Abstract:This study investigated the impact of micropollutants on fish health from Segredo hydroelectric reservoir (HRS) along the Iguaçu River, Southern Brazil, contaminated by urban, industrial, and agricultural activities. This is the first comprehensive study assessment in the river after the severe drought in the 2020s in three fish species from different trophic levels Astyanax spp. (water column depth/omnivorous), Hypostomus commersoni (demersal/herbivorous), and Pimelodus maculatus (demersal/omnivorous). Animals, water, and sediment samples were collected from three distinct sites within the reservoir: Floresta (upstream), Iratim (middle), and Station (downstream). The chemical analysis revealed elevated concentrations of metals (Al, Cu, Fe) and the metalloid As in water, or Cu, Zn, and As in sediment, surpassing Brazilian regulatory limits, while the organic pollutants as DDT, PAHs, PCBs, and PBDEs were found under the Brazilian regulatory limits. The metal bioaccumulation was higher in gills with no significant differences among sites. The species Astyanax spp and H. commersoni displayed variations in hepatosomatic index (HSI) and P. maculatus in the condition factor index ( K ) between sites, while adverse effects due to micropollutants bioaccumulation were observed by biochemical, genotoxic, and histopathological biomarkers. The principal component analysis and integrated biomarker response highlighted the upstream site Floresta as particularly inhospitable for biota, with distinctions based on trophic level. Consequently, this multifaceted approach, encompassing both fish biomarkers and chemical analyses, furnishes valuable insights into the potential toxic repercussions of micropollutant exposure. These findings offer crucial data for guiding management and conservation endeavors in the Iguaçu River.

Abstract:The rivers and streams of the large urban centers in Southeast Brazil are increasingly being degraded, demanding expanded conservation efforts. This study was conducted in the Grande River, one of the main tributaries of the Billings Complex, a reservoir that is a strategic fresh water resource for the São Paulo metropolitan region. Water quality, habitat features and fish fauna were investigated at seven sites along the longitudinal gradient with the aim of identifying the distribution patterns and relative contributions of the environmental factors. The water samples and environmental characteristics were recorded, and fish were collected during the rainy (January to March) and dry seasons (July and August) of 2009. The water quality varied along the river, with higher values of conductivity, fecal coliforms and total phosphorus in the lower reach, indicating a strong influence of the urban area. Twenty-two fish species were recorded, two of which are considered endangered. A canonical correspondence analysis (CCA) indicated marked differences in species composition between the river’s upper and lower reaches, which was mainly attributed to vegetation cover and the presence of different meso-habitats, such as riffles and pools. Trychomycterus spp. and Astyanax paranae were associated with the upper reaches, while Astyanax fasciatus and Astyanax bockmanni, Cyphocharax modestus, Hoplias malabaricus and Hypostomus ancistroides occurred in the lower reaches. Despite the disturbance in water quality and riparian vegetation in the lower river section, no detectable changes in community structure were observed. However, the presence of some tolerant species, such as Astyanax fasciatus, Hoplosternum littorale and Hypostomus ancistroides, may indicate that the community is experiencing initial stages of disturbance.

Abstract:The amino acid sequence and structure of a uterotonic polypeptide extracted from the African plant Oldenlandia affinis DC have been determined. The peptide, kalata B1, consists of 29 amino acid residues and is rich in cysteine (6), threonine (5), and glycine (5). Enzyme cleavage studies show that the polypeptide backbone is cyclic. The three-dimensional solution structure has been determined using two-dimensional nuclear magnetic resonance (NMR) spectroscopy and distance-restrained simulated annealing. Kalata B1 is composed mainly of beta-strands connected by tight turns, forming regions of beta-sheet, except in the case of one section which forms a longer, less structured loop. The tertiary fold, together with the disulfides that form a sulfur core, produces a striking and unusual surface in which the majority of the hydrophobic residues form a solvent-exposed patch. The hydrophobic side of kalata B1 is flanked by two diametrically opposed and opposite-charged residues. The structure calculations have been used to predict the previously unknown disulfide bond connectivities using two approaches. In the first, a family of structures was calculated on the basis of NOE constraints without the assumption of a specific disulfide connectivity. The resultant structures were examined to determine whether the calculated position of the sulfur atoms suggested that one set of disulfide connectivities was more likely than the other, theoretically possible, sets. In the second approach, a separate family of structures (50 per set) was calculated for each of the 15 possible disulfide-bonded molecules. The resultant families of structures were compared to see whether one was favored over the others. Both approaches led to the same global fold, and the most likely disulfide connectivity is predicted to be 5-22, 13-27, and 17-29. In the calculated structure the cyclic peptide backbone is folded back onto itself and braced with disulfide pairs across diagonally opposed beta-strands. This structure involves one of the disulfide bonds (5-22) threading through the eight amino acid loop formed by the other two disulfide bonds and the peptide fragments connecting them.

