Abstract:Organic weak acid, such as gluconic acid, acetic acid, formic acid koto-acetic acid and sorbic acid, can be separated by ion chromatography using 1 mmol/L NaHCO 3 as eluent at a flow-rate of 1 mL/min. The detection limits (S/N=3) of the organic weak acids are 0.500 μg/mL、0.145 μg/mL、0.097 μg/mL、0.195 μg/mL、1.078 μg/mLL, respectively, experiment results show that linear relationship and repeatability are good. The method is used to determine organic weak acid in drinks and the results are satisfactory.

Abstract:A simple method based on direct saponification followed by RP-HPLC analysis was developed for quantification of free and conjugated sterols in sugarcane. Acid hydrolysis prior to alkaline saponification was used to determined acylated steryl glycoside and steryl glycoside in sugarcane. The applicability and generality of this method were improved with intensive investigation. Compared to traditional solvent extraction method, this method was more time saving and appropriate for characterization of sterol fractions in sugarcane. This method was successfully applied for determination of free and conjugated sterols in different sugarcane samples. The results exhibited that stigmasterol (varied from 883.3 ± 23.5 to 1823.9 ± 24.5 μg/g dry weigh) and β-sitosterol (varied from 117.6 ± 19.9 to 801.4 ± 33.5 μg/g dry weight) were major phytosterols in the sugarcane sample, and their glycosylated forms accounted for almost 87.0% of stigmasterol and 87.5% of β-sitosterol in sugarcane, respectively. In addition, among other parts of sugarcane, tips contained the greatest amount of phytosterols.

Abstract:A sensitive high performance liquid chromatography (HPLC) assay was developed for the qualitative and quantitative determination of carbohydrate sweeteners and organic acids in oral fluid. To separate these compounds, an ion-moderated partition resin HPLC column (Aminex HPX-87H) was used. All components of the HPLC system were interconnected using stainless steel capillary tubing. Isocratic elution with 0.01 N sulfuric acid provided the profile of both compound classes. The compounds were detected using a refractive index detector. The method employed computerized data collection and integration (Omega-2 system) with a detection sensitivity of 0.1 µg compound per HPLC assay (80 µl). This method is useful in caries research, because it detects minute amounts of sugars and organic acids in oral fluid during clearance studies of various foods in the mouth.

Liquid chromatography of short chain carboxylic acids using a glutamic acid surfactant coated C18 stationary phase

Abstract:A C18 column was modified with the anionic amino acid surfactant lauroyl-l-glutamate (LLG) to facilitate the separation of ten short-chain aliphatic carboxylic acids (oxalic, tartaric, malic, malonic, lactic, acetic, maleic, citric, fumaric, and succinic). The developed method was proven to be fast, versatile, and environmentally friendly. After the coating of the column using 1% LLG solution and optimizing chromatographic conditions such as pH and temperature, near baseline resolution of the ten carboxylic acids within 4 min with excellent peak shape at pH = 1.8 using 100% H2O acidified with sulfuric acid was possible. Although the design of this stationary phase, with the hydrophilic group at the end of the alkyl chain, seems to be in contrast to such columns designed for a totally aqueous mobile phase that have a polar (often amide) group embedded near the silica surface, no evidence of phase collapse was noted. Linear relationships of ln retention factor (k) versus 1/Temperature (T) (van't Hoff plots) were generated for all the acids indicating a single retention mechanism was likely. As the pH of the mobile phase decreased, the analyte retention factors increased due to the increase of the fraction of the analyte with neutral charge (alpha zero). The surfactant amide linkage, being electron donating, increased the pKa of the more acidic carboxyl group of glutamic acid so both carboxyl groups were protonated (neutral) at pH 1.8. The exact nature of the retention mechanism is uncertain but there certainly seems to be a pronounced hydrophobic component due to the large difference in retention of fumaric acid and methyl fumarate at pH 1.8. In addition, eleven beverage samples were analyzed for their aliphatic carboxylic acid contents. The results showed that malic, fumaric, and citric acids were the most common carboxylic acids in natural beverages with concentrations as high as 6432 ppm of malic acid in organic apple juice, 64 ppm of fumaric acid in organic concord juice, and 6543 ppm citric acid in strawberry lemonade juice.

