Abstract:Objective The transformation of BZR1, a BR signal transduction component to tomato and its effect on characters of fruit of tomato was investigated in present study. Method The bzr1 gene was introduced into tomato cultivar Zhongshusihao via tumefaciens-mediated transformation and confirmed by molecular test and genetic analysis. The content of main quality components and pigments, as well as ethylene evolution rate was analyzed in fruits of transgenic tomato line and non-transgenic line (control) at four typical developmental stages. The expression of BR biosynthetic and regulated genes, as well as ethylene biosynthetic, response, and receptor genes was also analyzed. Result Significantly higher contents of soluble solids, organic acid, soluble proteins, vitamin C, lycopene and total carotenold, as well as the ethylene evolution rate in fruit of turning stage were observed in transgenic tomato line compared with the control. The expression of BR regulated gene LeBR1 was higher in transgenic line than in the control at the stage of seedling and early three developmental stages of fruit. The expression of LeACS4 gene encoding ACC synthase, which catalyzes the rate-limiting step of ethylene biosynthesis, was higher in the fruit of transgenic line than that of control from the stage of mature green to turning. The expression of ethylene response gene ERF1 and receptor gene LeETR1 and LeETR3 in fruit of breaker stage was significantly higher in transgenic line than in the control. Conclusion It was concluded from the study that the transformation of bzr1gene to tomato increased the content of various nutrients in fruit of turning stage, which is the edible part of tomato, and therefore improved the nutrient quality of tomato.
Regulation of Fruit Ripening by the Brassinosteroid Biosynthetic Gene SlCYP90B3 Via an Ethylene-Dependent Pathway in Tomato

Abstract:The essential role of ethylene in fruit ripening has been thoroughly studied. However, the involvement of brassinosteroids (BRs) in the regulation of fruit ripening and their relationship with the ethylene pathway are poorly understood. In the current study, we found that BRs were actively synthesized during tomato fruit ripening. We then generated transgenic lines overexpressing or silencingSlCYP90B3, which encodes a cytochrome P450 monooxygenase that catalyzes the rate-limiting step of BR synthesis. The expression level ofSlCYP90B3was positively related to the contents of bioactive BRs as well as the ripening process in tomato fruit, including enhanced softening and increased soluble sugar and flavor volatile contents. Both carotenoid accumulation and ethylene production were strongly correlated with the expression level ofSlCYP90B3, corroborated by the altered expression of carotenoid biosynthetic genes as well as ethylene pathway genes in transgenic tomato fruits. However, the application of the ethylene perception inhibitor 1-methycyclopropene (1-MCP) abolished the promotion effect ofSlCYP90B3overexpression on carotenoid accumulation. Taken together, these results increase our understanding of the involvement ofSlCYP90B3in bioactive BR biosynthesis as well as fruit ripening in tomato, thus makingSlCYP90B3a target gene for improvement of visual, nutritional and flavor qualities of tomato fruits with no yield penalty.
Regulation of Tomato Fruit Autophagic Flux and Promotion of Fruit Ripening by the Autophagy-Related Gene SlATG8f

Abstract:Autophagy is a highly conserved self-degradation process that involves the degradation and recycling of cellular components and organelles. Although the involvement of autophagy in metabolic changes during fruit ripening has been preliminarily demonstrated, the variations in autophagic flux and specific functional roles in tomato fruit ripening remain to be elucidated. In this study, we analyzed the variations in autophagic flux during tomato fruit ripening. The results revealed differential expression of the SlATG8 family members during tomato fruit ripening. Transmission electron microscopy observations and dansylcadaverine (MDC) staining confirmed the presence of autophagy at the cellular level in tomato fruits. Furthermore, the overexpression of SlATG8f induced the formation of autophagosomes, increased autophagic flux within tomato fruits, and effectively enhanced the expression of ATG8 proteins during the color-transition phase of fruit ripening, thus promoting tomato fruit maturation. SlATG8f overexpression also led to the accumulation of vitamin C (VC) and soluble solids while reducing acidity in the fruit. Collectively, our findings highlight the pivotal role of SlATG8f in enhancing tomato fruit ripening, providing insights into the mechanistic involvement of autophagy in this process. This research contributes to a better understanding of the key factors that regulate tomato fruit quality and offers a theoretical basis for tomato variety improvement.
Transcriptome profiles reveal new regulatory factors of anthocyanin accumulation in a novel purple-colored cherry tomato cultivar Jinling Moyu