Abstract:This study aimed to isolate and identify pathogenic bacteria in the intestinal tract, skin, and muscles of Sciades herzbergii ; detect histopathological changes in the gill and liver; and use these biomarkers for the assessment of potential risks to human health. Fish were sampled during the rainy and dry seasons at two points in São Marcos Bay, Maranhão, Brazil: Ilha dos Caranguejos (IC) and Porto Grande (PG). Isolation and quantification were carried out using COLItest®. Colonies were subjected to identification and phenotypic investigation of antimicrobial resistance using Vitek®. Gill and liver samples were subjected to routine histological examination. The results indicated the presence of Klebsiella pneumoniae and Escherichia coli , the latter of which showed phenotypic resistance to norfloxacin and gentamicin. Fish caught at PG exhibited more extensive gill and liver damage than fish caught at IC. The findings suggest that histological changes in target organs of S. herzbergii may be influenced by infection with pathogenic bacteria

Abstract:Two of the largest water reservoirs in the Metropolitan Region of São Paulo, Brazil (MRSP), named Billings and Guarapiranga, are facing high levels of anthropic impact. This is evidenced by the presence of contaminants and pollutants, which are deteriorating their water quality. Therefore, this study evaluated antioxidant defense enzymes, lipoperoxidation and genotoxicity, in adult females of a native species, Astyanax altiparanae from the Guarapiranga and Billings reservoirs. The study also aimed to evaluate these biomarkers during two different periods of the year, the rainy (summer) and dry (winter) seasons. The oxidative stress was evaluated by the activity of enzymes such as glutathione peroxidase, glutathione S-transferases, superoxide dismutase, and catalase in the gills and liver, and the occurrence of lipoperoxidation was also evaluated in both organs. The genotoxicity was assessed by performing comet assay, micronucleus, and nuclear abnormality tests on blood samples. The results showed that fish from both reservoirs are subjected to oxidative stress and genotoxic damage, mainly during winter, but fish living in Billings showed greater alterations than fish from Guarapiranga. Likewise, the results of the principal component analysis suggested that caffeine, nitrogenous compounds, and some metals might be triggering these toxic effects in fish.

Abstract:Coastal environments are faced with constant habitat modification and biodiversity loss due to human occupation. Estuaries are considered to be highly productive and shelter heterogeneous habitats such as mangrove forests, tidal creeks, tidal mud flats and sandy beaches, which are important for the maintenance of natural resources. The present study aims to evaluate how different shallow juvenile habitats (sandy beaches, mangrove shoreline habitats and dense macrophyte habitats) influence fish during their early life stages in a tropical estuary of Brazil. Monthly samples using a beach seine net from July 2012 to June 2013 were collected in eight shallow areas of the São Mateus River estuary. The fish assemblage comprised mainly juvenile and small-sized fish, which included ecologically and commercially important species. Small recruits of Atherinella brasiliensis and Ctenogobius boleosoma were most abundant in the autumn, while Rhinosardinia bahiensis and Centropomus undecimalis were highly abundant during the winter. The water salinity recorded in the mid and lower estuarine portions was the most important variable for the distribution of A. brasiliensis and Sphoeroides spengleri , whereas Anchovia clupeoides , C. undecimalis , C. parallelus and Gobionellus oceanicus were positively correlated with the water depth of the upper estuarine portion. The fish species T. paulistanus , Genidens genidens and Achirus lineatus were related to the water turbidity in the upper portion. Thus, the heterogeneous shallow water habitats (and their associated environmental parameters) of the São Mateus River estuary are potential nursery grounds for the early life stages of a variety of fish species, which highlights the importance of these areas for local biodiversity management.

