Abstract:In this chapter, the effects of high pressure treatment on the quality characteristics and rancidity of brown rice during storage are investigated. Our group has been studying brown rice quality modifications through innovative processing technologies. In addition to the high pressure treatment and germination-parboiling techniques, another novel brown rice processing method has been explored—the freeze–thaw cycle. Therefore, the effects of these three different treatments on the physicochemical properties of brown rice are also investigated with emphasis. The effect of high pressure treatment on the properties of rice bran proteins is also investigated.

Abstract:Different extrusion temperatures (90, 100, 110, and 120 degrees C) were used to investigate changes in the expansion ratios, die pressures, phytochemical contents and antioxidant activities of extrusion products of black rice. The results showed that the die pressure significantly decreased with the increasing extrusion temperature, and the expansion ratio reached a peak value at 100 degrees C. The soluble-free and total phenolic acid contents gradually increased, whereas portions of soluble-free and soluble-conjugated phenolic acids transformed into insoluble-bound phenolic acids. The soluble-free (52.45) and insoluble-bound (73.59 mg GAE/100 g DF) total phenolic contents (TPC) reached peak values at 110 degrees C. The soluble-conjugated TPC values remained similar. Antioxidant activity occurred at higher levels in the range from 100 degrees C to 120 degrees C. The anthocyanin content decreased after extrusion possibly because some anthocyanin remained in the residue after extraction and could not be completely extracted. The content of free sterols increased from 90 degrees C to 110 degrees C and decreased at 120 degrees C. However, the content of bound sterols showed an opposite trend and reached a minimum value at 110 degrees C.

Abstract:Brown rice (BR) contains more lipids, as compared with white rice, but its indigestibility and rancidity limit the consumer acceptability. Therefore, the objective of this study was to compare the effect of high-pressure (HP), freeze–thaw cycles (FTC) and germination-parboiling (GP) treatments on lipid in vitro digestibility and rancidity of BR. GP treated BR released the most fatty acids (especially palmitic acid and stearic acid) after in vitro digestion, followed by FTC treated BR. FTC treated BR kept the highest value of fat acidity during storage, while opposite results were observed in GP treated BR. Although HP treatment increased fat acidity value immediately, it slowed down the increase of fat acidity with storage. The results of conjugated dienes and malonaldehydes content in BR stored under accelerated conditions indicated better lipid oxidation stability after HP and GP treatment, and that it’s necessary for FTC treated BR products to be stored under anaerobic conditions.
Comparative Evaluation of the Nutritional, Antinutritional, Functional, and Bioactivity Attributes of Rice Bran Stabilized by Different Heat Treatments

Abstract:The objective of this study was to evaluate the effects of different stabilization treatments—namely, dry-heating, infrared-radiation, and microwave-heating—on the nutritional, antinutritional, functional, and bioactivity attributes of rice bran (RB). Among the heating treatments, infrared-radiation exerted the strongest inactivation, resulting in 34.7% residual lipase activity. All the stabilization methods were found to be effective in the reduction of antinutrients, including phytates, oxalate, saponins, and trypsin inhibitors. No adverse effect of stabilization was noted on chemical composition and fatty acid profile of RB. Instead, stabilization by all heat treatments caused a significant decrease of vitamin E and total phenolics content in RB; the same trend was observed for the antioxidant activity as evaluated by the DPPH test. The antioxidant activity, as evaluated by ABTS and FRAP tests, and water absorption capacity were improved by the stabilization of RB, whereas the oil absorption capacity and emulsifying properties decreased. Microwave-heating enhanced the foaming properties, whereas infrared-radiation improved the water solubility index and swelling power of RB. Consequently, treatment of RB with infrared-radiation has a potential for industrialization to inactivate the lipase and improve some functional properties of this material for uses as a nutraceutical ingredient in food and cosmetic products.

Abstract:Over the years, extrusion has been a multi-step thermal technique that has proven to be the most effective process to stabilize rice bran (RB). This study aimed to investigate the effects of extrusion treatment and temperature (15, 25, and 40 °C) on the storage stability, lipid oxidation, peroxidase, and peroxide values, free fatty acids, fatty acid composition, and protein variations of RB over 60 days. The study offers novel insights into the changes in RB's protein and amino acid compositions during extrusion and storage, which has not been extensively explored in prior research. After extrusion processing, peroxidase activity (POD) and lipase activity (LPS) were significantly reduced. However, peroxide value (PV), free fatty acids (FFA), and malondialdehyde content (MDA) observed a significantly increased by 0.64 mEqO2/kg, 8.3 mg/100 g, and 0.0005 μmol/L respectively. The storage stability of RB after extrusion shows that the POD, LPS, FFA, PV, and MDA were positively correlated with storage duration and temperature. The oleic acid/linoleic acid ratio in processed RB by extrusion had no significant changes during storage. The total and essential/non-essential amino acid ratios showed a downward trend of 5.26% and 8.76%, respectively. The first-order kinetics was the best-fitting model to describe the enzymatic inactivation and degradation of extruded RB during storage. The extrusion treatment did not affect the crude protein and the essential subunits of protein. Overall, the optimized extrusion procedure exhibited promising results in stabilizing the RB.