Abstract:Anthocyanins are important secondary metabolites with high nutraceutical value. They represent the major red, purple, violet and blue pigments in higher plants but are usually not present in the fruits of domesticated tomatoes. Recently, several genes that regulate anthocyanin accumulation have been identified in wild tomatoes or mutation lines; however, whether there are other new factors involved in the regulation of anthocyanin biosynthesis is still unknown. “Jinling Moyu” (MY) is a novel purple cherry tomato that accumulates considerable anthocyanins in its ripe fruit. A transcriptome comparison analysis of MY fruits with pink and green fruit revealed that several structural genes assigned to the anthocyanin biosynthetic pathway were highly expressed in MY tomato, but all six known regulatory genes of anthocyanin accumulation in tomato fruits were rarely expressed among the samples in this study. However, four other R2R3–MYB and five other bHLH transcription factors were highly correlated with the differentially expressed structural genes in the anthocyanin pathway, indicating that these transcription factors might regulate anthocyanin biosynthesis in MY tomato. Our study will aid in understanding the complex mechanism regulating anthocyanin pigmentation in tomato fruits .

Abstract:Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality. However, the use of a single gene to impact several metabolic pathways is difficult. Here, we show that overexpression of the single gene SlMYB75 (SlMYB75-OE) is effective at improving multiple fruit quality traits. In these engineered fruits, the anthocyanin content reached 1.86 mg g−1 fresh weight at the red-ripe stage, and these SlMYB75-OE tomatoes displayed a series of physiological changes, including delayed ripening and increased ethylene production. In addition to anthocyanin, the total contents of phenolics, flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6, 4, and 1.2 times, respectively, compared to those of wild-type (WT) fruits. Interestingly, a number of aroma volatiles, such as aldehyde, phenylpropanoid-derived and terpene volatiles, were significantly increased in SlMYB75-OE fruits, with some terpene volatiles showing more than 10 times higher levels than those in WT fruits. Consistent with the metabolic assessment, transcriptomic profiling indicated that the genes involved in the ethylene signaling, phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits. Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC, AADC2 and TPS genes. The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.

Abstract:Background Tomato quality is a complex trait influenced by multiple genes involved in various complicated metabolic pathways. Materials and Methods This study sought to improve tomato taste and quality by grafting on goji rootstock. We integrated physiological and biochemical indicators, as well as metabolic and transcriptomic analysis, to evaluate tomato quality. Results The grafted tomatoes showed significant increases in soluble solids (51.48%), vitamin C (16.86%), soluble protein (16.19%), titratable acid (11.73%), anthocyanin (11.43%), and polysaccharide (9.43%) content compared to those of the control. Metabolomics analysis identified 234 differentially accumulated metabolites and transcriptome analysis identified 4213 differentially expressed genes between grafted and control tomatoes at three ripening stages (mature green, turning, and fully red). A total of 93 phenolic compounds, including flavonoids and phenolic acids, showed differential accumulation patterns between grafted and control tomatoes at the fully red stage. These changes were attributed to the upregulation of key genes (F3'H, F3'5'H, DFR, and ANS) associated with flavonoids and anthocyanin synthesis in the goji rootstock compared to those of the control. The expression of genes involved in sucrose hydrolysis and starch synthesis, including invertase (INV), sucrase synthase (SUS), and beta-amylase (BAM) genes, were suppressed by goji grafting, resulting in increased levels of sugars. In addition, the consistency between the transcriptomic and metabolomic data provided a robust validation of the observed quality changes. Conclusions Taken together, our results demonstrate that grafting onto goji rootstock improves tomato quality by modulating multiple genes involved in phenylpropanoid, sucrose, and starch pathways during fruit development, providing valuable insights for improving the quality and taste of tomato. Arabidopsis thaliana MYB75/PAP1 transcription factor induces anthocyanin production in transgenic tomato plants

Abstract:Tomato (Solanum lycopersicum L.) cv. Micro-Tom plants were transformed with the Arabidopsis thaliana (L.)Heyhn. MYB75/PAP1 (PRODUCTION OF ANTHOCYANIN PIGMENT 1) gene. This gene encodes for a well known transcription factor, which is involved in anthocyanin production and is modulated by light and sucrose. Transgenic tomato plants expressing AtMYB75 were characterised by a significantly higher anthocyanin production in leaves, stems, roots and flowers under normal growth conditions. Further, they also exhibited anthocyanins in fruits. Anthocyanin accumulation was not widespread but took place in specific groups of cells located in epidermal or cortical regions or in proximity of vascular bundles. In all the organs of the transgenic plants, where AtMYB75 overexpression was determined, a clear increase in the accumulation of DFR (DIHYDROFLAVONOL 4-REDUCTASE) transcript was also detected. The expression of the tomato MYB-gene ANT1 (ANTHOCYANIN1), which had previously been identified as a transcriptional endogenous regulator of anthocyanin biosynthesis, was not altered. The higher basal content of anthocyanins in the leaves of the transgenic plants could be further increased in the presence of high light conditions and contributed to mitigate photobleaching damages under high irradiance.
Constitutive expression of SlMX1 gene improves fruit yield and quality, health-promoting compounds, fungal resistance and delays ripening in transgenic tomato plants

