Abstract:The aim of this work was to develop a novel strategy for improving the vanillin tolerance and ethanol fermentation performances of Saccharomyces cerevisiae strains. Isogeneic diploid, triploid, and tetraploid S. cerevisiae strains were generated by genome duplication of haploid strain CEN.PK2-1C. Ploidy increments improved vanillin tolerance and diminished proliferation capability. Antimitotic drug methyl benzimidazol-2-ylcarbamate (MBC) was used to introduce chromosomal aberrations into the tetraploid S. cerevisiae strain. Interestingly, aneuploid mutants with DNA contents between triploid and tetraploid were more resistant to vanillin and showed faster ethanol fermentation rates than all euploid strains. The physiological characteristics of these mutants suggest that higher bioconversion capacities of vanillin and ergosterol contents might contribute to improved vanillin tolerance. This study demonstrates that genome duplication and MBC treatment is a powerful strategy to improve the vanillin tolerance of yeast strains.
Optimization of vanillin biosynthesis in Escherichia coli K12 MG1655 through metabolic engineering

Abstract:Vanillin is an important flavouring agent applied in food, spices, pharmaceutical industries and other fields. Microbial biosynthesis of vanillin is considered a sustainable and economically feasible alternative to traditional chemical synthesis. In this study, Escherichia coli K12 MG1655 was used for the de novo synthesis of VAN by screening highly active carboxylic acid reductases and catechol O-methyltransferases, optimising the protocatechuic acid pathway, and regulating competitive metabolic pathways. Additionally, major alcohol by-products were identified and decreased by deleting three endogenous aldo-keto reductases and three alcohol dehydrogenases. Finally, a highest VAN titer was achieved to 481.2 mg/L in a 5 L fermenter from glucose. This work provides a valuable example of pathway engineering and screens several enzyme variants for the first time in E. coli.

Abstract:AbstractVanillin is one of the most commonly used natural-occurring flavors in the world. This study successfully constructed an efficient whole-cell catalytic system for vanillin biosynthesis from ferulic acid by regulating feruloyl-CoA synthetase (FCS) and enoyl-CoA hydratase (ECH). First, we constructed an efficient cell-free catalytic system with FCS-Str (fcs from Streptomyces sp. V-1) and ECH-Str (ech from Streptomyces sp. V-1) combination at 1:1. The efficient cell-free catalytic system provided necessary strategies for optimizing the whole-cell catalytic system. Then, we constructed the recombinant Escherichia coli by heterologously expressing the fcs-Str and ech-Str combination. Moreover, E. coli JM109 was a better recombinant Escherichia coli than E. coli BL21 with higher vanillin production. Finally, we first adjusted the ratio of FCS and ECH in E. coli JM109 to 1:1 using two copies of fcs-Str. For higher vanillin production, we further optimized the induction conditions of E. coli JM109 to increase the amount of FCS and ECH. The optimized E. coli JM109-FE-F constructed in this study has the highest vanillin synthesis rate of converting 20 mM ferulic acid to 15 mM vanillin in 6 h among all of the E. coli catalytic systems. Abstract:Vanillin is one of the world's most extensively used flavoring agents with high application value. However, the yield of vanillin biosynthesis remains limited due to the low efficiency of substrate uptake and the inhibitory effect on cell growth caused by vanillin. Here, we screened high-efficiency ferulic acid importer TodX and vanillin exporters PP_0178 and PP_0179 by overexpressing genes encoding candidate transporters in a vanillin-producing engineered Escherichia coli strain VA and further constructed an autoregulatory bidirectional transport system by coexpressing TodX and PP_0178/PP_0179 with a vanillin self-inducible promoter ADH7. Compared with strain VA, strain VA-TodX-PP_0179 can efficiently transport ferulic acid across the cell membrane and convert it to vanillin, which significantly increases the substrate utilization rate efficiency (14.86%) and vanillin titer (51.07%). This study demonstrated that the autoregulatory bidirectional transport system significantly enhances the substrate uptake efficiency while alleviating the vanillin toxicity issue, providing a promising viable route for vanillin biosynthesis.

