Abstract:Silver nanoparticles (AgNPs) are one of the most investigated metal-based nanomaterials. Their biocidal activity boosted their application in both diagnostic and therapeutic medical systems. It is therefore crucial to provide sound evidences for human-related safety of AgNPs. This study aimed to enhance scientific knowledge with regard to biomedical safety of AgNPs by investigating how their different surface properties affect human immune system. Methods: preparation, characterization and stability evaluation was performed for four differently coated AgNPs encompassing neutral, positive and negative agents used for their surface stabilization. Safety aspects were evaluated by testing interaction of AgNPs with fresh human peripheral blood mononuclear cells (hPBMC) by means of particle cellular uptake and their ability to trigger cell death, apoptosis and DNA damages through induction of oxidative stress and damages of mitochondrial membrane. Results: all tested AgNPs altered morphology of freshly isolated hPBMC inducing apoptosis and cell death in a dose- and time-dependent manner. Highest toxicity was observed for positively-charged and protein-coated AgNPs. Cellular uptake of AgNPs was also dose-dependently increased and highest for positively charged AgNPs. Intracellularly, AgNPs induced production of reactive oxygen species (ROS) and damaged mitochondrial membrane. Depending on the dose, all AgNPs exhibited genotoxic potential. Conclusions: this study provides systematic and comprehensive data showing how differently functionalized AgNPs may affect the human immune system. Presented results are a valuable scientific contribution to safety assessment of nanosilver-based blood-contacting medical products.

Abstract:Silver nanoparticles (AgNPs) are known to affect the physiology and morphology of plants in various ways, but the exact mechanism by which they interact with plant cells remains to be elucidated. An unresolved question of silver nanotoxicology is whether the interaction is triggered by the physical features of the particles, or by silver ions leached from their surface. In this study, we germinated and grew Arabidopsis thaliana seedlings in synthetic medium supplemented with sub-morbid concentrations (4 μg/mL) of AgNPs and silver nitrate (AgNO3). This treatment led to in planta accumulation of 106 μg/g and 97 μg/g of silver in the AgNO3- and AgNP-exposed seedlings, respectively. Despite the statistically indistinguishable silver accumulation, RNA sequencing data demonstrated distinct changes in the transcriptome of the AgNP-exposed, but not in the AgNO3-exposed plants. AgNP exposure induced changes in the expression of genes involved in immune response, cell wall organization, photosynthesis and cellular defense against reactive oxygen species. AgNO3 exposure, on the other hand, caused the differential expression of only two genes, neither of which belonged to any AgNP-enriched gene ontology categories. Moreover, AgNP exposure led to a 39% reduction (p < 0.001) in total chlorophyll concentration relative to untreated plants which was associated with a 56.9% and 56.2% drop (p < 0.05) in carbon assimilation rate at ambient and saturating light, respectively. Stomatal conductance was not significantly affected by AgNP exposure, and limitations to carbon assimilation, as determined through analysis of light and carbon dioxide (A/Ci) curves, were attributed to rates of electron transport, maximum carboxylation rates and triose phosphate use. AgNO3-exposure, on the other hand, did not lead to significant reduction either in chlorophyll concentration or in carbon assimilation rate. Given these data, we propose that the impact of AgNPs cannot be simply attributed to the presence of the metal in plants, but is innate to the particulate nature of nanosilver.
Cellular uptake and toxicity effects of silver nanoparticles in mammalian kidney cells

Abstract:The rapid progress and early commercial acceptance of silver-based nanomaterials is owed to their biocidal activity. Besides embracing the antimicrobial potential of silver nanoparticles (AgNPs), it is imperative to give special attention to the potential adverse health effects of nanoparticles owing to prolonged exposure. Here, we report a detailed study on the in vitro interactions of citrate-coated AgNPs with porcine kidney (Pk15) cells. As uncertainty remains whether biological/cellular responses to AgNPs are solely as a result of the release of silver ions or whether the AgNPs themselves have toxic effects, we investigated the effects of Ag(+) on Pk15 cells for comparison. Next, we investigated the cellular uptake of both AgNPs and Ag(+) in Pk15 cells at various concentrations applied. The detected Ag contents in cells exposed to 50 mg l(-1) AgNPs and 50 mg l(-1) Ag(+) were 209 and 25 µg of Ag per 10(6) cells, respectively. Transmission electron microscopy (TEM) images indicated that the Pk15 cells internalized AgNPs by endocytosis. Both forms of silver, nano and ionic, decreased the number of viable Pk15 cells after 24 h in a dose-dependent manner. In spite of a significant uptake into the cells, AgNPs had only insignificant toxicity at concentrations lower than 25 mg l(-1) , whereas Ag(+) exhibited a significant decrease in cell viability at one-fifth of this concentration. The Comet assay suggested that a rather high concentration of AgNP (above 25 mg l(-1) ) is able to induce genotoxicity in Pk15 cells. Further studies must seek deeper understanding of AgNP behavior in biological media and their interactions with cellular membranes.