Abstract:An HPLC method was developed for the simultaneous separation and determination of sugars, ascorbic acid, 5-HMF (5-hydroxymethylfurfural), furfural and four other furanic compounds as the possible degradation products of sugars and ascorbic acid on heating and by Maillard reaction. Sucrose, glucose, fructose, ascorbic acid, dehydroascorbic acid, 5-HMF, furfural, DMHF (2,5-dimethyl-4-hydroxy-3(2H)-furanone), 2-furoic acid, 2-acetylfuran and furfuryl alcohol were separated on an Aminex HPX-87H column (300×7.8 mm). The mobile phase consisted of acetonitrile and 0.005 mol l−1 sulfuric acid aqueous solution (16:84, v/v). Two detectors, a refractive index detector and a photodiode array detector, were used to detect these compounds. Several fruit juice and drink samples were analyzed using this HPLC method. For alcoholic beverages, the content of alcohol was also simultaneously determined by refractive index detector.

Abstract:The application of column switching method was studied in an attempt to separate organic acids and cations in beverages by ion chromatography with an eluent which can be used for anion and cation exchange columns. The columns and valves were assembled in such a way that the columns could be operated both in series and parallel. For precutting operation, a cation exchange column (Yokogawa ICS-C25, 4.6 mm i.d. x 125 mm) and an anion exchange column (Yokogawa SAM3-075, 4.9 mm i.d. x 75 mm) were connected in series. Three and a half minutes after sample injection, when all anions in the sample has been transferred into the anion exchange column, flow of the eluent is changed by valve operation, so that the effluent from the cation exchange column directly enters the detector. When all cations are eluted from the cation exchange column, the valve is turned to the first position so that the effluent from the cation exchange column passes through the anion exchange column to serve as an eluent for separation of anions. The chromatograms for anions and cations can be recorded on a single chart with only one sample injection. Organic acids and five cations were separated at ambient temperature within 60 min when 1.5 mM oxalic acid at a flow rate of 0.75 ml/min was used as an eluent. The proposed method was applied to simultaneous determination of both organic acids and cations in wine, juice and coffee with good reproducibilities.

Abstract:A high performance liquid chromatography post column derivatization was developed for the determination of 19 free amino acids in tobacco.Samples were extracted by ultrasonic with 0.005 mol/L hydrochloric acid,separated by a high performance lithium cation exchange column,derivatiz by OPA with post column,and detected by a fluorescence detector.Each amino acid showed a good linear relationship in the content range of sample with the correlation coefficient(R2) more than 0.999,and the spike recoveries in tobacco samples were in the range of 87.73%~100.02%with relative standard deviations of 2.7%~6.0%,and the detection limits were 0.13~2 μmol/L.This method can be used for the analysis of free amino acids in tobacco.

Abstract:We address the exponential consensus problem for the linearized Vicsek model which was introduced by Jadbabaie et al. in [10] under a joint rooted leadership via the $(sp)$ matrices. This model deals with self-propelled particles moving in the plane with the same speed but different headings interacting with neighboring agents by a linear relaxation rule. When the time-varying switching topology of the neighbor graph satisfies some weak connectivity condition, namely, `` joint connectivity condition'' in the spatial-temporal domain, it is well known that the consensus for the linearized Vicsek model can be achieved asymptotically. In this paper, we extend the theory of $(sp)$ matrices and apply it to revisit this asymptotic consensus problem and give an explicit estimate on the maximum Lyapunov exponent, when the underlying network topology satisfies the joint rooted leadership which is directed and non-symmetric.

Abstract:The separation and purification of sucrose6-acetate from acetylation product of sucrose by PUROLITE PCR(Ca) ion exchange resin were invesigaged.It was found that the adsorption velocity of sucrose-6-acetate on ion exchange resin was corresponding to sucrose,but the dynamic permeable adsorption capacity of sucrose was larger than that of sucrose-6-caetate.The resolution of the samples on column chromatography could generally be enhanced with the increase of the ration of column height to diameter,decreased with the increase of the loading quantity and the flowing velocity.Under the typical separation operation conditions,the content of sucrose-6-acetate could reach about 94 from 80.7%,and the average separation yield was about 85.0%.

Abstract:An HPLC-based method for quantitative determination of the 4 metabolites of gluconic acid(GlcA),2-keto-D-gluconic acid(2-KDG),2,5-diketo-D-gluconic acid(2,5-DKG)and 2-keto-L-gulonic acid(2-KLG)from direct fermentation of D-glucose to 2-KLG by using HPLC was established.The optimum separation of the 4 substances could be achieved under the conditions of using Aminex HPX-87H column(size 300 mm×7.8 mm)with the temperature of 30 ℃,sulfuric acid(50 mmol/L) as mobile phase at the flow rate of 0.2 mL/min,RID-10A detector,and malonic acid as internal standard.The analytical results show that the method has average analytical deviation of 1.53%,0.89%and 1.84%on 2-KDG,2,5-DKG and 2-KLG,respectively.