Abstract:With special fragrance and various biological properties, linalool has wide applications in food, pharmaceuticals and cosmetics industries. Microbial biosynthesis has become a promising approach to linalool production, however its efficiency is limited by the limited precursor supply and poor efficiency of linalool synthase. With the goal of enhancing heterologous linalool production in Saccharomyces cerevisiae, protein engineering and metabolic engineering were simultaneously employed to solve these bottlenecks. Linalool-producing yeast was first constructed by introduction of t670McLIS selected out from a collection of linalool synthases. The efficiency of linalool biosynthesis was then improved by strengthening the supply of geranyl pyrophosphate (GPP) as precursor via overexpression of the complete mevalonate (MVA) pathway and a GPP synthase variant. To accelerate the conversion of the accumulated GPP to linalool, t670McLIS was engineered by means of directed evolution after development of a competition-based color-indicated high-throughput screening method. Expression of the t670McLIS variant in the engineered S. cerevisiae with enhanced precursor supply yielded 53.14 mg/L of linalool in biphasic shake-flask culture. This study delivered an efficient linalool-producing yeast cell factory and meanwhile provided efficient strategies for biosynthesis of other valuable monoterpenes.

Abstract:Linalool, a valuable monoterpene alcohol, is widely used in cosmetics and flavoring ingredients. However, its scalable production by microbial fermentation is not yet achieved. In this work, considerable increase in linalool production was obtained in Saccharomyces cerevisiae by dual metabolic engineering of the mevalonic acid (MVA) pathway in both mitochondria and cytoplasm. A farnesyl pyrophosphate synthase mutant ERG20(F96W/N127W) and a linalool synthase from Cinnamomum osmophloeum (CoLIS) were introduced and meanwhile the endogenous ERG20 was down-regulated to prevent the competitive loss of precursor. In addition, overexpression of the proteins of CoLIS and ERG20(F96W/N127W) and another copy of the same enzymes CoLIS/ERG20(F96W/N127W) with mitochondrial localization signal (MLS) were carried out to further pull the flux to linalool. Finally, a maximum linalool titer of 23.45 mg/L was obtained in a batch fermentation with sucrose as carbon source. This combinatorial engineering strategy may provide hints for biosynthesis of other monoterpenes.

Abstract:Vanillin holds significant importance as a flavoring agent in various industries, including food, pharmaceuticals, and cosmetics. The CoA-dependent pathway for the biosynthesis of vanillin from ferulic acid involved feruloyl-CoA synthase (Fcs) and enoyl-CoA hydratase/lyase (Ech). In this research, the Fcs and Ech were derived from Streptomyces sp. strain V-1. The sequence conservation and structural features of Ech were analyzed by computational techniques including sequence alignment and molecular dynamics simulation. After detailed study for the major binding modes and key amino acid residues between Ech and substrates, a series of mutations (F74W, A130G, A130G/T132S, R147Q, Q255R, ΔT90, ΔTGPEIL, ΔN1-11, ΔC260-287) were obtained by rational design. Finally, the yield of vanillin produced by these mutants was verified by whole-cell catalysis. The results indicated that three mutants, F74W, Q147R, and ΔN1-11, showed higher yields than wild-type Ech. Molecular dynamics simulations and residue energy decomposition identified the basic residues K37, R38, K561, and R564 as the key residues affecting the free energy of binding between Ech and feruloyl-coenzyme A (FCA). The large changes in electrostatic interacting and polar solvating energies caused by the mutations may lead to decreased enzyme activity. This study provides important theoretical guidance as well as experimental data for the biosynthetic pathway of vanillin.