Silver Nanoparticles Effects on In Vitro Germination, Growth, and Biochemical Activity of Tomato, Radish, and Kale Seedlings

Abstract:The interactions between nanoparticles and plant cells are still not sufficiently understood, and studies related to this subject are of scientific and practical importance. Silver nanoparticles (AgNPs) are one of the most commonly produced and used nanomaterials. This study aimed to investigate the influence of AgNPs applied at the concentrations of 0, 50, and 100 mg·L-1 during the process of in vitro germination as well as the biometric and biochemical parameters of developed seedlings in three vegetable species: Solanum lycopersicum L. 'Poranek', Raphanus sativus L. var. sativus 'Ramona', and Brassica oleracea var. sabellica 'Nero di Toscana'. The application of AgNPs did not affect the germination efficiency; however, diverse results were reported for the growth and biochemical activity of the seedlings, depending on the species tested and the AgNPs concentration. Tomato seedlings treated with nanoparticles, particularly at 100 mg·L-1, had shorter shoots with lower fresh and dry weights and produced roots with lower fresh weight. Simultaneously, at the biochemical level, a decrease in the content of chlorophylls and carotenoids and an increase in the anthocyanins content and guaiacol peroxidase (GPOX) activity were reported. AgNPs-treated radish plants had shorter shoots of higher fresh and dry weight and longer roots with lower fresh weight. Treatment with 50 mg·L-1 and 100 mg·L-1 resulted in the highest and lowest accumulation of chlorophylls and carotenoids in the leaves, respectively; however, seedlings treated with 100 mg·L-1 produced less anthocyanins and polyphenols and exhibited lower GPOX activity. In kale, AgNPs-derived seedlings had a lower content of chlorophylls, carotenoids, and anthocyanins but higher GPOX activity of and were characterized by higher fresh and dry shoot weights and higher heterogeneous biometric parameters of the roots. The results of these experiments may be of great significance for broadening the scope of knowledge on the influence of AgNPs on plant cells and the micropropagation of the vegetable species. Future studies should be aimed at testing lower or even higher concentrations of AgNPs and other NPs and to evaluate the genetic stability of NPs-treated vegetable crops and their yielding efficiency.

Abstract:Abstract Background Silver nanoparticles (AgNPs) are currently one of the most manufactured nanomaterials. A wide range of toxicity studies have been performed on various AgNPs, but these studies report a high variation in toxicity and often lack proper particle characterization. The aim of this study was to investigate size- and coating-dependent toxicity of thoroughly characterized AgNPs following exposure of human lung cells and to explore the mechanisms of toxicity. Methods BEAS-2B cells were exposed to citrate coated AgNPs of different primary particle sizes (10, 40 and 75 nm) as well as to 10 nm PVP coated and 50 nm uncoated AgNPs. The particle agglomeration in cell medium was investigated by photon cross correlation spectroscopy (PCCS); cell viability by LDH and Alamar Blue assay; ROS induction by DCFH-DA assay; genotoxicity by alkaline comet assay and γH 2 AX foci formation; uptake and intracellular localization by transmission electron microscopy (TEM); and cellular dose as well as Ag release by atomic absorption spectroscopy (AAS). Results The results showed cytotoxicity only of the 10 nm particles independent of surface coating. In contrast, all AgNPs tested caused an increase in overall DNA damage after 24 h assessed by the comet assay, suggesting independent mechanisms for cytotoxicity and DNA damage. However, there was no γH 2 AX foci formation and no increased production of intracellular reactive oxygen species (ROS). The reasons for the higher toxicity of the 10 nm particles were explored by investigating particle agglomeration in cell medium, cellular uptake, intracellular localization and Ag release. Despite different agglomeration patterns, there was no evident difference in the uptake or intracellular localization of the citrate and PVP coated AgNPs. However, the 10 nm particles released significantly more Ag compared with all other AgNPs (approx. 24 wt% vs . 4–7 wt%) following 24 h in cell medium. The released fraction in cell medium did not induce any cytotoxicity, thus implying that intracellular Ag release was responsible for the toxicity. Conclusions This study shows that small AgNPs (10 nm) are cytotoxic for human lung cells and that the toxicity observed is associated with the rate of intracellular Ag release, a ‘Trojan horse’ effect.