Abstract:Initiating licensure and medical education in Croatia was not only schooling itself, but struggle for national identity and institution of an academic setting, which, by itself, is paramount for cultural development anywhere. Throughout history, this struggle mostly ended with administrative discouragement and opposition to all such efforts of higher medical education by contemporary authorities. The paper elaborates on the first initiatives in this direction focused on the establishment of a degree of medical education in Dalmatia. Doctor Jakov Mirković (1748(?)-1824) was the first to become instrumental in the area. The aims, outlines and academic basis of those first initiatives were presented in a publication of his, leading eventually to the establishment of Split School of Medicine, University of Split, finally realized in 1997.

Abstract:Aquatic biota are exposed to toxic substances resulting from human activities, reducing environmental quality and can compromise the health of the organisms. This study aimed to employ Danio rerio as an environmental bioindicator, analyzing the effects of water from distinct urban aquatic environments. An active biomonitoring system was set up to compare the temporal dynamics of histological biomarkers for gill and liver and the patterns of non-protein thiols (NPSH) in muscle in specimens exposed for 3, 6, and 12 days. Three large urban basins in the city of Campo Grande (Midwest of Brazil) were selected. Two sites are in a very populous area (i.e Lagoa and Bandeira) and another on in an area with agricultural activities (i.e Anhanduí). All the streams displayed distinct qualitative characteristics. The presence of metals, including Mn, Zn, Fe, and Al, as well as pH, temperature, and dissolved oxygen, accounted for 38% of the variability (PC1), while total solids, conductivity, ammonia, nitrite, and explained 24 % (PC2). Degree tissue changes index (DTC) in gill and the concentration of NPSH increased in all streams during 3, 6 and 12 days of exposure. DTC in liver increases in all exposure times in most populous stream (i.e Lagoa and Bandeira). Histopathological evidence in the gill, including proliferation, desquamation, and necrosis of the primary lamellar epithelium; fusion and aneurysms in the secondary lamellar epithelium were observed after three days of exposure. Degenerative nuclear figures were noted in the liver after three days of exposure, followed by hepatocellular hypertrophy, lipidosis, and necrosis at twelve days. Our findings showing time-dependent effects of urban aquatic environments in histopathological (i.e DTC) and biochemical biomarkers in zebrafish. The biomonitoring model enabled a comparison of the temporal dynamics of various health markers, using zebrafish as bioindicator. Future studies might use this experimental model and biomarkers for environmental biomonitoring program.
Hatchery fish stocking: case study, current Brazilian state, and suggestions for improvement

Abstract:Hatchery fish stocking is one of the main actions adopted as a form of conservation and replacement of fishing stocks. Under Brazilian law, the release of fish has become mandatory and is seen as one of the main ways of mitigating the negative effects on fish populations and preserving the ichthyofauna. Since its institution, the efficiency of this method has been questioned by the Brazilian scientific community, due to the lack of monitoring based on scientific criteria. The objective of this work was to discuss the few available reports and to analyze their deficiencies and analyze the real risks and/or benefits arising from the methodologies employed in hatchery stocking activities developed in Brazil. For this, three different studies developed in the Paranapanema River, Southeast/South of Brazil, were evaluated seeking evidence of the efficiency or inefficiency of these actions, such as management of the conservation of fish species and stocks. Our analysis shows that this management policy is poorly evaluated, and review of the planning of the stocking programs is essential, in order to improve, update, modernize, and unify the knowledge of the producing stations with respect to the existing ecological and genetic studies, aiming at better monitoring and greater effectiveness in the results advance. Also, a protocol is suggested to standardize and guide new conservationist policies of hatchery fish stocking.