Abstract:Vanillin is a popular flavoring agent widely used around the world. Vanillin is generated by natural extraction, chemical synthesis, or tissue culture technology, but these production methods no longer meet the increasing worldwide demand for vanillin. Accordingly, a biotechnological approach may provide an effective replacement route to obtaining vanillin. Processes for environmentally friendly production of vanillin in microorganisms from different carbon sources, such as eugenol, isoeugenol, lignin, ferulic acid, sugars, and waste residues, with high productivity and yield have been developed. However, challenges remain for optimizing the vanillin biosynthesis process and further improving production titer and yield. In this review, successful and applicable strategies for increasing vanillin titer and yield in different microorganisms are summarized. Additionally, perspectives for further optimizing the production of vanillin are discussed.Graphical abstract

Abstract:minal Gln amide transfer (GAT) domain and is promoted by the allosteric effector GTP. 3] The resulting nascent NH3 is subsequently transferred to the N-terminal synthase domain via an NH3 tunnel, [4–6] where it reacts with UTP that has been activated by ATP-dependent phosphorylation at the 4-position (i.e. , UTP-4-P). The enzyme exhibits positive cooperativity for ATP and UTP, and these nucleotides act synergistically to promote tetramerization of the enzyme to its active form. The product CTP is a feedback inhibitor of the enzyme and plays an important role in the biosynthesis of nucleic acids, phospholipids, 12] and sialic acid. Consequently, CTPS is a recognized target for the development of antineoplastic, antiviral, and antiprotozoal agents. Although many studies have focused on delineating the regulatory properties of CTPS, very few have focused on the development of nucleotide analogues as inhibitors of this enzyme. 24] 3-Deazauridine 5’-triphosphate (IC50 ~18 mm) and cyclopentenyl cytosine (CPEC) 5’-triphosphate (IC50 ~6 mm) are substrate and product analogues, respectively, that are the most well-studied inhibitors of CTPS. In addition, the 5’-triphosphate of 2’,2’-difluorodeoxycytidine (gemcitabine) is also believed to exert its chemotherapeutic effects, in part, through the inhibition of CTPS. 28] However, mutations in CTPS lead to the loss of feedback inhibition by CTP and resistance to the cytotoxic effects of these and other chemotherapy drugs. Therefore, the development of new, potent, and selective CTPS inhibitors such as transition state/intermediate analogues or multivalent inhibitors is required. Enzymes often exhibit high affinity for structural and electronic mimics of transition states or reactive intermediates generated during catalysis. We rationalized that CTPS inhibitors might be developed by employing methylene or difluoromethylene phosphonate groups, or a phosphoramidate group as isosteric and/or isoelectronic replacements for the 4-phosphate group of the UTP-4-P intermediate (Scheme 1). Alternatively, the use of multivalent ligands (e.g. , substrate– product analogues) is often an effective strategy to develop potent and specific enzyme inhibitors, as such inhibitors may interact with multiple binding determinants on the enzyme to afford an entropic advantage in binding. Abstract:Vanillin (4-hydroxy-3-methoxybenzaldehyde) is one of the most popular flavors with wide applications in food, fragrance, and pharmaceutical industries. However, the high cost and limited yield of plant extraction failed to meet the vast market demand of natural vanillin. Vanillin biotechnology has emerged as a sustainable and cost-effective alternative to supply vanillin. In this review, we explored recent advances in vanillin biosynthesis and highlighted the potential of vanillin biotechnology. In particular, we addressed key challenges in using microorganisms and provided promising approaches for improving vanillin production with a special focus on chassis development, pathway construction and process optimization. Future directions of vanillin biosynthesis using inexpensive precursors are also thoroughly discussed.