Abstract:Hepatitis B virus (HBV) recurrence after orthotopic liver transplantation (OLT) is associated with poor graft‐ and patient‐survival. Treatment with HBV‐specific immunoglobulins (HBIG) in combination with nucleos(t)ide analogs is effective in preventing HBV reinfection of the graft and improving OLT outcome. However, the role of HBV‐specific cellular immunity in viral containment in immune suppressed patients in general and in OLT recipients in particular is unclear. To test whether or not OLT recipients maintain robust HBV‐specific cellular immunity, the cellular immune response against HBV was assessed in 15 OLT recipients and 27 individuals with chronic and 24 subjects with self‐limited HBV infection, respectively; using an overlapping peptide set spanning the viral nucleocapsid‐ and envelope‐protein sequences. The data demonstrate that OLT recipients mounted fewer but stronger clusters of differentiation (CD)8 T cell responses than subjects with self‐limited HBV infection and showed a preferential targeting of the nucleocapsid antigen. This focused response pattern was similar to responses seen in chronically infected subjects with undetectable viremia, but significantly different from patients who presented with elevated HBV viremia and who mounted mainly immune responses against the envelope protein. In conclusion, virus‐specific CD4 T cell–mediated responses were only detected in subjects with self‐limited HBV infection. Thus, the profile of the cellular immunity against HBV was in immune suppressed patients similar to subjects with chronic HBV infection with suppressed HBV‐DNA. Liver Transpl 2008. © 2008 AASLD.

Abstract:Background: Silver nanoparticles (AgNPs) are used in industrial applications as catalysts, sanitary materials, and health supplements. Generally, AgNPs have shown cytotoxicity such as cell membrane damage. However, the mechanisms of their toxicity have not been completely elucidated. Methods: The cellular effects (cell viability, induction of chemokine and cellular oxidative stress) of two AgNP water suspensions (AgNP-A for cosmetic application and AgNP-B for industrial application) on epithelial-like A549 cells and macrophage-like differentiated THP-1 (dTHP-1) cells were examined. Results: AgNPs caused enhancement of IL-8 expression and oxidative stress. The cellular uptake of AgNP-A cells was observed. However, the cellular uptake of AgNP-B into A549 cells was hardly observed. Moreover, the intracellular Ag level was increased by AgNP suspensions exposure. Cell viability was not affected by AgNP suspensions exposure. Conclusions: AgNPs induce chemokine expression and cellular oxidative stress on culture cells. The intracellular Ag level may be important for these cellular effects.
Effects of Exposure Pathways on the Accumulation and Phytotoxicity of Silver Nanoparticles in Soybean and Rice

Abstract:Abstract The widespread use of silver nanoparticles (AgNPs) raises concerns both about their accumulation in crops and human exposure via crop consumption. Plants take up AgNPs through their leaves and roots, but foliar uptake has been largely ignored. To better understand AgNPs–plant interactions, we compared the uptake, phytotoxicity and size distribution of AgNPs in soybean and rice following root versus foliar exposure. At similar AgNP application levels, foliar exposure led to 17–200 times more Ag bioaccumulation than root exposure. Root but not foliar exposure significantly reduced plant biomass, while root exposure increased the malondialdehyde and H2O2 contents of leaves to a larger extent than did foliar exposure. Following either root or foliar exposure, Ag-containing NPs larger (36.0–48.9 nm) than the originally dosed NPs (17–18 nm) were detected within leaves. These particles were detected using a newly developed macerozyme R-10 tissue extraction method followed by single-particle inductively coupled plasma mass spectrometry. In response to foliar exposure, these NPs were stored in the cell wall and plamalemma of leaves. NPs were also detected in planta following Ag ion exposure, indicating their in vivo formation. Leaf-to-leaf and root-to-leaf translocation of NPs in planta was observed but the former did not alter the size distribution of the NPs. Our observations point to the possibility that fruits, seeds and other edible parts may become contaminated by translocation processes in plants exposed to AgNPs. These results are an important contribution to improve the risk assessment of NPs under environmental exposure scenarios.
Prolonged Effects of Silver Nanoparticles on p53/p21 Pathway-Mediated Proliferation, DNA Damage Response, and Methylation Parameters in HT22 Hippocampal Neuronal Cells