Abstract:Chromatographic analysis of orange and apple juices is provided using novel covalently-bonded poly(styrene-divinylbenzene)-based (PS-DVB) hyperbranched anion exchanger for suppressed ion chromatography (IC) with improved selectivity toward inorganic anions and organic acids. The obtained stationary phase provides baseline resolution of weakly retained organic acids such as glycolate, acetate, lactate, and formate, which are not separated to baseline with modern commercially available anion exchangers. The proposed method is validated with respect to linearity, recovery, limits of detection, and intra-day and inter-day precision.

Abstract:Ion exchange chromatography with integral pulse amperometric detection was applied to the determination of glucose,fructose,sucrose,lactose and maltose in food.Retention property of the 5 saccharides in the elution with NaOH solution was studied.CarboPacTM PA 10 column was used as separation column,and mixtures of pure water and 0.2 mol·L-1 NaOH solution in different ratio were used as mobile phase in the gradient elution.The flow rate taken was 0.5 mL·min-1.In the amperometric detection,gold electrode was used as working electrode,Ag/AgCl electrode was used as reference electrode.Under the optimum conditions,the 5 saccharides in a sample were separated and determined effectively and simultaneously.Linearity ranges for all the 5 saccharides were obtained in the range of 0.5-10.0 mg·L-1.Detection limit(3S/N) found were in the range of 0.8-2.5 μg·L-1.Test for recovery was made by addition of the standards of the 5 saccharides to a substantial sample as the matrix,values of recovery obtained were in the range of 91.1%-104.8%.

Abstract:The method for separation and determination of several di- and tricarboxylic acids, including oxalic, malic, tartaric and citric acids (OX, MA, TA and CI), found in fruit juices by capillary zone electrophoresis with amperometric detection at a copper electrode was developed. Based on cyclic voltammetry (CV) experiments, the detection mechanism for the analytes at a copper electrode was proved to be the formation of complexes between di- and tricarboxylic acids and Cu(II) ion on the electrode surface. Separation was carried out in a pH 6.2 phosphate buffer solution (PBS) containing 0.4mmol/l cetylpridinium bromide (CPB) and 20mmol/l β-cyclodextrin (β-CD) as an electro-osmotic flow (EOF) modifier to reverse the direction of the EOF and a complexing reagent for CPB to improve the separation, respectively. At the working potential of 0.12V (versus SCE), the calibration curves for the analytes were linear with about three orders of magnitude and the detection limits (S/N=3) for them were between 4 and 8fmol. By the method developed here, di- and tricarboxylic acids in four fruit juices, apple, grape, pear and orange juices, were successfully separated and determined.

Abstract:Produced in Madeira Island from regional sugarcane cultivars through a traditional manufacturing and storage process, sugarcane honey (SCH) is a black syrup recognized by its excellent quality. Its economic value has led to the emergence of adulterated SCH, whereby the identification of molecular markers became an essential task in order to overcome the fraudulent activities, protect its authenticity, and guarantee the consumer safety. In the present study, an analytical strategy based on ultrasound-assisted liquid-liquid extraction (USA-LLE) followed by reversed-phase liquid chromatography with a refractive index detector (LC-RI) was developed for the determination of sugars (glucose, fructose, sucrose, xylose, and mannose) in SCH samples from certified producers, supported on analytical quality-by-design (AQbD) approach, as a useful tool to establish its typicality. The application of AQbD was based on analytical risk assessment, multivariate statistics and quality control procedures for definition of the Method Operable Design Region (MODR). The optimal conditions into MODR were accomplished using BEH Amide column operating at a temperature of 80 °C and a flow rate of 300 μL min−1, with a mobile phase composed by acetone and water (85:15, v v−1) at a flow rate of 1.0 mL min−1. The robustness was determined by Monte Carlo simulation and capability analysis. The concentration-response function for all sugars was described by polynomial models. Accuracy was presented by recovery values between 98.2 and 119.5%. The analytical figures of merit validated the utility of AQbD in the systematic design of a LC-RI method with fine sensitivity for sugar analysis in SCH.

Abstract:Sugar can be directly determined without derivation reaction by ion exchange chromatography-pulsed ampere detector, in which the eluent is 22.5 mmol/L NaOH, a gold electrode is work electrode and Ag/AgCl electrode is reference electrode. L-rhamnose, L-arabinose, D-galactose, glucose, xylose ,D-fructose can be well separated. Experimental results show that the linear relationship and repeatability are good. The method is used to determine low component and complicated samples, and the results are satisfactory.