Integrated analysis of the quality of water bodies from the lower Paraná River basin with different productive uses by physicochemical and biological indicators

Abstract:The Paraná River basin is one of the most important in South America and is affected by human activities that take place on its margins. In particular, the De la Cruz stream flows through an industrial pole and the Arrecifes River goes mainly through agricultural fields. The aim of this study was to evaluate the water quality of the De la Cruz stream (S1) and the Arrecifes River (S2) by means of physicochemical parameters, including metals and pesticides concentrations. Since amphibians are good indicators of environmental quality, bioassays with Rhinella arenarum were carried on. For lethal and sublethal parameters, embryos and larvae were exposed to a dilution gradient of water samples and AMPHITOX Solution (AS) as negative control for 504 h. For the determination of oxidative stress biomarkers (Catalase -CAT-, Glutathione S-Transferase -GST-, Reduced Glutathione -GSH-, and lipid peroxidation -TBARS-), embryos and larvae were exposed to undiluted water samples and AS. For the determination of micronuclei, larvae at hind limb bud stage (S.28) were exposed to undiluted water samples, simultaneously with negative and positive controls (AS and cyclophosphamide 40 mg/L, respectively). Dissolved oxygen was low in both sites and the copper levels exceeded the Argentine limit for the protection of aquatic life. In embryos exposure, water sample from S1 caused lethal effects (504h-LC50 = 49 (28–71.6)%), increased TBARS levels, and GST and CAT activities. In larvae exposure, water sample from this site decreased CAT activity, while the water sample from S2 caused important lethal effects (504h-LC50 = 98.72 (60.60–302.52)%), low GSH levels and increased GST activity. Water samples from both sites induced higher micronuclei frequency than the negative control. This study alerts about the degradation of water quality of the studied sites including lethal and sublethal effects in R. arenarum that can jeopardize the native populations of this species.

Abstract:Biodiversity is declining worldwide due to anthropogenic impacts, especially noxious for freshwater ecosystems, considering their close relationship with human activities. Damming is one of the most harmful human impacts that leads to the loss of several riverine fish through habitat loss or change. Herein, we aimed to assess the relationship between the composition of fish assemblages in reservoirs and the availability of littoral habitats, and whether these patterns differ between native and non-native species. Fish assemblages of fifteen reservoirs from up to downstream the Paraíba do Sul river basin were evaluated. Habitat availability was more important for native than for non-native species, after controlling for the influence of reservoir features. Local drivers were crucial for the occurrence of non-native species which thrived in the absence of complex habitats. Macrophytes seemed to play two key but quite antagonistic roles for fish species. They were positively related to native small characids, probably acting as shelter against predation, but they were also related to hypoxic conditions in eutrophic reservoirs. These findings stress the importance of preserving or restoring physically complex habitats as beneficial for native fish species through providing grounds for feeding (catfishes on leaves) and recruitment (juveniles of characids in macrophytes banks).

Multiple biomarker responses in Prochilodus lineatus subjected to short-term in situ exposure to streams from agricultural areas in Southern Brazil

Abstract:In order to assess the quality of streams susceptible to contamination by pesticides we apply biochemical and genotoxic biomarkers in the Neotropical fish Prochilodus lineatus submitted to in situ tests. Fish were caged, for 96 h, in two streams located in areas with intensive use of pesticides, the Apertados (AP) and the Jacutinga (JC), and in a small stream (Godoy stream — GD) found inside a forest fragment adjacent to a State Park. Biochemical parameters, such as biotransformation enzymes 7-ethoxyresorufin-O-deethylase (EROD) and glutathione-S-transferase (GST), non-protein thiols (NPSH), lipoperoxidation (LPO), protein carbonylation (PCO) and acetylcholinesterase (AChE) were evaluated in various fish organs, as well as genotoxic biomarkers (damage to DNA and occurrence of micronuclei and erythrocyte nuclear abnormalities). Samples of water and sediment were collected for analysis of metals (Cu, Cr, Pb, Ni, Mn, Cd and Zn), organochloride pesticides, and triazine and glyphosate herbicides. We observed an increase in liver GST activity in fish at AP and gill GST activity in fish at JC. An increase in liver LPO was also observed in fish exposed to AP and JC. The same animals also exhibited increased DNA damage and erythrocyte nuclear abnormalities (ENAs) compared to the fish kept in GD. A number of compounds showed concentrations higher than the permitted levels, in particular, dichlorodiphenyltrichloroethane (DDT), its metabolites dichlorodiphenyldichloroethylene (DDE) and dichlorodiphenyldichloroethane (DDD), hexachlorocyclohexanes (HCH), heptachloride, diclofluanid and aldrins. These pesticides were detected at higher concentrations in water and sediment samples from AP, followed by JC and GD. The Integrated Biomarker Response Index (IBR) indicated that AP and JC (AP: 21.7 > JC: 18.5 > GD: 12.6) have the worst environmental quality. Integrated biomarker analysis revealed that the alterations observed related well with the levels of environmental contaminants, demonstrating the effectiveness of this biomonitoring approach.