Abstract:It is widely accepted that silver nanoparticles (AgNPs) are toxic to biological systems. However, little is known about their actions at molecular level and the cytophysiological effects after AgNP removal. As nanoparticles are suggested a promising tool to transport drugs to the brain for use in neurological conditions, we used HT22 mouse hippocampal neuronal cells as a model to study AgNP-mediated effects after their removal from the cell culture medium. We selected a relatively low concentration of AgNPs, 5 μg/ml, treated the cells for 48 h, and evaluated AgNP-induced cytophysiological effects after 96 h of AgNP removal. AgNP removal did not result in cytotoxicity. In contrast, AgNPs modulated HT22 cell cycle and proliferation and induced oxidative stress and 53BP1 recruitment, which were accompanied by elevated levels of p53 and p21. AgNP-associated diminution in lamin B1 pools did not significantly affect the structure of the nucleus. No disruption in F-actin dynamics was observed upon AgNP treatment. Moreover, we showed for the first time that AgNPs stimulated changes in DNA methylation: the augmentation in 5-methylcytosine (5-mC) and DNMT1, DNMT2, DNMT3a, and DNMT3b levels were observed. The upregulation of DNMT2 may be a part of cellular stress response to AgNP treatment. Taken together, AgNP removal resulted in p53/p21-mediated inhibition of cell proliferation, oxidant-based DNA damage response, and changes in DNA methylation patterns, which suggests that more attention should be paid to the possible outcomes in individuals exposed to nano-sized biomaterials.

Abstract:Nanomaterials, including zinc oxide nanoparticles (ZnO NPs), have a great application potential in many fields, such as medicine, the textile industry, electronics, and cosmetics. Their impact on the environment must be carefully investigated and specified due to their wide range of application. However, the amount of data on possible negative effects of ZnO NPs on plants at the cellular level are still insufficient. Thus, we focused on the effect of ZnO NPs on tobacco BY-2 cells, i.e., a widely accepted plant cell model. Adverse effects of ZnO NPs on both growth and biochemical parameters were observed. In addition, reactive oxygen and nitrogen species visualizations confirmed that ZnO NPs may induce oxidative stress. All these changes were associated with the lipid peroxidation and changes in the plasma membrane integrity, which together with endoplasmatic reticulum and mitochondrial dysfunction led to autophagy and programmed cell death. The present study demonstrates that the phytotoxic effect of ZnO NPs on the BY-2 cells is very complex and needs further investigation.

How Can Biological and Chemical Silver Nanoparticles Positively Impact Physio-Chemical and Chloroplast Ultrastructural Characteristics of Vicia faba Seedlings?

Abstract:Through interactions with plant cells, silver nanoparticles (AgNPs) with both biological and chemical origins can stimulate physiological and metabolic processes in plants. To ensure their safe application in the food chain, it is necessary to investigate their effects on plant systems. Therefore, the effects of chemical AgNPs (chem-AgNPs) and biologically synthesized AgNPs (bio-AgNPs) at different levels (i.e., 0, 10, and 50 ppm) on physiological and biochemical traits {i.e., root and shoot growth traits, photosynthetic pigments (Chl a, Chl b, carotenoids, and total pigments), soluble sugars, total carbohydrates, starch, H2O2, and antioxidant enzyme activities} of Vicia faba L. seedlings were investigated. AgNPs were biosynthesized from silver nitrate (AgNO3) by a green synthesis approach using Jatropha curcas seed extract. The synthesized AgNPs were characterized by UV-vis spectroscopy, transmission electron microscopy (TEM), zeta potential, Fourier-transform infrared spectra (FT-IR), and X-ray diffraction (XRD). The results showed that bio-AgNPs at 10 ppm resulted in the highest growth, physiological, and biological traits of faba bean seedlings in comparison with those obtained from both AgNO3 and chem-AgNPs treatments. On the other hand, all AgNPs treatments adversely affected the chloroplast ultrastructure, however, fewer negative effects were obtained with the application of 10 ppm bio-AgNPs. In addition, the roots and shoots of seedlings contained the lowest Ag content under different treatments at 10 ppm AgNPs in comparison to the highest level of AgNPs (50 ppm), which indicates that additional studies should be incorporated to ensure safe use of lower concentrations of bio-AgNPs in seed priming. In conclusion, the application of biogenic nanoparticles at 10 ppm can be recommended to enhance plant growth and the productivity of strategic crops.