Abstract:AIMS As statin therapy has been demonstrated to augment endothelial function in sedentary hypercholesterolaemia (HC), we aimed to investigate the effects of atorvastatin therapy on endothelial function in physically active, HC men. METHOD AND RESULTS Eleven physically active, HC males were recruited. Endothelial function [forearm blood flow response to brachial artery infusion of acetylcholine (Ach)] was assessed twice in each subject following atorvastatin or no therapy in a randomized crossover design. In addition, endothelial function was compared with an active, normolipidaemic control group (C). Atorvastatin therapy reduced total and LDL cholesterol, but had no effect on basal blood flow or endothelial function (peak ACh mean difference +/- standard error 0.75 +/- 1.75 ml min(-1) per 100 ml tissue) [95% confidence interval (CI) -3.1, 4.6]. In addition, there was no difference in endothelial function between the HC and C groups (-1.14 +/- 2.60 ml min(-1) per 100 ml tissue; CI -6.53, 4.25). CONCLUSION Statin therapy in HC patients with normal endothelial function does not augment endothelial function.

Abstract:Currently, the biotechnological preparation of fragrances using natural materials attracted growing attention. Enzymatic synthesis of vanillin from isoeugenol by recombinant isoeugenol monooxygenase from Pseudomonas nitroreducens Jin1 was systematically investigated herein. With series of work on the construction of recombinant E. coli over-expressing isoeugenol monooxygenase, optimization of the culture conditions for enzyme production and reaction process for converting isoeugenol into vanillin, an increase of 22-fold in the enzyme activity (2050 U/L) was obtained, and the conversion was significantly increased at high substrate concentration with the aid of magnetic chitosan membrane for product isolation in situ. Under optimal conditions, the product concentration and space-time yield reached 252 mM and 115 g/L/d, respectively, and vanillin was obtained in 82.3% yield and > 99% purity in the gram preparative scale. The developed bioprocess showed application potential for efficient preparation of vanillin from inexpensive natural resources.

Abstract:Engineering Escherichia coli for efficient methanol assimilation is important for developing methanol as an emerging next-generation feedstock for industrial biotechnology. While recent attempts to engineer E. coli as a synthetic methylotroph have achieved great success, most of these works are based on the engineering of the prokaryotic ribulose monophosphate (RuMP) pathway. In this study, we introduced a hybrid methanol assimilation pathway which consists of prokaryotic methanol dehydrogenase (Mdh) and eukaryotic xylulose monophosphate (XuMP) pathway enzyme dihydroxyacetone synthase (Das) into E. coli and reprogrammed E. coli metabolism to improve methanol assimilation by combining rational design and adaptive laboratory evolution. By deletion and down-regulation of key genes in the TCA cycle and glycolysis to increase the flux toward the cyclic XuMP pathway, methanol consumption and the assimilation of methanol to biomass were significantly improved. Further improvements in methanol utilization and cell growth were achieved via adaptive laboratory evolution and a final evolved strain can grow on methanol with only 0.1 g/L yeast extract as co-substrate. C-13-methanol labeling assay demonstrated significantly higher labeling in intracellular metabolites in glycolysis, TCA cycle, pentose phosphate pathway, and amino acids. Transcriptomics analysis showed that the expression of fba, dhak, and part of pentose phosphate pathway genes were highly up-regulated, suggesting that the rational engineering strategies and adaptive evolution are effective for activating the cyclic XuMP pathway. This study demonstrated the feasibility and provided new strategies to construct synthetic methylotrophy of E. coli based on the hybrid methanol assimilation pathway with Mdh and Das.

Abstract:Efficient and economical supply of cis-abienol has attracted extensive attention due to its use as a precursor in the synthesis of ambrox (a cardinal odor of rare ambergris). Increasing awareness in environmental issues and advantages of microbial production–sustainable and green manufacture, have motivated efforts to advance the cis-abienol biosynthesis in more affordable ways. Herein, Escherichia coli cells were employed to scale up cis-abienol production via expressing the concise isopentenol utilization pathway (IUP). Alcohol kinases and isopentenyl phosphate kinases (IPKs) were screened and coordinated to increase dimethylallyl diphosphate (DMAPP) supply, which resulted in approximately 37-fold improvements in the cis-abienol yield, compared with the titer obtained by endogenous MEP pathway. Next, we constructed a double-phosphatase-deficient strain (BD203), resulting in a 40% increase in the cis-abienol titer relative to wild-type strains. By optimization of addition strategies, a cis-abienol titer of 311.8 ± 1.0 mg/L was obtained in strain BD203, which is the highest cis-abienol titer reported in shake flasks and 14.7% of the maximum theoretical yield. Finally, further optimization of the fed-batch fermentation process enabled a cis-abienol titer of 1375.7 mg/L in a 1.3 L bioreactor. Overall, BD203 achieved a substantial increase in the cis-abienol titer in this study and engineering microbes were expected to facilitate progress toward industrial production.