Abstract:Tomato is one of the major economically domesticated crops, and it is extensively used in different ways and purposes worldwide. Cell metabolism is the central core of all the biological processes to sustain life including cell growth, differentiation, maintenance, and response to environmental stress. To evaluate how genetic engineering can improve tomato fruit metabolome, the transcriptomic and metabolomic datasets of two transgenic tomatoes (SlMX1 overexpression and RNAi lines) have been compared with wild-type. The combined results demonstrated that the constitutive expression of SlMX1 not only increased trichome formation, carotenoids, and terpenoids as has been stated in several studies, but has also up- and down-regulated the expression of multiple genes related to cell growth (cell wall turnover), primary (carbohydrates, vitamins, and phytohormones), and secondary (phenylpropanoids, carotenoids, and terpenoids) metabolism, cell signaling, and stress responses. These changes in gene expression due to the constitutive expression of SlMX1 promote the most important agroeconomic traits such as fruit yield and quality, biosynthesis of health-promoting phytochemicals (including phenolic acids, flavonoids, and anthocyanins), and finally, activate resistance to Botrytis cinerea and repress the expression of over-ripening-related genes, thus extending the fruit shelf-life. In conclusion, the traits improvement achieved by SlMX1 overexpression can be harnessed in molecular breeding programs to engineer fruit size and yield, induce health-promoting secondary metabolites, promote fungal resistance, and finally extend the fruit shelf-life.

Heterologous Expression of BoPAP1 in Tomato Induces Stamen Specific Anthocyanin Accumulation and Enhances Tolerance to a Long-Term Low Temperature Stress

Abstract:Anthocyanins are ubiquitous plant secondary products that possess nutritive values and serve essential functions in plant defense. The purple kale (Brassica oleracea var. acephala f. tricolor) BoPAP1 gene, which encodes a MYB transcription factor associated with anthocyanin biosynthesis, was introduced into tomato (Solanum lycopersicum) under the control of the CaMV 35S promoter. Transgenic tomato plants exhibited restricted accumulation of anthocyanins in stamens under normal growth conditions. qPCR analysis revealed that the expression of the anthocyanin biosynthetic genes such as SlF3′5′H, SlDFR, and SlANS, as well as a bHLH gene SlTT8, was specifically induced in transgenic stamens. A yeast two-hybrid assay revealed that BoPAP1 was a transcription activator and could interact with SlTT8. Despite the absence of anthocyanins in seedlings under normal conditions, overexpression of BoPAP1 led to high levels of anthocyanins and chlorophylls in the transgenic seedlings that were exposed to low temperature for 2 weeks, which conferred chilling tolerance on transgenic tomato plants. The consistency of SlTT8 expression and anthocyanin accumulation was observed in both purple stamens and low temperature treated seedlings in transgenic lines. These results suggest that the endogenous bHLH gene SlTT8 could be a candidate bHLH gene involved in the tomato anthocyanin pathway. Considering that the inducible type of anthocyanins apparently does not interfere with the normal growth of the plant; it provides an optimized transgenic strategy for crop improvement.