Abstract:Silver nanoparticles (AgNPs) have gained significant attention in various fields due to their unique properties, but their release into the environment has raised concerns about their environmental and biological impacts. Silver nanoparticles can enter plants following their exposure to roots or via stomata following foliar exposure. Upon penetrating the plant cells, AgNPs interact with cellular components and alter physiological and biochemical processes. One of the key concerns associated with plant exposure to AgNPs is the potential of these materials to induce oxidative stress. Silver nanoparticles can also suppress plant growth and development by disrupting essential plant physiological processes, such as photosynthesis, nutrient uptake, water transport, and hormonal regulation. In crop plants, these disruptions may, in turn, affect the productivity and quality of the harvested components and therefore represent a potential threat to agricultural productivity and ecosystem stability. Understanding the phytotoxic effects of AgNPs is crucial for assessing their environmental implications and guiding the development of safe nanomaterials. By delving into the phytotoxic effects of AgNPs, this review contributes to the existing knowledge regarding their environmental risks and promotes the advancement of sustainable nanotechnological practices.

Abstract:BACKGROUND: Silver exposures are rising because of the increased use of silver nanoparticles (AgNPs) in consumer products. The monovalent silver ion (Ag+) impairs neurodevelopment in PC12 cells and zebrafish.OBJECTIVES AND METHODS: We compared the effects of AgNPs with Ag+ in PC12 cells for neurodevelopmental end points including cell replication, oxidative stress, cell viability, and differentiation. First, we compared citrate-coated AgNPs (AgNP-Cs) with Ag+, and then we assessed the roles of particle size, coating, and composition by comparing AgNP-C with two different sizes of polyvinylpyrrolidone-coated AgNPs (AgNP-PVPs) or silica nanoparticles.RESULTS: In undifferentiated cells, AgNP-C impaired DNA synthesis, but to a lesser extent than an equivalent nominal concentration of Ag+, whereas AgNP-C and Ag+ were equally effective against protein synthesis; there was little or no oxidative stress or loss of viability due to AgNP-C. In contrast, in differentiating cells, AgNP-C evoked robust oxidative stress and impaired differentiation into the acetylcholine phenotype. Although the effects of AgNP-PVP showed similarities to those of AgNP-C, we also found significant differences in potencies and differentiation outcomes that depended both on particle size and coating. None of the effects reflected simple physical attributes of nanoparticles, separate from composition or coating, as equivalent concentrations of silica nanoparticles had no detectable effects.CONCLUSIONS: AgNP exposure impairs neurodevelopment in PC12 cells. Further, AgNP effects are distinct from those of Ag+ alone and depend on size and coating, indicating that AgNP effects are not due simply to the release of Ag+ into the surrounding environment.

Evaluation of Biological Effects and Toxicity of Cetyltrimethylammonium Bromide Stabilized Silver Nanoparticles and Cetyltrimethylammonium Bromide Alone Following Intravenous Injection in Mice