Abstract:Over the past few decades, South American Rivers, including the Paraná River system, have been intensely impacted by dam constructions. To mitigate adverse impacts, fish passages were incorporated into two large dams on the Paraná River (Itaipu and Porto Primavera) to reestablish longitudinal connectivity, which is very important for conserving populations of migratory fish species. The aim of this study was to estimate genetic diversity, population differentiation and gene flow, using microsatellites markers and mitochondrial haplotypes (d-loop), in populations of Prochilodus lineatus, a long distance migratory species commonly found in Paraná River basin drainages. Samples were obtained from six locations in the Upper and Middle Paraná River: Porto Primavera Reservoir, downstream of the Porto Primavera Reservoir, four tributaries of this reservoir (Pardo, Peixe, Verde and Aguapeí Rivers) and in the Iguaçu River, downstream of the Itaipu dam. High genetic diversity was observed in all samples, as expected for species with large population sizes and high migration rates. However, the influence of other factors, such as secondary contacts, fish passages and restocking were also suggested. In general, evidence of spatial population structuring was not found, even though there were two large dams in the study area. This could also be attributed to the brief period that has elapsed since the dams were built, the influence of migratory and reproductive patterns and the possible contributory effects of the fish passages. Furthermore, the absence of population structuring also corroborates existing information that emphasizes the importance of unimpeded tributaries for conserving migratory fish populations in fragmented watersheds.

Abstract:Rivers interlinking by transposition projects have multiplied in the world, altering the landscape and flow regime of continental water systems. Recently, an intermittent river in northeastern Brazil has been artificially connected to the São Francisco River basin in a water transfer project. Thus, this study aims to provide an inventory of the fish parasites from the Jaguaribe River basin in the scenarios of pre- and post-transposition, and to evaluate the short-term impacts on the parasite fauna. Of the analyzed fishes, belonging to 31 species, about 30% were parasitized. A total of 47 parasite taxa were recovered, including monogeneans (17), digeneans (9), nematodes (9), acanthocephalans (3) and crustaceans (6), and unidentified cysts of Acanthocephala, Cestoda and Digenea, representing the description of 30 new geographic records and 104 new parasite-host associations. By expanding the range of the geographic distribution of fish parasites and increasing the list of hosts, the study contributes to the knowledge of fish parasites biodiversity in the semi-arid region of Brazil and in the Caatinga domain. Considering all parasite species, significant differences were observed in total prevalence, total mean abundance, diversity index and host specificity index; the highest values of total prevalence and total mean abundance were observed in the post-transposition, while the diversity index and host specificity index was higher in the pre-transposition period. This is the first study evaluating fish parasites before and after a major water transfer event and the results will allow further evaluation of middle and long-term changes in the specific composition of fish and parasites from the receiver drainages.