Abstract:Valorizing lignocellulosic biomass (LCB) into platform aromatic chemicals could contribute to a sustainable biomass-based bioeconomy. However, lacking simultaneous bioconversion route of lignin and carbohydrates hinders the LCB utilization. The artificial biological funneling pathway (ABFP) was hence designed to convert full components of LCB into vanillin. The results showed that combinational engineering strategies enabled the successful accumulation of vanillin in Saccharomyces cerevisiae . The funneling pathways of lignin-derived monomers to vanillin were constructed. Enzyme fusion and S-adenosylmethionine metabolism regulation of heterologous rate-limiting enzyme further promoted the vanillin production to 2.49 and 1.94 mmol/L from ferulic acid and p -coumaric acid, respectively The construction of funneling pathways of hexose and pentose promoted the carbon flux towards vanillin biosynthesis. Together with the regulation of branch and shikimic acid pathways, the microbial cell factory with a novel ABFP produced 1.97 mmol/L vanillin from real LCB hydrolysates with a recorded yield of 10.5 mg vanillin/g carbon source Overall, the engineered S. cerevisiae simultaneously valorized full components into fine aromatic chemicals, holding the promise to transform biorefinery mode and contribute to the biomass-based economy.

Abstract:L-valine is an essential amino acid and an important amino acid in the food and feed industry. The relatively low titer and low fermentation yield currently limit the large-scale application of L-valine. Here, we constructed a chromosomally engineered Escherichia coli to efficiently produce L-valine. First, the synthetic pathway of L-valine was enhanced by heterologous introduction of a feedback-resistant acetolactate acid synthase from Bacillus subtilis and overexpression of other two enzymes in the L-valine synthetic pathway. For efficient efflux of L-valine, an exporter from Corynebacterium glutamicum was subsequently introduced. Next, the precursor pyruvate pool was increased by knockout of GTP pyrophosphokinase and introduction of a ppGpp 3′-pyrophosphohydrolase mutant to facilitate the glucose uptake process. Finally, in order to improve the redox cofactor balance, acetohydroxy acid isomeroreductase was replaced by a NADH-preferring mutant, and branched-chain amino acid aminotransferase was replaced by leucine dehydrogenase from Bacillus subtilis. Redox cofactor balance enabled the strain to synthesize L-valine under oxygen-limiting condition, significantly increasing the yield in the presence of glucose. Two-stage fed-batch fermentation of the final strain in a 5 L bioreactor produced 84 g/L L-valine with a yield and productivity of 0.41 g/g glucose and 2.33 g/L/h, respectively. To the best of our knowledge, this is the highest L-valine titer and yield ever reported in E. coli. The systems metabolic engineering strategy described here will be useful for future engineering of E. coli strains for the industrial production of L-valine and related products.