Abstract:Lysophospholipids (LPLs) represent a major class of polar lipids crucial for rice's nutritional and functional properties. This study investigates the impact of varying storage temperatures (20, 30, and 40 °C) and humidity (50 and 95%) on the nonstarch and starch LPLs of paddy and milled rice. The findings revealed that the average nonstarch LPL content in paddy rice aged at 20 °C (82.6 μg/g) and 40 °C (83.6 μg/g) was significantly lower than that at 30 °C (95.0 μg/g). The nonstarch LPL content of milled rice aged at 20 °C (78.0 μg/g) was significantly higher than that at 30 and 40 °C. High storage temperature (40 °C) and humidity (95%) resulted in a significant reduction in rice total starch LPC and LPE content when compared to low humidity (50%). The ratio of rice starch/nonstarch LPL components such as LPC16:0 and LPC18:2 remarkably increased with increased storage temperature and humidity.

Abstract:Stabilization of rice bran after milling is a necessary step to avoid subsequent oxidation of lipids in the bran. Selected moving-bed drying methods, i.e., hot-air fluidized bed drying (HAFBD), superheated-steam fluidized bed drying (SSFBD), and infrared vibrated bed drying (IRVD), were used to reduce the moisture content of the bran and, at the same time, inactivate deleterious enzymes, which are the cause of oxidation. Drying kinetics, oxidative stability parameters (i.e., lipase activity, free fatty acid content, and peroxide value), oil extraction yield, and contents of phenolic compounds and γ-oryzanol as well as color changes of the bran were determined. SSFBD could reduce the drying time by 8–22 and 76–79% in comparison with HAFBD and IRVD, respectively. Drying method significantly affected the total phenolic content (TPC) and total color changes but did not significantly affect the γ-oryzanol content of the bran. SSFBD resulted in the lowest levels of all oxidative stability parameters and to the highest TPC and oil extraction yield. SSFBD at 140°C resulted in the bran with the longest shelf life of 55 days and is suggested as an alternative method to shorten the stabilization process and maintain the stability of rice bran.

Abstract:Hydrolytic and oxidative rancidity are the main cause of brown rice quality deterioration during storage. Brown rice was soaked in water and treated with high pressure (HP) at 100-400 MPa, for 0-10 min. The effect of HP treatment on moisture content, fat acidity, conjugated dienes (CD) and 2-thiobarbituric acid (TBA) value during 3 months storage at room temperature, was evaluated and compared with soaked (0.1 MPa) brown rice and untreated brown rice. After storage, moisture content of HP treated brown rice were significantly higher than the untreated but lower than the soaked, apart from the brown rice treated at 400 MPa-0 min, which was the lowest in moisture content. HP treatment at 400 MPa enhanced the fat acidity immediately after the treatment, while samples treated at 200 MPa-0 min showed lower level of hydrolytic rancidity during storage. Better stabilities based on CD content and TBA value were observed in brown rice when HPP at 200 MPa - 10 min was applied.
Storage characteristics of infrared radiation stabilized rice bran and its shelf-life evaluation by prediction modeling

Abstract:Background: Rice bran is a nutrient-dense and resource-rich byproduct produced from the rice milling. The limitation of rice bran utilization is mainly caused by oxidative deterioration. Improvement of stability to prolong rice bran shelf-life has thus become an urgent requirement. Results: The present study aimed to determine the characteristics of infrared radiation heat treatment of rice bran (IRRB) and raw rice bran stored under different temperatures. The effects of heating and storage time on physicochemical, microbial, storage stability and structural properties were investigated. Additionally, the prediction model for the shelf-life of rice bran was established based on free fatty acids and the peroxide value by fitting the curve of bran lipid oxidation. The results obtained demonstrated that infrared radiation heating at 300 °C for 210 s resulted in decreased lipase activity and peroxidase activity of 73.05% and 81.50%, respectively. The free fatty acids and peroxide value of IRRB stored at 4 and 25 °C for 8 weeks were only reached at 2.35% and 3.17% and 2.53 and 3.64 meq kg-1 , respectively. The shelf-life prediction model showed the the shelf-life of infrared radiation-treated samples increased to 71.6 and 25.8 weeks under storage at 4 and 25 °C, respectively. Conclusion: The stabilizing process could effectively suppress microbial growth and had no prominent effect on the physicochemical and microstructure properties of rice bran and, simultaneously, storage life was greatly extended. © 2020 Society of Chemical Industry.
Hot air-assisted radio frequency heating for stabilization of rice bran: Enzyme activity, phenolic content, antioxidant activity and microstructure