Abstract:The use of biomarkers in fish for biomonitoring is a valuable approach to reveal effects of human impacts on biota health. Top predator fish are effective models for monitoring human activities' impacts on aquatic ecosystems. The Guaraguaçu River is the largest river-system on coastal region of South Brazil and a World Heritage site. The river receives contaminants from disorderly urban growth, including discharges of domestic sewage and small fishery boats, particularly during the tourist season. Our study aimed to assess impact of anthropogenic activities on water quality in the Guaraguaçu River by analyzing environmental contamination biomarkers in the top fish predator Hoplias malabaricus . Fish were collected using a fyke net trap across sectors representing a gradient of anthropic impact: sector 1 - pristine; sector 2 – impacted; and sector 3 - less impacted. Water samples were collected to analyze the presence of trace elements and pesticide. Biomarkers of the antioxidant system, histopathology, genotoxicity, neurotoxicity, and concentration of trace elements were analyzed in fish tissues. In water samples Al, Fe and Mn were detected, but no pesticides were found. In fish muscle, zinc and iron were detected. Brain acetylcholinesterase activity decreased in impacted sectors, indicating neurotoxic effects. The antioxidant system increased activity in gills and liver, and damage from lipoperoxidation was observed, particularly in sector 2 when compared to sector 1, suggesting oxidative stress. Histopathological biomarkers revealed lesions in the liver and gills of fish in impacted sectors. Micronuclei, a genotoxicity biomarker, were observed in organisms from all sectors. Our results demonstrate detrimental effects of poor water quality on biota health, even when contaminants are not detected in water.

Abstract:The most recent dam rupture in Brazil released tons of mining tailings into the upper course of the Paraopeba River, affecting this river in an unprecedented way. The present study aimed to evaluate the influence of heavy metals on Prochilodus costatus , an important commercial species in Brazil, four years after the dam colapse. To this end, biomarkers of heavy metals, oxidative stress, and environmental stress were analyzed, and histological analyses of target organs were performed. The results demonstrated critical contamination of fish from the Paraopeba River. Increased expression of Metallothioneins - MTs, Heat Shock Protein - HSP70, and inducible nitric oxide synthase - iNOS, as well as greater rates of histological changes in the liver, spleen, and gonads, were observed in P. costatus . These findings demonstrate that, despite past contamination, the metals present in mining tailings have significantly increased the contamination of the Paraopeba River basin.

Abstract:Agriculture is one of the main factors associated with global biodiversity loss. The present study assesses the impact of horticultural land use on water quality and fish assemblage composition in lowland Pampean streams. Four samplings were conducted with seasonal frequency during 2018 and 2019 in three horticultural stream reaches (HSR) and three grassland reaches (GSR) with low anthropic intervention, two also located in a biosphere reserve area. Environmental parameters were determined, and water and sediment samples were taken for nutrient and pesticide analysis, respectively. Fish assemblages were sampled in two habitat types: open water (OW) and vegetated patches (VP). The concentration of soluble reactive phosphorus and the detection frequency and concentration of pesticides were significatively higher in HSR mainly attributed to the runoff events following application of fertilizers and pesticides to the surrounding crops. Specific richness was significantly higher in GSR for both OW and VP environments. Diversity and evenness in the VP were significantly higher in GSR, while abundance was significantly higher in HSR due to the dominance of the environmental pollution-tolerant taxa of the order Cyprinodontiformes. The multivariate analysis recorded a major contribution of the order Cyprinodontiformes in the HSR, while the GSR species of the order Characiformes, common in environments of low environmental impact, were better represented. Our study demonstrated water quality deterioration associated with horticultural land use resulting in a ruderal fish community. On-going horticulture expansion is expected to cause further deterioration within the region, unless managerial practices are implemented.

Abstract:The introduction of non-native aquarium fish species has modified the composition of fish assemblages worldwide. This anthropogenic impact alters not only the richness/abundance of local assemblages but also the taxonomic and functional composition among them. To investigate this phenomenon, we conducted a standardized sampling program in degraded headwater creeks located in the largest ornamental aquaculture center of Brazil, with the aim of quantifying spatial and temporal changes in taxonomic and functional diversity induced by fish escapes. Data were organized in two periods: "historical", which included the native species pool of the region, and "current" (divided in "past" and "present"), which included all native and non-native species recorded during 2015–2017 (past) and 2022 (present). Fish assemblages showed taxonomic and functional differentiation through time, and the continuous introduction of non-native fish from 2015 to 2022 contributed to increase species richness in local assemblages. Trait diversity did not differ across native/non-native fish assemblages, and non-native assemblages had smaller body sizes than natives. Our study provides strong evidence that escapes of non-native ornamental fish induces changes in taxonomic and functional fish diversity in small creeks, representing a significant threat to these fragile ecosystems.