Abstract:Flavonols are one class of flavonoids that play a crucial role in regulating plant growth and promoting stress resistance. They are also important dietary components in horticultural crops due to their benefits for human health. However, flavonol levels in commonly consumed horticultural crops are insufficient to provide optimal benefits, and their role in regulating postharvest pathogen resistance in fruit remains poorly understood. In this study, overexpression of MrMYB12 or MrMYB12/MrF3'5'H from Morella rubra in tomato (Solanum lycopersicum) resulted in high accumulation of flavonols in fruit up to 3.2 g kg-1 by upregulating expression of genes involved in the phenylpropanoid pathway including SlPAL, SlC4H, Sl4CL, SlCHS1, SlCHI-like, SlF3H, and SlFLS. Transgenic tomato fruit showed high free radical scavenging activities including 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric ion reducing antioxidant power (FRAP) and 2,2 '-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and they also possessed excellent inhibitory effects against alpha-glucosidase. Postharvest storage experiments indicated that the flavonol-rich tomato fruit exhibited less surface shrinkage than wild type (WT), and they also showed decreased weight loss and increased antioxidant activity. Experiments of Botrytis cinerea infection showed that overexpression of MrMYB12 or MrMYB12/MrF3'5'H in tomato improved their resistance against B. cinerea during postharvest storage. Improvement of B. cinerea resistance in the flavonol-rich tomato fruit was associated with increase of superoxide dismutase (SOD) and catalase (CAT) enzyme activities and upregulated expression of defense-related genes such as SlPAL, Sl beta CA3, SlNPR1, SlPR1a, SlPR1b1, and SlPR3a. This study provides new insights for improving the nutritional value and postharvest pathogen resistance of horticultural crops by metabolic engineering.
Different Fruit-Specific Promoters Drive AtMYB12 Expression to Improve Phenylpropanoid Accumulation in Tomato

Abstract:Tomato is an economically crucial vegetable/fruit crop globally. Tomato is rich in nutrition and plays an essential role in a healthy human diet. Phenylpropanoid, a critical compound in tomatoes, reduces common degenerative and chronic diseases risk caused by oxidative stress. As an MYB transcription factor, ATMYB12 can increase phenylpropanoid content by activating phenylpropanoid synthesis related genes, such as PAL, C4H, 4CL, CHS. However, the heterologous expression of AtMYB12 in tomatoes can be altered through transgenic technologies, such as unstable expression vectors and promoters with different efficiency. In the current study, the efficiency of other fruit-specific promoters, namely E8S, 2A12, E4, and PG, were compared and screened, and we determined that the expression efficiency of AtMYB12 was driven by the E8S promoter was the highest. As a result, the expression of phenylpropanoid synthesis related genes was regulated by AtMYB12, and the phenylpropanoid accumulation in transgenic tomato fruits increased 16 times. Additionally, the total antioxidant capacity of fruits was measured through Trolox equivalent antioxidant capacity (TEAC) assay, which was increased by 2.4 times in E8S transgenic lines. TEAC was positively correlated with phenylpropanoid content. Since phenylpropanoid plays a crucial role in the human diet, expressing AtMYB12 with stable and effective fruit-specific promoter E8S could improve tomato’s phenylpropanoid and nutrition content and quality. Our results can provide genetic resources for the subsequent improvement of tomato varieties and quality, which is significant for human health.

Abstract:Polyamines play an important role in plant response to abiotic stress. S-adenosyl-l-methionine decarboxylase (SAMDC) is one of the key regulatory enzymes in the biosynthesis of polyamines. In order to better understand the effect of regulation of polyamine biosynthesis on the tolerance of high-temperature stress in tomato, SAMDC cDNA isolated from Saccharomyces cerevisiae was introduced into tomato genome by means of Agrobacterium tumefaciens through leaf disc transformation. Transgene and expression was confirmed by Southern and Northern blot analyses, respectively. Transgenic plants expressing yeast SAMDC produced 1.7- to 2.4-fold higher levels of spermidine and spermine than wild-type plants under high temperature stress, and enhanced antioxidant enzyme activity and the protection of membrane lipid peroxidation was also observed. This subsequently improved the efficiency of CO2 assimilation and protected the plants from high temperature stress, which indicated that the transgenic tomato presented an enhanced tolerance to high temperature stress (38 degrees C) compared with wild-type plants. Our results demonstrated clearly that increasing polyamine biosynthesis in plants may be a means of creating high temperature-tolerant germplasm.

Abstract:Over the past decade, progress has been made in the characterization of anthocyanin synthesis in fruits of plants belonging to the tomato clade. The genomic elements underlying the activation of the process were identified, providing the basis for understanding how the pathway works in these species. In this review we explore the genetic mechanisms that have been characterized to date, and detail the various wild relatives of the tomato, which have been crucial for recovering ancestral traits that were probably lost during evolution from green-purple to yellow and red tomatoes. This knowledge should help developing strategies to further enhance the status of the commercial tomato lines on sale, based on both genome editing and breeding techniques.