Abstract:<p>Stabilization treatment of rice bran (RB) for reducing enzyme activity and improving storability is a key step to develop its value-added utilization. Hot air-assisted radio frequency (HA-RF) heating was investigated as a stabilization method for RB, and associated quality attributes were evaluated and compared with samples subjected to extrusion stabilization. Results showed that two HA-RF stabilization treatments (low-temperature HA-RF: 100–105 °C, 15 min; high-temperature HA-RF: 110–115 °C, 6 min) reduced lipase and polyphenol oxidase activities to 20–30% of fresh RB. HA-RF stabilized RB was darker than that of fresh RB. HA-RF stabilization had no significant (<em>p</em> > 0.05) effect on protein, lipid, total tocotrienol and total tocopherol contents, and fatty acid composition, but reduced free phenolic content of RB. HA-RF stabilized RB also showed higher free flavonoid content, higher antioxidant ability, and less damage to the microstructure than extruded RB. This study provides useful information for making HA-RF heating as an industrial stabilization method for RB.</p>
The Effect of Anaerobic Treatment on Polyphenols, Antioxidant Properties, Tocols and Free Amino Acids in White, Red, and Black Germinated Rice (oryza Sativa L.)

Abstract:The effects of anaerobic treatment on the content of polyphenols, antioxidant capability, tocols, and free amino acids (FAAs) in the germinated brown rice (GBR) were investigated. The values of polyphenols and antioxidant capability significantly increased in treated groups, in which the three phenolic acids, i.e. ferulic, p-coumaric, and sinapic acids, showed the most pronounced increase. No significant change was observed in tocols. Among the evaluated FAAs, only the content of aspartic and glutamic acids in the white, red and black GBRs decreased by 75, 65 and 69%, and by 30, 18 and 39% after anaerobic treatment, respectively. While the contents of serine and threonine remained unchanged, all others significantly increased. In particular, the contents of γ-aminobutyric acid (GABA) increased 4.2-, 9.7- and 4.8-folds in the white, red and black GBRs, respectively. The results indicated that additional anaerobic treatment could be used in the production of GBR as a functional food.
Effects of Radio Frequency Heating on the Stability and Antioxidant Properties of Rice Bran

Abstract:Radio frequency (RF) technology is considered as a rapid heating method. Lipase in rice bran could highly accelerate lipid oxidation. The objectives of this study were to establish the radio frequency heating conditions for lipase inactivation and to evaluate the stability and antioxidant capacity. The results showed that the suitable electrode gap for a 1 kg sample load was 6 cm, and it only took 2 min to heat rice bran from 25 °C to 100 °C. Besides, there were no significant differences in the total phenolic content, flavonoid content and color between the untreated and RF-treated group, and the DPPH free radical scavenging activity of the RF treatment reached 84.8%. The acid value, free fatty acid content and peroxide value of the RF-treated rice bran met the quality standard after 8 weeks of storage at 4, 25 and 37 °C. In summary, this study provides valuable information about the RF heating procedure, and shows the great potential of RF technology for stabilizing rice bran efficiently.

Abstract:Three treatments, namely germination-parboiling (PG), freeze-thaw cycle (FTC) and high pressure processing (HPP) were compared for phytochemical content and antioxidant activity of brown rice (BR). These were determined in raw (uncooked), cooked, and in-vitro digested BR and compared with those from untreated BR and white rice (WR). PG showed the highest retention of phytochemicals after cooking (87-100%) while it dropped to 59-72% with FTC and 64-76% with HPP. After in-vitro digestion, the highest amount of phenolics was found in PG-24 h and flavonoids in FTC for two cycles. The antioxidant activity, as determined by oxygen radical absorbance capacity and ABTS methods, showed the highest value to be associated with in-vitro digested sample of PG-24 h, and lowest in WR. The results of this study show that these three treatments could improve or retain the phenolic content and antioxidant activity in cooked BR after in-vitro digestion.