Abstract:Background:: The exciting benefits of Silver nanoparticles (AgNPs) in the biomedical field necessitate generating knowledge on the safety concerns which have been raised over the applications of these NPs. Objective:: To understand the biological effects and mechanism of toxicity induction of Cetyltrimethylammonium bromide (CTAB) stabilized AgNPs as well as CTAB alone in mice following intravenous injection. Methods:: The investigations were carried out by measuring hematological and serum biochemical parameters, oxidative stress, genotoxicity, and histopathology. Results:: AgNPs’ treatment was found to induce a marked decrease (p<0.05) in platelet and lymphocyte count, Serum glutamate oxaloacetate transaminase (SGOT), and increase (p<0.05) in granulocytes count and Serum glutamate pyruvate transaminase (SGPT) whereas CTAB treatment-induced a decrease in platelet count. The decrease in glutathione (GSH) and an increase in lipid peroxidation (LPO) levels in the liver, spleen, and kidney of mice suggest the potential role of AgNPs in inducing oxidative stress. Genotoxicity apparent from the increased comet parameters and micronuclei formation observed in the liver, spleen, and kidney of mice treated with AgNPs and CTAB. Histological examination in mice treated with AgNPs and CTAB showed (i) diffused venous congestion in the liver (AgNPs) and focal venous congestion in the liver (CTAB); (ii) mild red pulp congestion in the spleen and (iii) acute tubular necrosis in the kidney. Histological examination in mice treated with AgNPs and CTAB showed diffused venous congestion and focal venous congestion respectively in the liver, while mild red pulp congestion in the spleen and acute tubular necrosis in the kidney were also observed.. Conclusion:: There is a need to develop methods to dissolve the toxicity of CTAB, thereby it induces inducing relatively low or no toxicity without affecting compromising the stability of nanosilver formulations formation. Such insights are believed to be fundamental in the synthesis of high-performance AgNPs demonstrated for outstanding biomedical applications.
Lepidopteran insect susceptibility to silver nanoparticles and measurement of changes in their growth, development and physiology

Abstract:Increased use of nanomaterials in various fields of science has lead for the need to study the impact of nanomaterial on the environment in general and on insect and plant life in particular. We studied the impact of silver nanoparticles (AgNPs) on growth and feeding responses of two lepidopteran pests of castor plant (Ricinus communis L.) namely Asian armyworm, Spodoptera litura F. and castor semilooper, Achaea janata L. Larvae were fed with PVP coated-AgNPs treated castor leaf at different concentrations and their activity was compared to that of silver nitrate (AgNO3) treated leaf diets. Larval and pupal body weights decreased along with the decrease in the concentrations of AgNPs and AgNO3 in both the test insects. Low amounts of silver were accumulated in the larval guts, but major portion of it was eliminated through the feces. Ultrastructural studies of insect gut cell using Transmission Electron Microscopy (TEM) showed accumulation of silver nanoparticles in cell organelles. Changes in the antioxidative and detoxifying enzymes of the treated larva were estimated. The effect of treatments showed differences in the activities of detoxifying enzymes, carboxylesterases (CarE), glucosidases (Glu) and glutathione S-transferases (GST) in the larval gut. Activities of superoxide dismutase, catalase, and peroxidase were also altered in the larval bodies due to the AgNPs treatments, suggesting that exposure of larvae to nanoparticles induces oxidative stress, which is countered by antioxidant enzymes. Induction of these enzymes may be an effective detoxification mechanism by which the herbivorous insect defends itself against nanoparticle treatment.
Evaluation of the Genotoxic and Oxidative Damage Potential of Silver Nanoparticles in Human NCM460 and HCT116 Cells

Abstract:Nano Ag has excellent antibacterial properties and is widely used in various antibacterial materials, such as antibacterial medicine and medical devices, food packaging materials and antibacterial textiles. Despite the many benefits of nano-Ag, more and more research indicates that it may have potential biotoxic effects. Studies have shown that people who ingest nanoparticles by mouth have the highest uptake in the intestinal tract, and that the colon area is the most vulnerable to damage and causes the disease. In this study, we examined the toxic effects of different concentrations of Ag-NPs on normal human colon cells (NCM460) and human colon cancer cells (HCT116). As the concentration of nanoparticles increased, the activity of the two colon cells decreased and intracellular reactive oxygen species (ROS) increased. RT-qPCR and Western-blot analyses showed that Ag NPs can promote the increase in P38 protein phosphorylation levels in two colon cells and promote the expression of P53 and Bax. The analysis also showed that Ag NPs can promote the down-regulation of Bcl-2, leading to an increased Bax/Bcl-2 ratio and activation of P21, further accelerating cell death. This study showed that a low concentration of nano Ag has no obvious toxic effect on colon cells, while nano Ag with concentrations higher than 15 μg/mL will cause oxidative damage to colon cells.