Abstract:Anthocyanins are specialized plant metabolites with significant dietary value due to their anti-inflammatory properties. Research indicates that dietary intake of these phenolic compounds contributes to preventing various chronic diseases. As the most consumed vegetable worldwide, tomato ( ) is an excellent candidate for anthocyanin-enrichment strategies. In tomato, activation of anthocyanin biosynthesis is light-dependent, but this mechanism has yet to be entirely characterized. We investigated the role of light in anthocyanin biosynthesis in fruits of the purple tomato, which is a near-isogenic line (NIL) derived from wild accessions into cv. Micro-Tom (MT). MT- starts accumulating anthocyanin early during fruit development but is restricted to the peel (exocarp and epicarp). Manipulating light incidence in different fruit tissues determined that the absence of anthocyanin accumulation in the flesh results from the sun-blocking effect of the cyanic epicarp on the mesocarp, thus preventing light from penetrating deeper into the fruit. Transcriptional analyses of the fruit peel and flesh revealed that the bHLH transcription factor SlAN1 (Solyc09g065100) is the limiting factor for light-dependent anthocyanin accumulation in both tissues. This research enhances our comprehension of the genetic and environmental regulation of anthocyanin accumulation in fruit tissues, offering valuable insights into plant breeding for human nutrition.

Abstract:Tomato fruit ripening is accompanied by carotenoid accumulation and color changes. To elucidate the regulatory mechanisms underlying carotenoid synthesis during fruit ripening, a combined transcriptomic and metabolomic analysis was conducted on red-fruited tomato (WP190) and orange-fruited tomato (ZH108). A total of twenty-nine (29) different carotenoid compounds were identified in tomato fruits at six different stages. The abundance of the majority of the carotenoids was enhanced significantly with fruit ripening, with higher levels of lycopene; (E/Z)-lycopene; and α-, β- and γ-carotenoids detected in the fruits of WP190 at 50 and 60 days post anthesis (DPA). Transcriptome analysis revealed that the fruits of two varieties exhibited the highest number of differentially expressed genes (DEGs) at 50 DPA, and a module of co-expressed genes related to the fruit carotenoid content was established by WGCNA. qRT-PCR analysis validated the transcriptome result with a significantly elevated transcript level of lycopene biosynthesis genes (including SlPSY2, SlZCIS, SlPDS, SlZDS and SlCRTSO2) observed in WP190 at 50 DPA in comparison to ZH108. In addition, during the ripening process, the expression of ethylene biosynthesis (SlACSs and SlACOs) and signaling (SlEIN3 and SlERF1) genes was also increased, and these mechanisms may regulate carotenoid accumulation and fruit ripening in tomato. Differential expression of several key genes in the fruit of two tomato varieties at different stages regulates the accumulation of carotenoids and leads to differences in color between the two varieties of tomato. The results of this study provide a comprehensive understanding of carotenoid accumulation and ethylene biosynthesis and signal transduction pathway regulatory mechanisms during tomato fruit development.

Abstract:Sugars act as signaling molecules to modulate various growth processes and enhance plant tolerance to various abiotic and biotic stresses. Moreover, sugars contribute to the post-harvest flavor in fleshy fruit crops. To date, the regulation of sugar metabolism and its effect in plant growth, fruit ripening, postharvest quality and resistance remains not fully understood. In this study, we investigated the role of tomato gene encoding a vacuolar invertase, hydrolyzing sucrose to glucose and fructose. SlVI is specifically expressed during the tomato fruit ripening process. We found that overexpression of SlVI resulted in increased leaf size and early flowering, while knockout of SlVI led to increased fruit sucrose content, enhanced fruit firmness, elevated resistance of postharvest fruit to Botrytis cinerea. Moreover, the content of naringenin and total soluble solids was significantly increased in SlVI knockout fruit at postharvest stage. Transcriptome analysis showed a negative feedback regulation triggered by sucrose accumulation in SlVI knockout fruit resulting in a downregulation of BAM3 and AMY2, which are critical for starch degradation. Moreover, genes associated with cell wall, cutin, wax, and flavonoid biosynthesis and pathogen resistance were upregulated in SlVI knockout fruit. Conversely, the expression levels of genes involved in cell wall degradation were decreased in knockout fruit. These results are consistent with the enhanced postharvest quality and resistance. Our findings not only provide new insights into the relationship between tomato fruit sucrose content and postharvest fruit quality, but also suggest new strategies to enhance fruit quality and extend postharvest shelf life.