Abstract:Water absorption, color, morphology and cooked texture of brown rice were evaluated after selected soaking (30–50 °C, 30 min) and high pressure treatment (HPT) (100–500 MPa; single or two cycle; total holding time 10 min). Water absorption ratio and lightness values of brown rice were increased by soaking and HPT. Hardness and gumminess values of cooked brown rice were reduced while springiness and cohesiveness were elevated by HPT. Scanning electron microscopy indicated that HPT improved the texture of brown rice by disrupting the structure of rice bran layer, which allowed easier water penetration into the rice grain during cooking. Moreover, the two cycle HPT resulted in lighter color and softer texture for cooked brown rice than single cycle HPT primarily caused by the more severe structural disruption of bran layer. Overall, two cycle HPT after soaking could potentially improve the quality of brown rice, taking about the same time as the single cycle HPT. Further, the quality improvements with the two cycle HPT were facilitated at lower pressure levels thereby providing better commercial processing opportunities.

Abstract:Nowadays,due to changes in consumer life style and health awareness,brown rice is receiving increased interest as a source of healthy attributes.Compared with white rice,brown rice is rich in several health beneficial nutrients such as vitamins,dietary fiber,minerals,antioxidants and essential amino acids.It is necessary to study the influence of processing on nutrients of brown rice,because these nutrients are easily lost during processing.High pressure processing (HPP) is a new non-thermal preservation technology that enhances food safety and retains the food original nutrients.Up to date,the research on the effect of HPP on nutritional property of brown rice is limited.Phenolic,flavonoid content and antioxidant activity of brown rice after HPP (100 ～400 MPa) by single step (5 min and 10 min of holding time) or two step process (two cycles,5 min of holding time each) were investigated.The antioxidant activity was evaluated by 1,l-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and 2,2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS + ·) scavenging assays.Results showed that phenolic,flavonoid contents and antioxidant activity of brown rice were increased with the increase of pressure.Moreover,under the same pressure level,the phenolic,flavonoid contents and antioxidant activity of brown rice under two step HPP were higher than those from single step HPP.Percentage retention of phenolic and flavonoid content in brown rice treated by two step process at 300 MPa was 97％.These findings suggested that the HPP was an effective non-thermal processing to retain antioxidants in brown rice.

Abstract:The effect of microwave heating, packaging, and storage temperature on the production of free fatty acids (FFA) in rice bran was examined. Freshly milled raw rice bran was adjusted to 21% moisture content and heated in a microwave oven at 850 W for 3 min. Raw and microwave-heated rice bran were packed in zipper-top bags or vacuum-sealed bags and stored at 4-5 or 25 degrees C for 16 weeks. FFA content of bran was measured at 4-week intervals. Total FFA increased rapidly over the 16-week period from the initial value of 2.5% in raw bran stored at 25 degrees C to 54.9% in vacuum bags and 48.1% in zipper-top bags. However, total FFA of raw bran stored at 4-5 degrees C increased at a slower rate from an initial value of 2. 5 to 25.4% in vacuum bags and 19.5% in zipper-top bags. After 16 weeks of storage, total FFA of microwave-heated bran stored at 25 degrees C increased from 2.8 to 6.9 and 5.2%, respectively, for samples stored in vacuum bags and zipper-top bags. Total FFA of microwave-heated samples stored at 4-5 degrees C did not change significantly with storage time. Results showed that hydrolytic rancidity of rice bran can be prevented by microwave heating and that the recommended storage condition for microwaved rice bran is 4-5 degrees C in zipper-top bags.

Fractionation of the rice bran layer and quantification of vitamin E, oryzanol, protein, and rice bran saccharide

Abstract:Value-added processing with respect to rice milling has traditionally treated the rice bran layer as a homogenous material that contains significant concentrations of high-value components of interest for pharmaceutical and nutraceutical applications. Investigators have shown that high-value components in the rice bran layer vary from differences in kernel-thickness, bran fraction, rice variety, and environmental conditions during the growing season. The objectives of this study were to quantify the amount of rice bran removed at pre-selected milling times and to correlate the amount of rice bran removed at each milling time with the concentration of vitamin E, gamma-oryzanol, rice bran saccharide, and protein obtained. The ultimate goal of this research is to show that rice bran fractionation is a useful method to obtain targeted, nutrient-rich bran samples for value-added processing. Two long grain rice cultivars, Cheniere and Cypress, were milled at discrete times between 3 and 40 seconds using a McGill mill to obtain bran samples for analysis. Results showed that the highest oryzanol and protein concentrations were found in the outer portion of the rice bran layer, while the highest rice bran saccharide concentration was found in the inner portion of the bran layer. Vitamin E concentration showed no significant difference across the bran layer within a variety, though the highest magnitude of concentration occurs within the first 10 seconds of milling for both varieties. To extract the higher concentration of oryzanol and protein only the outer portion of the bran layer requires processing, while to extract the higher concentration of rice bran saccharide, only the inner portion of the bran layer requires processing. Rice bran fractionation allows for the selective use of portions of the bran layer and is advantageous for two reasons: (1) bran fractions contain higher concentrations of components of interest with respect to the overall bran layer average, and (2) less bran needs to be processed to obtain components of interest.

