isoleucine absorption was more efficient under high protein and amino acid-supplemented diet. Additionally, plasma levels of branched-chain amino acids were elevated under high protein and amino acid diet. Taken together, our experiments did not reveal an acute amino acid-induced regulation of B0AT1 but revealed a chronic dietary adaptation mainly restricted to the proximal segment of the small intestine.

Abstract:Objective To observe the relationship between homocysteine and arterial smooth muscle cells(SMC) proliferation. To reveal machine of hyperhomocysteinemia result in atherosclerosis.Methods Rat arterial smooth muscle cells of primary culture promoted by different types of homocysteine and different concentration, to observe the degree of smooth muscle cells proliferation gy MTT assay.Results The arterial smooth muscle cells proliferation induced by Hcy has no significant discrepancy to control by One-Way ANOVA test. One-WAY ANOVA test showed that there were significant differnce when the concentration of D, L-Hcy and L-Hcy is higher. Univariate test showed that there were significant difference between D-Hcy and L-Hcy among the several types of Hcy. Conclusion The proliferation of SMC was positively correlated with the D, L-Hcy and L-Hcy concentration. There is no sifnificant effect when D-Hcy promotes the proliferation of VSMC. There is no great difference between the SMC proliferation of induced by L-Hcy and D, L-Hcy. But there is significant difference between the proliferation induced by L-Hcy and D-Hcy.
Histological and histochemical study of effect of dietary fibre in motility of stomach in male mice

Abstract:Objective: To investigate the relationship between dietary fibres and muscularis externa layer. Methods: The experimental study was conducted at the Anatomy Department of the College of Medicine, Al- Nahrain University, Iraq, from November 2020 to April 2021, and comprised adult healthy males. They were divided randomly into experimental group A and control group B. Group A was fed high-fibre diet, and group B was fed standard pellet diet. The tissue samples were harvested at day 30 post-surgery. The stomach samplings were placed in 10% neutral formalin for 24 hours to obtain paraffin sections for routine histological and immunohistochemical staining. The protein expression in stomach's smooth muscle of each group was detected by immunohistochemical staining. Data was analysed using SPSS 24. Results: Of the 20 mice, 10(50%) were in each of the two groups. Group A exhibited significant weight-gain compared to group B (p≤0.01). There was significant reduction in the thickness of the muscularis layer in group A compared to group B (p≤0.01). Anoctamin 1 expression in group A was significantly lower than that in group B (p≤0.01). Conclusions: The stress induced by an unstable fibre diet significantly affected the stomach by decreasing the muscularis layer thickness and Anoctamin 1 expression in interstitial cells of Cajal.

Abstract:Recently, magnetic nanoparticles of iron oxide (Fe3O4, γ-Fe2O3) have shown an increasing number of applications in the field of biomedicine, but some questions have been raised about the potential impact of these nanoparticles on the environment and human health. In this work, the three types of magnetic nanoparticles (DMSA-Fe2O3, APTS-Fe2O3, and GLU-Fe2O3) with the same crystal structure, magnetic properties, and size distribution was designed, prepared, and characterized by transmission electronic microscopy, powder X-ray diffraction, zeta potential analyzer, vibrating sample magnetometer, and Fourier transform Infrared spectroscopy. Then, we have investigated the effect of the three types of magnetic nanoparticles (DMSA-Fe2O3, APTS-Fe2O3, and GLU-Fe2O3) on smooth muscle cells (SMCs). Cellular uptake of nanoparticles by SMC displays the dose, the incubation time and surface property dependent patterns. Through the thin section TEM images, we observe that DMSA-Fe2O3is incorporated into the lysosome of SMCs. The magnetic nanoparticles have no inflammation impact, but decrease the viability of SMCs. The other questions about metabolism and other impacts will be the next subject of further studies.
The Functional Proteome Preliminary Study of Rat Arterial Smooth Muscle Cells Promoted by Homocysteine

Abstract:Objective:We investigated the changes of the proteins in the rat arterial smooth muscle cells induced by homocysteine.Detected the mechanism by which homocysteinemia leads to atherosclerosis. Methods:The rat arterial smooth muscle cells of primary culture were planned to be the experimental systems.Extracted the proteins from the smooth muscle cells which were induced by homocysteine and control.After 2-D electrophoresis,the map was analyzed by imaging systems. Results:After cells promoted by homocysteine, twenty-eight proteins of the smooth muscle cells changed.Twelve proteins were found to differ in quantity(11 higher and 1 lower).Sixteen proteins were seen to be unique in one region or the others(12 points of protein appear after Hcy stimulating,4 points of protein disapear after Hcy stimulating). Conclusion:The protein of rat arterial muscle cells changed after promoted by homocysteine.We considered that the proteins were related to the proliferation of the smooth muscle cells.
The Effect of Weichangshu on Plasma MTL,SP and SS in Rats of Gastrointestinal Motility Disorder

Abstract:Objective To investigate the mechanism of Weichangshu promots gastrointestinal motility.Methods Select SD rats were randomly divided into normal control group,L-Arg group,L-Arg plus Weichangshu group and L-Arg plus xishabili group.In addition to the normal control group,the other three groups were Intraperitoneal injected L-Arg,Each group was given equal volume of drug administered,abdominal aortic blood,radioimmunoassay determination of plasma MTL,SP,SS content.Results L-Arg model contral group MTL,SP,SS were significantly lower,compared with the normal contral group(P0.01).After treatment,Weichangshu group MTL,SP,SS in plasma increased significantly,compared with L-Arg model contral group(P0.05).Conclusion The mechanism of Weichangshu promote gastrointestinal motility is may be:①Regulate plasma gastrointestinal hormone balance and facilitate the coordination,then Promote gastrointestinal motility.②Elevate levels of plasma MTL and SP,Increase Ca2+ concentration within gastrointestinal smooth muscle cells,Ca2+ signal system trigger a series of physiological functions and then promote the effect of gastrointestinal smooth muscle contraction.

Abstract:Bioactive peptides derived from both exogenous and endogenous origins have been studied extensively to use their beneficial effects in humans and animals. Bioactive peptides exhibit beneficial bodily functions and contribute to a healthy gastrointestinal system by influencing barrier functions, immune responses, and gut microbiota. Gut microbiota is a diverse microbial community that significantly influences the overall well-being and homeostasis of the body. Factors such as diet, age, lifestyle, medication, and environmental circumstances can affect the composition and diversity of the gut microbiota. The disturbances or imbalances in the gut microbiota have been associated with various health problems. The interplays between bioactive peptides and gut microbiota are not fully understood, but bioactive peptides hold promise as modulators of the gut microbiota to promote gut health. Almost all the bioactive research on human health, including the development of therapeutics and nutritional interventions, uses cell culture, even though their direct biofunctional activities can only occur when absorbed in the intestine and into the blood system. This review focuses on the current understanding of bioactive peptides in gut microbiota and their impact and mechanisms on gut and human health. The novelty of this review lies in its comprehensive analysis of the multifaceted interactions between bioactive peptides and gut microbiota, integrating knowledge from diverse disciplines between microbiology and nutrition. By elucidating the underlying mechanisms and identifying current research gaps, this review offers an outlook on the potential of bioactive peptides in promoting gut health and shaping future therapeutic and nutritional interventions.