Abstract:Tomato is a horticultural crop with an incomplete flavonoid metabolic pathway that does not typically accumulate anthocyanins in the fruit. In recent years, intensive studies of the loci Anthocyanin fruit (Aft) and atroviolacium (atv) have clarified the functions of positive regulators (R2R3-MYBs) and a negative regulator (CPC-MYB) in anthocyanin biosynthesis in the fruits. However, little is known about the R2R3-MYB repressors. Here, we used transient overexpression analysis to show that SlMYB7, a subgroup 4 AtMYB4-like R2R3-MYB, inhibited anthocyanin accumulation and reduced expression of anthocyanin synthase genes in the 'black pearl' tomato fruits, which usually accumulate high concentrations of anthocyanins. These findings revealed that SlMYB7 served as a repressor of anthocyanin production. Furthermore, SlMYB7 actively repressed SlANS expression by binding its promoter and passively inhibited anthocyanin synthesis by interacting with the basic helix-loop-helix (bHLH) proteins SlJAF13 and SlAN1, which are involved in the formation of MBW complexes. Thus, SlMYB7 and the MBW complex may coregulate the anthocyanin content of 'black pearl' tomato fruits via a negative feedback loop. These findings provide a theoretical basis for the future enhancement of tomato anthocyanin contents through genetic manipulation of the biosynthetic regulatory network.
Defective mutations of SlSGR1 , SlPSY1 and MYB12 genes lead to formation of green ripe fruit in tomato

Abstract:Modern tomatoes produce colourful mature fruits, but many wild tomato ancestors form green or gray green ripe fruits. In this study, tomato cultivar ' Lvbaoshi' (LBS) that produces green ripe fruits was found to contain three recessive loci that were responsible for development of green ripe fruits. The colourless peel of LBS fruits was caused by a 603-bp deletion in the promoter of SlMYB12. The candidate genes of the remaining two loci were identified as Stay-Green 1 ( SlSGR1 ) and Phytoene Synthase 1 ( SlPSY1 ). A single nucleotide substitution was present in sgr1 of LBS, resulting in disruption of mRNA splicing and truncation of the translated protein. A retrotransposon was found in the first exon of the psy1 locus in LBS, leading to the absence of any detectable PSY1 transcripts. Co-suppression of SGR1 and PSY1 by RNA interference (RNAi) converted the pink fruits into green ripe fruits in transgenic plants. An amino acid change in PSY1 and a deletion in the promoter of SGR1 were identified in many wild tomatoes bearing green or gray ripe fruits. Transgenic lines expressing ProSGR1 :: SGR1 from Solanum pennellii failed to convert the purple-flesh fruits into the red-flesh fruits. Overexpression of PSY1 from green ripe fruit wild tomatoes in LBS plants could only partially rescue the green ripe fruit phenotype of LBS. Downregulation of the Ripening Inhibitor ( RIN ) gene in the green-flesh ( gf ) plants led to development of green ripe fruits. This work uncovers a novel regulatory mechanism by which SlMYB12 , SlPSY1 and SlSGR1 regulate fruit colour in cultivated tomatoes and some wild tomato species.

Abstract:Background: Tomatoes are an important source of human health-promoting compounds, and efforts have been made to enhance their nutritional quality through conventional plant breeding or biotechnology. This study assessed the composition of volatile compounds, phenolics and carotenoids in two allele-introgressed tomato lines, an anthocyanin-rich purple tomato and a β-carotene-rich orange tomato, as well as a red tomato. Results: The purple tomato peel accumulated a high amount of anthocyanins, mainly petunidin 3-(p-coumaroyl)-rutinoside-5-glucoside, responsible for the purple color, and other flavonoids such as rutin and kaempferol. The orange tomato did not undergo changes in the flavonoid profile but accumulated a high amount of β-carotene, with impairment on lycopene. A total of 27 volatile compounds were detected in purple tomato, 38 in orange tomato and 39 in red tomato. They comprise terpenes, carbonyls, alcohols, esters and hydrocarbons. The difference in the volatile compound profiles of ripe fruits can be related to differences in some precursor contents in the introgression lines. Orange tomato accumulates volatiles from β-carotene cleavage, not detected in the red fruits. Otherwise, volatiles from lycopene were absent in orange tomato as a result of the inhibition on lycopene accumulation. Phenolic volatiles were higher in the purple tomato, which has the highest total phenolic content. Conclusion: The introgessed alleles seem to have a positive effect on the enrichment of ripe tomato in bioactive compounds such as anthocyanins and β-carotene, improving nutritional quality. However, the allele introgression resulted in marked changes in volatile compound profiles, whose impact on tomato flavor and consumer acceptability needs to be evaluated. © 2019 Society of Chemical Industry.