Abstract:Abstract Although the stability of rice bran oil (RBO) has been showed on several studies, the factors which make it capable on maintaining its stability under thermal oxidation has not been sure yet. We hypothesized that its fatty acid composition [high composition of oleic acid (OA), lower composition of linoleic acid (LA) and α-linolenic acid (LnA)] and/or its antioxidant agents [γ-oryzanol (OZ)] and vitamin E [tocopherol (Toc), tocotrienol (T3)] might be the biggest factor. To prove the hypothesis, we thermally oxidized RBO under 40 °C for 17 days to mimic the harsh daily storage condition, and compared it with soybean oil (SO) and rapeseed oil (RPO) then monitoring their primary oxidation products [triacylglycerol hydroperoxide (TGOOH)] from easily oxidized fatty acid contained in triacylglycerol (TG) and the amount loss of antioxidant agents. As a result, RBO showed the lowest TGOOH/TG ratio, followed by RPO and SO. The superior stability RPO compared SO might occur due to because of the influence of the fatty acid profile (higher OA and lower LA). For RBO’s case, besides its fatty acid profile, the existence of OZ and the synergistic effect of OZ and vitamin E might have a greater contribution in maintaining its stability under thermal oxidation.

Abstract:Development of Free Fatty Acids (FFA) is a major problem which affects the use of rice bran as a feed ingredient. The purpose of this research was to evaluate the effect of microwave heating and pan roasting on FFA development of rice bran during storage, proximate composition and to study the available phosphorous (P) content. Rice bran from raw and parboiled rice obtained from paddy variety BG 358 was treated using microwave heat and pan roasting. An experiment was conducted in a two-factor factorial with 2×3 factorial design. Treated samples were studied for FFA content up to 8 weeks in weekly intervals. Proximate composition was studied immediately after heat treatment and at the eighth week. Availability of P in rice bran was studied after heat treatment. Both the treatments significantly reduced the formation of FFA in rice bran during storage (P<0.05), while microwave heat treatment showed a higher effect on reduction of the formation of FFA content in raw microwave heated rice bran from 2.68 to 12.44 g 100 g˗1 and in parboiled microwave heated rice bran from 1.8 to 9.54 g 100 g˗1 during eight weeks of storage. There was no significant (P>0.05) difference in proximate composition of rice bran during the period of storage in any of the treatments. Both treatments improved P availability; however, pan-roasting had a high effect on P availability where raw pan roasted rice bran and parboiled pan roasted rice bran showed an increment of 17.67 and 19.43%, respectively. It is concluded that pan roast and microwave heat retard development of rancidity in rice bran, although at different degrees. These findings could be used with suitable modifications to the milling and storage processes to improve the quality of the rice bran available for animal feeding in Sri Lanka.

Abstract:This research aimed to stabilize rice bran to reduce enzymatic reactions and optimize rice bran oil extraction using ultrasound-assisted extraction (UAE). The optimization was performed using response surface methodology (RSM). The results showed that rice bran oil has three primary fatty acids (oleic, linoleic, palmitic). Stabilization of rice bran using an autoclave (121 °C) for 10 min was recommended, with oil yield (14.2%), free fatty acid/FFA (0.41%), and gamma oryzanol (16.1 ppm). The storage significantly affected oil yield and gamma oryzanol but not FFA (p>0.05). The untreated and stabilized rice bran FFA during 18-day storage ranged from 1.33 to 5.73% and 0.41 to 1.12%. This confirmed that stabilization could inactivate lipase enzymes by reducing FFA. The optimization showed that the linear model best explained oil yield, total phenol content (TPC), and antioxidant activity (IC50), while the 2FI model was best fitted for gamma oryzanol. The optimal condition was found for UAE extraction at an amplitude of 90% and a time of 30 min. Verifying optimal conditions resulted in oil yield (13.6%), TPC (44.8 mg GAE/g), IC50 (207.2 ppm), and gamma oryzanol (15.8 ppm). Further in-depth studies are required to investigate using green solvents in the UAE and gamma oryzanol purification.