Abstract:For efficiently enhancing the activity of isoeugenol monooxygenase, a whole cell overproducing active aggregate IEM720-18A was successfully fabricated via the fusion of amphiphilic short peptide 18A (EWLKAFYEKVLEKLKELF) and isoeugenol monooxygenase and then efficiently expressed in E. coli BL21 (DE3). Such resulting bacteria, E. coli BL21 (DE3) harboring pET30a-IEM720-18A was applied in the biotransformation of isoeugenol to vanillin with the optimization of cultivation conditions. Our results revealed that the vanillin concentration reached to the highest level (14.5 mmol/L) under the optimized reaction conditions including 1.5-g cells containing active aggregate of IEM720-18A, 10% (v/v) dimethyl sulfoxide (DMSO), 100 mmol/L isoeugenol, 50 mmol/L glycine-sodium hydroxide buffer (pH 10.5) in 10 mL reaction volume, and 200 rpm at 25 °C for 36 h. Moreover, the active aggregate IEM720-18A was immobilized with 100 mmol/L glutaraldehyde at 4 °C to improve the operational stability. The highest activity could be achieved when the reactions were carried out at 25 °C and the relative activity of the immobilized enzyme maintained over 60% after seven recycles. Our study provides a new approach to the biotransformation of isoeugenol into vanillin.

Abstract:Vanillylamine, as an important drug precursor and fine chemical intermediate, has great economic value. By constructing a strategy of double enzyme co-expression, one newly constructed recombinant E. coli HNIQLE-AlaDH expressing ω-transaminase from Aspergillus terreus and alanine dehydrogenase from Bacillus subtilis was firstly used aminate lignin-derived vanillin to vanillylamine by using a relatively low dosage of amine donors (vanillin:L-alanine:isopropylamine = 1:1:1, mol/mol/mol). In addition, in a two-phase system (water:petroleum ether = 80:20 v/v), the bioconversion of vanillin to vanillylamine was catalyzed by HNIQLE-AlaDH cell under the ambient condition, and the vanillylamine yield was 71.5%, respectively. This double-enzyme HNIQLE-AlaDH catalytic strategy was applied to catalyze the bioamination of furfural and 5-hydroxymethylfurfural with high amination efficiency. It showed that the double-enzyme catalytic strategy in this study promoted L-alanine to replace D-alanine to participate in bioamination of vanillin and its derivatives, showing a great prospect in the green biosynthesis of biobased chemicals from biomass.

Abstract:In hydrothermal liquefaction (HTL) of biomass, only the oil and the char phases are considered imperative while the aqueous phase is discarded. However, the aqueous phase is rich in phenolic compounds like guaiacol and vanillyl alcohol that can be upgraded to vanillin. Commercially vanillin is synthesized from phenolic compounds derived from fossil fuels using chemical synthesis methods. In this investigation, the method was developed to use raw materials and enzymatic conversion of substrates which is sustainable and renewable. The purpose of this investigation is to evaluate the applicability of enzymatic vanillin synthesis within the HTL aqueous phase major components. Vanillyl alcohol oxidase was expressed in E. coli BL21 (DE3) bacteria cells and purified to characterize the production of vanillin from vanillyl alcohol. The effect on the enzyme reaction of each of the major components of the aqueous phase as detected by HPLC (High Performance Liquid Chromatography) analysis was evaluated. The components evaluated in this study included different phenolic compounds namely guaiacol, vanillic acid, vanillyl alcohol, phenol, catechol and acetic acid. These components were evaluated individually as well as in different combinations. Vanillic acid increased the reaction rate while guaiacol inhibited the reaction rate. Acetic acid exhibited the largest inhibition of the enzyme reaction after 15 min. It is known that acids are often used to stop enzyme reactions as it results in precipitation of the enzyme. Therefore, to successfully convert vanillin from vanillyl alcohol using a recombinantly expressed vanillyl alcohol oxidase, the acetic acid would first need to be removed from the HTL aqueous phase.

Abstract:SAMs based on a novel dendron-thiol system, which maintain the alkanethiols' active site, but with the -SH group connected to independently variable groups by a dendron-linker, showed a controllable surface pattern and wetting properties. The precisely tailored structure of dendron-thiols with locally controlled hydrophobic and hydrophilic peripheries allows the formation of designed surface structures on a gold surface, e.g. nano-stripes, honeycomb and homogeneous structures.