Abstract:A novel and reliable high-performance liquid chromatographic method for trace short-chain organic acids measurement in wheat root exudates under aluminum stress has been optimized and validated. The chromatographic separation of the short-chain organic acids (citric, oxalic, malonic, succinic, tartaric, malic, and acetic acids) was achieved with Bio-rad Aminex HPX-87H cation exchange resin column. These seven organic acids were identified and quantified in 25 min. Well-shaped peaks were obtained for trace organic acids using dilute sulfuric acid as mobile phase. Under optimum conditions, Bio-rad Aminex HPX-87H column showed distinct advantages of the ability to well separate different short-chain organic acids (especially for tartaric and malic acids, as well as malonic and acetic acids) in wheat root exudates under aluminum stress, and offered accurate and precise results for the analysis of these organic acids. This HPLC method can efficiently eliminate the aluminum's interference and is quite suitable to the trace detection of trace organic acids in wheat root exudates under aluminum stress.

Abstract:Assessing central carbon metabolism in plants can be challenging due to the dynamic range in pool sizes, with low levels of important phosphorylated sugars relative to more abundant sugars and organic acids. Here, we report a sensitive liquid chromatography-mass spectrometry method for analysing central metabolites on a hybrid column, where both anion-exchange and hydrophilic interaction chromatography (HILIC) ligands are embedded in the stationary phase. The liquid chromatography method was developed for enhanced selectivity of 27 central metabolites in a single run with sensitivity at femtomole levels observed for most phosphorylated sugars. The method resolved phosphorylated hexose, pentose, and triose isomers that are otherwise challenging. Compared with a standard HILIC approach, these metabolites had improved peak areas using our approach due to ion enhancement or low ion suppression in the biological sample matrix. The approach was applied to investigate metabolism in high lipid-producing tobacco leaves that exhibited increased levels of acetyl-CoA, a precursor for oil biosynthesis. The application of the method to isotopologue detection and quantification was considered through evaluating 13C-labeled seeds from Camelina sativa. The method provides a means to analyse intermediates more comprehensively in central metabolism of plant tissues.

Abstract:Analysis of glucose and other carbohydrates are often performed by use of normal phase HPLC methods with acetonitrile as major eluent coupled with evaporative light-scattering detector (ELSD) or by use of anion-exchange ion chromatography (IC) methods with NaOH as eluent coupled with pulsed amperimetric electrochemical detector. In this work, a novel method for the determination of carbohydrates by IC in conjunction with a self-regenerating suppressor and an ELSD detector was investigated. Three carbohydrates (glucose, fructose, and sucrose) were separated using a KOH eluent generator to avoid the effect of carbon dioxide absorption in the alkaline eluent. Due to the use of the suppressor, non-volatile components were removed and a low salt background (K+ approximately 0.070 microg/mL) can be obtained so the suppressed eluent could directly go into an ELSD detector without obvious interference of inorganic salts. After examining the changes in retention and resolution, an optimized method was established (for IC: using 32 mM KOH as the eluent at a flow rate of 1 mL/min; for ELSD: operated at 95 degrees C, 4.0 bar nitrogen with a gas flow rate of 2.0 L/min) and the linearity, reproducibility, and the limit of detection (LOD) for the three carbohydrates were further evaluated. Regression equations revealed acceptable linearity (correlation coefficients=0.994-0.998) across the working-standard range (100-1000 microg/mL for glucose and sucrose, 150-1000 microg/mL for fructose) and LODs of glucose, fructose, and sucrose were 93, 126, and 90 microg/mL, respectively. This method has successfully been applied to the determination of the three carbohydrates in carbonated cola drinks and fruit juices. The recoveries were between 95 and 113% (n=3) for different carbohydrates.

Abstract:This paper presents a simple analytical method for determining sugars in soybean ( Glycine max (L.) Merr.) tissues. Sample preparation was modified from several early published methods. High-performance liquid chromatography (HPLC) equipped with an evaporative light scattering detector (ELSD) was used to separate, identify, and quantify seven sugars, including glucose, galactose, fructose, sucrose, melibiose, raffinose, and stachyose. Two mobile phases were programed into a gradient elution. Mobile phase A is pure water and mobile phase B is a mixture of acetonitrile and acetone 75 : 25 (v/v). Total chromatographic retention time is less than 20 minutes. This method has been validated for detection limit, calibration range, and intraday and interday repeatability. This method has been used analyzing more than 5000 soybean samples in the experiments studying natural genetic variation of sugar contents and components in soybean seeds and other tissues.