Abstract:With the advancement of nanotechnology and the increasing utilization of nanoparticles (NPs), their production and release into the environment are on the rise. Consequently, it is crucial to continuously monitor the toxicity of nanoparticles for humans, animals, and plants, as well as their impact on the environment. This is particularly significant in relation to human health and food production, given the escalating use of nanomaterials in agriculture and horticulture. The aim of the study was to investigate the response of rapeseed seedlings to silver nanoparticles (AgNPs) and gold nanoparticles (AuNPs) over different periods of exposure. This research analyzed the impact of these nanoparticles on the biochemical response of rapeseed seedlings after 7, 14, and 21 days of growth in their presence. This study assessed the activity of guaiacol peroxidase (GPOX), pyrogallol peroxidase (PPOX), superoxide dismutase (SOD), and free protein content, as well as the interactions between key elements responsible for oxidative stress and the antioxidant response. The findings demonstrated a significant effect of AgNPs and AuNPs on stimulating the response of rapeseed seedlings, with the activity of PPOX, GPOX, and SOD being dependent on the exposure time and the type and dose of nanoparticles used. Enzyme activity increased with the length of exposure time, while the content of free protein decreased over the weeks. The most intense reaction of seedlings was observed in the case of GPOX, with the lowest activity observed in PPOX and SOD. High effects of the nanoparticle type and rate were also observed in the correlation matrix. This study suggests that a comprehensive analysis of plant reactions to nanoparticles could have a significant impact on the proper and effective use of nanoparticles in agriculture and horticulture. This could lead to the environmentally friendly production of high-quality plant material.

Abstract:The use of silver nanoparticles (AgNPs) in commercial products has increased significantly in recent years. Although there have been some attempts to determine the toxic effects of AgNPs in mammalian and human cell-lines, there is little information on plants which play a vital role in ecosystems. The study reports the use of Vicia faba root-tip meristem to investigate the genotoxicity of AgNPs under modified GENE-TOX test conditions. The root tip cells of V. faba were treated with four different concentrations of engineered AgNPs dispersion to study toxicological endpoints such as mitotic index (MI), chromosomal aberrations (CA) and micronucleus induction (MN). For each concentration, five sets of microscopy observations were carried out. The results demonstrated that AgNPs exposure significantly increased (p < 0.05) the number of chromosomal aberrations, micronuclei, and decreased the MI in exposed groups compared to control. From this study we infer that AgNPs might have penetrated the plant system and may have impaired mitosis causing CA and MN. The results of this study demonstrate that AgNPs are genotoxic to plant cells. Since plant assays have been integrated as a genotoxicity component in risk assessment for detection of environmental mutagens, they should be given full consideration when evaluating the overall toxicological impact of the nanoparticles in the environment.

Abstract:Bioactive, oxygenated metabolites of polyunsaturated fatty acids (PUFAs) are important indicators of inflammation and oxidative stress but almost nothing is known about their interactions with nanomaterials (NMs). To investigate the effects of nano-sized materials (n-TiO<sub>2</sub>, n-ZnO, n-Ag) and their bulk-sized or ionic (b-TiO<sub>2</sub>, b-ZnO, i-Ag) counterpart, we studied the status of oxidative stress and PUFA metabolism in THP-1 cells at low-toxic concentrations (<15% cytotoxicity) 6 h or 24 h after the particle exposures by LC/MS and microarray. N-Ag had a significant and sustained impact on cellular antioxidant defense, seen as incremental synthesis and accumulation of glutathione (GSH) in the cell, and reduction of superoxide dismutase (SOD) activity. The cellular particle doses were largely dependent on exposure duration and particle dissolution, and active transporter mechanisms controlled the concentration of Zn in cytosol. Even at these sub-toxic concentrations, n-Ag was able to induce statistically significant elevation in the 5-HETE: arachidonic acid ratio at 24 h, which suggests association to oxidative stress and induction of pro-inflammatory responses. This was supported by the enhanced gene expression of chemotaxis-related genes. Overall, THP-1 cells internalized all tested particles, but only n-Ag led to low level of oxidative stress through ROS production and antioxidant balance disruption. N-Ag stimulated arachidonic acid oxidation to form 5-HETE which further magnified the inflammatory responses by enhancing the production of mitochondrial superoxide and leukocyte chemokines. Since the sustained n-Ag uptake was detected, the effects may last long and function as a trigger for the low-grade inflammation playing role in the chronic inflammatory diseases.
